RESUMO

In laser-driven inertial confinement fusion (ICF) facilities, nonuniform laser irradiation can cause significant challenges, such as hydrodynamics instability and laser plasma instability, which hinder the success of fusion. This article presents a new idea for improving the uniformity of far-field laser irradiation through a method of single-beam polarization smoothing. The method involves modulating full Poincaré beams using stress-engineered optics made from fused silica. We designed a stress birefringence system and conducted opto-mechanical modeling and analysis on it. The article elaborates on the mechanism and principles of generating large-aperture full Poincaré beams by stress birefringence, as well as the mechanism of polarization smoothing by full Poincaré beams. Near-field polarization measurements were conducted to verify these mechanisms, and the effectiveness of this method in improving the uniformity of laser irradiation in the target area was evaluated through far-field optical tests.

OBJECTIVE: To integrate the qualitative research on the self-management experience of breast cancer patients and conduct a systematic review of their self-management experience. METHODS: Using a computer to search a series of databases such as CNKI, Wanfang, VIP, and China Biomedical Database, systematically collect and integrate qualitative research on the self-management experience of breast cancer patients, and the search time is limited to January 2010 to December 2022. The qualitative research quality evaluation standard of the Joanna Briggs Institute Centre for Evidence-Based Health Care in Australia was used as the evaluation standard of this project to complete the accurate evaluation of the literature; Meta-analysis was used to complete the effective integration of the results. RESULTS: 17 pieces of literature were included in this project, and 37 research results with strong integrity were extracted accordingly. On this basis, 7 different categories were summarised, and three integrated results were obtained: the experience of maintaining self-management, symptom recognition, and self-management. CONCLUSION: In the different stages of self-management of breast cancer patients, medical staff should give targeted guidance to help patients obtain a good prognosis.

INTRODUCTION: LncRNA rhabdomyosarcoma 2-associated transcript (RMST) serves as a key regulator in neural stem cell fate and is involved in the progression of different neurological diseases. In this research, the serum level and clinical value of RMST in Parkinson's disease (PD) patients were detected, and the underlying mechanism was explored. METHODS: Ninety-nine PD patients and 93 healthy individuals were collected for clinical experiments. SH-SY5Y cells were treated with the dopaminergic neurotoxin 1-methyl-4-phenylpyridinium (MPP+) to establish PD cell models. qRT-PCR was used for the detection of mRNA levels. CCK-8 and flow cytometry were used to detect neuronal viability and apoptosis. The target relationship of RMST with miR-15a-5p was confirmed applying luciferase reporter assay. RESULTS: RMST was present at high levels in both serum of PD patients and PD cell models. Serum RMST had a certain clinical value for the diagnosis of PD with the AUC of 0.892 at a cutoff value of 1.225. Serum RMST was positively associated with the levels of TNF-α (r = 0.421, p < 0.001) and IL-1ß (r = 0.567, p < 0.001) in PD patients. Knockdown of RMST alleviated the apoptosis and inflammatory response of SH-SY5Y cells induced by MPP+. miR-150-5p was the target gene of RMST and less expressed in the clinical serum samples and PD cell models. CONCLUSION: Serum RMST serves as a promising biomarker for the diagnosis of PD. RMST downregulation may regulate the occurrence and development of PD through inhibiting neuron cell apoptosis and the release of inflammatory cytokines via targeting miR-150-5p.

BACKGROUND Epithelial-mesenchymal transition (EMT) is an essential progress for tumor cell invasion to both epithelial and non-epithelial cancers, and zinc finger E-box-binding homeobox 1/2 (ZEB1/2) is a well-known promoter of EMT. In glioma cell lines, both ZEB1 and ZEB2 have been demonstrated to facilitate cancer cell proliferation and invasion with experiments in vitro. However, the clinical significance of ZEB1 and ZEB2 in glioblastoma (GBM) is still controversial. MATERIAL AND METHODS We detected the expression of ZEB1 and ZEB2 in 91 cases of GBM with immunohistochemistry and investigated the correlation between clinicopathological factors and ZEB family expression with Fisher test. By univariate analysis with Kaplan-Meier test, we explored the prognostic significance of ZEB1/2 expression and the clinicopathological factors in GBM. By multivariate analysis with the Cox regression model, we identified the independent prognostic factors in GBM. RESULTS The percentages of ZEB1 high expression and ZEB2 high expression were 31.9% (29/91) and 41.9% (36/91), respectively. High expression of ZEB2 was significantly associated with lower survival rate of GBM patients (P=0.001). ZEB2, lower KPS score (P=0.004), gross total resection (P<0.001) and higher Ki67 percentage (P=0.001) were notably correlated to worse prognosis of GBM. With multivariate analysis, high expression of ZEB2 was demonstrated to be an independent prognostic factor indicating unfavorable prognosis of GBM (P=0.001, HR=3.86, and 95%CI=1.61-9.23). CONCLUSIONS High expression of ZEB2 is an independent prognostic factor predicting unfavorable prognosis of GBM, indicating that ZEB2 or its downstream proteins may be potential drug targets of GBM therapy.

RESUMO

A new relay-imaged off-axial eight-pass laser amplifier with several joules energy and 1 Hz repetition rate was demonstrated. The extraction efficiency and pulse-to-pulse energy stability were greatly improved. Under the single-pass small-signal gain of 3.6, a net gain of 900 and an effective extraction efficiency of 42.4% in the beam aperture were realized. Pulse-to-pulse energy stability of 0.83% (peak-valley) and 0.17% (root-mean-square) was achieved by the significant saturation of eight-pass amplification, which, to the best of our knowledge, is the best energy stability in several-joules-class amplifiers. The far-field quality was 2.52 times the diffraction limit, and the near-field modulation of the 90% beam aperture was 1.28. No parasitic oscillations or pencil beams were observed. Moreover, another key feature of the proposed amplifier was the ability to remarkably improve the pulse contrast with a unique design.

Due to the low concentration of indoor air contaminants, photocatalytic technology shows low efficiency for indoor air purification. The application of TiO2 for photocatalytic removal of formaldehyde is limited, because TiO2 can only absorb ultraviolet (UV) light. Immobilization of TiO2 nanoparticles on the surface of graphene can improve the visible light photocatalytic activity and the adsorption capacity. In this study, rGO (reduced graphene oxide)/TiO2 was synthesized through a hydrothermal method using titanium tetrabutoxide and graphene oxide as precursors, and was used for the degradation of low concentration formaldehyde in indoor air under visible light illumination. Characterization of the crystalline structure and morphology of rGO/TiO2 revealed that most GO was reduced to rGO during the hydrothermal treatment, and anatase TiO2 nanoparticles (with particle size of 15-30nm) were dispersed well on the surface of the rGO sheets. rGO/TiO2 exhibited excellent photocatalytic activity for degradation of formaldehyde in indoor air and this can be attributed to the role of rGO, which can act as the electron sink and transporter for separating photo-generated electron-hole pairs through interfacial charge transfer. Furthermore, rGO could adsorb formaldehyde molecules from air to produce a high concentration of formaldehyde on the surface of rGO/TiO2. Under visible light irradiation for 240min, the concentration of formaldehyde could be reduced to 58.5ppbV. rGO/TiO2 showed excellent moisture-resistance behavior, and after five cycles, rGO/TiO2 maintained high photocatalytic activity for the removal of formaldehyde (84.6%). This work suggests that the synthesized rGO/TiO2 is a promising photocatalyst for indoor formaldehyde removal.

RESUMO

We report a kind of structured light beam with periodical polarization and phase singularities. It is generated from a setup consisting of conical refraction transformation and 4f-system. By this setup, the periodical structures are produced without any change of intensity distributions. We analyze both theoretically and experimentally the polarization and phase structures of the periodically structured light beam. The dependence of period is demonstrated on the length of crystal and the focal length. It is shown that the polarization of the input beam can be used to control the polarization and phase structures of the output beam.

[Purpose] The aim of this study was to determine which is better in the rehabilitation of stroke patients, core stability exercises or conventional exercises. [Subjects and Methods] Forty participants with hemiplegia were recruited in the Department of Neurology of Yidu Central Hospital of Weifang between January 2014 and February 2015 and randomly divided into either an experimental or control group. The patients in the control group performed conventional exercises for six weeks, and those in the experiment group performed core stability exercises for six weeks. The outcomes were evaluated using Modified Barthel Index and Berg Balance Scale. [Results] After treatment, the Modified Barthel Index and Berg Balance Scale were significantly increased in both groups when compared with the baseline. The Modified Barthel Index was significantly lower in the control group compared with the experimental group. The Berg Balance Scale scores in the control group were relatively lower than those in the experimental group, but there was no significant difference between the two groups. [Conclusion] Core stability exercises have a better effect on patients with hemiplegia than conventional exercises.

BACKGROUND: The mortality rate of patients with traumatic brain injury (TBI) is still high even while undergoing decompressive craniectomy (DC), and the expensive treatment costs bring huge economic burden to the families of patients. OBJECTIVE: The aim of this study was to identify preoperative indicators that influence patient outcomes and to develop a risk model for predicting patient mortality by a retrospective analysis of TBI patients undergoing DC. METHODS: A total of 288 TBI patients treated with DC, admitted to the First Affiliated Hospital of Shantou University Medical School from August 2015 to April 2021, were used for univariate and multivariate logistic regression analysis to determine the risk factors for death after DC in TBI patients. We also built a risk model for the identified risk factors and conducted internal verification and model evaluation. RESULTS: Univariate and multivariate logistic regression analysis identified four risk factors: Glasgow Coma Scale, age, activated partial thrombin time, and mean CT value of the superior sagittal sinus. These risk factors can be obtained before DC. In addition, we also developed a 3-month mortality risk model and conducted a bootstrap 1000 resampling internal validation, with C-indices of 0.852 and 0.845, respectively. CONCLUSIONS: We developed a risk model that has clinical significance for the early identification of patients who will still die after DC. Interestingly, we also identified a new early risk factor for TBI patients after DC, that is, preoperative mean CT value of the superior sagittal sinus (p < .05).

The expression level of SLC35A3 is associated with the prognosis of many cancers, but its role in colorectal cancer (CRC) is unclear. The purpose of our study was to elucidate the role of SLC35A3 in CRC. The expression levels of SLC35A3 in CRC were evaluated through tumor immune resource assessment (TIMER), The Cancer Genome Atlas (TCGA), Gene Expression Omnibus (GEO), International Cancer Genome Consortium (ICGC), Human Protein Atlas (HPA), qRT-PCR, and immunohistochemical evaluation. TCGA, GEO, and ICGC databases were used to analyze the diagnostic and prognostic value of SLC35A3 in CRC. A overall survival (OS) model was constructed and validated based on the expression level of SLC35A3 and multivariable analysis results. The cBioPortal tool was used to analyze SLC35A3 mutation in CRC. The UALCAN tool was used to analyze the promoter methylation level of SLC35A3 in colorectal cancer. In addition, the role of SLC35A3 in CRC was determined through GO analysis, KEGG analysis, gene set enrichment analysis (GSEA), immune infiltration analysis, and immune checkpoint correlation analysis. In vitro experiments validated the function of SLC35A3 in colorectal cancer cells. Compared with adjacent normal tissues and colonic epithelial cells, the expression of SLC35A3 was decreased in CRC tissues and CRC cell lines. Low expression of SLC35A3 was associated with N stage, pathological stage, and lymphatic infiltration, and it was unfavorable for OS, disease-specific survival (DSS), recurrence-free survival (RFS), and post-progression survival (PPS). According to the Receiver Operating Characteristic (ROC) analysis, SLC35A3 is a potential important diagnostic biomarker for CRC patients. The nomograph based on the expression level of SLC35A3 showed a better predictive model for OS than single prognostic factors and TNM staging. SLC35A3 has multiple types of mutations in CRC, and its promoter methylation level is significantly decreased. GO and KEGG analysis indicated that SLC35A3 may be involved in transmembrane transport protein activity, cell communication, and interaction with neurotransmitter receptors. GSEA revealed that SLC35A3 may be involved in energy metabolism, DNA repair, and cancer pathways. In addition, SLC35A3 was closely related to immune cell infiltration and immune checkpoint expression. Immunohistochemistry confirmed the positive correlation between SLC35A3 and helper T cell infiltration. In vitro experiments showed that overexpression of SLC35A3 inhibited the proliferation and invasion capability of colorectal cancer cells and promoted apoptosis. The results of this study indicate that decreased expression of SLC35A3 is closely associated with poor prognosis and immune cell infiltration in colorectal cancer, and it can serve as a promising independent prognostic biomarker and potential therapeutic target.

China became the world leader in crude oil processing capacity in 2021. However, petroleum refining generates significant volatile organic compound (VOC) emissions, and the composite source profile, source-specific emission factors, and emission inventories of VOCs in the petroleum refining industry remain poorly understood. In this study, we focused on Guangdong, China's major province for crude oil processing, and systematically evaluated the historical emissions and reduction of VOCs in the petroleum refining industry from 2001 to 2020. We accomplished this by establishing local source-specific emission factors and composite source profiles. Finally, we quantitatively assessed the potential impact of these emissions on ozone and secondary organic aerosol formation. Our results revealed that VOC emissions from the petroleum refining industry in Guangdong followed an increasing-then-decreasing trend from 2001 to 2020, peaking at 37.3 Gg in 2016 and declining to 18.7 Gg in 2020. Storage tanks and wastewater collection and treatment remained the two largest sources, accounting for 41.9 %-53.4 % and 20.6 %-27.5 % of total emissions, respectively. Initially, Guangzhou and Maoming made the most significant contributions, with Huizhou becoming a notable contributor after 2008. Emission reduction efforts for VOCs in Guangdong's petroleum refining industry began showing results in 2017, with an average annual VOC emission reduction of 21.5 Gg from 2017 to 2020 compared to the unabated scenario. Storage tanks, wastewater collection and treatment, and loading operations were the primary sources of emission reduction, with significant contributions from Maoming, Huizhou, and Guangzhou. Alkanes made the largest contribution to VOC emissions, while alkenes/alkynes and aromatics comprised the most significant portions of ozone formation potential (OFP) and secondary organic aerosol formation potential (SOAP). We also estimated VOC emissions and reduction from petroleum refining for China from 2001 to 2020, and measures such as "one enterprise, one policy" and deep control strategies could reduce emissions by at least 103.9 Gg.

BACKGROUND: Heat-clearing and detoxifying drugs has protective effect on colorectal cancer (CRC). Given the complicated features of Traditional Chinese medicine formulas, network pharmacology is an effective approach for studying the multiple interactions between drugs and diseases. AIM: To systematically explore the anticancer mechanism of heat-clearing and detoxifying drug JC724. METHODS: This study obtained the active compounds and their targets in JC724 from Traditional Chinese Medicine System Pharmacology Database. In addition, the CRC targets were obtained from Drugbank, TTD, DisGeNET and GeneCards databases. We performed transcriptome analysis of differentially expressed genes in CRC treated with JC724. Venn diagram was used to screen the JC724-CRC intersection targets as candidate targets. Core targets were selected by protein-protein interaction network and herb ingredient-target-disease network analysis. The functional and pathway of core targets were analysed by enrichment analysis. RESULTS: We found 174 active ingredients and 283 compound targets from JC724. 940 CRC-related targets were reserved from the four databases and 304 CRC differentially expressed genes were obtained by transcriptome analysis. We constructed the network and found that the five core ingredients were quercetin, ß Beta sitosterol, wogonin, kaempferol and baicalein. The core JC724-CRC targets were CYP1A1, HMOX1, CXCL8, NQO1 and FOSL1. JC724 acts on multiple signaling pathways associated with CRC, including the Nrf2 signaling pathway, oxidative stress, and the IL-17 signaling pathway. CONCLUSION: In this study, we systematically analyzed the active ingredients, core targets and main mechanisms of JC724 in the treatment of CRC. This study could bring a new perspective to the heat-clearing and detoxifying therapy of CRC.

Printing industry is one of the most important sources of industrial volatile organic compound (VOC) emissions in China, and is thus a key industrial sector in terms of VOC control. However, process-based VOC emission and speciation from the printing industry have not been well identified, mainly owing to the poor emission factors (EFs) and diversity of source profiles. In this study, we systematically characterized process-based VOC emissions from the printing industry for the period of 2010-2019, through the establishment of improved emission factors and composite source profiles. VOC emissions from the printing industry were found to continuously increase from 2010 to 2018, reaching their maximum in 2018 at 939.8 Gg, but started to decrease afterwards. The substantial growth is driven by the large demand for ink and adhesive and the absence of effective control measures in the printing industry. The total VOC emissions and ozone formation potential (OFP) in China in 2019 were 916.1 Gg and 1834.5 Gg, respectively. Gravure printing and the compound process were the processes that contributed the most to both emissions and OFP. Rapidly developing provinces such as Guangdong, Jiangsu, and Zhejiang were the largest contributors to emissions. Oxygenated VOCs (OVOCs) accounted for most of the VOC emissions, followed by alkanes and aromatics, while aromatics were the dominant groups for total OFP, followed by alkenes/alkynes and OVOCs. Ethyl acetate, toluene, isopropanol, isopentane, and n-pentane were the top five VOC species in terms of emissions, while toluene, ethyl acetate, 1,3-butadiene, isopentane, and 1-butene were the top five species in terms of OFP. Scientific and precise control policy were proposed, involving four aspects: environmental access, emission standards, classification and management, and research on source substitution. We believe our study will provide an important reference for the systematic characterization and control policy of VOC emissions from other industries.

The printing industry has always been the key source of volatile organic compound(VOC) emissions in China. However, owing to the complexity of raw materials and processes, the fine emission inventory and its future emission reduction potential of VOCs from the printing industry have not been well characterized. In this study, the existing VOCs emission factors of the printing industry were improved, considering the neglected semi/intermediate VOCs(S/IVOCs). An emissions inventory of VOCs from the printing industry in the period of 2011-2020 in China was compiled. Through scenario analysis, the emission of VOCs under different scenarios in 2030 was predicted, and the emission reduction potential was analyzed. VOCs emissions from the printing industry in China increased first and then decreased in the period of 2011-2020. Compared with that in 2011, VOCs emissions increased by 29.6% in 2020, with an average annual growth rate of 3.0%. This was mainly due to the increasing consumption demand in the printing industry market and the lack of effective measures for integrated management of VOCs. The VOCs emission of the printing industry in China in 2020 was 861 Gg. Gravure printing and packaging processing were the two most important processes, accounting for 52.0% and 28.7%, respectively. Guangdong, Jiangsu, and Zhejiang were the largest contributors to VOC emissions, accounting for 44.12% of the total emissions. VOCs emissions of the printing industry in 2030 were 1187 Gg, 684 Gg, and 362 Gg for the baseline scenario, the general control scenario, and the strict control scenario, respectively. Compared to that in 2020, emissions under different control scenarios in 2030 increased by 37.9% and decreased by 20.6% and 57.9%, respectively. Gravure printing and packaging processing are still the focus of emission reduction.

Breast cancer has the highest incidence rate among all cancer types worldwide, seriously threatening women's health. The present retrospective study explored differences in serum lipid contents in different breast cancer (BC) subcategories and their correlation with Ki-67 expression levels in patients with invasive BC with the aim of identifying novel diagnostic and prognostic indicators for personalized BC treatment. The study included 170 patients diagnosed with BC who were diagnosed with invasive BC by postoperative pathological examination. Data on patient age, body mass index and menopausal status were collected, in addition to estrogen receptor, progesterone receptor, human epidermal growth factor receptor 2 (HER2) and antigen Ki-67 expression levels and pathological tumor type. Preoperative circulating lipid levels, specifically the levels of total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), triglycerides (TG) and apolipoproteins A1 (ApoA1) and B (ApoB) were also obtained. Molecular subcategories of BC were grouped based on their immunohistochemistry. Differences in serum lipid levels between the groups were assessed, and correlations between serum lipid and Ki-67 expression levels were explored. While TC, LDL-C, HDL-C and ApoA1 levels differed significantly among molecular subcategories. TG and ApoB levels did not. Circulating TC and LDL-C levels were considerably higher in patients with triple-negative BC (TNBC) and HER2-positive [hormone receptor (HR)-negative] BC than in those with luminal A and B (HER2-negative) BC. Serum HDL-C levels were significantly diminished in the TNBC and HER2-positive (HR-negative) groups compared with the luminal A and B (HER2-negative) groups. ApoA1 levels were significantly reduced in cases of TNBC and HER2-positive (HR-negative) BC compared with luminal A and B BC. Ki-67 expression levels were positively correlated with circulating TC and LDL-C levels and inversely correlated with circulating HDL-C and ApoA1 levels but exhibited no correlation with serum ApoB and TG levels. The results indicate that elevated TC and LDL-C levels and diminished HDL-C and ApoA1 levels were high-risk factors in patients with TNBC and HER2-positive (HR-negative) BC, but not patients with luminal subcategories of BC. Abnormal serum lipid levels were correlated with Ki-67 expression levels, with elevated circulating TC and LDL-C levels and reduced circulating HDL-C and ApoA1 levels indicating a poor prognosis in patients with BC.

Objective: Bazhen Decoction (BZD) is a common adjuvant therapy drug for colorectal cancer (CRC), although its anti-tumor mechanism is unknown. This study aims to explore the core components, key targets, and potential mechanisms of BZD treatment for CRC. Methods: The Traditional Chinese Medicine Systems Pharmacology (TCMSP) was employed to acquire the BZD's active ingredient and targets. Meanwhile, the Drugbank, Therapeutic Target Database (TTD), DisGeNET, and GeneCards databases were used to retrieve pertinent targets for CRC. The Venn plot was used to obtain intersection targets. Cytoscape software was used to construct an "herb-ingredient-target" network and identify core targets. GO and KEGG pathway enrichment analyses were conducted using R language software. Molecular docking of key ingredients and core targets of drugs was accomplished using PyMol and Autodock Vina software. Cell and animal research confirmed Bazhen Decoction efficacy and mechanism in treating colorectal cancer. Results: BZD comprises 173 effective active ingredients. Using four databases, 761 targets related to CRC were identified. The intersection of BZD and CRC yielded 98 targets, which were utilized to construct the "herb-ingredient-target" network. The four key effector components with the most targets were quercetin, kaempferol, licochalcone A, and naringenin. Protein-protein interaction (PPI) analysis revealed that the core targets of BZD in treating CRC were AKT1, MYC, CASP3, ESR1, EGFR, HIF-1A, VEGFR, JUN, INS, and STAT3. The findings from molecular docking suggest that the core ingredient exhibits favorable binding potential with the core target. Furthermore, the GO and KEGG enrichment analysis demonstrates that BZD can modulate multiple signaling pathways related to CRC, like the T cell receptor, PI3K-Akt, apoptosis, P53, and VEGF signaling pathway. In vitro, studies have shown that BZD dose-dependently inhibits colon cancer cell growth and invasion and promotes apoptosis. Animal experiments have shown that BZD treatment can reverse abnormal expression of PI3K, AKT, MYC, EGFR, HIF-1A, VEGFR, JUN, STAT3, CASP3, and TP53 genes. BZD also increases the ratio of CD4+ T cells to CD8+ T cells in the spleen and tumor tissues, boosting IFN-γ expression, essential for anti-tumor immunity. Furthermore, BZD has the potential to downregulate the PD-1 expression on T cell surfaces, indicating its ability to effectively restore T cell function by inhibiting immune checkpoints. The results of HE staining suggest that BZD exhibits favorable safety profiles. Conclusion: BZD treats CRC through multiple components, targets, and metabolic pathways. BZD can reverse the abnormal expression of genes such as PI3K, AKT, MYC, EGFR, HIF-1A, VEGFR, JUN, STAT3, CASP3, and TP53, and suppresses the progression of colorectal cancer by regulating signaling pathways such as PI3K-AKT, P53, and VEGF. Furthermore, BZD can increase the number of T cells and promote T cell activation in tumor-bearing mice, enhancing the immune function against colorectal cancer. Among them, quercetin, kaempferol, licochalcone A, naringenin, and formaronetin are more highly predictive components related to the T cell activation in colorectal cancer mice. This study is of great significance for the development of novel anti-cancer drugs. It highlights the importance of network pharmacology-based approaches in studying complex traditional Chinese medicine formulations.

PURPOSE: To study the expression of survivin and VEGF-C in breast cancer tissue and its relation to lymphatic metastasis. METHODS: The expression level of survivin and VEGF-C in breast cancer tissue is determined by immunohistochemistry and reverse transcriptase polymerase chain reaction (Rt-PCR). RESULTS: Among the 60 breast cancer tissues, the percentage of positive expression of survivin or VEGF-C was 85.0% and 78.3%, respectively. The expression rate of VEGF-C was 84.3% (43/51), 44.4% (4/9) in the survivin positive and negative expression group, respectively. Linear correlation analysis showed a correlation coefficient of survivin and VEGF-C in breast cancer was 0.80 (p = 0.035), which indicates a positive correlation between the two biomarkers. CONCLUSIONS: Survivin and VEGF-C are highly expressed in breast cancer tissues with a positive correlation. The regulation of survivin expression can cause excessive expression of VEGF-C, leading to the generation of breast cancer lymphangiogenesis, thus causing lymphatic metastasis of breast cancer.

Severe pneumonia in children is a group of inflammatory diseases of respiratory tract caused by pathogenic microorganisms. Increasing evidence suggested the crucial effects of microRNA on inflammatory diseases. This study aimed to reveal the expression and role of miR-483-3p in the serum of children with severe pneumonia, and to explore the effect of miR-483-3p on the biological function of lipopolysaccharide (LPS)-induced MRC-5 cells. MRC-5 cells were disposed with LPS to construct an in vitro pneumonia cell model. The relative expression level of miR-483-3p was measured by qRT-PCR. ROC curve was used to evaluate the diagnostic value of miR-483-3p in severe pneumonia. The Kaplan-Meier curve was performed to test the characteristics of survival distribution of different miRNA classifications. Cell viability and apoptosis were performed by CCK-8 assay and flow cytometry. IL-1ß, TNF-α, and IL-6 were detected by ELISA. Luciferase reporter gene assay and western blot analysis were performed to detect the interaction between miR-483-3p and IGF-1. The expression of serum miR-483-3p in severe pneumonia patients was higher than in controls. The AUC value of the ROC curve was 0.919, indicating that miR-483-3p had diagnostic value for severe pneumonia. The survival curve showed that patients with high expression of miR-483-3p had higher mortality. Cell viability and apoptosis assay showed that overexpression of miR-483-3p suppressed cell proliferation and promoted apoptosis. And upregulation of miR-483-3p promoted generation of inflammatory cytokines. Luciferase report gene assay and western blot assay both illustrated that IGF-1 might be the target gene of miR-483-3p. Serum miR-483-3p can be used as a biomarker for the diagnosis of severe pneumonia. High expression of miR-483-3p promoted the development of severe pneumonia.

OBJECTIVE: Investigative studies report contradictory results of the relationship between serum lipid levels and the risk of colorectal cancer (CRC). We conducted a meta-analysis of prospective published studies to clarify the relationship between serum lipid and CRC risk. DESIGN: Systematic review and meta-analysis. DATA SOURCES: PubMed and Embase from inception until December 2020. ELIGIBILITY CRITERIA: We considered prospective cohort and case-control studies that evaluated differences in serum lipid levels with the risk of developing CRC. DATA EXTRACTION AND SYNTHESIS: Two independent reviewers screened and included the studies using standardised electronic data extraction forms. The relative risks of the studies were combined with random-effect and fixed-effect models and were analysed for heterogeneity, publication bias and sensitivity. RESULTS: Twenty-four prospective studies, including 4 224 317 individuals with 29 499 CRC cases, were included in the meta-analysis. The total pooled risk ratio (RR) for high vs low concentrations of triglyceride (TG) concentrations was reported at 1.21 (95% CI 1.09 to 1.34; I2=46.8%), total cholesterol (TC) was at 1.15 (95% CI 1.08 to 1.22; I2=36.8%), high-density lipoprotein cholesterol (HDL-C) was 0.86 (95% CI 0.77 to 0.97; I2=28.8%) and low-density lipoprotein cholesterol (LDL-C) was observed at 1.03 (95% CI 0.75 to 1.41; I2=69.4%). CONCLUSIONS: This meta-analysis shows that high levels of serum TG and TC are positively correlated with the incidence rate of CRC, while high levels of serum HDL-C are negatively correlated with CRC incidence rate. Furthermore, no association was found between LDL-C and the risk of developing CRC. Nevertheless, the heterogeneity brought about by comparative methods, demographic differences and pathological differences between the research subjects limits the effectiveness of the overall pooled results.

CONTEXT: Western diet and unhealthy lifestyle have contributed to the continued growth of type 2 diabetes mellitus (T2DM). T2DM is associated with dysbacteriosis, and studies have found that altering the gut microbiota has a positive effect on treatment. OBJECTIVE: In addition to hyperglycaemia, T2DM often causes damage to multiple organs. However, there are few studies on organ damage from faecal microbiota transplantation (FMT). MATERIALS AND METHODS: T2DM mice were divided into four groups and were given phosphate buffered saline (PBS) (T2DM group), FMT (FMT group), Lactobacillus (LAB group), and Bifidobacterium (BIO group) by gavage for six weeks, respectively. Mice on a normal diet (control group) were gavaged with PBS for six weeks. RESULTS: After gavage treatment, FMT, LAB, and BIO groups were similar in lowering glucose, endotoxemia was slightly reduced, and the colonic mucus layer and liver lobules developed towards normal tissue. Surprisingly, we found that the FMT group had unique effects on islet cell regeneration, increased functional ß cells, and insulin sensitivity. DISCUSSION AND CONCLUSION: Lactobacillus has the best glucose-lowering effect, but FMT has obvious advantages in ß-cell regeneration, which provides new treatment ideas for tissue damage caused by T2DM.
