RESUMO

The isotopic composition of iron, zinc, copper, and cadmium (δ56Fe, δ66Zn, δ65Cu, and δ114Cd) are novel and promising tools to study the metabolism and homeostasis of trace metals in the human body. Serum δ65Cu has been proposed as a potential tool for diagnosis of cancer in liquid biopsy, and other metals may have similar utility. However, accurate analysis of trace metal isotopes is challenging because of the difficulties in purifying the metals from biological samples. Here we developed a simple and rapid method for sequential purification of Cu, Fe, Zn, and Cd from a single blood plasma sample. By using a combination of 11 M acetic acid and 4 M HCl in the first steps of column chemistry on AG-MP1 resin, we dramatically improve the separation of Cu from matrix elements compared to previous methods which use concentrated HCl alone. Our new method achieves full recovery of Cu, Fe, Zn, and Cd to prevent column-induced isotope fractionation effects, and effectively separates analytes from the matrix in order to reduce polyatomic interferences during isotope analysis. Our methods were verified by the analysis of isotope standards, a whole blood reference material, and a preliminary sample set including five plasma samples from healthy individuals and five plasma samples from cancer patients. This new method simplifies preparation of blood samples for metal isotope analysis, accelerating multi-isotope approaches to medical studies and contributing to our understanding of the cycling of Fe, Zn, Cu, and Cd in the human body. Atomically-flat surfaces are obtained after thin GaAsSb buffer layer growth on GaAs substrates with regular-distributed nano-holes formed after oxide desorption of the local atomic-force-microscopy anode oxidation. Different from the samples with GaAsSb buffer layers, increasing surface root-mean-square roughness is observed for the GaAs-buffered samples with increasing GaAs buffer layer thickness. The phenomenon is attributed to the enhanced adatom migration resulting from the incorporation of Sb atoms. By using the substrates with nano-holes after buffer layer growth, site-controlled self-assembled InAs quantum dots (QDs) are observed with the deposition of a below-critical-thickness InAs coverage of 1.3 monolayer (ML).

Little is known about temporal changes in sedimentary organic matter (OM) in the East China Sea (ECS) due to the lack of continuous high-resolution records. In this study, we used a multi-proxy approach to reconstruct sedimentary OM variations over the past 100years from three sediment cores collected from the coastal areas of the ECS. For the three cores, total organic carbon (TOC) normalized concentrations of phytoplankton biomarkers (brassicasterol+dinosterol+alkenones) and a marine archaea biomarker (crenarchaeol) showed gradual increases over the past 100years with a greater increase after the 1980s. These increases are attributed to eutrophic conditions caused by monsoonal variation and intensified human activities in the Yangtze River Basin (YRB) over the past century. Conversely, TOC-normalized concentrations of higher plant biomarker long-chain n-alkanes (∑(C27+C29+C31n-alkanes)) showed no obvious trend over the past 100years. The δ13C values of TOC (δ13CTOC), the terrestrial and marine biomarker ratio (TMBR) index, and the branched and isoprenoid tetraether (BIT) index for the three cores record a gradual decrease in the contribution of terrestrial organic matter (TOM) since the 1960s. This decrease is likely linked to enhanced dam construction in the YRB from the 1960s onwards, leading to a reduction in TOM input to the ECS. A three end-member mixing model based on δ13CTOC values and the TMBR index was used to further divide sedimentary OM into three factions: marine organic matter (MOM), plant OM, and soil OM. The MOM contribution gradually increased over the past 100years, while the contribution of soil OM gradually decreased over the past 100years with a significant decline during the past 50years. This trend can also be related to intensify dam construction in the middle and upper reaches of the YR since the 1960s.
