RESUMO

Natural astaxanthin is a high-value ketone carotenoid mainly derived from Haematococcus pluvialis, which is an excellent antioxidant for human consumption. To study the role of lipids in accumulation of astaxanthin, the H. pluvialis-derived astaxanthin synthesis pathway genes (ß-carotene ketolase gene, BKT and ß-carotene hydroxylase gene, BCH) and fatty acid elongation gene (mitochondrial trans-2-enoyl-coa reductase gene, MECR) were heterologously co-expressed in C. reinhardtii. Zeaxanthin, the precursor of astaxanthin synthesis, was significantly increased after BKT and BCH were expressed. In contrast, the α-carotene that competes with astaxanthin synthesis for lycopene decreased significantly. This redistribution of carbon flow was conducive to the synthesis of astaxanthin. In addition, the transformant only expressed astaxanthin metabolism related genes (BKT, BCH) would lead to an increase in total lipid, a decrease in monounsaturated fatty acids and an increase in polyunsaturated fatty acids. On this basis, the expression of MECR gene further increased the total lipid, and the relative content of different fatty acids also changed. The astaxanthin content of algal strains transformed with BKT and BCH genes was nearly 50% higher than that of the wild type. On this basis, the astaxanthin content of transformants expressing MECR gene related to long-chain fatty acid synthesis was increased by 227.5%. In this study, an astaxanthin production model similar to H. pluvialis by combining carotenoid metabolism and fatty acid metabolism was constructed in C. reinhardtii. The results suggest that the increase in astaxanthin is indeed linked to the regulation of fatty acid metabolism, and this link may involve the type of fatty acids and the dynamics of astaxanthin ester in cells. The strategy of promoting the synthesis of fatty acids has potential to achieve efficient production of astaxanthin in C. reinhardtii.

The two title crystalline compounds, viz. meso-bis{η(5)-1-[1-(dimethylamino)ethenyl]-3-(trimethylsilyl)cyclopentadienyl}iron(II), [Fe(C(12)H(20)NSi)(2)], (II), and meso-bis{η(5)-1-[1-(dimethylamino)ethenyl]-3-(trimethylsilyl)cyclopentadienyl}cobalt(II), [Co(C(12)H(20)NSi)(2)], (III), were obtained by the reaction of lithium 1-[1-(dimethylamino)ethenyl]-3-(trimethylsilyl)cyclopentadienide with FeCl(2) and CoCl(2), respectively. For (II), the trimethylsilyl- and dimethylaminoethenyl-substituted cyclopentadienyl (Cp) rings present a nearly eclipsed conformation, and the two pairs of trimethylsilyl and dimethylaminoethenyl substituents on the Cp rings are arranged in an interlocked fashion. In the case of (III), the same substituted Cp rings are perfectly staggered leading to a crystallographically centrosymmetric molecular structure, and the two trimethylsilyl and two dimethylaminoethenyl substituents are oriented in opposite directions, respectively, with the trimethylsilyl group of one Cp ring and the dimethylaminoethenyl group of the other Cp ring arranged more closely than in (II).

Two new 2D Zn(ii) metal-organic frameworks (MOFs) {[Zn2(L)2(µ2-O)(H2O)3]·3DMA}n (Zn-1) and [Zn2(L)2(2,2'-bipy)(µ2-O)(H2O)2]n (Zn-2) (H2L = 2,5-thiophene dicarboxylic acid, bipy = 2,2'-dipyridyl) were designed and synthesized. Through the strategy of introducing bipy as a "lock", Zn-1 was transferred to Zn-2via a crystal-to-crystal process, which showed significantly enhanced solvent and air stability compared to that of Zn-1. Without post modification, Zn-2 was used as an electrode modified material to construct sensors for the electrochemical recognition and distinction of S-containing amino acids, including l-cysteine, l-methionine and l-cystine.

The reactions of 2-pyridinecarboxaldehyde-phenylhydrazonatolithium C5H4Npy-CH[double bond, length as m-dash]Ni-NaLi(Ph) (abbreviated as Li) with a 1/2 equivalent of anhydrous metal (Zn, Sn, Fe and Co) chlorides or NiCl2(DME) (DME = 1, 2-dimethoxyethane) produced the corresponding mononuclear metal(ii) complexes ( and ), in which each ligand acts as a bidentate ligand and the coordination geometries around the metals are shown to be tetrahedral within the complexes , , and , respectively, and a tetragonal pyramid in the complex . The reaction of Li successively with sodium tert-butoxide and anhydrous ZrCl4 afforded the unanticipated bizirconium complex , in which each monoanionic ligand behaves as a tridentate bridge. Whereas treatment of Li with NiCl2 and then CH2Cl2 led to an interesting methylene-bridged bis(2-pyridyl-phenylhydrazone) compound in moderate yield; a comparative experiment showed that when the Li reacted with CH2Cl2, the coupling compound was also obtained but in very low yield. A plausible mechanism of compound formation was also proposed and supported by the density functional theory (DFT) calculations. All the synthesized compounds were characterized by single-crystal X-ray diffraction.

Deprotonation of the aminofulvene 1 with LiCH(SiMe3)2 provides 2, which reacts with MCl4 (M=Zr or Hf) by intramolecular NMe2H elimination and coupling of the fulvene moieties to form complex 3 or 4. This reaction is a useful synthetic pathway to allyl-bridged ansa-metallocenes, especially those with a conjugated 14 π electron system. R'=dimethylamino.

Decomposition of soil organic matter plays an important role in the regulation of carbon (C) cycles at ecosystem or regional scales, and is closely related to temperature, moisture, and land-use types. The influences of soil temperature, moisture, and land-use types on soil C mineralization in Citrus reticulata and Pinus elliottii forests were investigated at the Qianyanzhou Ecological Experiment Station, Chinese Academy of Sciences, by conducting incubation experiments at 5-level temperatures (5, 10, 15, 20 and 25 degrees C) and 3-level moistures (30%, 60% and 90% saturated soil moisture, SSM). The results showed that soil temperature, moisture, and land-use types had significant effects on soil C mineralization and they had significant interaction effects. Soil C mineralization was positively correlated with incubation temperature in the two forests, and the maximum of soil C mineralization was in the 60% SSM treatment. The accumulation of soil C mineralization was higher in the C. reticulata forest than in the P. elliottii forest under the same temperature and moisture conditions. The temperature sensitivity (Q10) of soil C mineralization was influenced by land-use type and soil moisture. Q10 increased with the increasing soil moisture in both C. reticulata and P. elliottii forests at incubation 7 and 42 d. Q10 in the C. reticulata forest was higher than in the P. elliottii forest in the same moisture level, and the deviation increased with the increasing soil moisture. The model including temperature and moisture could depict the response of soil C mineralization to temperature and moisture. Temperature and moisture together explained 79.9% -91.9% of the variation in soil C mineralization.

Lithium N-[(N,N-dimethylamino)dimethylsilyl]-2-pyridylamidate (2) was prepared from the reaction of new compound N-[(N,N-dimethylamino)dimethylsilyl]-2-pyridylamine (1) and LiBu(n). Treatment of the lithium salt (2) with an equal equivalent of MgBr(2)(THF)(2), FeCl(2) and CoCl(2) afforded the corresponding dinuclear complexes , , and , in which metal atoms possess similar trigonal bipyramidal geometries and each ligand functions as a bimetallic bridging binding aminopyridinato moiety with N-donation from the dimethylamino group. While the stoichiometric reaction of with ZrCl(4) gave the mononuclear zirconium complex (6); the seven coordinated zirconium atom adopts a distorted pentagonal bipyramid geometry and the ligand acts as monoanionic η(2)-aminopyridinato moiety with the pendant arm coordinated via N(CH(3))(2). The reaction of with one equivalent of TiCl(4)(THF)(2) produced the interesting dinuclear titanium complex (7) owing to the elimination of a (N,N-dimethylamino)dimethylsilyl group from the original ligand, and the two titanium centers present different coordination geometries. The molecular structures of the crystalline metal complexes have been confirmed by X-ray single crystal diffraction analysis. Upon activation with methylaluminoxane (MAO), both complexes and exhibited moderate catalytic activities toward ethylene polymerization and produced high molecular weight polyethylenes with broad molecular weight distributions.

The tetracyclic dilithio-Si,Si'-oxo-bridged bis(N,N'-methylsilyl-beta-diketiminates) 2 and 3, having an outer LiNCCCNLiNCCCN macrocycle, were prepared from [Li{CH(SiMe(3))SiMe(OMe)(2)}](infinity) and 2 PhCN. They differ in that the substituent at the beta-C atom of each diketiminato ligand is either SiMe(3) (2) or H (3). Each of and has (i) a central Si-O-Si unit, (ii) an Si(Me) fragment N,N'-intramolecularly bridging each beta-diketiminate, and (iii) an Li(thf)(2) moiety N,N'-intermolecularly bridging the two beta-diketiminates (thf = tetrahydrofuran). Treatment of [Li{CH(SiMe(3))(SiMe(2)OMe)}](8) with 2Me(2)C(CN)(2) yielded the amorphous [Li{Si(Me)(2)((NCR)(2)CH)}](n) [R = C(Me)(2)CN] (4). From [Li{N(SiMe(3))C(Bu(t))C(H)SiMe(3)}](2) (A) and 1,3- or 1,4-C(6)H(4)(CN)(2), with no apparent synergy between the two CN groups, the product was the appropriate (mu-C(6)H(4))-bis(lithium beta-diketiminate) 6 or 7. Reaction of [Li{N(SiMe(3))C(Ph)=C(H)SiMe(3)}(tmeda)] and 1,3-C(6)H(4)(CN)(2) afforded 1,3-C(6)H(4)(X)X' (X =CC(Ph)N(SiMe3)Li(tmeda)N(SiMe3)CH; X' = CN(SiMe3)Li(tmeda)NC(Ph)=C(H)SiMe3)(9). Interaction of A and 2[1,2-C(6)H(4)(CN)(2)] gave the bis(lithio-isoindoline) derivative [C6H4C(=NH)N{Li(OEt2)}C=C(SiMe3)C(Bu(t))=N(SiMe3)]2 (5). The X-ray structures of 2, 3, 5 and 9 are presented, and reaction pathways for each reaction are suggested.

The following crystalline, or microcrystalline (4), metal diamides have been prepared under mild conditions from the N,N'-disubstituted 1,2-diaminobenzene [{N(R)H}2C6H4-1,2] (H(2): R = CH2But; H2L': R = SiMe2NPri2): [Li(thf)(mu-L)(mu-I)Ca(thf)] (1), [Li(thf)4][{Zn(mu-L)}3(mu3-Cl)] (2), [Li(thf)4][Zn(L)2] (3), [{Li(OEt2)(mu-L)Zn}2(mu-L)] (4), [Li(OEt2)(mu-L)Zn(mu-L)Zn(LH)] (5) and [Li(thf)(mu-L')Li(thf)2] (6). Compounds 1-5 were obtained from [Li2(L)] and CaI2 (1) or ZnCl2 (2-5) while 6 was derived from H2(L') and LiBun. Compound 5 was isolated as a very minor by-product from the synthesis of 4, and is assumed to have been formed therefrom by adventitious hydrolysis. The green salt 3 was paramagnetic with the negative charge uniformly delocalised on the two ligands. The other compounds were colourless and diamagnetic. The X-ray structures of each, except 4, are reported and discussed.

The following crystalline, X-ray-characterised heterometallic oligomeric diamides have been prepared in good yield under mild conditions in diethyl ether from the dilithio or disodio derivative of the N,N'-dineopentyl-1,2-diaminobenzene [{N(H)(CH2Bu(t))}2C6H4-1,2] (abbreviated as H2L):[Y(L)(mu-Cl)2Li(OEt2)2]2 (1), [Li(OEt2)2Li(mu2-Cl)4(mu3-Cl)2{Zr(L)}2]2 (2), [Zr(L)2(mu-Cl){Li(OEt2)2}(mu2-Cl)2Zr(L)] (3), [Ce{(mu-L)M}3(OEt2)(1/2)] (3M = Li(1.82)Na(1.18)) (4), [Ce{(mu-L)Na}3(OEt2)] (5) and [Ce{(mu-L)Na}3] (6). Compounds 1-3 were obtained from Li2(L) and YCl3 (the colourless 1) or ZrCl4 (the red 2 and 3), while the red 4 and 5 were isolated from CeCl3 and M2(L) (3M = Li(1.82)Na(1.18)) (4) or Na2(L) (5). Attempted oxidation of 5 with Br2 in hexane yielded the black 6. The ligand is N,N'-chelating to each of the d- or f-block metals in 1-6; and in 4-6 L is also acting as a bridge between Ce and the alkali metal, to which L is thus also chelating.

The yttrium, cerium and magnesium bis(trimethylsilyl)methyls [Ln[CH(SiMe3)2]3][Ln = Y (1), Ce (2)], and the known compound Mg[[CH(SiMe3)2]2 (C) and [Mg(mu-Br)[CH(SiMe3)2](OEt2)]2 (D) formed the crystalline nitrile adducts [1(NCBut)2] (5), [2(NCPh)] (6), [C(NCR)2][R = But (8), Ph (9), C6H3Me2-2,6 (10)] and [Mg(mu-Br)[CH(SiMe3)2](NCR)]2 [R = But (11), Ph (12), C6H3Me2-2,6 (13)], rather than beta-diketiminato-metal insertion products. The beta-diketiminato-cerium complex [Ce[(N(SiMe3)C(C6H4But-4))2CH][N(SiMe3)2]2] (16) was obtained from [Ce[N(SiMe3)2]3] and the beta-diketimine H[[N(SiMe3)C(C6H4But-4)]2CH]]. The cerium alkyl 2 and [Ln[CH(SiMe3)(SiMe2OMe)]3][Ln = Y (3), Ce (4)] were obtained from the appropriate lithium alkyl precursor and [Ce(OC6H2But2-2,6-Me-4)3] or LnCl3, respectively. Heating complex 3 with benzonitrile in toluene afforded 2,2-dimethyl-4,6-diphenyl-5-trimethylsilyl-1,3-diaza-2-silahexa-1,3-diene (7), a member of a new class of heterocycles. The X-ray structures of the crystalline compounds, D, [Mg[CH(SiMe3)2]2(OEt2)2], the known [Ce(Cl)[(N(SiMe3)C(Ph))2CH]2] (E) and 16 are reported. The cerium alkyl (like 1) has one close Ce...C contact for each ligand, attributed to a gamma-C-Ce agostic interaction. The Ln alkyls and have a trigonal prismatic arrangement of the chelating ligands (each of the same chirality at Calpha) around the metal. In an arene solution at 313 K exists as two isomers, as evident from detailed NMR spectroscopic experiments.
