RESUMO

Myocardial fibrosis (MF) is the manifestation of a variety of cardiovascular diseases. Salidroside (SAL) has been proved to have a certain effect on anti-fibrosis in various organs. However, the mechanism of SAL in the treatment of MF remains unclear. Network pharmacology showed that there were 1228 SAL-related target genes and 2793 MF-related target genes. The intersection of these genes resulted in 271 drug-disease interactions, and 15 core active targets were filtered from protein-protein interaction mapping. The top 20 Gene ontology biological processes analysis showed that the involved processes were close to the pathogenesis of MF. Among the top 20 enriched KEGG pathways, Wnt/ß-catenin and TGF-ß1/Smad3 signaling pathways were identified. In vivo, MI rats exhibited thinning of the myocardial region and the formation of fibrous scars, the expression of smad3 and ß-catenin were increased. After SAL treatment, there was a significant reduction in collagen area and a decrease in the ratio of collagen type I to type III. The expression of smad3 and ß-catenin was suppressed and positively correlated with the dosage of SAL. SAL may contribute to the progression of MF through the TGF-ß1/Smad3 and Wnt/ß-catenin signaling pathways.

RESUMO

Metallic chromium pollution in soil is widespread, which aroused intensive research in recent decades. In mainstream research, most studies use materials with a reducing ability to adsorb and reduce hexavalent chromium. However, comprehensive analyses and systematic verifications of these different materials are scarce. Therefore, this study conducted a meta-analysis of relevant papers published from 2013 to October 2024 to compare and analyze the performance and usage conditions of some common materials, such as iron-based materials, mineral inorganic materials, organic materials, and layered double hydroxide materials. We synthesized 31 papers for 186 pairwise comparisons and selected the Standardized Mean Difference (SMD) as the appropriate effect size for mean-to-mean comparisons. Fe-based materials had the most stable performance based on its numerous data support, while organic materials had the worst performance. The difference in performance between inorganic mineral materials was the greatest, which was closely related to the selection of components. The difference in the effectiveness of inorganic materials was the greatest, which was closely related to the selection of components and there was room for further improvement. Through further analysis of the impact of environmental factors on material performance, it can be concluded that the effect of the material was better under alkaline, non-sandy, low organic matter, and high CEC soil conditions.

RESUMO

A terahertz metamaterial microfluidic sensing chip for ultrasensitive detection is proposed to investigate the response of substances to terahertz radiation in liquid environments and enhance the molecular fingerprinting of trace substances. The structure consists of a cover layer, a metal microstructure, a microfluidic channel, a metal reflective layer, and a buffer layer from top to bottom, respectively. The simulation results show that there are three obvious resonance absorption peaks in the range of 1.5-3.0 THz and the absorption intensities are all above 90%. Among them, the absorption intensity at M1 = 1.971 THz is 99.99%, which is close to the perfect absorption, and its refractive index sensitivity and Q-factor are 859 GHz/RIU and 23, respectively, showing excellent sensing characteristics. In addition, impedance matching and equivalent circuit theory are introduced in this paper to further analyze the physical mechanism of the sensor. Finally, we perform numerical simulations using refractive index data of normal and cancer cells, and the results show that the sensor can distinguish different types of cells well. The chip can reduce the sample pretreatment time as well as enhance the interaction between terahertz waves and matter, which can be used for early disease screening and food quality and safety detection in the future.

RESUMO

OBJECTIVE: To investigate the pharmacological mechanism of Qili Qiangxin Capsule (QLQX) improvement of heart failure (HF) based on miR133a-endoplasmic reticulum stress (ERS) pathway. METHODS: A left coronary artery ligation-induced HF after myocardial infarction model was used in this study. Rats were randomly assigned to the sham group, the model group, the QLQX group [0.32 g/(kg·d)], and the captopril group [2.25 mg/(kg·d)], 15 rats per group, followed by 4 weeks of medication. Cardiac function such as left ventricular ejection fraction (EF), fractional shortening (FS), left ventricular systolic pressure (LVSP), left ventricular end diastolic pressure (LVEDP), the maximal rate of increase of left ventricular pressure (+dp/dt max), and the maximal rate of decrease of left ventricular pressure (-dp/dt max) were monitored by echocardiography and hemodynamics. Hematoxylin and eosin (HE) and Masson stainings were used to visualize pathological changes in myocardial tissue. The mRNA expression of miR133a, glucose-regulated protein78 (GRP78), inositol-requiring enzyme 1 (IRE1), activating transcription factor 6 (ATF6), X-box binding protein1 (XBP1), C/EBP homologous protein (CHOP) and Caspase 12 were detected by RT-PCR. The protein expression of GRP78, p-IRE1/IRE1 ratio, cleaved-ATF6, XBP1-s (the spliced form of XBP1), CHOP and Caspase 12 were detected by Western blot. TdT-mediated dUTP nick-end labeling (TUNEL) staining was used to detect the rate of apoptosis. RESULTS: QLQX significantly improved cardiac function as evidenced by increased EF, FS, LVSP, +dp/dt max, -dp/dt max, and decreased LVEDP (P<0.05, P<0.01). HE staining showed that QLQX ameliorated cardiac pathologic damage to some extent. Masson staining indicated that QLQX significantly reduced collagen volume fraction in myocardial tissue (P<0.01). Results from RT-PCR and Western blot showed that QLQX significantly increased the expression of miR133a and inhibited the mRNA expressions of GRP78, IRE1, ATF6 and XBP1, as well as decreased the protein expressions of GRP78, cleaved-ATF6 and XBP1-s and decreased p-IRE1/IRE1 ratio (P<0.05, P<0.01). Further studies showed that QLQX significantly reduced the expression of CHOP and Caspase12, resulting in a significant reduction in apoptosis rate (P<0.05, P<0.01). CONCLUSION: The pharmacological mechanism of QLQX in improving HF is partly attributed to its regulatory effect on the miR133a-IRE1/XBP1 pathway.

RESUMO

This paper proposes to detect heavy metal pollutants in wheat using terahertz spectroscopy and deep support vector machine (DSVM). Five heavy metal pollutants, arsenic, lead, mercury, chromium, and cadmium, were considered for detection in wheat samples. THz spectral data were pre-processed by wavelet denoising. DSVM was introduced to further enhance the accuracy of the SVM classification model. According to the relationship between the accuracy and the training time with the number of hidden layers ranging from 1 to 4, the model performs the best when the hidden layer network has three layers. Besides, using the back-propagation algorithm to optimize the entire DSVM network. Compared with Deep neural network (DNN) and SVM models, the comprehensive evaluation index of the proposed model optimized by DSVM has the highest accuracy of 91.3 %. It realized the exploration enhanced the classification accuracy of the heavy metal pollutants in wheat.

RESUMO

The structure of the grain-and-oil-food-supply chain has the characteristics of complexity, cross-regionality, a long cycle, and numerous participants, making it difficult to maintain the safety of supply. In recent years, some phenomena have emerged in the field of grain procurement and sale, such as topping the new with the old, rotating grains, the pressure of grades and prices, and counterfeit oil food, which have seriously threatened grain-and-oil-food security. Blockchain technology has the advantage of decentralization and non-tampering Therefore, this study analyzes the characteristics of traceability data in the grain-and-oil-food-supply chain, and presents a blockchain-based traceability model for the grain-and-oil-food-supply chain. Firstly, a new method combining blockchain and machine learning is proposed to enhance the authenticity and reliability of blockchain source data by constructing anomalous data-processing models. In addition, a lightweight blockchain-storage method and a data-recovery mechanism are proposed to reduce the pressure on supply-chain-data storage and improve fault tolerance. The results indicate that the average query delay of public data is 0.42 s, the average query delay of private data is 0.88 s, and the average data-recovery delay is 1.2 s. Finally, a blockchain-based grain-and-oil-food-supply-chain traceability system is designed and built using Hyperledger Fabric. Compared with the existing grain-and-oil-food-supply chain, the model constructed achieves multi-source heterogeneous data uploading, lightweight storage, data recovery, and traceability in the supply chain, which are of great significance for ensuring the safety of grain-and-oil food in China.

RESUMO

Background: Myocardial infarction (MI) is an acute and serious cardiovascular disease. Arrhythmia after MI can lead to sudden cardiac death, which seriously affects the survival outcome of patients. WenXin KeLi is a Chinese patent medicine for the treatment of arrhythmia in a clinic, which can significantly improve symptoms of palpitation and play an important role in reducing the risk of arrhythmia after MI. In this study, we aimed to explore the pharmacological mechanism of WenXin KeLi in protecting the heart. Methods: The MI model was established by ligating the left coronary artery and the ventricular fibrillation threshold (VFT) was measured by electrical stimulation. The expression of connexin43 (CX43) and autophagy-related protein were measured by Western Blot, and correlation analysis was conducted to study the relationship between cardiac autophagy, CX43, and arrhythmia in rats after MI. The effects of WenXin KeLi on arrhythmia, cardiac structure, and function in MI rats were respectively observed by electrical stimulation, cardiac gross section, Masson staining, and cardiac ultrasound. The effects of WenXin KeLi on the expression of phosphoinositide 3 kinase-protein kinase B-mammalian targets of rapamycin (PI3K-AKT-mTOR) autophagy pathway and CX43 were observed by Western Blot. Results: After 4 weeks of MI, the VFT in the model group was significantly reduced, the expression levels of yeast ATG6 homolog (Beclin1), microtubule-associated protein 1A/1B-light chain 3 (LC3II/LC3I), and p-CX43 (S368) significantly increased, the expression of sequestosome-1(P62) and CX43 significantly decreased. LC3II/LC3I and Beclin1 expression were significantly negatively correlated with the VFT, and the expression of P62 and CX43 were significantly positively correlated with the VFT. LC3II/LC3I and Beclin1 expression were negatively correlated with CX43 expression, while P62 expression was positively correlated with CX43 expression. WenXin KeLi could significantly increase the VFT, reduce the deposition of collagen fibers, and increase the index levels of the left ventricular end-diastolic anterior wall (LVEDAW), interventricular septum end-diastolic (IVSED), left ventricular end-systolic anterior wall (LVESAW), interventricular septum end-systolic (IVSES), left ventricular end-diastolic posterior wall (LVEDPW), left ventricular end-systolic posterior wall (LVESPW), left ventricular ejection fraction (LVEF) and left ventricular fractional shortening (LVFS), and reduce the index levels of the left ventricular end-diastolic dimension (LVEDD), left ventricular end-systolic dimension (LVESD), left ventricular end-diastolic volume (LVEDV) and left ventricular end-systolic volume (LVESV). WenXin KeLi could increase the expression of CX43, P62, AKT, p-PI3K, p-AKT (308), p-AKT (473), and p-mTOR and decrease the expression of LC3II/LC3I and Beclin1. Conclusion: WenXin KeLi can activate the PI3K-AKT-mTOR signaling pathway, improve cardiac autophagy and Cx43 expression in rats after MI, reduce the risk of arrhythmia after MI, and play a cardioprotective role.

RESUMO

Background. Arrhythmia after myocardial infarction is the leading cause of death in clinical heart disease. Increasing studies have shown that the response to endoplasmic reticulum (ER) stress (ERS) caused by myocardial infarction is related to prognosis and the development of arrhythmias. The unfolded protein response (UPR) could serve as an important regulatory signaling pathway following myocardial infarction. The traditional Chinese medicine Wenxin Granules improve arrhythmias following myocardial infarction, which may be related to ERS intervention and the activation of the UPR and apoptosis. We aimed to investigate the involvement of Wenxin Granules in the activation of the UPR and apoptosis following myocardial infarction. Left coronary artery ligation was established as a rat model of myocardial infarction. The rats were randomly divided into the model group, low-dose Wenxin Granule group, high-dose Wenxin Granule group, and metoprolol group. Rats with only wire insertion and no ligature were used as the sham group. Small animal ultrasound systems were used to detect changes in heart structure and function, and the electrical stimulation threshold for ventricular fibrillation was detected. The expression of glucose-regulated protein (GRP)78, activating transcription factor (ATF)6, X-box binding protein (XBP)1, protein kinase-like ER kinase (PERK), phosphorylated (p)-PERK, Bax, Bcl2, C/EBP homologous protein (CHOP), caspase 12, caspase 8, and caspase 3 were detected by western blot, and terminal deoxynucleotidyl transferase dUTP Nick end labeling (TUNEL) was used to determine the cardiomyocyte apoptosis index. Compared with the sham group, rats in the model group displayed immediate ST-segment elevation and pathological Q waves after 24 hours. After 2 weeks, the left ventricular (LV) anterior wall thickness (LVAW) became thinner, and the inner diameter (LVID) increased. The end-diastolic LVAW (LVAWd), end-systolic LVAW (LVAWs), ejection fraction (EF), and fractional shortening (FS) were significantly reduced (P < 0.01), whereas the LVIDd, LVIDs, diastolic LV volume (LV Vold), and systolic LV volume (LV Vols) significantly increased (P < 0.01). The ventricular fibrillation threshold decreased significantly (P < 0.01). ERS proteins GRP78, p-PERK, PERK, ATF6, and XBP1 and apoptotic proteins CHOP, Bax, caspase 12, caspase 8, and caspase 3 significantly increased (P < 0.01, P < 0.05), whereas Bcl-2 expression and the Bcl-2/Bax ratio decreased (P < 0.01). Compared with the sham group, LVAWd, LVAWs, FS, and Bcl-2 protein expression were significantly increased in the low-dose Wenxin Granule group (P < 0.01, P < 0.05), and p-PERK and ATF6 decreased (P < 0.01, P < 0.05). Compared with the sham group, LVAWd, LVAWs, EF, FS, and the ventricular fibrillation threshold significantly increased in the high-dose Wenxin Granule and metoprolol groups (P < 0.01, P < 0.05), whereas LVIDs, LV Vols, and ERS proteins were significantly decreased (P < 0.01, P < 0.05). CHOP, Bax, caspase 12, caspase 8, and caspase 3 protein expression decreased in the Wenxin Granule group (P < 0.01, P < 0.05), whereas Bcl-2 and the Bcl-2/Bax ratio increased (P < 0.01, P < 0.05). LVIDd and Bax decreased in the metoprolol group (P < 0.01, P < 0.05), and the Bcl-2/Bax ratio increased (P < 0.05). The cardiomyocyte apoptosis index values for the low- and high-dose Wenxin Granule and metoprolol groups were significantly reduced (P < 0.05). This study suggested that the UPR is an essential mechanism underlying pathological injury after myocardial infarction. Wenxin Granule treatment can improve ventricular remodeling and cardiac function and inhibit arrhythmia by preventing excessive ERS from activating the UPR and apoptosis.

RESUMO

Gap junctions are the main form of interaction between cardiomyocytes, through which the electrochemical activities between cardiomyocytes can be synchronized to maintain the normal function of the heart. Connexins are the basis of gap junctions. Changes in the expression, structural changes (e.g., phosphorylation and dephosphorylation), and distribution of connexins can affect the normal electrophysiological activities of the heart. Myocardial infarction (MI) and concurrent arrhythmia, shock, or heart failure can endanger life. The structural and functional damage of connexin (Cx) 43 in cardiomyocytes is a central part of the pathological progression of MI and is one of the main pathological mechanisms of arrhythmia after MI. Therefore, increasing Cx43 expression has become one of the main measures to prevent MI. Also, intervention in Cx43 expression can improve the structural and electrical remodeling of the myocardium to improve MI prognosis. Here, research progress of Cx43 in MI and its prevention and treatment using Traditional Chinese Medicine formulations is reviewed.

RESUMO

Environment-friendly medium-density fiberboards (MDFs) prepared using sodium lignosulfonate/chitosan adhesives (L/C) show potential in environment-friendly wood-based panel application. However, the synthesis mechanism of this adhesive and the relationships between synthesis mechanism and bonding performance were not discussed in depth. Herein, the synthesis mechanism of L/C was explored in detail based on characterizations of L/C with different mass ratios of sodium lignosulfonate to chitosan by Fourier-transform infrared spectroscopy, thermogravimetric analysis, and X-ray diffraction. For L/C with different mass ratios of sodium lignosulfonate to chitosan, the corresponding bonding performance was also determined based on characterizations of mechanical and dimensional performance of MDFs. Results showed a 3D network structure of L/C formed through the hydrogen linkages among hydroxyl groups in sodium lignosulfonate and hydroxyl and amino groups in chitosan, amide linkages resulted from reaction between carbonyl groups in sodium lignosulfonate and amino groups in chitosan, and sulfonamide linkages originated from reaction between sulfonic groups in sodium lignosulfonate and amino groups in chitosan. The mechanical performance of MDF was closely related to the 3D network and amino groups of L/C, while the dimensional performance of MDF was negatively affected by sodium lignosulfonate. The MDFs with 1:3 and 1:2 mass ratios of sodium lignosulfonate to chitosan showed superior mechanical properties and comparable dimensional performance with a commercial panel.

RESUMO

Graphitic carbon nitride (g-C3N4) hybrid aerogels (GHAs) with different proportions of g-C3N4 were prepared using g-C3N4, carboxymethyl cellulose, and ß-cyclodextrin. GHAs have demonstrated high porosity, large specific surface area, rich three-dimensional network structure. GHAs demonstrated an excellent synergistic effect on the photocatalytic and adsorptive properties. Especially, GHA1 (the ratio of g-C3N4 and aerogel component was 1:1) exhibited a good synergy in photodegradation and adsorption, with a Rhodamine B (Rh B) removal up to 97.99% in 90 min. In five cycling experiments, the GHA1 showed a good long-term stability. The aerogels can be easily isolated and maintains its excellent photocatalytic properties. Compared to pure g-C3N4, GHA1 has a stronger photocurrent, which is due to the transmission of light to the surface of the catalyst, promoted photo-generated electron-hole pairs, and inhibited recombination of electros and holes. Finally, mechanistic studies reveal that the superoxide radicals (O2-) are the main active groups in the photocatalytic degradation process. This work provides a useful perspective in design and fabrication of hybrid aerogel composites with an enhanced photocatalytic activity.

RESUMO

The main aim of this work was to systematically investigate the effects of time on hydrothermal carbonization of holocellulose coming from the forestry residues. Based on the results, reaction time has a significant impact on the structural, chemical, fuel characteristics and combustion behavior of hydrochars. With increasing reaction time, holocellulose is gradually converted into the amorphous hydrochars with aromatic structure and abundant oxygen-containing functional groups; the O/C and H/C decrease from 0.85 and 1.60 to 0.23 and 0.71, respectively. The HHVs of some hydrochars have reached the quality grade of medium-high quantity calorific coal. The combustion stage and characteristic parameters of holocellulose are predominantly modified after HTC, and HC-14 could be regarded as a good fuel. Additionally, the structural and chemical formation processes of holocellulose-derived hydrochar are proposed. These results revealed that HTC is an effective and simple strategy for converting holocellulose into functional, value-added, and energy-dense hydrophobic solid fuels.

RESUMO

With the growing depletion of wood-based materials and concerns over emissions of formaldehyde from traditional wood fibre composites, there is a desire for environment-friendly binders. Herein, we report a green wood fibre composite with specific bonding strength and water resistance that is superior to a commercial system by using wood fibres and chitosan-based adhesives. When the mass ratio of solid content in the adhesive and absolute dry wood fibres was 3%, the bonding strength and water resistance of the wood fibre composite reached the optimal level, which was significantly improved over that of wood fibre composites without adhesive and completely met the requirements of the Chinese national standard GB/T 11718-2009. Fourier transform infrared (FTIR), X-ray photoelectron spectroscopy (XPS) and X-ray diffraction (XRD) characterizations revealed that the excellent performance of the binder might partly be due to the amide linkages and hydrogen bonding between wood fibres and the chitosan-based adhesive. We believe that this strategy could open new insights into the design of environment-friendly wood fibre composites with high bonding strength and water resistance for multifunctional applications.

RESUMO

An efficient method was developed for preparing medium-density fiberboard (MDF) adhesives with high performance, using chitosan as the main component and glutaraldehyde as a crosslinking agent. The effects of glutaraldehyde content on the bonding strength and water resistance were investigated. The results indicated that the optimum internal bonding strength (IB, 1.22MPa), modulus of elasticity (MOE, 3162.69MPa), modulus of rupture (MOR, 29.10MPa), water absorption (WA, 22.23%) and thickness swell (TS, 26.17%) of the MDF complied with the requirement of the Chinese national standard for MDF. The chitosan-based adhesive was characterized by Fourier transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA), and X-ray diffractometry (XRD). The thermal stability and water holding capacity of the chitosan-based adhesives were influenced by the extent of crosslinking. The excellent properties were attributed to the network structure of the chitosan-based adhesives, formed by crosslinking of chitosan with self-polymerized glutaraldehyde or glutaraldehyde monomers through CN bonds.
