RESUMO

Bismuth halogenoxide (BiOX)-based heterojunctions have garnered considerable attention recently due to their potential to enhance photocatalytic performance. However, the predominant focus on II-type heterojunctions has posed challenges in achieving the requisite band edge positions for efficient water splitting. In this investigation, stable van der Waals SbPO4/BiOClxBr1-x heterojunctions were constructed theoretically by using density-functional theory (DFT). Our findings demonstrate that SbPO4 can modulate the formation of Z-scheme heterojunctions with BiOClxBr1-x. The structural properties of BiOX were preserved, while reaching excellent photocatalytic capabilities with high redox capacities. Further investigation unveiled that the band edge positions of the heterojunctions fully satisfy the oxidation-reduction potential of water. Moreover, these heterojunctions exhibit notable absorption efficiency in the visible range, with absorption increasing as x decreases. Our research provides valuable theoretical insights for the experimental synthesis of high-performance BiOX-based photocatalysts for water splitting, leveraging the unique properties of SbPO4. These insights contribute to the advancement of clean energy technology.

RESUMO

Alzheimer's disease (AD) is the most prevalent neurodegenerative disorder characterized by cognitive impairments. Despite the limited efficacy of current treatments for AD, the 1,2,4-oxadiazole structure has garnered significant attention in medicinal chemistry due to its potential impact on mGluR1 and its association with AD therapy. In this study, a series of novel 1,2,4-oxadiazole derivatives were designed, synthesized, and evaluated for the neuroprotective effects in human neuroblastoma (SH-SY5Y) cells. Among all the derivatives tested, FO-4-15 (5f) existed the lowest cytotoxicity and the highest protective effect against H2O2. Based on these in vitro results, FO-4-15 was administered to 3×Tg mice and significantly improved the cognitive impairments of the AD mice. Pathological analysis showed that FO-4-15 significantly reduced Aß accumulation, Tau hyper-phosphorylation, and synaptic impairments in the 3×Tg mice. Dysfunction of the CaMKIIα/Fos signaling pathway in 3×Tg mice was found to be restored by FO-4-15 and the necessity of the CaMKIIα/Fos for FO-4-15 was subsequently confirmed by the use of a CaMKIIα inhibitor in vitro. Beyond that, mGluR1 was identified to be a potential target of FO-4-15, and the interaction of FO-4-15 and mGluR1 was displayed by Ca2+ flow increase, molecular docking, and interaction energy analysis. The target of FO-4-15 was further confirmed in vitro by JNJ16259685, a nonselective inhibitor of mGluR1. These findings suggest that FO-4-15 may hold promise as a potential treatment for Alzheimer's disease.

RESUMO

Low-temperature curable negative-tone photosensitive polyimide (n-LTPI) viscous solutions were prepared by dissolving photo-crosslinkable poly (amic ester) (pc-PAE) resin, photophotocrosslinker, photoinitiator, and the heteroaromatic base as curing catalysts, and other additives in organic solvents. Among them, the pc-PAE resin was synthesized by polycondensation of aromatic diacid chloride and diester of 2-ethoxymathacrylate, aromatic diamines in aprotic solvents. After being spun-coated on a silicon wafer surface, soft-baked, exposed to UV light, and developed, the n-LTPI with 2% of imidazole (IMZ) as a curing catalyst produced high-quality photo-patterns with line via resolution of 5 µm at 5 µm film thickness. The photo-patterned polymer films thermally cured at 230 °C/2 h in nitrogen showed 100% of the imidization degree (ID) determined by in situ FT-IR spectroscopy. The thermally cured polymer films exhibited great combined mechanical and thermal properties, including mechanical properties with tensile strength of as high as 189.0 MPa, tensile modulus of 3.7 GP, and elongation at breakage of 59.2%, as well as glass transition temperature of 282.0 °C, showing great potential in advanced microelectronic packaging applications.

RESUMO

When pathological hypertrophy progresses to heart failure (HF), the prognosis is often very poor. Therefore, it is crucial to find new and effective intervention targets. Here, myocardium-specific Trim44 knockout rats were generated using CRISPR-Cas9 technology. Cardiac phenotypic observations revealed that Trim44 knockout affected cardiac morphology at baseline. Rats with Trim44 deficiency exhibited resistance to cardiac pathological changes in response to stimulation via isoproterenol (ISO) treatment, including improvement of cardiac remodeling and dysfunction by morphological and functional observations, reduced myocardial fibrosis and reduced expression of molecular markers of cardiac stress. Furthermore, signal transduction validation associated with growth and hypertrophy development in vivo and in vitro demonstrated that Trim44 deficiency inhibited the activation of signaling pathways involved in myocardial hypertrophy, especially response to pathological stress. In conclusion, the present study indicates that Trim44 knockout attenuates ISO-induced pathological cardiac remodeling through blocking the AKT/mTOR/GSK3ß/P70S6K signaling pathway. This is the first study to demonstrate the function and importance of Trim44 in the heart at baseline and under pathological stress. Trim44 could be a novel therapeutic target for prevention of cardiac hypertrophy and HF.

RESUMO

Aeration tanks from activated sludge wastewater treatment plants (WWTPs) can release a large amount of bioaerosols that can pose health risks. However, risk characterization of bioaerosols emissions form wastewater treatment plants is currently not systematically carried out and still in its infancy. Therefore, this study investigated emission characteristic of two indicator model bioaerosols Staphylococcus aureus and Escherichia coli, emitted from aeration tanks of a municipal WWTP. Monte Carlo simulation was then used to quantitatively assess microbial risk posed by different aeration modes under optimistic and conservative estimates. Further to this, two different exposure scenarios were considered during 3 days sampling campaign in autumn and winter. Results showed that the bioaerosol concentration from microporous aeration tank (20-262 CFU m-3) was one order of magnitude lower than rotating disc aeration tank. Average aerosolization rate was 7.5 times higher with mechanical aeration mode. Health risks of exposed populations were 0.4 and 9.6 times higher in winter than in autumn for E. coli and S. aureus bioaerosol, respectively. Health risks of staff members were 10 times higher than academic visitors. Interesting results were observed for academic visitors without personal protective equipment (PPE) respectively exposed to S. aureus and E. coli bioaerosol in autumn and winter: while the derived infection risk met the United States Environmental Protection Agency (U.S. EPA) benchmark under optimistic estimation, the disease risk burden was over the World Health Organization (WHO) benchmark under conservative estimation. These revealed that only satisfying one of the two benchmarks didn't mean absolute acceptable health risk. This study could facilitate the development of better understanding of bioaerosol quantitative assessment of risk characterizations and corresponding appropriate risk control strategies for wastewater utilities.

RESUMO

Wastewater treatment plants (WWTPs) could emit a large amount of bioaerosols containing pathogenic bacteria. Assessing the health risks of exposure to these bioaerosols by using quantitative microbial risk assessment (QMRA) is important to protect workers in WWTPs. However, the relative impacts of the stochastic input variables on the health risks determined in QMRA remain vague. Hence, this study performed a Monte Carlo simulation-based QMRA case study for workers exposing to S. aureus or E. coli bioaerosols and a sensitivity analysis in two WWTPs with various aeration modes. Results showed that when workers equipped without personal protective equipment (PPE) were exposed to S. aureus or E. coli bioaerosol in the two WWTPs, the annual probability of infection considerably exceeded the U.S. EPA benchmark (≤10E-4 pppy), and the disease burden did not satisfy the WHO benchmark (≤10E-6 DALYs pppy) (except exposure to E. coli bioaerosol for disease health risk burden). Nevertheless, the use of PPE effectively reduced the annual infection health risk to an acceptable level and converted the disease health risk burden to a highly acceptable level. Referring to the sensitivity analysis, the contribution of mechanical aeration modes to the variability of the health risks was absolutely dominated in the WWTPs. On the aeration mode that showed high exposure concentration, the three input exposure parameters (exposure time, aerosol ingestion rate, and breathing rate) had a great impact on health risks. The health risks were also prone to being highly influenced by the various choices of the dose-response model and related parameters. Current research systematically delivered new data and a novel perspective on the sensitivity analysis of QMRA. Then, management decisions could be executed by authorities on the basis of the results of this sensitivity analysis to reduce related occupational health risks of workers in WWTPs.

RESUMO

ETHNOPHARMACOLOGICAL RELEVANCE: Ephedrae Herba (EH, Ephedra sinica Stapf.) and Armeniacae Semen Amarum (ASA, Prunus armeniaca L. var. ansu Maxim.) have been used to treat asthma, cold, fever, and cough in China for thousands of years. AIM OF THE STUDY: In this study, we aimed to investigate the optimal ratio of EH and ASA compatibility (EAC) to reduce airway injury in asthmatic rats and its possible mechanism. METHODS: Rats were sensitized with a mixture of acetylcholine chloride and histamine bisphosphate 1 h before sensitization by intragastric administration of EAC or dexamethasone or saline for 7 days. Subsequently, the ultrastructure of rat airway epithelial tissue changes, apoptosis of the airway epithelial cells, and the expression of mRNA and protein of EGRF and Bcl-2 were detected. RESULTS: Transmission electron microscope: EAC (groups C and E) had the most prominent effect on repairing airway epithelial cells' ultrastructural changes in asthmatic rats. TUNEL: dexamethasone and EAC (groups B、C、E and F) inhibited the apoptosis of airway epithelial cells in asthmatic rats (P < 0.05). In situ hybridization: EAC (group E) inhibited the overexpression of EGFR and Bcl-2 mRNA (P < 0.05).Western Blotting: EAC (groups A、B、C、E and F) inhibited the upregulation of airway epithelial EGFR and Bcl-2 protein expression (P < 0.01). CONCLUSIONS: Our findings indicate that EAC can inhibit abnormal changes in airway epithelial structure and apoptosis of airway epithelial cells, thereby alleviating airway injury. In this study, the best combination of EH and ASA to alleviate airway epithelial injury in asthmatic rats was group E (EH: ASA = 8: 4.5).

RESUMO

MicroRNAs have been reported as related to multiple diseases and have potential applications in diagnosis and therapeutics. However, detection of miRNAs remains improvable, given their complexity, high cost, and low sensitivity as of currently. In this study, we attempt to build a novel platform that detects miRNAs at low cost and high efficacy. This detection system contains isothermal amplification, detecting and reporting process based on rolling circle amplification, CRISPR-Cas9, and split-horseradish peroxidase techniques. It is able to detect trace amount of miRNAs from samples with mere single-base specificity. Moreover, we demonstrated that such scheme can effectively detect target miRNAs in clinical serum samples and significantly distinguish patients of non-small cell lung cancer from healthy volunteers by detecting the previously reported biomarker: circulating let-7a. As the first to use CRISPR-Cas9 in miRNA detection, this method is a promising approach capable of being applied in screening, diagnosing, and prognosticating of multiple diseases.

RESUMO

We have shown that the slow eye movements extracted from electrooculogram (EOG) signals can be used to estimate human vigilance in our previous work. However, the traditional method for recording EOG signals is to place the electrodes near the eyes of subjects. This placement is inconvenient for users in real-world applications. This paper aims to find a more practical placement for acquiring EOG signals for vigilance estimation. Instead of placing the electrodes near the eyes, we place them on the forehead. We extract EOG features from the forehead EOG signals using both independent component analysis and support vector machines. The performance of our proposed method is evaluated using the correlation coefficients between the forehead EOG signals and the traditional EOG signals. The results show that a correlation of 0.84 can be obtained when the users make 14 different face movements and for merely eye movements it reaches 0.93.

RESUMO

This study aims at using electrooculographic (EOG) features, mainly slow eye movements (SEM), to estimate the human vigilance changes during a monotonous task. In particular, SEMs are first automatically detected by a method based on discrete wavelet transform, then linear dynamic system is used to find the trajectory of vigilance changes according to the SEM proportion. The performance of this system is evaluated by the correlation coefficients between the final outputs and the local error rates of the subjects. The result suggests that SEMs perform better than rapid eye movements (REM) and blinks in estimating the vigilance. Using SEM alone, the correlation can achieve 0.75 for off-line, while combined with a feature from blinks it reaches 0.79.
