RESUMO

The diversity of bacteria associated with alpine lichens was profiled. Lichen samples belonging to the Umbilicariaceae family, commonly known as rock tripe lichens, were gathered from two distinct alpine fellfields: one situated on Mt. Brennkogel located in the Eastern European Alps (Austria), and the other on Mt. Stanley located in the Rwenzori mountains of equatorial Africa (Uganda). The primary aim of this research was to undertake a comparative investigation into the bacterial compositions, and diversities, identifying potential indicators and exploring their potential metabolisms, of these lichen samples. Bulk genomic DNA was extracted from the lichen samples, which was used to amplify the 18S rRNA gene by Sanger sequencing and the V3-V4 region of the 16S rRNA gene by Illumina Miseq sequencing. Examination of the fungal partner was carried out through the analysis of 18S rRNA gene sequences, belonging to the genus Umbilicaria (Ascomycota), and the algal partner affiliated with the lineage Trebouxia (Chlorophyta), constituted the symbiotic components. Analyzing the MiSeq datasets by using bioinformatics methods, operational taxonomic units (OTUs) were established based on a predetermined similarity threshold for the V3-V4 sequences, which were assigned to a total of 26 bacterial phyla that were found in both areas. Eight of the 26 phyla, i.e. Acidobacteriota, Actinomycota, Armatimonadota, Bacteroidota, Chloroflexota, Deinococcota, Planctomycetota, and Pseudomonadota, were consistently present in all samples, each accounting for more than 1% of the total read count. Distinct differences in bacterial composition emerged between lichen samples from Austria and Uganda, with the OTU frequency-based regional indicator phyla, Pseudomonadota and Armatimonadota, respectively. Despite the considerable geographic separation of approximately 5430 km between the two regions, the prediction of potential metabolic pathways based on OTU analysis revealed similar relative abundances. This similarity is possibly influenced by comparable alpine climatic conditions prevailing in both areas.

This paper demonstrates a compressed sensing-based single-shot hyperspectral imaging system for combustion diagnostics. The hyperspectral system can capture well-resolved spectra in a 2D plane through a single shot, i.e., converting a 3D data cube of 2D spatial and 1D spectral information into a compressed 2D hyperspectral image. Experimentally, the light emissions are first coded by a random binary pattern to generate the hyperspectral content, which is then sent through a spectrometer. The resulting compressed hyperspectral image is computationally analyzed to recover original 2D spatial and 1D spectral information. C2∗ and CH∗ chemiluminescence emissions of a methane/air flame at various equivalence ratios are measured using the compressed hyperspectral imaging technique. Comparison to traditional measurements shows good agreement in the correlation of emission ratio to equivalence ratio. The technique can be further applied to other laser-based combustion diagnostics.

Lichens are not only fungal-algal symbiotic associations but also matrices for association with bacteria, and the bacterial diversity linked to lichens has been receiving more attention in studies. This study compares the diversity and possible metabolism of lichen-associated bacteria from saxicolous foliose and fruticose taxa Alectoria, Canoparmelia, Crocodia, Menegazzia, Usnea, and Xanthoparmelia from the Venezuelan Guiana Shield and the South African Highveld Plateau. We used DNA extractions from the lichen thalli to amplify the eukaryotic 18S rRNA gene (rDNA) and the V3-V4 region of the bacterial 16S rDNA, of which amplicons were then Sanger- and MiSeq-sequenced, respectively. The V3-V4 sequences of the associated bacteria were grouped into operational taxonomic units (OTUs) ascribed to twelve bacterial phyla previously found in the rock tripe Umbilicaria lichens. The bacterial OTUs emphasized the uniqueness of each region, while, at the species and higher ranks, the regional microbiomes were shown to be somewhat similar. Nevertheless, regional biomarker OTUs were screened to predict relevant metabolic pathways, which implicated different regional metabolic features.

A fundamental feature of vocal communication is that animals produce vocalizations with different acoustic features in different behavioral contexts (contact calls, territorial calls, courtship calls, etc.). The midbrain periaqueductal gray (PAG) is a key region that regulates vocal production, and artificial activation of the PAG can elicit the production of multiple species-typical vocalization types.1,2,3,4,5,6,7,8,9 How PAG circuits are organized to regulate the production of different vocalization types remains unknown. On the one hand, studies have found that partial PAG lesions abolish the production of some vocalization types while leaving others intact,3,8,10,11 suggesting that different populations of PAG neurons might control the production of different vocalization types. On the other hand, electrophysiological recordings have revealed individual PAG neurons that increase their activity during the production of multiple vocalization types,12,13,14 suggesting that some PAG neurons may regulate the production of more than one vocalization type. To test whether a single population of midbrain neurons regulates the production of different vocalization types, we applied intersectional methods to selectively ablate a population of midbrain neurons important for the production of ultrasonic vocalizations (USVs) in mice. We find that, although ablation of these PAG-USV neurons blocks USV production in both males and females, these neurons are not required for the production of distress calls. Our findings suggest that distinct populations of midbrain neurons control the production of different vocalization types.

Vision-and-language navigation requires an agent to navigate in a photo-realistic environment by following natural language instructions. Mainstream methods employ imitation learning (IL) to let the agent imitate the behavior of the teacher. The trained model will overfit the teacher's biased behavior, resulting in poor model generalization. Recently, researchers have sought to combine IL and reinforcement learning (RL) to overcome overfitting and enhance model generalization. However, these methods still face the problem of expensive trajectory annotation. We propose a hierarchical RL-based method-discovering intrinsic subgoals via hierarchical (DISH) RL-which overcomes the generalization limitations of current methods and gets rid of expensive label annotations. First, the high-level agent (manager) decomposes the complex navigation problem into simple intrinsic subgoals. Then, the low-level agent (worker) uses an intrinsic subgoal-driven attention mechanism for action prediction in a smaller state space. We place no constraints on the semantics that subgoals may convey, allowing the agent to autonomously learn intrinsic, more generalizable subgoals from navigation tasks. Furthermore, we design a novel history-aware discriminator (HAD) for the worker. The discriminator incorporates historical information into subgoal discrimination and provides the worker with additional intrinsic rewards to alleviate the reward sparsity. Without labeled actions, our method provides supervision for the worker in the form of self-supervision by generating subgoals from the manager. The final results of multiple comparison experiments on the Room-to-Room (R2R) dataset show that our DISH can significantly outperform the baseline in accuracy and efficiency.

RESUMO

In this article, a novel hybrid multirobot motion planner that can be applied under no explicit communication and local observable conditions is presented. The planner is model-free and can realize the end-to-end mapping of multirobot state and observation information to final smooth and continuous trajectories. The planner is a front-end and back-end separated architecture. The design of the front-end collaborative waypoints searching module is based on the multiagent soft actor-critic (MASAC) algorithm under the centralized training with decentralized execution (CTDE) diagram. The design of the back-end trajectory optimization module is based on the minimal snap method with safety zone constraints. This module can output the final dynamic-feasible and executable trajectories. Finally, multigroup experimental results verify the effectiveness of the proposed motion planner.

BACKGROUND: Cancer cell lines are invaluable model systems for biomedical research because they provide an almost unlimited supply of biological materials. However, there is considerable skepticism regarding the reproducibility of data derived from these in vitro models. RECENT FINDINGS: Chromosomal instability (CIN) is one of the primary issues associated with cell lines, which can cause genetic heterogeneity and unstable cell properties within a cell population. Many of these problems can be avoided with some precautions. Here we review the underlying causes of CIN, including merotelic attachment, telomere dysfunction, DNA damage response defects, mitotic checkpoint defects and cell cycle disturbances. CONCLUSION: In this review we summarize studies highlighting the consequences of CIN in various cell lines and provide suggestions on monitoring and controlling CIN during cell culture.

RESUMO

Lichens are mutually symbiotic systems consisting of fungal and algal symbionts. While diverse lichen-forming fungal species are known, limited species of algae form lichens. Plasticity in the combination of fungal and algal species with different eco-physiological properties may contribute to the worldwide distribution of lichens, even in extreme habitats. Lichens have been studied systematically for more than 200 years; however, plasticity in fungal-algal/cyanobacterial symbiotic combinations is still unclear. In addition, the association between non-cyanobacterial bacteria and lichens has attracted attention in recent years. The types, diversity, and functions of lichen-associated bacteria have been studied using both culture-based and culture-independent methods. This review summarizes the history of systematic research on lichens and lichen-associated bacteria and provides insights into the current status of research in this field.

The cooperative navigation algorithm is the crucial technology for multirobot systems to accomplish autonomous collaborative operations, and it is still a challenge for researchers. In this work, we propose a new multiagent reinforcement learning algorithm called multiagent local-and-global attention actor-critic (MLGA2C) for multiagent cooperative navigation. Inspired by the attention mechanism, we design the local-and-global attention module to dynamically extract and encode critical environmental features. Meanwhile, based on the centralized training and decentralized execution (CTDE) paradigm, we extend a new actor-critic method to handle feature encoding and make navigation decisions. We also evaluate the proposed algorithm in two cooperative navigation scenarios: static target navigation and dynamic pedestrian target tracking. The multiple experimental results show that our algorithm performs well in cooperative navigation tasks with increasing agents.

Increased research attention is being given to bacterial diversity associated with lichens. Rock tripe lichens (Umbilicariaceae) were collected from two distinct Antarctic biological regions, the continental region near the Japanese Antarctic station (Syowa Station) and the maritime Antarctic South Orkney Islands (Signy Island), in order to compare their bacterial floras and potential metabolism. Bulk DNA extracted from the lichen samples was used to amplify the 18S rRNA gene and the V3-V4 region of the 16S rRNA gene, whose amplicons were Sanger- and MiSeq-sequenced, respectively. The fungal and algal partners represented members of the ascomycete genus Umbilicaria and the green algal genus Trebouxia, based on 18S rRNA gene sequences. The V3-V4 sequences were grouped into operational taxonomic units (OTUs), which were assigned to eight bacterial phyla, Acidobacteriota, Actinomyceota, Armatimonadota, Bacteroidota, Cyanobacteria, Deinococcota, Pseudomonadota and the candidate phylum Saccharibacteria (also known as TM7), commonly present in all samples. The OTU floras of the two biological regions were clearly distinct, with regional biomarker genera, such as Mucilaginibacter and Gluconacetobacter, respectively. The OTU-based metabolism analysis predicted higher membrane transport activities in the maritime Antarctic OTUs, probably influenced by the sampling area's warmer maritime climatic setting.

The diversity of microorganisms associated with speleological sources has mainly been studied in limestone caves, while studies in silicate caves are still under development. Here, we profiled the microbial diversity of opal speleothems from a silicate cave in Guiana Highlands. Bulk DNAs were extracted from three speleothems of two types, i.e., one soft whitish mushroom-like speleothem and two hard blackish coral-like speleothems. The extracted DNAs were amplified for sequencing the V3-V4 region of the bacterial 16S rRNA gene by MiSeq. A total of 210,309 valid reads were obtained and clustered into 3184 phylotypes or operational taxonomic units (OTUs). The OTUs from the soft whitish speleothem were mostly affiliated with Acidobacteriota, Pseudomonadota (formerly, Proteobacteria), and Chloroflexota, with the OTUs ascribed to Nitrospirota being found specifically in this speleothem. The OTUs from the hard blackish speleothems were similar to each other and were mostly affiliated with Pseudomonadota, Acidobacteriota, and Actinomycetota (formerly, Actinobacteria). These OTU compositions were generally consistent with those reported for limestone and silicate caves. The OTUs were further used to infer metabolic features by using the PICRUSt bioinformatic tool, and membrane transport and amino acid metabolism were noticeably featured. These and other featured metabolisms may influence the pH microenvironment and, consequently, the formation, weathering, and re-deposition of silicate speleothems.

Humans are extraordinarily social, and social isolation has profound effects on our behavior, ranging from increased social motivation following short periods of social isolation to increased anti-social behaviors following long-term social isolation. Mice are frequently used as a model to understand how social isolation impacts the brain and behavior. While the effects of chronic social isolation on mouse social behavior have been well studied, much less is known about how acute isolation impacts mouse social behavior and whether these effects vary according to the sex of the mouse and the behavioral context of the social encounter. To address these questions, we characterized the effects of acute (3-day) social isolation on the vocal and non-vocal social behaviors of male and female mice during same-sex and opposite-sex social interactions. Our experiments uncovered pronounced effects of acute isolation on social interactions between female mice, while revealing more subtle effects on the social behaviors of male mice during same-sex and opposite-sex interactions. Our findings advance the study of same-sex interactions between female mice as an attractive paradigm to investigate neural mechanisms through which acute isolation enhances social motivation and promotes social behavior.

In this paper, we study the quantum self-contained refrigerator [Linden et al., Phys. Rev. Lett. 105, 130401 (2010)PRLTAO0031-900710.1103/PhysRevLett.105.130401] in the strong internal coupling regime with engineered reservoirs. We find that if some modes of the three thermal reservoirs can be properly filtered out, the efficiency and the working domain of the refrigerator can be improved in contrast to the those in the weak internal coupling regime, which indicates one advantage of the strong internal coupling. In addition, we find that the background natural vacuum reservoir could cause the filtered refrigerator to stop working and the background natural thermal reservoir could greatly reduce the cooling efficiency.
