RESUMO

Cellular senescence associates with pathological aging and tissue dysfunctions. Studies utilizing mouse models for cell lineage tracings have emphasized the importance of senescence heterogeneity in different organs and cell types. Here, we constructed a p21- (Akaluc - tdTomato - Diphtheria Toxin Receptor [DTR]) (ATD) mouse model to specifically study the undefined mechanism for p21-expressing senescent cells in the aged and liver injury animals. The successful expressions of these genes enabled in vitro flow cytometric sorting, in vivo tracing, and elimination of p21-expressing senescent cells. During the natural aging process, p21-expressing cells were found in various tissues of p21-ATD mice. Eliminating p21-expressing cells in the aged p21-ATD mice recovered their multiple biological functions. p21-ATD/Fah-/- mice, bred from p21-ATD mice and fumarylacetoacetate hydrolase (Fah)-/- mice of liver injury, showed that the majority of their senescent hepatocytes were the phenotype of p21+ rather than p16+. Furthermore, eliminating the p21-expressing hepatocytes significantly promoted the engraftment of grafted hepatocytes and facilitated liver repopulation, resulting in significant recovery from liver injury. Our p21-ATD mouse model serves as an optimal model for studying the pattern and function of p21-expressing senescent cells under the physical and pathological conditions during aging.

OBJECTIVES: Heart rate variability (HRV) is becoming more prevalent as a measurable parameter in wearable sleep-monitoring devices, which are simple and effective instruments for illness evaluation. Currently, most studies on investigating OSA severity and HRV have measured heart rates during wakefulness or sleep. Therefore, the objective of this study was to investigate the circadian rhythm of HRV in male patients with OSA and its value for the estimation of OSA severity using group-based trajectory modeling. METHODS: Patients with complaints of snoring were enrolled from the Sleep Center of Shandong Qianfoshan Hospital. Patients were divided into 3 groups according to apnea hypopnea index (AHI in events/h), as follows: (<15, 15≤AHI<30, and ≥30). HRV parameters were calculated using 24 h Holter monitoring, which included time-domain and frequency-domain indices. Circadian differences in the standard deviation of normal to normal (SDNN) were evaluated for OSA severity using analysis of variance, trajectory analysis, and multinomial logistic regression. RESULTS: A total of 228 patients were enrolled, 47 with mild OSA, 48 moderate, and 133 severe. Patients with severe OSA exhibited reduced triangular index and higher very low frequency than those in the other groups. Circadian HRV showed that nocturnal SDNN was considerably higher than daytime SDNN in patients with severe OSA. The difference among the OSA groups was significant at 23, 24, 2, and 3 o'clock sharp between the severe and moderate OSA groups (all P<0.05). The heterogeneity of circadian HRV trajectories in OSA was strongly associated with OSA severity, including sleep structure and hypoxia-related parameters. Among the low-to-low, low-to-high, high-to-low, and high-to-high groups, OSA severity in the low-to-high group was the most severe, especially compared with the low-to-low and high-to-low SDNN groups, respectively. CONCLUSIONS: Circadian HRV in patients with OSA emerged as low daytime and high nocturnal in SDNN, particularly in men with severe OSA. The heterogeneity of circadian HRV revealed that trajectories with low daytime and significantly high nighttime were more strongly associated with severe OSA. Thus, circadian HRV trajectories may be useful to identify the severity of OSA.

AIMS: Sub-therapeutic use of antibiotics as a growth promoter in animal diets has either been banned or voluntarily withdrawn from use in many countries to help curb the emergence of antibiotic-resistant pathogens. Probiotics may be an alternative to antibiotics as a growth promoter. We investigated the effects of a novel probiotic strain, Bacillus amyloliquefaciens H57 (H57) on the performance and microbiome-associated metabolic potential. METHODS AND RESULTS: Broiler chickens were fed either sorghum- or wheat-based diets supplemented with the probiotic H57. The growth rate, feed intake, and feed conversion in supplemented birds were compared with those in non-supplemented control. Caecal microbial metabolic functions were studied with shotgun metagenomic sequencing. H57 supplementation significantly increased the growth rate and daily feed intake of meat chickens relative to the non-supplemented controls without any effect on feed conversion ratio. In addition, relative to the non-supplemented controls, gene-centric metagenomics revealed that H57 significantly altered the functional capacity of the caecal microbiome, with amino acid and vitamin synthesis pathways being positively associated with H57 supplementation. CONCLUSIONS: Bacillus amyloliquefaciens H57 improves the performance of meat chickens or broilers and significantly modifies the functional potential of their caecal microbiomes, with enhanced potential capacity for amino acid and vitamin biosynthesis.

BACKGROUND: To investigate if the correlation between left and right cerebral tissue oxygen saturation (SctO2) was affected by one-lung ventilation (OLV) in patients undergoing lung cancer surgery. METHODS: Patients who underwent surgery for lung cancer were enrolled. Left and right SctO2 were collected during anesthesia. The primary outcome was the correlation between left and right SctO2 at 30 min after OLV which was analysed by Pearson correlation and linear regression model. Secondary outcomes included the trend of left-right SctO2 change over the first 30 min after OLV, correlation of left-right SctO2 during OLV for each patient; maximal difference between left-right SctO2 and its relationship with postoperative delirium. RESULTS: Left-right SctO2 was moderately correlated at baseline (r = 0.690, P < 0.001) and poorly correlated at 30 min after OLV (r = 0.383, P < 0.001) in the Pearson correlation analysis. Linear regression analysis showed a poor correlation between left and right SctO2 at 30 min after OLV (r = 0.323, P < 0.001) after adjusting for confounders. The linear mixed model showed a change in left-right SctO2 over the first 30 min after OLV that was statistically significant (coefficient, -0.042; 95% CI, -0.070--0.014; P = 0.004). For the left-right SctO2 correlation during OLV in each patient, 62.9% (78/124) patients showed a strong correlation, 19.4% (24/124) a medium correlation, and the rest a poor correlation. The maximal difference between the left and right SctO2 was 13.5 (9.0, 20.0). Multivariate analysis showed that it was not associated with delirium (odds ratio [OR], 1.023; 95% CI, 0.963-1.087; P = 0.463). CONCLUSIONS: The correlation between left and right SctO2 was affected by one-lung ventilation in patients undergoing lung cancer surgery. This result indicates the requirement of bilateral SctO2 monitoring to reflect brain oxygenation. TRIAL REGISTRATION: This study was a secondary analysis of a cohort study approved by the Clinical Research Review Board of Peking University First Hospital (#2017-1378) and was registered in the Chinese Clinical Trial Registry on 10/09/2017 ( http://www.chictr.org.cn , ChiCTR-ROC-17012627).

BACKGROUND: Non-communicable diseases (NCDs) pose a major challenge to health economic cost and residents' health status. Community health workers (CHWs) are the gatekeeper of primary health care. OBJECTIVE: This study aimed to conduct a situational analysis of current human resource and requirements of NCDs-related training among CHWs in Chengdu with regard to address to understand the suggestions for improvement of challenges and barriers. METHODS: A descriptive online cross-sectional survey was conducted among CHWs (doctors and nurses) from 23 districts and counties in Chengdu. Sociodemographic and NCDs-related variables were collected. Univariate analysis and multiple response analysis were used to describe the characteristics of these variables. RESULTS: 711 doctors and 637 nurses completely responded. There were significant differences among gender, age, educational levels, professional title, working year, type of institution, urban circle and registration in general practice between doctors and nurses (P < 0.001). 60.6% of doctors were female, compared to 98.0% for nurses. 58.2% of doctors held a bachelor's degree compared with 45.4% of nurses, while 48.3% of nurses held a junior college degree compared with 25.7% of doctors. Higher levels of professional title and registration in general practice were found in doctors compared with nurses. The proportions of NCDs' category, NCDs-related roles and tasks, NCDs-related training contents and forms that CHWs have attend and hoped to gain more were significantly different between doctors and nurses (P < 0.001). In general, the proportions in nurses were much lower than those of doctors (P < 0.05). The top five diseases managed by CHWs were hypertension, diabetes, cerebrovascular disease, chronic respiratory diseases and mental diseases. The five most reported roles performed among doctors included the distribution of health education (91.4%), following up (85.9%), establishing archives (71.3%), medicine adjustment (64.7%) and treatment implementation (52.0%). The top three diseases managed by nurses were same with doctors. The top four and five tasks were contact with patients or health services (39.6%) and referral (16.6%) in nurses. Most CHWs had received primary and common diseases-related trainings, but they had few opportunities to study in a tertiary hospital (40.4% in doctors and 20.9% in nurses, respectively), attend domestic academic conferences (26.9% in doctors vs. 9.7% in nurses), and take part in training courses (44.9% in nurses). CHWs hoped that the above-discussed training contents and forms could be provided more in the future. Besides basic skills related trainings, some specific skills related trainings should be strengthened. CONCLUSION: The qualifications in doctors were much better than those of nurses. The roles performed by CHWs in NCDs management are varied form common and frequent disease management to subsequent follow up and supervision. CHWs hope to receive more desired and oriented trainings. There is a need for building capacity of CHWs, optimizing and defining CHWs' role, facilitating postgraduate medical education support and strengthening multidisciplinary collaboration would be effective in NCDs management.

Sweetpotato (Ipomoea batatas) is one of the most important crops in China. To gain a clearer picture of the occurrence of diseases on sweetpotato, 50 fields (100 plants/field) were randomly surveyed in prominent sweetpotato growing areas of Lulong county, Hebei Province in the years 2021-2022. Plants showing chlorotic leaf distortion with mildly twisted young leaves and stunted vines were observed frequently. It was similar to the symptoms of chlorotic leaf distortion of sweetpotato (Clark et al. 2013). The incidence of disease with patch pattern ranged from 15% to 30%. Ten symptomatic leaves were excised, surface disinfested with 2% sodium hypochlorite for 1 min, rinsed three times in sterilized ddH2O, and cultured on potato dextrose agar (PDA) at 25°C. Nine fungal isolates were obtained. A pure culture of representative isolate FD10 obtained after serial hyphal tip transfer was examined for morphological and genetic characters. Colonies of isolate FD10 on PDA at 25°C were slow growing (4±0.1mm/day) with aerial, white-to-pink mycelium. Lobed colonies had greyish-orange pigmentation in reverse and conidia aggegated in false heads. Conidiophores were prostrate and short. Phialides were mostly monophialidic but occasionally polyphialidic. Polyphialidic openings often denticulate in a rectangular arrangement. Microconidia were abundant, long, oval to allantoid, mostly none or one septate, and 4.79 to 9.53 × 2.08 to 3.22 µm (n = 20). Macroconidia were fusiform to falcate with a beaked apical cell and a footlike basal cell, 3 to 5 septate, and 25.03 to 52.92 × 2.56 to 4.49 µm. Chlamydospores were absent. All in agreement with the morphology of Fusarium denticulatum (Nirenberg and O'Donnell 1998). Genomic DNA of isolate FD10 was extracted. The EF-1α and ß-tubulin genes were amplified and sequenced (O'Donnell and Cigelnik 1997; O'Donnell et al. 1998). The obtained sequences were deposited in GenBank (accession nos. OQ555191 and OQ555192). BLASTn revealed that those sequences showed 99.86% (EF-1α) and 99.93% (ß-tubulin) homology with the relative sequences of F. denticulatum type strain CBS407.97 (accession nos. MT011002.1 and MT011060.1), respectively. Furthermore, a neighbor-joining phylogenetic tree based on the EF-1α and ß-tubulin sequences revealed that the isolate FD10 clustered with F. denticulatum. Based on morphological characteristics and sequence analysis, the isolate FD10 associated with chlorotic leaf distortion of sweetpotato was identified as F. denticulatum. Pathogenicity tests were performed by immersing ten 25-cm-long vine-tip cuttings of cultivar Jifen 1 origin from tissue culture in a conidial suspension (1 × 106 conidia per ml) of isolate FD10. Vines immersed in sterile distilled water used as a control. All inoculated plants in 25-cm plastic pots were incubated in a climate chamber at 28℃ and 80% RH for two and half months and the control were incubated in a separate climate chamber. Nine inoculated plants developed chlorotic terminals, moderate interveinal chlorosis and slight leaf distortion. No symptoms were observed on the control plants. The pathogen was reisolated from inoculated leaves and matched the morphological and molecular characteristics of the original isolates, thus fulfilling Koch's postulates. To our knowledge, this is the first report of F. denticulatum causing chlorotic leaf distortion of sweetpotato in China. Its identification will promote the management of this disease in China.

The state of angle cocks determines the air connectivity of freight trains, and detecting their state is helpful to improve the safety of the running trains. Although the current research for fault detection of angle cocks has achieved high accuracy, it only focuses on the detection of the closed state and non-closed state and treats them as normal and abnormal states, respectively. Since the non-closed state includes the fully open state and the misalignment state, while the latter may lead to brake abnormally, it is very necessary to further detect the misalignment state from the non-closed state. In this paper, we propose a coarse-to-fine localization method to achieve this goal. Firstly, the localization result of an angle cock is obtained by using the YOLOv4 model. Following that, the SVM model combined with the HOG feature of the localization result of an angle cock is used to further obtain its handle localization result. After that, the HOG feature of the sub-image only containing the handle localization result continues to be used in the SVM model to detect whether the angle cock is in the non-closed state or not. When the angle cock is in the non-closed state, its handle curve is fitted by binarization and window search, and the tilt angle of the handle is calculated by the minimum bounding rectangle. Finally, the misalignment state is detected when the tilt angle of the handle is less than the threshold. The effectiveness and robustness of the proposed method are verified by extensive experiments, and the accuracy of misalignment state detection for angle cocks reaches 96.49%.

Dragon fruit (Hylocereus undatus) is a tropical and subtropical fruit that undergoes multiple ripening cycles throughout the year. Accurate monitoring of the flower and fruit quantities at various stages is crucial for growers to estimate yields, plan orders, and implement effective management strategies. However, traditional manual counting methods are labor-intensive and inefficient. Deep learning techniques have proven effective for object recognition tasks but limited research has been conducted on dragon fruit due to its unique stem morphology and the coexistence of flowers and fruits. Additionally, the challenge lies in developing a lightweight recognition and tracking model that can be seamlessly integrated into mobile platforms, enabling on-site quantity counting. In this study, a video stream inspection method was proposed to classify and count dragon fruit flowers, immature fruits (green fruits), and mature fruits (red fruits) in a dragon fruit plantation. The approach involves three key steps: (1) utilizing the YOLOv5 network for the identification of different dragon fruit categories, (2) employing the improved ByteTrack object tracking algorithm to assign unique IDs to each target and track their movement, and (3) defining a region of interest area for precise classification and counting of dragon fruit across categories. Experimental results demonstrate recognition accuracies of 94.1%, 94.8%, and 96.1% for dragon fruit flowers, green fruits, and red fruits, respectively, with an overall average recognition accuracy of 95.0%. Furthermore, the counting accuracy for each category is measured at 97.68%, 93.97%, and 91.89%, respectively. The proposed method achieves a counting speed of 56 frames per second on a 1080ti GPU. The findings establish the efficacy and practicality of this method for accurate counting of dragon fruit or other fruit varieties.

Ditylenchus destructor is a plant-parasitic nematode that seriously infests sweet potato crop in China. Thus, fast and accurate detection of D. destructor in soil and plant tissue samples is of great significance. In this study, a real-time recombinase polymerase amplification (RPA) assay was developed for the rapid and accurate detection of D. destructor in various samples. The RPA assay could be easily operated and detected as low as 1/500 individual J4 nematode DNA per reaction in 20 min at 39 °C with high specificity. The assay meets the requirements of rapid detection prior to port quarantine as well as on-site real-time detection and can be applied to detect the parasite in soil and plant samples. The modified gDNA extraction method for a single nematode established in this study significantly reduced the time of detection and improved the applicability of the real-time RPA assay for on-site detection in different environments. The real-time RPA assay to detect D. destructor will be useful for epidemiological investigations in the field as well as for quarantine processes in the sweet potato and potato trade.

Sugarcane is the main industrial crop for sugar production, and its growth status is closely related to fertilizer, water, and light input. Unmanned aerial vehicle (UAV)-based multispectral imagery is widely used for high-throughput phenotyping, since it can rapidly predict crop vigor at field scale. This study focused on the potential of drone multispectral images in predicting canopy nitrogen concentration (CNC) and irrigation levels for sugarcane. An experiment was carried out in a sugarcane field with three irrigation levels and five fertilizer levels. Multispectral images at an altitude of 40 m were acquired during the elongating stage. Partial least square (PLS), backpropagation neural network (BPNN), and extreme learning machine (ELM) were adopted to establish CNC prediction models based on various combinations of band reflectance and vegetation indices. The simple ratio pigment index (SRPI), normalized pigment chlorophyll index (NPCI), and normalized green-blue difference index (NGBDI) were selected as model inputs due to their higher grey relational degree with the CNC and lower correlation between one another. The PLS model based on the five-band reflectance and the three vegetation indices achieved the best accuracy (Rv = 0.79, RMSEv = 0.11). Support vector machine (SVM) and BPNN were then used to classify the irrigation levels based on five spectral features which had high correlations with irrigation levels. SVM reached a higher accuracy of 80.6%. The results of this study demonstrated that high resolution multispectral images could provide effective information for CNC prediction and water irrigation level recognition for sugarcane crop.

Lithium-rich manganese-based layered cathode materials are considered to be one of the best options for next-generation lithium-ion batteries, owing to their ultra-high specific capacity (>250 mAh·g−1) and platform voltage. However, their poor cycling stability, caused by the release of lattice oxygen as well as the electrode/electrolyte side reactions accompanying complex phase transformation, makes it difficult to use this material in practical applications. In this work, we suggest a molybdenum surface modification strategy to improve the electrochemical performance of Li1.2Mn0.54Ni0.13Co0.13O2. The Mo-modified Li1.2Mn0.54Ni0.13Co0.13O2 material exhibits an enhanced discharge specific capacity of up to 290.5 mAh·g−1 (20 mA·g−1) and a capacity retention rate of 82% (300 cycles at 200 mA·g−1), compared with 261.2 mAh·g−1 and a 70% retention rate for the material without Mo modification. The significantly enhanced performance of the modified material can be ascribed to the formation of a Mo-compound-involved nanolayer on the surface of the materials, which effectively lessens the electrolyte corrosion of the cathode, as well as the activation of Mo6+ towards Ni2+/Ni4+ redox couples and the pre-activation of a Mo compound. This study offers a facile and effective strategy to address the poor cyclability of lithium-rich manganese-based layered cathode materials.

When organic electron donor (D) and acceptor (A) chromophores are linked together, an electron transfer (ET) state can take place. When a short bridge such as one Sigma bond is used to link the donor and the acceptor, complete charge separation is difficult to access and one usually observes an intramolecular charge transfer (CT) state instead. Due to the inevitable coupling between the donor and the acceptor in compact organic chromophores, the most common decay pathway for the CT state is charge recombination, which may lead to a distinct longer wavelength fluorescence emission or non-radiative dissipation of the excited state energy. However, recent studies have shown that unique excited state dynamics can be observed when the CT state is involved during both forward and backward intersystem crossing (ISC) from singlet excited states to triplet excited states in organic chromophores. Analysis of the mechanism for ISC involving the CT state has received much attention over the last decade. In this perspective, we present a collection of molecular design rationales, spectroscopy and theoretical investigations that provide insights into the mechanism of the ISC involving the CT state in compact organic chromophores. We hope that this perspective will prove beneficial for researchers to design novel compact organic chromophores with a predictable ISC property for future biochemical and optoelectronic applications.

OBJECTIVE: We aimed to evaluate masticatory dysfunction after two different types of ablation on the Gasserian ganglion for the treatment of idiopathic trigeminal neuralgia. We hypothesized that low-temperature plasma radiofrequency ablation (LTP-RFA) was noninferior to radiofrequency thermocoagulation (RFT) with respect to initial efficacy. METHODS: In the randomized, single-blind, parallel-group, noninferiority trial, 204 participants with idiopathic trigeminal neuralgia were randomly allocated to receive plasma ablation in the LTP-RFA group and radiofrequency ablation in the RFT group in a 1:1 ratio, with random block sizes of four or six. Participants were examined at baseline (T0), on the day of discharge (T1), and at the 6-month follow-up (T2). The primary end point was the clinincal effective rate in the LTP-RFA group compared with that in the RFT group after intervention on the day of discharge. Noninferiority was prespecified at -10%. RESULTS: The intention-to-treat analysis revealed that the initial efficacy rates were 91.2% in LTP-RFA group and 93.1% in RFT group (rate ratio [RR] = 0.979, 95% confidence interval [CI]: 0.904-1.061, P = 0.795). The difference between the two groups was 1.9% (95% CI: -5.6% to 9.4%), which showed that LTP-RFA demonstrated noninferiority compared with RFT in initial efficacy. Compared with the RFT group, the LTP-RFA group exhibited a significantly greater improvement in the maximum voltage of the masseter muscles with mean differences of 11.40 (95% CI: 10.52 to 12.27, P < 0.001) at T1 and 17.41 (95% CI: 14.68 to 20.13, P < 0.001) at T2, respectively. Similar results were observed for the asymmetry index of occlusion, the maximum voltage of the anterior temporalis, and the activity index of anterior temporalis / masseter muscles. No serious adverse events were observed in either group. CONCLUSIONS: Compared with the RFT group, noninferior efficacy for pain relief and improvement of masticatory function was revealed in the LTP-RFA group.

RESUMO

Computational ghost imaging is difficult to apply under low sampling rate. We propose high-speed computational ghost imaging based on an auto-encoder network to reconstruct images with high quality under low sampling rate. The auto-encoder convolutional neural network is designed, and the object images can be reconstructed accurately without labeled images. Experimental results show that our method can greatly improve the peak signal-to-noise ratio and structural similarity of the test samples, which are up to 18 and 0.7, respectively, under low sampling rate. Our method only needs 1/10 of traditional deep learning samples to achieve fast and high-quality image reconstruction, and the network also has a certain generalization to the gray-scale images.

In this paper, a novel method, to the best of our knowledge, of structured light fields based on point cloud adaptive repair is proposed to realize 3D reconstruction for highly reflective surfaces. We have designed and built a focused light field camera whose spatial and angular resolution can be flexibly adjusted as required. Then the subaperture image extraction algorithm based on image mosaic is deduced and presented to obtain multidirectional images. After that, the 3D reconstruction of structured light field imaging based on point cloud adaptive repair is presented to accurately reconstruct for highly reflective surfaces. In addition, a method based on smoothness and repair rate is also proposed to objectively evaluate the performance of the 3D reconstruction. Experimental results demonstrate the validity of the proposed method to perform high-quality depth reconstruction for highly reflective surfaces. Generally, our method takes advantage of the multidirectional imaging of the light field camera and can ensure good modulation effect of structured light while avoiding hardware complexity, which makes it application more convenient.

In this paper, a novel, to the best of our knowledge, iterative approach for highlight removal is proposed using lenselet-based plenoptic cameras without multiple exposures. An unsupervised k-means clustering approach that relates unsaturated pixels to chromatic dispersion based on the intrinsic decomposition and dichromatic reflection model is proposed to recover unsaturated highlights. Meanwhile, an adaptive direction method along with a Gaussian probability distribution model is designed to recover the saturated highlights. Finally, a method that combines the specular residual ratio with information entropy is built to quantitatively evaluate the quality of highlight removal. Generally, our method not only fully removes specular highlights, but also has low spatial complexity of image acquisition, more stability, and outstanding restoration for complex scenes.

OBJECTIVE: To evaluate risk factors associated with recurrence after radiofrequency thermocoagulation (RFT) of the gasserian ganglion among patients with ophthalmic trigeminal neuralgia (TN) and prognostic factors in terms of recurrence-free survival (RFS) during a long-term follow-up. METHODS: From January 2005 to December 2017, 300 patients with ophthalmic TN underwent RFT. A retrospective analysis of 14-year outcomes was performed. Kaplan-Meier analysis was used for RFS after the procedure. Univariate and multivariate Cox regression analyses were performed to identify risk factors for pain recurrence. RESULTS: The initial effective rate of RFT for ophthalmic TN was 92%. The mean follow-up time was 77.38 ± 43.24 months. The cumulative probability of RFS was 86.94% at 1 year, 80.03% at 2 years, 77.27% at 3 years, 74.01% at 5 years, and 59.92% at 10 years after RFT. The mean duration of RFS was 114.67 months (95% confidence interval [CI] 106.27 to 123.06 months). In multivariate analysis, atypical pain (hazard ratio [HR] = 2.831, 95% CI 1.759 to 4.554, P < 0.001) and mild facial hypesthesia (HR = 2.540, 95% CI 1.309 to 4.931, P = 0.006) before RFT were independently associated with pain recurrence. Patients with a prognostic index (PI) > 1.27 were at high risk for pain recurrence. Major complications included troublesome dysesthesia (0.7%), keratitis (10.9%), diplopia (0.4%), facial paresthesia (6.2%), and masseter weakness (12.7%). Masseter weakness was more common in patients with V3 branch involvement. Three patients lost their sight due to keratitis. CONCLUSION: Our study investigated long-term outcomes and complications of RFT for ophthalmic TN. Patients at high risk for pain recurrence were identified, which might provide a basis for clinical decision making before RFT.

The present study aimed to investigate the effects of high-mobility group box-1 (HMGB-1) on mechanical pain hypersensitivity and the underlying mechanism. Mouse primary astrocytes were isolated and treated as specified. A CCK-8 assay was used to determine cytotoxicity and a gap junctional communication assay was performed. Ethidium bromide (EtBr) uptake was used to evaluate the hemichannel activity of primary astrocytes. A mouse model of neuropathic pain was developed and paw withdrawal threshold was used to evaluate hind paw sensitivity. RT-qPCR and Western blot were used to determine mRNA and protein expression of genes, respectively. ELISA was used to measure the release of inflammatory cytokines. Treatment with HMGB-1 increased the expression of both toll-like receptor-4 (TLR-4) and connexin 43 (Cx43) in mouse primary astrocytes. HMGB-1 also promoted gap junctional intercellular communication and hemichannel function. Our results also demonstrated that HMGB-1-regulated Cx43 through the JNK signaling pathway, and Cx43 was involved in HMGB-1-mediated inflammation in astrocytes. In vivo analysis supported the idea that HMGB-1-induced mechanical hypersensitivity was associated with Cx43. We therefore conclude that HMGB-1-induced mechanical pain hypersensitivity occurs through modulating astrocytic Cx43 via the TLR-4/JNK signaling pathway.

The objective of this study was to identify molecular responses in Ditylenchus destructor to cold and desiccation by means of transcriptomes analyses. A total of 102,517 unigenes were obtained, with an average length of 1,076 bp, in which 58,453 (57%) had a functional annotation. A total of 1154 simple sequence repeats (SSRs) distributed over 1078 unigenes were detected. Gene expression profiles in response to cold and desiccation stress and the expression of specific stress-related genes were compared. Gene ontology analysis and pathway-based analysis were used to further investigate the functions of the differentially expressed genes. The reliability of the sequencing data was verified through quantitative real-time PCR analysis of 19 stress-related genes. RNA interference used to further assess the functions of the cold-related unigenes 15628 and 15596 showed that the knockdown of each of these genes led to decreased cold tolerance of D. destructor. Hence, this study revealed molecular processes and pathways active in cold- or dessication-treated nematodes. The transcriptome profiles presented in this study provide insight into the transcriptome complexity and will contribute to further understand stress tolerance in D. destructor.

Kennedy's disease, resulted from the expansion of a CAG repeat in exon 1 of androgen receptor (AR) gene, is a motor neuron degenerative disease in the brainstem and spinal cord with the slow development of facial, bulbar, and limb muscle degeneration. To investigate the clinical manifestations and gene mutations in Han Chinese patients with Kennedy's disease. The clinical manifestations of 5 male Han Chinese patients including 2 probands and their relatives from 2 families and 1 sporadic case were retrospectively studied. The CAG repeats in the first exon of AR were screened in 5 Han Chinese people including 2 probands and their healthy relatives from 2 families and 1 sporadic case by polymerase chain reaction (PCR) and direct sequencing. The average age at onset of Kennedy's disease was 48.20 ± 8.70 (mean ± SD) years and the average duration was 7.60 ± 5.32 years. All the patients showed slow onset and progressive weakness, wasting, and fasciculations of the whole body. Four patients demonstrated decreased fertility and 1 patient showed mild gynecomastia. Serum creatine kinase and testosterone levels were elevated mildly in 2 and 1 patients, respectively. The electromyogram showed neurogenic abnormalities. Muscle magnetic resonance demonstrated reduced muscle volume and fatty infiltration. Three different enlarged CAG domains were discovered in the 2 families and 1 sporadic patient with Kennedy's disease, and the CAG repeat number was 48, 43, and 44, respectively. The clinical manifestations of Kennedy's disease in Han Chinese middle-aged men were progressive weakness and atrophy in the bulbar and spinal muscles, occasionally demonstrating incomplete androgen insensitivity syndrome. These patients were also characterized with enlarged CAG repeat number in the first exon of AR, indicating that CAG number could be used in the diagnosis of Han Chinese patients with Kennedy's disease.
