PURPOSE: To describe serum irisin and fibroblast growth factor-21 (FGF-21) concentrations in healthy female adolescents with different training activity patterns and their associations with bone mineral properties and metabolic markers. METHODS: A total of 62 adolescent girls aged 14-18 years were recruited: 22 rhythmic gymnasts, 20 swimmers, and 20 untrained controls. Bone mineral characteristics by dual-energy X-ray absorptiometry, daily energy intake by dietary recall, serum irisin, FGF-21, undercarboxylated osteocalcin, and C-terminal telopeptide of type I collagen were measured in all girls. RESULTS: Whole body and lumbar spine areal bone mineral density and lumbar spine bone mineral content were higher in the rhythmic gymnasts group compared with swimmers and untrained controls groups (P < .05). Serum irisin, FGF-21, undercarboxylated osteocalcin, and C-terminal telopeptide of type I collagen levels were not significantly different between the groups. In the rhythmic gymnasts group, serum FGF-21 concentration was positively correlated with lumbar spine areal bone mineral density independently of confounding factors (r = .51; P = .027). CONCLUSIONS: Serum irisin and FGF-21 levels were not different between adolescent eumenorrheic girls with different training activity patterns. FGF-21 was positively associated with lumbar spine areal bone mineral density, which predominantly consists of trabecular bone in adolescent rhythmic gymnasts.

Transient insulin resistance seen during puberty is expected to favour body growth, but our results show that increment in insulin resistance even in physiological ranges during puberty might compromise lumbar spine bone mineral density accrual independently of body composition parameters, and therefore adult bone quality might be challenged. INTRODUCTION: Insulin resistance (IR) might have a compromising effect on growing bone, and therefore adult bone quality might be challenged. The aim of the present study was to identify whether increases in IR during puberty contribute to bone mineral characteristics in males independently of body composition parameters. METHODS: This is a retrospective cohort-based longitudinal observational study. Data from 85 subjects were included. Boys were studied annually during their pubertal years (12 years at baseline) and at follow-up at the age of 18 years. Anthropometry, bone age, fasting blood samples, body composition, total body, and lumbar spine bone mineral characteristics were measured. Insulin resistance was determined by homeostatic model assessment of IR (HOMA-IR). Multiple regression analysis was performed to determine the effect of changes in HOMA-IR during pubertal years as a longitudinal predictor to fixed bone mineral outcome variables at the age of 18 years. All models were adjusted to potential clinically justified confounding variables. RESULTS: After adjustment to baseline bone indices and body composition-related predictors, the pubertal increment in the HOMA-IR was a negative independent predictor of lumbar spine bone mineral areal density (ß = - 0.202, p = 0.005) and lumbar spine bone mineral apparent density (ß = - 0.235, p = 0.005) in 18-year-old males. CONCLUSIONS: Pubertal increment in IR has a potential diminishing effect on lumbar spine bone mineral density accrual independently of body composition parameters. Further studies are needed to clarify whether monitoring HOMA-IR during puberty may identify subjects at increased risk of low peak bone mass and possible osteoporosis in the future.

Session rating of perceived exertion is different for similar intensity and duration prescribed low-intensity sessions and has a different effect on performance in young cross-country skiers. J Strength Cond Res 37(1): 187-193, 2023-The aim of this study was to analyze whether the rating of perceived exertion (RPE) and the resulting internal load (sRPE) could differ among subjects when volume and intensity are matched in young cross-country (XC) skiers. Second, we analyzed whether subjects with a high or low RPE response and sRPE have different adaptation after a 1-week low-intensity high-volume training period. Well-trained national level XC skiers (n = 13) aged 13.4 ± 1.9 years took part in a 1-week training period with double poling performance tested pretraining and after 1-week training. All subjects trained under a similar training program. Rating of perceived exertion was measured after each training session on a 10-point scale, and the total training load was calculated by the sRPE method by multiplying training session's duration and the RPE. Subjects were divided into Glow (n = 6) or Ghigh (n = 7) groups based on median rating of RPE (3.78 ± 1.24) in low-intensity trainings. Total training load in low-intensity trainings was higher in Ghigh compared with Glow (4,010 ± 765 vs. 2,499 ± 193 arbitrary units, respectively), although total training time with (16.3 ± 0.8 hours vs. 16.0 ± 0.6 hours) or without (13.1 ± 0.9 hours vs. 13.3 ± 0.7 hours, p > 0.05, respectively to Glow and Ghigh) high-intensity sessions was not different. No significant differences were found for time spent in different heart rate zones between the 2 groups. Increases in performance were significant for both groups (p< 0.05), while the effect was large in Ghigh (+12.5%; ES = 0.74), and medium in Glow (+10.7%; ES = 0.34). Differences in the internal load and the further quantification of training load for different target intensity sessions could provide important practical information to stimulate expected adaptation in young endurance athletes.

Long-term rowing performance development in male Olympic and World Championship medal winners compared with nonmedalists. J Strength Cond Res 37(9): e521-e526, 2023-The purpose of this study was, first, to investigate individual longitudinal 2,000 m rowing ergometer performance (2,000 erg) development of world class male rowers from the beginning of their career until reaching the elite level and to compare ELITE performance development with those who did not reach podium places and, second, to provide 2,000 erg milestones for talent prediction in rowing. Individual annual 2,000 erg performances of 54 male rowers were analyzed from age 15 and throughout their career. Olympic or World Championships medal winners (ELITE; n = 11) were compared with those who did not reach international podium places. Two thousand erg increased ( p ≤ 0.05) until the age of 24 in ELITE, while plateaued earlier for less successful rowers. No differences in the rate of performance improvement were found between ELITE and lower performers ( p > 0.05). At the age of 20, performance between ELITE and international level nonmedal winners reached significant difference (480.7 ± 20.9 W vs. 435.3 ± 33.9 W, respectively) and remained different onwards ( p ≤ 0.05). However, the average of the 10 best seasons cross-sectional performances of less successful rowers were significantly higher until age 18, compared with ELITE. ELITE rowers were able to improve their 2,000 erg performance to higher age; therefore, it is difficult to predict elite performers in junior rowers. Cross-sectional performances of less successful rowers may indicate temporary high-level performers, who might not be able to advance the elite level.

INTRODUCTION: The specific aims of the study were to compare possible differences in sclerostin and preadipocyte factor-1 (Pref-1) between rhythmic gymnasts (RG), swimmers (SW) and untrained controls (UC), and to investigate the relationships of sclerostin and Pref-1 with bone mineral characteristics in studied groups. MATERIALS AND METHODS: This study included 62 eumenorrheic adolescents (RG = 22; SW = 20; UC = 20). Bone mineral and body composition characteristics were measured by dual-energy X-ray absorptiometry, and sclerostin, Pref-1, osteocalcin and C-terminal telopeptide of type I collagen (CTx) were measured. RESULTS: Sclerostin was higher (P = 0.001) in RG (129.35 ± 51.01 pg/ml; by 74%) and SW (118.05 ± 40.05 pg/ml; by 59%) in comparison with UC (74.32 ± 45.41 pg/ml), while no differences (P = 0.896) were seen in Pref-1 (RG: 1.42 ± 0.16 ng/ml; SW: 1.41 ± 0.20 ng/ml; UC: 1.39 ± 0.26 ng/ml) between groups. Osteocalcin (RG: 7.74 ± 4.09 ng/ml; SW: 8.05 ± 4.18 ng/ml; UC: 7.04 ± 3.92 ng/ml; P = 0.843) and CTx (RG: 0.73 ± 0.22 ng/ml; SW: 0.64 ± 0.16 ng/ml; UC: 0.62 ± 0.20 ng/ml; P = 0.173) were not different between groups. Sclerostin correlated (P < 0.05) with whole-body bone mineral content (r = 0.61) and lumbar spine (LS) areal bone mineral density (aBMD) (r = 0.43) in RG, and femoral neck aBMD (r = 0.45) in UC. No correlation was found between sclerostin and bone mineral values in SW, and Pref-1 was not correlated with any bone mineral characteristics in studied groups. Sclerostin was the independent variable that explained 14% of the total variance (R2 × 100) in LS aBMD value only in RG. CONCLUSIONS: Adolescent athletes have higher sclerostin compared to UC. Sclerostin was correlated with bone mineral values and predicted areal bone mineral density in RG.

This study examined sclerostin and cytokine responses to an endurance training session in male single sculling rowers. Sixteen highly trained rowers performed a 2-h rowing exercise (distance: 23.8 ± 0.9 km; heart rate: 138 ± 8 beats.min-1; intensity: 79.8 ± 2.1% of the anaerobic threshold). Rowing resulted significant increment (p < 0.05) in sclerostin (36%), interleukin (IL)-6 (345%), IL-10 (129%) and monocyte chemoattractant protein-1 (MCP-1) (132%), while increases in irisin (6%) and tumour necrosis factor-α (3%) were not significant (p > 0.05). There was a correlation between the pre-to-post-exercise increase in sclerostin and the distance covered (r = 0.67; p = 0.005) as a marker of energy expenditure, and weekly training volume (r = 0.66; p = 0.005) as a marker of training stress. Post-exercise gain in MCP-1 was the most important predictor of post-exercise gain in sclerostin (ß = 0.543; p = 0.040). In conclusion, acute non-impact rowing training session with a total body mode induced increases in sclerostin and also in IL-6, IL-10 and MCP-1 concentrations, while post-exercise gain in MCP-1 was the main determinant of post-exercise gain in sclerostin. Exercise-induced increase in sclerostin could be regarded as a signal for metabolic reaction to the energy cost of acute exercise in rowers.

Internal load from hard training sessions is related to changes in performance after a 10-week training period in adolescent swimmers. J Strength Cond Res 35(10): 2846-2852, 2021-The purpose of this study was to investigate the association between session rating of perceived exertion (sRPE) and its categorization with the changes in swimming performance in adolescent swimmers. National level swimmers (age 16.4 ± 2.1 years, VÌo2max 50.0 ± 4.0 ml·min-1·kg-1) participated in a 10-week period preceding the National Winter Championships. Pre-training and post-training physiological parameters and swimming performance were measured. All training bouts with sRPE data were recorded. In addition, trainings were categorized as light, moderate, or hard. For calculating the corresponding internal training load (ITL), 2 fixed categorizations were applied based on earlier published sources and an individualized categorization based on ventilatory thresholds. During the 10-week training period, large to very large increases were found in aerobic (effect size [ES] = 1.58) and anaerobic threshold power output W·kg-1 (ES = 2.46), respectively, and smaller, but significant, improvement in 100-m swimming performance (ES = 0.36). Total ITL during the 10-week period was significantly related to changes in anaerobic threshold (r = 0.81). Fixed and individualized sRPE methods from hard sessions were associated with changes in VÌo2max (r = 0.77) and performance of only 100-m leg kicks (r = 0.76), respectively. Internal training load was related to changes in performance variables during the 10-week study period. Using individualized sRPE values, the accumulation of the ITL from hard training sessions was associated with changes in VÌo2max and swimming performance. The aforementioned categorization could help coaches in appropriate application of ITL for assessing the total training load in adolescent swimmers.

Regular physical activity (PA) has been associated with decreased risk of many chronic diseases and increased longevity among older adults. In addition, ageing has been associated with higher levels of different inflammatory biomarkers while the association between inflammatory biomarkers and PA has remained controversial. The aim of the current investigation was to examine the association between blood biomarkers and objectively assessed PA among a sample of healthy older women with different levels of PA engagement. A total of 81 healthy women were recruited. Study participants were allocated to three groups according to accelerometer-obtained PA data. Body composition was assessed with dual-energy X-ray absorptiometry. Fasting blood samples were collected for the measurement of resistin, leptin, tumour necrosis factor alpha (TNFα) and C-reactive protein (CRP) concentrations. There were no significant differences between groups for resistin, leptin, TNFα and for CRP concentrations; however, higher moderate-to-vigorous physical activity (MVPA) groups tended to have lower level of blood biomarker concentrations. There was a significant negative relationship between resistin and steps per day. Inverse association between leptin and MVPA was significant after controlling for age. In multivariate stepwise linear regression analysis, steps per day were the strongest independent predictor for resistin, whereas for leptin, TNFα and CRP the strongest independent predictor was whole body fat mass. In conclusion, this study demonstrated negative association between resistin concentration and steps per day. Sedentary time and light PA had no relationship with resistin, leptin, TNFα or CRP concentrations.

Background and objectives: Studies indicate that genetic and lifestyle factors influence optimal bone development. Adaptations in bone mineral characteristics related to physical activity (PA) are most often observed in pre- and peri-puberty. Longitudinal associations between bone mineral accrual and objectively measured PA in puberty are poorly understood. The present study aims to investigate whether pubertal PA at different intensities is related to bone mineral characteristics in individuals at 18 years of age. Materials and Methods: Anthropometrics, pubertal stage, bone age and PA by accelerometer were measured in 88 boys at the mean age of 12.1 (T1), 13.1 (T2), 14.0 (T3) and 18.0 years (T4). Different bone mineral parameters were measured by dual-energy X-ray at T4. Stepwise multiple regression analysis was performed to determine the effect of bone age, body mass and PA characteristics on measured bone mineral parameters at 18 years of age. Results: Total PA in puberty together with mean pubertal body mass predicted 35.5% of total body (TB) bone mineral density (BMD), 43.0% of TB less head (LH) bone mineral content (BMC) and 48.1% of BMC/height in individuals at 18 years of age. Vigorous PA and body mass in puberty predicted 43.2% of femoral neck (FN) BMD; bone age at T1, vigorous PA and body mass in puberty predicted 47.3% of FN BMC at 18 years of age. No associations between pubertal PA levels and lumbar spine bone mineral characteristics in individuals at 18 years of age were found. Conclusions: Physical activity in puberty has a significant impact on bone mineral characteristics in individuals at 18 years of age, with total PA being a significant predictor of TB BMD and TB LH BMC as well as BMC/height, whereas vigorous PA is a significant predictor of FN BMD and FN BMC.

This investigation examined the effects of acute rowing exercise on a panel of 12 different inflammatory cytokines. Fifteen female rowers (18.3±1.6 yrs; 172.0±5.0 cm; 67.5±8.8 kg; maximal oxygen consumption [VO2max]: 47.2±7.9 ml.min.-1kg-1) completed a 1-h endurance exercise (distance: 12.1±1.1 km; energy expenditure [EE]: 639±69 kcal; heart rate: 151±7 beats.min-1; intensity: 79.6±3.5% of the second ventilatory turn point). Venous blood samples were analysed for interleukin (IL)-2, IL-4, IL-6, IL-8, IL-10, vascular endothelial growth factor (VEGF), interferon-gamma (IFN-γ), tumor necrosis factor-alpha (TNF-α), IL-1α, IL-1ß, monocyte chemoattractant protein-1 (MCP-1) and epidermal growth factor (EGF) concentrations. Rowing exercise resulted increment (P<0.05) in IL-6, IL-8, VEGF and MCP-1. Exercise metabolic demand variables such as rating of perceived exertion (r=0.61), distance covered (r=0.60) and EE (r=0.57) were related (P<0.05) to changes in VEGF concentration. Cardiorespiratory fitness as measured by VO2max was correlated with changes in IL-6 (r=-0.55; P<0.05) level. In conclusion, acute exercise-induced inflammatory reaction was induced by a significant increase in IL-6, IL-8, VEGF and MCP-1 concentrations. Variance in exercise-induced increases in inflammatory cytokines in response to prolonged endurance exercise was characterised by exercise metabolic demand and cardiorespiratory fitness measures in female rowers.

PURPOSE: To investigate the differences in the pattern of changes in serum inflammatory cytokines measured annually over a 24-month period, between less active and more active overweight boys. PARTICIPANTS/METHODS: In total, 25 pubertal overweight boys were divided by their moderate to vigorous physical activity (MVPA) levels into 2 groups: less active group (LAG; n = 10; MVPA < 60 min/d) and more active group (MAG; n = 15; MVPA > 60 min/d). Physical activity was measured by 7-day accelerometry. Serum concentration of 13 inflammatory cytokines [interleukin (IL)-2, IL-4, IL-6, IL-8, IL-10, IL-1α, IL-1ß, vascular endothelial growth factor, interferon-γ, tumor necrosis factor-α, monocyte chemotactic protein-1, epidermal growth factor, and C-reactive protein] was measured at baseline (T0), after 12 months (T1), and after 24 months (T2) from fasting blood samples. RESULTS: Serum IL-6 level was significantly higher [LAG: 1.27 (0.86, 1.98) pg/mL; MAG: 0.80 (0.52, 0.84) pg/mL] at T0 and IL-8 level [LAG: 10.26 (8.80, 11.64) pg/mL; MAG: 7.42 (6.10, 9.54) pg/mL] at T2 in LAG compared with MAG. The changes over the study period varied between different inflammatory markers. None of the slopes of any measured markers were statistically different between the LAG and MAG, although the slopes of interferon-γ and IL-10 tended to be different between the groups. CONCLUSIONS: The pattern of changes over the study period varied between different inflammatory markers, but these changes were not different between the MVPA groups. More longitudinal studies are needed to investigate whether IL-6, IL-8, IL-10, and interferon-γ would be the choice of inflammatory markers to study the associations between obesity and physical activity in future.

OBJECTIVES: The purpose of this study was to estimate the state of tension (tone) and the biomechanical and viscoelastic properties of skeletal muscle in aging rats during the administration of different doses of dexamethasone and to find the relationships among the state of muscle atrophy, muscle strength, and the abovementioned muscle properties. METHODS: Muscle state of tension, biomechanical (elasticity, dynamic stiffness) and viscoelastic (mechanical stress relaxation time, Deborah number) properties (using MyotonPRO, Myoton Ltd, Tallinn, Estonia), lean body mass (BM), and hind limb grip strength were measured before and after the administration of a 10-day treatment with dexamethasone 100 µg/100 g BM (young and old group) and 50 µg/100 g BM (old group). RESULTS: Muscle elasticity (logarithmic decrement) was lower in old animals (1.86 ± 0.03) in comparison with young adult rats (1.38 ± 0.04) (P < .01). After the 10-day treatment with dexamethasone 100 µg/100 g BM, young adult rats had 10% lower muscle elasticity (P < .01). The same dose of dexamethasone in old rats increased tone (frequency of natural oscillation) from 29.13 ± 0.51 Hz to 38.50 ± 0.95 Hz (P < .001). There were dose-dependent differences in dynamic stiffness and tone of muscle; changes in elasticity were independent of the dose in old animals. In old rats, the muscle's viscoelastic properties decreased after dexamethasone administration. Significant correlation was found between changes in muscle logarithmic decrement and stiffness (rs = 0.90; P < .05) in old animals. CONCLUSIONS: Biomechanical and viscoelastic properties of skeletal muscle indicate changes in the main function of muscle during glucocorticoid-induced muscle atrophy and are in agreement with changes in hind limb strength. The myometric measurements indicate the direction and magnitude of change in muscle tissue after different doses of dexamethasone administration easily and quickly.

The aim of this 3-year prospective study was to examine changes in bone mineral characteristics during pubertal maturation in boys with different BMI values at the beginning of puberty and with different BMI increments during puberty. 26 boys with overweight and obesity (OWB) and 29 normal weight boys (NWB) were studied yearly for 3 years from the age of 11 years to measure the changes in different bone mineral characteristics. The OWB group was further divided into two subgroups according to extensive or non-extensive BMI increment during 3-year period. OWB had higher (P < 0.01) baseline total body (TB) bone mineral density (BMD), TB bone mineral content (BMC), TB BMC for height, lumbar spine (LS) BMD, and LS BMC compared to NWB. Throughout the study period, OWB gained more TB BMD (P = 0.0001), TB BMC (P = 0.0048), TB BMC for height (P = 0.0124), LS BMD (P = 0.0029), and LS BMC (P = 0.0022) compared to NWB. Also during the study period, TB BMD (P = 0.0065), TB BMC (P = 0.0141), TB BMC for height (P = 0.0199), LS BMD (P = 0.0066), LS apparent volumetric BMD (BMAD) (P = 0.0075), and LS BMC (P = 0.017) increased significantly less in those OWB whose BMI increased more extensively. Extensive BMI gain is associated with lower increments in bone mineral characteristics in boys with overweight and obesity. Unfavorable increment in total body fat mass and percentage during pubertal years could be one reason for that.

The purpose of this study was to evaluate the differences in body composition, neuromuscular performance, and mobility in healthy, regularly exercising and inactive older women, and examine the relationship between skeletal muscle indices and mobility. Overall, 32 healthy older women participated. They were divided into groups according to their physical activity history as regularly exercising (n = 22) and inactive (n = 10) women. Body composition, hand grip strength, leg extensor muscle strength, rapid force development, power output, and mobility indices were assessed. Regularly exercising women had lower fat mass and higher values for leg extensor muscle strength and muscle quality, and also for mobility. Leg extensor muscle strength and power output during vertical jumping and appendicular lean mass per unit of body mass were associated with mobility in healthy older women. It was concluded that long-term regular exercising may have beneficial effects on body composition and physical function in older women.

The aim of this study was to investigate the differences in associations of serum acylated and des-acylated ghrelin, peptide YY (PYY) and leptin levels with physical activity (PA) and cardiorespiratory fitness (CReF) in adolescent boys (mean age of 14.0 years) with overweight (OWB; n=55) and with normal weight (NWB; n=154). METHODS: Total PA was measured by 7-day accelerometry (counts/min) and CReF by peak oxygen consumption (VO2peak/kg). RESULTS: No differences were found in serum PYY, acylated ghrelin or des-acyl ghrelin levels, whereas mean leptin (11.6±10.6 vs. 2.0±2.7 ng/ml; p<0.05) and insulin (18.1±8.7 vs. 11.0±6.2 mU/l; p<0.05) levels were significantly higher in OWB compared to NWB. Mean CReF was significantly lower in OWB compared to NWB (39.7±8.7 vs. 50.5±6.8 ml/min/kg; p<0.05). Leptin was negatively correlated with CReF in both groups (r=-0.43; p<0.05), des-acylated ghrelin with CReF only in OWB (r =-0.36; p<0.05). In OWB leptin was negatively correlated with total PA (r=-0.32; p<0.05) and positively with sedentary time of PA (r=0.35; p<0.05). In NWB 28.1% of the variability of CReF was determined by leptin and insulin resistance index (HOMA-IR), whereas in OWB 71.9% was determined by trunk FM and BMI. CONCLUSIONS: Leptin concentration was inversely associated with CReF in adolescent boys independently of BMI in both groups, while des-acylated ghrelin was associated with CReF only in OWB. Low PA in OWB was associated with high serum leptin level.

Elevated blood lactate has been shown to influence subsequent anaerobic exercise due to an inhibition of glycolysis. The aim of the present study was therefore to investigate the influence of a short and high-intensity anaerobic arm crank pre-load exercise (HIE) added to a low-intensity warm-up on cardio-respiratory and metabolic responses on a subsequent all out rowing exercise. Nine well-trained college level male rowers (24.6 ± 7.1 yrs; 1.87 ± 0.07 m; 88.9 ± 9.8 kg; 18.5 ± 3.7% body fat) volunteered to participate in the study. The subjects performed a maximal 2000-m rowing ergometer performance tests (MPT) twice. One MPT was preceded by a normal low intensity warm-up (MPTlow), while another one was performed with the additional inclusion of the HIE protocol (MPThigh). Overall rowing performance in the MPTlow was significantly faster (p = 0.004) by 3.7 ± 2.8 sec compared to the MPThigh condition (401.7 ± 23.0 s v. 405.4 ± 23.3 s) but the reduction in speed was found only for the first 1000-m (p = 0.017). Net La increase from rest to the end of the MPTlow was 11.9 ± 2.3 mmol·l-1 which was significantly higher (p = 0.0001) compared to the MPThigh condition (6.3 ± 1.8 mmol·l-1). Carbon dioxide output was significantly lower in the second (p = 0.041), third (p = 0.009), fourth (p = 0.036) and fifth (p = 0.028) 250-m split in the MPThigh compared to the MPTlow test. In conclusion, HIE upper-body anaerobic pre-load added to a standard low intensity warm-up protocol decreased anaerobic performance only in the early stages of the MPThigh but the latter part was unaffected. The inhibition of glycolysis in the first minute of the workout might allow a different race strategy, which needs to be investigated in further studies.

Subjective energy intake (sEI) is often misreported, providing unreliable estimates of energy consumed. Therefore, relating sEI data to health outcomes is difficult. Recently, Börnhorst et al. compared various methods to correct sEI-based energy intake estimates. They criticised approaches that categorise participants as under-reporters, plausible reporters and over-reporters based on the sEI:total energy expenditure (TEE) ratio, and thereafter use these categories as statistical covariates or exclusion criteria. Instead, they recommended using external predictors of sEI misreporting as statistical covariates. We sought to confirm and extend these findings. Using a sample of 190 adolescent boys (mean age=14), we demonstrated that dual-energy X-ray absorptiometry-measured fat-free mass is strongly associated with objective energy intake data (onsite weighted breakfast), but the association with sEI (previous 3-d dietary interview) is weak. Comparing sEI with TEE revealed that sEI was mostly under-reported (74 %). Interestingly, statistically controlling for dietary reporting groups or restricting samples to plausible reporters created a stronger-than-expected association between fat-free mass and sEI. However, the association was an artifact caused by selection bias - that is, data re-sampling and simulations showed that these methods overestimated the effect size because fat-free mass was related to sEI both directly and indirectly via TEE. A more realistic association between sEI and fat-free mass was obtained when the model included common predictors of misreporting (e.g. BMI, restraint). To conclude, restricting sEI data only to plausible reporters can cause selection bias and inflated associations in later analyses. Therefore, we further support statistically correcting sEI data in nutritional analyses. The script for running simulations is provided.

BACKGROUND: We investigated longitudinal relationships between the biochemical markers of bone and adipose tissue with bone mineral content (BMC), bone mineral density (BMD), moderate-to-vigorous physical activity (MVPA) and sedentary time (SED) in pubertal boys. METHODS: Ninety-six boys (11.9 ± 0.6 years old) were measured at baseline, after 12 and 24 months. Body composition (fat mass [FM], lean body mass [LBM]), and whole body (WB), lumbar spine (LS) and femoral neck (FN) BMD and BMC were assessed. Additionally, serum leptin, adiponectin, osteocalcin (OC) and C-terminal telopeptide of type I collagen (CTX) were measured. RESULTS: OC had a strong longitudinal inverse effect on changes in WB_BMD (p < 0.001) and LS_BMD (p = 0.021), while CTX had an inverse effect only on changes in FN_BMD (p = 0.011). Leptin had an inverse effect on changes in WB_BMC/WB_BMD (p = 0.001), FN_BMD (p = 0.002) and LS_BMD (p = 0.001). MVPA showed a longitudinal inverse effect on changes in leptin (p = 0.030), however no longitudinal effect of SED to biochemical markers of bone and adipose tissue was found. CONCLUSIONS: Bone metabolism markers have negative effect on bone mineral accrual during puberty. Increases in MVPA affect leptin, suggesting a positive link of MVPA through leptin metabolism on increases in bone mineralization during puberty.

One of the key determinants of adult skeletal health is the maximization of bone mass during the growth period. Physical activity (PA) in combination with lean mass and fat mass contribute to a great extent to bone mineral accrual; however, PA changes significantly during puberty. The aim of the present study was to examine PA exposure relative to bone mass acquisition during a longer observation period. Daily PA was measured with 7-day accelerometry and bone mineral parameters by DXA in 11- to 13-year-old peripubertal boys (n = 169). Similar testing was done after 1 calendar year. Changes in sedentary time were negatively related to changes in whole-body bone mineral density (BMD), lumbar spine bone mineral content (BMC), lumbar spine bone area (BA), femoral neck (FN) BMD, and FN BMC (r > -0.157; p < 0.05). Sedentary time emerged as the main PA level in predicting changes in FN BMC (p = 0.027) and in combination with vigorous PA predicting changes in FN BMD (p < 0.024). In addition to the effect of body composition on the skeleton, increase in sedentary time emerged as one main physical activity predictor (in addition to vigorous PA) of bone mineral acquisition during a 12-month period in peripubertal boys.

Skinfold Calipers are widely used to obtain subcutaneous adipose tissue thickness because of its non-invasive, simple and inexpensive technique. Nevertheless, Caliper skinfold thicknesses have the disadvantage of measuring compressed adipose tissue and double layers of skin, which might reduce the precision of these results. In contrast, the computerized optical device Lipometer was developed to permit a quick, precise and non-invasive determination of non-compressed mono layers of subcutaneous adipose tissue thickness. In the present paper we investigate the hypothesis that Caliper skinfold thicknesses are significantly different from subcutaneous adipose tissue thicknesses in mm, which can be measured by Lipometer. Caliper and Lipometer results were obtained from 371 Estonian boys aged between 9.0 and 12.8 years. Measurements were performed at six different body sites: triceps, biceps, upper back, upper abdomen, hip and front thigh. Caliper measurements were systematically higher than Lipometer results in a range between 1.2 mm (hip) and 11.08 mm (front thigh). The limits of agreement analysis provided intervals from 7.5 mm (biceps) up to 30.14 mm (front thigh). Comparing Caliper and Lipometer results very low measurement agreement was found. The two methods provided very poor interchangeability.
