RESUMO

There is considerable potential for nuclear genomic material in environmental DNA (eDNA) to inform us of population genetic structure within aquatic species. We tested if nuclear allelic composition data sourced from eDNA can resolve fine scale spatial genetic structure of the cichlid fish Astatotilapia calliptera in Lake Masoko, Tanzania. In this ∼35 m deep crater lake the species is diverging into two genetically distinguishable ecomorphs, separated by a thermo-oxycline at ∼15 m that divides biologically distinct water masses. We quantified population genetic structure along a depth transect using single nucleotide polymorphisms (SNPs) derived from genome sequencing of 530 individuals. This population genetic structure was reflected in a focal set of SNPs that were also reliably amplified from eDNA - with allele frequencies derived from eDNA reflecting those of fish within each depth zone. Thus, by targeting known genetic variation between populations within aquatic eDNA, we measured genetic structure within the focal species.

Climate change seriously affects habitat suitability for coldwater fish. Amur whitefish (Coregonus ussuriensis) is a commercially important coldwater fish species found in northeast China that is particularly sensitive to water temperature. We use water temperature models for Songhua (Harbin to Yilan) and Mayi rivers to predict habitat suitability for Amur whitefish and other coldwater fish species during the months of July and August in 2014 and 2100-2 months during which river temperatures can reach extremes. The predict-results showed that fish habitat was to deteriorate over time, and to be less suitable for survival of coldwater fish species in 2100 than that in 2014. Under scenario RCP4.5 (Representative Concentration Pathway 4.5), the time period during which habitat for Amur whitefish was unsuitable increased continually; under scenario RCP8.5 (Representative Concentration Pathway 8.5), Amur whitefish might even disappear from the river basin. A significant increase in river water temperature will compromise coldwater fish survival and possibly lead to their disappearance. Practical measures to control greenhouse gas emissions and reduce the impact of global warming on aquatic ecosystems must be taken.

Rainwater chemistry was investigated at a semi-rural site in Ya'an, Sichuan basin with rain samples collected from May 2013 to July 2014. The rainwater pH values ranged from 3.25 to 6.86, with an annual volume-weighted mean (VWM) of 4.38, and the acid rain frequency was 74 %. Such severe acidification, 15 % of the total events showed a pH below 4.0, attributed to the deficiency of Ca(2+), significant anthropogenic pollution contribution, and rainy pattern to this area. The annual VWM of total ions concentration was 477.19 µeq/L. NH4 (+) was the most abundant ionic species, followed by SO4 (2-), NO3 (-), Ca(2+), Cl(-), Na(+), K(+), Mg(2+), and F(-) in a descending order. The total ionic concentrations presented a seasonal trend of lower values in autumn and summer but higher ones in winter and spring. Based on enrichment factor, correlation analysis and principle component analysis, three factors were identified: factor 1 (NH4 (+), SO4 (2-), NO3 (-), K(+), and Cl(-), 47.45 % of the total variance) related to anthropogenic sources (coal/fuel combustion, biomass burning and agriculture), factor 2 (Ca(2+), Mg(2+), Na(+), and Cl(-), 34.01 % of the total variance) associated with natural sources, and factor 3 (H(+), 11.78 % of the total variance) related to free acidity. Back trajectory analysis indicates that the rainwater chemistry in Ya'an was mainly affected by regional air masses from Sichuan basin. Long-range transported air masses from southwest with heavy anthropogenic pollution increased the total ion concentration and acidity of rainwater. Considering its special topography, anthropogenic emissions from regional and long-range transport (especially from southwest) must be controlled effectively to improve the acid rain condition of non-urban areas in Sichuan basin.

Integrated PM2.5 aerosol samples were collected at Baima Spring Scenic Area, a forest site of Yaan, Sichuan Province, during the summer of 2010. Organic speciation including isoprene oxidation products (2-methyltetrols, C5-alkene trols, 2-methylyceric acid), alpha-/beta-pinene oxidation products (norpinic acid, 3-hydroxyglutaric acid, 3-methy-1,2,3-butanetricarboxylic acid), and small molecular carboxylic acid (malic acid, 2-hydroxyglutaric acid) were analyzed. The generation mechanisms of SOA as well as their influencing factors were particularly discussed. Results show that average concentrations of 2-methyltetrols, C5-alkene triols, 2-methyglyceric acid, norpinic acid, 3-hydroxyglutaric acid and 3-methy-1,2,3-butanetricarboxylic acid are 63.3, 45.0, 4.4, 4.1, 5.0, 5.3 ng x m(-3) respectively, of 24-hour lapse samples. SOA compounds are consistent with higher concentrations in the day than during the night only except for norpinic acid. Relatively high level of biogenic SOA at the study area is concerned with many environmental factors, i. e. local abundant vegetations, warm and humid climate, sunken valley topography, the atmospheric pollution state, etc.
