RESUMO

Microorganisms inhabiting freshwater environments are an integral part of the aquatic ecosystems. Very few data are available regarding the profiles of the microbial communities in the reservoirs in Bulgaria, despite their key role in the biogeochemical processes. In the present study, we provide the first comprehensive metagenomic analysis on the planktonic bacterial diversity of two large and economically important Bulgarian reservoirs (Batak and Tsankov Kamak) using next-generation sequencing of 16S ribosomal RNA gene (16S rRNA). Analysis of the metagenomic amplicon datasets, including quality filtering, clustering of Operational Taxonomic Units and taxonomy assignment revealed that 78.45% of the microbial communities between the two reservoirs were overlapping. The diversity (H) and Pielou's evenness (J) indices declined along the longitudinal axis of both reservoirs. The estimated values for the Shannon diversity index are typically observed in oligotrophic lakes. The microbial communities of both reservoirs were dominated by Proteobacteria, followed by Actinobacteria and Bacteroidetes all comprised over 95% of the relative abundance, regardless of the reservoir's large hydrogeological differences. The bacterioplankton was characterized by high phylogenetic heterogeneity in the taxonomic structure, being distributed among 211 genera. The genera Limnohabitans and Rhodoferax held the absolute predominance, implying their significance in the aquatic food webs. The obtained data can contribute to the better systematic understanding of the microbial diversity of freshwater environments.

Alternating magnetic fields (MF) with Schumann resonance frequencies accompanied the development of living organisms throughout evolution, but today it remains unclear whether they can have a special biological effect in comparison with surrounding non-resonant frequencies. This work shows some stimulating effect of extremely low-frequency MFs on morphometric parameters and the activity of physiological processes in wheat (Triticum aestivum L.). It is shown that the MF effect is more pronounced for transient processes - photosynthesis reactions and changes in electrical potential caused by turning on light. For light-induced electrical reactions, the dependence of the severity of the effect on the frequency of the applied MF was demonstrated. It is shown that the most pronounced effect occurs in the 14.3 Hz field, which corresponds to the second harmonic of the Schumann resonance. The predominant sensitivity of signal-regulatory systems gives reason to assume the influence of MFs with Schumann resonance frequencies on the interaction of plants with environmental factors under conditions of a changed electromagnetic environment. Such conditions can occur, for example, with an increase in lightning activity caused by climate change, which serves as the basis for the generation of Schumann resonances, and with the development of artificial ecosystems outside the Earth's atmosphere.

RESUMO

The manufacturing of orthodontic archwires made from NiTi alloy has undergone numerous changes from the second half of the last century to modern times. Initially, superelastic-active austenitic NiTi alloys were predominant, followed by thermodynamic-active martensitic NiTi alloys, and, finally, the most recent development was graded thermodynamic alloys. These advancements have been the subject of extensive investigation in numerous studies, as they necessitated a deeper understanding of their properties. Furthermore, it is imperative that we validate the information provided by manufacturers regarding these archwires through independent studies. This review evaluates existing studies on the subject with a specific focus on the Bio-active multi-force NiTi archwire, by examining its mechanical, thermal, and physicochemical properties before and after clinical use. This archwire consists primarily of Ni and Ti, with traces of Fe and Cr, which release graduated, biologically tolerable forces which increase in a front-to-back direction and are affected by the temperature of the environment they are in. The review provides information to practicing orthodontists, facilitating informed decisions regarding the selection and use of Bio-active™ archwires for individual patient treatments.

BACKGROUND: Specifically designed peptide mimetics offer higher selectivity regarding their toxicity to mammalian cells. In addition to the α-helix conformation, the specific activity is related to the peptide's ability to penetrate the cell membrane. The alterations in lipid membrane properties were addressed in the presence of the peptide KLAKLAK-NH2 and analogs containing ß-alanine, strengthening the antibacterial activity and/or naphtalimide with proven anticancer properties. METHODS: The molecular interactions of the peptide mimetics with POPC bilayers were studied using FTIR-ATR spectroscopy. The thermal shape fluctuation analysis of quasispherical unilamellar vesicles was applied to probe the membrane bending elasticity. The impedance characteristics of bilayer lipid membranes were measured using fast Fourier-transform electrochemical impedance spectroscopy. RESULTS: A lateral peptide association with the membrane is reported for ß-alanine-containing peptides. The most pronounced membrane softening is found for the NphtG-KLßAKLßAK-NH2 analog containing both active groups that corroborate with the indications for 1,8-naphthalimide penetration in the lipid hydrophobic area obtained from the FTIR-ATR spectra analysis. The ß-alanine substitution induces strong membrane-rigidifying properties even at very low concentrations of both ß-alanine-containing peptides. CONCLUSIONS: The reported results are expected to advance the progress in tailoring the pharmacokinetic properties of antimicrobial peptides with strengthened stability towards enzymatic degradation. The investigation of the nonspecific interactions of peptides with model lipid membranes is featured as a useful tool to assess the antitumor and antimicrobial potential of new peptide mimetics.

Multiforce orthodontic archwires are thermodynamic wires made of nickel-titanium alloy (Ni-Ti). They release biologically tolerable forces along their length, progressively increasing from front to back. The frontal archwires' segments distribute the weakest force: the premolar, the greater, and the molar, the greatest. The aim of the present study was to determine the influence of clinical use on the mechanical properties of two types of multi-force orthodontic archwires (TriTanium®, American orthodontics; Bio-Active®, GC) with dimensions of 0.016 × 0.022 inches for periods of up to 8 weeks and over 8 weeks of in-vivo use. A three-point bending test was used, and the data gained is statistically analyzed through a multi-variance comparison Mann-Whitney test. We found that after uses of up to 8 weeks and over 8 weeks, the shape memory effect and superelasticity are preserved, as well as the tendency for differential force release along the length of the archwires is kept.

Pooled Immunoglobulin G (IgG), hematin and the membrane-disruptive amphipathic peptide melittin have received attention as powerful biomacromolecules for biomedical and pharmacology applications. Their action on surface properties, oxidation status and epifluorescence properties measured in vitro provide useful information about the functional activity of upper biomacromolecules in erythrocytes in vivo. The hemolysis of erythrocyte membranes, as well as changes in hematocrit and the morphology of erythrocytes, was investigated here via fluorescence microscopy using FITC-concanavalin A binding to cells. The effect of melittin on the membrane capacitance and resistance of model lipid bilayers was probed via electrochemical impedance spectroscopy. Lipid bilayer capacitance was higher in the presence of 0.10 g/L melittin compared to that in the control, which is likely related to bilayer thinning and alterations of the dielectric permittivity of melittin-treated membranes. The biomolecule interactions with red blood cells were probed in physiological media in which the surface of erythrocyte membranes was negatively charged. Surface parameters of erythrocytes are reported upon IgG/hematin and IgG/melittin treatment. Pooled IgG in the presence of melittin, preincubated IgG/hematin preparations promoted a significant decrease in the electrokinetic potential of erythrocytes (Rh-positive). A malondialdehyde (MDA) assay revealed a high rate of lipid peroxidation in erythrocytes treated with IgG/hematin or IgG/melittin preparations. This finding might be a result of pooled IgG interactions with the hematin molecule and the subsequent conformational changes in the protein molecule altering the electrokinetic properties of the erythrocyte membrane related to the Rh group type of erythrocytes. The pooled IgG and hematin are reported to have important consequences for the biophysical understanding of the immunopathological mechanisms of inflammatory, autoimmune and antibody-mediated pathological processes.

Multiforce nickel-titanium (NiTi) orthodontic archwires release progressively increasing forces in a front-to-back direction along their length. The properties of NiTi orthodontic archwires depend on the correlation and characteristics of their microstructural phases (austenite, martensite and the intermediate R-phase). From a clinical and manufacturing point of view, the determination of the austenite finish (Af) temperature is of the greatest importance, as in the austenitic phase, the alloy is most stable and exhibits the final workable form. The main purpose of using multiforce orthodontic archwires is to decrease the intensity of the applied forces to the teeth with a small root surface area, such as the lower central incisors, and also provide forces high enough to move the molars. With the optimally dosed forces of multiforce orthodontic archwires in the frontal, premolar and molar segments, the feeling of pain can be reduced. This will contribute to the greater cooperation of the patient, which is of utmost importance to achieve optimal results. The aim of this research was to determine the Af temperature at each segment of as-received and retrieved Bio-Active® and TriTanium® archwires with dimensions of 0.016 × 0.022 inches, investigated by the differential scanning calorimetry (DSC) method. A classical Kruskal-Wallis one-way ANOVA test and multi-variance comparison based on the ANOVA test statistic using the Bonferroni corrected Mann-Whitney test for multiple comparisons were used. The incisor, premolar and molar segments have different Af temperatures, and they decrease from the anterior to posterior so that the posterior segment has the lowest Af. Bio-Active® and TriTanium® with dimensions of 0.016 × 0.022 inches can be used as first leveling archwires by additional cooling and are not recommended for use on patients with mouth breathing.

Two lineages of bivalve transmissible neoplasia (BTN), BTN1 and BTN2, are known in blue mussels Mytilus. Both lineages derive from the Pacific mussel M. trossulus and are identified primarily by their unique genotypes of the nuclear gene EF1α. BTN1 is found in populations of M. trossulus from the Northeast Pacific, while BTN2 has been detected in populations of other Mytilus species worldwide but not in M. trossulus itself. Here we examined M. trossulus from the Sea of Japan (Northwest Pacific) for the presence of BTN. Using hemocytology and flow cytometry of the hemolymph, we confirmed the presence of disseminated neoplasia in our specimens. Cancerous mussels possessed the BTN2 EF1α genotype and two mitochondrial haplotypes with different recombinant control regions, similar to that of common BTN2 lineages. This is the first report of BTN2 in its original host species M. trossulus. A comparison of all available BTN and M. trossulus COI sequences suggests a common and recent origin of BTN2 diversity in populations of M. trossulus outside the Northeast Pacific, possibly in the Northwest Pacific.

Simple carbohydrates are associated with the enhanced risk of cardiovascular disease and adverse changes in lipoproteins in the organism. Conversely, sugars are known to exert a stabilizing effect on biological membranes, and this effect is widely exploited in medicine and industry for cryopreservation of tissues and materials. In view of elucidating molecular mechanisms involved in the interaction of mono- and disaccharides with biomimetic lipid systems, we study the alteration of dielectric properties, the degree of hydration, and the rotational order parameter and dipole potential of lipid bilayers in the presence of sugars. Frequency-dependent deformation of cell-size unilamellar lipid vesicles in alternating electric fields and fast Fourier transform electrochemical impedance spectroscopy are applied to measure the specific capacitance of phosphatidylcholine lipid bilayers in sucrose, glucose and fructose aqueous solutions. Alteration of membrane specific capacitance is reported in sucrose solutions, while preservation of membrane dielectric properties is established in the presence of glucose and fructose. We address the effect of sugars on the hydration and the rotational order parameter for 1-palmitoyl-2-oleoyl-sn-glycero-3- phosphocholine (POPC) and 1-stearoyl-2-oleoyl-sn-glycero-3- phosphocholine (SOPC). An increased degree of lipid packing is reported in sucrose solutions. The obtained results provide evidence that some small carbohydrates are able to change membrane dielectric properties, structure, and order related to membrane homeostasis. The reported data are also relevant to future developments based on the response of lipid bilayers to external physical stimuli such as electric fields and temperature changes.

PURPOSE: Most of the orthodontic archwires used in the clinical practice nowadays contain nickel (Ni), however, many patients, especially kids, are allergic to Ni. One possible Ni-free alternative is the Titanium-Niobium (Ti-Nb) archwire. Unfortunately, there is not enough information about its mechanical properties in the literature, especially after clinical usage. Therefore, the aim of this work was to investigate and compare the mechanical properties, chemical composition, structure and morphology of as received and used in clinical practice Ti-Nb orthodontic archwires. MATERIALS AND METHODS: We investigated and compared as received and clinically retrieved after 4 and 6 weeks respectively Ti-Nb archwires with dimensions 0.43 × 0.64 mm (0.017 in. × 0.025 in.). The following methods were used: instrumented indentation testing (nanoindentation), X-ray diffraction, scanning electron microscopy, energy-dispersive X-ray spectroscopy and X-ray photoelectron spectroscopy. RESULTS: The nanoindentation investigations of as received and used Ti-Nb archwires revealed a decreasing in their indentation hardness with increased periods of use in the patient's mouth. Moreover, an increasing of the concentration of Ti in the content of the TiNb alloy was associated with an increased period of use in the oral cavity. The SEM analysis showed changes in surface morphology with increasing the period of use of the archwires. CONCLUSIONS: The results showed that there are slight changes in the mechanical and physicochemical properties of the investigated wires after their use in the patient's mouth. That is why we do not recommend them for recycling.

The conditions for formation of the I(3)(-)-starch compound and measuring its absorbance have been found, and a spectrophotometric method has been developed for the determination of the oxygen content in YBa(2)Cu(3)O(y) superconducting bulk samples. The method involves the following stages: a decomposition of the sample in an acid medium in the presence of iodide ions under inert atmosphere; formation of a complex between Cu(II) and glycine; binding the I(3)(-)-complex with a starch and the absorbance measurement of the colored I(3)(-)-starch compound. The coefficient of the active oxygen is calculated by the ratio of the absorbances of two solutions and the method does not require both calibration and precise measuring sample mass. The accuracy of the results is confirmed applying the comparative spectrophotometric method that uses the yellow I(3)(-)-complex. The precision of the results evaluated by the relative standard deviation is 2%. The developed method is sensitive and allows a sample mass about 2mg to be used. The analysis is rapid and requires a simple and inexpensive apparatus. Thus the new method would be useful for an express analytical control of the oxygen content of YBCO-superconducting materials produced for the electronics.
