A 41-year-old man developed rapidly progressive cranial neuropathies and muscle weakness followed by respiratory failure, requiring ventilation support. On examination, there was marked bilateral ptosis and ophthalmoplegia with bulbar, neck and proximal upper limb weakness. He had a recent open left humeral fracture that eventually required amputation. Despite immunoglobulin therapy, his progressive weakness continued. Multiple investigation results were inconclusive. Eventually, botulinum type A toxin was found positive, by which time the therapeutic window for antitoxin had passed. He continued on supportive management and was treated for concomitant infections and nosocomial illnesses. He was subsequently weaned from respiratory support and has made a good neurological recovery.

The ArcLight observatory provides an hourly continuous time series of all-sky images providing light climate data (intensity, spectral composition, and photoperiod) from the Arctic (Svalbard at 79°N). Until recently, no complete annual time series of light climate relevant for biological processes has been provided from the high Arctic because of insufficient sensitivity of commercial light sensors during the Polar Night. The ArcLight set up is unique, as it provides both all-sky images and the corresponding integrated spectral irradiance in the visible part of the solar electromagnetic spectrum (E P A R ). Here we present a further development providing hourly diel-annual dynamics from 2020 of the irradiance partitioned into the red, green, and blue parts of the solar spectrum and illustrate their relation to weather conditions, and sun and moon trajectories. We show that there is variation between the RGB proportions of irradiance throughout the year, with the blue part of the spectrum showing the greatest variation, which is dependent on weather conditions (i.e., cloud cover). We further provide an example of the biological impact of these spectral variations in the light climate using in vivo Chl a-specific absorption coefficients of diatoms (mean of six low light acclimated northern-Arctic bloom-forming species) to model total algal light absorption (AQ t o t a l ) and the corresponding fraction of quanta used by Photosystem II (AQPSII) (O 2 production) in RGB bands and the potential impacts on the photoreceptor response, suggesting periods where repair and maintenance functions dominate activity in the absence of appreciable levels of red or green light. The method used here can be applied to light climate data and spectral response data worldwide to give localized ecological models of AQ.

The optical chain and logger (OptiCAL) is an autonomous ice-tethered observatory equipped with multiple light sensors for mapping the variation of light with depth. We describe the instrument and present an ensemble calibration for downwelling irradiance E P A R in [µm o l m -2 s -1]. Results from a long-term deployment in the Arctic Ocean demonstrate that the OptiCAL can cover the high dynamic range of under-ice light levels from July to November and produce realistic values in terms of magnitude when compared to modeled surface irradiance. Transient features of raised light levels at specific depths associated with nearby leads in the ice underline the importance of depth-resolved light measurements.

A series of Monte Carlo and HydroLight radiative transfer simulations are used to demonstrate that the traditional form of the Fresnel transmission across the water-air interface is accurate. This contradicts assertions to the contrary in a recent paper [Opt. Express25, 27086 (2017)10.1364/OE.25.027086] that suggested that the impact of multiple surface interactions had previously been ignored and that the transmission factor was dependent upon the turbidity of the water.

Until now, no complete annual time series of biologically relevant light has been provided from the high Arctic due to insufficient sensitivity of commercial light sensors during the Polar Night. We describe a camera system providing all-sky images and the corresponding integrated spectral irradiance (EPAR) in energy or quanta units, throughout a complete annual cycle. We present hourly-diel-annual dynamics from 2017 to 2020 of irradiance and its relation to weather conditions, sun and moon trajectories.

The editorial team are delighted to present this Special Issue of Sensors focused on Remote Sensing of Ocean Color: Theory and Applications. We believe that this is a timely opportunity to showcase current developments across a broad range of topics in ocean color remote sensing (OCRS). Although the field is well-established, in this Special Issue we are able to highlight advances in the applications of the technology, our understanding of the underpinning science, and its relevance in the context of monitoring climate change and engaging public participation.

The nature and magnitude of measurement uncertainties (precision and accuracy) associated with two approaches for measuring absorption by turbid waters (b(532 nm) ranging from 0.20 m-1 to 22.89 m-1) are investigated here: (a) point source integrating cavity absorption meters (PSICAM), and (b) reflective tube absorption meters (AC-9 and AC-s - both WET Labs Inc., USA). Absolute measurement precision at 440 nm was quantified using standard deviations of triplicate measurements for the PSICAM and de-trended, bin averaged time series for the AC-9/s, giving comparable levels (< 0.006 m-1) for both instruments. Using data collected from a wide range of UK coastal waters, PSICAM accuracy was assessed by comparing both total non-water absorption and absorption by coloured dissolved organic material (CDOM) measured on discrete samples by two independent PSICAMs. AC-9/s performance was tested by comparing total non-water absorption measured in situ by an AC-9 and an AC-s mounted on the same frame. Results showed that the PSICAM outperforms AC-9/s instruments with regards to accuracy, with average spread in the PSICAM total absorption data of 0.006 m-1 (RMSE) compared to 0.028 m-1 for the AC-9/s devices. Despite application of a state of the art scattering correction method, the AC-9/s instruments still tend to overestimate absorption compared to PSICAM data by on average 0.014 m-1 RMSE (AC-s) and 0.043 m-1 RMSE (AC-9). This remaining discrepancy can be largely attributed to residual limitations in the correction of AC-9/s data for scattering effects and limitations in the quality of AC-9/s calibration measurements.

A Mie-based forward modeling procedure was developed to reconstruct bulk inherent optical properties (IOPs) from particle size distributions (PSDs) and real refractive index distributions (PRIDs) obtained using a previously developed flow cytometric (FC) method [Appl. Opt.57, 1705 (2018)APOPAI0003-693510.1364/AO.57.001705]. Given the available PSDs, extrapolations for the particle fraction outside the detection limits of the method and a complex refractive index input (with real part nr directly estimated and imaginary part ni adapted from the literature separately for organic and inorganic components), the model produces volume scattering functions that are integrated to produce scattering and backscattering coefficients, and absorption efficiencies that are used to calculate absorption coefficients. The procedure was applied to PSDs and PRIDs derived from natural samples retrieved in UK coastal waters and analyzed using a CytoSense flow cytometer (CytoBuoy b.v., The Netherlands). Optical closure analysis was carried out between reconstructed IOPs and in situ IOPs measured using an ac-9 spectrophotometer and a BB9 backscattering meter (WET Labs Inc., OR) in the same waters. The procedure is shown to achieve broad agreement with particulate scattering (bp) and backscattering (bbp) [root mean square percentage error (RMS%E): 35.3% and 44.5%, respectively) and to a lesser degree with backscattering ratio (bËbp) (RMS%E: 77%). The procedure, however, generally overestimated particulate absorption (ap) (RMS%E: 202.3%). This degree of closure was dependent on applying recently developed scattering error corrections to both absorption and attenuation in situ measurements. Not only do these results indirectly validate the FC method as a useful tool for PSD and PRID determination in natural particle populations, they also suggest that Mie theory may be a sufficient model for bulk IOP determination, with previously reported difficulties potentially being caused by inadequately corrected IOP measurements. Finally, in a feature unique to the FC method, the concurrent size and refractive index retrieval enabled assessment of the relative contributions that organic versus inorganic, fluorescent versus non-fluorescent fractions of the particle populations had on the IOPs, and identified which size classes had the largest influence on each of these properties.

A flow cytometric (FC) method was developed to retrieve particle size distributions (PSDs) and real refractive index (nr) information in natural waters. Geometry and signal response of the sensors within the flow cytometer (CytoSense, CytoBuoy b.v., Netherlands) were characterized to form a scattering inversion model based on Mie theory. The procedure produced a mesh of diameter and nr isolines where each particle is assigned the diameter and nr values of the closest node, producing PSDs and particle real refractive index distributions. The method was validated using polystyrene bead standards of known diameter and polydisperse suspensions of oil with known nr, and subsequently applied to natural samples collected across a broad range of UK shelf seas. FC PSDs were compared with independent PSDs produced from data of two LISST-100X instruments (type B and type C). PSD slopes and features were found to be consistent between the FC and the two LISST-100X instruments, but LISST concentrations were found in disagreement with FC concentrations and with each other. FC nr values were found to agree with expected refractive index values of typical marine particle components across all samples considered. The determination of particle size and refractive index distributions enabled by the FC method has potential to facilitate identification of the contribution of individual subpopulations to the bulk inherent optical properties and biogeochemical properties of the particle population.

In situ absorption measurements collected with a WET Labs ac-9 employing a reflective tube approach were scatter corrected using several possible methods and compared to reference measurements made by a PSICAM to assess performance. Overall, two correction methods performed best for the stations sampled: one using an empirical relationship between the ac-9 and PSICAM to derive the scattering error (ε) in the near-infrared (NIR), and one where ε was independently derived from concurrent measurements of the volume scattering function (VSF). Application of the VSF-based method may be more universally applicable, although difficult to routinely apply because of the lack of commercially available VSF instrumentation. The performance of the empirical approach is encouraging as it relies only on the ac meter measurement and may be readily applied to historical data, although there are inevitably some inherent assumptions about particle composition that hinder universal applicability. For even the best performing methods, residual errors of 20% or more were commonly observed for many water types. For clear ocean waters, a conventional baseline subtraction with the assumption of negligible near-IR absorption performed as well or better than the above methods because propagated uncertainties were lower than observed with the proportional method.

Measurements of the absorption coefficient of chromophoric dissolved organic matter (ay) are needed to validate existing ocean-color algorithms. In the surface open ocean, these measurements are challenging because of low ay values. Yet, existing global datasets demonstrate that ay could contribute between 30% to 50% of the total absorption budget in the 400-450 nm spectral range, thus making accurate measurement of ay essential to constrain these uncertainties. In this study, we present a simple way of determining ay using a commercially-available in-situ spectrophotometer operated in underway mode. The obtained ay values were validated using independent collocated measurements. The method is simple to implement, can provide measurements with very high spatio-temporal resolution, and has an accuracy of about 0.0004 m-1 and a precision of about 0.0025 m-1 when compared to independent data (at 440 nm). The only limitation for using this method at sea is that it relies on the availability of relatively large volumes of ultrapure water. Despite this limitation, the method can deliver the ay data needed for validating and assessing uncertainties in ocean-colour algorithms.

Long path length liquid waveguide capillary cell (LWCC) systems using simple spectrometers to determine the spectral absorption by colored dissolved organic matter (CDOM) have previously been shown to have better measurement sensitivity compared to high-end spectrophotometers using 10 cm cuvettes. Information on the magnitude of measurement uncertainties for LWCC systems, however, has remained scarce. Cross-comparison of three different LWCC systems with three different path lengths (50, 100, and 250 cm) and two different cladding materials enabled quantification of measurement precision and accuracy, revealing strong wavelength dependency in both parameters. Stable pumping of the sample through the capillary cell was found to improve measurement precision over measurements made with the sample kept stationary. Results from the 50 and 100 cm LWCC systems, with higher refractive index cladding, showed systematic artifacts including small but unphysical negative offsets and high-frequency spectral perturbations due to limited performance of the salinity correction. In comparison, the newer 250 cm LWCC with lower refractive index cladding returned small positive offsets that may be physically correct. After null correction of measurements at 700 nm, overall agreement of CDOM absorption data at 440 nm was found to be within 5% root mean square percentage error.

To investigate the genetics of late-onset myasthenia gravis (LOMG), we conducted a genome-wide association study imputation of>6 million single nucleotide polymorphisms (SNPs) in 532 LOMG cases (anti-acetylcholine receptor [AChR] antibody positive; onset age≥50 years) and 2,128 controls matched for sex and population substructure. The data confirm reported TNFRSF11A associations (rs4574025, P = 3.9 × 10-7, odds ratio [OR] 1.42) and identify a novel candidate gene, ZBTB10, achieving genome-wide significance (rs6998967, P = 8.9 × 10-10, OR 0.53). Several other SNPs showed suggestive significance including rs2476601 (P = 6.5 × 10-6, OR 1.62) encoding the PTPN22 R620W variant noted in early-onset myasthenia gravis (EOMG) and other autoimmune diseases. In contrast, EOMG-associated SNPs in TNIP1 showed no association in LOMG, nor did other loci suggested for EOMG. Many SNPs within the major histocompatibility complex (MHC) region showed strong associations in LOMG, but with smaller effect sizes than in EOMG (highest OR ~2 versus ~6 in EOMG). Moreover, the strongest associations were in opposite directions from EOMG, including an OR of 0.54 for DQA1*05:01 in LOMG (P = 5.9 × 10-12) versus 2.82 in EOMG (P = 3.86 × 10-45). Association and conditioning studies for the MHC region showed three distinct and largely independent association peaks for LOMG corresponding to (a) MHC class II (highest attenuation when conditioning on DQA1), (b) HLA-A and (c) MHC class III SNPs. Conditioning studies of human leukocyte antigen (HLA) amino acid residues also suggest potential functional correlates. Together, these findings emphasize the value of subgrouping myasthenia gravis patients for clinical and basic investigations and imply distinct predisposing mechanisms in LOMG.

Filter pad light absorption measurements are subject to two major sources of experimental uncertainty: the so-called pathlength amplification factor, ß, and scattering offsets, o, for which previous null-correction approaches are limited by recent observations of non-zero absorption in the near infrared (NIR). A new filter pad absorption correction method is presented here which uses linear regression against point-source integrating cavity absorption meter (PSICAM) absorption data to simultaneously resolve both ß and the scattering offset. The PSICAM has previously been shown to provide accurate absorption data, even in highly scattering waters. Comparisons of PSICAM and filter pad particulate absorption data reveal linear relationships that vary on a sample by sample basis. This regression approach provides significantly improved agreement with PSICAM data (3.2% RMS%E) than previously published filter pad absorption corrections. Results show that direct transmittance (T-method) filter pad absorption measurements perform effectively at the same level as more complex geometrical configurations based on integrating cavity measurements (IS-method and QFT-ICAM) because the linear regression correction compensates for the sensitivity to scattering errors in the T-method. This approach produces accurate filter pad particulate absorption data for wavelengths in the blue/UV and in the NIR where sensitivity issues with PSICAM measurements limit performance. The combination of the filter pad absorption and PSICAM is therefore recommended for generating full spectral, best quality particulate absorption data as it enables correction of multiple errors sources across both measurements.

Optical closure using radiative transfer simulations can be used to determine the consistency of in situ measurements of inherent optical properties (IOPs) and radiometry. Three scattering corrections are applied to in situ absorption and attenuation profile data for a range of coastal and oceanic waters, but are found to have only very limited impact on subsequent closure attempts for these stations. Best-fit regressions on log-transformed measured and modelled downwards irradiance, Ed, and upwards radiance, Lu, profiles have median slopes between 0.92 - 1.24, revealing a tendency to underestimate Ed and Lu with depth. This is only partly explained by non-inclusion of fluorescence emission from CDOM and chlorophyll in the simulations. There are several stations where multiple volume scattering function related data processing steps perform poorly which suggests the potential existence of unresolved features in the modelling of the angular distribution of scattered photons. General optical closure therefore remains problematic, even though there are many cases in the data set where the match between measured and modelled radiometric data is within 25% RMS%E. These results are significant for applications that rely on optical closure e.g. assimilating ocean colour data into coupled physical-ecosystem models.

AIM: Despite recognition that regular physical activity is essential for good health, many children do not accumulate sufficient daily physical activity. The aim of this study was to examine the effect of a classroom-based activity break on accelerometer-determined moderate-to-vigorous intensity physical activity (MVPA) and adiposity in primary school children. METHODS: One hundred twenty children from seven primary schools in Northern Ireland participated in the study. In each school, one class of children was randomly assigned to an intervention group and another class to a control group. Teachers of the intervention classes led a 5-min activity break three times per day for 12 weeks. Accelerometer-determined MVPA, height, weight and four skinfolds were measured at baseline and post-intervention. RESULTS: Compared with the control group, the intervention group significantly increased weekday MVPA (+9.5 min) from baseline to post-intervention. There were no significant changes in BMI; however, an increase in sum-of-skinfolds of the intervention group was observed. CONCLUSIONS: Classroom-based activity breaks led by the teacher are successful in increasing children's physical activity levels. The programme shows a positive step in improving overall physical activity levels and contributing to the goal of 60 min daily MVPA.

The BP09 experiment conducted by the Centre for Maritime Research and Experimentation in the Ligurian Sea in March 2009 provided paired vertical profiles of nadir-viewing radiances L(u)(z) and downward irradiances E(d)(z) and inherent optical properties (IOPs, absorption, scattering and backscattering coefficients). An inversion algorithm was implemented to retrieve IOPs from apparent optical properties (AOPs, radiance reflectance R(L), irradiance reflectance R(E) and diffuse attenuation coefficient K(d)) derived from the radiometric measurements. Then another inversion algorithm was developed to infer vertical profiles of water constituent concentrations, including chlorophyll-a concentration, non-algal particle concentration, and colored dissolved organic matter from the retrieved IOPs based on a bio-optical model. The algorithm was tested on a synthetic dataset and found to give reliable results with an accuracy better than 1%. When the algorithm was applied to the BP09 dataset it was found that good retrievals of IOPs could be obtained for sufficiently deep waters, i.e. for L(u)(z) and E(d)(z) measurements conducted to depths of 50 m or more. This requirement needs to be satisfied in order to obtain a good estimation of the backscattering coefficient. For such radiometric measurements a correlation of 0.88, 0.96 and 0.93 was found between retrieved and measured absorption, scattering and backscattering coefficients, respectively. A comparison between water constituent values derived from the measured IOPs and in-situ measured values, yielded a correlation of 0.80, 0.78, and 0.73 for chlorophyll-a concentration, non-algal particle concentration, and absorption coefficient of colored dissolved organic matter at 443 nm, respectively. This comparison indicates that adjustments to the bio-optical model are needed in order to obtain a better match between inferred and measured water constituent values in the Ligurian Sea using the methodology developed in this paper.

BACKGROUND: The emergence of influenza A(H1N1) 2009 was met with increased reports of associated neurological manifestations. We aimed to describe neurological manifestations of influenza in adults and children in the United Kingdom that presented at this time. METHODS: A 2-year surveillance study was undertaken through the British adult and pediatric neurological surveillance units from February 2011. Patients were included if they met clinical case definitions within 1 month of proven influenza infection. RESULTS: Twenty-five cases were identified: 21 (84%) in children and 4 (16%) in adults. Six (29%) children had preexisting neurological disorders. Polymerase chain reaction of respiratory secretions identified influenza A in 21 (81%; 20 of which [95%] were H1N1) and influenza B in 4 (15%). Twelve children had encephalopathy (1 with movement disorder), 8 had encephalitis, and 1 had meningoencephalitis. Two adults had encephalopathy with movement disorder, 1 had encephalitis, and 1 had Guillain-Barré syndrome. Seven individuals (6 children) had specific acute encephalopathy syndromes (4 acute necrotizing encephalopathy, 1 acute infantile encephalopathy predominantly affecting the frontal lobes, 1 hemorrhagic shock and encephalopathy, 1 acute hemorrhagic leukoencephalopathy). Twenty (80%) required intensive care, 17 (68%) had poor outcome, and 4 (16%) died. CONCLUSIONS: This surveillance study described a cohort of adults and children with neurological manifestations of influenza. The majority were due to H1N1. More children than adults were identified; many children had specific encephalopathy syndromes with poor outcomes. None had been vaccinated, although 8 (32%) had indications for this. A modified classification system is proposed based on our data and the increasing spectrum of recognized acute encephalopathy syndromes.

The light absorption coefficient of water is dependent on temperature and concentration of ions, i.e. the salinity in seawater. Accurate knowledge of the water absorption coefficient, a, and/or its temperature and salinity correction coefficients, Ψ(T) and Ψ(S), respectively, is essential for a wide range of optical applications. Values are available from published data only at specific narrow wavelength ranges or at single wavelengths in the visible and infrared regions. Ψ(T) and Ψ(S) were therefore spectrophotometrically measured throughout the visible, near, and short wavelength infrared spectral region (400 to ~2700 nm). Additionally, they were derived from more precise measurements with a point-source integrating-cavity absorption meter (PSICAM) for 400 to 700 nm. When combined with earlier measurements from the literature in the range of 2600 - 14000 nm (wavenumber: 3800 - 700 cm(-1)), the coefficients are provided for 400 to 14000 nm (wavenumber: 25000 to 700 cm(-1)).
