Several essential nutrients such as Fe, Co, Cu, I, Se, and Zn are vital to grazing cattle as they play a significant role in intracellular enzyme systems, with antioxidant and repair functions of DNA lesions. Feeds may supply most essential nutrients in adequate amounts. However, essential nutrient deficiencies are common and are mainly related to the geochemical characteristics of the soils, inherited from parent rocks. This study aims to assess the concentrations of the selected essential nutrients in topsoils and pasture grass from São Miguel Island (Azores) and discuss the possible effects of its deficiency in grazing cattle health. Sixty-eight samples of agricultural (pastures) topsoil and thirty samples of pasture grass were collected throughout São Miguel Island in areas with basaltic and trachytic parent rocks. Soil physicochemical properties and geochemical baselines of selected elements were determined on composite samples of soil and pasture grass in each site. Results indicate that the highest concentrations of the selected essential nutrients were observed in areas with basaltic soils, while the lowest values were found in areas with trachytic soils. These differences are considered of geogenic origin since they result from the weathering of volcanic parent rocks with different geochemical compositions. Data indicate a lack of some essential nutrients in topsoils (Co and Se) and/or pasture grass (Co, Cu, Se, and Zn) that can contribute to harmful health effects on grazing cattle. The uneven distribution of essential nutrients in topsoil and pasture grass and their deficiency can lead to several health problems in the cattle since these elements regulate physiological functions in biological systems. The identification of these deficit sites is important as it can help farmers to implement soil and/or animal supplementation programs to diminish possible health problems.

Antibiotics are successful drugs used in human and animal therapy; however, they must be considered as environmental pollutants. This study aims to isolate and characterize the extended-spectrum ß-lactamase (ESBL) producing Escherichia coli soil from Azores Archipelago subjected to livestock agricultural practices. Twenty-four soil samples were collected from three different pasture systems with different number of cattle heads, and from a control site. Antibiotic susceptibility method was performed by Kirby-Bauer disk diffusion method against 16 antibiotics, and the presence of genes encoding lactamases, antimicrobial resistance genes, virulence factors, and phylogenetic groups was determined by polymerase chain reaction (PCR). Nine ESBLs were recovered from the three grazing sites, and all isolates presented the beta-lactamase genes blaCTX-M-3 and blaSHV. E. coli isolates were resistance to tetracycline and streptomycin and harbored the tetB, strA, and strB genes. One isolate also showed resistance to sulfonamides, and the genes sul1 and sul2 were detected. The isolates were grouped into the following phylogenic groups: B1 (n = 6), D (n = 2), and A (n = 1). The presence of antibiotics and resistance genes in soils may be the source to the development of antimicrobial resistance, which may have negative consequences in human and animal health.

Hydrothermal areas are potentially hazardous to humans as volcanic gases such as radon (222Rn) are continuously released from soil diffuse degassing. Exposure to radon is estimated to be the second leading cause of lung cancer, but little is known about radon health-associated risks in hydrothermal regions. This cross-sectional study was designed to evaluate the DNA damage in the buccal epithelial cells of individuals chronically exposed to indoor radon in a volcanic area (Furnas volcano, Azores, Portugal) with a hydrothermal system. Buccal epithelial cells were collected from 33 individuals inhabiting the hydrothermal area (Ribeira Quente village) and from 49 individuals inhabiting a non-hydrothermal area (Ponta Delgada city). Indoor radon was measured with Ramon 2.2 detectors. Chromosome damage was measured by micronucleus cytome assay, and RAPD-PCR was used as a complementary tool to evaluate DNA damage, using three 10-mer primers (D11, F1 and F12). Indoor radon concentration correlated positively with the frequency of micronucleated cells (r s = 0.325, p = 0.003). Exposure to radon is a risk factor for the occurrence micronucleated cells in the inhabitants of the hydrothermal area (RR = 1.71; 95% CI, 1.2-2.4; p = 0.003). One RAPD-PCR primer (F12) produced differences in the banding pattern, a fact that can indicate its potential for detecting radon-induced specific genomic alterations. The observed association between chronic exposure to indoor radon and the occurrence of chromosome damage in human oral epithelial cells evidences the usefulness of biological surveillance to assess mutations involved in pre-carcinogenesis in hydrothermal areas, reinforcing the need for further studies with human populations living in these areas.

Concentrations of cadmium (Cd), mercury (Hg), and lead [Pb (µg g(-1) wet weight)] were determined in liver and muscle samples of 15 bigeye (Thunnus obesus) and 15 skipjack tunas (Katsuwonus pelamis) caught over an active volcanic region in the Mid-Atlantic Ocean (Azores, Portugal) and evaluated regarding consumption safety. None of the muscle samples (edible part) exceeded the European Union (EU) maximum limits (MLs) for Hg and Pb. Cd concentrations in muscle were much greater than EU MLs with 53 and 26 % of the bigeye tuna and skipjack tuna, respectively, in exceedance of the limits. Results obtained in this work, together with other studies in the same region, support the existence of an important volcanic source of Cd in waters of the Mid-Atlantic region, which should be carefully monitored given the importance of many commercial marine species for human consumption, mainly in Europe.

Worldwide, disorders of the thyroid gland are a growing concern; such can be caused by exposure to contaminants, including agrochemicals used in conventional agriculture, which act as endocrine disruptors. The purpose of this study is to evaluate whether or not exposure to an environment with conventional agriculture leads to thyroid disruption. Mus musculus were used as bioindicator species, captured in two sites: a farm where conventional agriculture is practiced, and a place without agriculture. Thyroid histomorphometric and morphologic data were analyzed. The impacts of the agricultural environment over the thyroid were revealed, as indications of hypothyroidism were observed in exposed mice: the area and volume of epithelial cells were much lower. Alterations in thyroid histomorphology were also observed: lower follicular sphericity, irregularly delimited epithelium and increased exfoliation into the colloid. These results highlight the need for transition from current conventional agricultural systems towards organic systems.

BACKGROUND: In areas where cattle graze outdoors, the environmental availability of trace elements is of utmost importance for an adequate intake. Cattle hair trace elements can be used as exposure biomarkers to assess animal intake and help ensure good nutrition and animal welfare. The aim of this study was to evaluate the environmental exposure to trace elements in cattle grazing outdoors in a volcanic region (São Miguel Island, Azores) and assess the corresponding bioaccumulation factors. METHODS: Twenty-nine samples of pasture topsoil were collected in two areas of São Miguel with different geological characteristics: Picos fissure system (basaltic area) and Povoação volcano (trachytic area). Hair from fifty-four animals (calves and dairy cows) was collected in two grazing sites: Ponta Delgada (Picos fissure system) and Povoação (Povoação volcano). All samples were analysed by inductively coupled plasma mass spectrometry (ICP-MS) after digestion in aqua regia. RESULTS: Basaltic soils of the Picos fissure system had significantly higher concentrations of most trace elements, except Mo, when compared with the trachytic soils of Povoação volcano. Hair of calves from Povoação showed higher concentrations of most essential trace elements, except Mg, Cu, and Zn, than hair from calves of Ponta Delgada (considered in this study as a control group since they are raised in a barn). On the other hand, hair from dairy cows grazing in Ponta Delgada showed higher concentrations of most essential trace elements, except Mg, As, and Mo, when compared with hair of cows grazing in Povoação. CONCLUSIONS: Cattle hair trace elements are suitable biomarkers for the assessment of the environmental exposure to trace elements in volcanic soils. The results from dairy cows support the uneven distribution of essential trace elements (and their bioavailability) in the different soils of São Miguel, reinforcing the need to graze animals outdoors from an early age. Such practice (grazing cattle outdoors) provides another means of accessing essential trace elements that complemented with feed can reduce the natural depletion of these essential elements in the body due to reproductive phenomena and milk production.

Iodine deficiency, the most common cause of preventable mental impairment worldwide, has been linked to poorer intellectual function in several studies. However, to our knowledge, no studies have been performed in moderate iodine-deficient schoolchildren using the complete form of Wechsler Intelligence Scale for Children (WISC-III; Portuguese version). The main purpose of this study was to ascertain whether moderate iodine deficiency would affect the cognitive function of schoolchildren (7-11 years old; 3rd and 4th grades). Raven's Colored Progressive Matrices (CPM; Portuguese version) were used for measuring the intelligence quotient (IQ) of the total population (n = 256; median UIC = 66.2 µg/L), and the WISC-III was used to study two selected subgroups: one moderately iodine-deficient (n = 30) and the other with adequate iodine intake (n = 30). WISC-III was shown to be the prime instrument for cognitive function assessment among moderate iodine-deficient schoolchildren; this subgroup had a Full-Scale IQ 15.13 points lower than the adequate iodine intake subgroup, with a magnitude effect of d = 0.7 (p = 0.013). Significant differences were also registered in 6 of the 13 Verbal-Performance IQ subtests. Moderate iodine deficiency has a substantial impact on mental development and cognitive functioning of schoolchildren, with significant impairment in both Performance IQ and Verbal IQ spectrum, adversely impacting their educational performance.

For the first time, the structure, dimensions, and composition of the epidermis of an invasive earthworm species that has successfully colonized hostile conditions in actively volcanic soil on São Miguel (Azores) have been measured. Metal concentrations in actively volcanic (Furnas) and volcanically inactive (Fajã) soils were similar; however, Furnas soil was characterised by elevated temperature (10°C differential), relative hypoxia, extremely high CO(2) tension, and accompanying acidity. The epidermis of earthworm's resident at Fajã was approximately twice the thickness of the epidermis of conspecifics resident in Furnas soil. Reference worms transferred to Furnas soil for 14 days experienced an epidermal thinning of approximately 51%. In comparison, when Furnas earthworms were transferred to mesocosms at the relatively benign Fajã site, their epidermal thickness increased by approximately 21% over 14 days. Earthworms resident in Furnas soil had higher goblet cell counts than the residents of volcanically inactive soil on a neighbouring island (S. Maria). Transferring worms from S. Maria to mesocosms at Furnas induced a significant increase in goblet cell counts. Clearly, the active volcanic environment at Furnas poses a multifactorial stress challenge to the epigeic A. gracilis colonizer.

Smaller oceanic islands, often hosting endangered native habitats, are particularly vulnerable to the impact of human activities. Using lichens as bioindicators, this study aimed to test if agricultural (AGR) and artificial (ART) land uses are noticeably more impacted than forest (FOR) land use on an oceanic island (São Miguel, Azores). Livestock and farming practices in AGR areas involve the intensive application of synthetical agrochemicals as well as organic fertilizers and manure. ART areas accommodate vehicular traffic besides industries dedicated to waste management, energy production or exploration and transformation of raw materials. Naturally occurring Parmotrema lichens were collected in 28 sampling sites distributed between each land use. The concentrations of 58 elements as well as the percentage (%N) and the isotopic composition of nitrogen (δ15N) were determined on lichen samples. An overall pattern of significant elemental enrichment was observed in lichens from AGR and ART sites compared with FOR lichens, including several rare-earth elements. FOR lichens were noticeably cleaner, thus providing background concentrations for the calculation of bioaccumulation ratios. Bioaccumulation levels were generally low to moderate in AGR lichens and moderate to high in ART lichens, including toxic heavy metals. %N was highest in AGR lichens and its isotopic signature was distinguishable from ART lichens by significantly lower δ15N values. This study provides a comprehensive baseline of bioaccumulation data across major land uses for comparison with other insular regions, highlighting the greater vulnerability of island ecosystems to anthropogenic impacts even if by relatively small-scale human activities.

Recent studies have shown that exposure to hydrothermal emissions has a negative impact on the respiratory system. Still, volcanogenic air pollution studies are still outnumbered when compared to anthropogenic studies which can result in an unknown risk to the human populations living near volcanically active areas. This study was carried out in São Miguel Island, with noneruptive volcanically active environments, such as the Furnas volcano caldera. Its noneruptive volcanism presents itself as hydrothermal emissions, mainly by the release of nearly 1000 T d-1 of CO2 along with H2S, and the radioactive gas radon; metals [e.g., mercury (Hg), cadmium (Cd), copper (Cu), and zinc (Zn)] and particulate matter are also released in a daily basis. We test the hypothesis whether chronic exposure to hydrothermal emissions causes pulmonary oxidative stress, using Mus musculus as a surrogate species. Mus musculus was live-captured in two villages with hydrothermal emissions and one village without any type of volcanic activity. The level of pulmonary oxidative stress was immunohistochemically assessed by using an OxyIHCTM Oxidative stress detection kit, and the detection of terminal deoxynucleotidyl transferase (TdT)-mediated dUTP nick end-labeling (TUNEL) was used to evaluate apoptosis in lung tissues. Mice chronically exposed to hydrothermal emissions presented increased levels of oxidative stress and amount of apoptotic cells. We demonstrate, for the first time, the high oxidative stress potential in the lungs of mice chronically exposed to hydrothermal emissions. This study highlights the usefulness of M. musculus as a bioindicator species and enforces the necessity of regularly biomonitor the inhabitants of hydrothermal areas to prevent respiratory pathologies.

The particularities of volcanic soils raise the need to better understand the link between soil agricultural management intensity and trace metal bioaccumulation. The Azores are a region characterized by volcanic soils, which were changed in different degrees according to the intensity of the agricultural practices. The main objective of this study was to assess the potential ecotoxicological effects of the trace metals present in volcanic pastureland soils along a gradient of management intensity (i.e., semi-natural, permanent and reseeded), using earthworms (Eisenia fetida) as biological indicators. For this purpose earthworms were exposed during 7, 14, 28 and 56 days to soils from the three types of pastures. At each exposure time, we quantified trace element bioaccumulation (As, Cd, Co, Cr, Cu, Hg, Li, Mn, Mo, Ni, Pb, Rb, U, V and Zn) and the activities of superoxide dismutase and acetylcholinesterase in earthworm tissues. Overall, the results showed that the type of pastureland management significantly increased the soil contents in trace metals: V, Co, Ni and Zn in semi-natural pasturelands; As, Cd and Hg in reseeded pasturelands; and, Rb and U in both permanent and reseeded pasturelands. The soil physicochemical properties observed in the reseeded pastureland systems (higher electric conductivity values associated with a moderately acid pH value) modulated the metal bioavailability, from soil to biota, leading to a greater Hg bioaccumulation in earthworm tissues. The long-term exposure (56 days) of earthworms to reseeded pastureland soil was associated with adverse biological effects (intensification of AChE activity and decrease of SOD activity), encompassing key processes such as neurotransmission and antioxidant defence mechanisms in resident soil biota (earthworms). This study point towards the increased importance of semi-natural and permanent pastureland management, over the intensive management (reseeded pasturelands), in favour of more sustainable ecosystems.

Abalones are considered a delicacy and exploited for food worldwide. In many places, overfishing has led to the decimation of wild stocks and many are now reared in aquaculture systems. In the Azores, there is no tradition of eating abalones and Haliotis tuberculata stocks are still virtually untouched. However, as tourism in the islands grows and the stocks of other shellfish diminish, there is an increasing pressure to find alternative edible resources, leading to a rising interest in abalones. According to previous studies performed in the region, other edible species, including the local highly appreciated limpets and the giant barnacle, present high concentration levels of some heavy metals, which has been attributed to the volcanic origin of the islands. Here we analysed the metal content in the edible tissue of Haliotis tuberculata from São Miguel Island, Azores. The potential human health risks due to its consumption was assessed by estimating the average daily intake (EDI) and target hazard quotient (THQ) of metals. Similarly to other organisms in the Azores, abalones have higher than normal levels of some heavy metals, particularly cadmium, reflecting a local natural source that should be closely monitored from a public health point of view.

BACKGROUND: Previous studies have provided some evidence of a possible association between cancer and metallothioneins. Whether this relates to an exposure to carcinogenic metals remains unclear. METHODS: In order to examine the association between the expression of metallothioneins and bladder tumors, and to compare the levels of arsenic, cadmium, chromium, lead and nickel in animals with bladder tumors and animals without bladder tumors, 37 cases of bovine bladder tumors and 17 controls were collected. The detection and quantification of metallothioneins in bladder tissue of both cases and controls was performed by immunohistochemistry. And the quantification of metals in tissue and hair was assessed by inductively coupled plasma - mass spectrometry. RESULTS: Increased expression of metallothioneins was associated with bladder tumors when compared with non-tumoral bladder tissue (OR = 9.3, 95% CI: 1.0 - 480). The concentrations of cadmium, chromium, lead and nickel in hair of cases were significantly higher than those of controls. However, as for the concentration of metals in bladder tissue, the differences were not significant. CONCLUSION: Though the sample size was small, the present study shows an association between bladder tumors and metallothioneins. Moreover, it shows that concentrations of metals such as cadmium, chromium, lead and nickel in hair may be used as a biomarker of exposure.

Fasciolosis, caused by the trematode Fasciola hepatica, is considered one of the most important parasitosis in dairy cattle due to significant economic losses. The main objectives of this study were to determine the prevalence of fasciolosis in dairy cattle slaughtered at São Miguel Island (SMI) slaughterhouse and to ascertain areas (parishes and dairy herds), within the island, with highest risk for fasciolosis. The association among cattle age and weight with the occurrence of fasciolosis was also determined. From a sample of 24,389 cattle slaughtered in two years (2015-2016), a total of 1134 dairy cattle livers were inspected and, during this period, 531 were positive to fasciolosis and 603 presented no typical fasciolosis lesions. This study showed that fasciolosis has a broad geographical distribution on SMI and that is also found in cattle from other islands of the Azores archipelago, namely Flores Island. Although there were no significant differences in weight between healthy animals and parasitized ones, we observed that the older animals present a greater level of liver lesions and a higher level of parasitization, reflecting their longer period of exposure to the risk factor (ingestion of metacercariae in pastures infested by Galba truncatula).

Human populations living in volcanically active areas are chronically exposed to volcanogenic air pollution, potentially contributing to long-term adverse health effects. However, mapping chronic exposure is difficult due to low spatial resolution of monitoring data on air pollutants and the need for time integration. To overcome these problems, lichens were tested as ecological indicators of hydrothermal volcanic air pollution, considering their bioaccumulation capacity over time, by transplanting them from a reference area to several sites (n = 39) in a volcanic area. The test was developed at Furnas volcano (Azores, Portugal). A stratified sampling design was followed using previous measurements of soil CO2 flux at ground level and the distance to the main fumarolic fields. After 6 months of exposure, lichen transplants were analyzed for S isotopic ratio (δ34S), which strongly related with the distance to fumarolic fields on a logarithmic regression, serving as an appropriate hydrothermal exposure biomarker. Considering kriging interpolated δ34S values as tracer of airborne hydrothermal emissions and habitational areas as proxy of ongoing human presence, a map was built relating both information per area unit to spatially model risk areas. It was estimated that 26% of habitational areas in the study area stand at high or very high risk of outdoors chronic exposure to airborne hydrothermal emissions. This methodologic approach to produce chronic exposure risk maps is applicable to other volcanically active and inhabited areas of the world, with time-integration and high spatial resolution, contributing in this way for spatially focusing future human health assessments.

Volcanic soils cover only approximately 1% of the Earth's surface, yet they support 10% of the world's population due to their inherent fertility. However, deep magmatic processes may lead to depletion of certain elements in volcanic rocks. The imbalance of essential elements, such as cobalt, in soil parent materials can affect the health of plants and grazing animals and, thus, humans. Within the particular geological context of the Azores, the present study aims to assess cobalt concentration in volcanic soils to predict the risk of cobalt deficiency in animals and humans. Samples from agricultural topsoils and pasture grass were collected in six volcanic regions of São Miguel Island, and their physicochemical properties were measured, including selected transition metal element contents, such as iron, manganese and cobalt. The soil cobalt concentration was below 5 mg/kg in Povoação< Furnas/Congro< Sete Cidades< Fogo and was higher than 10 mg/kg in Picos< Nordeste. Cobalt concentrations were very low in pasture grass. Values below 0.1 mg/kg were observed in Furnas/Congro

We have evaluated buccal epithelial cell micronucleus formation as a biomarker of genotoxic damage following occupational exposure to low doses of ionizing radiation. Buccal epithelial cells were collected from 42 occupationally exposed medical personnel and 39 non-exposed individuals (reference group). Each individual in the exposed group was routinely monitored with a personal radiation badge. The frequency of micronucleated cells (MNC) was significantly higher in the exposed group (5.26 vs. 1.33 MNC/2000 cells) and correlated strongly with accumulated radiation dose (Annual Deep Dose, mSv). Occupational exposure to low doses of ionizing radiation was associated with an 80% relative increase in the frequency of MNC (MRR = 1.8; 95% CI: 1.1-2.8), suggesting that such exposures cause DNA damage. This non-invasive biomonitoring method should be generally applicable to studies of workers who are exposed occupationally to low doses of IR.

Volcanoes produce and release large amounts of toxicants, and the concentration of metals in hair can be used as a biomarker of exposure to trace metals. In order to investigate whether humans exposed to volcanic emissions demonstrate higher concentrations of essential and non-essential trace metals, scalp hair of men (3-89 years) living in two areas of the Azores, one exposed to volcanic emissions and the other not, was sampled, and their content in Cd, Cu, Pb, Rb, Se, and Zn was quantified by using High Resolution Inductively Coupled Plasma Mass Spectrometry. In average, men exposed to volcanic emissions presented higher concentrations of Cd (96.9 ppb), Cu (16.2 ppm), Pb (3417.6 ppb), Rb (216.3 ppb), and Zn (242.8 ppm), but not Se (469.6 ppb). Also, strong and significant correlations were found between Cd-Rb and Pb-Rb in the exposed men. In conclusion, humans living chronically exposed to volcanic emissions show high concentrations of essential and non-essential trace metals in scalp hair, and is suggested that this type of exposure may be as harmful as living close to industrial facilities.

Organisms living in volcanic environments are chronically exposed to metals, either as particles or associated with gases, from volcanic emissions, being therefore potential sentinels of the effects derived from such exposure. Concentrations of Ca, Cd, Cu, Mg, Mn, Pb, Rb, and Zn were measured in soil, grass (Lolium perenne), and larvae of Pseudaletia uninpuncta captured in sites exposed and non-exposed to volcanic activity. The midgut epithelial cell morphometry and apoptosis of P. unipuncta larvae were also analyzed. Larvae from the site with volcanic activity showed higher levels of Cu, Mn, Rb and Zn. Metals such as Pb, Cd and Mg levels of P. unipuncta larvae were similar between sites. Apoptosis was higher in cells from digestive epithelium of larvae exposed to volcanic activity. Soils and grass not exposed to volcanic activity showed higher levels for most of the analyzed elements with the exception of Rb. Such result when compared with metal levels of larvae may reveal that bioavailability of elements differs between sites. The higher levels of Cd, Zn and Mg in soils and grass from the site with no volcanic activity are probably related to the severe artificial fertilization in the studied pastures. Such result, when compared with metal levels of larvae, suggest that the bioavailability of metals differs between sites.

The pressure exerted by shallow water hydrothermal vents on edible gastropods and their cellular responses triggered by these stresses are almost unknown. The aims of this study were to evaluate the bioavailability of metals in the Macaronesian endemic limpet Patella candei gomesii living close to shallow water hydrothermal vents, and the structural differences in their digestive gland as well as the levels of apoptosis in that organ. Limpets were sampled in four sites, two with the presence of hydrothermalism and the other two without it. Whole body concentrations of several metals (Ca, Cd, Cs, Co, Cu, Fe, Hg, Mg, Mn, Pb, Rb, Se, Sr, and Zn) were obtained, morphometry analysis of the digestive gland and TUNEL test for apoptosis were also performed. Results revealed that the presence of shallow water hydrothermal vents is a source of chronic metal stress to limpets, imposing modifications in the morphometry and cell composition of the digestive gland of those limpets that may constitute cell and tissue adaptations to the environment they live in. This study sets up new baseline data for further research on the influence of shallow water hydrothermal vents over communities living in these habitats.
