RESUMO

Cryptosporidiosis is one of the main causes of diarrhea in children and young livestock. The interaction of the parasite with the intestinal host cells has not been characterized thoroughly yet but may be affected by the nutritional demand of the parasite. Hence, we aimed to investigate the impact of C. parvum infection on glucose metabolism in neonatal calves. Therefore, N = 5 neonatal calves were infected with C. parvum on the first day of life, whereas a control group was not (N = 5). The calves were monitored clinically for one week, and glucose absorption, turnover and oxidation were assessed using stable isotope labelled glucose. The transepithelial transport of glucose was measured using the Ussing chamber technique. Glucose transporters were quantified on gene and protein expression level using RT-qPCR and Western blot in the jejunum epithelium and brush border membrane preparations. Plasma glucose concentration and oral glucose absorption were decreased despite an increased electrogenic phlorizin sensitive transepithelial transport of glucose in infected calves. No difference in the gene or protein abundance of glucose transporters, but an enrichment of glucose transporter 2 in the brush border was observed in the infected calves. Furthermore, the mRNA for enzymes of the glycolysis pathway was increased indicating enhanced glucose oxidation in the infected gut. In summary, C. parvum infection modulates intestinal epithelial glucose absorption and metabolism. We assume that the metabolic competition of the parasite for glucose causes the host cells to upregulate their uptake mechanisms and metabolic machinery to compensate for the energy losses.

Vitamin D3 has an integral part in calcium and phosphorus homoeostasis, which in turn plays a key role in egg production of hens. The present study aimed to investigate whether an additional vitamin D3 supplementation improves the laying performance and egg quality of hens according to their genetic potential. For this purpose, four layer lines (low performing: R11 and L68; high performing: WLA and BLA) supplemented either with 300 or 3000 IU vitamin D3 per kg feed were compared concerning serum 25-hydroxyvitamin D3 (25-OHD3), calcium, phosphorus and alkaline phosphatase (ALP), laying performance and egg quality. The higher supplementation of vitamin D3 increased 25-OHD3 serum concentrations in all genotypes, except for R11 and WLA hens in week 49, and also elevated vitamin D3 and 25-OHD3 content in the egg yolk (p < 0.05). In week 29, 3000 IU vitamin D3 decreased pooled least squares means (LSMeans) of serum calcium concentrations considering all genotypes and increased the ALP concentrations in BLA hens (p < 0.05). Considering the whole experimental period daily egg mass of R11 hens was increased by an additional vitamin D3 supplementation (p < 0.001). Regarding all genotypes and the whole experimental period the pooled LSMeans of breaking strength of eggs from hens fed 3000 IU vitamin D3 were higher than those of hens fed 300 IU (p = 0.044). In conclusion, present results give evidence that the higher vitamin D3 supplementation might have genotype-dependently beneficial effects on calcium and phosphorus homoeostasis of hens, which might improve feed efficiency in the early laying period and promote the persistence of the laying period irrespectively of genotype. The increase of serum 25-OHD3 by the higher vitamin D supplementation supported the higher transfer of vitamin D in the egg yolk and improved genotype-dependently the breaking strength of the eggshell.

Although commercial broiler feed is usually differently conditioned before pelleting, the nutritional consequences of fine grinding or expanding as pre-pelleting treatments are poorly defined so far. Therefore, the effects of these two pre-treatments on nutrient digestibility, broiler performance and digestive tract of broilers were tested. In order to investigate possible interactions between pre-treatments and diet composition two diets differing in rapeseed expeller proportion were tested in a two by two factorial design. Thus, four diets were designed including two diets containing 6% rapeseed expeller (RSE) which were pre-treated by fine grinding (6%FgP) or expanding (6%ExP), and two corresponding diets containing 12% RSE (12%FgP and 12%ExP). For the experiments, 864 male broilers were used. There was a significant diet-by-technical feed treatment (TFT) interaction in case of the digestibility of all considered crude nutrients (p < 0.05). Diet 6%ExP showed higher crude protein digestibility compared to other feeds (p < 0.001). The highest digestibility of organic matter, ether extract, crude fibre and N-free extractives achieved diet 12%FgP. Diets 6%ExP and 12%FgP showed higher N-corrected metabolisable energy content (p < 0.001). TFT affected daily feed intake (DFI) and body weight (BW) gain in a diet-dependent manner (p < 0.001). Feeding of 6%FgP enhanced DFI and BW gain compared to other feeds but 6%ExP reduced both parameters (p < 0.001). Weights of proventriculi and gizzards of animals fed 6%ExP were increased compared with 6%FgP (p < 0.01). In contrast, proventricular length in animals fed 6%FgP was increased compared with diet 6%ExP (p = 0.042). Moreover, animals fed 6%FgP had wider proventriculi than animals fed 12%FgP (p = 0.023). Feed 6%ExP increased proventricular weight compared to 12%ExP (p = 0.001). With regard to the strong relationships between diet and TFT no specific processing method can be recommended according to considered nutritional aspects. A marked prevention of proventricular dilatation due to pellet feeding could not be realised by various used TFT or feed formulations. Used amounts of RSE had no obvious adverse effects on considered nutritional aspects.

The aim of the experiment on 100 cross-bred barrows was to compare commercial diets for fattening pigs based on either soya bean meal (SBM) imported from non-European countries with diets based on a mixture of locally produced rape seed meal, distillers' dried grains with solubles and soya beans as main protein sources. In addition, these both types of diets were processed by two different technical feed treatments, i.e. coarse grinding without hydrothermal treatment or fine grinding and pelleting. With only few exceptions, nutrients of the diet without SBM were more digestible (p < 0.05) resulting in a higher metabolisable energy (ME) content. Fine grinding and pelleting increased also the ME content and the nutrient digestibility with the exception of crude fibre. Higher feed intake of animals that fed diets without SBM (p < 0.01) resulted in higher average daily gain (p < 0.01). However feeding this diet, the higher digestibility was not reflected in a decreased feed-to-gain ratio (FGR), but fine grinding and pelleting reduced FGR (p < 0.001). A higher pH value and a lower DM content of caecal chymus were detected in animals that received coarsely ground feed (p < 0.05). Animals that fed finely ground and pelleted feed had higher slaughter and relative liver weights and higher blood cholesterol concentrations (p = 0.040). The urea concentrations of blood were lower (p = 0.019) after feeding diets without SBM. In conclusion, SBM imported from non-European countries can be replaced by alternative local protein sources without compromising digestibility or performances of animals. Although fine grinding and thermal treatment particularly seemed to be advantageous for digestibility and performance, the possible risk of development of stomach lesions should be considered.

Effects of grinding and hydro-thermal treatment of feed on growth performance, slaughter traits, nutrient digestibility, stomach content and stomach health were examined by using 96 crossbred fattening pigs. Pigs were fed a grain-soybean meal-based diet processed by various technical treatments. Feeding groups differed in particle size after grinding (finely vs. coarsely ground feed) and hydro-thermal treatment (without hydro-thermal treatment, pelleting, expanding, expanding and pelleting). Fine grinding and hydro-thermal treatment showed significant improvements on the digestibility of crude nutrients and content of metabolisable energy. Hydro-thermal treatment influenced average daily gain (ADG) and average daily feed intake (DFI) significantly. Finely ground pelleted feed without expanding enhanced performances by increasing ADG and decreasing feed-to-gain ratio (FGR) of fattening pigs. Coarsely ground feed without hydro-thermal treatment resulted in the highest ADG and DFI, however also in the highest FGR. Expanded feed decreased DFI and ADG. Slaughter traits were not affected by treatments. Coarsely ground feed without hydro-thermal treatment had protective effects on the health of gastric pars nonglandularis, however, pelleting increased gastric lesions. Hydro-thermal treatment, especially expanding, resulted in clumping of stomach content which possibly induced satiety by slower ingesta passage rate and thus decreased feed intake. Pigs fed pelleted feed showed less pronounced development of clumps in stomach content compared with expanded feed.

One of the leading causes of infectious diarrhea in newborn calves is the apicomplexan protozoan Cryptosporidium parvum (C. parvum). However, little is known about its immunopathogenesis. Using next generation sequencing, this study investigated the immune transcriptional response to C. parvum infection in neonatal calves. Neonatal male Holstein-Friesian calves were either orally infected (N = 5) or not (CTRL group, N = 5) with C. parvum oocysts (gp60 subtype IIaA15G2R1) at day 1 of life and slaughtered on day 7 after infection. Total RNA was extracted from the jejunal mucosa for short read. Differentially expressed genes (DEGs) between infected and CTRL groups were assessed using DESeq2 at a false discovery rate < 0.05. Infection did not affect plasma immunohematological parameters, including neutrophil, lymphocyte, monocyte, leucocyte, thrombocyte, and erythrocyte counts as well as hematocrit and hemoglobin concentration on day 7 post infection. The immune-related DEGs were selected according to the UniProt immune system process database and were used for gene ontology (GO) and pathway enrichment analysis using Cytoscape (v3.9.1). Based on GO analysis, DEGs annotated to mucosal immunity, recognizing and presenting antigens, chemotaxis of neutrophils, eosinophils, natural killer cells, B and T cells mediated by signaling pathways including toll like receptors, interleukins, tumor necrosis factor, T cell receptor, and NF-KB were upregulated, while markers of macrophages chemotaxis and cytosolic pattern recognition were downregulated. This study provides a holistic snapshot of immune-related pathways induced by C. parvum in calves, including novel and detailed feedback and feedforward regulatory mechanisms establishing the crosstalk between innate and adaptive immune response in neonate calves, which could be utilized further to develop new therapeutic strategies.

The aim of the present study was to determine if colostrum and the equipment for harvesting and feeding colostrum are sources of fecal ESBL/AmpC-producing Escherichia coli (ESBL/AmpC-E. coli) in calves. Therefore, 15 male calves fed with pooled colostrum on a dairy farm and held individually in an experimental barn, the colostrum pool and the equipment for harvesting and feeding colostrum were sampled and analyzed for the occurrence of ESBL/AmpC-E. coli. The ESBL-AmpC-E. coli suspicious isolates were subjected to whole-genome sequence analysis. Forty-three of 45 fecal samples were tested positive for ESBL/AmpC-E. coli. In the colostrum sample and in the milking pot, we also found ESBL/AmpC-E. coli. All 45 E. coli isolates were ESBL-producers, mainly commensal sequence type (ST) 10, but also human-extraintestinal pathogenic E. coli ST131 and ST117 were found. The clonal identity of six fecal isolates with the ESBL-E. coli isolate from the colostrum and of five fecal isolates with the strain from the milking pot demonstrates that the hygiene of colostrum or the colostrum equipment can play a significant role in the spread of ESBL-E. coli. Effective sanitation procedures for colostrum harvesting and feeding equipment are crucial to reduce the ESBL-E. coli shedding of neonatal dairy calves.

Fistulation is a helpful procedure in animal nutritional research and also common practise in human medicine. However, there are indications that alterations in the upper gastrointestinal tract contribute to intestinal immune modulations. The present study aimed to investigate effects of a rumen cannulation in week 3 of life on the intestinal and tissue specific immune system of 34-week old heifers. Nutrition influences the development of the neonatal intestinal immune system to a high extent. Therefore, rumen cannulation was investigated in combination with different pre-weaning milk feeding intensities (20% (20MR) vs. 10% milk replacer feeding (10MR). Heifers of 20MR without rumen cannula (NRC) showed higher cluster of differentiation (CD)8+ T cell subsets in mesenteric lymph nodes (MSL) compared to heifers with rumen cannula (RC) and 10MRNRC heifers. CD4+ T cell subsets in jejunal intraepithelial lymphocytes (IELs) were higher in 10MRNRC heifers compared to 10MRRC heifers. CD4+ T cell subsets in ileal IELs were lower and CD21+ B cell subsets were higher in NRC heifers compared to RC heifers. CD8+ T cell subsets in spleen tended to be lower in 20MRNRC heifers compared to all other groups. Splenic CD21+ B cell subsets were higher in 20MRNRC heifers compared to RC heifers. Splenic toll like receptor (TLR) 6 expression was increased and IL4 expression tended to be increased in RC heifers than NRC heifers. Splenic TLR2, 3 and 10 gene expression was higher in 20MR compared to 10MR heifers. Jejunal prostaglandin endoperoxide synthase 2 expression was higher in RC heifers than NRC heifers, and MUC2 expression tended to increase in 20MR heifers compared to 10MR heifers. In conclusion, rumen cannulation modulated T and B cell subsets in the down streaming gastrointestinal tract and spleen. Pre-weaning feeding intensity seemed to affect intestinal mucin secretion and T and B cell subsets in MSL, spleen and thymus until several month later. Interestingly, in MSL, spleen and thymus the 10MR feeding regime evoked similar modulations of T and B cell subsets like rumen cannulation.

Conjugated linoleic acids (CLAs) modulate the fatty acid composition in dairy cow milk, which represents the most important nutrient source of neonatal calves. In turn, dietary fatty acids are known to influence the gut microbiota. The current preliminary study investigated effects of a maternal fatty acid supplementation (MFAS) during transition period with coconut oil (CON, control), CLA (Lutalin®), or CLA + EFA (Lutalin® + essential fatty acids-linseed oil; safflower oil) on physico-chemical characteristics of jejunal content and microbiota of 5-day-old calves. MFAS of CLA + EFA increased α-linolenic, eicosapentaenoic, docosapentaenoic, and n-3 fatty acid proportions in jejunum compared to the other groups (P < 0.05). Proportions of n-6 and polyunsaturated fatty acids increased by MFAS of CLA + EFA compared to CON (P < 0.05). Most abundant phyla in the jejunum were Proteobacteria, Firmicutes, and Bacteroidota. CLA + EFA decreased the relative abundance of Diplorickettsiales (Proteobacteria) compared to CON and CLA (P < 0.05). CLA calves showed a lower abundance of Enterobacterales (Proteobacteria) compared to CON calves (P = 0.001). The abundance of Veillonellales-Selenomonadales and RF39 (Firmicutes) decreased in CLA + EFA calves compared to CON calves (P < 0.05). Bacteroidales (Bacteroidota) decreased in CLA + EFA calves compared to CLA calves (P < 0.05). The relative abundance of Cyanobacteria and Euryarchaeota decreased and the abundance of Chloroflexi increased in CLA + EFA calves compared to CON and CLA calves (P < 0.05). MFAS alters the fatty acid composition and microbial milieu in the intestinal content of neonatal calves due to their ability to modulate colostral fatty acid composition of dams.

Mycotoxin contaminated feed has been associated with colic of horses caused by intestinal disorders. Whether such disease conditions alter the intestinal toxin metabolism and transfer across a compromised mucosal barrier is unknown. A screening approach was used to relate blood residue levels of DON, ZEN and their metabolites to the status of the horses (sick vs. healthy). A total of 55 clinically healthy horses from 6 different farms with varying feeding background served as control for sick horses (N = 102) hospitalized due to colic. ZEN, alpha-zearalenol (ZEL), beta-ZEL and DON were detectable in peripheral blood as indicators for the inner exposure with significant farm effects for alpha- and beta-ZEL. However, the levels in sick horses were similar to all farms. Moreover, the proportion of beta-ZEL of all detected ZEN metabolites as an indicator for the degree of metabolism of ZEN was not different for sick horses but differed amongst the control farms. Although the incidence of DON in blood was generally low and not significantly different amongst healthy and sick horses, the positive samples were nearly exclusively found in sick horses suggesting either a higher toxin transfer, an association of DON with the development of colic or a different feeding background.

Fatty acids are known for their regulatory role in inflammation and oxidative stress. The present study investigated 38 calves born from dams, abomasally supplemented with coconut oil, essential fatty acids (EFA), conjugated linoleic acid (CLA) or EFA + CLA, according to immunological traits and the oxidative and anti-oxidative status for the first 5 days of life. On day 2 of life, plasma total bilirubin, cholesterol, interleukin 1-ß and ferric ion reducing anti-oxygen power (FRAP) were lower in calves with than without maternal EFA supplementation, and FRAP additionally on day 4. On day 3, the concentrations of reactive oxygen metabolites were higher in calves with than without maternal EFA supplementation and additionally on day 5 together of retinol. Total leucocyte counts were decreased in the EFA group compared to the CLA group on day 5. Lymphocyte proportions decreased from day 1 to 5 only in the EFA + CLA group. On day 2, plasma total protein was higher in CLA and EFA + CLA than in EFA calves. Similarly, CLA calves had higher interleukin 1-ß concentrations compared to EFA + CLA calves. FRAP was decreased by CLA on day 4. Overall, the maternal fatty acid supply affected the inflammatory response and the oxidative and anti-oxidative status of the neonatal offspring.

Pelleted feed is associated with improved broiler performance but also with a higher incidence of proventricular dilatation and ascites. The present study aimed to investigate influences of expanded and pelleted (ExP) or finely ground and pelleted feeds (FgP) containing either 6% rapeseed expeller (RSE) or 12% RSE on these adverse effects by studying performance, visceral organ, and immunological traits in 36 broilers. ExP reduced daily feed intake compared to FgP when feeding a 6% RSE diet (P < 0.05) but did not affect the daily feed intake when feeding a 12% RSE diet, which was also reflected in the body weight gain. There were no significant differences in the size of proventriculus and gizzard between feeding groups but significant diet-by-technical feed treatment interactions in case of proventricular and gizzard weights and the proventricular length (P < 0.05). Proventriculi and gizzards were heavier in birds fed 6%ExP than proventriculi or gizzards of animals from all other groups except for birds of the group 12%FgP. A total of three animals (1 from 6%ExP, 1 from 6%FgP, and 1 from 12%ExP) developed ascites during the study. Pooled LsMeans of peripheral blood leucocyte proportions of CD3+/CD4-/CD8- cells were increased in birds fed FgP compared to birds fed ExP (P = 0.048). Pooled LsMeans of CD3+/CD4+/CD8+ T cell subsets in jejunal lamina propria were higher in birds fed 12% RSE compared to birds fed 6% RSE (P = 0.024). Concluding, technical feed treatment or diet did not inhibit adverse effects of pelleting on gizzard and proventricular development. Morphometric alterations of proventriculus and gizzard might modify the local immune system of the distal digestive tract and promote the development of ascites; however, further studies are required to confirm this hypothesis since in the present study only three birds developed ascites.

Conjugated linoleic acid (CLA) isomers are known for their health-promoting effects in mammals and metabolic functions in dairy cows and are synthesized in the forestomach depending on essential fatty acid (EFA) intake. The current preliminary study investigated effects of a maternal fatty acid supplementation (MFAS) during late pregnancy and early lactation with coconut oil (CON, control), CLA (Lutalin®), or CLA + EFA (Lutalin® linseed oil; safflower oil) on plasma fatty acid composition and T and B cell subsets in mesenteric lymph nodes (MLN) and the small intestine of 5-day-old calves. MFAS of CLA + EFA increased α-linolenic, eicosapentaenoic, docosapentaenoic, and n-3 fatty acid proportions in calf plasma fat on days 1 and 5 after birth (P < 0.05). On day 5, CLA and CLA + EFA calves showed higher plasma fat trans-10, cis-12 CLA proportions, and CLA calves had higher plasma cis-9, trans-11 CLA proportions compared with CON calves (P < 0.1). MFAS of CLA tended to increase CD4+ T cell subsets in MLN and increased CD21+ B cell subsets in ileal lamina propria compared with CON but decreased CD2+ T cell subsets in jejunal lamina propria (P < 0.05). CLA + EFA decreased CD4+ T cell subsets in MLN compared with CLA (P < 0.05). MFAS of CLA seemed to affect the intestinal adaptive immune system of calves, but additional EFA supplementations reversed CLA effects. Possible direct CLA and EFA effects or whether changes in milk composition affected this immune modulation must be clarified in further studies.

The influence of variously processed feedstuffs on the gastrointestinal tract, various blood parameters, and T cell subsets of different localizations was investigated in 36 broilers. Birds were fed a similarly composed feed mixture differing in particle size (coarse vs. fine). Feedstuffs with different particle size distribution were additionally hydrothermally treated (HTT) (without-non-compacted, pelleted, expanded, and pelleted). The HTT affected BW at slaughtering day and feed intake (P < 0.001). Birds fed non-compacted feed consumed less feed and were lighter than animals of other feeding groups irrespective of grinding fineness. Proventricular size was influenced by HTT (P < 0.001) but not the weight. Generally, feeding of pelleted feed increased proventricular size. Gizzard weight and length was affected by particle size and HTT (P < 0.01). Non-compacted feed resulted in heavier but shorter gizzards compared to other feeds except for finely ground expanded and pelleted feed. Organ traits of gizzard and proventriculus were strongly related to the feed intake. The jejunum was heavier in birds fed expanded and pelleted feed compared to animals fed solely pelleted feed (P = 0.030). The intestinal segment weight to length ratio (WL ratio) of duodenum, jejunum, and total small intestine was affected by HTT (P < 0.01). Generally, non-compacted feed resulted in lowest WL ratios regardless of the particle size distribution. Feeding variously processed feedstuffs also affected white blood cells, blood sodium, and chloride concentrations (P < 0.05) and tended to influence blood carbon dioxide partial pressure and blood total carbon dioxide levels (P < 0.1) in venous blood. Subsets of CD4/CD8 double negative T cells of caecal tonsils were influenced by HTT (P < 0.043). Non-compacted feed tended to increase these subsets in this localization compared to other feeds. In conclusion, the present study shows that an increased feed intake provoked by feed compaction evoke proventricular and ventricular alterations. Furthermore, feeding of variously processed feedstuffs influences the acid base homeostasis and the local immune system of caecal tonsils.

The present study investigated clinical and immunological modulations due to intramuscular injection of Escherichia coli LPS in 49-wk-old laying hens over 48 h post injection (p.i.). LPS induced characteristic sickness behavior but no significant body temperature alterations ( P > 0.05). During experimental period decreases in blood albumin, calcium, phosphorus and tryptophan concentrations, hyperglycemia, increased plasma nitrite concentrations, leucopenia, decreased thrombocyte counts, lymphopenia, heterophilia and an increased heterophilic granulocyte/lymphocyte (H/L) ratio were observed after LPS administration. Time-dependent effects were shown on T and B cell subsets in caecal tonsils (CT) and on splenic CD3+/CD4+/CD8+ proportions, on IL-1ß and -10 and inducible NO synthase mRNA expression in peripheral blood lymphocytes (PBL), liver, spleen and CT, and on the mRNA expression of the TLR4 in PBL, liver and spleen p.i. ( P < 0.05). The main responding period of mentioned alterations due to LPS appears to include the period from 2 until 8 h p.i. According to the H/L ratio, the most stressful phase was 5 h p.i. T and B cell subsets in CT, the IL-1ß and TLR4 mRNA expression in liver and plasma nitrite concentrations seemed to be affected for a longer period.

Influences of feed particle size (coarse, fine) and hydro-thermal processing methods (HTPM) (without-non-compacted feed, pelleting, expanding and pelleting) on feeding value and the performance and digestive tract of 624 broilers were studied. HTPM increased the starch disintegration of feed. Starch disintegration and electron microscopy indicated the highest degree of starch modification in expanded and pelleted feed. HTPM affected ether extract digestibility (p < 0.05). A grinding-by-HTPM interaction was found in case of crude protein digestibility (p = 0.008). Non-compacted feed reduced daily feed intake (DFI) and body weight gain and increased the feed to gain ratio compared to compacted feeds (p < 0.001). Compacted feeds increased proventricular size and the risk of Isthmus gastrici dilatation compared to coarsely ground non-compacted feed, except for finely ground expanded and pelleted feed. Finely ground feed reduced proventricular weights compared to coarsely ground feed and pelleted feed compared to other feeds. Non-compacted feed increased gizzard weights compared to compacted feeds. Relationships between proventricular size and Isthmus gastrici dilatation and the DFI were detected. Summarizing, the beneficial effects of pelleted feed were mainly based on the reduction of feed wastage and selection. However, the high DFI caused by pellet feeding is also a main risk factor for proventricular dilatation.
