RESUMO

The purpose of the study was to measure the cement thickness obtained when ceramic rods were luted to dentin and to analyze the relation between cement thickness and the previously published tensile bond strength of similar test specimens. In addition, the ISO standard 4049:2019 method was used to determine the film thickness of the used cements. Zirconia (n = 100) and lithium disilicate (n = 50) rods were cemented to bovine dentin using one of five different resin-based cements. The ceramic-dentin test specimens were cut into two slices and the cement thickness was measured using a scanning electron microscope and compared to the bond strength values of similar specimens already published. The mean cement thickness recorded for ceramic rods cemented to dentin was in the range 20-40 µm, which was larger than the cement film thickness found by the ISO method. The cement film thickness determined according to ISO standard methods did not concur with the results obtained when cementing ceramic rods to dentin. For cementing ceramic restorations, a cement thickness in the range 25-35 µm seems to be favorable for the bond strength.

OBJECTIVE: Despite new strategies in tissue engineering, cartilage repair remains a major challenge. Our aim is to treat patients with focal lesions of articular cartilage with autologous hyaline cartilage implants using a scaffold-free approach. In this article, we describe experiments to optimize production of scaffold-free cartilage discs. DESIGN: Articular chondrocytes were expanded in vitro, seeded in transwell inserts and redifferentiated using established chondrogenic components. Experimental variables included testing 2 different expansion media, adding bone morphogenetic protein 2 (BMP2), insulin-like growth factor 1 (IGF1), growth/differentiation factor 5 (GDF5), or fibroblast growth factor 18 (FGF18) to the differentiation medium and allowing the disc to float freely in large wells. Cartilage discs were analyzed by weight and thickness, real-time RT-qPCR (reverse transcriptase qualitative polymerase chain reaction), fluorescence immunostaining, transmission electron microscopy, second harmonic generation imaging, and measurement of Young's modulus. RESULTS: Addition of BMP2 to the chondrogenic differentiation medium (CDM) was essential for stable disc formation, while IGF1, GDF5, and FGF18 were redundant. Allowing discs to float freely in CDM on a moving platform increased disc thickness compared with discs kept continuously in transwell inserts. Discs cultured for 6 weeks reached a thickness of almost 2 mm and Young's modulus of >200 kPa. There was abundant type II collagen. Collagen fibrils were 25 nm thick, with a tendency to be organized perpendicular to the disc surface. CONCLUSION: Scaffold-free engineering using BMP2 and providing free movement in CDM produced firm, elastic cartilage discs with abundant type II collagen. This approach may potentially be used in clinical trials.

OBJECTIVES: Fracture strength measured in vitro indicates that most all-ceramic crowns should be able to withstand mastication forces. Nevertheless, fractures are one of the major clinical problems with all-ceramic restorations. Furthermore, the fracture mode of all-ceramic crowns observed in clinical use differs from that found in conventional fracture strength tests. The aim of the present study was to develop and investigate a method that simulates clinical fracture behavior in vitro. METHODS: 30 crowns with alumina cores were made to fit a cylindrical model with a molar-like preparation design. These crowns were randomly allocated to 3 tests groups (n=10). The crowns in group 1 were cemented to abutment models of epoxy and subsequently fractured by occlusal loading without contact damage. The crowns in group 2 were fractured by cementation with expanding cement. The crowns in group 3 were cemented on an abutment model of epoxy split almost in two and fractured by increasing the diameter of the model in the bucco-lingual direction. The fractured crowns were analyzed by fractographic methods and compared to a reference group of 10 crowns fractured in clinical use. RESULTS: The fracture modes of all the in vitro crowns were similar to clinical fracture modes. The fracture modes in group 1 were most closely matched to the clinical fractures. These crowns also fractured at clinically relevant loads. CONCLUSION: Laboratory tests that induce a distortion of the abutment model during occlusal loading without occlusal contact damage can simulate clinical fractures of all-ceramic crowns.

Debonding of cemented bone implants is regarded as a major contributor to complications. The relationship between shear bond strength and surface roughness has been investigated, however there are inconsistencies in the trends reported in different studies. The shear strength between poly(methyl methacrylate) bone-cement and sand blasted cobalt-chromium and titanium alloy surfaces was measured to investigate the relationship between interfacial shear strength and surface topology. Surface roughness was quantified by a power law relationship fitted to Fourier spectra as well as three traditional parameters (arithmetical average roughness (Ra), volume of interdigitation (Rr), and RMS slope (Rdq)). We found that the interfacial shear strength is directly proportional to the exponent of the surfaces power spectra (P2) and Rdq, but not to Ra and Rr. However, Rdq is shown to be critically dependent on sampling frequency, making it sensitive to measurement settings. P2 was found to be a robust measure of the surface roughness being independent of sampling frequency.

The expansion of the European Union is bringing new types of metal-ceramic alloys to the market, i.e. alloys probably unknown in Western-European dentistry. The aim of this study was to investigate recent developments and "classic" alloy compositions (one iron and two cobalt alloys, unalloyed titanium and an experimental titanium-zirconium alloy, and one gold alloy containing copper and zinc). The alloys and titanium were subject to static immersion in a 0.1 mol l(-1) solution of saline lactic acid before and after oxidation, hence simulating the temperature cycles for the application of ceramic to metal. The greatest amounts of released metal ions were found in the electrolytes of the oxidized gold alloy and of a cobalt alloy not exposed to high-temperature oxidation. Corrosion of the titanium and alloy surfaces was related to the condition of the specimen. The irritation potentials of some metal ions found in the electrolyte were investigated by performing the hen's egg test-chorio-allantoic membrane (HET-CAM) procedure with 1 mmol l(-1) solutions of Ce(3+), Co(2+), Cu(2+), Zn(2+), Fe(2+), and Ti(4+) ions. The irritation potential of the electrolyte of the oxidized gold alloy with a high concentration of metal elements was also investigated. Of these solutions, only the 1 mmol l(-1) Cu(2+) solution was graded as slightly irritating.
