AIM: The purpose of this study was to evaluate and compare the cytotoxicity and genotoxicity of two bioceramic root canal sealers: EndoSequence BC and iRoot SP with zinc oxide eugenol sealers on fibroblast cell line. MATERIALS AND METHODS: The sealers tested were zinc oxide eugenol, EndoSequence BC, and iRoot SP. Each material was mixed according to the manufacturer's instructions and mounted into sterile polyethylene color-coded rings, for cytotoxicity and genotoxicity evaluation. After 48 hours, the set materials were transferred to previously marked wells and cytotoxicity evaluation to L929 murine fibroblast cells was done by 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. The percentages of viable cells were then calculated and values were statistically analyzed by Kruskal-Wallis test. The evaluation of genotoxicity of the materials to L929 murine fibroblast cells was carried out by Comet assay. To quantify deoxyribonucleic acid (DNA) damage, the following comet parameters were evaluated in the assay using Comet scoring software: tail length, tail moment, and Olive moment. The values were statistically analyzed using Kruskal-Wallis test with a significance value set to p < 0.05. RESULTS: The results of the study showed that both cytotoxicity and genotoxicity evaluation by MTT assay and Comet assay can be done on L929 murine fibroblast cell line. Among the three tested materials, zinc oxide eugenol showed maximum cytotoxicity to the cells (30.64% viable cells), followed by EndoSequence BC (71.33% viable cells) and iRoot SP (75.11% viable cells). The evaluation of DNA damage by genotoxicity assessment showed iRoot SP to be least genotoxic followed closely by EndoSequence BC. Zinc oxide eugenol was genotoxic and induced more DNA damage on the fibroblast cell line studied. The statistical analyses for both the assays were nonsignificant. CONCLUSION: All the three tested sealers showed varying degrees of cytotoxicity and genotoxicity while using fibro-blast cell line. Zinc oxide eugenol was most toxic in both the assays and iRoot SP showed least toxicity, followed closely by EndoSequence BC.

AIM: Restorative dentists employ different bonding systems between the resin and the dentin and other dentinal tissues to achieve the goal of micromechanical retention. Studies have shown that the bond between composite and dentin degrades over time because of the action of matrix metalloproteinases (MMPs) on collagen fibrils left unprotected by acid etching. The MMPs may be partially responsible for hybrid layer degradation. Since chlorhexidine (CHX) inhibits MMPs, we hypothesized that CHX would decelerate the loss of resin-dentin bonds. Hence, this in vitro study is intended to evaluate the effects of 2% CHX on hybrid layer and bonding interface. MATERIALS AND METHODS: Totally, 40 freshly extracted molars were randomly divided into four experimental groups. In all 40 specimens, class II cavities were prepared to a depth of 1 mm below the dentinoenamel junction with no axial wall, but the elimination of the proximal enamel ridge. The teeth were then randomly divided into four experimental groups, i.e., All Bond 2 without 2% CHX (group I), All Bond 2 with 2% CHX (group II), One Coat 7.0 without 2% CHX (group III), and One Coat 7.0 with 2% CHX (group IV). All the specimens were derooted and sectioned mesiodistally into two halves and placed under water at 37°C for 3 months and observed under scanning electron microscope for the hybrid layer and resin tag formation. RESULTS: Groups I and II showed statistically significant difference when the presence/absence of resin tags was compared. When groups III and IV were compared for the presence/absence of hybrid layer and resin tags, the results were statistically significant. CONCLUSION: Between all the four experimental groups, irrespective of the bonding systems used, we concluded that groups with 2% CHX usage showed promising results with presence/ absence of hybrid layer and resin tags formation. CLINICAL SIGNIFICANCE: Studies suggest that the bond between composite and dentin degrades over time because of the action of MMPs on collagen fibrils left unprotected by acid etching. Measures should be taken to prevent this from happening and thus allow bond between composite and dentin last longer.

AIM: The aim of this study was to evaluate the tensile strength of die stone incorporated with sodium and calcium hypochlorite as disinfectants. MATERIALS AND METHODS: Two commercially available type IV die stone (Kalrock: Kalabhai Karson Pvt., Ltd and Pearlstone: Asian Chemicals) and two commercially available disinfectant solutions (sodium hypochlorite and calcium hypochlorite: Beachem Laboratory Chemical Private Limited, Chennai and Leo Chem Private Limited, Bengaluru) were used in this study, and the tensile strength was measured using Lloyd's Universal Testing Machine. RESULTS: The results show that incorporating the disinfecting solutions decreases the tensile strength of both products. The effect of decreasing tensile strength on type IV gypsum product is seen more in calcium hypochlorite when compared with sodium hypochlorite disinfecting solution, and the tensile strength of Kalrock specimens is higher than Pearlstone specimens after disinfecting with sodium hypochlorite and calcium hypochlorite solution. The statistical results also show significant results in all the groups when compared with the control group. CONCLUSION: The incorporation of sodium and calcium hypochlorite disinfecting solutions is not an encouraging method for both die materials as it reduces the tensile strength of type IV gypsum product. Tensile strength of Kalstone® die material is superior than Pearlstone® die material after mixing with sodium hypochlorite and calcium hypochlorite. CLINICAL SIGNIFICANCE: According to the recommendations of Americans with Disability Act (ADA) and the Centers for Disease Control and Prevention, disinfecting the whole cast without or minimal changes in physical and mechanical properties was the motto of the study. The tensile strength in type IV gypsum product plays a most important role in retrieval of cast from impression, especially in narrow tooth preparation. This study reveals that incorporating method of disinfecting solutions is not recommended as it reduces the tensile strength.

Whole cell based screens to identify hits against Mycobacterium tuberculosis (Mtb), carried out under replicating and non-replicating (NRP) conditions, resulted in the identification of multiple, novel but structurally related spiropiperidines with potent antitubercular properties. These compounds could be further classified into three classes namely 3-(3-aryl-1,2,4-oxadiazol-5-yl)-1'-alkylspiro[indene-1,4'-piperidine] (abbr. spiroindenes), 4-(3-aryl-1,2,4-oxadiazol-5-yl)-1'-alkylspiro[chromene-2,4'-piperidine] (abbr. spirochromenes) and 1'-benzylspiro[indole-1,4'-piperidin]-2(1H)-one (abbr. spiroindolones). Spiroindenes showed ⩾ 4 log10 kill (at 2-12 µM) on replicating Mtb, but were moderately active under non replicating conditions. Whole genome sequencing efforts of spiroindene resistant mutants resulted in the identification of I292L mutation in MmpL3 (Mycobacterial membrane protein Large), required for the assembly of mycolic acid into the cell wall core of Mtb. MIC modulation studies demonstrated that the mutants were cross-resistant to spirochromenes but not to spiroindolones. This Letter describes lead identification efforts to improve potency while reducing the lipophilicity and hERG liabilities of spiroindenes. Additionally, as deduced from the SAR studies, we provide insights regarding the new chemical opportunities that the spiroindolones can offer to the TB drug discovery initiatives.

One of many hazardous workplaces includes the construction sites as they involve several dangerous tasks. Many studies have revealed that material handling equipment is a major cause of accidents at these sites. Though safety measures are being followed and monitored continuously, accident rates are still high as either workers are unaware of hazards or the safety regulations are not being strictly followed. This paper analyses the safety management systems at construction sites through means of questionnaire surveys with employees, specifically referring to safety of material handling equipment. Based on results of the questionnaire surveys, two construction sites were selected for a safety education program targeting worker safety related to material handling equipment. Knowledge levels of the workers were gathered before and after the program and results obtained were subjected to a t-test analysis to mark significance level of the conducted safety education program.

Pollination and reproductive biology of a dioecious tree Canarium strictum Roxb. (Burseraceae) was extensively studied within the Agumbe forest range of Western Ghats, Karnataka to identify primary pollen vectors and to enumerate interrelationship with the pollinators. The study also investigated phenology, floral biology, pollen production, pollen viability, stigma receptivity and nectar production. Trees produced functionally unisexual flowers with white petals, organized densely on inflorescences. Staminate flowers produced high percentage of viable pollen and relatively abundant nectar (15.75 µl) as a reward to the pollinators, while pistillate flowers produced only nectar (12 µl). Successful fruit set with wind pollination was facilitated by synchronization of flowering male and female trees, long term receptivity of stigma in female flowers and extended lifespan of flowers. The highest mean percent of fruit set with hand cross-pollination (µ = 91.06) suggests the influence of local male tree density, as well as, frequency and abundance of pollinator community on fruit set by open pollination.

Scaffold hopping from the thiazolopyridine ureas led to thiazolopyridone ureas with potent antitubercular activity acting through inhibition of DNA GyrB ATPase activity. Structural diversity was introduced, by extension of substituents from the thiazolopyridone N-4 position, to access hydrophobic interactions in the ribose pocket of the ATP binding region of GyrB. Further optimization of hydrogen bond interactions with arginines in site-2 of GyrB active site pocket led to potent inhibition of the enzyme (IC50 2 nM) along with potent cellular activity (MIC=0.1 µM) against Mycobacterium tuberculosis (Mtb). Efficacy was demonstrated in an acute mouse model of tuberculosis on oral administration.

PURPOSE: To evaluate the curing depth and compressive strength of dental composite using halogen light curing unit and light emitting diode light curing unit. MATERIALS AND METHODS: Eighty cylindrical composite specimens were prepared using posterior composite P60(3M). Forty specimens, out of which 20 samples (group A) cured with halogen light and 20 samples (group B) cured using light emitting diode (LED) light were checked for curing depth according to ISO 4049. Remaining 40 samples out of which 20 samples (group I) cured using halogen light and 20 samples (group II) cured using LED light were checked for compressive strength using Instron universal testing machine. RESULTS: Twenty samples (group A) cured with halogen light showed better curing depth than 20 samples (group B) cured with LED light. Twenty samples (group I) cured with halogen light showed almost similar results as 20 samples (group II) cured with LED light for compressive strength. CONCLUSION: Halogen light commonly used to cure composite resin have greater depth of cure, when compared to LED light, while both the lights produced compressive strength which is almost similar. CLINICAL SIGNIFICANCE: Lower depth of cure with the LED unit, compared to the QTH unit, is associated with different light scattering due to differences in spectral emission. LED technology differs from QTH by the spectral emission that favorably matches the absorption spectrum of camphorquinone.

BACKGROUND: Oral submucous fibrosis (OSF) is a chronic oral mucosal condition with pre-malignant potential. The association between psychiatric morbidity and OSF has been sparsely studied. METHODS: This cross-sectional study included 33 consecutive subjects clinically diagnosed with OSF who attended an academic tertiary care dental centre. General Health Questionnaire-28 was administered to assess the psychological morbidity. Dependence to areca products was assessed using the ICD-10 criteria. Patient group was compared with an age and gender matched control group (n=33) from the same hospital that comprised patients who had dental diseases other than OSF and had no areca/tobacco habit. RESULTS: Total mean scores (SD) of GHQ-28 for study group and control group respectively were 48.2 (18.1) vs. 24.2 (6.5) (P<0.001). Among the patient group, the mean total GHQ scores were significantly higher in those with functional stage C (those with worst functional staging) of OSF 63.4 (15.2) when compared with those with stage B 54 (14.2) and stage A 38.7 (17) (d.f.=2; P=0.005). 22 (66.7%) of patients met ICD-10 criteria for dependence to areca products. CONCLUSIONS: OSF may be associated with considerable psychological morbidity. This morbidity may be significantly more in patients with worst functional staging. It is difficult to determine whether this is the cause or effect of the disease. Further systematic studies are urgently needed to address this issue.

A method for the fabrication of flexible electrical circuits on polyaramid substrates is presented based on laser-induced carbonization followed by copper electroplating. Locally carbonized flexible sheets of polyaramid (Nomex), by laser radiation, create rough and highly porous microstructures that show a higher degree of graphitization than thermally carbonized Nomex sheets. The found recipe for laser-induced carbonization creates conductivities of up to ∼45 S cm-1, thereby exceeding that observed for thermally pyrolyzed materials (∼38 S cm-1) and laser carbon derived from Kapton using the same laser wavelength (∼35 S cm-1). The electrical conductivity of the carbonized tracks was further improved by electroplating with copper. To demonstrate the electrical performance, fabricated circuits were tested and improvement of the sheet resistance was determined. Copper films exhibit antimicrobial activity and were used to fabricate customized flexible antibacterial coatings. The integration of laser carbonization and electroplating technologies in a polyaramid substrate points to the development of customized circuit designs for smart textiles operating in high-temperature environments.

During high temperature pyrolysis of polymer thin films, nanocrystalline graphene with a high defect density, active edges and various nanostructures is formed. The catalyst-free synthesis is based on the temperature assisted transformation of a polymer precursor. The processing conditions have a strong influence on the final thin film properties. However, the precise elemental processes that govern the polymer pyrolysis at high temperatures are unknown. By means of time resolved in situ transmission electron microscopy investigations we reveal that the reactivity of defects and unsaturated edges plays an integral role in the structural dynamics. Both mobile and stationary structures with varying size, shape and dynamics have been observed. During high temperature experiments, small graphene fragments (nanoflakes) are highly unstable and tend to lose atoms or small groups of atoms, while adjacent larger domains grow by addition of atoms, indicating an Ostwald-like ripening in these 2D materials, besides the mechanism of lateral merging of nanoflakes with edges. These processes are also observed in low-dose experiments with negligible electron beam influence. Based on energy barrier calculations, we propose several inherent temperature-driven mechanisms of atom rearrangement, partially involving catalyzing unsaturated sites. Our results show that the fundamentally different high temperature behavior and stability of nanocrystalline graphene in contrast to pristine graphene is caused by its reactive nature. The detailed analysis of the observed dynamics provides a pioneering overview of the relevant processes during ncg heating.

RESUMO

A straightforward one-step process was developed, in which CO2 gas is directly converted into multi-layer graphene via atmospheric pressure chemical vapor deposition (APCVD). A bimetallic alloy film based on Cu and Pd was employed as the catalyst and substrate. In this study, we found that the quantity of Cu required for the CO2 conversion process is high (>82 at %). The findings gained in this study serve as a foundation for further studies of metallic alloys for the thermo-reduction of CO2 to graphene under CVD conditions.

Glassy carbon is a graphene-rich form of elemental carbon obtained from pyrolysis of polymers, which is composed of three-dimensionally arranged, curved graphene fragments alongside fractions of disordered carbon and voids. Pyrolysis encompasses gradual heating of polymers at ≥ 900 °C under inert atmosphere, followed by cooling to room temperature. Here we report on an experimental method to perform in situ high-resolution transmission electron microscopy (HR-TEM) for the direct visualization of microstructural evolution in a pyrolyzing polymer in the 500-1200 °C temperature range. The results are compared with the existing microstructural models of glassy carbon. Reported experiments are performed at 80 kV acceleration voltage using MEMS-based heating chips as sample substrates to minimize any undesired beam-damage or sample preparation induced transformations. The outcome suggests that the geometry, expansion and atomic arrangement within the resulting graphene fragments constantly change, and that the intermediate structures provide important cues on the evolution of glassy carbon. A complete understanding of the pyrolysis process will allow for a general process tuning specific to the precursor polymer for obtaining glassy carbon with pre-defined properties.

Based on magnetic-field-assisted growth of carbon nanofibers in an open ethanol flame we fabricated arrays of carbon nanofibers with different degrees of orientation. Inspired by the dry adhesive system of geckos we investigated the adhesive properties of such carbon nanofiber arrays with ordered and random orientation. AFM-based force spectroscopy revealed that adhesion force and energy rise linear with preload force. Carbon nanofibers oriented by a magnetic field show a 68% higher adhesion (0.66 N/cm2) than the randomly oriented fibers. Endurance tests revealed that the carbon nanofiber arrays withstand 50.000 attachment/detachment cycles without observable wear.

Graphitization of polymers is an effective way to synthesize nanocrystalline graphene on different substrates with tunable shape, thickness and properties. The catalyst free synthesis results in crystallite sizes on the order of a few nanometers, significantly smaller than commonly prepared polycrystalline graphene. Even though this method provides the flexibility of graphitizing polymer films on different substrates, substrate free graphitization of freestanding polymer layers has not been studied yet. We report for the first time the thermally induced graphitization and domain growth of free-standing nanocrystalline graphene thin films using in situ TEM techniques. High resolution transmission electron microscopy (HRTEM), selected area electron diffraction (SAED) and electron energy loss spectroscopy (EELS) techniques were used to analyze the graphitization and the evolution of nanocrystalline domains at different temperatures by characterizing the crystallinity and domain size, further supported by ex situ Raman spectroscopy. The graphitization was comparable to the substrate supported heating and the temperature dependence of graphitization was analyzed. In addition, the in situ analysis of the graphitization enabled direct imaging of some of the growth processes taking place at different temperatures.

The approval of bedaquiline to treat tuberculosis has validated adenosine triphosphate (ATP) synthase as an attractive target to kill Mycobacterium tuberculosis (Mtb). Herein, we report the discovery of two diverse lead series imidazo[1,2-a]pyridine ethers (IPE) and squaramides (SQA) as inhibitors of mycobacterial ATP synthesis. Through medicinal chemistry exploration, we established a robust structure-activity relationship of these two scaffolds, resulting in nanomolar potencies in an ATP synthesis inhibition assay. A biochemical deconvolution cascade suggested cytochrome c oxidase as the potential target of IPE class of molecules, whereas characterization of spontaneous resistant mutants of SQAs unambiguously identified ATP synthase as its molecular target. Absence of cross resistance against bedaquiline resistant mutants suggested a different binding site for SQAs on ATP synthase. Metal matrix composites (MMCs) exhibit improved strength but suffer from low ductility. Metal particles reinforcement can be an alternative to retain the ductility in MMCs (Bauri and Yadav, 2010; Thakur and Gupta, 2007) [1,2]. However, processing such composites by conventional routes is difficult. The data presented here relates to friction stir processing (FSP) that was used to process metal particles reinforced aluminum matrix composites. The data is the processing parameters, rotation and traverse speeds, which were optimized to incorporate Ni particles. A wide range of parameters covering tool rotation speeds from 1000 rpm to 1800 rpm and a range of traverse speeds from 6 mm/min to 24 mm/min were explored in order to get a defect free stir zone and uniform distribution of particles. The right combination of rotation and traverse speed was found from these experiments. Both as-received coarse particles (70 µm) and ball-milled finer particles (10 µm) were incorporated in the Al matrix using the optimized parameters.

Structure-activity relationship (SAR) exploration on the left-hand side (LHS) of a novel class of bacterial topoisomerase inhibitors led to a significant improvement in the selectivity against hERG cardiac channel binding with concomitant potent antimycobacterial activity. Bulky polar substituents at the C-7 position of the naphthyridone ring did not disturb its positioning between two base pairs of DNA. Further optimization of the polar substituents on the LHS of the naphthyridone ring led to potent antimycobacterial activity (Mtb MIC = 0.06 µM) against Mycobacterium tuberculosis (Mtb). Additionally, this knowledge provided a robust SAR understanding to mitigate the hERG risk. This compound class inhibits Mtb DNA gyrase and retains its antimycobacterial activity against moxifloxacin-resistant strains of Mtb. Finally, we demonstrate in vivo proof of concept in an acute mouse model of TB following oral administration of compound 19.

RESUMO

We present the exact rogue wave solutions of the quasi-one-dimensional inhomogeneous Gross-Pitaevskii equation by using similarity transformation. Then, by employing the exact analytical solutions we have studied the controllable behavior of rogue waves in the Bose-Einstein condensates context for the experimentally relevant systems. Additionally, we have also investigated the nonlinear tunneling of rogue waves through a conventional hyperbolic barrier and periodic barrier. We have found that, for the conventional nonlinearity barrier case, rogue waves are localized in space and time and get amplified near the barrier, while for the dispersion barrier case rogue waves are localized in space and propagating in time and their amplitude is reduced at the barrier location. In the case of the periodic barrier, the interesting dynamical features of rogue waves are obtained and analyzed analytically.

BACKGROUND: To evaluate the efficacy of topical antiinflammatory agent (amlexanox 5%), along with topical antiseptic, analgesic, and anesthetic agent (benzalkonium chloride 0.01%, choline salicylate 8.7% and lidocaine hydrochloride 2%), in promoting ulcer healing, decreasing ulcer size, erythema, pain and recurrence in minor RAS. MATERIALS & METHODS: A randomized control trial was conducted on 100 patients of RAS who fulfilled the inclusion criteria. The number, size, erythema and pain with the ulcer were recorded. Visual analogue scale (VAS) and erythema scale were used to record pain and erythema. 50 patients comprising the study group received anti inflammatory paste (amlexanox 5%) applied four times daily and the control group of 50 patients received topical antiseptic, analgesic, and anesthetic agent (benzalkonium chloride 0.01%, choline salicylate 8.7% and lidocaine hydrochloride 2%) paste, patients were evaluated after 3rd, 6th, 9th and on 30th, 60th day for recurrence. RESULTS: The study group had reduction in ulcer number, size; erythema, pain and frequency of ulcers during follow up. The healing period and recurrence of ulceration reduced in both the groups but the study group had significant reduction in 30th and 60th day follow up for recurrence of ulcers. CONCLUSION: Amlexanox 5% can reduce the frequency, duration and symptoms associated with the aphthous ulcers with no sideeffects attributed to the drug.
