RESUMO

The genus Burkholderia represents a challenge to the fields of taxonomy and phylogeny and, especially, to the understanding of the contrasting roles as either opportunistic pathogens or bacteria with biotechnological potential. Few genomes of nonpathogenic strains, especially of diazotrophic symbiotic bacteria, have been sequenced to improve understanding of the genus. Here, we contribute with the complete genome sequence of Burkholderia phenoliruptrix strain BR3459a (CLA1), an effective diazotrophic symbiont of the leguminous tree Mimosa flocculosa Burkart, which is endemic to South America.

Lectins constitute a class of glycoproteins, which are capable of selectively and reversibly binding to carbohydrates, distinguishing small structural differences in complex oligosaccharides. Studies have shown that the binding of lectins to cell-surface carbohydrates can lead to various effects such as cellular proliferation, histamine release and cytokine production. Canavalia brasiliensis lectin (ConBr) is a (D-mannose) D-glucose lectin. In this study, murine splenocytes were cultured to determine the effect of ConBr on cell proliferation, nitric oxide (NO) release and cytokine secretion. In addition, cellular viability assays were performed to evaluate any mitogenic activity induced by this lectin. ConBr significantly increased cell proliferation with minimal cell damage. This lectin was able to induce an increased production of cytokines such as IL-2, IL-6 and IFN-γ and a decreased production of IL- 10. The release of NO was also observed. The results of this study indicate that ConBr could potentially be used as an immunomodulator.

Surface colonization is an essential step in biofilm development. The ability of oral pathogens to adhere to tooth surfaces is directly linked with the presence of specific molecules at the bacterial surface that can interact with enamel acquired pellicle ligands. In light of this, the aim of this study was to verify inhibitory and antibiofilm action of lectins from the Diocleinaesubtribe against Streptococcus mutans and Streptococcus oralis. The inhibitory action against planctonic cells was assessed using lectins from Canavaliaensi formis (ConA), Canavalia brasiliensis (ConBr), Canavalia maritima (ConM), Canavalia gladiata (CGL) and Canavalia boliviana (ConBol). ConBol, ConBr and ConM showed inhibitory activity on S. mutans growth. All lectins, except ConA, stimulated significantly the growth of S. oralis. To evaluate the effect on biofilm formation, clarified saliva was added to 96-well, flat-bottomed polystyrene plates, followed by the addition of solutions containing 100 or 200 µg/mL of the selected lectins. ConBol, ConM and ConA inhibited the S. mutans biofilms. No effects were found on S. oralis biofilms. Structure/function analysis were carried out using bioinformatics tools. The aperture and deepness of the CRD (Carbohydrate Recognition Domain) permit us to distinguish the two groups of Canavalia lectins in accordance to their actions against S. mutans and S. oralis. The results found provide a basis for encouraging the use of plant lectins as biotechnological tools in ecological control and prevention of caries disease.

Lectins are sugar-binding proteins widely distributed in nature with many biological functions. Although many lectins have a remarkable biotechnological potential, some of them can be cytotoxic. Thus, the aim of this study was to assess the toxicity of five lectins, purified from seeds of different species of Canavalia genus. In order to determine the toxicity, assays with Artemia nauplii were performed. In addition, a fluorescence assay was carried out to evaluate the binding of lectins to Artemia nauplii. In order to verify the relationship between the structure of lectins and their cytotoxic effect, structural analysis was carried out to evaluate the volume of the carbohydrate recognition domain (CRD) of each lectin. The results showed that all lectins exhibited different toxicities and bound to a similar area in the digestive tract of Artemia nauplii. Concerning the structural analysis, differences in spatial arrangement and volume of CRD may explain the variation of the toxicity exhibited by each lectin. To this date, this is the first study that establishes a link between toxicity and structure of CRD from Diocleinae lectins.

Sementes quiescentes de Bauhinia forficata foram submetidas à caracterizaçao bioquímica por medio de análise de ácidos graxos, (fraccionamiento de proteínas e atividade hemaglutinante específica. A análise elementar da semente mostrou grande quantidade de proteína total e de lipídeos com 21,24 por cento e 19.45 por cento, respectivamente. Na fraçao lipídica, o ácido linolêico foi o mais abundante com 46,47 por cento. Com exceçao das prolaminas, as diferentes françoes protéicas (albuminas, globulinas, glutelinas ácidas e básicas) apresentaram actividade hemaglutinante contra hemácias de coelho tratadas e nao tratadas com enzimas proteolíticas. A maior actividade hemaglutinante específica foi evidenciada na fraçao glutelinas ácidas (1.072,25 U.H./mgP) contra sangue de coelho tratado com tripsina. Os aminoácidos presentes em maior teor foram glutamina (16,20 por cento) e valina (11,07 por cento). Assim, por apresentarem alto valor energético as sementes de Bauhinia forficata sao uma possível fonte apcional na alimentaçao
