Ontogeny of the digestive tract and its accessory organs and their further development in the Indian walking catfish (Clarias magur) were examined in larvae, starting from the day of hatching until 35 days post-hatching (dph) reared at 28-29 °C. Many organs at their primordial stage were seen on the day of hatching. These include opened oral cavity with monolayered epithelial lining and very few newly emerging taste buds and goblet cells, primordial pharyngeal teeth on slightly stratified epithelia of the pharyngeal plate, stomach anlage with some degree of the mucosal fold, and a few newly forming gastric glands embedded under its mucosa, primordial anterior and posterior intestine with the smooth mucosal surface, anal opening, and primordial liver and pancreas. At 1 dph, the stomach appeared to be bilobed with the first evidence of food particle in it, and the intestine had some initial folding. On the day of hatching, goblet cells appeared in all lengths of the gut, but not densely, except in the stomach; on it, they appeared at 2 dph. Pancreatic zymogen granules also appeared on this day. Supranuclear vesicles first appeared on 4-5 dph (7.9 ± 0.5-8.6 ± 0.8 mm TL), and they continue to exist until 35 dph. The developmental sequence in this fish confirmed it as an altricial species with some major histomorphological events after the onset of feeding; these include-the appearance of fully developed-pharyngeal teeth at 4 dph and onwards, pyloric sphincter, anterior to posterior intestinal sphincter at 6 dph, and the continuous development of buccopharyngeal cavity and stomach in their shape, size, and functionality until the completion of metamorphosis. Overall, the information on gastrointestinal development in the early life stage of C. magur will be useful for understanding its larval digestive physiology, and this, in turn, will help in designing effective larval feed for growth and survival.

The study was conducted to investigate the expression and activity of key lipolytic enzymes during the ontogenetic development of Clarias magur. After partial characterization, the messenger RNA (mRNA) expression analysis of lipoprotein lipase (LPL), pancreatic triacylglycerol lipase (PL), and bile salt-activated lipase (BAL) genes along with the specific lipase activity were performed in larvae from Day 1 after hatching till 34-day posthatch (dph). Heterogeneous patterns of mRNA expression were shown by the important lipolytic enzymes and were detected before first exogenous feeding during the yolk-sac stage. LPL started increasing from 13 dph and peaked at 16 dph followed by a declining trend till 34 dph. However, the PL observed to be peaking at 9, 22, and 30 dph. Similarly, BAL showed an increasing trend from 11 to 22 dph with a significantly high level of mRNA expression at 16 dph. Later, the specific lipase activity was evaluated which appears at Day 1 after hatching with a progressive increase from 7 to 16 dph and a further declining trend afterwards with a peak at 22 dph. The results indicated the development of exocrine pancreas at 16 dph. Furthermore, the transcript levels and the activity of lipases were regulated with the age. Hence, the present study can be helpful in devising different strategies containing optimum lipid levels at a suitable stage of development for improving the survival during larval rearing. Furthermore, the study could be a baseline for elucidating the optimized dietary lipid levels of this catfish during its larval rearing.

Monogenea (gill parasite) is a major problem in aquaculture that reduces the growth of cultured fish and adversely affects the economy. The present study was performed to evaluate the impact of various degrees of Thaparocleidus sp. (dactylogyrids, monogenean) infestation on haematological and serum biochemical parameters of Pangasianodon hypophthalmus. A standard cohabitation study, following complete randomized design in triplicate, was conducted to obtain low, moderate and high degrees of infestation in P. hypophthalmus along with the control (uninfested) group. Blood and serum were studied for haematological (total erythrocyte count (TEC), haemoglobin (Hb), packed cell volume (PCV), total leucocyte count (TLC) and indices viz. mean cell volume (MCV), mean cell haemoglobin (MCH), mean cell haemoglobin concentration (MCHC) and erythrocyte osmolarity brittleness (EOB)) and serum biochemical parameters (serum glutamate oxaloacetate transaminase (SGOT), serum glutamate pyruvate transaminase (SGPT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), lactate, total bilirubin and creatinine. Significant (p < 0.05) increase in TEC, Hb, TLC, EOB, SGOT, SGPT, LDH, lactate, bilirubin, and creatinine were noticed in moderate to high monogenean-infested group in comparison to the control group. However, significant (p < 0.05) reduction in MCH, and MCV and no difference (P > 0.05) in PCV were noticed in high degree parasitized group in comparison to the control group. The results of altered haematological and serum biochemical parameters in various degrees of monogenean-infested groups signify the density dependent physiological responses and changes in cells of the blood. The data of serum enzymes in the present study would be valuable for assessing the health status of the host and facilitate as a potential biomarker in relation to various degrees of monogenean infestation.

Modulation of innate immune activity and oxidative stress response of Pangasianodon hypophthalmus through experimental infection with (Thaparocleidus sp.) dactylogyrid monogenean was studied. A standard cohabitation method was used to infect healthy experimental fish. After 14 days, dactylogyrid (gill monogenean) infested fish were sampled and categorised into three different infected groups namely (T1) low (<10 mean dactylogyrid per gill arch per fish), (T2) moderate (10-49 mean dactylogyrid per gill arch per fish) and (T3) high (>50 mean dactylogyrid per gill arch per fish) along with a control group T0 (un-infested fish). Serum and tissues (gills and liver) were collected from experimental fish and analyzed for markers of innate immune and oxidative stress, respectively. The results showed that respiratory burst activity, myeloperoxidase level, serum lysozyme, α-2 macroglobulin and total serum immunoglobulin level were significantly (p < 0.05) elevated in fish with different degrees of parasite infestation compared to the control (un-infested group). Similarly, cellular oxidative biomarkers superoxide dismutase, catalase, glutathione-S-transferase and Na+-K+-ATPase activities of gills and liver were significantly (p < 0.05) elevated in dactylogyrid infested fish in comparison to the control. However, significantly decreased level of albumin, albumin to globulin ratio, total serum antiprotease and ceruloplasmin were observed in fish infested with low degree of dactylogyrids, while no significant differences in these parameters were observed between moderately infested and the control groups. The results suggested that varying degree of gill monogenean dactylogyrid infestation not only modulated the innate immune response of P. hypophthalmus by lowering albumin, total serum antiprotease and ceruloplasmin and inducing respiratory burst activity, phagocytic activity, myeloperoxidase, lysozyme, α-2 macroglobulin and total immunoglobulins, but also the oxidative stress biomarkers. The baseline data obtained in the present study will be valuable in understanding the host-parasite relationship and the dynamics of innate, oxidative stress responses and susceptibility of P. hypophthalmus to different degrees of parasitosis.

The effect of letrozole and tamoxifen on the specific growth rate (SGR; % day(-1)), gonado-somatic index (GSI), total haemoglobin (g%), gonadal and serum protein as well as lipid, sex differentiation and 17ß-oestradiol levels were studied in sexually undifferentiated Cyprinus carpio fingerlings 30 days post fertilisation (30 dpf) for 60 days. Results showed decreased GSI with tamoxifen treatment whereas letrozole increased it. There were reduced protein, lipid, triglyceride and cholesterol levels after treatment with tamoxifen and letrozole during gonadal development. Tamoxifen (200mgkg(-1) feed) induced 82.5% masculinisation, whereas letrozole in the same dose produced 98.5% males. Gonadal 17ß-oestradiol significantly declined from 86.0±1.41pg per 100mg (control) to 45.5±1.94pg per 100mg with tamoxifen and 36.0±0.72pg per 100mg with letrozole treatment. Similarly, serum 17ß-oestradiol levels also decreased after tamoxifen and letrozole treatments. Testicular development in 37.8% of fish treated with tamoxifen and letrozole was found to be more advanced (spermatocytes) than in the control (spermatogonium); however, there was reduced ovarian growth and increased atresia. It was concluded that letrozole and tamoxifen both significantly affect sex differentiation and gonadal maturity in C. carpio leading to the production of sex-reversed males, yet the effect of letrozole was more potent.

Chronic diseases have been treated using the phytochemical concepts of ethnomedicinal plant-derived herbal products. Terminalia arjuna, a significant ethnomedicinal plant, was revisited and reconnoitred for antioxidant, free radical scavenging, and DNA nicking inhibiting activity under H2O2 conditions using 21 solvent extracts. Ferric reducing antioxidant power (FRAP), 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid, and nitrous oxide scavenging (%) were found to have a strong positive association and interaction (PCA 1 explains 84.54% variation) with ethanol bark (Etoh-AB) (Meoh-AF). TPC (144.67-1794 µg/mL GAE) and TFC (2.5-34 µM Fe(II)/g were highest in Etoh-AB. In a pattern of combined solvent extracts, Etoh-AB had the highest antioxidant capacity, accompanied by Etoh-AL ≥ Meoh-AB ≥ Dw-AF. With R2 = 0.94, the DNA nicking inhibition behaviour parameters relative front, relative quantity, band (%), and lane (%) formed a positive significant (p < 0.01) connection. For the first time, we show that Etoh-AB nicks supercoiled, circular plasmid DNA in a way that is comparable to normal antioxidants. Normal antioxidants with the ability to prevent DNA nicking include Butylated hydroxy anisole < Butylated hydroxy toluene < ascorbic acid < and Gallic acid. Gallic acid (m/z 170.0208 g/mol) and Ellagic acid (m/z 302.0063 g/mol were present in high concentrations in solvent extracts. 0.48 mg was found to be the effective concentration for inhibiting relative DNA nicking. The current study is the first of its kind to show that steroid concentrations are higher in bark fractions of acetone, ethanol, and methanol. Furthermore, T. arjuna solvent extracts provide a wealth of information on phytochemical profiling, antioxidant ability, and DNA nicking inhibition, which may be useful for exploring the natural way and further research to develop a remedy against geriatric chronic disease. Despite the fact that ethanol is very close to methanol in terms of solvent toxicity, the current study identified it as the preferred solvent. Thus, the current research revisits previous studies and explores the potentiality of non-polar and polar aprotic and polar protic solvent systems, which lend credence to bioactive compounds that may be useful in isolating and formulating safe and cost effective herbal medicament for livestocks and aquaculture, and drugs for deoxygenerative human diseases, and can also be investigated further to instil environmental frugality.

Evaluation of genetic resources is an essential prerequisite for their effective utilization. In India, the tropical climate prevails in most of the sericultural belts, where temperature goes beyond the ambient during summer, adversely affecting the silkworm rearing. Although polyvoltine silkworms are poor in silk content, they are mostly tolerant to tropical conditions and diseases. With an aim to identify potential silkworm races/breeds specific to thermo-tolerance for their effective utilization in breeding programme, 15 selected polyvoltine silkworm races were studied for their thermo-tolerance behaviour. Their genomic DNA samples were analyzed for ISSR-PCR using 15 selected primers. The UPGMA analysis based on Nei and Li algorithm has clustered the 15 silkworm races into five groups and one isolate. ALSCAL-multidimensional scaling has not only supported the information generated by the dendrogram, but it has made the genetic distance among races more clear and substantiating their status in terms of thermal stress where pupation rate was taken as indicator. Further, discriminant function analysis (DFA) was done with three groups of silkworms classified for thermal stress viz. susceptible, moderately tolerant and tolerant. The canonical correlation value was estimated to be 0.987 (Wilk's lambda = 0.004; chi2 = 36.044, p < 0.05). DFA clearly discriminated the above three groups. Beta statistics with t value and its significance for the markers identified through stepwise multiple regression analysis (MRA) revealed a total of five bands (807(1300), 808(3000), 808(4000), 834(4000), and 834(3000)) showing correlation with pupation rate after thermal treatment. Out of them, marker 8083000 showed maximum and highly significant correlation (r = 0.757, p < 0.001, t = 4.182) with pupation rate among the silkworm races. The identified putative markers are being used to develop DNA marker to be used in evolving thermo-tolerant silkworm breeds using marker assisted selection programme.

Capsaicin exerts a stabilizing effect on erythrocytes making them more resistant to lysis under hypotonic stress. The protective action of capsaicin on osmotic fragility (OF) was not receptor mediated since no dose responsive effect was observed. The results suggest that this protective effect of capsaicin on OF is due to a direct interaction of capsaicin with the erythrocyte membrane rather than due to any alteration in the intracellular metabolism of erythrocytes.

Species identification in mulberry (Morus) continues to be a point of great debate among scientists despite the number of criteria such as floral characters, wood, and leaf anatomical and biochemical characters used to identify the species within this genus. However, no consensus system of classification has emerged. Hence, an investigation was undertaken with inter-simple sequence repeat (ISSR) and random amplified polymorphic DNA (RAPD) markers to find out the possibility of using these DNA markers to confirm the identity of genotypes in a particular species. Fifteen ISSR and 15 RAPD primers generated 86% and 78% polymorphism, respectively, among 19 mulberry genotypes. The polymorphism among the species varied from 50% to 57% in ISSR markers and 31% to 53% in RAPD markers. Similarity coefficients were higher among the genotypes of M. latifolia, M. bombycis and M. alba. Cluster analyses separated genotypes of M. laevigata and M. indica from those of the other species. Population structure analysis of these species further showed high genetic differentiation coefficients (GST), high heterozygosity between two species (DST), and total heterozygosity among populations (Ht) coupled with considerably low gene flow (Nm) when M. laevigata was paired with other species. Based on these parameters and the result of cluster analysis it is concluded that M. laevigata can be considered as a separate species of mulberry, whereas the other four species may be grouped together and treated as subspecies.

Morus spp., commonly known as mulberry, is significantly associated with human civilization and spread of silk-culture from Asia to Europe, Africa and Latin America. One of its species, Morus laevigata, traditionally well known for its timber value, forage use and silkworm's feed, is widely distributed in India extending from Himalayan foothill to Andaman islands. The variability occurring for 12 morpho-biochemical parameters and RAPD profiles, generated with 13 selected RAPD primers, for M. laevigata accessions from six different zones were investigated. Analyses revealed high degree of genotypic similarity of collection from Himalayan foothill (West Bengal) with those from Andaman Islands. Specific accessions from central India and south India also revealed genotypic similarities with specific accessions from north-east India. These observations are discussed in the context of clonal propagation of mulberry and evolutionary perspective of dispersal of this species, through human activities

India is one of the countries where sericulture is being practiced traditionally. Due to the higher economic return and the greater employment potential, attempts are being made to increase the productivity by developing high yielding mulberry varieties. At the present, Mysore local, Bomaypiasbari, Kanva-2, Bilidevalaya, Kajli, S1, BC(2)59, C776, RFS-175, S-36 and Victory-1 are being cultivated extensively in different parts of India for rearing the silkworm Bombyx mori L. Using 17 random amplified polymorphic DNA (RAPD) and 11 inter-simple sequence repeat (ISSR) primers the genetic relationships among these varieties were analyzed. The RAPD and ISSR primers revealed more than 75% polymorphism among the varieties. The genetic similarity estimated from RAPD markers varied from 0.645, between Kajli and Victory-1 to 0.887, between Kanva-2 and Bilidevalaya. Similarly, the genetic similarity estimated from the ISSR markers ranged from 0.600, between Kajli and Victory-1, to 0.873 between Kanva-2 and BC(2)59. The dendrogram constructed from these markers grouped the varieties into three major groups comprising the low yielding, medium yielding and high yielding. The low genetic similarity between the group of varieties originating from the eastern regions with that of the southern region encourages formation of extensive breeding programs between these groups as to transfer the high yield potential of the southern varieties to the low yielding but highly adaptive eastern varieties.
