RESUMO

TLR4 is activated by the bacterial endotoxin lipopolysaccharide (LPS) and triggers two proinflammatory signaling cascades: a MyD88-dependent one in the plasma membrane, and the following TRIF-dependent one in endosomes. An inadequate inflammatory reaction can be detrimental for the organism by leading to sepsis. Therefore, novel approaches to therapeutic modulation of TLR4 signaling are being sought after. The TLR4 activity is tightly connected with the presence of CD14, a GPI-anchored protein that transfers LPS monomers to the receptor and controls its endocytosis. In this study we focused on CD14 trafficking as a still poorly understood factor affecting TLR4 activity. Two independent assays were used to show that after endocytosis CD14 can recycle back to the plasma membrane in both unstimulated and stimulated cells. This route of CD14 trafficking can be controlled by sorting nexins (SNX) 1, 2 and 6, and is important for maintaining the surface level and the total level of CD14, but can also affect the amount of TLR4. Silencing of these SNXs attenuated especially the CD14-dependent endosomal signaling of TLR4, making them a new target for therapeutic regulation of the inflammatory response of macrophages to LPS.

Chimeric antigen receptor (CAR) T-cell therapy has had considerable success in the treatment of B-cell malignancies. Targeting the B-lineage marker CD19 has brought great advances to the treatment of acute lymphoblastic leukemia and B-cell lymphomas. However, relapse remains an issue in many cases. Such relapse can result from downregulation or loss of CD19 from the malignant cell population or expression of alternate isoforms. Consequently, there remains a need to target alternative B-cell antigens and diversify the spectrum of epitopes targeted within the same antigen. CD22 has been identified as a substitute target in cases of CD19-negative relapse. One anti-CD22 antibody-clone m971-targets a membrane-proximal epitope of CD22 and has been widely validated and used in the clinic. Here, we have compared m971-CAR with a novel CAR derived from IS7, an antibody that targets a central epitope on CD22. The IS7-CAR has superior avidity and is active and specific against CD22-positive targets, including B-acute lymphoblastic leukemia patient-derived xenograft samples. Side-by-side comparisons indicated that while IS7-CAR killed less rapidly than m971-CAR in vitro, it remains efficient in controlling lymphoma xenograft models in vivo. Thus, IS7-CAR presents a potential alternative candidate for the treatment of refractory B-cell malignancies.

A new coordination compound of copper(II) with a tolfenamate ligand of the paddle-wheel-like structure [CuII2(Tolf)4(MeOH)2]∙2MeOH was obtained and structurally characterized. Chemical bonds of Cu(II)∙∙∙Cu(II) and Cu(II)-O were theoretically analyzed and compared with the results for selected similar structures from the CSD database. QTAIM analysis showed that the Cu(II)∙∙∙Cu(II) interaction has a strength comparable to a hydrogen bond, as indicated by the electron density at a critical point. The remaining QTAIM parameters indicate stability of the Cu(II)∙∙∙Cu(II) interaction. Other methods, such as NCI and NBO, also indicate a significant strength of this interaction. Thus, the Cu(II)∙∙∙Cu(II) interaction can be treated as one of the noncovalent interactions that affects the structure of the coordination compound, the packing of molecules in the crystal, and the general properties of the compound.

Despite the unquestionable success achieved by rituximab-based regimens in the management of diffuse large B-cell lymphoma (DLBCL), the high incidence of relapsed/refractory disease still remains a challenge. The widespread clinical use of chemo-immunotherapy demonstrated that it invariably leads to the induction of resistance; however, the molecular mechanisms underlying this phenomenon remain unclear. Rituximab-mediated therapeutic effect primarily relies on complement-dependent cytotoxicity and antibody-dependent cell cytotoxicity, and their outcome is often compromised following the development of resistance. Factors involved include inherent genetic characteristics and rituximab-induced changes in effectors cells, the role of ligand/receptor interactions between target and effector cells, and the tumor microenvironment. This review focuses on summarizing the emerging advances in the understanding of the molecular basis responsible for the resistance induced by various forms of immunotherapy used in DLBCL. We outline available models of resistance and delineate solutions that may improve the efficacy of standard therapeutic protocols, which might be essential for the rational design of novel therapeutic regimens.

In order to improve pharmaceutical properties of drugs, complexes are synthesized as combinations with other chemical substances. The complexes of fenamic acid and its derivatives, such as mefenamic-, tolfenamic- and flufenamic acid, with acridine were obtained and the X-ray structures were discussed. Formation of the crystals is determined by the presence of the intermolecular O-H…N hydrogen bond that occur between fenamic acids and acridine. Intermolecular interactions stabilizing the crystals such as π…π stacking, C-H…X (X = O, Cl) intermolecular hydrogen bonds as well as C-H…π and other dispersive interactions were analyzed by theoretical methods: the quantum theory of atoms in molecules (QTAIM) and noncovalent interaction (NCI) approaches.

The first discrete hexanuclear metal clusters with cores adopting the M6(µ-O)12 cubo-octahedral topology have been synthesized in the course of a simple one-pot reaction. We present a new class of rhenium clusters which are the first hexanuclear rhenium complexes with 12 bridging ligands and the first clusters with octahedrally arranged Re atoms bridged only by O atoms forming a unique cube-like Re6(µ-O)12 unit. Our synthetic strategy demonstrates a new approach to the syntheses of polynuclear rhenium complexes under mild conditions. We discovered that the [Re6(µ-O)12(3-Mepy)6]BPh4 cluster compound has the ability to undergo reversible or/and quasireversible redox reactions without changing spatial structure and overall geometry. Subsequently, a reduction reaction of [Re6(µ-O)12(3-Mepy)6]BPh4 was performed successfully and almost quantitatively resulting in the formation of the molecular [Re6(µ-O)12(3-Mepy)6] complex.

Rituximab (RTX) plus chemotherapy (R-CHOP) applied as a first-line therapy for lymphoma leads to a relapse in approximately 40% of the patients. Therefore, novel approaches to treat aggressive lymphomas are being intensively investigated. Several RTX-resistant (RR) cell lines have been established as surrogate models to study resistance to R-CHOP. Our study reveals that RR cells are characterized by a major downregulation of CD37, a molecule currently explored as a target for immunotherapy. Using CD20 knockout (KO) cell lines, we demonstrate that CD20 and CD37 form a complex, and hypothesize that the presence of CD20 stabilizes CD37 in the cell membrane. Consequently, we observe a diminished cytotoxicity of anti-CD37 monoclonal antibody (mAb) in complement-dependent cytotoxicity in both RR and CD20 KO cells that can be partially restored upon lysosome inhibition. On the other hand, the internalization rate of anti-CD37 mAb in CD20 KO cells is increased when compared to controls, suggesting unhampered efficacy of antibody drug conjugates (ADCs). Importantly, even a major downregulation in CD37 levels does not hamper the efficacy of CD37-directed chimeric antigen receptor (CAR) T cells. In summary, we present here a novel mechanism of CD37 regulation with further implications for the use of anti-CD37 immunotherapies.

The title salt, (C(12)H(12)N(4)O(2))[ReCl(6)], consists of 2,2'-[(1E,2E)-1,2-bis-(hy-droxy-imino)-ethane-1,2-di-yl]dipyridinium cations and [ReCl(6)](2-) anions which are both located on inversion centres. Each cation consists of a glyoxime moiety attached to two protonated pyridine rings in ortho positions. In the crystal, E,E isomers of the cation are observed which differ in the spatial arrangement of the pyridine rings. These rings are positionally disordered over two positions with site-occupancy factors of 0.786 (7) and 0.214 (7). The geometry of the cation is compared with that of a recently reported dipyridyl-glyoxime with the same configuration. The cations and anions are involved in a network of inter-molecular O-H⋯Cl, N-H⋯Cl and C-H⋯Cl hydrogen bonds.

RESUMO

Chimeric antigen receptor (CAR)-T cell therapy is undeniably a promising tool in combating various types of hematological malignancies. However, it is not yet optimal and a significant number of patients experience a lack of response or relapse after the treatment. Therapy improvement requires careful analysis of the occurring problems and a deeper understanding of the reasons that stand behind them. In this review, we summarize the recent knowledge about CAR-T products' clinical performance and discuss diversified approaches taken to improve the major shortcomings of this therapy. Especially, we prioritize the challenges faced by CD19 CAR-T cell-based treatment of B cell-derived malignancies and revise the latest insights about mechanisms mediating therapy resistance. Since the loss of CD19 is one of the major obstacles to the success of CAR-T cell therapy, we present antigens that could be alternatively used for the treatment of various types of B cell-derived cancers.

BACKGROUND: Immune checkpoint inhibitors and chimeric antigen receptor (CAR)-based therapies have transformed cancer treatment. Recently, combining these approaches into a strategy of PD-L1-targeted CAR has been proposed to target PD-L1high tumors. Our study provides new information on the efficacy of such an approach against PD-L1low targets. METHODS: New atezolizumab-based PD-L1-targeted CAR was generated and introduced into T, NK, or NK-92 cells. Breast cancer MDA-MB-231 and MCF-7 cell lines or non-malignant cells (HEK293T, HMEC, MCF-10A, or BM-MSC) were used as targets to assess the reactivity or cytotoxic activity of the PD-L1-CAR-bearing immune effector cells. Stimulation with IFNγ or with supernatants from activated CAR T cells were used to induce upregulation of PD-L1 molecule expression on the target cells. HER2-CAR T cells were used for combination with PD-L1-CAR T cells against MCF-7 cells. RESULTS: PD-L1-CAR effector cells responded vigorously with degranulation and cytokine production to PD-L1high MDA-MB-231 cells, but not to PD-L1low MCF-7 cells. However, in long-term killing assays, both MDA-MB-231 and MCF-7 cells were eliminated by the PD-L1-CAR cells, although with a delay in the case of PD-L1low MCF-7 cells. Notably, the coculture of MCF-7 cells with activated PD-L1-CAR cells led to bystander induction of PD-L1 expression on MCF-7 cells and to the unique self-amplifying effect of the PD-L1-CAR cells. Accordingly, PD-L1-CAR T cells were active not only against MDA-MD-231 and MCF-7-PD-L1 but also against MCF-7-pLVX cells in tumor xenograft models. Importantly, we have also observed potent cytotoxic effects of PD-L1-CAR cells against non-malignant MCF-10A, HMEC, and BM-MSC cells, but not against HEK293T cells that initially did not express PD-L1 and were unresponsive to the stimulation . Finally, we have observed that HER-2-CAR T cells stimulate PD-L1 expression on MCF-7 cells and therefore accelerate the functionality of PD-L1-CAR T cells when used in combination. CONCLUSIONS: In summary, our studies show that CAR-effector cells trigger the expression of PD-L1 on target cells, which in case of PD-L1-CAR results in the unique self-amplification phenomenon. This self-amplifying effect could be responsible for the enhanced cytotoxicity of PD-L1-CAR T cells against both malignant and non-malignant cells and implies extensive caution in introducing PD-L1-CAR strategy into clinical studies.

Oxidative stress, caused by the imbalance between reactive species generation and the dysfunctional capacity of antioxidant defenses, is one of the characteristic features of cancer. Here, we quantified hydrogen peroxide in the tumor microenvironment (TME) and demonstrated that hydrogen peroxide concentrations are elevated in tumor interstitial fluid isolated from murine breast cancers in vivo, when compared with blood or normal subcutaneous fluid. Therefore, we investigated the effects of increased hydrogen peroxide concentration on immune cell functions. NK cells were more susceptible to hydrogen peroxide than T cells or B cells, and by comparing T, B, and NK cells' sensitivities to redox stress and their antioxidant capacities, we identified peroxiredoxin-1 (PRDX1) as a lacking element of NK cells' antioxidative defense. We observed that priming with IL15 protected NK cells' functions in the presence of high hydrogen peroxide and simultaneously upregulated PRDX1 expression. However, the effect of IL15 on PRDX1 expression was transient and strictly dependent on the presence of the cytokine. Therefore, we genetically modified NK cells to stably overexpress PRDX1, which led to increased survival and NK cell activity in redox stress conditions. Finally, we generated PD-L1-CAR NK cells overexpressing PRDX1 that displayed potent antitumor activity against breast cancer cells under oxidative stress. These results demonstrate that hydrogen peroxide, at concentrations detected in the TME, suppresses NK cell function and that genetic modification strategies can improve CAR NK cells' resistance and potency against solid tumors.

A family of oxidovanadium(iv) compounds containing linear diaminebis(phenolate (salans) L1-5 ligands (L1 = [MeNCH2CH2NMe(CH2-4-CMe2CH2CMe3-C6H3O)2]2-; L2 = [MeNCH2CH2NMe(CH2-4-CH3-C6H3O)2]2-; L3 = [MeNCH2CH2NMe(CH2-4-Cl-C6H3O)2]2-; L4 = {MeNCH2CH2NMe[CH2-4,6-(CH3)2-C6H2O]2}2-; and L5 = {MeNCH2CH2NMe[CH2-4,6-(Br)2-C6H2O]2}2-) and non-oxidovanadium(iii) with L2,4 and acac ligands has been prepared and characterized by chemical and physical techniques. Reactions of [VO(acac)2] with ligand precursors H2L2,4 in toluene or hexane afforded vanadium(iii) compounds [V(L-κ4ONNO)(acac)] (1, L2; 2, L4), while the use of acetonitrile or ethanol led to the formation of dimeric oxidovanadium(iv) [(VO)2(µ-L-κ4ONNO)2] (3, L1; 4, L2; 5, L3) and monomeric [VO(L-κ4ONNO)] (6, L4, 7, L5) compounds. As shown by X-ray crystallography, compounds 1 and 2 are monomeric, in which the chelating ligands afford octahedral cis-α geometry at the vanadium center. In the dimeric structures of 3-5, the six-coordinate vanadium centers are bridged via two oxygen atoms of the L1-3 ligands while the L4,5 ligands generate square pyramidal structures of the monomeric 6 and 7 compounds. HFEPR studies allowed the determination of the spin Hamiltonian parameters of the S = 1 spin state of the monomeric V(iii) and dimeric V(iv), and S = ½ in monomeric V(iv) compounds. Magnetic measurements of 3-5 indicated weak ferromagnetic metal-metal exchange interactions. A reaction course for the deoxygenation and reduction of vanadyl-salan compounds is proposed.

BACKGROUND: The skeletal muscle reconstruction occurs thanks to unipotent stem cells, i.e., satellite cells. The satellite cells remain quiescent and localized between myofiber sarcolemma and basal lamina. They are activated in response to muscle injury, proliferate, differentiate into myoblasts, and recreate myofibers. The stem and progenitor cells support skeletal muscle regeneration, which could be disturbed by extensive damage, sarcopenia, cachexia, or genetic diseases like dystrophy. Many lines of evidence showed that the level of oxygen regulates the course of cell proliferation and differentiation. METHODS: In the present study, we analyzed hypoxia impact on human and pig bone marrow-derived mesenchymal stromal cell (MSC) and mouse myoblast proliferation, differentiation, and fusion. Moreover, the influence of the transplantation of human bone marrow-derived MSCs cultured under hypoxic conditions on skeletal muscle regeneration was studied. RESULTS: We showed that bone marrow-derived MSCs increased VEGF expression and improved myogenesis under hypoxic conditions in vitro. Transplantation of hypoxia preconditioned bone marrow-derived MSCs into injured muscles resulted in the improved cell engraftment and formation of new vessels. CONCLUSIONS: We suggested that SDF-1 and VEGF secreted by hypoxia preconditioned bone marrow-derived MSCs played an essential role in cell engraftment and angiogenesis. Importantly, hypoxia preconditioned bone marrow-derived MSCs more efficiently engrafted injured muscles; however, they did not undergo myogenic differentiation.

The machinability of hard-to-cut CoCr alloys manufactured by Selective Laser Melting (SLM) technology is not yet sufficiently studied. Therefore, this work focuses on evaluation of surface texture formation during face turning of CoCr alloy. As part of the research, two specimen types were subject to comparison: made with the application of conventional casting and manufactured by additive manufacturing-SLM. A number of analytical and experimental methods were employed to describe the specimen composition and morphology, as: X-Ray Diffraction Analysis (XRD), optical metallurgical microscopy, confocal optical microscopy, and Vickers hardness HV0.1 measurements. In the next stage, the measurements of surface topographies formed during turning in a range of variable cutting speeds and feeds were carried out. Ultimately the multi-factor MANOVA (Multivariate Analysis of Variance) illustrating the influence of manufacturing technology, cutting speed, and feed ratio on selected surface parameters of samples was made. It has been demonstrated that during face turning with feeds up to 0.15 mm/rev, the similar values of surface roughness height and material ratio curve parameters were reached for both tested CoCr alloys. However, in a range of higher feed values, the surface quality of CoCr samples fabricated by SLM was lower than that reached for CoCr after casting process.

In the title salt, [ReO(2)(C(5)H(5)N)(4)]I(3), the cation and anion are both located on centres of symmetry. The Re(V) atom adopts a trans-ReO(2)N(4) octa-hedral coordination and short intra-molecular C-H⋯O contacts occur within the cation. In the crystal, the cations form layers perpendicular to [100] and a weak C-H⋯O inter-action links the cations.

The one-dimensional polymeric structure of sodium diaquafenamate-water (1/1) was studied by X-ray diffraction. The sodium cation is coordinated to one oxygen atom of the carboxylate group and to four water oxygen atoms. To characterize the Na-O bonds, the quantum theory of atoms in molecules (QTAIM) and noncovalent interaction (NCI) approaches have been used. Both methods confirmed that the Na-O bonds are very weak, comparable with the weak N-H...O intramolecular hydrogen bond. The polymeric structure is stabilized by the interaction of the sodium cation with the surrounding water molecules.

RESUMO

α-Lipoic acid (LA) is a unique antioxidant that is not only effective in affording protection against oxidative stress but also plays an essential role in metabolic processes of all living organisms. Therefore, the determination of LA and its metabolites content is crucial for understanding their physiological and pathophysiological functions. Most of the recently developed methods for the detection and determination of LA and its metabolites in various biological samples have focused on sample preparation procedures involving but not limited to sampling, extraction and storage. The main goal of this review is to summarize and critically evaluate the current state of the art of analytical procedures applied to the determination of LA and related compounds in biological samples.
