Disruption of photosynthesis and carbon transport due to damage to the tree crown and stem cambial cells, respectively, can cause tree mortality. It has recently been proposed that fire-induced dysfunction of xylem plays an important role in tree mortality. Here, we simultaneously tested the impact of a lethal fire dose on nonstructural carbohydrates (NSCs) and xylem hydraulics in Pinus ponderosa saplings. Saplings were burned with a known lethal fire dose. Nonstructural carbohydrates were assessed in needles, main stems, roots and whole plants, and xylem hydraulic conductivity was measured in the main stems up to 29 d postfire. Photosynthesis and whole plant NSCs declined postfire. Additionally, all burned saplings showed 100% phloem/cambium necrosis, and roots of burned saplings had reduced NSCs compared to unburned and defoliated saplings. We further show that, contrary to patterns observed with NSCs, water transport was unchanged by fire and there was no evidence of xylem deformation in saplings that experienced a lethal dose of heat from fire. We conclude that phloem and cambium mortality, and not hydraulic failure, were probably the causes of death in these saplings. These findings advance our understanding of the physiological response to fire-induced injuries in conifer trees.

Understanding tree physiological responses to fire is needed to accurately model post-fire carbon processes and inform management decisions. Given trees can die immediately or at extended time periods after fire, we combined two experiments to assess the short- (one-day) and long-term (21-months) fire effects on Pinus ponderosa sapling water transport. Native percentage loss of conductivity (nPLC), vulnerability to cavitation and xylem anatomy were assessed in unburned and burned saplings at lethal and non-lethal fire intensities. Fire did not cause any impact on nPLC and xylem cell wall structure in either experiment. However, surviving saplings evaluated 21-months post-fire were more vulnerable to cavitation. Our anatomical analysis in the long-term experiment showed that new xylem growth adjacent to fire scars had irregular-shaped tracheids and many parenchyma cells. Given conduit cell wall deformation was not observed in the long-term experiment, we suggest that the irregularity of newly grown xylem cells nearby fire wounds may be responsible for decreasing resistance to embolism in burned plants. Our findings suggest that hydraulic failure is not the main short-term physiological driver of mortality for Pinus ponderosa saplings. However, the decrease in embolism resistance in fire-wounded saplings could contribute to sapling mortality in the years following fire.

Wildfire is an essential earth-system process, impacting ecosystem processes and the carbon cycle. Forest fires are becoming more frequent and severe, yet gaps exist in the modeling of fire on vegetation and carbon dynamics. Strategies for reducing carbon dioxide (CO2 ) emissions from wildfires include increasing tree harvest, largely based on the public assumption that fires burn live forests to the ground, despite observations indicating that less than 5% of mature tree biomass is actually consumed. This misconception is also reflected though excessive combustion of live trees in models. Here, we show that regional emissions estimates using widely implemented combustion coefficients are 59%-83% higher than emissions based on field observations. Using unique field datasets from before and after wildfires and an improved ecosystem model, we provide strong evidence that these large overestimates can be reduced by using realistic biomass combustion factors and by accurately quantifying biomass in standing dead trees that decompose over decades to centuries after fire ("snags"). Most model development focuses on area burned; our results reveal that accurately representing combustion is also essential for quantifying fire impacts on ecosystems. Using our improvements, we find that western US forest fires have emitted 851 ± 228 Tg CO2 (~half of alternative estimates) over the last 17 years, which is minor compared to 16,200 Tg CO2 from fossil fuels across the region.

Acoustic impulse events have long been used as diagnostics for discrete phenomena in the natural world, including the detection of meteor impacts and volcanic eruptions. Wildland fires display an array of such acoustic impulse events in the form of crackling noises. Exploratory research into the properties of these impulse events revealed information regarding the specific properties of plant material. Unique acoustic frequency bands in the upper end of the sonic spectrum correlated to changes in vegetation properties. The signature of acoustic impulse events as they relate to plant species and plant water stress, were investigated in controlled laboratory combustion experiments. Correlation in the frequency range of 6.0-15.0 kHz was found for both species and water stress, indicating the possibility that a digital filter may be capable of identifying vegetation properties during wildland fire events.

RESUMO

Wildland fire management has reached a crossroads. Current perspectives are not capable of answering interdisciplinary adaptation and mitigation challenges posed by increases in wildfire risk to human populations and the need to reintegrate fire as a vital landscape process. Fire science has been, and continues to be, performed in isolated "silos," including institutions (e.g., agencies versus universities), organizational structures (e.g., federal agency mandates versus local and state procedures for responding to fire), and research foci (e.g., physical science, natural science, and social science). These silos tend to promote research, management, and policy that focus only on targeted aspects of the "wicked" wildfire problem. In this article, we provide guiding principles to bridge diverse fire science efforts to advance an integrated agenda of wildfire research that can help overcome disciplinary silos and provide insight on how to build fire-resilient communities.

Geofences are virtual boundaries based on geographic coordinates. When combined with global position system (GPS), or more generally global navigation satellite system (GNSS) transmitters, geofences provide a powerful tool for monitoring the location and movements of objects of interest through proximity alarms. However, the accuracy of geofence alarms in GNSS-radio frequency (GNSS-RF) transmitter receiver systems has not been tested. To achieve these goals, a cart with a GNSS-RF locator was run on a straight path in a balanced factorial experiment with three levels of cart speed, three angles of geofence intersection, three receiver distances from the track, and three replicates. Locator speed, receiver distance and geofence intersection angle all affected geofence alarm accuracy in an analysis of variance (p = 0.013, p = 2.58 × 10(-8), and p = 0.0006, respectively), as did all treatment interactions (p < 0.0001). Slower locator speed, acute geofence intersection angle, and closest receiver distance were associated with reduced accuracy of geofence alerts.

There is growing interest in the development of methods capable of non-invasive characterization of stem cells prior to their use in cell-based therapies. Raman spectroscopy has previously been used to detect biochemical changes commensurate with the osteogenic, cardiogenic, and neurogenic differentiation of stem cells. The aim of this study was to characterize the adipogenic differentiation of live adipose derived stem cells (ASCs) under aseptic conditions. ASCs were cultured in adipogenic or basal culture medium for 14 days in customized culture flasks containing quartz windows. Raman spectra were acquired every 3 days. Principal component analysis (PCA) was used to identify spectral changes in the cultures over time. Adipogenic differentiation was confirmed using quantitative reverse transcription polymerase chain reaction for the marker genes PPARγ and ADIPOQ and Oil red O staining performed. PCA demonstrated that lipid associated spectral features varied throughout ASC differentiation with the earliest detection of the lipid associated peak at 1,438 cm(-1) after 3 days of induction. After 7 days of culture there were clear differences between the spectra acquired from ASCs in adipogenic or basal culture medium. No changes were observed in the spectra acquired from undifferentiated ASCs. Significant up-regulation in the expression of both PPARγ and ADIPOQ genes (P < 0.001) was observed after 14 days of differentiation as was prominent Oil red O staining. However, the Raman sampling process resulted in weaker gene expression compared with ASCs that had not undergone Raman analysis. This study demonstrated that Raman spectroscopy can be used to detect biochemical changes associated with adipogenic differentiation in a non-invasive and aseptic manner and that this can be achieved as early as three days into the differentiation process.

There is an unmet need for the non-invasive characterisation of stem cells to facilitate the translation of cell-based therapies. Raman spectroscopy has proven utility in stem cell characterisation but as yet no method has been reported capable of taking repeated Raman measurements of living cells aseptically over time. The aim of this study was to determine if Raman spectroscopy could be used to monitor changes in a well characterised cell population (human dental pulp stromal cells (DPSCs)) by taking repeated Raman measurements from the same cell populations in osteoinductive culture over time and under aseptic conditions. DPSCs were isolated from extracted premolar teeth from 3 consenting donors. Following in vitro expansion, DPSCs were maintained for 28 days in osteo-inductive medium. Raman spectra were acquired from the cells at days 0, 3, 7, 10, 14 and 28. Principal component analysis (PCA) was carried out to assess if there was any temporal spectral variation. At day 28, osteoinduction was confirmed using alizarin red staining and qRT-PCR for alkaline phosphatase and osteocalcin. Alizarin red staining was positive in all samples at day 28 and significant increases in alkaline phosphatase (p < 0.001) and osteocalcin (p < 0.05) gene expression were also observed compared with day 0. PCA of the Raman data demonstrated trends in PC1 from days 0-10, influenced by protein associated features and PC2 from days 10-28, influenced by DNA/RNA associated features. We conclude that spectroscopy can be used to monitor changes in Raman signature with time associated with the osteoinduction of DPSCs using repeated measurements via an aseptic methodology.

Ectomycorrhizal fungi (EcMF) typically colonize nursery seedlings, but nutritional and growth effects of these communities are only partly understood. To examine these effects, Picea glauca seedlings collected from a tree nursery naturally colonized by three dominant EcMF were divided between fertilized and unfertilized treatments. After one growing season seedlings were harvested, ectomycorrhizas identified using DNA sequencing, and seedlings analyzed for leaf nutrient concentration and content, and biomass parameters. EcMF community structure-nutrient interactions were tested using nonmetric multidimensional scaling (NMDS) combined with vector analysis of foliar nutrients and biomass. We identified three dominant species: Amphinema sp., Atheliaceae sp., and Thelephora terrestris. NMDS + envfit revealed significant community effects on seedling nutrition that differed with fertilization treatment. PERMANOVA and regression analyses uncovered significant species effects on host nutrient concentration, content, and stoichiometry. Amphinema sp. had a significant positive effect on phosphorus (P), calcium and zinc concentration, and P content; in contrast, T. terrestris had a negative effect on P concentration. In the unfertilized treatment, percent abundance of the Amphinema sp. negatively affected foliar nitrogen (N) concentration but not content, and reduced foliar N/P. In fertilized seedlings, Amphinema sp. was positively related to foliar concentrations of N, magnesium, and boron, and both concentration and content of manganese, and Atheliaceae sp. had a negative relationship with P content. Findings shed light on the community and species effects on seedling condition, revealing clear functional differences among dominants. The approach used should be scalable to explore function in more complex communities composed of unculturable EcMF.

Fire is a major cause of tree injury and mortality worldwide, yet our current understanding of fire effects is largely based on ocular estimates of stem charring and foliage discoloration, which are error prone and provide little information on underlying tree function. Accurate quantification of physiological performance is a research and forest management need, given that declining performance could help identify mechanisms of-and serve as an early warning sign for-mortality. Many previous efforts have been hampered by the inability to quantify the heat flux that a tree experiences during a fire, given its highly variable nature in space and time. In this study, we used a dose-response approach to elucidate fire impacts by subjecting Pinus monticola var. minima Lemmon and Pseudotsuga menziesii (Mirb.) Franco var. glauca (Beissn.) Franco saplings to surface fires of varying intensity doses and measuring short-term post-fire physiological performance in photosynthetic rate and chlorophyll fluorescence. We also evaluated the ability of spectral reflectance indices to quantify change in physiological performance at the individual tree crown and stand scales. Although physiological performance in both P. monticola and P. menziesii declined with increasing fire intensity, P. monticola maintained a greater photosynthetic rate and higher chlorophyll fluorescence at higher doses, for longer after the fire. Pinus monticola also had complete survival at lower fire intensity doses, whereas P. menziesii had some mortality at all doses, implying higher fire resistance for P. monticola at this life stage. Generally, individual-scale spectral indices were more accurate at quantifying physiological performance than those acquired at the stand-scale. The Photochemical Reflectance Index outperformed other indices at quantifying photosynthesis and chlorophyll fluorescence, highlighting its potential use to quantify crown scale physiological performance. Spectral indices that incorporated near-infrared and shortwave infrared reflectance, such as the Normalized Burn Ratio, were accurate at characterizing stand-scale mortality. The results from this study were included in a conifer cross-comparison using physiology and mortality data from other dose-response studies. The comparison highlights the close evolutionary relationship between fire and species within the Pinus genus, assessed to date, given the high survivorship of Pinus species at lower fire intensities versus other conifers.

In radiology departments with multiple geographically separated reporting areas, locating radiologists can be challenging. We have developed an in-house solution to minimise the time spent looking for radiologists utilising near real-time data stored with our radiology information system (RIS). An auto updating Extensible Markup Language (XML) data feed from our RIS provider provides information about users logged into the RIS. It includes user names, their contact details and specialty interests, their location within the department, and a time stamp of last recorded dictation or report verification. The information is then displayed on our internal intranet and on a self-refreshing screen in our main department corridor. In order to estimate time savings made through the tools creation, usage statistics were calculated and combined with assessments of time taken to find a named radiologist prior to the tools implementation. Over the month of April 2009, there were 2,798 hits on the locator page. Radiologists were responsible for 1,248 hits and radiology administration staff for 1,550 hits. The average time for using the tool was calculated at 5 s. Reviewing a roster and calling/paging a radiologist took on average 30 s, and a full walk around of the department took 195 s. Through utilisation of near real-time data available within our RIS system and display of these data in an accessible form, we have developed a tool that has realised savings of up to 16 h of radiologist reporting time per week.

Quantum computers are becoming increasingly accessible and may soon outperform classical computers for useful tasks. However, qubit readout errors remain a substantial hurdle to running quantum algorithms on current devices. We present a scheme to more efficiently mitigate these errors on quantum hardware and numerically show that our method consistently gives advantage over previous mitigation schemes. Our scheme removes biases in the readout errors, allowing a general error model to be built with far fewer calibration measurements. Specifically, for reading out n-qubits, we show a factor of 2n reduction in the number of calibration measurements without sacrificing the ability to compensate for correlated errors. Our approach can be combined with, and simplify, other mitigation methods, allowing tractable mitigation even for large numbers of qubits.

BACKGROUND: Psychological difficulties such as anxiety, depression, and irritability are common in Huntington's disease, even for premanifest individuals. However, very little evidence exists of psychological approaches to manage this distress. We have conducted a feasibility study with an embedded qualitative component to investigate the possibility of using mindfulness-based cognitive therapy (MBCT) and present here the findings from the qualitative data. OBJECTIVE: To investigate the experience of premanifest individuals learning and practising mindfulness through completing a course of MBCT. METHODS: Twelve premanifest individuals completed a course of MBCT and attended three follow up reunion meetings over the following year. Eleven participants agreed to be interviewed post-course and ten participants one year post-course about their experience of the course and any impact on their lives. Seven participants nominated a friend or relative (supporter) to be involved in the research, of whom six agreed to be interviewed post-course and two at one year about the impact of the course on the participants. Data were analysed using reflexive thematic analysis. RESULTS: Four themes were constructed from the data: 1) A meeting of minds: the group facilitating learning and support; 2) Mindfulness is hard, but enables more effective emotional management; 3) Mindfulness can change the relationship with self and others; and 4) Benefiting from mindfulness: the importance of persistence. CONCLUSION: The participants who completed the course found it beneficial. Some participants reported reductions in psychological distress, a greater sense of calm and better emotion regulation, with some of these positive changes also noticed by supporters. MBCT is worthy of further investigation for this population.

BACKGROUND: Huntington's disease (HD) is an inherited neurodegenerative condition which affects movement, coordination and cognitive functioning. Psychological difficulties are commonly experienced; however, psychological interventions have been little researched with this population. We investigated the feasibility of conducting a randomised controlled trial (RCT) of mindfulness-based cognitive therapy (MBCT) with people with the HD genetic mutation, either pre-manifest (before onset of movement symptoms) or at an early disease stage. Specifically, we evaluated the willingness of participants to be recruited into and complete the intervention; the acceptability of the study measures in relation to completion; the feasibility of offering the standard MBCT course to people with HD; the acceptability of the intervention and the estimated effect sizes. METHODS: Participants were recruited from two UK HD centres and took part in an 8-week course of MBCT, with three reunions throughout the following year. Stress, depression, anxiety, and mindfulness were measured pre-, mid-, and post-course, at 3 months and at 1 year. Sleep, quality of life, positive affect and coping were measured pre- and post-course, at 3 months and at 1 year. Descriptive data and approximate effect sizes were calculated. Interviews were conducted post-course and at 1 year and data pertaining to the acceptability of the course were extracted. RESULTS: Twelve participants took part in two groups; all were pre-manifest. Levels of depression and anxiety were low pre-course leaving little room for improvement. Changes in stress and in some aspects of mindfulness were medium to large. The qualitative data suggested participants rated the course highly and found it helpful and no changes to the standard course were needed. Recruitment levels were below those anticipated. Most measures were found to be acceptable. CONCLUSIONS: Although the course was acceptable to those who took part, given the difficulties in recruiting and the rarity of HD, conducting an RCT of MBCT teaching groups in person does not seem feasible. However, alternative modes of course delivery (e.g. online) would allow the recruitment of people from a greater geographical area and may make an RCT feasible; this revised focus would be suitable for future feasibility studies. OBJECTIVES: The study aimed (1) to investigate changes in older adults' emotional wellbeing (specifically depression, anxiety and stress levels) and mindful ability following a mindfulness-based cognitive therapy (MBCT) course; (2) to explore correlations between mindfulness (measured as an overall ability and as individual components; observe, describe, act with awareness and accept without judgement) and changes in depression, anxiety and stress levels. METHOD: Twenty-two participants took an eight-week MBCT course. Levels of depression, anxiety and stress were recorded pre- and post-intervention, as was mindfulness ability (measured both as an overall ability and as individual components). RESULTS: Significant improvements in emotional wellbeing and mindfulness were reported post-MBCT, with large to moderate effect sizes. Increased mindfulness was moderately and significantly associated with improved emotional wellbeing. Increases on all four components of mindfulness were positively associated with greater emotional wellbeing, however only act with awareness and accept without judgement were significantly correlated (with reduced depression). Older adults in our sample reported higher scores on observe and act with awareness than other populations. CONCLUSION: This study adds to a growing evidence-base indicating the efficacy of MBCT for depression, anxiety and stress, and extends these finding to older adults. This study found older adults to have elevated levels of certain facets of mindfulness and recommendations are made for researching the possibility that mindfulness may be an extension of the developmental process.

AIM: Pre-hospital triage strategies aim to identify the type and extent of patient injuries and ensure that they are transferred to the most appropriate trauma centres. Despite the importance of appropriate pre-hospital transport, there is little evidence base to assist medical staff on optimal destination policy for emergent pre-hospital transport. This paper explores the spatial relationship of patient transfers prior to the implementation of the Midland Pre-Hospital Trauma Destination Matrix in New Zealand, and is a retrospective view of practice against a destination policy that was applied after the study period. METHODS: We use data obtained from the Midland Trauma Registry merged with Global Positioning System (GPS) data from St John and Land Information New Zealand Data Service on major trauma occurring in 2014 and 2015. Using ArcGIS, data were analysed for spatial relationships between factors associated with major trauma events and pre-hospital transportation. RESULTS: In the retrospective analysis of 162 major trauma patients, 107 (66%) were transported to a hospital that matched the destination specified in the Matrix, and 55 (34%) were transported to a non-Matrix designated hospital. CONCLUSION: Approximately one-third of patients were not directly transported to the preferred definitive care hospital subsequently defined in the Midland Pre-Hospital Trauma Destination Matrix. Ongoing monitoring of the pre-hospital transportation system and the implementation of a formal pre-hospital transport policy may improve the efficiency of the Midland Trauma System. Future studies should examine the possible reasons for variations in triage decisions across the Midland Region.

The anaesthetic lidocaine is metabolised quickly to produce a series of metabolites, including several hydroxylated metabolites, which are further metabolised by addition of a glucuronic acid moiety. Analysis of these glucuronide metabolites in urine is performed indirectly by cleaving the glucuronic acid group using ß-glucuronidase. However, direct analysis of intact glucuronide conjugates is a more straightforward approach as it negates the need for long hydrolysis incubations, and minimises the oxidation of sensitive hydrolysis products, while also distinguishing between the two forms of hydroxylated metabolites. A method was developed to identify three intact glucuronides of lidocaine in sheep urine using LC-MS/MS, which was further confirmed by the synthesis of glucuronide derivatives of 3OH-MEGX and 4OH-LIDO. Direct analysis of urine allowed the detection of the glucuronide metabolites of hydroxylidocaine (OH-LIDO), hydroxyl-monoethylglycinexylidide (OH-MEGX), and hydroxy-2,6-xylidine (OH-XYL). Analysis of urine before and after ß-glucuronidase digestion showed that the efficiency of hydrolysis of these glucuronide metabolites may be underestimated in some studies. Analysis of urine in the current study from three different sheep with similar glucuronide metabolite concentrations resulted in different hydrolysis efficiencies, which may have been a result of different levels of substrate binding by matrix components, preventing enzyme cleavage. The use of direct analysis of intact glucuronides has the benefit of being less influenced by these matrix effects, while also allowing analysis of unstable metabolites like 4OH-XYL, which rapidly oxidises after hydrolysis. Additionally, direct analysis is less expensive and less time consuming, while providing more information about the status of hydroxylated metabolites in urine.

BACKGROUND: Automated clinical decision support systems (CDSS) are associated with improvements in health care delivery to those with long-term conditions, including diabetes. A CDSS was introduced to two Scottish regions (combined diabetes population ~30 000) via a national diabetes electronic health record. This study aims to describe users' reactions to the CDSS and to quantify impact on clinical processes and outcomes over two improvement cycles: December 2013 to February 2014 and August 2014 to November 2014. METHODS: Feedback was sought via patient questionnaires, health care professional (HCP) focus groups, and questionnaires. Multivariable regression was used to analyze HCP SCI-Diabetes usage (with respect to CDSS message presence/absence) and case-control comparison of clinical processes/outcomes. Cases were patients whose HCP received a CDSS messages during the study period. Closely matched controls were selected from regions outside the study, following similar clinical practice (without CDSS). Clinical process measures were screening rates for diabetes-related complications. Clinical outcomes included HbA1c at 1 year. RESULTS: The CDSS had no adverse impact on consultations. HCPs were generally positive toward CDSS and used it within normal clinical workflow. CDSS messages were generated for 5692 cases, matched to 10 667 controls. Following clinic, the probability of patients being appropriately screened for complications more than doubled for most measures. Mean HbA1c improved in cases and controls but more so in cases (-2.3 mmol/mol [-0.2%] versus -1.1 [-0.1%], P = .003). DISCUSSION AND CONCLUSIONS: The CDSS was well received; associated with improved efficiencies in working practices; and large improvements in guideline adherence. These evidence-based, early interventions can significantly reduce costly and devastating complications.

Extreme wildfires have substantial economic, social and environmental impacts, but there is uncertainty whether such events are inevitable features of the Earth's fire ecology or a legacy of poor management and planning. We identify 478 extreme wildfire events defined as the daily clusters of fire radiative power from MODIS, within a global 10 × 10 km lattice, between 2002 and 2013, which exceeded the 99.997th percentile of over 23 million cases of the ΣFRP 100 km-2 in the MODIS record. These events are globally distributed across all flammable biomes, and are strongly associated with extreme fire weather conditions. Extreme wildfire events reported as being economically or socially disastrous (n = 144) were concentrated in suburban areas in flammable-forested biomes of the western United States and southeastern Australia, noting potential biases in reporting and the absence of globally comprehensive data of fire disasters. Climate change projections suggest an increase in days conducive to extreme wildfire events by 20 to 50% in these disaster-prone landscapes, with sharper increases in the subtropical Southern Hemisphere and European Mediterranean Basin.

We studied selected tissues from ewes and their lambs that were grazing pastures fertilized with either sewage sludge (treated) or inorganic fertilizer (control) and determined concentrations of alkylphenols and phthalates in these tissues. Mean tissue concentrations of alkylphenols were relatively low (< 10-400 microg/kg) in all animals and tissues. Phthalates were detected in tissues of both control and treated animals at relatively high concentrations (> 20,000 microg/kg in many tissue samples). The use of sludge as a fertilizer was not associated with consistently increased concentrations of either alkylphenols or phthalates in the tissues of animals grazing treated pastures relative to levels in control animal tissues. Concentrations of the two classes of chemicals differed but were of a similar order of magnitude in liver and muscle as well as in fat. Concentrations of each class of compound were broadly similar in tissues derived from ewes and lambs. Although there were significant differences (p < 0.01 or p < 0.001) between years (cohorts) in mean tissue concentrations of both nonylphenol (NP) and phthalate in each of the tissues from both ewes and lambs, the differences were not attributable to either the age (6 months or 5 years) of the animal or the duration of exposure to treatments. Octylphenol concentrations were generally undetectable. There was no consistent cumulative outcome of prolonged exposure on the tissue concentrations of either class of pollutant in any ewe tissue. Mean tissue concentrations of phthalate were higher (p < 0.001) in the liver and kidney fat of male compared with female lambs. We suggest that the addition of sewage sludge to pasture is unlikely to cause large increases in tissue concentrations of NP and phthalates in sheep and other animals with broadly similar diets and digestive systems (i.e., domestic ruminants) grazing such pasture.
