In 2019, an oil spill of unprecedented dimensions and unknown source reached the Brazilian coast. Such incident, which occurred somewhere in the ocean and was detected only when the first oil patches reached the beaches in the northeastern region, is recognized as one of the most serious oil spills in the country. From the chromatographic analysis of the oily residues using n-alkanes, polycyclic aromatic hydrocarbons (PAH), and oil biomarkers, it was possible to characterize their coverage. The results confirmed that an oil of the same specification reached a length of coastline higher than 3,000 kilometers. However, were also found oily residues that did not match with the product involved in that incident, evidencing the occurrence of different discharges. This study documents the extension of the affected coastline during the incident and assesses the possible origin of such different oils from their geochemical characteristics by selecting 21 samples from more than 300 samples previously collected and analyzed during the response actions. The unexpected occurrence of these residues suggest chronic events. The results highlighted that unrelated oily residues were found even in a region far from the coast, indicating that discharges of oil in the open sea occur more regularly than initially expected.

Copper (Cu) contents in vineyard soils due to the application of cupric fungicides cause changes in the native covering flora. Under these conditions, the surviving individuals accumulate the metal in and decrease its availability in the soil, reducing the potential toxicity to grapevine. We have identified spontaneous plant species and their phytoremediation potential from vineyards of Isabella (Vitis labrusca) on two distinct soil types (Inceptisol and Entisol) contaminated with Cu. The results demonstrated that wild species displayed higher Cu contents in the roots than in the shoot, but had low bioaccumulation potential. During summer, the plants were unable to extract and stabilize the metal, although during the winter, Lolium multiflorum, Cyperus compressus and Chrysanthemum leucanthemum demonstrated phytostabilization potential. Among the investigated species, dry matter production and Cu accumulation by Lolium multiflorum indicated that the species is effective to decrease Cu availability in the soil.

BACKGROUND: The combination of near-infrared (NIR) spectroscopy and chemometrics can be used to group or discriminate soils based on spectral response. In this study, we conducted discrimination and classification analyses on soils managed with different sources of fertilization and plant species grown in organic and conventional farming systems. RESULTS: Principal component analysis explained 96% (PC1) and 3% (PC2) of the data variability and separated the soil samples of organic and conventional management systems. The wavenumbers that contributed most to the separation of the management systems were in the range of 3600 and 7300 cm-1 , especially the absorption peaks of 3700 and 4600 cm-1 (characteristic of CH and NH combinations), and 5200 and 7000 cm-1 (typical of OH combinations). Machine learning analysis using k-nearest neighbor and random forest algorithms was efficient in classifying soil samples according to management system with an accuracy of 97.8% and can therefore be used for future classification studies. CONCLUSION: Based on the results, we strongly recommend the use of NIR spectroscopy associated with chemometrics for discriminating soils grown with Malus domestica, Musa spp., Oryza sativa and Solanum tuberosum L. under organic and conventional management systems through spectral response. © 2021 Society of Chemical Industry.

Fractionation of rare earth elements (REE) and other trace metal concentrations (Th, U, Cd, Cr, Cu, Ni, Pb, and Zn) between mangrove sediments and claw muscles and shells of male crabs (Ucides cordatus) from a subtropical estuary highly impacted by fertilizer industry activities was investigated. This is the first record of REE distribution in these organisms, and the results showed higher accumulations of these metals, U and Th in shells, probably related to the replacement of Ca during molting. Contents of Cd, Cr and Ni were similar in both tissues, but Cu, Zn and Pb were mostly accumulated in the claw muscle with concentrations above those considered safe for human consumption according to the Brazilian legislation. REE fractionation was different in the analyzed tissues being softer in the shells. The results provided evidences that the water absorbed during molting controls the chemistry of REE in shells. In contrast, the chemistry of REE in the claw muscle, in which was observed preferential absorption of light REE, is controlled by diet. REE fractionation obtained for the claw muscles was closely correlated to the observed in the contaminated substrate and in materials related to the production of phosphate fertilizers (contamination source), which supports their transference to this Ucides cordatus tissue without fractionation by the ingestion of sediments. Our results showed the potential use of crab tissues for monitoring REE and trace element sources in mangrove areas, with claw muscle exhibiting the contaminant source fingerprint.

AIM: This study evaluated the effects of scaling and root planing (SRP) on gingival crevicular fluid (GCF) and serum levels of adipokines in patients with chronic periodontitis (CP) with or without obesity. METHODS: Twenty patients with obesity and 20 patients without obesity, all with CP, received SRP. Serum and GCF levels of resistin, adiponectin, leptin, tumour necrosis factor [TNF]-α and interleukin [IL]-6 were evaluated by enzyme-linked immunosorbent assay at baseline, 3, 6 and 12 months post-therapy. RESULTS: SRP reduced the amounts of TNF-α in deep sites and increased the concentration of adiponectin in shallow sites of non-obese patients (p < 0.05). SRP increased the concentrations of TNF-α and leptin in patients with obesity (p < 0.05). GCF levels of TNF-α were higher in patients with obesity than in patients without obesity at all time-points (p < 0.05). There were no changes in serum levels of any adipokines for any group after therapy (p > 0.05). Patients with obesity exhibited higher serum levels of leptin at all time-points and IL-6 at 3 months post-therapy (p < 0.05). CONCLUSIONS: Obesity may modulate systemic and periodontal levels of adipokines in favour of pro-inflammation, independently of periodontal therapy. SRP did not affect the circulating levels of adipokines in patients with or without obesity.

Onychomycosis is a common fungal nail infection for which new antifungals are needed to overcome antimicrobial resistance and the limitations of conventional treatments. This study reports the development of antifungal nail lacquers containing oregano essential oil (OEO), rosemary essential oil (REO), and biogenic silver nanoparticles (bioAgNPs). The formulations (F) were tested against dermatophytes using agar diffusion, ex vivo nail infection, and scanning electron microscopy techniques. They were evaluated for their pharmacotechnical characteristics and by FTIR-PAS to assess permeation across the nail. F-OEO and F-OEO/bioAgNPs were promising candidates for the final nail lacquer formulation, as they permeated through the nail and showed antifungal efficacy against dermatophytes-contaminated nails after 5 days of treatment. Treated nails exhibited decreased hyphae and spores compared to the untreated control; the hyphae were atypically flattened, indicating loss of cytoplasmic content due to damage to the cytoplasmic membrane. The formulations were stable after centrifugation and thermal stress, maintaining organoleptic and physicochemical characteristics. Both F-OEO and F-OEO/bioAgNPs had pH compatible with the nail and drying times (59-90 s) within the reference for nail lacquer. For the first time, OEO and bioAgNPs were incorporated into nail lacquer, resulting in a natural and nanotechnological product for onychomycosis that could combat microbial resistance.

Selection of adjuvant to be combined with the antigen is an extremely important point for formulating effective vaccines. The aim of this study was to evaluate reactogenicity, levels of IgM, IgG and subclasses (IgG1, IgG2b and IgG3), and protection elicited by vaccine formulations with association of chitosan coated alginate or Montanide ISA 61 with γ-irradiated Brucella ovis. The alginate/chitosan biopolymers as well as the Montanide ISA 61 emulsion elicited intense and long-lasting local response, especially when associated with the antigen. However, Montanide ISA 61 induced less intense reactogenicity when compared to alginate/chitosan. Furthermore, γ-irradiated B. ovis with Montanide ISA 61 induced higher levels of IgG2b an important marker of cellular immune response. In conclusion, Montanide ISA 61 resulted in milder reactogenicity when compared to the alginate/chitosan, while it induced a high IgG2b/IgG1 ratio compatible with a Th1 profile response.

We perform a spectroscopic analysis of asphaltene in solution and in crude oil with the goal of designing an optical probe of asphaltene precipitation inside high-pressure cells. Quantitative analysis of steady-state spectroscopic data is employed to identify fluorescence and Raman contributions to the observed signals. Time-resolved fluorescence spectroscopy indicates that fluorescence lifetime can be used as a spectroscopic probe of asphaltene in crude oil. Quantitative confocal laser-scanning microscopy studies of asphaltene in n-heptane are used to calculate particle-size distributions as a function of time, both at the sample surface and asphaltene interior. The resulting precipitation kinetics is well described by stochastic numerical simulations of diffusion-limited aggregation. Based on these results, we present the design and construction of an apparatus to optically probe the in situ precipitation of asphaltene suitable for studies inside high pressure cells. Design considerations include the use of a spatial light modulator for aberration correction in microscopy measurements, together with the design of epi-fluorescence spectrometer, both fiber-based and for remote sensing fluorescence spectroscopy.

The burden of disease attributable to obesity is rapidly increasing and becoming a public health challenge globally. Using a nationally representative sample in Australia, this study aims to examine the association of obesity with healthcare service use and work productivity across outcome distributions. We used Household, Income and Labour Dynamics Australia (HILDA) Wave 17 (2017-2018), including 11,211 participants aged between 20 and 65 years. Two-part models using multivariable logistic regressions and quantile regressions were employed to understand variations in the association between obesity levels and the outcomes. The prevalence of overweight and obesity was 35.0% and 27.6%, respectively. After adjusting for socio-demographic factors, low socioeconomic status was associated with a higher probability of overweight and obesity (Obese III: OR = 3.79; 95% CI 2.53-5.68) while high education group was associated with a lower likelihood of being high level of obesity (Obese III OR = 0.42, 95% CI 0.29-0.59). Higher levels of obesity were associated with higher probability of health service use (GP visit Obese, III: OR = 1.42 95% CI 1.04-1.93,) and work productivity loss (number of paid sick leave days, Obese III: OR = 2.40 95% CI 1.94-2.96), compared with normal weight. The impacts of obesity on health service use and work productivity were larger for those with higher percentiles compared to lower percentiles. Overweight and obesity are associated with greater healthcare utilisation, and loss in work productivity in Australia. Australia's healthcare system should prioritise interventions to prevent overweight and obesity to reduce the cost on individuals and improve labour market outcomes.

The aim of this study was to evaluate the influence of various opacities and thicknesses of lithium disilicate on the degree of conversion (DC) of two resin cements and on their bond strength (BS) to the ceramic. Two hundred and forty ceramic samples of lithium disilicate with high translucency (HT), low translucency (LT), and medium opacity (MO) were obtained from IPS e. max CAD in five different thicknesses. Light-cured and dual-cured resin cements were used for DC (n = 9) and BS (n = 8). Cement samples were light-cured under ceramic samples and analyzed using a Raman confocal spectrometer to determine the DC. For BS, resin cement cylinders were fabricated using ceramic samples which were thsen subjected to a microshear bond strength test. The mean values were provided for statistical analysis. The increase in thickness resulted in a decrease in DC for both cements under all experimental conditions, but only affected the BS of the light-cured cement for the MO ceramic. Comparing the opacities, the most translucent ceramics with particular thicknesses exhibited higher DC values than the less translucent ceramics. The LT and MO ceramics with certain thicknesses exhibited the highest BS values than the HT. The dual-cured cement generally showed highest values in both analyses than the light-cured cement. A thicker and more opaque ceramic material can reduce the DC of the cement. The BS decreases with the increasing thickness of the more-opaque ceramics.

RESUMO

This study included 47 free-ranging bats from the State of Minas Gerais, Brazil. Six bats (12.8%) had genital inflammatory lesions, and two of them (one Artibeus lituratus and one Glossophaga soricina, a frugivorous and a nectarivorous, respectively) were diagnosed with Brucella sp. infection through PCR, and antigens in intralesional macrophages were detected using immunohistochemistry.

BACKGROUND: Onychomycosis (OM) is a common nail plate disorder caused by dermatophyte molds, yeasts, and non-dermatophyte molds, which use keratin in the nail plate as an energy source. OM is characterized by dyschromia, increased nail thickness, subungual hyperkeratosis, and onychodystrophy, and is typically treated with conventional antifungals despite frequent reports of toxicity, fungal resistance, and OM recurrence. Photodynamic therapy (PDT) with hypericin (Hyp) as a photosensitizer (PS) stands out as a promising therapeutic modality. When excited by a specific wavelength of light and in the presence of oxygen, to lead to photochemical and photobiological reactions on the selected targets. METHODS: OM diagnosis was made in three suspected cases, and the causative agents were identified by classical and molecular methods, and confirmed by attenuated total reflectance-Fourier transform infrared spectroscopy (ATR-FTIR). Susceptibility of planktonic cells of the clinical isolates to conventional antifungals and PDT-Hyp was evaluated, and photoacoustic spectroscopy (PAS) of Hyp permeation in nail fragments ex vivo was analyzed. Furthermore, the patients opted to undergo PDT-Hyp treatment and were subsequently followed up. The protocol was approved by the human ethics committee (CAAE, number 14107419.4.0000.0104). RESULTS: The etiological agents of OM in patients ID 01 and ID 02 belonged to the Fusarium solani species complex, being identified as Fusarium keratoplasticum (CMRP 5514) and Fusarium solani (CMRP 5515), respectively. For patient ID 03, the OM agent was identified as Trichophyton rubrum (CMRP 5516). PDT-Hyp demonstrated a fungicidal effect in vitro, with reductions of p3 log10 (p < 0.0051 and p < 0.0001), and the PAS analyses indicated that Hyp could completely permeate through both healthy and OM-affected nails. After four sessions of PDT-Hyp, mycological cure was observed in all three cases, and after seven months, clinical cure was confirmed. CONCLUSION: PDT-Hyp showed satisfactory results in terms of efficacy and safety, and thus can be considered a promising therapy for the clinical treatment of OM.

Species of the Candida genus represent the third most common cause of onychomycosis, the most frequent and difficult to treat nail infection. Onychomycosis has been attributed to fungi organized in biofilm and some natural products have proved promising for its treatment. This study aimed to evaluate the antibiofilm activity of propolis extract (PE) and its by-product (WPE) on 7-day preformed biofilms produced by Candida albicans in polystyrene microplates, as well as in an ex vivo model on human nail fragments. The cytotoxicity and permeation capacity were also assessed. Firstly, multiple parameters were evaluated over 7 days to elucidate the dynamics of biofilm formation by C. albicans. The cell viability and total biomass did not vary much from the beginning; however, days 3 and 4 were crucial in terms of metabolic activity, which was significantly increased, and the levels of extracellular matrix components, wherein proteins and nucleic acids experienced an increase, but polysaccharide levels dropped. Architecturally, one-day biofilm showed a monolayer of organized cells (blastoconidia, hyphae, and pseudohyphae), while in the seven-day biofilm there was a three-dimensional well-structured and complex biofilm. This yeast was also able to form a biofilm on both surfaces of the nail, without an additional nutritional source. Both extracts showed excellent antibiofilm activity against the 7-day preformed biofilm and were not toxic to Vero cells at concentrations compatible with the antifungal and antibiofilm activities. Both extracts permeated the experimentally infected nail, with WPE being more efficient. The results of this study, taken together, reinforce the potential of these natural products, containing propolis, as a safe option for the topical treatment of onychomycosis.

INTRODUCTION: Previous studies exploring associations of physical inactivity, obesity, and out-of-pocket expenditure (OOPE) mainly used traditional linear regression, and little is known about the effect of both physical inactivity and obesity on OOPE across the percentile distribution. This study aims to assess the effects of physical inactivity and obesity on OOPE in China using a quantile regression approach. METHODS: Study participants included 10,687 respondents aged 45 years and older from the recent wave of the China Health and Retirement Longitudinal Study in 2015. Linear regression and quantile regression models were used to examine the association of physical activity, body weight with annual OOPE. RESULTS: Overall, the proportion of overweight and obesity was 33.2% and 5.8%, respectively. The proportion of individuals performing high-level, moderate-level, and low-level physical activity was 55.2%, 12.7%, and 32.1%, respectively. The effects of low-level physical activity on annual OOPE were small at the bottom quantiles but more pronounced at higher quantiles. Respondents with low-level activity had an increased annual OOPE of 26.9 USD, 150.3 USD, and 1,534.4 USD, at the 10th, 50th, and 90th percentiles, respectively, compared with those with high-level activity. The effects of overweight and obesity on OOPE were also small at the bottom quantiles but more pronounced at higher quantiles. CONCLUSION: Interventions that improve the lifestyles and unhealthy behaviors among people with obesity and physical inactivity are likely to yield substantial financial gains for the individual and health systems in China.

Some species of cover crops produce phenolic compounds with allelopathic potential. The use of math, statistical and computational tools to analyze data obtained with spectrophotometry can assist in the chemical profile discrimination to choose which species and cultivation are the best for weed management purposes. The aim of this study was to perform exploratory and discriminant analysis using R package specmine on the phenolic profile of Secale cereale L., Avena strigosa L. and Raphanus sativus L. shoots obtained by UV-vis scanning spectrophotometry. Plants were collected at 60, 80 and 100 days after sowing and at 15 and 30 days after rolling in experiment in Brazil. Exploratory and discriminant analysis, namely principal component analysis, hierarchical clustering analysis, t-test, fold-change, analysis of variance and supervised machine learning analysis were performed. Results showed a stronger tendency to cluster phenolic profiles according to plant species rather than crop management system, period of sampling or plant phenologic stage. PCA analysis showed a strong distinction of S. cereale L. and A. strigosa L. 30 days after rolling. Due to the fast analysis and friendly use, the R package specmine can be recommended as a supporting tool to exploratory and discriminatory analysis of multivariate data.

OBJECTIVE: To evaluate the early stages of the setting process of chemically activated restorative glass-ionomer cements (GICs). MATERIAL AND METHODS: Five GICs were evaluated (n = 5): Equia Forte (GC), Equia Forte HT (GC), Ketac Universal (3M ESPE), Maxxion R (FGM) and Riva Self Cure (SDI) by Thermography, Fourier Transform Infrared Attenuated Total Reflectance Spectroscopy (FTIR-ATR) and Gillmore needle indentation mechanical testing. The FTIR-ATR spectra showed the formation of metal carboxylates within the cements and enabled the stabilization time (ST) to be determined and the thermographic camera measured the temperature field images in the sample. Data were statistically analyzed by ANOVA and Tukey-Kramer (α = 5%). RESULTS: The Gillmore needle test showed that the order of hardening was opposite to the order of ST values determined by FTIR. The results with the thermographic camera showed two stages of temperature variation, which coincided with the evolution of specific infrared bands. The exception was Maxxion R, which showed only a single step change in temperature. CONCLUSION: The early stages of the GIC setting reaction show temperature changes, both endothermic and exothermic, at specific times, confirming the occurrence of individual chemical reactions. The early setting involves reactions other than carboxylate formation.Significance: This study gives further detail of the early stages of the setting of GICs, and past research regarding the setting reaction of GIC.

The antifungal application of photodynamic therapy (PDT) has been widely explored. According to superficial nature of tinea capitis and the facility of application of light sources, the use of nanoencapsulated hypericin in P-123 associated with PDT (P123-Hy-PDT) has been a poweful tool to treat this pathology. Thus, the aim of this study was to evaluate the efficiency of P123-Hy-PDT against planktonic cells and in a murine model of dermatophytosis caused by Microsporum canis. In vitro antifungal susceptibility and in vivo efficiency tests were performed, including a skin toxicity assay, analysis of clinical signs by evaluating score, and photoacoustic spectroscopy. In addition, tissue analyses by histopathology and levels of pro-inflammatory cytokines, such as quantitative and qualitative antifungal assays, were employed. The in vitro assays demonstrated antifungal susceptibility with 6.25 and 12.5 µmol/L P123-Hy-PDI; these experiments are the first that have used this treatment of animals. P123-Hyp-mediated PDT showed neither skin nor biochemical alteration in vivo; it was safe for dermatophytosis treatment. Additionally, the treatment revealed rapid improvement in clinical signs at the site of infection after only three treatment sessions, with a clinical score confirmed by photoacoustic spectroscopy. The mycological reduction occurred after six treatment sessions, with a statistically significant decrease compared with untreated infected animals. These findings showed that P123-Hy-PDT restored tissue damage caused by infection, a phenomenon confirmed by histopathological analysis and proinflammatory cytokine levels. Our results reveal for the first time that P123-Hy-PDT is a promising treatment for tinea capitis and tinea corporis caused by M. canis, because it showed rapid clinical improvement and mycological reduction without causing toxicity.

Selective catalytic reduction of NO with CO (CO-SCR) was investigated based on optimizing the operating conditions by response surface methodology (RSM) and by appropriately choosing the supported SBA-15 catalysts. The effects of the CO-SCR reaction parameters such as NO:CO molar ratios and oxygen concentrations on the catalytic performance were determined by RSM to evaluate the NO conversion using a first-order polynomial model. The CuO/SBA-15 and Fe2O3/SBA-15 catalysts were synthesized by a hydrothermal method and characterized by X-ray diffraction (XRD), atomic absorption spectroscopy (AAS), N2 adsorption-desorption (BET), scanning electron microscopy coupled to energy dispersive X-Ray spectroscopy (SEM-EDS), and Fourier transform infrared spectroscopy (FTIR) to investigate the physicochemical properties of the solids. The RSM showed a very good agreement between predicted values and experimental results with the Pareto analysis confirming the accuracy and reliability of the model. The optimized results indicated the maximum NO conversion at 500 °C with using the NO to CO molar ratio of 1:2 (500:1000 ppm) in the absence of oxygen. Under these conditions, CuO/SBA-15 catalyst achieved 99.7% of NO conversion, whereas Fe2O3/SBA-15 had 98.1% of the catalytic parameter. Catalytic tests in CO-SCR reaction were performed on both catalysts at optimum operating conditions with CuO/SBA-15 exhibiting better performance compared to that of Fe2O3/SBA-15. The results revealed that CuO/SBA-15 was a promising catalyst for CO-SCR of NO due to the well-dispersed CuO phase on SBA-15 surface that allows the solid being more tolerant to the presence of oxygen.

Although Melanaphis sacchari Zehntner (Hemiptera: Aphididae) is known worldwide for its injury to sugarcane, Saccharum officinarum L., and sorghum, Sorghum bicolor (L.) Moench, M. sacchari is reported to infest several plants in the family Poaceae, including important agronomic crops. It is not clear how M. sacchari interacts with other crops in the Louisiana agroecoscape but there is potential for these plants to be bridging hosts prior to sorghum colonization. Thus, this study determined the population dynamics of M. sacchari on sugarcane, sorghum, rice (Oryza sativa L.), sweetpotato (Ipomea batatas L.), maize (Zea mays L.), Johnsongrass (S. halepense L.), and wheat (Triticum aestivum L.) using life table studies. Melanaphis sacchari was able to complete its life cycle on sorghum, sugarcane, rice, and wheat, having the highest intrinsic rate of increase (rm) on sorghum (0.466). Population growth was negative on rice (rm = -0.020) and intermediately positive on sugarcane and Johnsongrass. The results demonstrate that there is the potential for multiple host use within the agricultural landscape. Using the information collected from this study, population estimations on host plants demonstrate that M. sacchari populations will increase more rapidly on sorghum than on the other host plants.
