BACKGROUND: Cyclin D1 (CCND1) plays a pivotal role in cancer susceptibility and the platinum-based chemotherapy response. This study aims to assess the relationship between a common polymorphism (rs9344 G > A) in CCND1 gene with cancer susceptibility, platinum-based chemotherapy response, toxicities and prognosis of patients with lung cancer. METHODS: This study involved 498 lung cancer patients and 213 healthy controls. Among them, 467 patients received at least two cycles of platinum-based chemotherapy. Unconditional logistical regression analysis and meta-analysis were performed to evaluate the associations. RESULTS: The lung adenocarcinoma risk was significantly higher in patients with AA than GG + GA genotype (adjusted OR = 1.755, 95%CI = 1.057-2.912, P = 0.030). CCND1 rs9344 was significantly correlated with platinum-based therapy response in patients receiving PP regimen (additive model: adjusted OR = 1.926, 95%CI = 1.029-3.605, P = 0.040; recessive model: adjusted OR = 11.340, 95%CI = 1.428-90.100, P = 0.022) and in the ADC subgroups (recessive model: adjusted OR = 3.345, 95%CI = 1.276-8.765, P = 0.014). Furthermore, an increased risk of overall toxicity was found in NSCLC patients (additive model: adjusted OR = 1.395, 95%CI = 1.025-1.897, P = 0.034; recessive model: adjusted OR = 1.852, 95%CI = 1.088-3.152, P = 0.023), especially ADC subgroups (additive model: adjusted OR = 1.547, 95%CI = 1.015-2.359, P = 0.043; recessive model: adjusted OR = 2.030, 95%CI = 1.017-4.052, P = 0.045). Additionally, CCND1 rs9344 was associated with an increased risk of gastrointestinal toxicity in non-smokers (recessive model: adjusted OR = 2.620, 95%CI = 1.083-6.336, P = 0.035). Non-significant differences were observed in the 5-year overall survival rate between CCND1 rs9344 genotypes. A meta-analysis of 5432 cases and 6452 control samples did not find a significant association between lung cancer risk and CCND1 rs9344 polymorphism. CONCLUSION: This study suggests that in the Chinese population, CCND1 rs9344 could potentially serve as a candidate biomarker for cancer susceptibility and treatment outcomes in specific subgroups of patients.

The excavation and utilization of dormancy loci in breeding are effective endeavors for enhancing the resistance to pre-harvest sprouting (PHS) of wheat varieties. CH1539 is a wheat breeding line with high-level seed dormancy. To clarify the dormant loci carried by CH1539 and obtain linked molecular markers, in this study, a recombinant inbred line (RIL) population derived from the cross of weak dormant SY95-71 and strong dormant CH1539 was genotyped using the Wheat17K single-nucleotide polymorphism (SNP) array, and a high-density genetic map covering 21 chromosomes and consisting of 2437 SNP markers was constructed. Then, the germination percentage (GP) and germination index (GI) of the seeds from each RIL were estimated. Two QTLs for GP on chromosomes 5A and 6B, and four QTLs for GI on chromosomes 5A, 6B, 6D and 7A were identified. Among them, the QTL on chromosomes 6B controlling both GP and GI, temporarily named QGp/Gi.sxau-6B, is a major QTL for seed dormancy with the maximum phenotypic variance explained of 17.66~34.11%. One PCR-based diagnostic marker Ger6B-3 for QGp/Gi.sxau-6B was developed, and the genetic effect of QGp/Gi.sxau-6B on the RIL population and a set of wheat germplasm comprising 97 accessions was successfully confirmed. QGp/Gi.sxau-6B located in the 28.7~30.9 Mbp physical position is different from all the known dormancy loci on chromosomes 6B, and within the interval, there are 30 high-confidence annotated genes. Our results revealed a novel QTL QGp/Gi.sxau-6B whose CH1539 allele had a strong and broad effect on seed dormancy, which will be useful in further PHS-resistant wheat breeding.

We firstly report a high pulse repetition rate (101.4 MHz) nonlinear post-compression based on the normal dispersion fiber (NDF) operating in 2-µm wavelength region. With only one-stage NDF-based nonlinear pulse compressor, the 2-µm ultrafast laser pulses are compressed from ∼460 fs down to 70 fs, corresponding to ∼10.4 optical oscillation cycle. With two-stage nonlinear pulse compressor, the input ultrafast laser pulses are further compressed to 28.3 fs (∼4.3 optical oscillation cycle). In each case, the average power of the compressed 2-µm laser pulses exceeds 1 W, which is believed to be the highest average power never achieved at ∼100-MHz pulse repetition rate. The efficiencies of the one-stage and two-stage nonlinear pulse compressors are 64% and 47% respectively.

We have experimentally observed an ultrashort conventional vector soliton in an erbium-doped fiber laser. The few-layered graphene oxide (GO) is used as a saturable absorber (SA). It is found that the saturable absorption characteristic of GO is polarization independent. Therefore, vector solitons can be obtained without polarization control by using such SA. By using a polarization beam splitter to split the mode-locked pulse obtained in the oscillator, two orthogonal polarization vector solitons with equal intensity and consistent characteristics can be obtained. It demonstrates that the initial soliton consists of two orthogonal polarization components. It is worth noting that these two orthogonal polarization component solitons improve the signal-to-noise ratio (SNR) of 3 dB compared with the initial soliton. The improvement in SNR is very significant and cannot be neglected. This phenomenon has not been reported before, to our knowledge. In addition, the conventional soliton generated by this mode-locked laser has a central wavelength of 1559 nm with 1.1 ps pulse duration. The mode-locking state of this laser can be self-started. After mode locking, the environmental stability is excellent. The experimental results indicate that GO as a broadband SA has great potential and application prospects in the field of vector soliton generation.

Green infrastructure (GI) is used as an alternative and complement to traditional urban drainage system for mitigating urban stormwater issues mainly caused by climate change and urbanization. The combination of hydrological model and optimization algorithm can automatically find the optimal solution under multiple objectives. Given the multi-functional characteristics of GI, choosing the optimization objectives of GI are critical for multiple stakeholders. This study proposes a GI optimization method considering spatial functional zoning. Based on the basic conditions, the study area is divided into the flood risk control zone (FRCZ) and the total runoff control zone (TRCZ). The integrated model coupling hydrological model and optimization algorithm is applied to obtain the Pareto fronts and corresponding non-dominated solutions. The Technique for Order of Preference by Similarity to Ideal Solution (TOPSIS) method is used to support the decision-making process. The optimal solution obtained for the FRCZ achieves a flood risk reduction rate of 60.49% with an average life cycle cost per year of 0.20 × 108 Chinese Yuan (CNY); The optimal solution obtained for the TRCZ achieves a total runoff reduction rate of 22.83% with an average life cycle cost per year of 0.17 × 108 CNY. This study provides a reference for stakeholders in GI planning and design.

We present a tunable, high-energy optical parametric chirped pulse amplification system with a front-end based on a femtosecond Cr:ZnS laser. By taking advantage of the broad emission spectrum of the femtosecond Cr:ZnS master oscillator, we are able to directly seed the holmium-based pump around 2 µm. At the same time, the signal pulses for the parametric process are generated via Raman self-frequency shifting of the red end of the spectrum centered at 2.4 µm. The solitons, generated in a fluoride fiber, are tunable over the wavelength range between 2.8 and 3.2 µm. The optical parametric amplifier operates at a 1 kHz repetition rate, and consists of two stages equipped with ZGP as nonlinear crystal. The generated idler pulses are tunable between 5.4 and 6.8 µm with a pulse energy of up to 400 µJ. Dispersion management using bulk material stretching and compression in combination with precise phase shaping prior to amplification enables idler pulses of a sub-100 fs duration, translating into a peak power as high as 4 GW.

A compact mid-wavelength infrared (MWIR) optical parametric chirped pulse amplification (OPCPA) system generates sub-150 fs pulses at wavelengths from 5.4 to 6.8 µm with >400µJ energy at a 1 kHz repetition rate. A femtosecond Cr:ZnS master oscillator emitting 40 fs pulses at 2.4 µm seeds both a Ho:YLF regenerative amplifier and a two-stage OPCPA based on ZnGeP2 crystals. The 2.05 µm few-picosecond pump pulses from the Ho:YLF amplifier have an energy of 13.4 mJ. Seed pulses for the OPCPA are generated by soliton self-frequency shifting in a fluoride fiber and are tunable between 2.8 and 3.25 µm with a sub-100 fs duration and few-nanojoule energy. The intense MWIR pulses hold strong potential for applications in ultrafast mid-infrared nonlinear optics and spectroscopy.

In this paper, a reverse-strip AlGaAs waveguide with three zero-dispersion wavelengths (ZDWs) is designed. The corresponding three ZDWs are located at 3.74, 6.56, and 8.89 µm. The nonlinearity coefficient of the proposed reverse-strip AlGaAs waveguide is calculated as 2.09W-1m-1 at wavelength 4.9 µm. The effects of pump pulse parameters, waveguide length, and noise coefficient on the nonlinear dynamics of supercontinuum (SC) generation are investigated. When the hyperbolic secant pump pulse with a wavelength of 4.9 µm, peak power of 900 W, and duration of 100 fs is launched into the proposed waveguide and propagated after a 3 mm length, highly coherent and multi-octave mid-infrared (MIR) SC spanning from 2.2 to 14.5 µm (more than 2.7 octaves, at -40dB level) is generated. Finally, a possible fabrication process of the reverse-strip AlGaAs waveguide is introduced. Our research results have important applications in MIR photonics, MIR spectroscopy, optical precision measurement, and more.

Signaling by the phytohormone abscisic acid (ABA) involves pre-mRNA splicing, a key process of post-transcriptional regulation of gene expression. However, the regulatory mechanism of alternative pre-mRNA splicing in ABA signaling remains largely unknown. We previously identified a pentatricopeptide repeat protein SOAR1 (suppressor of the ABAR-overexpressor 1) as a crucial player downstream of ABAR (putative ABA receptor) in ABA signaling. In this study, we identified a SOAR1 interaction partner USB1, which is an exoribonuclease catalyzing U6 production for spliceosome assembly. We reveal that together USB1 and SOAR1 negatively regulate ABA signaling in early seedling development. USB1 and SOAR1 are both required for the splicing of transcripts of numerous genes, including those involved in ABA signaling pathways, suggesting that USB1 and SOAR1 collaborate to regulate ABA signaling by affecting spliceosome assembly. These findings provide important new insights into the mechanistic control of alternative pre-mRNA splicing in the regulation of ABA-mediated plant responses to environmental cues.

Rapid urbanization has dramatically increased the impermeable surface in urban area, which led to urban severe flooding and waterlogging in the world, especially in China and southeast Asia. There are more than 100 cities that suffered from urban flood every year since 2006, and more than 100 million citizens are involved in China. Urban flood mitigation is one of the most critical issues for both water administration and city management agency, in which urban flood modeling is vital and necessary. Whereas, there are relatively few data of waterlogging and runoff in urban area during flooding episodes to calibrate and validate the models, for there are usually few observation facilities installed in the cities. This paper used a combination of experiment and modelling to overcome the lack of reliable discharge data and be able to characterize the urban flooding problems in Xiamen Island, China. This paper simulated the urban flooding in Xiamen Island based on a hydrodynamic model coupled with hydrological model. The datasets of underlying surfaces were input to the model, including the terrain data, building plan, land use, etc. The uncertainty of the urban flood model was analyzed based on the generalized likelihood uncertainty estimation (GLUE) method with shuffled complex evolution Metropolis (SCEM-UA) sampling algorithm. The key parameters were evaluated by on-site experiment to reduce the uncertainties of the model, which could improve the accuracy of the model. If using the recommended parameter value range, the average relative error of flood depth was less than 27.2% at 90% confidence level. A typical rain pattern of 50 years return event was used for flood simulation. The results show that the main inundated areas (flooded depth more than 40 cm) are located in three groups: southeast to the Yundang Lake, around the Hubian Reservoir, along the Exhibition Road. The other inundated areas that less than 40 cm deep are scattered in some low-lying land of Xiamen Island. The main inundated areas simulated are consistent with the point survey of urban flooding, which verifies that the suggested model and the on-site experiment is effective and reliable for urban flood prediction.

The translation initiation factor eIF4E-binding protein-mediated regulation of protein translation by interfering with assembly of mRNA cap-binding complex eIF4F is well described in mammalian and yeast cells. However, it remains unknown whether a signaling regulator or pathway interacts directly with any translation initiation factor to modulate assembly of eIF4F in plant cells. Here, we report that the two isoforms of Arabidopsis eIF4G, eIFiso4G1 and eIFiso4G2, interact with a cytoplasmic-nuclear dual-localized pentatricopeptide repeat protein SOAR1 to regulate abscisic acid (ABA) signaling. SOAR1 inhibits interactions of eIFiso4E, eIF4Es, eIF4A1, eIF4B2 and poly(A)-binding protein PAB6 with eIFiso4G1 and eIFiso4G2, thereby inhibiting eIFiso4F/mixed eIF4F assembly and repressing translation initiation. SOAR1 binds mRNA of a key ABA-responsive gene ABI5 and cooperates with eIFiso4G1/2 to repress translation of ABI5. The binding of SOAR1 to ABI5 mRNA is likely to inhibit the interaction of SOAR1 with eIFiso4G1/2, suggesting a regulatory loop. Our findings identify a novel translation initiation repressor interfering with cap-binding complex assembly, and establish a link between cap-binding complex assembly and ABA signaling.

In this paper, we design a polarization-maintaining CS2-core photonic crystal fiber (PM-CCPCF). The two air holes in the x direction are infiltrated with C2H5OH in order to introduce birefringence. By optimizing the structure parameters of the PM-CCPCF, it is demonstrated that the x-polarization fundamental mode has an all-normal dispersion profile and the corresponding y-polarization fundamental mode has an anomalous dispersion profile for a pump wavelength of 1.76 µm. Then, we investigate the supercontinuum (SC) generations when different fiber lengths, pump peak powers, and pump pulse widths are chosen, respectively. Simulation results show that for the x-polarization and y-polarization fundamental modes, highly coherent SCs can be generated by appropriately choosing the fiber length and pump pulse parameters. Finally, nonlinear propagation dynamics are analysed when the optimized fiber length and pump pulse parameters are used. The bandwidth of the SCs generated for the x-polarization and y-polarization fundamental modes can be up to 0.82 and 1.26 octave, respectively.

In this paper, we numerically investigate the mid-infrared supercontinuum (SC) generation in a suspended ${{\rm As}_2}{{\rm Se}_3}$As2Se3 ridge waveguide, which is designed with the two zero-dispersion wavelengths. Simulation results show that when the pump pulses at wavelength 3.3 µm with width of 100 fs and peak power of 900 W are launched into the anomalous dispersion region of the designed waveguide with a length of 0.87 mm, the SC can be generated in the wavelength range from 1.76 to 14.42 µm (more than three octaves), extending deep into the "fingerprint" region. The stability of the generated SC is confirmed by the first-order coherence. Moreover, we demonstrate the performance of the SC-based frequency comb by assuming a 50 pulse pump source at a repetition rate of 100 MHz.

RESUMO

Although the concept of body image was first introduced to Taiwan nursing in 1975, its application in the context of clinical nursing with regard to promoting humanistic patient-centered care has yet to be examined. This paper adopts a field-study approach to reinterpret the concept of body image with deeper and broader strength using patient vignettes, nursing case studies, and the findings of prior nursing research. The four main issues covered in this paper are: 1. The body image concept, 2. Building body schema and a postural model based on body experiences, 3. The characteristics of body image, and 4. The extended meaning of body image, including self-concept and the role function of relationship. This paper is expected to help nurses better understand the concept of body image and further apply this concept in clinical practice in order to improve the provision of humanistic patient-centered care.

To investigate the active fraction from Bletilla striata in Caco-2 cell monolayer,so as to explore its absorption mechanism of oral administration preliminarily.Active fraction from B.striata in Caco-2 cell monolayer was analyzed by UPLC-Q-TOF and detected by UPLC-MS/MS,and the effects of different concentrations,pH and P-glycoprotein inhibitors on Caco-2 cells Monolayer were investigated.Six compounds were isolated from the active fraction of B.striata in Caco-2 cell monolayer by UPLC-Q-TOF,and identified as B6,B12,B14,B17,B19 and B23,with concentration dependence.Within the 0-180 min,the uptake of B12 and B14 had a time dependence,while B6,B17,B19 and B23 tended to saturate after 60 min.All of the components had a good absorption in an acidic environment.B6 had a good absorption at pH 6.0,while the other components B12,B14,B17,B19 and B23 had a good absorption at pH4.0.The absorption of the 6 main components of B.striata were not be affected by P-glycoprotein inhibitors(verapamil/cyclosporin A).Compared with the control group,there was no difference in the absorption of B6 and B12,and the absorption of B14,B17,B19 and B23 increased,but with no significant difference.The absorption characteristic of B.striata extract across the Caco-2 cell monolayer is probably passive diffusion,and the absorption process was not affected by P-glycoprotein.

Platinum (Pt)-based nanostructures are the most efficient catalysts for the oxygen reduction reaction (ORR) in acid media. Here, Pt9 Ni wavelike nanowires (W-NWs) have been synthesized by etching Pt3 Ni@PtNi2 core-shell nanowires with 2,5-dihydroxyterephthalic acid for 24 hours. Compared to the commercial Pt/C catalyst, the free-standing Pt9 Ni W-NWs show improvements of up to 9.3 times for mass activity and 12.6 times for specific activity, respectively.

In this Letter, we demonstrate experimentally for the first time, to the best of our knowledge, discrete ultraviolet (UV) wavelength generation by cascaded intermodal FWM when femtosecond pump pulses at 800 nm are launched into the deeply normal dispersion region of the fundamental guided mode of a multimode photonic crystal fiber (MPCF). For pump pulses at average input powers of Pav=450, 550, and 650 mW, the first anti-Stokes waves are generated at the visible wavelength of 538.1 nm through intermodal phase matching between the fundamental and second-order guided mode of the MPCF. The first anti-Stokes waves generated then serve as the secondary pump for the next intermodal FWM process. The second anti-Stokes waves in the form of the third-order guided mode are generated at the UV wavelength of 375.8 nm. The maximum output power is above 10 mW for Pav=650 mW. We also confirm that the influences of fiber bending and intermodal walk-offs on the cascaded intermodal FWM-based frequency conversion process are negligible.

Whereas several mitochondrial/chloroplast pentatricopeptide repeat (PPR) proteins have been reported to regulate plant responses to abiotic stresses, no nucleus-localized PPR protein has been found to play role in these processes. In the present experiment, we provide evidence that a cytosol-nucleus dual-localized PPR protein SOAR1, functioning to negatively regulate abscisic acid (ABA) signaling in seed germination and postgermination growth, is a crucial, positive regulator of plant response to abiotic stresses. Downregulation of SOAR1 expression reduces, but upregulation of SOAR1 expression enhances, ABA sensitivity in ABA-induced promotion of stomatal closure and inhibition of stomatal opening, and plant tolerance to multiple, major abiotic stresses including drought, high salinity and low temperature. Interestingly and importantly, the SOAR1-overexpression lines display strong abilities to tolerate drought, salt and cold stresses, with surprisingly high resistance to salt stress in germination and postgermination growth of seeds that are able to potentially germinate in seawater, while no negative effect on plant growth and development was observed. So, the SOAR1 gene is likely useful for improvement of crops by transgenic manipulation to enhance crop productivity in stressful conditions. Further experimental data suggest that SOAR1 likely regulates plant stress responses at least partly by integrating ABA-dependent and independent signaling pathways, which is different from the ABI2/ABI1 type 2C protein phosphatase-mediated ABA signaling. These findings help to understand highly complicated stress and ABA signalling network.

A dominant suppressor of the ABAR overexpressor, soar1-1D, from CHLH/ABAR [coding for Mg-chelatase H subunit/putative abscisic acid (ABA) receptor (ABAR)] overexpression lines was screened to explore the mechanism of the ABAR-mediated ABA signalling. The SOAR1 gene encodes a pentatricopeptide repeat (PPR) protein which localizes to both the cytosol and nucleus. Down-regulation of SOAR1 strongly enhances, but up-regulation of SOAR1 almost completely impairs, ABA responses, revealing that SOAR1 is a critical, negative, regulator of ABA signalling. Further genetic evidence supports that SOAR1 functions downstream of ABAR and probably upstream of an ABA-responsive transcription factor ABI5. Changes in the SOAR1 expression alter expression of a subset of ABA-responsive genes including ABI5. These findings provide important information to elucidate further the functional mechanism of PPR proteins and the complicated ABA signalling network.

AIM: Myeloperoxidase (MPO) and glutathione S-transferase pi 1 (GSTP1) are important carcinogen-metabolizing enzymes. The aim of this study was to investigate the association between the common polymorphisms of MPO and GSTP1 genes and lung cancer risk in Chinese Han population. METHODS: A total of 266 subjects with lung cancer and 307 controls without personal history of the disease were recruited in this case control study. The tagSNPs approach was used to assess the common polymorphisms of MOP and GSTP1 genes and lung cancer risk according to the disequilibrium information from the HapMap project. The tagSNP rs7208693 was selected as the polymorphism site for MPO, while the haplotype-tagging SNPs rs1695, rs4891, rs762803 and rs749174 were selected as the polymorphism sites for GSTP1. The gene polymorphisms were confirmed using real-time PCR, cloning and sequencing. RESULTS: The four GSTP1 haplotype-tagging SNPs rs1695, rs4891, rs762803 and rs749174, but not the MPO tagSNP rs7208693, exhibited an association with lung cancer susceptibility in smokers in the overall population and in the studied subgroups. When Phase 2 software was used to reconstruct the haplotype for GSTP1, the haplotype CACA (rs749174+rs1695 + rs762803+rs4891) exhibited an increased risk of lung cancer among smokers (adjust odds ratio 1.53; 95%CI 1.04-2.25, P=0.033). Furthermore, diplotype analyses demonstrated that the significant association between the risk haplotype and lung cancer. The risk haplotypes co-segregated with one or more biologically functional polymorphisms and corresponded to a recessive inheritance model. CONCLUSION: The common polymorphisms of the GSTP1 gene may be the candidates for SNP markers for lung cancer susceptibility in Chinese Han population.
