KEY MESSAGE: In our study, we discovered a fragment duplication autoregulation mechanism in 'ZS-HY', which may be the reason for the phenotype of red foliage and red flesh in grapes. In grapes, MYBA1 and MYBA2 are the main genetic factors responsible for skin coloration which are located at the color loci on chromosome 2, but the exact genes responsible for color have not been identified in the flesh. We used a new teinturier grape germplasm 'ZhongShan-HongYu' (ZS-HY) which accumulate anthocyanin both in skin and flesh as experimental materials. All tissues of 'ZS-HY' contained cyanidin 3-O-(6″-p-coumaroyl glucoside), and pelargonidins were detected in skin, flesh, and tendril. Through gene expression analysis at different stage of flesh, significant differences in the expression levels of VvMYBA1 were found. Gene amplification analysis showed that the VvMYBA1 promoter is composed of two alleles, VvMYBA1a and 'VvMYBA1c-like'. An insertion of a 408 bp repetitive fragment was detected in the allele 'VvMYBA1c-like'. In this process, we found the 408 bp repetitive fragment was co-segregated with red flesh and foliage phenotype. Our results revealed that the 408 bp fragment replication insertion in promoter of 'VvMYBA1c-like' was the target of its protein, and the number of repeat fragments was related to the increase of trans-activation of VvMYBA1 protein. The activation of promoter by VvMYBA1 was enhanced by the addition of VvMYC1. In addition, VvMYBA1 interacted with VvMYC1 to promote the expression of VvGT1 and VvGST4 genes in 'ZS-HY'. The discovery of this mutation event provides new insights into the regulation of VvMYBA1 on anthocyanin accumulation in red-fleshed grape, which is of great significance for molecular breeding of red-fleshed table grapes.

RESUMO

Forward collision warning (FCW) is a critical technology to improve road safety and reduce traffic accidents. However, the existing multi-sensor fusion methods for FCW suffer from a high false alarm rate and missed alarm rate in complex weather and road environments. For these issues, this paper proposes a decision-level fusion collision warning strategy. The vision algorithm and radar tracking algorithm are improved in order to reduce the false alarm rate and omission rate of forward collision warning. Firstly, this paper proposes an information entropy-based memory index for an adaptive Kalman filter for radar target tracking that can adaptively adjust the noise model in a variety of complex environments. Then, for visual detection, the YOLOv5s model is enhanced in conjunction with the SKBAM (Selective Kernel and Bottleneck Attention Mechanism) designed in this paper to improve the accuracy of vehicle target detection. Finally, a decision-level fusion warning fusion strategy for millimeter-wave radar and vision fusion is proposed. The strategy effectively fuses the detection results of radar and vision and employs a minimum safe distance model to determine the potential danger ahead. Experiments are conducted under various weather and road conditions, and the experimental results show that the proposed algorithm reduces the false alarm rate by 11.619% and the missed alarm rate by 15.672% compared with the traditional algorithm.

Mangosteen (Garcinia mangostana Linn.) is an evergreen fruit tree grown in Southeast Asia. The fruit of Mangosteen is dark purple spherical with a diameter of 5 to 8 centimeter, a hard rind and white flesh. The seeds and pericarps of the fruit have long been utilized as a traditional medicine in Southeast Asia (Ovalle-Magallanes et al. 2017). Surveys were conducted during the months from March to April 2019 of fruit markets located in Wujing Town, Shanghai (31°02'33″ N, 121°28'04″ E), one noteworthy postharvest disease was observed. Overall 116 fruit samples were collected from 5 different supermarkets, 10.3% (12) of the fruits were found infected with the same symptoms during cold storage. The infected fruits were subjected to pathogen isolation and characterization. The junction areas between diseased and healthy tissues were cut into small pieces of diameter 5 mm, which were surface sterilized with 0.5% NaClO solution for 3 min, followed by rinsing in sterile water thrice, and then the tissues were placed on sterilized potato dextrose agar (PDA) plates supplemented with 50 µg/ml streptomycin. The colony edges of resulted fungal cultures were cut and removed to new PDA plates for purification after 2 days incubation at 28°C. After 3 rounds of subculture, a total of 19 purified isolates were obtained and these isolates showed similar phenotypes in morphological characters. The fungal colonies were fast growing (25 mm per day at 28°C on PDA plates) and were circular shaped with massive aerial hyphae, white to ochraceous color. Non-septate hyphae were hyaline and the septa were found only at the branching points. Sporangiophores arose from hyphae or from stolons. Sporangiospores were hyaline, smooth-walled, mostly subspherical to ellipsoidal, and measured 1.34 to 2.73 µm (width) and 3.12 to 6.19 µm (length) (1.98 ± 0.36×4.46 ± 0.69, n=100)(Alvarez et al. 2011). Three independent isolates (GM-07, GM-16, GM-19) were selected used for further genetic analysis and pathogenicity test. The internal transcribed spacer (ITS) and the D1/D2 region of the nuclear large subunit ribosomal RNA gene (LSU) were amplified by primers ITS1/ITS4 and primers NL1/LR3, respectively (Walther et al. 2013). BLASTn analyses of ITS (MN081612, MN081613, MN087659) and LUS (MN080423, MN080422, MN080432) sequences exhibited 99 to 100% identify to Mucor irregularis strains HZ640, extr25 and CBS977.68 (Accession numbers KC461926, MH510278 of ITS and JX976214 of LUS). These observed morphological features and molecular identification indicated that the pathogen possessed identical characteristics as Mucor irregularis Stchigel, Cano, Guarro & Ed. Alvarez (Alvarez et al. 2011). To conduct a pathogenicity test (Al-Sadi et al. 2014), 20 µl of spore suspension (106 spores ml-1) were inoculated into five healthy mangosteen fruits by sterile syringes, meanwhile another five healthy fruits were injected with sterilized water as controls. All inoculated fruits were kept in sealed plastic boxes at 28°C with a 12L/12D illumination cycle and 90% relative humidity. After about 3 to 7 days, the inoculated materials showed similar symptoms to those on the original diseased fruits, while control samples remained symptomless. M. irregularis was successfully re-isolated from symptomatic fruits, fulfilling Koch's postulates. Only three plant species that can be infected by M. irregularis have been reported so far, including key lime from Oman (Al-Sadi et al. 2014), corn from China (Peng et al. 2015), and sorghum from Brazil (Lima et al. 2018). To our knowledge, this is the first report of M. irregularis causing the postharvest rot on mangosteen fruit in China.

OBJECTIVE: Dexmedetomidine (DEX) can strengthen the analgesic effects of local anesthetics (LAs) when used as an adjuvant through intrathecal, perineural, and intraperitoneal routes. Many studies have used intraperitoneal instillation of DEX with LAs in laparoscopic cholecystectomy (LC) to relieve postoperative pain. We performed a systematic review and meta-analysis to synthesize evidence of the efficacy and safety of intraperitoneal instillation of DEX as an adjuvant of LAs in patients undergoing LC. METHODS: A comprehensive literature search of the MEDLINE, PubMed, EMBASE, and Cochrane Library databases was performed to identify randomized controlled trials in which patients received intraperitoneal instillation of DEX combined with LAs during LC. A meta-analysis and sensitivity analysis of the results were conducted. We also performed a subgroup analysis to investigate the source of heterogeneity. The Egger test was used to check for publication bias. RESULTS: Eleven randomized controlled trials involving 890 patients were analyzed. We found that the addition of DEX to LAs significantly decreased pain scores at six postoperative time points (0.5, 1, 2, 4, 12, and 24 h) and significantly prolonged the time to the first analgesic request by patients. In addition, 24-hour postoperative analgesic consumption was decreased in the experimental group, and no significant difference in the incidence of nausea and vomiting was observed. CONCLUSION: Our findings indicate that intraperitoneal instillation of DEX with LAs can reduce postoperative pain and prolong the time to first request analgesia after LC.

Coal gasification crude slag (CGCS) is the side-product of the coal gasification process, and its effective utilization has attracted great attention. A novel flocculant of poly-aluminum-ferric-acetate-chloride (PAFAC) was synthesized based on the recovery of CGCS by a two-step acid leaching process, namely HCl-acid leaching and HAc-acid leaching, which was optimized by an acid leaching liquor volume ratio of HCl to HAc of 3 : 2, polymerization pH of 3.5, and reaction temperature and time of 70 °C and 3.0 h, respectively. The performance of PAFAC was further evaluated by kaolin simulated wastewater, domestic sewage, river water, and aquaculture wastewater. The results revealed that PAFAC was feasible for the removal of turbidity, chemical oxygen demand (COD) and total phosphorus (TP). Moreover, PAFAC was characterized by X-ray Diffraction (XRD), Fourier transform infrared spectroscopy (FT-IR), X-ray fluorescence spectrometry (XRF) and scanning electron microscopy (SEM), which proved that PAFAC was a kind of amorphous polyionic composite. Additionally, the acid leaching kinetics and flocculation mechanisms were further investigated. It was found that the acid leaching process was followed by the unreacted shrinkage core model, and the flocculation process was dominated by charge neutralization, adsorption bridging and precipitation net trapping. The work is expected to develop a new method for the safe disposal of CGCS and provide a novel way for the preparation of Fe-Al composite flocculants, especially, offering a potential strategy for the promotion of the additional value of the coal chemical industry.

A novel magnetic carbon-silicon composite (Fe-HH-CGFS) was prepared from solid waste coal gasification fine slag (CGFS) by a two-step acid leaching and one-step chemical co-precipitation process, which was optimized using a 3-factor, 3-level Box-Behnken design and then analyzed for correlation. Fe-HH-CGFS was characterized by scanning electron microscopy (SEM), X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), Brunauer-Emmett-Teller (BET), thermal gravimetric analysis (TGA), and vibrating sample magnetometer (VSM) measurements. The results demonstrated that Fe-HH-CGFS had a reverse spinel structure with an average particle size of 5.14 nm, exhibiting a microporous/mesoporous structure with a specific surface area (SSA) of 196.84 m2 g-1 and pore volume of 0.346 cm3 g-1. Furthermore, Fe-HH-CGFS could achieve 97.59% removal efficiency of rhodamine B (RhB) under the optimal conditions: an initial concentration of RhB of 100 mg L-1, an adsorption time of 60 min, and a dosage of Fe-HH-CGFS of 1.0 g L-1. The pseudo-second-order model and the Langmuir isotherm satisfactorily described the adsorption behavior. The results indicated that the RhB removal process was a single-molecule layer endothermic adsorption, which is dominated by chemical adsorption reactions. This work is expected to provide an alternative route for the high-value utilization of CGFS and offer a valuable insight for the recycling of other solid wastes, aligning with the green development concept of "treating wastes with wastes".

Fresh Allium mongolicum Regel (FA) and dried A. mongolicum Regel (DA) are significantly different in antioxidant activity. However, the relevant mechanisms have not yet been explored. We evaluated the antioxidant activities of two varieties of FA and DA and characterized their metabolites using targeted metabolomics. The effect of different metabolites on the antioxidant activity of A. mongolicum Regel was investigated by multivariate analysis. A total of 713 metabolites were detected in all samples. Pearson correlation analysis demonstrated that the key primary metabolites were directly and significantly correlated with the total phenolic content (TPC) and total flavonoid content (TFC), while the secondary metabolites were directly correlated with antioxidant activity. The higher antioxidant activity of DA may be mainly attributed to the higher TPC and TFC. This study revealed the potential mechanism by which drying enhances the antioxidant activity of A. mongolicum Regel.

AIMS: Alzheimer's disease (AD) is a significant global health concern, and it is crucial that we find effective methods to prevent or slow down AD progression. Recent studies have highlighted the essential role of blood vessels in clearing Aß, a protein that contributes to AD. Scientists are exploring blood biomarkers as a potential tool for future AD diagnosis. One promising method that may help prevent AD is remote ischemic conditioning (RIC). RIC involves using sub-lethal ischemic-reperfusion cycles on limbs. However, a comprehensive understanding of how RIC can prevent AD and its long-term effectiveness is still lacking. Further research is essential to fully comprehend the potential benefits of RIC in preventing AD. METHODS: Female wild-type (WT) and APP/PS1 transgenic rats, aged 12 months, underwent ovariectomy and were subsequently assigned to WT, APP/PS1, and APP/PS1 + RIC groups. RIC was conducted five times a week for 4 weeks. The rats' depressive and cognitive behaviors were evaluated using force swimming, open-field tests, novel objective recognition, elevated plus maze, and Barnes maze tests. Evaluation of the neurovascular unit (NVU), synapses, vasculature, astrocytes, and microglia was conducted using immunofluorescence staining (IF), Western blot (WB), and transmission electron microscopy (TEM). Additionally, the cerebro-vasculature was examined using micro-CT, and cerebral blood flow (CBF) was measured using Speckle Doppler. Blood-brain barrier (BBB) permeability was determined by measuring the Evans blue leakage. Finally, Aß levels in the rat frontal cortex were measured using WB, ELISA, or IF staining. RESULTS: RIC enhanced memory-related protein expression and rescued depressive-like behavior and cognitive decline in APP/PS1 transgenic rats. Additionally, the intervention protected NVU in the rat frontal cortex, as evidenced by (1) increased expression of TJ (tight junction) proteins, pericyte marker PDGFRß, and glucose transporter 1 (GLUT1), as well as decreased VCAM1; (2) mitigation of ultrastructure impairment in neuron, cerebral vascular, and astrocyte; (3) upregulation of A2 astrocyte phenotype markers and downregulation of A1 phenotype markers, indicating a shift toward a healthier phenotype. Correspondingly, RIC intervention alleviated neuroinflammation, as evidenced by the decreased Iba1 level, a microglia marker. Meanwhile, RIC intervention elevated CBF in frontal cortex of the rats. Notably, RIC intervention effectively suppressed Aß toxicity, as demonstrated by the enhancement of α-secretase and attenuation of ß-secretase (BACE1) and γ- secretase and Aß1-42 and Aß1-40 levels as well. CONCLUSION: Chronic RIC intervention exerts vascular and neuroprotective roles, suggesting that RIC could be a promising therapeutic strategy targeting the BBB and NVU during AD development.

The aim of this study was to investigate the effects of dietary Allium mongolicum Regel powder (AMRP) supplementation on the growth performance, meat quality, antioxidant capacity and muscle fibre characteristics of fattening Angus calves. Growth performance data and longissimus thoracis (LT) samples were collected from four groups of fattening Angus, which were fed either a basal diet (CON) or a basal diet supplemented with an AMRP dose of 10 (LAMR), 15 (MAMR), or 20 g/animal/day AMRP (HAMR) for 120 days before slaughter. AMRP addition to the feed improved growth performance and meat quality and altered muscle fibre type. Some responses to AMRP supplementation were dose dependent, whereas others were not. Together, the results of this study demonstrated that dietary supplementation with 10 g/animal/day AMRP was the optimal dose in terms of fattening calf growth performance, while 20 g/animal/day AMRP supplementation was the optimal dose in terms of meat quality.

Coal gasification fine slag (FS), a kind of by-product of coal chemical industry, was recovered for the preparation of functional adsorbents by acid leaching process, which was orthogonally optimized by HCl, HNO3, HF, HAc, and H2SO4. Methylene blue (MB) was used to evaluate the performance of functional adsorbents. The results demonstrated that 57.6% of the leaching efficiency (RLE) and 162.94 mg/g of adsorption capacity (CAC) of MB were achieved under the optimal conditions of HNO3 of 2.0 mol/L, acid leaching time of 2.0 h, and acid leaching temperature of 293K. The detections on X-ray Diffraction (XRD), Scanning Electron Microscope (SEM), Fourier Transform Infrared Spectroscopy (FTIR), and BET surface area (SBET) indicated that the synthesized functional adsorbents were characterized by mesoporous materials. The good fitting of adsorption process using pseudo-second-order and Langmuir models demonstrated that the chemisorption contributed to MB removal. The results of thermodynamics further revealed that the adsorption process of MB occurred spontaneously due to the exothermic properties. The work is expected to develop a novel and cost-effective strategy for the safe disposal of FS, and potentially offer an alternative pathway to increase the additional value for the coal chemical industry.
