RESUMO

The objective of this network meta-analysis was to determine the efficacy of different mycotoxin binders (MTB) to reduce aflatoxin M1 (AFM1) in milk. A literature search was conducted to identify in vivo research papers from different databases. Inclusion criteria were in vivo, dairy cows, description of the MTB used, doses of MTB, aflatoxin inclusion in the diet, and concentration of AFM1 in milk. Twenty-eight papers with 131 data points were selected. Binders used in the studies were hydrated sodium calcium aluminosilicate (HSCAS), yeast cell wall (YCW), bentonite, and mixes of several MTB (MX). The response variables were AFM1 concentration, AFM1 reduction in milk, total AFM1 excreted in milk, and transfer of aflatoxin from feed to AFM1 in milk. Data were analyzed with CINeMA and GLIMMIX procedures with the WEIGHT statement of SAS (SAS Inst. Inc.). The AFM1 concentration in milk decreased for bentonite (0.3 µg/L ± 0.05; mean ± SE) and HSCAS (0.4 µg/L ± 0.12), and tended to decrease for MX (0.6 µg/L ± 0.13) but was similar for YCW (0.6 µg/L ± 0.12), compared with control (0.7 µg/L ± 0.12). The percentage reduction of AFM1 in milk was similar for all MTB and different from control with a range of reduction from 25% for YCW to 40% for bentonite. The excretion of AFM1 in milk was lower in YCW (5.3 µg/L ± 2.37), HSCAS (13.8 µg/L ± 3.31), and MX (17.1 µg/L ± 5.64), and not affected by bentonite (16.8 µg/L ± 3.33) compared with control (22.1 µg/L ± 5.33). The transfer of aflatoxin B1 from feed into AFM1 in milk was lowest in bentonite (0.6% ± 0.12), MX (1.04% ± 0.27), and HSCAS (1.04% ± 0.21), and not affected in YCW (1.4% ± 0.10), compared with control (1.7% ± 0.35). The meta-analysis results indicate that all MTB reduced the AFM1 transfer into milk, where bentonite had the highest capacity and YCW the lowest.

A network meta-analysis was conducted to determine the effects of feeding feed-grade urea (FGU) or slow-release urea (SRU) as a replacement for true protein supplements (control; CTR) in high-producing dairy cattle diets. Research papers were selected (n = 44) from experiments published between 1971 and 2021 based on the following criteria: dairy breed, detailed description of the isonitrogenous diets fed, provision of FGU or SRU (or both), high-yielding cows (>25 kg/cow per day), and results that included at least milk yield and composition, but data on nutrient intake, digestibility, ruminal fermentation profile, and N utilization were also considered. Most studies compared only 2 treatments, and a network meta-analysis approach was adopted to compare the effects among CTR, FGU, and SRU. Data were analyzed using a generalized linear mixed model network meta-analysis. Forest plots of milk yield were used to visualize the estimated effect size of treatments. Cows included in the study produced 32.9 ± 5.7 L/d of milk, 3.46 ± 5.0% of fat, and 3.11 ± 0.2% of protein with an intake of 22.1 ± 3.45 kg of dry matter. Average diet composition was 1.65 ± 0.07 Mcal of net energy for lactation, 16.4 ± 1.45% CP, 30.8 ± 5.91% neutral detergent fiber, and 23.0 ± 4.62% starch. Average supply of FGU was 209 g/cow per day, whereas the average supply of SRU was 204 g/cow per day. With some exceptions, feeding FGU and SRU did not affect nutrient intake and digestibility, N utilization, and milk yield and composition. However, the FGU reduced the acetate proportion (61.6 vs. 59.7 mol/100 mol) and the SRU reduced the butyrate proportion (12.4 vs. 11.9 mol/100 mol) compared with CTR. Ruminal ammonia-N concentration increased from 8.47 to 11.5 and 9.3 mg/dL in CTR, FGU, and SRU, respectively. Urinary nitrogen excretion increased from 171 to 198 g/d in CTR versus the 2 urea treatments, respectively. The use of moderate doses of FGU in high-producing dairy cows may be justified based on its lower cost.

BACKGROUND: Several randomized clinical trials were done to determine whether supplementation with a high caloric diet, either through carbohydrate or lipid supplementation, is safe, tolerable and improves survival. However, most of these trials are small and the results are conflicting. METHODS: Randomized prospective trials utilizing high caloric supplementation among patients with amyotrophic lateral sclerosis (ALS) were searched using the terms [("amyotrophic lateral sclerosis" or "motor neuron disease" or "ALS" or "MND") and ("high calorie" or "high fat" or "high protein" or "high carbohydrate" or "supplementation")] in Medline, Cochrane, Embase, Scopus, Prospero and Herdin by two independent neurologists. Journal articles deemed relevant were assessed for eligibility. MAIN RESULTS: There were 57 articles obtained from databases, 49 of which were excluded. Four articles were further excluded since all of them had different interventions. Overall, there were 311 ALS patients included in the study, 176 of them were from the intervention group while 135 were used as controls. Overall, high caloric supplementation in ALS was deemed safe and tolerable, and also when adverse events, tolerability and mortality are combined using meta-analysis. Although in most publications the efficacy of giving high caloric supplementation has been generally beneficial, some of the outcome parameters are not statistically different from controls when studies are combined using meta-analysis. CONCLUSIONS: Current evidence suggests that high calorie supplementation is generally safe and tolerable for patients with ALS. However, it has not been shown to be efficacious in improving weight and functional disability.

PURPOSE: This study evaluated the effect of coating traditional and translucent Y-TZP with an industrial nanometric colloidal silica or glaze before or after sintering on the adhesion of zirconia with various ytrria concentration. MATERIALS AND METHODS: Specimens of Y-TZP with 3% and 5% yttria were subdivided into 5 groups (n=10), according to the coating applied and moment of application (before or after Y-TZP sintering): Control (no coating), Colloidal Silica/Sintering, Sintering/Colloidal Silica, Glaze/Sintering, Sintering/ Glaze. Lithium disilicate (LD) was used as positive control. Except for Y-TZP controls, groups were conditioned with silane before cementation with a self-adhesive resin cement. After 24 hours, the shear bond strength and failure analysis were performed. Also, analysis of specimens' surface was accomplished with SEM-EDX. Kruskal-Wallis and Dunn tests were applied to analyze differences between groups (p⟨0.05). RESULTS: Overall, the worst and best values of shear bond strength test were control and glaze after sintering groups. Different morphological and chemical aspects were observed in SEM-EDX analysis. CONCLUSIONS: Coating Y-TZP with colloidal silica showed unsatisfactory results. In 3Y-TZP, the surface treatment associated with the best adhesion values was the application of glaze after zirconia sintering. However, in 5Y-TZP, glaze application can be performed before or after the zirconia sintering to optimize clinical steps.

The objective of this study was to identify changes in prepartum behavior associated with the incidence of postpartum diseases in dairy cows. Multiparous Holstein cows (n = 489) were monitored with accelerometers for 3 wk prepartum. Accelerometers measured steps, time at the feed bunk, frequency of meals, lying bouts, and lying time. Postpartum health was monitored from 0 to 30 d in milk and cases of metritis, mastitis, retained placenta, displaced abomasum (DA), ketosis, and hypocalcemia were recorded. A multivariate linear mixed model was used to assess differences in behavior between diseased and not diagnosed diseased cows. A multivariate logistic regression was used to predict the occurrence of diseases. Predictors were selected using a manual backward stepwise selection process of variables until all remaining predictors had a P < 0.10. Models were submitted to a leave-one-out cross-validation process, and sensitivity, specificity, false discovery rate, and false omission rate were calculated. On average, over the 3-wk prepartum period, cows not diagnosed diseased (n = 345) took 1,613 ± 38 steps, spent 181 ± 7.1 min at the feed bunk, had 8.3 ± 0.17 meals, had 9.8 ± 0.32 lying bouts, and spent 742 ± 11.3 min lying per day. Behavior of diseased cows (n = 144) did not differ from those not diagnosed diseased. However, differences for specific diseases were observed, being significant in the week prepartum. When considering changes in behavior for only the week before calving, cows with metritis had more lying bouts (+21%), cows with DA had fewer meals (-24%) and tended to take fewer steps (-18%), and cows with ketosis had fewer meals (-22%) and spent less time at the feed bunk (-40%). Prediction models with the best outcomes were found for DA and ketosis using data of the prepartum week only. The model for DA included time at the feed bunk. Cross-validation resulted in a 80% sensitivity, 58.1% specificity, 59.2% accuracy, 91.2% false discovery rate, and 1.7% false omission rate. The model for ketosis included time at the feed bunk and number of meals. Cross-validation resulted in 64.3% sensitivity, 59.3% specificity, 59.5% accuracy, 93.0% false discovery rate, and 2.8% false omission rate. Prepartum behavior of cows affected with metritis, DA, and ketosis was different from that of cows not diagnosed with diseases. Prediction equations were able to classify cows at high or low risk of ketosis and DA and can be used in taking management decisions, but the high false discovery rates requires further refinement.

BACKGROUND AND PURPOSE: Cranial nerve palsy is occasionally present in patients with chronic inflammatory demyelinating polyneuropathy (CIDP), but its prevalence, characteristics and relations with the CIDP subtypes have rarely been investigated. The aim of this study was to systematically assess cranial nerve involvement in typical and atypical CIDP. METHODS: Clinical data were reviewed in 132 consecutive patients with CIDP, including typical CIDP (n = 89), multifocal acquired demyelinating sensory and motor neuropathy (MADSAM) (n = 31), distal acquired demyelinating symmetric (DADS) (n = 9) and others (n = 3). RESULTS: The frequency of cranial nerve palsy was 11% in typical CIDP, 48% in MADSAM and 11% in DADS. Facial and bulbar palsy was most frequently present (9%), followed by ocular motor nerve palsy (5%). Bilateral involvement was seen in all typical CIDP and DADS patients, whereas 80% of MADSAM patients had unilateral palsy. The presence of cranial nerve involvement was associated with more severe limb muscle weakness in typical CIDP, but not in MADSAM. Cranial nerve palsy fully recovered in 90% of typical CIDP and in 67% of MADSAM patients. CONCLUSION: Amongst the CIDP subtypes, cranial palsy is frequent and unilateral in MADSAM, and less frequent and bilateral in typical CIDP and DADS. In typical CIDP, facial and bulbar palsy reflects more severe and extensive inflammation.

The objective of this study was to evaluate the capacity of 6 mycotoxin binders (MTB) to adsorb 3 AA and 4 water-soluble vitamins (WSV). Two experiments were conducted in in vitro conditions to simulate postruminal digestion with pepsin, malic acid, citric acid, acetic acid, and lactic acid at pH 3.0 and intestinal digestion with bile salts and pancreatin extract at pH 6.5. Experiment 1 was conducted with AA, and experiment 2 was conducted with WSV. Within experiment, main factors were the MTB (bentonite, clinoptiolite, sepiolite, montmorillonite, activated carbon, and yeast cell walls), the substrate (AA: Lys, Met, and Thr; WSV: B1, B2, B3, and B6), and the incubation strategy (substrates alone or mixed). Data were analyzed for the effects of main factors and their interactions. In experiment 1, the adsorption average for AA when incubated separately was 44.3%, ranging from 62.4% for Thr by clinoptiolite to 20.0% for Thr by activated carbon. When incubated together, the average adsorption was reduced to 19.9%, suggesting competition among substrates for adsorption. Adsorption ranged from 29.8% for Thr by yeast cell walls to 5.6% for Met by clinoptiolite, but there were significant interactions among MTB and AA. In experiment 2, the average adsorption of WSV when incubated separately or together was 34.1 and 45.1%, respectively, suggesting possible synergies among substrates. When vitamins were incubated separately, adsorption ranged from 90.5% for vitamin B1 to 4.0% for vitamin B3 by montmorillonite. Vitamins B1 (except by yeast cell walls) and B6 (except by bentonite, sepiolite, and montmorillonite) were absorbed the most, and vitamin B3 was absorbed the least (except by activated carbon and yeast cell walls, which were least together with vitamin B2). When vitamins were incubated together, adsorption ranged from 97.0% for vitamin B1 by montmorillonite to 0% for vitamin B2 by activated carbon and vitamin B3 by bentonite. Vitamins B1 by all MTB and B6 by clinoptiolite, sepiolite, and yeast cell walls were the most adsorbed, and vitamin B3 (except by activated carbon and yeast cell wall) was the least absorbed. There were significant interactions among MTB and WSV. Mycotoxin binders have a high degree of adsorption of the AA and WSV tested in in vitro conditions, which may limit their bioavailability. Results also suggest that when substrates were incubated together some interactions for adsorption occurred, which were competitive among AA and synergic among vitamins.

The objective of this study was to analyze whether changes in behavior can be a good early predictor of sickness in calves. Friesian males calves (n = 325; 30 ± 9 d of age; 65 ± 15 kg) were monitored with an activity-monitoring device from 30 to 90 d of life in 4 periods corresponding to 4 seasons. The activity-monitoring device measured number of steps, number of lying bouts, lying time, and frequency and time of visits to the feed bunk. Calf health status was monitored daily and all incidences were recorded. To compare sick and healthy calves, a matched pair design was used to assign calves into the healthy group. Day 0 was defined as the day of sickness diagnosis. For each sick calf, 3 calves with no signs of sickness during the entire period (healthy calves) on the same date, in the same season, and of similar age (±4 d) and weight at entry were identified. A multivariate linear mixed model was used from d -10 to +10 relative to the sickness diagnosis to describe differences between sick and healthy calves. A multivariate logistic regression model was used for predicting sick calves on the days before the diagnosis. Significance was declared at P < 0.05. Daily, healthy calves had 1,476 ± 195 steps, spent 185 ± 32.5 min at the feed bunk, consumed 10 ± 1.1 meals, had 19.5 ± 1.8 lying bouts, and spent an average of 978 ± 30.5 min lying. The difference in behavior between sick (n = 33) and healthy calves (n = 99) began to be evident on d -10. Sick calves had fewer steps and numbers of visits to the feed bunk on d -1 and 0 and spent less time at the feed bunk on d -10 and -1 compared with healthy calves. From d -2 to d 9, sick calves had 15% fewer lying bouts, with no difference in lying time except on d -10, when sick calves spent more time lying. The best prediction model was for d -1 and included season and age at entry as qualifying variables, and frequency of visits to the feed bunk, steps, and lying time as behavior predictors (69% sensitivity, 72% specificity, 72% accuracy, 55% false discovery rate, and 12% false omission rate). However, an earlier prediction would be more useful to reduce the negative effect of sickness on production and welfare. The prediction model for d -10 had 67% sensitivity, 67% specificity, 67% accuracy, 60% false discovery rate, and 14% false omission rate. Results indicate that the occurrence of sickness can be predicted in advance, and an automated alarm system could be used to identify calves at risk of becoming sick and apply a preventive treatment.

AIM: To answer the following focused question: 'As regards microorganism load reduction for patients undergoing root canal treatment, is the use of ozone therapy comparable to conventional chemomechanical techniques using sodium hypochlorite (NaOCl)?' DATA SOURCES: A systematic review was conducted using controlled vocabulary and free-text key words in the following databases: PubMed, Science Direct, Scopus, Web of Science and Open Grey until 2 November 2018. Additional studies were sought through hand searching of endodontic journals. STUDY ELIGIBILITY CRITERIA, PARTICIPANTS AND INTERVENTIONS: The inclusion criteria comprised studies that compared microbial reduction in root canals after treatments with ozone and NaOCl in extracted mature human teeth or randomized clinical trials. STUDY APPRAISAL AND SYNTHESIS METHODS: The quality assessment of included laboratory studies was performed with the following parameters: (i) sample size calculation, (ii) samples with similar dimensions, (iii) control group, (iv) standardization of procedures, (v) statistical analysis and (vi) other risk of bias. For randomized clinical trials, the qualitative analysis of the studies was performed from the bias risk assessment using the tool 'Bias Risk Assessment of Randomized Controlled Studies' Cochrane Handbook 5.0.2. RESULTS: The search resulted in 180 published studies. After removal of duplicate studies and full-text analysis, eight studies were selected and seven were considered low risk of bias (seven ex vivo studies and one random clinical trial). Overall, the results demonstrated that ozone therapy provides significantly less microbial load reduction than NaOCl. As an adjunct in chemomechanical preparation, ozone was ineffective in increasing the antimicrobial effect of NaOCl. Ozone performance was strongly associated with the application protocol used: it is dose, time and bacterial strain dependent, besides the correlation with the use of complementary disinfection sources. LIMITATIONS: A restricted number of randomized clinical trial was found, and the difference amongst the methodology of the studies did not allow a meta-analysis to be performed. CONCLUSIONS AND IMPLICATIONS OF KEY FINDINGS: Although the selected studies had limitations, this review reached a satisfactory methodological and moderate evidence quality contributing to important preliminary information regarding ozone therapy. As regards load reduction of microorganisms for patients undergoing root canal treatment, ozone is not indicated neither to replace nor to complement the antimicrobial action of NaOCl.

AIM: To evaluate the influence of ultraconservative endodontic access cavities (UEC) on gaps and void formation in resin composite restorations in extracted two-rooted maxillary premolars after root canal treatment. Traditional endodontic access cavities (TEC) were used as a reference for comparison. METHODOLOGY: Two-rooted maxillary premolars were scanned in a micro-computed tomographic (micro-CT) device, matched based on similar anatomical features and allocated into two groups (n = 10) according to the design of the access cavity: TEC or UEC. Teeth were mounted on a mannequin head and a single operator performed the treatment, including endodontic access cavity preparation, root canal detection, preparation, filling and restoration procedures, under magnification. For restorative procedure, a bulk fill flowable layer was applied initially followed by an overlaying of regular composite. After restoration procedures, a new micro-CT scan was performed to check the quality of the coronal restoration by analysing the percentage volume of empty spaces present in the tooth-restoration interface (gaps) and inside the restoration (voids). Data were analysed statistically using Shapiro-Wilk and Student's t tests with a significance level of 5%. RESULTS: All specimens had gaps and voids. There were significant differences between the access cavity designs regarding the formation of voids with significantly more voids associated with the UEC (P < 0.05). Gap formation did not differ between groups (P > 0.05). CONCLUSION: The access cavity design used during root canal treatment interfered with the adaptation of the restorative material. The minimally invasive access cavity design was associated with a significantly greater number of voids within restorations.

OBJECTIVE: The objective of this study was to determine whether characteristics of positive results in the indirect immunofluorescence assay on HEp-2 cells for anti-cell antibodies (HEp-2 IFA) differ between patients with non-autoimmune diseases (NADs) and patients with systemic autoimmune rheumatic diseases (SARDs). METHODS: Cross-sectional observational study comparing HEp-2 IFA test results in three groups: (a) 558 NAD patients comprising four subgroups (cancer ( n = 95), infectious diseases ( n = 148), psychiatric diseases ( n = 163), common non-infectious chronic diseases ( n = 152)); (b) 194 SARD patients; (c) 1217 healthy individuals (HIs). Sera were tested at 1:80 dilution and diluted to the end titer. Slides were analyzed by two independent blinded examiners. RESULTS: A positive HEp-2 IFA test occurred in 102 (18.3%) NAD patients, 170 (87.6%) SARD patients and 150 (12.3%) HIs. The four NAD subgroups did not differ regarding HEp-2 IFA frequency, titer or pattern. HEp-2 IFA titer was higher in NAD patients than in HIs and both had lower titer than SARD patients. Nuclear dense fine speckled pattern was more frequent in NAD patients and HIs than in SARD patients ( p < 0.001). Nuclear homogeneous and nuclear coarse speckled patterns were more frequent in SARD patients than in the other groups ( p < 0.001). The nuclear fine speckled pattern was prevalent in all three groups, but presented a gradient in titer across them; HIs and NAD patients had low and intermediary titers, which were significantly lower than in SARD patients ( p < 0.001). CONCLUSION: Positive HEp-2 IFA frequency, pattern and titer present differential features in NAD and SARD patients, and this attribute adds value to the test in the diagnosis of SARDs.

AIM: To investigate the reliability of using bovine teeth as an alternative to human teeth in an intra-tooth push-out model by comparing the bond strength of three root canal sealers in both tooth substrates. METHODOLOGY: From human and bovine incisors (12 per group), one 1-mm disc was obtained from each root. On each disc, three standard 0.8-mm-diameter holes were drilled through the dentine. After irrigation, the holes were filled with one of three root canal sealers: AH Plus, MTA Fillapex or Total Fill BC Sealer. Tooth slices were kept in contact with sterile gauze moistened in PBS solution (pH = 7.2) for 7 days at 37 °C, and finally, a push-out load was applied until sealer dislocation. The bonded interface area was calculated to obtain data in MPa. The push-out data for each sealer and the overall push-out data for human and bovine teeth were compared (Mann-Whitney U). Kruskal-Wallis was used to study the sealers behaviour at each of the dental substrates. Multiple comparisons were performed using Mann-Whitney U after Bonferroni correction (P < 0.05). RESULTS: Dentine substrate did not influence the push-out of either individual sealers or overall sealers tested (Mann-Whitney U, P > 0.05). In both human and bovine samples, AH Plus performed better than BC Sealer and MTA Fillapex, whilst BC Sealer also provided a superior push-out compared to MTA Fillapex (Mann-Whitney U, P < 0.05). CONCLUSION: When using an intra-tooth model, bovine teeth did not significantly influence the bond strength of sealers.

WHAT IS KNOWN AND OBJECTIVE: CYP2C19 genotypes presumably allow the prediction of the metabolizer phenotypes: poor (PMs), extensive (EMs) and ultra-rapid (UMs). However, evidence from previous studies regarding this predictive power is unclear, which is important because the benefits expected by healthcare institutions and patients are based on this premise. Therefore, we aimed to complete a formal evaluation of the diagnostic value of CYP2C19 and CYP3A4 genes for predicting metabolizer phenotypes established by omeprazole (OME) administration in 118 healthy children from Jalisco (western Mexico). METHODS: The genotypes for CYP3A4*1B and CYP2C19*2, *3, *4, *5 and *17 alleles were determined. CYP2C19 and CYP3A4 phenotypes were obtained after 20 mg OME administration and HPLC quantification in plasma to estimate the Hydroxylation Index (HI = OME/HOME) and Sulfonation Index (SI = OME/SOME), respectively. RESULTS AND DISCUSSION: The distribution of genotypes and phenotypes for CYP2C19 and CYP3A4 was similar to previous studies in Mexico and Latin America. We estimated the CYP2C19 UM, EM and PM phenotype frequency in 0.84%, 96.61% and 2.54%, respectively. Although differences in the HI distribution were observed between CYP2C19 genotypes, they showed a poor diagnostic ability to predict the CYP2C19 metabolizer phenotype. Similarly, the number of CYP2C19 and CYP3A4 functional alleles was correlated with the HI distribution, but also their diagnostic ability to predict the CYP2C19 phenotype was poor. WHAT IS NEW AND CONCLUSION: The CYP2C19 phenotype is not predicted by the number of functional alleles of CYP2C19 and CYP3A4 genes. Phenotyping is still the most valuable alternative to dose individualization for CYP2C19 substrate drugs.

AIM: To analyze possible factors associated with the quality of life (QoL) of mothers of preterm infants with very low birth weight (VLBW) during the first 3 years after delivery. METHODS: The World Health Organization Quality of Life (WHOQOL)-bref scores were compared and correlated with maternal and infant-related sociodemographic and clinical variables at maternal discharge (T0) and at 6 (T1), 12 (T2), 24 (T3), and 36 (T4) months after delivery. Multiple linear regression models were fitted to investigate the effect of these variables on the mothers' QoL. RESULTS: The WHOQOL-BREF physical domain scores were higher at T1 and T2 compared to T0 (p = 0.013). Maternal variables that contributed to maternal QoL scores (p < 0.05) were stable marital union (b = 13.60; T0), family income (b = -12.75; T3), Evangelical religion (b = 8.11; T4), and beck depression inventory (BDI) score (-1.42 ≤ b ≤ -0.36; T0, T1, T2, T3, and T4). Infants' variables that most affected maternal QoL (p < 0.05) were posthemorrhagic hydrocephalus (PHH) (-18.84 ≤ b ≤ -10.05; T1, T2, and T4), bronchopulmonary dysplasia (BPD) (b = -7.41; T2), female gender (b = 8.09; T2), and SNAPPE severity score (b = -0.23; T3). CONCLUSION: Mothers of preterm infants with VLBW exhibited transient improvements in physical well-being during the first year after delivery. The presence of depressive symptoms in mothers and the diagnosis of PHH or BPD were negatively associated with QoL. Social, religious, and economic aspects were also important factors for the QoL of mothers of preterm infants with VLBW.

AIM: To evaluate the effect of 17% ethylenediaminetetraacetic acid (EDTA), 2.25% peracetic acid (PA) or 10% citric acid (CA) on the push-out bond strength of calcium silicate-based endodontic sealers [MTA Fillapex (Angelus, Londrina, Brazil) and Total Fill BC Sealer (FKG, La Chaux-de-Fonds, Switzerland)] to root dentine after different periods of immersion in phosphate-buffered saline (PBS). AH Plus (Dentsply DeTrey, Konstaz, Germany) was used as a reference material for comparison. METHODOLOGY: Thirty human maxillary incisors were selected and three 1-mm discs were obtained from the middle third of each root. On the axial surface of each disc, three 0.8-mm-diameter holes were drilled through the dentine. After standardized irrigation, each dentine disc from the same root was treated for smear-layer removal with one of the three tested chelating solutions: EDTA, PA or CA. After that, the holes were dried with paper points and filled with one of the three test root canal sealers. The filled dental slices were further immersed in PBS solution (pH 7.2) for 7 days (T1) or 30 days (T2) at 37 °C, before the push-out assay. Friedman, Kruskal-Wallis and Mann-Whitney tests were used respectively to estimate the impact of the independent variables (solutions, sealers and period of PBS immersion) on the push-out bond strength. The level for significance was set at α = 5% and data were further subjected to a Bonferroni correction. RESULTS: Solutions and periods of immersion did not significantly influence the displacement resistance (P = 0.285, and P = 0.104, respectively) of sealers to root dentine. On the other hand, the type of sealer significantly affected the push-out bond strength (P = 0.000). AH Plus was associated with greater resistance to dislodgment compared with Total Fill BC Sealer and MTA Fillapex (P = 0.000). MTA Fillapex resulted in the lowest push-out bond strength (P = 0.000). CONCLUSION: The use of different chelating agents did not influence the push-out bond strength of endodontic sealers. Calcium silicate-based sealers had lower push-out bond strength values compared with a conventional epoxy resin-based sealer (AH Plus).

PURPOSE: To determine the prevalence of the gestational trophoblastic disease (GTD) and to evaluate the risk factors for persistence of the disease. MATERIALS AND METHODS: A retrospective study with 152 women diagnosed with GTD at University of Campinas from 2006 to 2013. Frequencies, average, and standard deviation were evaluated for each variable and an exact Fisher test was utilized to study persistent disease risk factors. RESULTS: The women were on average 25.81 ± 8.02 years old. The most prevalent blood type was O positive (49.6%). The diagnosis of complete mole was found in 64.8%, followed by partial mole in 32%, and choriocarcinoma in 3%. Complete mole developed invasive mole in 12.3%, the same occurred with 7.5% of the partial moles. No significant association was found between age, number of gestations, race, BMI or blood type and persistent mole. CONCLUSION: The complete mole is the most fre- quent GTD, and is at high risk for developing malignant forms.

Tonically active cholinergic interneurons (TANs) from the nucleus accumbens (NAc) are centrally involved in reward behavior. TANs express a vesicular glutamate transporter referred to as VGLUT3 and thus use both acetylcholine and glutamate as neurotransmitters. The respective roles of each transmitter in the regulation of reward and addiction are still unknown. In this study, we showed that disruption of the gene that encodes VGLUT3 (Slc17a8) markedly increased cocaine self-administration in mice. Concomitantly, the amount of dopamine (DA) release was strongly augmented in the NAc of VGLUT3(-/-) mice because of a lack of signaling by metabotropic glutamate receptors. Furthermore, dendritic spines and glutamatergic synaptic transmission on medium spiny neurons were increased in the NAc of VGLUT3(-/-) mice. Increased DA and glutamate signaling in the NAc are hallmarks of addiction. Our study shows that TANs use glutamate to reduce DA release and decrease reinforcing properties of cocaine in mice. Interestingly, we also observed an increased frequency of rare variations in SLC17A8 in a cohort of severe drug abusers compared with controls. Our findings identify VGLUT3 as an unexpected regulator of drug abuse.

Currently, food allergies are an important health concern worldwide. The presence of undeclared allergenic ingredients or the presence of traces of allergens due to contamination during food processing poses a great health risk to sensitized individuals. Therefore, reliable analytical methods are required to detect and identify allergenic ingredients in food products. The present review addresses the recent developments regarding the application of DNA- and protein-based methods for the detection of allergenic ingredients in foods. The fitness-for-purpose of reviewed methodology will be discussed, and future trends will be highlighted. Special attention will be given to the evaluation of the potential of newly developed and promising technologies that can improve the detection and identification of allergenic ingredients in foods, such as the use of biosensors and/or nanomaterials to improve detection limits, specificity, ease of use, or to reduce the time of analysis. Such rapid food allergen test methods are required to facilitate the reliable detection of allergenic ingredients by control laboratories, to give the food industry the means to easily determine whether its product has been subjected to cross-contamination and, simultaneously, to identify how and when this cross-contamination occurred.

AIM: To evaluate the resistance to vertical root fracture of root filled teeth using different root canal preparation concepts: apical stop and continuously tapering preparation, with different foraminal enlargement. In addition, the effect of filling associated with these different concepts was assessed. METHODOLOGY: Ninety single-rooted premolars were used. The crowns were removed to standardize the root length at 11 mm. Ten roots were not instrumented and not filled (control group). The other roots were divided into four groups (n = 20): apical stop to finishing file size 25, 0.08 taper; apical stop to finishing file size 45, 0.02 taper; continuously tapering preparation to finishing file size 25, 0.08 taper; and continuously tapering preparation to finishing file size 45, 0.02 taper. Afterwards, 10 roots of each group were root filled using Gutta-percha and AH Plus. A continuous wave of condensation was used as the filling technique. The roots were evaluated as regards resistance to vertical fracture using a universal testing machine. The data were evaluated using the Kruskal-Wallis and Mann-Whitney tests. RESULTS: No significant difference in performance was observed between continuously tapering preparation size 25, 0.08 taper and apical stop size 45, 0.02 taper groups. Lower resistance values were found in continuously tapering preparation size 45, 0.02 taper group and the highest in apical stop size 25, 0.08 taper group. After filling, a significant increase in resistance values was observed in all groups. In addition, the continuously tapering preparation size 45, 0.02 taper group had values comparable with those of the control. CONCLUSION: The different canal preparation techniques reduced resistance to fracture when compared with the control group; however, after root filling, there was a significant increase in resistance.
