RESUMO

Various management practices can influence milk quality traits in dairy cattle. As an example, an increasing investment in automatic milking system to substitute milking parlors has been observed in the last 2 decades in dairy farms which could have affected certain bulk milk quality traits. What is more, milking practices can also affect certain milk parameters; as an example, teat disinfectants containing I are used in commercial farms where pre- or postdipping is performed, leading to presence of some I in the bulk milk. However, this trace mineral is also supplied in cows' diet to fulfill their nutritional requirements, partly contributing to the milk I final concentration. Therefore, the aim of this study was to evaluate the sources of variation of milk I along with other traditional milk quality traits. A total of 91 dairy farms in northeastern Italy were enrolled in the study. In each farm, diet and bulk milk samples were collected on the same day for chemical analysis. Concentration of I, in particular, was determined in both milk and feed with gold standard. Pearson correlations were calculated among the traits available for milk and diet, and a general linear model was used to test significance of fixed effects (feeding system, milking system, farming system, herd size, herd stage of lactation, and sampling month) on milk quality traits including the I concentration. In the case of milk I, diet I and presence of I-based predipping and postdipping teat disinfect application were also tested as fixed effects. Results showed a positive linear correlation between milk and diet I content (correlation coefficient [r] = 0.78). Although milk I was also positively correlated with lactose content (r = 0.25), dietary I was not correlated with other milk traits. Milk I content was significantly affected by dietary I, I-based predipping teat disinfectant application, and herd composition. Compared with conventional farms, organic farms showed lower protein content and greater somatic cell score (SCS) but similar milk I. Milking system significantly affected only lactose content and SCS of milk. Sampling month was only significant for milk urea nitrogen and herd composition, feeding system, herd size, and herd average days in milk did not modify milk gross composition and SCS. In conclusion, dietary supply of I is the main factor affecting milk I concentration and findings suggest that I level in milk can be naturally improved in dairy cows by modulating the I content in the diet administered. However, further research is needed to evaluate the effect of I-based sanitizers on milk I.

Moving from conventional (CMS) to automatic (AMS) milking systems could impact milk quality. Moreover, the type and preservation methods of the forages used in the total mixed ration (TMR) (such as alfalfa hay -HTMR- or corn silage -STMR-) have been demonstrated to modify milk composition. Thus, this study investigated the effect of implementing AMS and different diet forage types on the quality of Italian Holstein-Friesian bulk milk. Milk samples (n = 168) were collected monthly from 21 commercial farms in northern Italy during a period of 8 mo. Farms were categorized into 4 groups according to their milking system (CMS vs AMS) and diet's forage type (HTMR vs STMR). Milk quality data were analyzed through the mixed procedure for repeated measurement of SAS with the milking system, diet's forage type, and sampling day as fixed effects. Milking through the AMS led to lower milk fat, freezing point and ß-lactoglobulin A, longer coagulation time, and higher K content, pH and ß-lactoglobulin B than CMS. Cows fed STMR produced milk with greater fat, protein, casein, Mg content, titratable acidity and ß-lactoglobulin A, while reduced curd firming time, freezing point and ß-lactoglobulin B than those fed with HTMR. In conclusion, milk quality is not only altered by the diet's forage type and characteristics but also by the milking system.

Italian hard cooked types of cheese, like Parmigiano Reggiano and Grana Padano, are characterised by positive nutritional qualities. In fact, they contain substances that have particular biological activities, and therefore they can be fully considered, according to the definition given by the European Unit, as 'functional' foods. This short review concisely describes these components and the beneficial effects related to their activities. The description of the biologically active components has been organised in the following paragraphs: protein and peptides, fat and lipids, carbohydrates and prebiotics, probiotic bacteria, vitamins, mineral salts, and components of dairy products active in disease prevention. In particular, several known bioactive peptides were found in Parmigiano Reggiano cheese samples: for example, phosphopeptides, which are known for their mineral-binding capacity and vehiculation activity, peptides with immunomodulatory activity, and angiotensin-converting enzyme-inhibitory peptides with anti-hypertensive effects. Among lipids, the role of conjugated linoleic acid and other fatty acids present in these cheese types was taken into consideration. The presence of oligosaccharides with prebiotic properties and probiotic bacteria was also described. Finally, particular emphasis was given to highly available calcium and its impact on bone health.

Nitrogen content in urine plays a crucial role in assessing the environmental impact of dairy farming. Urine acidifications avoid urine nitrogen volatilization, but potentially lead to a degradation of creatinine, the most dependable marker for quantifying total urine excretion volume, affecting its measurement. This study aimed to assess how acidifying urine samples affects the concentration and detection of creatinine in dairy cattle. In this trial, individual urine samples from 20 Holstein lactating dairy cows were divided into three subsamples, allocated to 1 of 3 groups consisting of 20 samples each. Samples were immediately treated as follows: acidification with H2SO4 (1 mL of acid in 30 mL of sample) to achieve a pH < 2 (Group 1)); addition of an equal volume of distilled water (1 mL of distilled water in 30 mL of sample) to investigate dilution effects (Group 2); or storage without any acid or water treatment (Group 3). An analysis of creatinine levels was carried out using the Jaffe method. The Friedman test was employed to compare urine groups across treatments, and the Bland-Altman test was used to assess the agreement between measurements in Group 1 and Group 3. Urinary creatinine values were statistically different (p < 0.001) between Group 1 (median 48.5 mg/dL; range 36.9-83 mg/dL), Group 2 (median 47.5 mg/dL; range 36.5-80.7 mg/dL), and Group 3 (median 48.9 mg/dL, range 37.2-84). Bland-Altman analysis demonstrates agreement between Group 3 and Group 1. The measurement of urinary creatinine using the Jaffe method is affected by sample acidification, but the use of creatinine as a marker for total urine output could remain a viable tool when urine samples are acidified.

Feed additives including natural bioactive compounds (BCs) in combination with vitamin E (VitE) and organic Se could mitigate animal stress associated with intensive livestock farming due to their anti-inflammatory and antioxidant properties. Yeast and yeast derivate are included in feed additives as probiotic products and digestion promoters. Scutellaria baicalensis is a source of bioactive compounds and has been tested in monogastrics, exhibiting many immunostimulating and hepato-protective activities. However, the literature lacks information regarding S. baicalensis effects on beef cattle performance and health status. The aim of the present study was to evaluate the impact on beef cattle's feed intake, health and oxidative status, and growth performances of the inclusion of a commercial supplement (CS) containing VitE, organic Se, yeast derivate, and S. baicalensis extract during the fattening and finishing period. A total of 143 Charolaise male cattle were allotted into 12 pens of 11-12 animals each and assigned to a control ( 463.9 ± 21.48 body weight - BW) or a treated ( 469.8 ± 17.91  BW) group. Each group included two replicates of three pens. The treated groups were supplemented with 20  g CS animal - 1 d - 1 . Feed intake was measured monthly on a pen base during two consecutive days. Total mixed ration and fecal samples were collected at three time points (monthly, from November to February) and pooled by replicate for the analyses to monitor digestibility. Blood samples were individually collected at the beginning and at the end of the trial for oxidative status and metabolic profile determination. Final BW and carcass weight were individually recorded to calculate average daily gain, feed conversion ratio, and carcass yield. Similar feed digestibility between groups were observed during the whole experiment. Feed intake, growth performances, final body weight, average daily gain, feed conversion rate, oxidative status, and metabolic profile were not affected by the dietary inclusion of the tested CS indicating no detrimental effect of the treatment. Different doses of this product should be tested in the future in order to provide a more complete report on the product efficacy.

There is limited information on the impact of dietary supplementation with separate rumen-protected (RP) amino acids (AA), or with their combination, on ewes' oxidative status. Sixty ewes were divided into five groups; C: basal diet (control); M: basal diet + 6 g/ewe RP methionine; L: basal diet + 5 g/ewe RP lysine; LML: basal diet + 6 g methionine and 5 g lysine/ewe; and HML: basal diet + 12 g methionine + 5 g lysine/ewe. Milk's fat content increased in RP-AA fed ewes, while that of protein in M and L only. In blood plasma, the malondialdehyde (MDA) content was reduced in the M, LML, and HML compared to C-fed ewes. An increase in glutathione transferase activity in the blood plasma of the M and LML compared to the C and HML-fed ewes were found. In milk, lower values of the ferric reducing ability of plasma (FRAP) in the LML and HML-fed ewes and of 2,2'-Azino-bis 3-ethylbenzthiazoline-6-sulfonic acid (ABTS) in LML only, were found. Lysine increased milk's FRAP values and MDA content. Both L and HML diets increased milk's protein carbonyls content. Methionine improves the organism's oxidative status, without adversely affecting milk's oxidative stability. Lysine dietary inclusion affects negatively the oxidative stability of milk.

This study aimed to determine the impact of sesame meal, selenium (Se), and vitamin E (VitE) on goats' oxidative status. Thirty mid-lactation crossbred goats were divided into five homogeneous groups, and were fed 1 kg of alfalfa hay and 1.2 kg of concentrates daily. The control group (C) received a basal diet. In the concentrates of the treated groups, 10% of the soybean meal was replaced by sesame meal and no extra VitE or Se (SM), or an extra 60 mg of VitE (SME), or 0.1 mg organic Se (SMSe), or their combination (60 mg VitE and 0,1 mg organic Se/kg of concentrate (SMESe). In the plasma of the goats, the dietary treatments did not affect glutathione reductase, glutathione peroxidase, glutathione transferase, catalase, superoxide dismutase activities, malondialdehyde (MDA) content, or the total antioxidant capacity. A reduction and a trend for lower protein carbonyls content was found in goats fed SM (p = 0.03) and SME (p = 0.06) compared to SMESe. In the milk, the lactoperoxidase activity decreased with SMSe and SMESe. A numerical decrease in the total antioxidant capacity and an increase in the MDA content in the milk of the SMESe group compared with the other treated groups was found. In mid-lactation goats, SM improves the oxidative status of both the organism and the milk.

Scientific evidence demonstrates that plant feed additives (PFA) can be a viable alternative to synthetic antioxidant vitamins in poultry nutrition. PFA are represented by plants, essential oils, plant extracts, and by-products from herbal or crop processing. The use of PFA in the feed industry has increased in recent years as their biologically active compounds (polyphenols) have demonstrated antimicrobial and antioxidant effects in food-producing animals. However, few trials have directly compared the effects of PFA with synthetic vitamins. After a systematic literature review of studies comparing the effects of PFA and synthetic vitamins on poultry products in the last 20 years (2000-2020), a total of 44 peer-reviewed articles were included in the present work. A positive effect of PFA on poultry products' oxidative stability during storage, organoleptic characteristics, and fatty acids profile has been observed without a specific impact on their performances. The effects of PFA are variable but often similar to those of vitamin E, suggesting the opportunity for a partial substitution of the latter in poultry diets.

In the last two decades, the interest in natural plant feed additives (PFA) as alternatives to synthetic vitamins in livestock nutrition has increased. After a systematic review, a total of 19 peer-reviewed papers published between 2000 and 2020 were retained to evaluate the antioxidant effects of PFA compared to synthetic antioxidant vitamins (mainly vitamin E; VitE) in livestock nutrition. These studies demonstrated that PFAs could be as efficient as VitE in counteracting oxidative stress in pigs, rabbits, and ruminants. However, PFAs only positively affected animals' growth performance and feed efficiency in some monogastric studies. The PFA can affect antioxidant enzyme activity in a dose- and method of administration-dependent manner. The antioxidant capacity of both PFA and VitE were depressed in cows fed with diets rich in polyunsaturated fatty acids. Variability among studies could be related to species differences. Despite the interest of the feed industry sector in PFA, there are still very few studies evaluating their antioxidant effect in species other than poultry.

Plant feed additives (PFA) such as essential oils, extracts, and by-products from plant processing can be included in poultry diets. A total of 39 peer-reviewed articles were selected from the literature published in the last 20 years (2000-2020) comparing PFA antioxidant effects with synthetic antioxidant vitamins (mainly vitamin E) in poultry nutrition. The PFA can be used as an effective nutritional strategy to face poultry's oxidative stress with positive impact also on their productivity and efficiency. They can partially or completely replace antioxidant synthetic vitamins (the latter administered at doses between 150 and 500 mg/kg) in animal diets, sometimes affecting important physiological functions or expressing synergistic effect with the synthetic antioxidants. It is crucial to take into consideration the issues related to the absorption and the metabolism of these additives and their interaction with gut microbiota. However, some form- and dose-dependent negative effects on growth performances are observed.

The interest for safe and natural foods of animal origin is currently increasing the use of plant feed additives (PFA) as antioxidants in animal nutrition. However, studies with livestock animals dealing with PFA as antioxidants are scarce. The aim of the present review was to evaluate the antioxidant impact of PFA compared with synthetic vitamins on animal food product yield and quality. For this purpose, peer-reviewed studies published between 2000 and 2020 were collected. Most papers were carried out on ruminants (n = 13), but PFA were also tested in swine (n = 6) and rabbits (n = 2). The inclusion of PFA in the diets of pigs, rabbits, and ruminants improved the products' quality (including organoleptic characteristics and fatty acids profile), oxidative stability, and shelf life, with some impacts also on their yields. The effects of PFA are diverse but often comparable to those of the synthetic antioxidant vitamin E, suggesting their potential as an alternative to this vitamin within the diet.

Rumen protected amino acids inclusion in ewes' diets has been proposed to enhance their innate immunity. The objective of this work was to determine the impact of dietary supplementation with rumen-protected methionine or lysine, as well as with a combination of these amino acids in two different ratios, on the expression of selected key-genes (NLRs, MyD88, TRIF, MAPK-1, IRF-3, JunD, TRAF-3, IRF-5, IL-1α, IL-10, IKK-α, STAT-3 and HO-1). Thus, sixty Chios dairy ewes (Ovis aries) were assigned to one of the following five dietary treatments (12 animals/ treatment): A: basal diet consist of concentrates, wheat straw and alfalfa hay (control group); B: basal diet +6.0 g/head rumen-protected methionine; C: basal diet + 5.0 g/head rumen-protected lysine; D: basal diet +6.0 g/head rumen-protected methionine + 5.0 g/head rumen-protected lysine and E: basal diet +12.0 g/head rumen-protected methionine + 5.0 g/head rumen-protected lysine. The results revealed a significant downregulation of relative transcript level of the IL-1α gene in the neutrophils of C and in monocytes of D ewes compared with the control. Significantly lower mRNA transcript accumulation was also observed for the MyD88 gene in the neutrophils of ewes fed with lysine only (C). The mRNA relative expression levels of JunD gene were highly induced in the monocytes, while those of IL-10 and HO-1 genes were declined in the neutrophils of ewes fed with the C and D diets compared with the control. Lower transcript levels of STAT-3 gene were observed in the neutrophils of ewes fed with either C or with E diets in comparison with the control. In conclusion, our results suggest that the dietary supplementation of ewes with rumen-protected amino acids, down regulate the expression of some genes involved in the pro-inflammatory signalling.

This study aimed to assess the feasibility of visible/near-infrared reflectance (Vis-NIR) and near-infrared transmittance (NIT) spectroscopy to predict total and gelatinized starch and fiber fractions in extruded dry dog food. Reference laboratory analyses were performed on 81 samples, and the spectrum of each ground sample was obtained through Vis-NIR and NIT spectrometers. Prediction equations for each instrument were developed by modified partial least squares regressions and validated by cross- (CrV) and external validation (ExV) procedures. All studied traits were better predicted by Vis-NIR than NIT spectroscopy. With Vis-NIR, excellent prediction models were obtained for total starch (residual predictive deviation; RPDCrV = 6.33; RPDExV = 4.43), gelatinized starch (RPDCrV = 4.62; RPDExV = 4.36), neutral detergent fiber (NDF; RPDCrV = 3.93; RPDExV = 4.31), and acid detergent fiber (ADF; RPDCrV = 5.80; RPDExV = 5.67). With NIT, RPDCrV ranged from 1.75 (ADF) to 2.61 (acid detergent lignin, ADL) and RPDExV from 1.71 (ADL) to 2.16 (total starch). In conclusion, results of the present study demonstrated the feasibility of at-line Vis-NIR spectroscopy in predicting total and gelatinized starch, NDF, and ADF, with lower accuracy for ADL, whereas results do not support the applicability of NIT spectroscopy to predict those traits.

The aim of the present study was to investigate the ability of a handheld near-infrared spectrometer to predict total and gelatinized starch, insoluble fibrous fractions, and mineral content in extruded dry dog food. Intact and ground samples were compared to determine if the homogenization could improve the prediction performance of the instrument. Reference analyses were performed on 81 samples for starch and 99 for neutral detergent fiber (NDF), acid detergent fiber (ADF), acid detergent lignin (ADL), and minerals, and reflectance infrared spectra (740 to 1070 nm) were recorded with a SCiO™ near-infrared (NIR) spectrometer. Prediction models were developed using modified partial least squares regression and both internal (leave-one-out cross-validation) and external validation. The best prediction models in cross-validation using ground samples were obtained for gelatinized starch (residual predictive deviation, RPD = 2.54) and total starch (RPD = 2.33), and S (RPD = 1.92), while the best using intact samples were obtained for gelatinized starch (RPD = 2.45), total starch (RPD = 2.08), and K (RPD = 1.98). Through external validation, the best statistics were obtained for gelatinized starch, with an RPD of 2.55 and 2.03 in ground and intact samples, respectively. Overall, there was no difference in prediction models accuracy using ground or intact samples. In conclusion, the miniaturized NIR instrument offers the potential for screening purposes only for total and gelatinized starch, S, and K, whereas the results do not support its applicability for the other traits.

Due to the increasing interest in organic farming, an overview of this research area is provided through a bibliometric analysis conducted between April and May 2019. A total of 320 documents were published up until 2018 on organic livestock farming, with an annual growth rate of 9.33% and a clear increase since 2005; 268 documents have been published in 111 journals. Germany is the country with the largest number of published papers (56 documents). Authors' top keywords (excluding keywords used for running the search) included: animal welfare (29 times), animal health (22 times), cattle (15 times), grazing (10 times), and sheep (10 times). This could indicate that more research has been done on cattle because of the importance of this species in Germany. Moreover, the prevalence of the terms 'animal welfare' and 'animal health' may indicate that the research on organic livestock production has been focused on these two areas. The bibliometric analysis indicates that: i) countries focused the organic livestock production research on their main production, and ii) more research in species other than cattle and sheep is needed.

: Our objectives were to determine prevalence and intensity of gastrointestinal parasites of dead or euthanized emaciated moose in central and southern Sweden ( n=50) and to assess parasite intensity as a major contributing factor in the poor condition of these moose. All animals were infected and most had gastrointestinal nematodes. Seven parasite species were found in the abomasa and 10 species were found in the small intestine. Coinfections were commonly found in the abomasum ( Ostertagia antipini and Mazamastrongylus dagestanica) and in the small intestine ( Nematodirella alcidis and Trichostrongylus capricola). Moose had many nematodes; the highest numbers were 224,400 and 11,760, in the abomasum and in the small intestine, respectively. Fourteen moose had more than 40,000 gastrointestinal nematodes (excluding large intestine nematodes represented by Trichuris spp. and Oesophagostomum venulosum). Additionally, a moderate prevalence (36%) of protostrongylid larvae (dorsal spine larvae) and a low prevalence (2-4%) of protozoal infections were identified at microscopic examination of feces. Adult moose had significantly more parasites than did subadults. The results did not show parasite load as a single or a major cause of the moose mortality, but they provided an update on the species composition of helminth parasites in moose in Sweden and illustrated the extreme infection intensities that free-ranging moose can have.

The royal python (Python regius) is commonly bred in captivity. To have a successful breeding season, accurate monitoring of the reproductive activity is necessary. The use of non-invasive monitoring methods in exotics is important in order to minimize stress. For this purpose ultrasound has been anecdotally used to monitor royal python reproductive activity. However, there is limited information regarding the reproductive cycle of this species. The aim of the present study is to monitor the female reproductive cycle of the royal python using ultrasonography and gonadal steroid metabolite measurements in the faeces. The reproductive activity of one hundred twenty-nine adult female P. regius was examined during two consecutive years. We performed brief scans on non-anaesthetized snakes using a portable ultrasound system and a 10-12 MHz linear array transducer (MyLab™ 30 Gold, Esaote). Ultrasound features, dimension and echogenicity of the reproductive structures were determined. During the second reproductive cycle, the hormonal profiles of 30 animals were also evaluated, with a monthly collection of faecal samples. These samples were classified according to reproductive stage, as identified by ultrasonographic examination, and the mean faecal progesterone and 17ß-estradiol levels were calculated using the results from an enzyme-linked immunosorbent assay (ELISA). Progesterone levels increased during the reproductive cycle. Estradiol levels showed greater variability, although they appeared to increase before coupling when compared to the levels between coupling and egg laying. The present study suggests that it is possible to identify different phases in the female royal python reproductive cycle: anovulatory phase, transition, folliculogenesis and embryogenesis. Ultrasound is also useful for identifying follicular regression or slugs. Gonadal steroid metabolite measurements from the faeces could help integrate reproductive information. The use of ultrasonography in addition to the steroid metabolite measurement in the faeces gives an accurate picture of ovarian activity in captive adult female royal pythons.

Masked mycotoxins are plant metabolites of mycotoxins that contaminate food and feed. They pose health concern as the shortage of toxicological data forces the lack of regulation worldwide. The present work investigated the toxicological relevance of the masked mycotoxin zearalenone-14-glucoside. In vitro, it shows a lower toxicity in respect to the parent compound. However, the major risks related to the consumption of masked mycotoxins depend on the possibility to undergo hydrolysis. Therefore, the hydrolysis and further transformation of zearalenone-14-glucoside in bovine blood and blood components (i.e. plasma, serum and serum albumin) were monitored using LC/MS-MS analysis to gain insights on the possible systemic fate. Hydrolysis was observed in all matrices, and both cell-dependent and -independent contributions were pointed out. Moreover, further metabolism was observed in the whole blood as zearalenol isomers were found. Serum albumin was identified among the active components, and the protein-ligand interaction was investigated via computational analysis. The blood has been pointed out as possible district of reversion and further activation of zearalenone-14-glucoside, and a similar fate cannot be excluded for other masked mycotoxins. Therefore, the systemic hydrolysis should be evaluated beside the absorption, bioavailability and bioaccessibility to deeply understand the toxicity of masked mycotoxins.

There are no reports of saccharolytic enzymes being used in the preparation of formulations for animal semen extenders. In the present study, the use of an innovative semen extender (Formula12®) in the long-term liquid storage of boar semen at 17°C was evaluated. The formulation included use of a disaccharide (sucrose) as the energy source precursor coupled to an enzymatic agent (invertase). The innovative extender was evaluated and compared in vitro to a commercial extender (Vitasem LD®) for the following variables: Total Motility (TM), Forward Progressive Motility (FPM), sperm morphology, membrane integrity, acrosome integrity, and chromatin instability. Boar sperm diluted in Formula12® and stored for 12 days at 17°C maintained a commercially acceptable FPM (>70%). Using the results from the in vitro study, an AI field trial was performed. A total of 170 females were inseminated (135 with Formula12® and 35 with Vitasem LD®). The pregnancy rates were 97.8% compared with 91.4%, and the farrowing rates were 96.3% compared with 88.6% when Formula12® and Vitasem LD® were used, respectively. The mean number of piglets born/sow were 14.92±0.46 compared with 13.83±0.70, and the number of piglets born alive/sow were 14.07±0.46 compared with 12.12±0.70 (P<0.05). The results obtained in this study demonstrated that use of the innovative concept to provide a precursor of glucose and fructose as energy sources for an enzymatic agent in an extender allowed for meeting the metabolic requirements of boar sperm during storage at 17°C. It is suggested that there was a beneficial effect on fertilizing capacity of boar sperm in the female reproductive tract with use of these technologies.

Content of cyclopropyl and ω-cyclohexyl fatty acids of microbial origin, respectively, from silage and rumen, was determined by GC-MS and confirmed by (1)H NMR in more than 200 milk samples from dairy cows fed with different forages. Cyclopropyl fatty acids (about 0.1% of milk fat) were detected for the first time in milk and they were present only in milk samples from cows fed with forages containing maize silage, which is not allowed to produce milk for some PDO cheeses as Parmigiano-Reggiano. Their determination can be proposed as a quality parameter of milk or feed and it can be useful especially to distinguish cheeses sold as Parmigiano Reggiano from others cheeses. The content of ω-cyclohexyl tridecanoic acid varied from 0.0% to 0.15% of milk fat, and it was higher in milk samples from cows fed with diets richer in cereal meals.
