RESUMO

A novel mangrove soil-derived actinomycete, strain S2-29T, was found to be most closely related to Saccharopolyspora karakumensis 5K548T based on 16 S rRNA sequence (99.24% similarity) and genomic phylogenetic analyses. However, significant divergence in digital DNA-DNA hybridization, average nucleotide identity, and unique biosynthetic gene cluster possession distinguished S2-29T as a distinct Saccharopolyspora species. Pan genome evaluation revealed exceptional genomic flexibility in genus Saccharopolyspora, with > 95% accessory genome content. Strain S2-29T harbored 718 unique genes, largely implicated in energetic metabolisms, indicating different metabolic capacities from its close relatives. Several uncharacterized biosynthetic gene clusters in strain S2-29T highlighted the strain's untapped capacity to produce novel functional compounds with potential biotechnological applications. Designation as novel species Saccharopolyspora mangrovi sp. nov. (type strain S2-29T = JCM 34,548T = CGMCC 4.7716T) was warranted, expanding the known Saccharopolyspora diversity and ecology. The discovery of this mangrove-adapted strain advances understanding of the genus while highlighting an untapped source of chemical diversity.

BACKGROUND: Nosocomial infections caused by Serratia marcescens mostly occurred in pediatrics and it was very rarely reported after adult surgery. Here, an intracranial abscess caused by Serratia marcescens was reported. We report a rare case of a postoperative intracranial abscess caused by Serratia marcescens in a 63-year-old male patient with a left parietal mass. The patient underwent resection of the mass on June 1, 2022, and the postoperative pathology revealed an angiomatous meningioma, WHO I. He then experienced recurrent worsening of right limb movements, and repeated cranial CT scans showed oozing blood and obvious low-density shadows around the operation area. Delayed wound healing was considered. Subsequently, a large amount of pus was extracted from the wound. The etiological test showed that Serratia marcescens infection occurred before the removal of the artificial titanium mesh. Antibiotics were initiated based on the results of drug susceptibility tests. At present, the patient is recovering well and is still closely monitored during follow-up. CONCLUSION: It is rare for Serratia marcescens to cause brain abscesses without any obvious signs of infection. This report provided in detail our experience of a warning postoperative asymptomatic brain abscess caused by an uncommon pathogen.

RESUMO

A robust five-degree-of-freedom (5-DOF) measurement system is proposed in this paper. The compact optical configuration with high resolution is designed based on lens combination and multiple reflections. Beam drift and dual-beam parallelism are monitored and compensated by autocollimator units and a polarizer unit respectively. In addition, a protection method is proposed to reduce the intensity of air turbulence by reducing the Reynolds number of the beam path. The performance of the system is verified by experiments. The experimental results show that the self-compensation methods and air turbulence protection can effectively improve the accuracy and stability of the system under the long-term interference of external environments. The proposed system has high precision, desirable robustness, and convenient pre-calibration, which can be used for error measurement of precision machines.

In this paper, a high-accuracy measurement method for rotor-stator axial clearance in narrow spaces is proposed. The optical path structure based on all-fiber microwave photonic mixing is established. To improve the accuracy and expand the measurement range, the total coupling efficiency over the entire measurement range at different working distances of fiber probe was evaluated by Zemax analysis tool and theoretical model. The performance of the system was verified by experiments. The experimental results show that the measurement accuracy of axial clearance is better than 10.5 um within the range of 0.5-20.5 mm. The measurement accuracy has been effectively improved compared to previous methods. Additionally, the probe size is reduced to a mere diameter of 2.78 mm, which is more suitable for axial clearance measurement in narrow spaces inside rotating machines.

Rotor-stator axial clearance is a crucial design parameter affecting rotating machines' efficiency and safety. To accurately measure the dynamic axial clearance in high-speed machinery, a precise method based on time division multiplexing with frequency domain interferometry has been proposed. This method has proven robust and accurate through simulations and experiments. The inclusion of an optical switch enables the utilization of dispersive interferometry(DPI) and time division multiplexing for multiple channels of the light source. It achieves a static accuracy of 1.5 µm for a 15 mm range and a dynamic accuracy of 9 µm at 3000 rpm.

OBJECTIVE: To evaluate the feasibility and quality of biliary-enteric reconstruction (BER) in laparoscopic radical resection of hilar cholangiocarcinoma (LsRRH) versus open surgery and propose technical recommendations. METHODS: Data of 38 LsRRH and 54 radical laparotomy resections of hilar cholangiocarcinoma (LtRRH) cases were collected from our institution. BER was evaluated via biliary residuals numbers, number of anastomoses, anastomosis manner, suture method, time consumption, and postoperative complication. RESULTS: In the LsRRH group, patients were relatively younger; Bismuth type I had a higher proportion while type IIIa and IV were less and required no revascularization. In LsRRH and LtRRH groups, respectively, the biliary residuals number was 2.54 ± 1.62 and 2.47 ± 1.46 (p > 0.05); the number of anastomoses was 2.04 ± 1.27 and 2.57 ± 1.33 (p > 0.05); the time of BER was 65.67 ± 21.53 and 42.5 ± 19.77 min (p < 0.05), 15.08 ± 3.64% and 11.76 ± 2.54% of the total operation time (p < 0.05); postoperative bile leakage incidence was 15.79% and 16.67% (p > 0.05); 14 ± 10.28 and 17 ± 9.73 days for healing (p < 0.05); anastomosis stenosis rate was 2.63% and 1.85% (p > 0.05). Neither group had a biliary hemorrhage or bile leakage-related death. CONCLUSION: The selection bias in LsRRH mainly affects tumor resection than BER. Our cohort study indicates that BER in LsRRH is technically feasible and equals anastomotic quality to open surgery. However, its longer and a more significant proportion of total operation time represent that BER has higher technical requirements and is one of the critical rate-limiting steps affecting the minimal invasiveness of LsRRH.

A novel actinomycete, designated strain S1-112 T, was isolated from a mangrove soil sample from Hainan, China, and characterized using a polyphasic approach. Strain S1-112 T showed the highest similarity of the 16S rRNA gene to Streptomonospora nanhaiensis 12A09T (99.24%). Their close relationship was further supported by phylogenetic analyses, which placed these two strains within a stable clade. The highest values of digital DNA-DNA hybridization (dDDH, 41.4%) and average nucleotide identity (ANI, 90.55%) were detected between strain S1-112 T and Streptomonospora halotolerans NEAU-Jh2-17 T. Genotypic and phenotypic characteristics demonstrated that strain S1-112 T could be distinguished from its closely related relatives. We also profiled the pan-genome and metabolic features of genomic assemblies of strains belonging to the genus Streptomonospora, indicating similar functional capacities and metabolic activities. However, all of these strains showed promising potential for producing diverse types of secondary metabolites. In conclusion, strain S1-112 T represents a novel species of the genus Streptomonospora, for which the name Streptomonospora mangrovi sp. nov. was proposed. The type strain is S1-112 T (= JCM 34292 T).

Two Gram-positive, aerobic and non-motile actinomycetes, designated S1-96T and N2-109T, were isolated from soils collected from a cotton field. They are described as representing two novel species of genera Actinophytocola and Streptomyces through a polyphasic approach. Analysis of 16S rRNA gene sequences revealed that strains S1-96T and N2-109T showed highest similarity to Actinophytocola xinjiangensis CGMCC 4.4663T (99.10 %) and Streptomyces iconiensis BNT558T (98.21 %), respectively. Phylogenetic analyses based on 16S rRNA and core genes confirmed the close relationships of these strains. Genomic analyses further supported the novel taxonomic delimitation of these two species based on digital DNA-DNA hybridization and average nucleotide identity. Strains S1-96T and N2-109T contained MK-9(H4) and MK-9(H6) as the most abundant menaquinone, respectively. High abundances of iso-fatty acids were detected in both strains, which was similar to their close relatives. Physiological and polar lipid analyses also revealed differences between these strains and their phylogenetic neighbours, supporting their taxonomic delimitation as novel species. The names Actinophytocola gossypii sp. nov. (type strain S1-96T=JCM 34412T=CGMCC 4.7707T) and Streptomyces gossypii sp. nov. (type strain N2-109T=JCM 34628T=CGMCC 4.7717T) are proposed.

BACKGROUND AND AIMS: Limited EST plus EPLBD has been widely used for the therapy of large CBDS; however, long-term complication-relevant studies suggested that it damaged the function of the sphincter of Oddi (SO) and resulted in recurrent choledocholithiasis. Thus, we designed Endoclip papillaplasty (ECPP) and previous studies have shown that it successfully restored the function of SO. In this study, we designed a prospective cohort and aimed to verify the safety and effectiveness of ECPP. METHODS: Eligible patients were divided into the ECPP group and the limited EST plus EPLBD group based on papillary morphology and the ratio of maximum size of stones to length of intramural segments of CBD. All participants in the ECPP group received endoscopy at 3 weeks to retrieve the biliary stent, perform SOM, and were divided into grade A and grade B based on the healing grade of SO. All patients were followed up every 6 months until recurrent choledocholithiasis, patient death, or at the 36-month follow-up end. The primary outcome was the incidence of recurrent choledocholithiasis. The secondary outcomes included mechanical lithotrip usage and adverse events. RESULTS: The incidences of recurrent choledocholithiasis in the ECPP group and limited EST plus EPLBD group were 13.6 and 22.1%, respectively (P = 0.204). The ECPP-A group had a lower incidence of recurrent choledocholithiasis than the limited EST plus EPLBD groups (5.1 vs. 22.1%, P = 0.020*), and certified the function of SO successfully restored in the ECPP-A group. CONCLUSION: The ECPP-A group had a decrease in recurrent choledocholithiasis, and ECPP was safe and effective for CBDS.

In the domain of frequency sweeping interferometry, the accurate extraction of distance information from nonlinear frequency scanning signals holds paramount significance in ensuring meticulous measurements of high precision. This paper presents a novel, to the best of our knowledge, high-speed distance extraction algorithm based on the table lookup method and validates its feasibility through theoretical models, simulations, and practical experiments. The proposed algorithm achieves comparable accuracy to traditional methods involving resampling and Hilbert transform. However, it outperforms them in robustness against noise and variations in sampling points. This method can accurately process signals sampled even below the Nyquist sampling rate. The simplicity and computational efficiency of the proposed approach make it suitable for various nonlinear sampling applications, promising broad applicability in scientific and engineering contexts.

Pseudomonas fragi and Escherichia coli are considered as common colonizers of fresh and spoilage meat, where they tend to live in the proximity. In this study, we primarily tested interplay patterns between different isolates of these two species in two-by-two combinations grown on stainless steel surfaces as dual-species biofilms. Results showed that these two species presented competition as major observed interplay patterns as biofilms progressed independent of bacterial strains and growth temperatures (15 °C and 25 °C). One dual-species combination was proposed as a representative to further explore dynamic patterns of interaction strength between these two species, with species colonization order taken into consideration as a biological effector. We firstly reported that prior colonization of one species significantly decreased the initiatively colonized cell counts of counterpart species by one to three orders of magnitude when competing for limited adhesion surface, under which E. coli was observed to be more aggressive in surface colonization as compared to P. fragi. However, the spatial structure and microbial composition of mature dual-species biofilms were not observed to be significantly affected. Our findings also shed new light on the evidence that E. coli and P. fragi, respectively, enhanced their biofilm formation capabilities by upregulating expression level of genes that encoded Type 1 fimbriae and phosphate response regulator as dual-species consortia progressed, which could serve as a crucial factor that improved the difficulty of food biocontrol.

PURPOSE: This study determined independent predictors and developed a predictive nomogram for failed correction of intertrochanteric fractures due to cut-out of the proximal femur nail anti-rotation (PFNA) device. METHODS: Demographic and radiological data of 592 adult patients with intertrochanteric fractures (AO 31A) treated by PFNA were collected retrospectively. Independent predictors of cut-out were obtained through univariate and multivariate analyses, and a predictive nomogram was established. The discrimination, calibration, and clinical utility of the nomogram were based on receiver operating characteristic curve (AUC), concordance index (C-index), calibration curve, and decision curve analysis, respectively. RESULTS: Overall, 18 (3.04%) cases of cut-out occurred. Independent predictors according to the multivariate analysis were body mass index (BMI), poor-to-acceptable quality of reduction, PFNA blade position, and tip-apex distance (TAD). AUC of the nomogram was 0.849, and C-index was 0.849 (95% CI [0.844-0.854]). Bootstrapping yielded a corrected C-index of 0.849. The calibration and decision curves indicated good agreement and clinical benefit of the nomogram. CONCLUSION: A reliable predictive nomogram was developed for cut-out of the PFNA in intertrochanteric fractures, based on BMI, quality of reduction, blade position, and TAD.

A non-spore-forming, motile and alkali-resistant actinobacterium, designated N2-46T, was isolated from an alkaline soil sample collected from a cotton field in the Xinjiang region of PR China. Strain N2-46T formed creamy colonies on tryptone soy agar and managed to survive in extreme alkaline conditions at a pH value of 11. Strain N2-46T displayed the highest 16S rRNA gene similarity of 99.65 % to Haloactinobacterium kanbiaonis HY164T, followed by Occultella aeris F300T (99.61%) and Occultella glacieicola T3246-1T (98.54 %). 16S rRNA-directed phylogenetic analysis showed that strain N2-46T was embedded in a subclade with O. aeris F300T with a bootstrap value of 71.8 %. The phylogenetic tree based on core genes of genome sequences showed that strain N2-46T formed a unique subclade next to H. kanbiaonis HY164T and O. aeris F300T with a bootstrap value of 100 %. Digital DNA-DNA hybridization and the average nucleotide identity analyses showed that strain N2-46T displayed the highest values of 67.1 % (63.2-70.7 %) and 91.82 % with H. kanbiaonis HY164T, respectively. Comparative genomic analysis indicated that strain N2-46T and its three closest neighbours exhibited comparable distribution patterns in heavy metal resistance genes and biosynthetic gene clusters, while displaying distinctions probably related to ecological adaptation. MK-8(H4) was identified as the predominant isoprenoid quinone. The main fatty acids were identified as iso-C14 : 0 and anteiso-C15 : 0. Polar lipids are composed of diphosphatidylglycerol, phosphatidylglycerol, phosphatidylinositol, mono and diacylated phosphatidylinositol dimannosides, as well as several uncharacterized polar lipid, glycolipid, and phospholipids. Genotypic and physiological analyses support the view that strain N2-46T (=JCM 34413T=CGMCC 1.18819T) should be classified as a novel species of the genus Occultella, for which the name Occultella gossypii sp. nov. is proposed.

A novel actinomycete strain, designated S2-4T, was isolated from a mangrove soil sample, and a polyphasic approach was employed to determine its taxonomic position. Phylogenetic analysis based on 16S rRNA gene indicated that strain S2-4T formed a unique clade next to that harboring Pseudonocardia dioxanivorans CB1190T, which shared the highest sequence similarity (98.20%) with the new isolate. Phylogenetic analysis based on core genes of genomic sequences displayed a different scenario, exhibiting closer phylogenetic relationship of strain S2-4T with several species with 16S rRNA gene sequence similarities ranging from 96.95 to 98.06%, which was confirmed by the phylogenetic tree reconstructed based on genomic sequences. Further, substantial differences between the genotypic properties of strain S2-4T and its closest neighbors, including digital DNA-DNA hybridization, average nucleotide identity, and distribution patterns of biosynthetic gene clusters (BGC), indicated the taxonomic position of strain S2-4T as a novel species of the genus Pseudonocardia. Accordingly, strain S2-4T was observed to show a different distribution pattern of a predicted BGC encoding ectoine by comparative genomic analysis, which could be strongly linked to its unique habitat distinct from where its close neighbors were isolated. The major cellular fatty acids were iso-C15:0, C21:0, and iso-C16:0. The predominant menaquinone was MK-8(H4). The polar lipids were composed of diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, phosphatidyl-N-monomethylethanolamine, phosphatidylcholine, phosphatidylinositol, phosphatidylinositol mannosides, and two unidentified glycolipids. Here, we propose a novel species of the genus Pseudonocardia: Pseudonocardia humida sp. nov. with the type strain S2-4T (= JCM 34291T = CGMCC 4.7706T).

Escherichia coli O157:H7 and Pseudomonas were considered as common colonizers of fresh and spoilage meat, where they tended to live in the proximity. In this study, we tested the interplay between different isolates of E. coli O157:H7 and Pseudomonas in random two-by-two combinations grown as dual-species consortia. Results showed that the growth fitness of E. coli was not facilitated by the presence of all tested Pseudomonas strains, and vice versa. Representative combinations were further selected to investigate the property changes following the time course of biofilms formation as compared to single species. Cell counting confirmed that the growth of E. coli O157:H7 was challenged by the presence of Pseudomonas strains as previously described. Our findings shed new light on the evidence that the pathogenicity of E. coli O157:H7 was negatively affected by the presence of Pseudomonas according to the evaluation of spatial organization and genetic expression of virulence factors, which might be a naturally existing biological phenomenon constraining the safety risk of former strains in meat processing and preservation. Intriguingly, we observed that E. coli managed to stably co-exist at low cellular abundance in the progress of dual-species consortia, indicating successful adaptive mechanisms that need further investigations to uncover.

Microsatellites have attracted a large number of scholars and engineers because of their portability and distribution characteristics. The ground station suitable for microsatellite service has become an important research topic. In this paper, we propose a networked ground station and verify it on our own microsatellite. The specific networked ground station system consists of multiple ground nodes. They can work together to complete data transmission tasks with higher efficiency. After describing our microsatellite project, a reasonable distribution of ground nodes is given. A cloud computing model is used to realize the coordination of multiple ground nodes. An adaptive communication system between satellites and ground stations is used to increase link efficiency. Extensive on-orbit experiments were used to validate our design. The experimental results show that our networked ground station has excellent performance in data transmission capability. Finally, the specific cloud-computing-based ground station network successfully completes our satellite mission.

Working memory is an important foundation for advanced cognitive function. The paper combines the spatiotemporal advantages of electroencephalography (EEG) and functional near-infrared spectroscopy (fNIRS) to explore the neurovascular coupling mechanism of working memory. In the data analysis, the convolution matrix of time series of different trials in EEG data and hemodynamic response function (HRF) and the blood oxygen change matrix of fNIRS are extracted as the coupling characteristics. Then, canonical correlation analysis (CCA) is used to calculate the cross correlation between the two modal features. The results show that CCA algorithm can extract the similar change trend of related components between trials, and fNIRS activation of frontal pole region and dorsolateral prefrontal lobe are correlated with the delta, theta, and alpha rhythms of EEG data. This study reveals the mechanism of neurovascular coupling of working memory, and provides a new method for fusion of EEG data and fNIRS data.

BACKGROUND: Accumulating evidence has demonstrated the close relation of SOX1 with tumorigenesis and tumor progression. Upregulation of SOX1 was recently shown to suppress growth of human cancers. However, the expression and role of SOX1 in cholangiocarcinoma (CCA) is not well characterized. METHODS: Expression levels of SOX1 in CCA tissues and normal bile duct tissues were examined using public GEO database. Western blot and immunohistochemistry were used to confirm the expression levels. Cell proliferation assay (CCK-8) and colony formation assay were performed to assess proliferation of CCA cells. A mouse model of subcutaneous transplantable tumors was used to evaluated proliferation of CCA in vivo. The putative regulating factor of SOX1 were determined using Targetscan and dual-luciferase reporter assay. RESULTS: SOX1 was downregulated in CCA tissues. Overexpression of SOX1 significantly inhibited cell proliferation in vitro and suppressed tumor growth in vivo. miR-155-5p directly targeted the 3'-untranslated region (3'UTR) of SOX1 and inhibited expression of SOX1, resulting in the activation of RAF, MEK and ERK phosphorylation, and thus CCA proliferation. However, restoration of SOX1 expression in miR-155-5p overexpressing cell lines decreased the phosphorylation level of RAF, MEK and ERK, as well as the proliferation of CCA cells. CONCLUSION: MiR-155-5p decreased the expression of SOX1 by binding to its 3'UTR, which activated the RAF/MEK/ERK signaling pathway and promoted CCA progression.

A Gram-negative bacterium, designated S1-65T, was isolated from soil samples collected from a cotton field located in the Xinjiang region of PR China. Results of 16S rRNA gene sequence analysis revealed that strain S1-65T was affiliated to the genus Steroidobacter with its closest phylogenetic relatives being 'Steroidobacter cummioxidans' 35Y (98.4 %), 'Steroidobacter agaridevorans' SA29-B (98.3 %) and Steroidobacter agariperforans KA5-BT (98.3 %). 16S rRNA-directed phylogenetic analysis showed that strain S1-65T formed a unique phylogenetic subclade next to 'S. agaridevorans' SA29-B and S. agariperforans KA5-BT, suggesting that strain S1-65T should be identified as a member of the genus Steroidobacter. Further, substantial differences between the genotypic properties of strain S1-65T and the members of the genus Steroidobacter, including average nucleotide identity and digital DNA-DNA hybridization, resolved the taxonomic position of strain S1-65T and suggested its positioning as representing a novel species of the genus Steroidobacter. The DNA G+C content of strain S1-65T was 62.5 mol%, based on its draft genome sequence. The predominant respiratory quinone was ubiquinone-8. The main fatty acids were identified as summed feature 3 (C16:1ω6c/C16:1ω7c), C16 : 0 and iso-C15 : 0. In addition, its polar lipid profile was composed of aminophospholipid, diphosphatidylglycerol, phosphatidylethanolamine and phosphatidylglycerol. Here, we propose a novel species of the genus Steroidobacter: Steroidobacter gossypii sp. nov. with the type strain S1-65T (=JCM 34287T=CGMCC 1.18736T).

Research on the roles of the bacteria in tumor development and progression is a rapidly emerging field. Increasing evidence links bacteria with the modification of the tumor immune microenvironment, which greatly influences the antitumor response. In view of the individual immune effects of various bacteria in various tumors, developing personalized bacteria-modulating therapy may be a key to successful antitumor treatment. This review emphasizes the critical role of the bacteria in immune regulation, including both the tumor bacteria and gut bacteria. Aiming at tumor-related bacteria, we focus on various precise modulation strategies and discuss their impact and potential for tumor suppression. Finally, engineered bacteria with tumor-targeting ability could achieve precise delivery of various payloads into tumors, acting as a precision tool. Therefore, a precise tumor-related bacteria therapy may be a promising approach to suppress the development of tumors, as well as an adjuvant therapy to improve the antitumor efficacy of other approaches. KEY POINTS: • The mini-review updates the knowledge on complex effect of bacteria in TME. • Insight into the interaction and adjustment of bacteria in gut for TME. • Prospects and limitations of bacteria-related personalized therapy in the clinical anticancer therapy.
