RESUMO

The structural features and optical properties of supramolecular cyanoiron salts containing bis-pyridinium-4-oxime Toxogonin® (TOXO) as an electron acceptor are presented. The properties of the new TOXO-based cyanoiron materials were probed by employing two cyanoiron platforms: hexacyanoferrate(II), [Fe(CN)6]4- (HCF); and nitroprusside, [Fe(CN)5(NO)]2- (NP). Two water-insoluble inter-ionic donor-acceptor phases were characterized: the as-prepared microcrystalline reddish-brown (TOXO)2[Fe(CN)6]·8H2O (1a) with a medium-responsive, hydrochromic character; and the dark violet crystalline (TOXO)2[Fe(CN)6]·3.5H2O (1cr). Complex 1a, upon external stimulation, transforms to the violet anhydrous phase (TOXO)2[Fe(CN)6] (1b), which upon water uptake transforms back to 1a. Using the NP platform resulted in the water-insoluble crystalline salt TOXO[Fe(CN)5(NO)]·2H2O (2). The structures of 1cr and 2, solved by single-crystal X-ray diffraction, along with a comparative spectroscopic (UV-vis-NIR diffuse reflectance, IR, solid-state MAS-NMR, Mössbauer), thermal, powder X-ray diffraction, and microscopic analysis (SEM, TEM) of the isolated materials, provided insight for the supramolecular binding, electron-accepting, and H-bonding capabilities of TOXO in the self-assembly of these functionalized materials.

RESUMO

The majority of bladder cancers in humans are non-muscle-invasive cancers that recur frequently after standard treatment procedures. Mouse models are widely used to develop anti-tumor treatments. The purpose of our work was to establish an orthotopic mouse bladder tumor model and to explore early stages of implantation of cancerous MB49 cells in vivo using various labeling and microscopic techniques. To distinguish cancer cells from normal urothelial cells in mouse urinary bladders, we performed molecular characterization of MB49 cells before intravesical injection experiments. In this new approach we applied internalized metal nanoparticles to unequivocally discriminate cancer cells from normal cells. This method revealed that cancer cells attached to the urothelium or basal lamina within just 1 hour of intravesical injection, whereas small tumors and localized hyperplastic urothelial regions developed within two days. We found that cancer cells initially adhere to normal urothelial cells through filopodia and by focal contacts with basal lamina. This is the first in vivo characterization of intercellular contacts between cancerous and normal urothelial cells in the bladder. Our study yields new data about poorly known early events of tumorigenesis in vivo, which could be helpful for the translation into clinic.

RESUMO

The limpet radula is a feeding organ, which contains more than 100 rows of teeth. During their growth the teeth mature and advance in position along the radula. The simpler doccoglossan radulae operate by grinding rocky substrates, extracting the algae by rasping and scraping with the teeth functioning as shovels. Less is known about the rhipidoglossan radulae, used as rakes or brooms that brush and collect loose marine debris. This type of radula is found in the giant keyhole limpet (Megathura crenulata). The large size of this organism suggests that the rhipidoglossan radula entails a technological superiority for M. crenulata in its habitat. The structure and function of the radulae teeth have however not been reported in detail. Using a combination of 2D and 3D microscopy techniques coupled with amino acid analysis and X-ray scattering, we reveal the working components of M. crenulata's radula. It is characterized by numerous marginal teeth surrounding a pair of major hook-like lateral teeth, two pairs of minor lateral teeth and a large central tooth. The mature major lateral teeth show pronounced signs of wear, which gradually increase towards the very front end of the radula and are evidence for scraping. An abrupt change in the amino acid composition in the major lateral teeth and the concurrent formation of a chitinous fiber-network mark the onset of tooth maturation. In comparison to the simpler rock-scraping doccoglossate limpets, the radula of M. crenulata forms an elaborate feeding apparatus, which can be seen as a natural harvest machine.

RESUMO

This work focuses in-depth on the quantitative relationships between primary first-order microstructural parameters (i.e., volume fractions of various phases and particle size distribution) with the more complex second-order topological features (i.e., connectivity of phases, three-phase boundary length (TPBL), interfacial areas, or tortuosity). As a suitable model material, a cermet nickel/samaria-doped ceria (Ni-SDC) is used as an anode in a solid oxide fuel cell (SOFC). A microstructure description of nano-sized Ni-SDC cermets, fabricated at various sintering conditions from 1100 °C to 1400 °C, was performed using FIB-SEM nanotomography. The samples were serially sectioned employing a fully automated slicing procedure with active drift correction algorithms and an auto-focusing routine to obtain a series of low-loss BSE images. Advanced image processing algorithms were developed and applied directly to image data volume. The microstructural-topological relationships are crucial for the microstructure optimisation and, thus, the improvement of the corresponding electrode performance. Since all grains of individual phases (Ni, SDC, or pores) did not percolate, special attention was given to the visualisation of the so-called active TPBL. Based on the determined microstructure characteristics of the prepared Ni-SDC cermets, including simulations of gas flow and pressure drop, thermal treatment at 1200 °C was recognised as the most appropriate sintering temperature.

RESUMO

Teeth exemplify architectures comprising an interplay of inorganic and organic constituents, resulting in sophisticated natural composites. Rodents (Rodentia) showcase extraordinary adaptations, with their continuously growing incisors surpassing human teeth in functional and structural optimizations. In this study, employing state-of-the-art direct atomic-scale imaging and nanoscale spectroscopies, we present compelling evidence that the release of material from ameloblasts and the subsequent formation of iron-rich enamel and surface layers in the constantly growing incisors of rodents are complex orchestrated processes, intricately regulated and independent of environmental factors. The synergistic fusion of three-dimensional tomography and imaging techniques of etched rodent́s enamel unveils a direct correlation between the presence of pockets infused with ferrihydrite-like material and the acid resistant properties exhibited by the iron-rich enamel, fortifying it as an efficient protective shield. Moreover, observations using optical microscopy shed light on the role of iron-rich enamel as a microstructural element that acts as a path for color transmission, although the native color remains indistinguishable from that of regular enamel, challenging the prevailing paradigms. The redefinition of "pigmented enamel" to encompass ferrihydrite-like infusion in rodent incisors reshapes our perception of incisor microstructure and color generation. The functional significance of acid-resistant iron-rich enamel and the understanding of the underlying coloration mechanism in rodent incisors have far-reaching implications for human health, development of potentially groundbreaking dental materials, and restorative dentistry. These findings enable the creation of an entirely different class of dental biomaterials with enhanced properties, inspired by the ingenious designs found in nature.

RESUMO

In the past, platinum-copper catalysts have proven to be highly active for the oxygen reduction reaction (ORR), but transferring the high activities measured in thin-film rotating disk electrodes (TF-RDEs) to high-performing membrane electrode assemblies (MEAs) has proven difficult due to stability issues during operation. High initial performance can be achieved. However, fast performance decay on a timescale of 24 h is induced by repeated voltage load steps with H2/air supplied. This performance decay is accelerated if high relative humidity (>60% RH) is set for a prolonged time and low voltages are applied during polarization. The reasons and possible solutions for this issue have been investigated by means of electrochemical impedance spectroscopy and distribution of relaxation time analysis (EIS-DRT). The affected electrochemical sub-processes have been identified by comparing the PtCu electrocatalyst with commercial Pt/C benchmark materials in homemade catalyst-coated membranes (CCMs). The proton transport resistance (Rpt) increased by a factor of ~2 compared to the benchmark materials. These results provide important insight into the challenges encountered with the de-alloyed PtCu/KB electrocatalyst during cell break-in and operation. This provides a basis for improvements in the catalysts' design and break-in procedures for the highly attractive PtCu/KB catalyst system.

RESUMO

We report on a detailed structural versus electrochemical property investigation of the corncob-derived non-graphitizable carbons prepared at different carbonization temperatures using a combination of structural characterization methodology unique to this field. Non-graphitizable carbons are currently the most viable option for the negative electrode in sodium-ion batteries. However, many challenges arise from the strong dependence of the precursor's choice and carbonization parameters on the evolution of the carbon matrix and its resulting electrochemistry. We followed structure development upon the increase in carbonization temperature with thorough structural characterization and electrochemical testing. With the increase of carbonization temperature from 900 to 1600 °C, our prepared materials exhibited a trend toward increasing structural order, an increase in the specific surface area of micropores, the development of ultramicroporosity, and an increase in conductivity. This was clearly demonstrated by a synergy of small- and wide-angle X-ray scattering, scanning transmission electron microscopy, and electron-energy loss spectroscopy techniques. Three-electrode full cell measurements confirmed incomplete desodiation of Na+ ions from the non-graphitizable carbons in the first cycle due to the formation of a solid-electrolyte interface and Na trapping in the pores, followed by a stable second cycle. The study of cycling stability over 100 cycles in a half-cell configuration confirmed the observed high irreversible capacity in the first cycle, which stabilized to a slow decrease afterward, with the Coulombic efficiency reaching 99% after 30 cycles and then stabilizing between 99.3 and 99.5%. Subsequently, a strong correlation between the determined structural properties and the electrochemical behavior was established.

RESUMO

Grapevine leaf mottling and deformation is a novel grapevine disease that has been associated with grapevine Pinot gris virus (GPGV). The virus was observed exclusively inside membrane-bound structures in the bundle sheath cells of the infected grapevines. As reported widely in the literature, many positive-sense single-stranded RNA viruses modify host-cell membranes to form a variety of deformed organelles, which shelter viral genome replication from host antiviral compounds. Morphologically, the GPGV-associated membranous structures resemble the deformed endoplasmic reticulum described in other virus-host interactions. In this study we investigated the GPGV-induced membranous structures observed in the bundle sheath cells of infected plants. The upregulation of different ER stress-related genes was evidenced by RT-qPCR assays, further confirming the involvement of the ER in grapevine/GPGV interaction. Specific labelling of the membranous structures with an antibody against luminal-binding protein identified them as ER. Double-stranded RNA molecules, which are considered intermediates of viral replication, were localised exclusively in the ER-derived structures and indicated that GPGV exploited this organelle to replicate itself in a shelter niche. Novel analyses using focussed ion-beam scanning electron microscopy (FIB-SEM) were performed in grapevine leaf tissues to detail the three-dimensional organisation of the ER-derived structures and their remodelling due to virus replication.

RESUMO

Nanostructured alloy powders of SmCo5 + 10 wt% Fe obtained using recycled material were studied for the first time. The SmCo5 precursor was obtained from commercial magnets recycled by hydrogen decrepitation. The results were compared with identically processed samples obtained using virgin SmCo5 raw material. The samples were synthesized by dry high-energy ball-milling and subsequent heat treatment. Robust soft/hard exchange coupling was observed-with large coercivity, which is essential for commercial permanent magnets. The obtained energy products for the recycled material fall between 80% and 95% of those obtained when using virgin SmCo5, depending on milling and annealing times. These results further offer viability of recycling and sustainability in production. These powders and processes are therefore candidates for the next generation of specialized and nanostructured exchange-coupled bulk industrial magnets.

RESUMO

This research aimed to study the influence of lyocell with incorporated ZnO (CLY) for antibacterial activity and biodegradation of fiber blends composed of viscose (CV), flax (LI), and CLY. Fiber blended samples with an increased weight fraction of CLY fibers were composed, and single CLY, CV and LI fibers were also used for comparison. Antibacterial activity was determined for the Gram-negative Escherichia coli and the Gram-positive Staphylococcus aureus bacteria. The biodegradation of fiber blends was investigated by the soil burial test. The results show that the single CLY fibers exhibited high antimicrobial activity against both E. coli and S. aureus bacteria and that the presence of LI fibers in the blended samples did not significantly affect antibacterial activity against E. coli, but drastically decreased the antibacterial activity against S. aureus. LI fibers strongly promoted the growth of S. aureus and, consequently, impaired the antimicrobial performance of ZnO against this bacterium. The presence of CLY fibers slowed down, but did not prevent, the biodegradation process of the fiber blends, even at the highest ZnO concentration. The soil that was in contact with the fiber blended samples during their burial was not contaminated to such an extent as to affect the growth of sprouts, confirming the sustainability of the fiber blends.

RESUMO

We report on a heterogeneity study, down to the atomic scale, on a representative multiple-element-modified ceramic based on potassium sodium niobate (KNN): 0.95(Na0.49K0.49Li0.02)(Nb0.8Ta0.2)O3-0.05CaZrO3 with 2 wt % MnO2. We show that different routes for incorporating the MnO2 (either before or after the calcination step) affect the phase composition and finally the functionality of the material. According to X-ray diffraction and scanning electron microscopy analyses, the ceramics consist of orthorhombic and tetragonal perovskite phases together with a small amount of Mn-rich secondary phase. The addition of MnO2 after the calcination results in better piezoelectric properties, corresponding to a ratio between the orthorhombic and tetragonal perovskite phases that is closer to unity. We also show, using microscopy techniques combined with analytical tools, that Zr-rich, Ta-rich and Mn-rich segregations are present on the nano and atomic levels. With this multi-scale analysis approach, we demonstrate that the functional properties are sensitive to minor modifications in the synthesis route, and consequently to different material properties on all scales. We believe that detecting and learning how to control these modifications will be a step forward in overcoming the irreproducibility problems with KNN-based materials.

RESUMO

Metallic lithium is considered to be one of the most promising anode materials since it offers high volumetric and gravimetric energy densities when combined with high-voltage or high-capacity cathodes. However, the main impediment to the practical applications of metallic lithium is its unstable solid electrolyte interface (SEI), which results in constant lithium consumption for the formation of fresh SEI, together with lithium dendritic growth during electrochemical cycling. Here we present the electrochemical performance of a fluorinated reduced graphene oxide interlayer (FGI) on the metallic lithium surface, tested in lithium symmetrical cells and in combination with two different cathode materials. The FGI on the metallic lithium exhibit two roles, firstly it acts as a Li-ion conductive layer and electronic insulator and secondly, it effectively suppresses the formation of high surface area lithium (HSAL). An enhanced electrochemical performance of the full cell battery system with two different types of cathodes was shown in the carbonate or in the ether based electrolytes. The presented results indicate a potential application in future secondary Li-metal batteries.

RESUMO

Zinc(II) oxide nanoparticles were used for the stabilization of dicyclopentadiene (DCPD)-water-based high internal phase emulsions (HIPEs), which were subsequently cured using ring-opening metathesis polymerization (ROMP). The morphology of the resulting ZnO-pDCPD nanocomposite foams was investigated in correlation to the nanoparticle loading and nanoparticle surface chemistry. While hydrophilic ZnO nanoparticles were found to be unsuitable for stabilizing the HIPE, oleic acid coated, yet hydrophobic ZnO nanoparticles were effective HIPE stabilizers, yielding polymer foams with ZnO nanoparticles located predominately at their surface. These inorganic/organic hybrid foam-materials were subsequently calcined at 550 °C for 15 min to obtain inorganic macroporous ZnO foams with a morphology reminiscent to the original hybrid foam, and a specific surface area of 1.5 m(2) g(-1). Longer calcination time (550 °C, 15 h) resulted in a sea urchin like morphology of the ZnO foams, characterized by higher specific surface area of 5.5 m(2) g(-1). The latter foam type showed an appealing catalytic performance in the catalytic wet air oxidation (CWAO) process for the destruction of bisphenol A.

RESUMO

OBJECTIVE: In this work, continuous and size-segregated aerosol measurements at Mt. Krvavec, Slovenia, during the Eyjafjallajökull volcanic eruption were performed. Based on chemical and morphological characteristics of size-segregated particles, the presence of the volcanic aerosols after long-range transport to Slovenia was to be confirmed. RESULTS AND CONCLUSIONS: Continuous measurements with the aethalometer and SMPS indicated the suspected volcanic ash plume passing over the sampling site. The aerosols collected by discrete sampling showed a chemical signature similar to the known elemental signature of the Icelandic volcanic ash. Coarse particles showed a composition typical for silicates rich in metals; in many cases also S was present. Morphological analysis showed particles with features indicative of an explosive volcanic eruption, e.g., pumice and pumice shards, glass shards, minerals, evidence of steam condensation, etc. The high sulfate concentration associated with the fine particles resulted in sulfate crystallization within the cascade impactor leading to the formation of large structures resembling a "fern". Mass size distributions for Fe, Ti, Mn, Ca, Na, and Mg showed one primary peak (for Fe, Mn, and Ti at 2.8 µm; for Ca, Na, and Mg at ca. 4 µm), which supports the fact that most of the particles in the coarse sizes were silicates rich in metals. The size distribution of the water-soluble SO(4)(2-) showed a maximum peak at 0.75 µm, which also confirms the high sulfate concentration in the fine particles. Chemical and morphological characterization of aerosols collected at Mt. Krvavec indeed confirmed that volcanic ash plume passed over Slovenia.
