RESUMO

The objectives were to evaluate the effects of injectable vitamin E during the last 3 wk prepartum on the incidence of retained fetal membranes (RFM) and reproductive performance. Dairy cows (n=890), 390 Holsteins (132 nulliparous and 258 parous) and 500 crossbred Holstein × Gyr (199 nulliparous and 301 parous), from 3 dairy farms in Brazil were assigned to the study. In all 3 farms, from October to March, prepartum cows grazed tropical grasses and received 2 kg/d of a mixture of finely ground corn, soybean meal, and minerals and vitamins. From April to September prepartum cows received a total mixed ration composed of corn silage, finely ground corn, soybean meal, and minerals and vitamins. During the prepartum period, cows were fed 280 (farm 1), 390 (farm 2), and 480 IU (farm 3) of supplemental vitamin E per day, and throughout postpartum, cows were fed 370 (farm 1), 500 (farm 2), and 600 (farm 3) IU of supplemental vitamin E. Within each farm, cows were randomly assigned to remain as untreated controls or to receive 3 i.m. injections of 1,000 IU each of dl-α-tocopherol administered at 19.2 ± 4.3, 12.9 ± 3.3, and 6.2 ± 2.9 d before calving (VitE). Blood was sampled from 141 cows immediately before enrollment to determine the α-tocopherol and cholesterol statuses. Blood was also sampled and analyzed for concentrations of cortisol and nonesterified fatty acids in the last 3 wk of gestation. The serum concentration of α-tocopherol or α-tocopherol:cholesterol ratio did not differ between treatments and averaged 2.97 ± 0.10 µg/mL and 4.46 ± 0.16 × 10(-3), respectively. In total, 53.2% of the cows had an inadequate concentration of serum α-tocopherol based on the 3.0 µg/mL cut-off for adequacy. The risk of RFM decreased as serum α-tocopherol increased. Milk production did not differ between controls and VitE cows. Treatment with injectable α-tocopherol decreased RFM from 20.1 to 13.5%, decreased incidence of stillbirth from 14.9 to 6.8%, and tended to decrease death by 200 d postpartum. VitE cows tended to have improved pregnancy per insemination at first AI (36.7 vs. 30.1%) because of decreased pregnancy loss from 31 to 62 d of gestation (12.5 vs. 20.5%). Despite a similar insemination rate, VitE cows had 22% greater pregnancy rate than control cows. Cows receiving vitamin E had decreased circulating cortisol and nonesterified fatty acids around calving. In summary, when cows were fed limited amounts of supplemental vitamin E, 28 to 48% of the recommendations, prepartum supplementation with injectable α-tocopherol decreased incidence of RFM and improved reproduction.

The existence of a pheromone emitted during copulation has been reported for Rhodnius prolixus (Hemiptera: Reduviidae) Stål. Adults possess one pair of metasternal glands (MGs) from which female R. prolixus release volatiles mainly at night. We investigated whether these volatiles emitted by adult R. prolixus can modulate sexual-related behaviours of opposite and/or same sex individuals. We first used a shelter bioassay to test if adult activity patterns can be affected by chemical signals emitted by opposite sex conspecifics. We observed that males left the shelter more frequently in the presence of females, showing higher activity and an increase of intrasexual copulation attempts. Females showed no shelter-related activation in the presence of male odours. Second, we used a locomotion compensator device to investigate whether females or males show oriented responses to odours emitted by adults. We found that males oriented towards air currents carrying female odours but females did not orient towards odours emitted by adults. Finally, we observed that males oriented towards the female MG compounds. Thus, a volatile chemical signal emitted by females from their MGs promoted the activation of sheltered males and modulated orientation to air currents.

We investigated whether metasternal gland (MG) products mediate male aggregation during mating in Rhodnius prolixus (Heteroptera: Reduviidae) Stål, 1859, through selective occlusion of female MGs. Pairs of insects presenting intact MGs mated and other males aggregated around them confirming conclusions from previous studies. However, when female MGs were occluded, males did not aggregate. Conversely, the occlusion of male MGs did not affect aggregation of males. The present experiments demonstrated that the products emitted by the MGs of a mating R. prolixus female induce other males to aggregate around the mating pair. Aggregation behaviour of males probably results from the eventual release of volatiles from the female's MGs. We reinforce the hypothesis suggesting that the compounds produced by the MGs of R. prolixus act as sexual communication signals for this species.

The present study investigated the association between mannose-binding lectin (MBL) gene polymorphism and the susceptibility to human T-cell lymphotropic virus (HTLV) infection in a group of 83 HTLV-infected asymptomatic subjects (62 HTLV-1 and 21 HTLV-2) and 99 healthy controls. Detection of MBL*A, MBL*B, and MBL*C was performed by amplifying a fragment of 349 bp (exon 1) and submitting the product to restriction fragment length polymorphism analysis with BanI and MboII endonucleases. Allele MBL*D was investigated by sequence-specific primer-polymerase chain reaction. The frequency of MBL*A, MBL*B, and MBL*D was 63%, 22%, and 15% among seropositive subjects and 70%, 14%, and 16% among healthy controls, respectively. Genotype differences were statistically significant (chi2 = 11.57; p = 0.04); the presence of genotype BB was 9.6% among HTLV-infected patients compared with 1% among controls (chi2 = 7.151; p = 0.019). A significant difference of the genotype frequencies between HTLV-1 and HTLV-2 infections was observed, but this result could be attributed to the number of investigated HTLV-1-infected subjects. The odds ratio to the presence of BB genotype was 10.453 (1.279 < or = IC95% < or = 85.40; p = 0.019). Results reveal a strong association between MBL polymorphism and HTLV infection. Presence of genotype BB may be associated with the susceptibility to HTLV, but further studies, with a larger number of individuals, will be necessary. MBL polymorphism could possibly have an impact on diseases associated with HTLV infection.

RESUMO

The first case of Kawasaki disease in Pernambuco, Brazil, is described. An 18-month-old by presenting with fever, adenomegaly and mucocutaneous rash, had right and left coronary arteries dilation and aneurysm detected on echocardiography. Treated with a high dosage of aspirin, the patient is asymptomatic after a 7-month follow-up, with regression of coronary lesions.

RESUMO

The present work tested whether Triatoma pseudomaculata Corrêa & Espínola shows behavioral traits indicating that it is capable of feeding on arthropods. We consistently observed nymphs extending their proboscis in the direction of cockroaches and attempting to bite. Insects presented a preference for biting specific cockroach body parts. Our results suggest that arthropod hemolymph represents an alternative source of food that increases the survival of T. pseudomaculata individuals undergoing long starvation.

RESUMO

Low molecular weight fragments of sulfated galactans (Boc-5 and Boc-10) from the red algae Botryocladia occidentalis significantly inhibited Crotalus durissus cascavella sPLA2 enzymatic activity. Equimolar ratios of sPLA2 to Boc-5 or Boc-10 resulted in allosteric inhibition of sPLA2. Under the conditions tested, we observed that both Boc-5 and Boc-10 strongly decreased edema, myonecrosis, and neurotoxicity induced by native sPLA2.

Antibodies to human T-cell lymphotropic virus-1 and 2 (HTLV-1 and 2) were tested in 259 inhabitants (98 males and 161 females) of four villages of the Marajó Island (Pará, Brazil) using enzyme immunoassays (ELISA and Western blot). Types and subtypes of HTLV were determined by nested polymerase chain reaction (PCR) targeting the pX, env and 5 LTR regions. HTLV-1 infection was detected in Santana do Arari (2.06%) and Ponta de Pedras (1%). HTLV-2 was detected only in Santana do Arari (1.06%). Sequencing of the 5 LTR region of HTLV-1 and the phylogenetic analysis identified the virus as a member of the Cosmopolitan Group, subgroup Transcontinental. Santana do Arari is an Afro-Brazilian community and the current results represent the first report of HTLV-1 infection in a mocambo located in the Brazilian Amazon region.

We evaluated the ability of human anti-lipopolysaccharide (LPS) O6 immunoglobulin G (IgG) and IgM antibodies to protect mice challenged with Escherichia coli serotype O6:K2ac. Purified whole IgG, commercial gammaglobulin, whole IgM-effluent, pool of normal human serum (NHS), agammaglobulinaemic serum (test groups) or phosphate-buffered saline (control group) was injected into adult male 18 h before a challenge with viable O6 E. coli. The mortality rate was assessed over a period of 72 h. To determine the opsonic and phagocytic activity of the antibody isotypes, we incubated peritoneal macrophages from the control and test groups collected at different times after challenge with the live bacteria with acridine orange for fluorescent analysis. Tumour necrosis factor (TNF)-alpha and interleukin (IL)-6 were quantified in serum of both the test and control groups. All mice that received commercial gammaglobulin or NHS survived. Purified whole IgG (containing 1.1 mg/l of anti-LPS O6 IgG antibodies) protected 87.5% of the animals tested in this experiment, while whole IgM-enriched effluent with 1.5 mg/l of anti-LPS O6 IgM antibodies protected only 12.5%. The agamma serum showed no protective capacity compared with PBS (serving as control). The minimal concentration of anti-LPS O6 IgG antibodies able to protect 50% of animals was 0.137 mg/l of purified whole IgG. Whole IgM-enriched effluent showed no protective capacity independently of the concentration tested (0.048-17.0 mg/l of anti-LPS O6 IgM antibodies). Fluorescent analysis of peritoneal macrophages from animals pretreated with purified whole IgG showed no bacteria at 8 h after the challenge. By contrast, whole IgM effluent showed an increasing number of live bacteria at the same time. Mice that had received whole IgM effluent (1.5 mg/l of anti-LPS O6 IgM antibodies) before the challenge with LPS O6 presented 20.5 microg/l of IL-6 and 1.5 microg/l of TNF-alpha. Serum from animals pretreated with purified IgG did not present any detectable pro-inflammatory cytokine. Our findings suggest that IgG but not IgM antibodies protect animals from a challenge with E. coli O6 serotype.

BACKGROUND: Lysozyme is a muramidase that acts on the peptideoglycan wall of Gram positive bacteria, causing cell death. It plays part in innate immunity and is present in blood, external fluid, as well in lysossomal granules of the phagocytes. Primary Immunodeficiencies are a diverse group of illnesses that, as a result of abnormalities of the immune system, increase susceptibility to infection. Among the examples of impaired natural immunity are defects in phagocytes and in the complement system. Innate immunity could be important in protecting mucosas against infections in patients with different forms of primary immunodeficiencies. The aim of this study was to investigate lysozyme concentrations in saliva from patients with primary immunodeficiencies. METHODS: Lysozyme levels in saliva samples from 34 patients with primary immunodeficiency (30 children and adolescents between the age of 3-13 years and 4 adults between the age of 20-33) and 60 age-matched healthy controls (49 children and adolescents between the ages of 3-15 and 11 adults between the ages of 22-42) were determined by the lysoplate method. RESULTS: There was no statistically significant difference between the lysozyme concentrations in the saliva of the immunodeficient subjects and those of the healthy controls. CONCLUSION: The results in the present work clearly show that salivary lysozyme levels in primary immunodeficient patients are equivalent to those found in healthy controls, suggesting that this enzyme still represents a remaining (but not a compensatory mechanism), contributing to the protection of there patients against infections.
