RESUMO

Cryopreservation has adverse effects on the post-thaw sperm quality due to oxidative stress and the presence of bacteria. To minimize such effects, plant extracts have been included in the composition of the semen diluents. The objective of the present study was to evaluate the antimicrobial and antioxidant effects of Moringa oleifera seed extract (MOSE) on cryopreserved ram semen, as well as its impact on in vitro fertilization. Semen from six hair rams was treated with five treatments before cryopreservation: Control (without any antibiotic), Standard (conventional antibiotic), 1.0, 10.0, and 50.0 mg/ml of MOSE. Post-thawing sperm characteristics were evaluated by the computer-assisted semen analysis. Antimicrobial activity was assessed by counting colony-forming units (CFU) and the antioxidant capacity by the ferric reducing antioxidant power method. A heterologous in vitro fertilization technique was implemented to measure the fertilization rate. Progressive and rapid motility, membrane and acrosome integrity, and active mitochondria were higher (p < .05) in the 10.0 mg/ml treatment compared with Standard after thawing. All M. oleifera treatments showed inhibition of CFU. The antioxidant capacity of M. oleifera seed extract was higher in the 10.0 and 50.0 mg/ml treatments. Fertilization rate (cleavage percentage) was higher (p < .05) in the 10.0 mg/ml (82.9 ± 10.0) and Control (82.5 ± 9.9) treatments compared with Standard (73.7 ± 9.1). The addition of 10.0 mg/ml of MOSE to ram semen inhibits the development of microorganisms and improves sperm characteristics and the in vitro fertility of the semen.

RESUMO

In total, 57 ticks were collected from six white-tailed deer (Odocoileus virginianus) and three mule deer (O. hemionus) in northern Mexico during the 2017, 2018 and 2019 hunting seasons. Morphological features of adult male and female ticks were observed and photographed using a stereo-microscope and scanning electron micrography. The ticks were identified as Dermacentor albipictus based on taxonomic keys. Molecular analysis using DNA amplification of the 16S rDNA and cytochrome oxidase 1 (COI) genes was employed to resolve the phylogenetic relationships from 18 strains of Dermacentor species. Bayesian phylogenetic analysis was performed in order to obtain a phylogenetic tree based on the concatenated sequence in the D. albipictus clade. The geometric morphometric analysis compared the body shape of ticks collected from specimens of two deer species by analyzing nine dorsal and ventral landmarks from both males and females. The results suggest that body shape variation in dorsal structures might be related to the host.

RESUMO

BACKGROUND & OBJECTIVES: Rickettsial and other zoonotic diseases are a latent risk for workers of veterinary clinics. The objective of this study was to evaluate the prevalence and the associated risk factors of parasitosis caused by Rhipicephalus sanguineus, and to estimate the seroprevalence of rickettsial diseases in workers of urban veterinary clinics of Juárez city, México. METHODS: The participants of the study were recruited from 63 private veterinary clinics and hospitals. The serological analysis of the blood samples collected was carried out using immunofluorescence assay (IFA). The statistical analysis for prevalences, risk factors, and correlation was performed with the SAS program. RESULTS: In total, 167 veterinary workers were included in the study. The prevalence of tick bites was 40% (67/167), and the risk factors associated with the occurrence of bites included the activities performed in the clinic and the number of labour hours spent per week. About 21% (35/167) of participants were seropositive to R. rickettsii, 28% (47/167) to Ehrlichia chaffeensis, and 24% (40/167) to Anaplasma phagocytophilum. A correlation was observed between: the number of workers in the clinics and the proportion of tick bites (r2 = 0.865); the prevalence of bites and the seropositivity of the participants to at least one pathogen (r2 = 0.924); and the number of bites per individual and infection to pathogens (r2 = 0.838). INTERPRETATION & CONCLUSION: Workers in urban veterinary clinics are highly exposed to tick bites and, therefore, to the diseases they transmit. Hence, it is important to implement prevention measures and perform constant monitoring of these diseases.

RESUMO

Influence of both the presence of a corpus luteum on the ovary and semen sex-sorting on development following in vitro fertilization is not yet conclusive. To determine the effect of these factors, 376 bovine oocytes were processed in vitro according to luteal presence on the ovary (CL+ and CL-) and type of semen used (sexed or conventional). Maturation rate was higher (P < 0.01) in CL- (136/138; 98.6%) than in CL+ (217/238; 91.2%). Cleavage rate was lower (P < 0.01) in CL+ with sexed semen (60/172; 34.9%) than in CL- with sexed semen (42/71; 59.1%), CL+ with conventional semen (47/66; 71.2%), and CL- with conventional semen (54/67; 85.1%). Compaction was similar (P = 0.69) in CL- (49/99; 49.4%) and CL+ (50/107; 46.7%). Blastulation rate was higher (P < 0.01) in CL- (26/99, 26.2%) than in CL+ (13/107; 12.1%) group. Expansion rate was higher (P = 0.01) in CL- (22/99; 22%) than in CL+ (11/107; 10.2%) group. Compaction rates were similar (P = 0.78) in sex-sorted (50/102; 49.0%) or conventional semen (49/104; 47.1%) groups. Blastulation was also similar (P = 0.91) with sex-sorted semen (19/102; 18.6%) and conventional semen (20/104; 19.2%). The rate of expanded blastocysts was similar (P = 0.89) in sex-sorted (16/102; 15.6%) and conventional (17/104; 16.3%) semen groups. In conclusion, the presence of CL can compromise maturation of the oocytes and their development, as a higher proportion of cleavage-stage embryos can be obtained with non-sexed semen with oocytes from ovaries without a CL.

RESUMO

This study evaluated whether the administration of 50 and 100 mg bovine somatotropin (bST) at the start of estrous synchronization and at the time of artificial insemination improves lambing rate and prolificacy in hair sheep. Four hundred eighty adult hair ewes (Pelibuey, Blackbelly, Dorper, Katahdin, and their crosses) were synchronized with intravaginal sponge containing 40 mg of fluorogestone acetate. On the day of sponge insertion, ewes were assigned to three treatments: the bST-100 treatment (n = 156) received 100 mg bST at the start of synchronization (d 0) and at the time of insemination (d 14), the bST-50 treatment (n = 159) received 50 mg bST in the same schedule as the previous group, and the control (n = 165) did not receive any bST. Lambing rate and percentage of multiple births were analyzed using the GENMOD procedure of SAS. Prolificacy data were analyzed using the MIXED procedure of SAS. The IGF-1 and insulin concentrations were analyzed with ANOVA for repeated measures. The bST application did not affect the lambing rate (P = 0.06). The proportion of ewes with multiple births (P = 0.01) and prolificacy (P = 0.04) were higher in the bST-50 (54.3% and 1.57 ± 0.1) than the bST-100 (18.2% and 1.25 ± 0.1) and control (33.3% and 1.28 ± 0.1) groups. The IGF-1 and insulin concentrations were higher (P < 0.05) in the bST-treated groups, but the insulin concentration was higher (P = 0.001) in the bST-100 group than in the bST-50 group. The administration of 50 or 100 mg bST at the start of synchronization and at the time of artificial insemination does not increase lambing rate. However, the dose of 50 mg increased the proportion of multiple births and prolificacy.

RESUMO

In beef and dairy cattle, the number of follicular waves affects endocrine, ovarian, and behavioral events during a normal estrous cycle. However, in Mexican-native Criollo cattle, a shortly and recently domesticated breed, the association between wave patterns and follicular development has not been studied. The objective of this study was to evaluate the effect of number of follicular waves in an estrous cycle on development of anovulatory and ovulatory follicles, corpus luteum (CL) development and functionality, as well as estrual behavior in Criollo cows. Ovarian follicular activities of 22 cycling multiparous Criollo cows were recorded daily by transrectal ultrasound examinations during a complete estrous cycle. Additionally, blood samples were collected daily to determine serum progesterone concentrations. Only two- (n = 17, 77.3%) and three-wave follicular (n = 5, 22.7%) patterns were observed. Duration of estrus, length of estrous cycle, and length of follicular and luteal phases were similar (P > 0.05) between cycles of two and three waves. Two-wave cows ovulated earlier (P < 0.05) after detection of estrus than three-wave cows. Detected day and maximum diameter of first anovulatory follicle were not affected (P > 0.05) by number of waves. Growth rate of first dominant follicle was higher (P < 0.05) in three-wave cycles. Onset of regression of the first dominant follicle was earlier (P < 0.01) in cycles with three waves than in those with two waves. In two-wave cycles, ovulatory follicles were detected earlier (P < 0.01) and had lower (P < 0.01) growth rate than in three-wave cycles. Development (i.e., maximum diameter and volume) and functionality (minimum and maximum progesterone concentration) of CL were similar (P > 0.05) between two- and three-wave patterns. In conclusion, Criollo cows have two or three follicular waves per estrous cycle, which alters partially ovulatory follicle development and ovulation time after detection of estrus. Length of estrous cycle, as well as CL development and functionality, was not affected by number of follicular waves.

RESUMO

Canine monocytic ehrlichiosis (CME) is the most common tick-borne disease affecting domestic dogs and other wild canids. It has a worldwide distribution and is associated with the presence of the brown dog tick. Few studies have been conducted in Mexico to identify and characterize Ehrlichia canis genetic variability. In the present study, 111 dogs of different sex, breed, and age from three geographic regions in Mexico were included. All of them had a previous history of tick infestation and/or the presence of one or more clinical signs compatible with CME. All dogs were tested by a commercial ELISA and nested PCR assay for the detection of E. canis. In addition, we analyzed the E. canis genetic diversity from the 16S rRNA gene sequences obtained in this study, along with 15 additional sequences described for E. canis in Mexico and obtained from GeneBank. Serological detection by commercial ELISA results showed overall infection rates of 85.58% (95/111), including 73.1% (30/41) in samples from Guerrero state; 75% (15/20) in Morelos; and 100% (50/50) in Chihuahua. On the other hand, molecular detection (nPCR assay) showed 31.5% (35/111) overall infection rate, with 41.4% (17/41) in Guerrero state; 55% (11/20) in Morelos; and 14% (7/50) in Chihuahua. We observed a high 16S rRNA gene sequence conservancy in most of the E. canis isolates in the three geographical areas from Mexico, including those analyzed in this research, suggesting a common geographic origin among isolates.

RESUMO

Tick-borne bacterial pathogens (TBBPs) show a worldwide distribution and represent a great impact on public health. The brown dog tick (Rhipicephalus sanguineus) is a vector of several pathogens that affect dogs and sometimes humans as well. In addition, TBBPs represent a diagnostic challenge and imply financial resources and medical treatment for long periods of time. In the present study, R. sanguineus s. l. was identified as the main tick species naturally parasitizing dogs that inhabit. Juárez City, Chihuahua, in the Paso del Norte region, Mexico-US Border, representing 99.8% of the cases. Additionally, an end-point PCR was performed to search for whether pathogens in R. sanguineus s. l. can transmit in DNA extracted from ticks and dog blood samples. This is the first molecular detection of Rickettsia rickettsi infecting domestic dogs in Mexico; however, other pathogens were also identified, such as Ehrlichia canis and Anaplasma platys in both ticks and dog blood samples, while Anaplasma phagocytophilum was identified only in dog blood samples. Moreover, co-detection in tick pools and co-infection in the analyzed dog blood samples could be found. Similarly, this research showed that dogs were found mostly parasitized by adult female ticks, increasing the possibility of transmission of E. canis.

RESUMO

BACKGROUND: Nowadays, Ehrlichia canis receives increasing attention because of its great morbidity and mortality in animals. Dogs in the subclinical and chronic phases can be asymptomatic, and serological tests show cross-reactivity and fail to differentiate between current and past infections. Moreover, there could be low parasitaemia, and E. canis might be found only in target organs, hence causing results to be negative by polymerase chain reaction (PCR) on blood samples. METHODS: We evaluated by PCR the prevalence of E. canis in blood, liver, spleen, lymph node and bone marrow samples of 59 recently euthanised dogs that had ticks but were clinically healthy. RESULTS: In total, 52.55% of the blood PCRs for E. canis were negative, yet 61.30% yielded positive results from tissue biopsies and were as follows: 63.15% from bone marrow; 52.63% from liver; 47.36% from spleen; and 15.78% from lymph node. In addition, 33% had infection in three tissues (spleen, liver and bone marrow). CONCLUSIONS: Our results show the prevalence of E. canis from tissues of dogs that were negative by blood PCR. Ehrlichia canis DNA in tissue was 30% lower in dogs that tested negative in PCR of blood samples compared to those that were positive. However, it must be taken into account that some dogs with negative results were positive for E. canis in other tissues.

RESUMO

Workers at veterinary clinics are exposed to zoonotic pathogens, which, though infrequently, can lead to serious consequences. The aim of the present study was to estimate the seroprevalence for rickettsial pathogens in veterinary clinic personnel in Ciudad Juárez, Chihuahua, Mexico. Voluntary participants included 106 veterinarians, 19 pet groomers, 36 veterinary assistants and 6 administrative workers who supplied blood samples taken by venipuncture. Detection of Rickettsia rickettsii, Ehrlichia spp. and Anaplasma phagocytophilum was conducted by indirect immunofluorescence assay (IFA) and polymerase chain reaction (PCR) techniques. To determine risk factors for the seroprevalence, the study considered social, demographic and occupational variables as well as the clinical signs and symptoms of the participants. Logistic regression procedures were performed, and the odds ratio (OR) was calculated with confidence intervals corresponding to a probability of 95%. The results indicate that 54% of the participants had a positive seroreaction to at least one of the pathogens evaluated and 2% were positive to all. The coexposure was as follows: 3% to R. rickettsii and A. phagocytophilum, 9% to Ehrlichia spp. and A. phagocytophilum, and 2% to Ehrlichia spp. and R. rickettsii. It was established that grooming posed the highest risk for R. rickettsii exposure. The implementation of appropriate management practices for zoonotic diseases is recommended to avoid health problems that may be life threatening to personnel in veterinary clinics.

RESUMO

Abstract Background: Villi morphology and function affect the absorption capacity of the small intestine. Most tissues are fragile and their morphology may change with excessive manipulation and inadequate sampling techniques. Intestinal sampling includes methodologies such as cutting longitudinally or transversely, keeping the intestinal content in it and preserving all in a 10% formalin solution; washing the intestinal sample in saline solution while emptying it by pressing downwards with two fingers, conserving the sample in a 10% formalin solution and knotting both ends of the sample, introducing 10% formalin into it and preserving it in the same solution. Objective: To compare height, area and desquamation caused by washing, pressing, and knotting used in sampling and conservation techniques of small intestine villi of pigs. Methods: Samples (n = 270) from duodenum, jejunum and ileum of 30 Landrace × Yorkshire crossed pigs, aged 7 to 8 months were randomly subjected to washing, soft pressing or knotting procedures, fixed in 10% formalin solution, embedded in paraffin, and stained with eosin and hematoxylin. Intestinal villi in each slide were observed to determine height, surface area and cellular desquamation of each villus. Results: Villi height from duodenum and ileum knotted samples was higher (p<0.05) compared with samples from the other procedures in the same anatomical portion, which were similar to each other (p>0.05). Villi from knotted jejunum samples were the shortest (p<0.05) compared to the other two procedures, which were similar to each other (p>0.05). Knotted samples from ileum had larger villi area compared with the rest of the procedures and intestinal portions (p<0.05). Villi desquamation was similar among procedures and portions of the intestine (p>0.05). Conclusion: Knotting is the recommended procedure for intestinal cell morphometry evaluation, as values of villi height and area are higher. Desquamation in the three procedures may be related to epithelial restoration processes.

Background: evaluation of reproductive behavior in Creole cows is important to determine several parameters, such as fertility, that show the physiologic reproductive mechanisms under different conditions. Therefore, the sequence of events such as ovulation and the subsequent formation of a corpus luteum need to be considered to establish genetic improvement programs under synchronized or naturally-occurring estrous conditions. Objective: to evaluate the ovarian and behavioral estrous characteristics before, during and after prostaglandin F2α-induced or naturally occurring estrous in Creole cows. Methods: thirty Creole cows were subjected to estrous synchronization with PGF2α and observed continuously over five days to determine estrous onset and duration. Seventeen days after synchronized estrous detection, cows were observed during seven d to detect the naturally-occurring estrous and its duration. After the onset of both types of estrous the ovaries of each cow were ultrasounded every 8 h to determine diameter of the preovulatory follicle at deviation and its maximum diameter prior to ovulation, as well as its growth rate, time to ovulation and maximum diameter of the later corpus luteum. Results: time of appearance and duration of synchronized and spontaneous estrous were similar (p>0.1). Diameters of the dominant follicle at deviation and prior to ovulation were similar in both types of estrous. Nevertheless, growth rate (mm/d) of the preovulatory follicle was higher (p<0.05) in synchronized estrous. However, time from onset of synchronized estrous to ovulation was longer (p<0.01). Diameter of corpus luteum 10 d after ovulation was similar in both types of estrous. Results indicate that growth rate of follicle and time from synchronized or spontaneous estrous to ovulation must be considered for estrous cycle manipulation and artificial insemination of Creole cows. Conclusion: a higher proportion of Creole cows show heat during nighttime, growth rate of ovulatory follicle is higher during synchronized estrous, and time from onset of synchronized estrous to ovulation is longer, when compared with spontaneous estrous.
