RESUMO

Overexpression of sirtuins (NAD(+)-dependent protein deacetylases) has been reported to increase lifespan in budding yeast (Saccharomyces cerevisiae), Caenorhabditis elegans and Drosophila melanogaster. Studies of the effects of genes on ageing are vulnerable to confounding effects of genetic background. Here we re-examined the reported effects of sirtuin overexpression on ageing and found that standardization of genetic background and the use of appropriate controls abolished the apparent effects in both C. elegans and Drosophila. In C. elegans, outcrossing of a line with high-level sir-2.1 overexpression abrogated the longevity increase, but did not abrogate sir-2.1 overexpression. Instead, longevity co-segregated with a second-site mutation affecting sensory neurons. Outcrossing of a line with low-copy-number sir-2.1 overexpression also abrogated longevity. A Drosophila strain with ubiquitous overexpression of dSir2 using the UAS-GAL4 system was long-lived relative to wild-type controls, as previously reported, but was not long-lived relative to the appropriate transgenic controls, and nor was a new line with stronger overexpression of dSir2. These findings underscore the importance of controlling for genetic background and for the mutagenic effects of transgene insertions in studies of genetic effects on lifespan. The life-extending effect of dietary restriction on ageing in Drosophila has also been reported to be dSir2 dependent. We found that dietary restriction increased fly lifespan independently of dSir2. Our findings do not rule out a role for sirtuins in determination of metazoan lifespan, but they do cast doubt on the robustness of the previously reported effects of sirtuins on lifespan in C. elegans and Drosophila.

Heavy metal pollution of sediments is a global concern and can be a serious problem in heavily industrialized parts of the world. Pollution by manganese is particularly common due to its ubiquitous natural occurrence, ease of mobilization, and extensive association with industry. In Ningxia, China, manganese pollution of Yellow River alluvial sediments was assessed by comparing manganese concentrations in 35 sediment samples with background values derived from similar sediments obtained at sites considered remote from potential sources of contamination. Natural background values of manganese were found to range from 192 to 323 mg/kg for surface sediments, and from 220 to 325 and 283 to 394 mg/kg for subsurface sediments at depths of 45­50 and 95­100 cm, respectively. In the study area, manganese content ranged from 565 to 1,363 mg/kg, indicating anthropogenic pollution extending to a depth of at least 1 m in the study area. All 35 samples were found to exceed the threshold effect concentration (TEC) of 460 mg/kg, below which adverse effects on sediment-dwelling organisms are not expected to occur, and one sample (T12) was found to exceed the probable effect concentration (PEC) of 1,100 mg/kg. PEC defines the threshold above which adverse effects are likely to be observed. Variogram analysis of the surface sediment manganese data revealed adherence to a Gaussian model, and ordinary kriging was used to generate a manganese distribution map. Analysis of the high nugget effect ratio indicates high, small-scale variations that are consistent with potential emissions from an adjacent electrolytic manganese plant.

The uniformity of remolded loess is crucial for engineering stability and in laboratory testing, as it affects physical and mechanical properties. It is important to have an index which can accurately and conveniently evaluate the uniformity of remolded loess. This study demonstrated and verified the feasibility of using hydraulic conductivity (K) as an indicator for evaluating the uniformity of remolded loess through laboratory experiments and theoretical analysis. In laboratory research, nine loess samples under different preparation conditions were meticulously prepared in duplicate, which were divided into three sets according to the whole dry density (WDD) of approximately 1.3 g/cm3, 1.4 g/cm3, and 1.5 g/cm3 respectively. For the nine duplicate samples, two procedures were performed for each of the sample. One is the uniformity analysis by cutting the soil column and weighing. The other is the hydraulic conductivity experiment. Results showed that sample uniformity is affected by sample preparation conditions, and there are differences in the uniformity of the same WDD samples. The values of K positively correlate with the degree of sample uniformity. In theoretical analysis, based on Darcy's Law and Kozeny-Carman equation, it is found K is inversely proportional to the variance ( σ 2 ) of the sample dry density. That is, K is positively proportional to the sample uniformity. Since K can be easily determined in the laboratory, the application of this new index in the field of geotechnical engineering makes it very convenient and simple to evaluate the uniformity of remolded loess.

Groundwater vulnerability assessments provide a measure of the sensitivity of groundwater quality to an imposed contaminant load and are globally recognized as an essential element of all aquifer management and protection plans. In this paper, the vulnerability of groundwaters underlying the Yinchuan Plain of Northwest China is determined using OREADIC, a GIS-based assessment tool that incorporates the key characteristics of the universally popular DRASTIC approach to vulnerability assessment but has been modified to consider important additional hydrogeological factors that are specific to the region. The results show that areas of high vulnerability are distributed mainly around Qingtongxia City, Wuzhong City, Lingwu City, and Yongning County and are associated with high rates of aquifer recharge, shallow depths to the water table, and highly permeable aquifer materials. The presence of elevated NO (3) (-) in the high vulnerability areas endorses the OREADIC approach. The vulnerability maps developed in this study have become valuable tools for environmental planning in the region and will be used for predictive management of the groundwater resource.

Pumping-induced leakage across aquitards may induce a deterioration of water quality in multi-layer aquifer systems. It is critical to understand long-term trends of water quality parameters when assessing the sustainability of groundwater abstraction. Daily drinking water needs of 2.2 million people in Yinchuan region of northwest China are solely met by groundwater resources, but long-term groundwater withdrawal has created an extensive cone of depression (294 km2 in area) in confined aquifer causing increased vertical recharge. In this study, a model was established and calibrated with head data, then was incorporated with field tracer tests to provide key information on the hydro-dispersive characteristics of the contaminant for assessing both the current and future state of the aquifer system. The results confirmed a close association between water quality deterioration and high downward fluxes of high chloride groundwater, most notably near the center of the cone of depression. On a temporal scale, water quality degradation remains slow, largely due to the high, pre-existing storage of good quality water. Modeling suggests that the water quality in the upper confined aquifer will lose its potability over a 25 km2 and 50 km2 area within 200 years under the current and intensified pumping conditions, respectively. Elevated chloride values were also detected toward the east of the cone, highlighting the impact of hydrological settings on the vertical groundwater flow. Modeling of potential aquifer remediation shows an even slower response with a further 250 years or more required for potability to be restored in affected areas. The findings can provide valuable guidance to for decision makers and support the sustainable management of aquifer exploitation.

In the arid to semi-arid district of Chengcheng, Weinan City, in central Shaanxi Province, diminishing groundwater reserves in the shallow Quaternary (QLB) aquifer and elevated fluoride in the similarly shallow Permo-Triassic (PTF) aquifer, have promoted interest in the development of groundwater resources in the deep but poorly understood Cambrian-Ordovician carbonate aquifer system (COC). To investigate the origin of the COC groundwaters and the relationship between the deep and shallower systems, a hydrochemical study was undertaken involving 179 major and minor ion analyses, 39 stable isotope analyses (δD and δ18 O), and 14 carbon isotope analyses (14 C and δ13 C). PHREEQC 3.0 was used to investigate mixing. Hydrochemical data support the presence of a well-connected regional flow system extending southwards from the more mountainous north. Stable isotope data indicate that the COC groundwaters originate as soil zone infiltration, under a much cooler regime than is found locally today. This is confirmed by 14 C, which indicates the groundwater to be palaeowater recharged during the late Pleistocene (∼10-12 ka B.P.). The presence of nitrate in the COC groundwaters suggests leakage from overlying shallow aquifers currently provides an additional source of COC recharge, with major faults possibly providing the primary pathways for downward vertical flow.

North America's fifth most populated municipality--the Greater Toronto Area (GTA)--is undergoing rapid urban development with serious questions being raised regarding the long-term impacts of urban growth on the quality and quantity of ground and surface water. Degradation of groundwater quality by NaCl de-icing salt is the primary concern since there are no cost effective alternatives to NaCl de-icing salt and there is little evidence that salt loadings to the subsurface can be significantly reduced. In 2001, the issue acquired a new sense of urgency when de-icing chemicals containing inorganic chloride salts (with or without ferrocyanide de-caking agents) were designated as toxic under the Canadian Environmental Protection Act. To heighten concerns, future growth in the GTA will inevitably take place in areas where groundwater is regularly used for potable supply. Studies using groundwater flow and transport models show that significant deterioration of groundwater quality can be expected in shallow aquifers as a result of urban development with chloride concentrations approaching the drinking water quality standard of 250 mg/l. Results demonstrate that urban planning needs a fresh approach that explicitly includes groundwater protection and aquifer management in the decision-making process, clearly defines acceptable environmental performance standards and makes greater use of groundwater models to evaluate alternative urban designs.

BACKGROUND: Lipid phosphate phosphatases (LPPs) are integral membrane proteins believed to dephosphorylate bioactive lipid messengers, so modifying or attenuating their activities. Wunen, a Drosophila LPP homologue, has been shown to play a pivotal role in primordial germ cell (PGC) migration and survival during embryogenesis. It has been hypothesised that LPPs may form oligomeric complexes, and may even function as hexamers. We were interested in exploring this possibility, to confirm whether LPPs can oligomerise, and if they do, whether oligomerisation is required for either in vitro or in vivo activity. RESULTS: We present evidence that Wunen dimerises, that these associations require the last thirty-five C-terminal amino-acids and depend upon the presence of an intact catalytic site. Expression of a truncated, monomeric form of Wunen in Drosophila embryos results in perturbation of germ cell migration and germ cell loss, as observed for full-length Wunen. We also observed that murine LPP-1 and human LPP-3 can also form associations, but do not form interactions with Wunen or each other. Furthermore, Wunen does not form dimers with its closely related counterpart Wunen-2. Finally we discovered that addition of a trimeric myc tag to the C-terminus of Wunen does not prevent dimerisation or in vitro activity, but does prevent activity in vivo. CONCLUSION: LPPs do form complexes, but these do not seem to be specifically required for activity either in vitro or in vivo. Since neither dimerisation nor the C-terminus seem to be involved in substrate recognition, they may instead confer structural or functional stability through dimerisation. The results indicate that the associations we see are highly specific and occur only between monomers of the same protein.
