RESUMO

NEW FINDINGS: What is the topic of this review? This review focuses on the physiological role of the cytokine interleukin-1ß in the CNS. What advances does it highlight? Traditionally, interleukin-1ß is known as a key mediator of inflammation and immunity. This review highlights the more recent findings describing how interleukin-1ß signalling is required to maintain homeostasis in the CNS. ABSTRACT: Since its discovery in the early 1940s, the interleukin-1 (IL-1) cytokine family has been associated primarily with acute and chronic inflammation. The family member IL-1ß is produced by different leucocytes, endothelial cells and epithelial cells. This cytokine has been characterized as a key modulator of inflammation and innate immunity because it induces the transcription of several downstream inflammatory genes. More recently, several groups have demonstrated that IL-1ß production is also required to maintain homeostasis in several organ systems. This review focuses on providing an overview of the more recently characterized role of IL-1ß in the physiology of the CNS. So far, IL-1ß signalling has been implicated in neuronal survival, neurite growth, synaptic pruning, synaptic transmission, neuroplasticity and neuroendocrine functions.

RESUMO

IL-1ß is a potent pro-inflammatory cytokine that promotes immunity and host defense, and its dysregulation is associated with immune pathology. Toxoplasma gondii infection of myeloid cells triggers the production and release of IL-1ß; however, the mechanisms regulating this pathway, particularly in human immune cells, are incompletely understood. We have identified a novel pathway of T. gondii induction of IL-1ß via a Syk-CARD9-NF-κB signaling axis in primary human peripheral blood monocytes. Syk was rapidly phosphorylated during T. gondii infection of primary monocytes, and inhibiting Syk with the pharmacological inhibitors R406 or entospletinib, or genetic ablation of Syk in THP-1 cells, reduced IL-1ß release. Inhibition of Syk in primary cells or deletion of Syk in THP-1 cells decreased parasite-induced IL-1ß transcripts and the production of pro-IL-1ß. Furthermore, inhibition of PKCδ, CARD9/MALT-1 and IKK reduced p65 phosphorylation and pro-IL-1ß production in T. gondii-infected primary monocytes, and genetic knockout of PKCδ or CARD9 in THP-1 cells also reduced pro-IL-1ß protein levels and IL-1ß release during T. gondii infection, indicating that Syk functions upstream of this NF-κB-dependent signaling pathway for IL-1ß transcriptional activation. IL-1ß release from T. gondii-infected primary human monocytes required the NLRP3-caspase-1 inflammasome, but interestingly, was independent of gasdermin D (GSDMD) cleavage and pyroptosis. Moreover, GSDMD knockout THP-1 cells released comparable amounts of IL-1ß to wild-type THP-1 cells after T. gondii infection. Taken together, our data indicate that T. gondii induces a Syk-CARD9/MALT-1-NF-κB signaling pathway and activation of the NLRP3 inflammasome for the release of IL-1ß in a cell death- and GSDMD-independent manner. This research expands our understanding of the molecular basis for human innate immune regulation of inflammation and host defense during parasite infection.

RESUMO

OBJECTIVE: To present a method to identify critical areas for selected infectious and parasitic diseases for the purpose of health surveillance and to analyze the association between these critical areas and poverty indicators in Brazil. METHOD: The following incidence rates were mapped: dengue, acute Chagas disease, schistosomiasis, Hansen's disease, hepatitis A, cutaneous leishmaniasis, visceral leishmaniasis, leptospirosis, malaria, and tuberculosis. The analyses were performed for the period from 2010 to 2017 based on a synthetic indicator calculated as the mean of mean incidence coefficients for each disorder, normalized by the mean and standard deviation during the period of analysis. A 2014 population estimate was used. The calculated coefficients were stratified for classification of municipalities into very high, high, medium, low, or very low criticality according to each disorder. Indicators expressing several socioeconomic dimensions and space segregation in Brazilian municipalities were also selected and tested regarding their association with the transmission of the diseases under study. RESULTS: The indicator showed that 40.5% of Brazilian municipalities had high criticality for the diseases of interest, especially in the North, parts of the Northeast, and Midwest. Indicators "proportion of poverty," "garbage in surroundings," and "families headed by women" increased the chance of higher criticality for the diseases. The indicator "adequate sewer system" was a potential protection factor. CONCLUSIONS: The technique used was adequate to guide surveillance actions in the country and allows articulation between local surveillance efforts and other sectors to resolve health problems caused by infectious and parasitic diseases and associated factors.

IL-1ß is produced by myeloid cells and acts as a critical mediator of host defense during infection and injury. We found that the intracellular protozoan parasite Toxoplasma gondii induced an early IL-1ß response (within 4 h) in primary human peripheral blood monocytes isolated from healthy donors. This process involved upregulation of IL-1ß, IL-1RN (IL-1R antagonist), and NLRP3 transcripts, de novo protein synthesis, and the release of pro- and mature IL-1ß from infected primary monocytes. The released pro-IL-1ß was cleavable to mature bioactive IL-1ß in the extracellular space by the protease caspase-1. Treatment of primary monocytes with the NLRP3 inhibitor MCC950 or with extracellular potassium significantly reduced IL-1ß cleavage and release in response to T. gondii infection, without affecting the release of TNF-α, and indicated a role for the inflammasome sensor NLRP3 and for potassium efflux in T. gondii-induced IL-1ß production. Interestingly, T. gondii infection did not induce an IL-1ß response in primary human macrophages derived from the same blood donors as the monocytes. Consistent with this finding, NLRP3 was downregulated during the differentiation of monocytes to macrophages and was not induced in macrophages during T. gondii infection. To our knowledge, these findings are the first to identify NLRP3 as an inflammasome sensor for T. gondii in primary human peripheral blood cells and to define an upstream regulator of its activation through the release of intracellular potassium.

RESUMO

Macrophages infected with Gram-negative bacteria expressing Type III secretion system (T3SS) activate the NLRC4 inflammasome, resulting in Gasdermin D (GSDMD)-dependent, but GSDME independent IL-1ß secretion and pyroptosis. Here we examine inflammasome signaling in neutrophils infected with Pseudomonas aeruginosa strain PAO1 that expresses the T3SS effectors ExoS and ExoT. IL-1ß secretion by neutrophils requires the T3SS needle and translocon proteins and GSDMD. In macrophages, PAO1 and mutants lacking ExoS and ExoT (ΔexoST) require NLRC4 for IL-1ß secretion. While IL-1ß release from ΔexoST infected neutrophils is also NLRC4-dependent, infection with PAO1 is instead NLRP3-dependent and driven by the ADP ribosyl transferase activity of ExoS. Genetic and pharmacologic approaches using MCC950 reveal that NLRP3 is also essential for bacterial killing and disease severity in a murine model of P. aeruginosa corneal infection (keratitis). Overall, these findings reveal a function for ExoS ADPRT in regulating inflammasome subtype usage in neutrophils versus macrophages and an unexpected role for NLRP3 in P. aeruginosa keratitis.

RESUMO

BACKGROUND: Percutaneous coronary interventions (PCI) are associated with quality of life (QoL) and health status improvements in stable angina patients. There are few studies assessing the magnitude of this effect and its predictors in contemporary daily practice. METHODS: Prospective cohort study with stable angina patients submitted to PCI in a tertiary interventional cardiology center. The clinical characteristics and the Seattle Angina Questionnaire (SAQ) were assessed before PCI, and patients were followed-up for 1 year. Mixed linear regression and ANOVA were used to compare SAQ indices, and multivariate analysis to identify predictors of QoL improvement. RESULTS: Between September 2006 and May 2007, 110 patients were included. The mean age of the study population was 62.8 ± 8.7 years, and 62% of the patients were of the male gender. Diabetes mellitus was present in 29%, arterial hypertension in 82%, previous myocardial infarction in 32%, and previous PCI in 29%. Before PCI, only 5% of the patients were free of angina, and this rate improved to 68% in the one-year followup (P < 0.001). There was improvement in all SAQ scales in the one-year followup, which was already shown in the 6-month assessment (P < 0.0001). Quality of life before the procedure was the main predictor of QoL improvement by multivariate analysis (P < 0.001). CONCLUSIONS: Patients with stable angina submitted to PCI in the real-world practice present significant improvement in one-year health status, as assessed by the SAQ. Quality of life before the procedure is the main determinant of improvement in QoL.

RESUMO

Toxoplasma gondii is an intracellular protozoan parasite that has the remarkable ability to infect and replicate in neutrophils, immune cells with an arsenal of antimicrobial effector mechanisms. We report that T. gondii infection extends the life span of primary human peripheral blood neutrophils by delaying spontaneous apoptosis, serum starvation-induced apoptosis, and tumor necrosis alpha (TNF-α)-mediated apoptosis. T. gondii blockade of apoptosis was associated with an inhibition of processing and activation of the apoptotic caspases caspase-8 and -3, decreased phosphatidylserine exposure on the plasma membrane, and reduced cell death. We performed a global transcriptome analysis of T. gondii-infected peripheral blood neutrophils using RNA sequencing (RNA-Seq) and identified gene expression changes associated with DNA replication and DNA repair pathways, which in mature neutrophils are indicative of changes in regulators of cell survival. Consistent with the RNA-Seq data, T. gondii infection upregulated transcript and protein expression of PCNA, which is found in the cytosol of human neutrophils, where it functions as a key inhibitor of apoptotic pro-caspases. Infection of neutrophils resulted in increased interaction of PCNA with pro-caspase-3. Inhibition of this interaction with an AlkB homologue 2 PCNA-interacting motif (APIM) peptide reversed the infection-induced delay in cell death. Taken together, these findings indicate a novel strategy by which T. gondii manipulates cell life span in primary human neutrophils, which may allow the parasite to maintain an intracellular replicative niche and avoid immune clearance.IMPORTANCEToxoplasma gondii is an obligate intracellular parasite that can cause life-threatening disease in immunocompromised individuals and in the developing fetus. Interestingly, T. gondii has evolved strategies to successfully manipulate the host immune system to establish a productive infection and evade host defense mechanisms. Although it is well documented that neutrophils are mobilized during acute T. gondii infection and infiltrate the site of infection, these cells can also be actively infected by T. gondii and serve as a replicative niche for the parasite. However, there is a limited understanding of the molecular processes occurring within T. gondii-infected neutrophils. This study reveals that T. gondii extends the life span of human neutrophils by inducing the expression of PCNA, which prevents activation of apoptotic caspases, thus delaying apoptosis. This strategy may allow the parasite to preserve its replicative intracellular niche.

RESUMO

Colletotrichum has been identified as responsible for the "dried fruit" disease in açaí (Euterpe oleracea). Besides concern for açaí pulp quality control, the characterization of Colletotrichum has been difficult, which has motivated the search for chemical markers in the Volatile Organic Compounds (VOCs) profile for use as a tool in the identification of açaí pulp contaminated by the fungus. Extracted VOCs by Headspace - Solid Phase Micro Extraction (HS-SPME) were identified through gas chromatography mass spectrometry (GC-MS). From GC-MS analyzes 26 VOCs were identified, with a predominance of the terpenoids. Chemometrically, menthol and menthone were assigned as potential markers of the genus. The analysis of VOCs in açaí pulps contaminated by Colletotrichum under different cultivation conditions enabled the detection of menthone. This result illustrated the selectivity of the culture medium and the potential of this tool for use in the quality control of açaí pulp.

RESUMO

In this study, 395 volunteers were enrolled to investigate the seroprevalence of hepatitis C virus, the immunological and the alanine aminotransferase (ALT) biomarkers amongst hemodialysis patients, living in Manaus, Brazil. An overall seroprevalence of 13.9% was found in the hemodialysis patients. Analysis of seroconversion patterns demonstrated that most patients with HCV seroconverted up to 10 years following the first hemodialysis session. Anti-NS5 antibody was detectable in 60.4% of patients with HCV. A lower percentage of circulating CD3(+) and CD4(+) T-cells was found in patients seronegative for HCV, whereas a higher frequency of CD8(+) T-cells was the hallmark of patients with HCV. An overall low activation state of monocytes and eosinophils were observed in hemodialysis patients. In contrast, a higher frequency of activated neutrophils was observed in patients with HCV, selectively in the NS5+ subgroup. All hemodialysis patients had a higher percentage of activated lymphocytes, with the higher activation state in patients with NS5- reactivity. Higher ALT levels were observed in patients with HCV, especially in the NS5+ subgroup. Interestingly, the ALT levels were correlated negatively with the lymphocyte activation state, selectively in the NS5- subgroup, suggesting a protective role of these activated lymphocytes in patients with HCV. These findings reinforce the importance of the transmission of HCV among hemodialysis patients, suggesting that apart from the HCV screening, the serological and ALT biomarkers may represent important predictors of morbidity and/or mortality among patients undergoing hemodialysis.

RESUMO

Toxoplasma gondii is an intracellular protozoan parasite of global importance that can remarkably infect, survive, and replicate in nearly all mammalian cells. Notably, 110 years after its discovery, Toxoplasmosis is still a neglected parasitic infection. Although most human infections with T. gondii are mild or asymptomatic, T. gondii infection can result in life-threatening disease in immunocompromised individuals and in the developing fetus due to congenital infection, underscoring the role of the host immune system in controlling the parasite. Recent evidence indicates that T. gondii elicits a robust innate immune response during infection. Interestingly, however, T. gondii has evolved strategies to successfully bypass or manipulate the immune system and establish a life-long infection in infected hosts. In particular, T. gondii manipulates host immunity through the control of host gene transcription and dysregulation of signaling pathways that result in modulation of cell adhesion and migration, secretion of immunoregulatory cytokines, production of microbicidal molecules, and apoptosis. Many of these host-pathogen interactions are governed by parasite effector proteins secreted from the apical secretory organelles, including the rhoptries and dense granules. Here, we review recent findings on mechanisms by which T. gondii evades host innate immunity, with a focus on parasite evasion of the human innate immune system.

RESUMO

Neutrophils are a major player in host immunity to infection; however, the mechanisms by which human neutrophils respond to the intracellular protozoan parasite Toxoplasma gondii are still poorly understood. In the current study, we found that, whereas primary human monocytes produced interleukin-1beta (IL-1ß) in response to T. gondii infection, human neutrophils from the same blood donors did not. Moreover, T. gondii inhibited lipopolysaccharide (LPS)-induced IL-1ß synthesis in human peripheral blood neutrophils. IL-1ß suppression required active parasite invasion, since heat-killed or mycalolide B-treated parasites did not inhibit IL-1ß release. By investigating the mechanisms involved in this process, we found that T. gondii infection of neutrophils treated with LPS resulted in reduced transcript levels of IL-1ß and NLRP3 and reduced protein levels of pro-IL-1ß, mature IL-1ß, and the inflammasome sensor NLRP3. In T. gondii-infected neutrophils stimulated with LPS, the levels of MyD88, TRAF6, IKKα, IKKß, and phosphorylated IKKα/ß were not affected. However, LPS-induced IκBα degradation and p65 phosphorylation were reduced in T. gondii-infected neutrophils, and degradation of IκBα was reversed by treatment with the proteasome inhibitor MG-132. Finally, we observed that T. gondii inhibited the cleavage and activity of caspase-1 in human neutrophils. These results indicate that T. gondii suppression of IL-1ß involves a two-pronged strategy whereby T. gondii inhibits both NF-κB signaling and activation of the NLRP3 inflammasome. These findings represent a novel mechanism of T. gondii evasion of human neutrophil-mediated host defense by targeting the production of IL-1ß.IMPORTANCEToxoplasma gondii is an obligate intracellular parasite that infects approximately one-third of humans worldwide and can invade virtually any nucleated cell in the human body. Although it is well documented that neutrophils infiltrate the site of acute T. gondii infection, there is limited understanding of how human neutrophils respond to T. gondii Neutrophils control infectious pathogens by a variety of mechanisms, including the release of the cytokine IL-1ß, a major driver of inflammation during infection. This study reveals that T. gondii is able to inhibit IL-1ß production in human neutrophils by impairing the activation of the NF-κB signaling pathway and by inhibiting the inflammasome, the protein complex responsible for IL-1ß maturation. This two-pronged strategy of targeting the IL-1ß pathway may facilitate the survival and spread of T. gondii during acute infection.

RESUMO

Patients with diabetes and coronary artery disease are frequently considered for myocardial revascularization procedures, aiming at cardiovascular events risk reduction and a better quality of life. In clinical practice, decisions concerning surgery or percutaneous coronary intervention are frequently difficult, because of cases' severity, disease extension and co-morbidities association. Beyond that, the bulk of literature information was generated by subgroup analysis of randomized clinical trials, which were designed for the general population, not for diabetics. The aim of this study was to review literature on coronary percutaneous intervention in diabetic patients, and also to show recent data from the experience in this procedure at the Catheterization Laboratory of the Cardiology Institute of RS.

RESUMO

Alzheimer's disease considerably compromises communication skills. Language changes become more prominent as the disease progresses. Deterioration of language and cognition reduces the ability of holding conversations, which has a negative impact on social interaction. OBJECTIVE: To conduct a systematic review of the literature for articles reporting interventions focused on the language and communication of people with Alzheimer's disease (AD) without use of medication. METHODS: We performed a search using the keywords Alzheimer's disease, language, communication, cognition, cognitive intervention, rehabilitation and therapy, and their corresponding Portuguese and Spanish terms, on the SciELO, LILACS, PubMed and PsychINFO databases. We analyzed intervention studies published from 1993 to 2016 that involved stimulation of language skills and/or communication with pre-and post-intervention quantitative results, and whose samples included at least 50% with a diagnosis of probable AD. Studies were analyzed and classified into four levels of evidence, according to the criteria described in the literature. RESULTS: Twenty-eight articles were included. The majority of the designs had medium-to-low scientific evidence. Most interventions showed benefits for at least one language or communicative skill. Eight types of interventions emerged from the analysis of the studies. Further research with higher levels of evidence is recommended in the investigation of interventions focused on language and communication skills of patients with dementia. CONCLUSION: Studies with high levels of evidence on the topic investigated are only being conducted on a small scale. Two intervention techniques seem potentially effective: lexical-semantic approaches and interventions that work with different cognitive skills (including language).

Neutrophils have a critical role in the innate immune response; these cells represent the primary line of defense against invading pathogens or tissue injury. Crotoxin (CTX), the major toxin of the South American rattlesnake (Crotalus durissus terrificus) venom, presents longstanding anti-inflammatory properties, inhibiting neutrophil migration and phagocytosis by peritoneal neutrophils for 14 days. Herein, to elucidate these sustained inhibitory effects induced by CTX, we performed in vitro and in vivo studies evaluating the functionality of bone marrow neutrophils and possible molecular mechanisms associated with these effects. CTX inhibited the processes of chemotaxis, adhesion to fibronectin, and phagocytosis of opsonized particles; however, it did not affect ROS production or degranulation in bone marrow neutrophils. To understand the molecular mechanisms that orchestrate this effect, we investigated the expression of CR3 on the neutrophil surface and the total expression and activity of signaling proteins from the Syk-GTPase pathway, which is involved in actin polymerization. CTX down-regulated both subunits of CR3, as well as, the activity of Syk, Vav1, Cdc42, Rac1 and RhoA, and the expression of the subunit 1B from Arp2/3. Together, our findings demonstrated that CTX inhibits the functionally of bone marrow neutrophils and that this effect may be associated with an impairment of the Syk-GTPase pathway. This study demonstrates, for the first time, that the sustained down-modulatory effect of CTX on circulating and peritoneal neutrophils is associated with functional modifications of neutrophils still in the bone marrow, and it also contributes to a better understanding of the anti-inflammatory effect of CTX.

RESUMO

Abstract Introduction: Immature forms of the antlion Myrmeleon brasiliensis (Neuroptera, Myrmeleontidae) build traps in dry sandy soil to capture prey. Objective: The aim of the present study was to investigate how the waterlogging of the soil due to rain affects the trap rebuilding and relocation behavior of M. brasiliensis of different sizes. Methods: The study was conducted between July and December 2019. Larvae M. brasiliensis were observed and collected from a forest reserve in the municipality of Aquidauana in the state of Mato Grosso do Sul, Brazil. Results: In the natural environment, most larvae rebuilt their traps in the same location seven days after the simulation of rain, with a smaller diameter than that observed prior to the simulation of rain. In the laboratory, the movements of M. brasiliensis larvae and rebuilding of the traps after the waterlogging of the soil was affected by body size. Larger larvae moved more and were more likely to rebuild their traps. Conclusions: The saturation of the soil affects the foraging of M. brasiliensis larvae, which are impeded from rebuilding their traps for a period. In situations of long periods of saturated soil, the mortality rate of the larvae is high and rebuilding of the traps occurs after the soil dries out, but with a smaller trap size. These data suggest that changes in the rainfall pattern can affect the population structure of M. brasiliensis larvae, with the selection of larger individuals in situations of more severe rains. In this process, the smaller larvae are more affected, as their foraging is impeded.

Abstract Sedentary behavior tends to increase with aging and several studies have focused on analyzing the components which may influence this pattern of behavior. Indirect and direct measuring have been used to determine the relative parameters to this phenomenon while there is no consensus about which parameter should be adopted to conceptualize sedentary behavior, making difficulty to establish comparisons among the studies on this population. The aim of this study was to systematically review the characterization of low level of physical activity and sedentary behavior in studies with older people. Electronic search on Scielo, LILACS, MEDLINE, PubMed, and ISI Web of Knowledge was carried out. Selection of the studies included: original manuscripts, with elderly analyzing sedentary behavior or low-level pf physical activity through direct and indirect measure. Search initially screened 190 manuscripts yielding 10 relevant studies. Questionnaire, self-reported questionnaire and accelerometer were the instruments used in the studies. Sedentary behavior or low level of physical activity was characterized by analyzing sitting time, physical activity on leisure time, counts per minute, engaging in moderate or vigorous intensity during day or performing physical activities with MET <1.5. There is no standardization of the parameters adopted to characterize sedentary behavior on studies, generating divergent results and making it difficult to establish comparisons.

Pesquisa eletrônica no Scielo, LILACS, MEDLINE, PubMed e ISI Web of Knowledge foi realizada.

ABSTRACT Objective. To present a method to identify critical areas for selected infectious and parasitic diseases for the purpose of health surveillance and to analyze the association between these critical areas and poverty indicators in Brazil. Method. The following incidence rates were mapped: dengue, acute Chagas disease, schistosomiasis, Hansen's disease, hepatitis A, cutaneous leishmaniasis, visceral leishmaniasis, leptospirosis, malaria, and tuberculosis. Results. The indicator showed that 40.5% of Brazilian municipalities had high criticality for the diseases of interest, especially in the North, parts of the Northeast, and Midwest. Indicators "proportion of poverty," "garbage in surroundings," and "families headed by women" increased the chance of higher criticality for the diseases. The indicator "adequate sewer system" was a potential protection factor. Conclusions. The technique used was adequate to guide surveillance actions in the country and allows articulation between local surveillance efforts and other sectors to resolve health problems caused by infectious and parasitic diseases and associated factors.(AU)

A qualitative investigation characterized as an exploratory study of individual case. The guiding point was inquiring into how the Public Health System (SUS) provides intensive home-based care. The study was aimed at analyzing how this health support is made available--that is, what kind of assistance is given and what is the equipment maintenance and warranty like--and at examining how the subject of this study obtained this service, that is, how the service reached him/her and how long it usually takes to do so. The results show that the paths to get life support are full of obstacles and that this path in search of home-based care through the SUS dynamics involves bureaucracy, professionals, and equipment that often make it difficult for the population to have access to public health services.

RESUMO

In cases of asymmetric competition, larger individuals of many animal species have a greater probability of acquiring territory, gaining initial access to resources and finding a mate in comparison to smaller individuals. The competition among larvae of the antlion Myrmeleon brasiliensis (Neuroptera, Myrmeleontidae) is observed in the search for space for the construction of traps, in the forage for prey, and in the occurrence of cannibalism. The body size of the larvae is proportional to the size of the traps and the success of predation. Thus, larger specimens are better competitors in terms of capturing preys (asymmetric competition). The aim of the present study was to determine the effects of asymmetric competition in M. brasiliensis regarding the distance among these larvae. The study was developed in a permanent reserve area, located in the municipality of Aquidauana, from the Mato Grosso do Sul state, Brazil. For the laboratory experiments, we collected M. brasiliensis larvae and placed two larvae in plastic pots. The observations occurred by the visual search of traps, when a larva was seen in the diameter of its trap and the trap of its closest neighbor. Next, the trap size and the distance between them were measured for a period of 15 days. The results of the present study demonstrate that the largest M. brasiliensis larvae were outside the clusters. Within these clusters, the larvae moved away from each other because of the size of their traps, as the larger the neighboring larvae the greater the distance between them. Thus, the asymmetric competition between M. brasiliensis (larger larvae are better competitors) suggests a spatial arrangement among the larvae that is guided by the variable size of the trap.
