Biosurfactants are naturally occurring, surface-active chemicals generated by microorganisms and have attracted interest recently because of their numerous industrial uses. Compared to their chemical equivalents, they exhibit qualities that include lower toxic levels, increased biodegradable properties, and unique physiochemical properties. Due to these traits, biosurfactants have become attractive substitutes for synthetic surfactants in the pharmaceutical industry. In-depth research has been done in the last few decades, demonstrating their vast use in various industries. This review article includes a thorough description of the various types of biosurfactants and their production processes. The production process discussed here is from oil-contaminated waste, agro-industrial waste, dairy, and sugar industry waste, and also how biosurfactants can be produced from animal fat. Various purification methods such as ultrafiltration, liquid-liquid extraction, acid precipitation, foam fraction, and adsorption are required to acquire a purified product, which is necessary in the pharmaceutical industry, are also discussed here. Alternative ways for large-scale production of biosurfactants using different statistical experimental designs such as CCD, ANN, and RSM are described here. Several uses of biosurfactants, including drug delivery systems, antibacterial and antifungal agents, wound healing, and cancer therapy, are discussed. Additionally, in this review, the future challenges and aspects of biosurfactant utilization in the pharmaceutical industry and how to overcome them are also discussed.

Pigments are coloring agents used widely in different industrial sectors. There is a demand for using natural pigments rather than synthetic dyes because of the health hazards caused by synthetic dyes. Many natural pigments have different medicinal activities which can contribute to the nutritional value of the product. This study was carried forward with marine yeasts which can produce pigments. A total of 4 marine yeast isolates were recovered from the mangrove area of Sundarbans, West Bengal, India. Among them, the isolate KSB1 produced 856 µg/g total concentration of carotenoid pigment and the dry mass weight was 3.56 g/L. The stability of the extracted pigments was checked using temperature, pH, UV light exposure time, and different saline conditions. The pigments were characterized using HPLC and FTIR analysis. All of the extracted pigments showed good antioxidant activity in DPPH, metal chelating, and reducing power assay. The pigments were also found to have good antibacterial activity against the bacterial pathogens Staphylococcus aureus, Listeria monocytogenes, and Escherichia coli. Carotenoid pigment from KSB1 was found to have maximum activity in all the pathogens. The cytogenotoxicity using onion roots and phytotoxicity analysis indicated that the pigments were non-toxic and safe for cells. Finally, the potential marine yeast was identified using 18 s rRNA sequencing and identified as Rhodotorula sp. KSB1 (Accession no. MH782232).

Microbial exopolysaccharides (EPS) are biocompatible, biodegradable, and less toxic substances secreted outside the cell. They adsorb metal cations to its surface, making it another captivating property, which helps in stabilizing and biosynthesizing metal nanoparticles. Owing to these properties, we adopted bacterial EPS toward the green synthesis of nanoparticles and its application in the removal of azo dyes. Extracted EPS weighed 2.6 mg/mL from the most potential isolate A07 with 385 µg/mg of the carbohydrate content. The top three isolates were subjected to nanoparticle synthesis via the intracellular method and, by their extracted EPS, silver nanoparticles (AgNP) with the size around 87 nm were successfully produced by both methods mediated by the most potent isolate. The nanoparticles were characterized by UV-Vis spectroscopy, X-ray diffraction studies, atomic force microscopy, and FT-IR analysis. The nanoparticles were employed for dye degradation of azo dyes, namely, Methyl Orange (MO) and Congo Red (CO). EPS-Ag NPs showed fair degradation capability determined by UV-Vis kinetic studies. The work suggests electron transfer from reducing agent to dye molecule mediated by nanoparticles, destroying the dye chromophore. This makes EPS-Ag NPs a suitable, cheap, and environment-friendly candidate for biodegradation of harmful azo dyes. The most potential isolate was identified as Bacillus stratosphericus by 16S rRNA sequencing and submitted to GenBank under the accession id MK968439.

The study was planned to isolate a serine protease inhibitor compound with anticancer potential against colorectal and breast cancer cells from marine yeast. Protease enzymes play a crucial role in the mechanism of life-threatening diseases like cancer, malaria and AIDS. Hence, blocking these enzymes with potential inhibitors can be an efficient approach in drug therapy for these diseases. A total of 12 marine yeast isolates, recovered from mangrove swamps of Sundarbans, India, showed inhibition activity against trypsin. The yeast isolate ABS1 showed highest inhibition activity (89%). The optimum conditions for protease inhibitor production were found to be glucose, ammonium phosphate, pH 7.0, 30 °C and 2 M NaCl. The PI protein from yeast isolate ABS1 was purified using ethyl acetate extraction and anion exchange chromatography. The purified protein was characterized using denaturing SDS-PAGE, Liquid Chromatography Electrospray Ionization Mass Spectrometry (LC-ESI-MS), Reverse Phase High Pressure Liquid Chromatography (RP-HPLC) and Fourier Transform Infra-red Spectroscopy (FTIR) analysis. The intact molecular weight of the PI protein was determined to be 25.584 kDa. The PI protein was further studied for in vitro anticancer activities. The IC50 value for MTT cell proliferation assay was found to be 43 µg/ml against colorectal cancer HCT15 cells and 48 µg/ml against breast cancer MCF7 cells. Hoechst staining, DAPI staining and DNA fragmentation assay were performed to check the apoptotic cells. The marine yeast was identified as Candida parapsilosis ABS1 (Accession No. MH782231) using 18s rRNA sequencing.

The contemporary global agriculture is beset with serious threats from diverse eco-environmental conditions causing decreases in crop yields by ~ 15%. These yield losses might increase further due to climate change scenarios leading to increased food prices triggering social unrest and famines. Urbanization and industrialization are often associated with rapid increases in greenhouse gases (GHGs) especially atmospheric CO2 concentration [(CO2)]. Increase in atmospheric [CO2] significantly improved crop photosynthesis and productivity initially which vary with plant species, genotype, [CO2] exposure time and biotic as well as abiotic stress factors. Numerous attempts have been made using different plant species to unravel the physiological, cellular and molecular effects of elevated [CO2] as well as drought. This review focuses on plant responses to elevated [CO2] and drought individually as well as in combination with special reference to physiology of photosynthesis including its acclimation. Furthermore, the functional role of nitrogen use efficiency (NUE) and its relation to photosynthetic acclimation and crop productivity under elevated [CO2] and drought are reviewed. In addition, we also discussed different strategies to ameliorate the limitations of ribulose-1,5-bisphosphate (RuBP) carboxylation and RuBP regeneration. Further, improved stomatal and mesophyll conductance and NUE for enhanced crop productivity under fast changing global climate conditions through biotechnological approaches are also discussed here. We conclude that multiple gene editing approaches for key events in photosynthetic processes would serve as the best strategy to generate resilient crop plants with improved productivity under fast changing climate.

Fall armyworm, Spodoptera frugiperda (J. E. Smith) is a polyphagous and highly destructive invasive insect pest of many crops. It was recently introduced into India and widely reported in almost all parts of India. Development of a temperature-based phenology model for predicting its rate of development and distribution will help in understanding the establishment and further spread of introduced invasive insect pests. Development, survival and reproduction parameters of S. frugiperda at six constant temperature conditions (15, 20, 25, 27, 30 and 35°C) were investigated and further validated with data generated under fluctuating temperature conditions. The estimated lower developmental threshold temperatures were 12.1°C for eggs, 11°C for larvae, 12.2°C for pupae, 15.13°C for males and 12.66°C for females. Degree-day (DD) requirements for the development of the different stages of S. frugiperda were 50, 250 and 200 DD for egg, larva and pupa, respectively. The best-fitted functions were compiled for each life stage to yield a phenology model, which was stochastically simulated to estimate the life table parameters. The developed phenology model predicted temperature ranges between 27 and 30°C as favourable for S. frugiperda development, survival and reproduction. The results revealed that maximum net reproductive rate (215.66 females/female/generation) and total fecundity (981.08 individuals/female/generation) were attained at 30°C constant temperature. The mean length of generations decreased from 74.29 days at 15°C to 38.74 days at 30°C. The maximum intrinsic rate of increase (0.138 females/female/day) and shortest doubling time (4.9 days) were also observed at 30°C. Results of simulated life table parameters showed high temperature-dependent development of S. frugiperda and complete development within all the tested constant temperature ranges (15-35°C). Simulated life table parameters for predicting risk indices of S. frugiperda in India indicated a significant increase in activity indices and establishment risk indices with a higher number of generations during future (2050 and 2070) climatic change scenarios compared to present conditions. Our results indicate that India will be highly suitable for the establishment and survival of S. frugiperda in future time periods.

Cerebral proliferative angiopathies (CPAs) are distinct vascular malformations with varied clinical presentations and radiological findings from arteriovenous malformations (AVM) (Lasjaunias et al. in Stroke 39(3):878-85, 2008). They usually manifest with seizures if present supratentorial, headache, and progressive neurological deficits due to steal phenomenon or rarely with hemorrhage (Lasjaunias et al. in Stroke 39(3):878-85, 2008). Most of the patients are usually young females. Pediatric cases are extremely rare, with few cases reported till now. Here we report a child who presented with cerebellar bleed and diagnosed as CPA. The child was managed medically, and there was no change in caliber of the vessels after 18 months of follow-up. A short review of cases of CPA in pediatric age group presentations and management was undertaken in this case report.

Azo dyes are recalcitrant compounds used as a colorant in various industries. The pollution caused by their extensive usage has adversely affected the environment for years. The existing physicochemical methods for dye pollution remediation are rather inefficient and hence there is a dearth of low-cost, potential systems capable of dye degradation. The current research studies the biodegradation potential of immobilized bacterial cells against azo dyes Reactive Orange 16 (RO-16) and Reactive Blue 250 (RB-250). Two indigenous dye degrading bacteria Bacillus sp. VITAKB20 and Lysinibacillus sp. KPB6 was isolated from textile sludge sample. Free cells of Bacillus. sp. VITAKB20 degraded 92.38% of RO-16 and that of Lysinibacillus sp. KPB6 degraded 95.36% of RB-250 within 72 h under static conditions. Upon immobilization with calcium alginate, dye degradation occurred rapidly. Bacillus. sp. VITAKB20 degraded 97.5% of RO-16 and Lysinibacillus sp. KPB6 degraded 98.2% of RB-250 within 48 h under shaking conditions. Further, the nature of dye decolorization was biodegradation as evident by high-performance liquid chromatography (HPLC), and Fourier-transform infrared spectroscopy (FTIR) results. Phytotoxicity and biotoxicity assays revealed that the degraded dye products were less toxic in nature than the pure dyes. Thus, immobilization proved to be a highly likely alternative treatment for dye removal.

Chromium, ranking the second most among toxic heavy metal pollutants in the world, causing respiratory, cardiovascular and renal problems in human beings is under study herein. We examined the biological remediation of the carcinogenic Cr (VI) polluted soils by indigenous yeast isolates. The total element analysis of the treated sample was determined by Energy Dispersion X-ray Micro Analysis (EDXMA). The sample under study was observed to have a high concentration of 458.29 mgKg-1 Cr (VI), determined by Atomic Absorption Spectroscopy (AAS) and DPC analysis. The most tolerant isolate designated as CSR was used for in vitro and ex-situ bioremediation studies of Cr (VI). The isolate achieved significant bioremediation of 86% in vitro and 75.12% in ex-situ method. The optimal conditions for in vitro bioremediation were found to be 28 °C and a pH of 6. The ITS1, 5.8S rRNA and D1, D2 domain of LSU rRNA gene characterization of the isolate CSR illustrated that it belongs to Ustilago genera. The isolate was deposited in NCBI GenBank as Ustilago sp. CSR (KY284846). Although, Ustilago is generally a pathogenic fungus, our study opens up the scope of using Ustilago spp. for bioremediation of the carcinogenic heavy metal Chromium.

The perpetual increase in the resistance offered by biofilm-forming nosocomial pathogens has become a critical clinical challenge. Marine Streptomyces sps present a promising future of novel compounds with novel applications. We focus on the anti-biofilm activity of marine Streptomyces against two major nosocomial pathogens from clinical samples, Pseudomonas aeruginosa and Staphylococcus aureus. Herein, Streptomyces griseoincarnatus, a species known to harbour alkaline protease inhibitors and anti-tumour compounds were found to exhibit anti-biofilm activity. The study progresses to decipher the anti-biofilm potential of the extract as 82.657 ±â€¯1.1002% against P. aeruginosa and 78.973 ±â€¯1.672% against S. aureus at 100 µg/mL. The strain under study, S. griseoincarnatus HK 12 (accession no MF100857) has revealed the presence of certain fatty acyl compounds namely, 13Z-Octadecenal, 9Z-Octadecenal, Arachidic acid, Tetracosanoic acid and Erucic acid by GC-MS screening. Furthermore, the active compounds were docked against the quorum sensing system, LasI. The compound 13Z-Octadecenal was found to bind to the conserved sites of substrate binding with a binding energy of -1.90 kcal/mol thus, affirming the inhibitory activity of the fatty acyl compound. These active compounds were previously reported to be a part of active extracts exhibiting relevant antagonistic activities, but this so far is the first time they are found possessing anti-biofilm activity. Interestingly, the toxicity level of the extract at a high concentration of 500 µg/mL is as low as 11.5% when tested against human lung cancer lines, A549. Thus the report highlights the evidence of the potential of S griseoincarnatus HK12 to be an active and safe anti-biofilm agent.

INTRODUCTION: Anterior and anterolaterally situated foramen magnum meningiomas are a technically complex subgroup of meningiomas. The need for an extensive exposure and bone work and their complex anatomy make them a difficult and challenging group of tumors to resect. The bone work has ranged from an extensive condylar resection to condylar preserving exposures. In this paper, we present our experience with condylar preserving or minimal condylar resection based approaches to these tumors. MATERIALS AND METHODS: All patients who underwent surgical resection of anterior and anterolaterally situated foramen magnum meningiomas were included in the analysis. The study period was more than 10 years from 2005 to 2015 at our institute; a tertiary referral centre in India. The records along with demographic profile, clinico-radiological features, surgical strategies, outcomes as well as mortality and morbidity were analysed. RESULTS: There were a total of 20 patients (9 males and 11 females) who were operated during the study period. The average age was 36.7 years. In 16 patients, gross-total or near-total resection could be achieved, four patients underwent subtotal resection. Eight patients had fresh morbidity in the form of new motor deficits, pseudomeningocele formation, worsening of the lower cranial nerve functions or post-operative adhesions leading to syrinx formation. The follow-up ranged from 6 months to 140 months. CONCLUSION: Foramen magnum meningiomas are an eminently treatable group of tumors. Condylar preservation provides a good visualization, while helping to preserve joint stability and in avoiding instrumental stabilization.

Primary malignant glioma cell lines are being used for initial screening of anticancer agents. We utilized a simple mechanical disaggregation method for deriving cell lines from tumor tissues; and a Coverslip Culture-Acridine Orange Staining method to study cellular damage. Cell lines could be grown for up to three passages within three weeks after surgery. Cell proliferation, total cellular damage, and MTT assay were studied as parameters of cytotoxic response. Frequencies of damaged cells varied in different cell lines; and increased after cytotoxic treatments under clinically relevant conditions. These methods could contribute to preclinical evaluation of treatment response before commencement of radio-chemotherapy.

In the present study the leishmanicidal effect of potential protease inhibitor producing marine actinobacterial isolate has been investigated against Leishmania donovani, the causative agent of visceral leishmaniasis. Among 89 marine actinobacteria isolated from a salt pan in Kanyakumari, only one isolate (BVK2) showed 97% of protease inhibition activity against trypsin. Moderate to high protease inhibitor activity was shown by isolate BVK2 on proteinase (30%) and chymotrypsin (85%). In optimization study for protease inhibitor production glucose as carbon source and casein as nitrogen source showed the best activity. In the in-vitro Fluorescence-activated cell sorting (FACS) assay, 100 µg/ml of BVK2 extract was active against amastigotes in infected J774A.1 macrophages and showed 87% of parasitic inhibition. The isolate BVK2 showed significant anti-parasitic activity with an IC50 of 27.1 µg/ml after double doses were administered. The potential isolate was identified by molecular 16S rRNA gene sequencing as Streptomyces sp. VITBVK2. The results obtained suggest that the marine actinobacterial extract which have novel metabolites can be considered as a potential source for the development of drugs.

'Early responsive to dehydration' (ERD) genes are a group of plant genes having functional roles in plant stress tolerance and development. In this study, we have isolated and characterized a Brassica juncea 'ERD' gene (BjERD4) which encodes a novel RNA binding protein. The expression pattern of ERD4 analyzed under different stress conditions showed that transcript levels were increased with dehydration, sodium chloride, low temperature, heat, abscisic acid and salicylic acid treatments. The BjERD4 was found to be localized in the chloroplasts as revealed by Confocal microscopy studies. To study the function, transgenic Arabidopsis plants were generated and analyzed for various morphological and physiological parameters. The overexpressing transgenic lines showed significant increase in number of leaves with more leaf area and larger siliques as compared to wild type plants, whereas RNAi:ERD4 transgenic lines showed reduced leaf number, leaf area, dwarf phenotype and delayed seed germination. Transgenic Arabidopsis plants overexpressing BjERD4 gene also exhibited enhanced tolerance to dehydration and salt stresses, while the knockdown lines were susceptible as compared to wild type plants under similar stress conditions. It was observed that BjERD4 protein could bind RNA as evidenced by the gel-shift assay. The overall results of transcript analysis, RNA gel-shift assay, and transgenic expression, for the first time, show that the BjERD4 is involved in abiotic stress tolerance besides offering new clues about the possible roles of BjERD4 in plant growth and development.

We have generated 3900 enhancer-based activation-tagged plants, in addition to 1030 stable Dissociator-enhancer plants in a widely cultivated indica rice variety, BPT-5204. Of them, 3000 were screened for water-use efficiency (WUE) by analysing photosynthetic quantum efficiency and yield-related attributes under water-limiting conditions that identified 200 activation-tagged mutants, which were analysed for flanking sequences at the site of enhancer integration in the genome. We have further selected five plants with low Δ13 C, high quantum efficiency and increased plant yield compared with wild type for a detailed investigation. Expression studies of 18 genes in these mutants revealed that in four plants one of the three to four tagged genes became activated, while two genes were concurrently up-regulated in the fifth plant. Two genes coding for proteins involved in 60S ribosomal assembly, RPL6 and RPL23A, were among those that became activated by enhancers. Quantitative expression analysis of these two genes also corroborated the results on activating-tagging. The high up-regulation of RPL6 and RPL23A in various stress treatments and the presence of significant cis-regulatory elements in their promoter regions along with the high up-regulation of several of RPL genes in various stress treatments indicate that they are potential targets for manipulating WUE/abiotic stress tolerance.

Traumatic brain injury (TBI) is a devastating neurological disorder that usually presents in acute and chronic forms. Brain edema and associated increased intracranial pressure in the early phase following TBI are major consequences of acute trauma. On the other hand, neuronal injury, leading to neurobehavioral and cognitive impairments, that usually develop months to years after single or repetitive episodes of head trauma, are major consequences of chronic TBI. The molecular mechanisms responsible for TBI-induced injury, however, are unclear. Recent studies have suggested that early mitochondrial dysfunction and subsequent energy failure play a role in the pathogenesis of TBI. We therefore examined whether oxidative metabolism of (13)C-labeled glucose, lactate or glutamine is altered early following in vitro mechanical percussion-induced trauma (5 atm) to neurons (4-24 h), and whether such events contribute to the development of neuronal injury. Cell viability was assayed using the release of the cytoplasmic enzyme lactate dehydrogenase (LDH), together with fluorescence-based cell staining (calcein and ethidium homodimer-1 for live and dead cells, respectively). Trauma had no effect on the LDH release in neurons from 1 to 18 h. However, a significant increase in LDH release was detected at 24 h after trauma. Similar findings were identified when traumatized neurons were stained with fluorescent markers. Additionally (13)C-labeling of glutamate showed a small, but statistically significant decrease at 14 h after trauma. However, trauma had no effect on the cycling ratio of the TCA cycle at any time-period examined. These findings indicate that trauma does not cause a disturbance in oxidative metabolism of any of the substrates used for neurons. Accordingly, such metabolic disturbance does not appear to contribute to the neuronal death in the early stages following trauma.

BACKGROUND: Aneurysmal rebleed is the most dreaded complication following subarachnoid hemorrhage. Being a cause of devastating outcome, the stratification of risk factors can be used to prioritize patients, especially at high volume centers. METHOD: A total of 99 patients with aneurysmal rebleed were analyzed in this study both prospectively and retrospectively from August 2010 to July 2014. In the control group, 100 patients were selected randomly from the patient registry. A total of 25 variables from the demographic, historical, clinical and radiological data were compared and analyzed by univariate and multivariate logistic regression analysis. RESULTS: Significant independent predictors of aneurysm rebleed were the presence of known hypertension (p = 0.023), diastolic blood pressure of >90 mmHg on admission (p = 0.008); presence of loss of consciousness (p = 0.013) or seizures (p = 0.002) at first ictus; history of warning headaches (p = 0.005); higher Fisher grade (p < 0.001); presence of multiple aneurysms (p = 0.021); irregular aneurysm surface (0.002). CONCLUSIONS: Identification of high risk factors can help in stratifying patients in the high risk group. The risk stratification strategy with early intervention can prevent rebleeds. This in turn may translate into better outcomes of patients with intracranial aneurysms.

The ungauged wet semi-arid watershed cluster, Seethagondi, lies in the Adilabad district of Telangana in India and is prone to severe erosion and water scarcity. The runoff and soil loss data at watershed, catchment, and field level are necessary for planning soil and water conservation interventions. In this study, an attempt was made to develop a spatial soil loss estimation model for Seethagondi cluster using RUSLE coupled with ARCGIS and was used to estimate the soil loss spatially and temporally. The daily rainfall data of Aphrodite for the period from 1951 to 2007 was used, and the annual rainfall varied from 508 to 1351 mm with a mean annual rainfall of 950 mm and a mean erosivity of 6789 MJ mm ha(-1) h(-1) year(-1). Considerable variation in land use land cover especially in crop land and fallow land was observed during normal and drought years, and corresponding variation in the erosivity, C factor, and soil loss was also noted. The mean value of C factor derived from NDVI for crop land was 0.42 and 0.22 in normal year and drought years, respectively. The topography is undulating and major portion of the cluster has slope less than 10°, and 85.3% of the cluster has soil loss below 20 t ha(-1) year(-1). The soil loss from crop land varied from 2.9 to 3.6 t ha(-1) year(-1) in low rainfall years to 31.8 to 34.7 t ha(-1) year(-1) in high rainfall years with a mean annual soil loss of 12.2 t ha(-1) year(-1). The soil loss from crop land was higher in the month of August with an annual soil loss of 13.1 and 2.9 t ha(-1) year(-1) in normal and drought year, respectively. Based on the soil loss in a normal year, the interventions recommended for 85.3% of area of the watershed includes agronomic measures such as contour cultivation, graded bunds, strip cropping, mixed cropping, crop rotations, mulching, summer plowing, vegetative bunds, agri-horticultural system, and management practices such as broad bed furrow, raised sunken beds, and harvesting available water using farm ponds and percolation tanks. This methodology can be adopted for estimating the soil loss from similar ungauged watersheds with deficient data and for planning suitable soil and water conservation interventions for the sustainable management of the watersheds.

BACKGROUND: Basmati rice, originated in the foothills of Himalayas, commands a premium price in the domestic and international markets on account of its unique quality traits. The complex genetic nature of unique traits of Basmati as well as tedious screening methodologies involved in quality testing have been serious constraints to breeding quality Basmati. In the present study, we made an attempt to identify the genomic regions governing unique traits of Basmati rice. RESULTS: A total of 34 Quantitative Trait Loci (QTLs) for 16 economically important traits of Basmati rice were identified employing F(2), F(3) and Recombinant Inbred Line (RIL) mapping populations derived from a cross between Basmati370 (traditional Basmati) and Jaya (semi-dwarf rice). Out of which, 12 QTLs contributing to more than 15 % phenotypic variance were identified and considered as major effect QTLs. Four major effect QTLs coincide with the already known genes viz., sd1, GS3, alk1 and fgr governing plant height, grain size, alkali spreading value and aroma, respectively. For the remaining major QTLs, candidate genes were predicted as auxin response factor for filled grains, soluble starch synthase 3 for chalkiness and VQ domain containing protein for grain breadth and grain weight QTLs, based on the presence of non-synonymous single nucleotide polymorphism (SNPs) that were identified by comparing Basmati genome sequence with that of Nipponbare. CONCLUSIONS: To the best of our knowledge, the current study is the first attempt ever made to carry out genome-wide mapping for the dissection of the genetic basis of economically important traits of Basmati rice. The promising QTLs controlling important traits in Basmati rice, identified in this study, can be used as candidates for future marker-assisted breeding.

RESUMO

Kappa opioid receptor (KOR) is an important mediator of pain signaling and it is targeted for the treatment of various pains. Pharmacophore based mining of databases led to the identification of 2-aminobenzimidazole derivative as KOR agonists with selectivity over the other opioid receptors DOR and MOR. A short SAR exploration with the objective of identifying more polar and hence less brain penetrant agonists is described herewith. Modeling studies of the recently published structures of KOR, DOR and MOR are used to explain the receptor selectivity. The synthesis, biological evaluation and SAR of novel benzimidazole derivatives as KOR agonists are described. The in vivo proof of principle for anti-nociceptive effect with a lead compound from this series is exemplified.
