RESUMO

As ubiquitous pathogens, bovine virus diarrhea viruses (BVDVs) in cattle have been reported several times in Turkey. Over time, the frequency and importance of this infection has increased for the livestock industries. A total of 1291 animals were sampled from a dairy herd in Turkey suspected of BVDV clinical signs, for instance, reproductive failures (abortion, congenital malformations in calves, repeat breeding, etc.) and interdigital phlegmon in adult animals. Reverse transcription polymerase chain reactions (RT-PCRs) were made by using targeted 5' untranslated region (UTR), Npro, E2, and NS2-3 pestiviral gene region primers for antigen ELISA-positive samples (n = 20). The obtained amplicons were sequenced. Sequence results showed the presence of a new subgroup in Pestivirus A species. This paper describes the nucleotide sequences of a new BVDV 1 (BVDV 1-v) subgroup member.

RESUMO

To determine why serum from small ruminants infected with ruminant pestiviruses reacted positively to classical swine fever virus (CSFV)-specific diagnostic tests, we analyzed 2 pestiviruses from Turkey. They differed genetically and antigenically from known Pestivirus species and were closely related to CSFV. Cross-reactions would interfere with classical swine fever diagnosis in pigs.

Feline leukaemia virus (FeLV) is a member of the Gammaretrovirus genus, which has two genotypes in cats: endogenous (replication-defective provirus) and exogenous (replication-competent). In this study, 550 cats were examined, and 112 of them (20.36%) were found to have the endogenous FeLV (enFeLV) genotype. EnFeLV-positive animals were also tested for additional viral infections, and 48 cats (42.85%) were discovered to be co-infected with other viruses. According to co-infection data, these cats were infected with feline coronavirus (FCoV) (27/112, 24.1%), feline panleukopenia virus (FPV) (14/112, 12.5%), feline immunodeficiency virus (FIV) (0/112, 0%), and domestic cat hepadnavirus (DCH) (13/112, 11.6%). Their age, sex, breed, clinical state, lifestyle (in/outdoor), and immunization data against FeLV were also evaluated. In line with our results, the prevalence of enFeLV and co-infection with other pathogens in cats admitted to the clinic for various reasons were discussed. The majority of positive animals in terms of FeLV (94/112, 83.93%) had clinical findings. We emphasized that the FeLV-positive situation of cats should be taken into consideration by veterinarians when planning treatment and vaccination programs. Additionally, in this study, we questioned the group in which our enFeLVs were phylogenetically located. Therefore, we performed a phylogenetic analysis based on a comparison with global FeLV sequences obtained from the GenBank database. The sequenced positive samples were in the AGTT subgroup within Group-II.

RESUMO

We report here the complete genome sequence of pestivirus strain Aydin/04-TR, which is the prototype of a group of similar viruses currently present in sheep and goats in Turkey. Sequence data from this virus showed that it clusters separately from the established and previously proposed tentative pestivirus species.

Recent worldwide serological and genetic studies of small ruminant lentiviruses (SRLV) have led to the description of new genotypes and the development of new diagnostic tests. This study investigated the detection and molecular characterization of visna/maedi virus (VMV) infection in serum and blood samples from pure and mixed sheep breeds acquired from different regions in Turkey using ELISA and PCR techniques. The prevalence of VMV was 67.8 % by ELISA and/or LTR-PCR with both assays showing a medium level of agreement (kappa: 0.26; ± 0.038 CI). Positivity of VMV in sheep increased according to the age of the animal, although PCR positivity was higher than ELISA in young individuals. Phylogenetic analysis of 33 LTR sequences identified two distinct clades that were closely related to American and Greek LTR sequences. Phylogenetic analysis of 10 partial gag gene sequences identified A2, A3, A5, A9, A11 subtypes of genotype A SRLVs. In vitro culture of all isolates in fetal sheep lung cells (FSLC) showed a slow/low phenotype causing less or no lytic infection compared with infection with the WLC-1 American strain characterized by a rapid/highly lytic phenotype. Phylogenetic analysis revealed that Turkish VMV sequences preceded the establishment of American or Greek strains that were associated with the migration of sheep from the Middle East to Western Europe several centuries ago. This is the first study that describes Turkish VMV sequences with the molecular characterization of LTR and gag genes, and it strongly suggests that SRLV-genotype A originated in Turkey.

RESUMO

Cases of orf virus infection in human in Turkey have been reported for many years. Scab material from a man was found positive by PCR using pan-parapox-specific primers for parapoxvirus infection. The amplicon was purified and sequenced. The present study provides for the first time a phylogenetic analysis of parapoxviruses from Turkey. The partial B2L gene sequence of a Turkish orf virus from a human presented here may be useful for characterization of parapoxvirus infections in Turkey based on the phylogenetic analysis studies.

BACKGROUND: Domestic cat hepadnaviruses (DCHs) have been described as a novel virus that can infect cats. OBJECTIVE: The aim of our study is the first identification and molecular characterizations of DCH infection in Turkish domestic cats. METHODS: The blood, organ and ascites fluid samples from 550 cats were randomly sampled. The presence of DCH nucleic acid was investigated by using both in the literature and newly designed primers. RESULTS: It was found that the hepadnavirus positivity rate is 4% (22/550) in Türkiye. The full genomic characterization was performed on 13 of 22 samples, and others were characterized as nearly full genome. In this study, we highlight that whole blood samples should be also screened for DCH, not only serum samples as has frequently been done in other studies. DCH-infected cats were also found positive (54.54%, 12/22) for Feline leukaemia virus infection. BLAST results revealed that Turkish DCHs have 86.32%-99.08% homology with strains in the GenBank database, enabling us to construct phylogenetic trees. CONCLUSIONS: According to this study's results, it is suggested that this infection should be added to veterinary diagnostic panels worldwide. Additionally, we suggest that our new synthesized primers for the amplification of X gene can also be used for diagnosis.

The rabies infection is a zoonotic viral disease in humans and is spread by both wild and domestic carnivores. This study aimed to molecularly characterize the field strains of the rabies virus circulating in Turkey between 2013 and 2020. Brain samples obtained from 16 infected animals (eight cattle, one donkey, three foxes, three dogs, and one marten) were tested. Full nucleoprotein (N) and glycoprotein (G) gene sequences were used to determine the genetic and antigenic characteristics of the rabies virus field strains. The phylogenetic analyses revealed that the 16 field strains identified in Turkey belonged to the Cosmopolitan lineage.

The aim of this work was to investigate the molecular characteristics of bovine leukemia viruses (BLVs) in Turkey. The variability of env and gag fragments of BLVs was examined using DNA from blood samples obtained for sequence analysis of BLVs in four cattle herds from three different geographical areas in Turkey. The env gene sequences were highly similar to those of Brasilian, Argentine, and Japanese BLV strains, while gag genes from Turkish BLV isolates showed greatest similarity to those of Iranian isolates. This paper is the first report on the partial characterisation of env and gag genetic fragments of BLVs from Turkey.

Papillomaviruses are oncogenic DNA viruses and induce hyperplastic benign lesions of both cutaneous and mucosal tissues in their various hosts, including many domestic and wild animals as well as humans. There are some Papillomavirus genotypes that can infect hosts different from their own, such as BPV 1 and BPV 2 originated from cattle, which can also infect horses and are responsible for fibroblastic tumours in horses. This review article summarizes the origin and evolution of papillomaviruses as an etiological agent in the historical process. The main focus in this review is the evaluation of the interactions between high-risk papillomavirus oncoproteins and programmed cell-death pathways. It further exemplifies the role of these interactions in the malignant cell transformation process. In parallel with this, the use and importance of the bovine model system to enlighten the papillomavirus-associated cancers is discussed with an in-depth examination. Furthermore, it focuses on the epidemiological situation of BPV infections in Turkey in the cattle herds.

Human adenoviruses (HAdVs) are responsible for various clinical diseases. Molecular epidemiological studies of respiratory HAdVs are limited in Turkey. To determine the main genotypes and epidemiological characteristics of HAdVs in patients with respiratory symptoms. HAdV PCR-positive extracts of nasal/nasopharyngeal specimens sent to the Turkish Public Health Institution from various cities of Turkey in 2015-2016 were investigated by seminested PCR. Partial sequence analysis of the hexon gene of HAdVs was performed. SPSSv.24.0 was used. A total of 23/68 (33.82%) HAdV-positive samples were amplified. Mastadenovirus B, C, D, and F were detected and mastadenovirus B (10/23; 43.5%) and C (10/23; 43.5%) were predominant strains. Interestingly, HAdV-F known to have gastrointestinal system tropism was detected in two patients with respiratory symptoms. HAdV-B3 was the most prevalent genotype (9/23; 39.1%). Also, HAdV-B7 is defined as a reemerging pathogen. It is noteworthy that there is a cluster of four HAdV-C strains showing a close paraphyletic relationship with HAdV-2/6 intertypic recombination. To our knowledge, this is the first study showing that HAdV-B7 reemerging pathogen circulating in patients with respiratory infections in our country. It is also necessary to emphasize that HAdV-2/6 recombinant strains were detected in this study for the first time in Turkey.

In this study, strains of feline immunodeficiency virus (FIV), designated TR-D, TR-Mo and TR-Mi, isolated from three cats in Turkey, were characterized. PCR products (859 bp) from the envelope (env) gene region were amplified and sequenced, and possible geographical differences in the env gene region of Turkish FIV strains are discussed. Phylogenetic analysis of two strains showed that FIV subtype B was present in Turkey. Phylogenetic analysis showed that one new Turkish FIV strain occupies a separate branch from known clusters (subtypes A to E) from the USA, Canada, Europe and Japan.

Five BVDV species 2 (BVDV-2) isolates were detected from cattle in Turkey. Phylogenetic analysis was performed based on the 5' untranslated regions (UTRs) and E2 coding gene regions, respectively. The isolates were closely related to BVDV-2a strains from North America and Canada used as references. This is the first report of the detection of BVDV-2 in naturally infected Turkish cattle. It is important to consider BVDV-2 for planning future BVDV control and vaccination programs in Turkey.

In this study a mink showing hard pad disease like symptoms was euthanised. Heart blood and various tissue samples collected during necroscopy and tested by specific RT-PCR were found positive for CDV. H and F gene segments of the CDV strain was also partially sequenced using the appropriate primers, and subsequently the sequences were analysed and compared with same gene fragment sequence of other CDV isolates from different countries. The results of the phylogenetic analysis showed that the Turkish-Mink distemper strain is closely related to European CDV strains of lineage 1. Additionally, the distemper antigen was also detected when the tissue samples were examined by histology or immunohistochemistry.

In this study, 26 blood samples were collected from 25 healthy cats and one cat with clinical signs suggestive of feline infectious peritonitis (FIP), namely, fever, weight loss, enlarged abdomen, and ascites. Blood samples were tested for feline coronavirus (FCoV) messenger RNA (mRNA) by an reverse transcriptase-polymerase chain reaction (RT-PCR) assay which has previously been described to have a high specificity in the diagnosis of clinical FIP [Simons AF, Vennema H, Rofina JE, Pol JM, Horzinek MC, Rottier PJM, Egberink HF (2005) A mRNA PCR for the diagnosis of feline infectious peritonitis. Journal of Virological Methods124, 111-116]. Overall we found 14 (54%) of the cats were positive for FCoV including the cat with clinical disease, but the high rate of positivity among healthy cats suggested a poor specificity for the clinical diagnosis of FIP among these cats. It was observed that the positivity rate was highest in cats aged between 6 months-1 year old. Our findings suggest that FCoVs may be present in the blood samples from healthy cats as well as cats with clinical FIP.

Papillomavirus (PV) infections often cause benign and malignant skin neoplasia in dogs. To date, twenty types of canine papillomaviruses (CPVs) have been described worldwide. A detailed molecular characterization of CPVs in Turkey is lacking. In the present study, oral and mucosal lesions from 13 dogs with suspected CPV infection from the Mediterranean and central Anatolian regions of Turkey were analyzed. The partial gene sequences of the L1, E6, and E7 regions were compared with those of CPV types in the GenBank database. The results showed that CPV-1 infection was the dominant type of canine papillomatosis in Turkey. In addition, there was no statistically significant association between the frequency of the disease and the age or gender of the dog (p>0.05). However, all the dogs were pedigree breeds, suggesting that the disease may be more prevalent among pure-bred dogs than mixed breeds.

Pseudocowpox virus (PCPV) infects cattle throughout the world and has zoonotic potential. However, it is not known to infect cattle in Turkey. In August 2013, we observed ulcerative nodular swelling and pustules on udder and teats of a cow in a small village nearly Lake of Bafa, Milas, Mugla locate in southwestern part of Turkey. Interestingly, the similar lesions were also observed on skin of milkier women's hand at the same time. A PCPV strain was characterized based on the major envelop gene sequence. The phylogenetic analysis showed that the isolated strain was closely related to the members of other parapoxvirus genus. This study provides the first description of PCPV infection in Turkey.

Equine adenovirus type 1 (EAdV-1) is a cause of repiratory tract infection in equids. In present study for the first time in Turkey, the prevalence of EAdV-1 in nasal swab samples obtained from horses showing respiratory symptoms was investigated by polymerase chain reaction (PCR), and molecular characterization of the hexon gene detected in the Turkish (TR) strain was performed. Overall, the prevalence of EAdV-1 was found low (1.4%) as indicated by a positive PCR reaction from the nasal swab extracts tested. Phylogenetic analysis based on the partial sequences of the hexon gene of a TR-EAdV-1 strain with those of previously isolated AdVs from different mammals and an EAdV-1 M1 strain showed that the EAdV-1 strains were placed into a unique cluster. Although the TR-EAdV-1 strain was closely related to CAV-1, CAV-2 and bat adenovirus reference strains, larger-scale studies are necessary to better understand the molecular epidemiology and population structure of EAdV-1 in Turkey.
