RESUMO

Coronary heart disease (CHD) is the leading cause of global morbidity, but the effect of plasticizers and antimicrobial additives on CHD is unknown. Here, we conducted a case-control study to investigate the mediating role of oxidative stress in the association between co-exposure to seven bisphenols, four parabens, triclosan (TCS), triclocarban, and CHD risk in Guangzhou, China. Quantile-based g-computation and weighted quantile sum regression were used to analyze mixture-outcome associations. Quantile-based g-computation showed a positive joint effect of a decile increase in exposure to all examined pollutants on CHD risk (OR: 1.52, 95% CI: 1.25-1.84), with bisphenol A (BPA), bisphenol F (BPF), n-butyl paraben (BuP), and TCS representing major contributors. The results also showed a decile nonmonotonic increase in the exposure mixtures, positively correlated with a 2.22 ng/mL (95% CI: 1.21-3.23 ng/mL) elevation of 8-hydroxy-2'-deoxyguanosine (8-OHdG), with BuP, TCS, bisphenol AP (BPAP), and BPF contributing dominantly. Mediation analysis showed that 8-OHdG mediated the relationship between BPA, BPF, BPAP, and TCS, and CHD risk. Moreover, the mediating role of high-density lipoprotein (HDL) between several bisphenols and CHD was also identified. It is yet to be verified, but bisphenols may elevate CHD risk by reducing HDL status and increasing oxidative stress.

In this study of extracts of 100 food contact material (FCM) samples collected from South China, we identified 21 organophosphate esters (OPEs) by suspect screening and seven novel OPEs by characteristic fragments-based nontarget screening. Six organophosphite antioxidants (OPAs) were further identified using a suspect list derived from these identified OPEs. Of these compounds, 18 OPEs and 3 OPAs were found for the first time in the extracts of FCMs. (Semi-)quantification revealed that seven of the OPEs [triphenyl phosphate, tris(2,4-di-tert-butylphenyl) phosphate (TDtBPP), bis(2,4-di-tert-butylphenyl) methyl phosphate, (2,4-di-tert-butylphenyl)pentaerythritol phosphate, triethyl phosphate, 2-ethylhexyl-diphenyl phosphate, and trimethyl phosphate] and two of the OPAs [tris(2,4-di-tert-butylphenyl) phosphite (TDtBPPi) and pentabutylated triphenyl phosphite] were present in more than 50 FCM samples and that TDtBPP and TDtBPPi were the dominant OPE and OPA in FCMs, respectively [with median concentrations of 7260 ng/g (range: <8.50-103,879 ng/g) and 31,920 ng/g (range: <9.80-657,399 ng/g), respectively]. A migration test revealed that the migration efficiencies of compounds from a plastic coffee cup to food simulants in the cup increased as the ethanol/water ratio in the food simulants increased. This study significantly enhanced our understanding on the diversity and occurrences of OPEs and OPAs in FCMs used in China and their FCM-to-food migration risk.

Benzophenone (BP)-type UV filters are commonly added to sunscreens and cosmetics to protect against UV radiation for human skin and hair. As a result, BPs are ubiquitous in the environment and human body, and their endocrine-disrupting characteristics have been a hot topic of discussion. However, our knowledge regarding the detrimental effects of prenatal exposure to BPs on pregnant women and their offspring remains limited. To fill this gap, we determined five BP derivatives in 600 serum samples obtained from pregnant women. All the target analytes, except 2,4-dihydroxybenzophenone (BP-1), have achieved a 100 % detection rate. The most prevalent compound was 2-hydroxy-4-methoxybenzophenone (BP-3), with a median concentration of 0.545 ng/mL. Significant and positive correlations were observed among BP derivatives, indicating both endogenous metabolism and common external sources. Utilizing Bayesian kernel machine regression (BKMR) and quantile-based g-computation (QGC) models, we found relationships between BP exposure and reduced neonatal birth weight (BW) and birth chest circumference (BC) during the third trimester. Notably, the adverse effect of BPs on birth size was sex-specific. Moreover, triglyceride (TG) was identified as a potential mediator of the effect of BPs on blood pressure, and co-exposure to BPs was linked to disruptions in thyroid hormone levels and glucose regulation. Further research is warranted to unravel the toxicity of BPs and their detrimental effects on pregnant women and fetuses.

Bisphenol analogs (BPs) are extensively employed in commercial and industrial products and they have been found in a variety of environmental matrices and human samples. The red swamp crayfish (Procambarus clarkii) has been a trendy food in China in recent decades. However, the levels of BPs in Chinese crayfish and the associated hazards of human exposure remain unknown. Thus, in this study, the levels of eight BPs in crayfish gathered from five major provinces engaged in crayfish within the Yangtze River Basin were analyzed. Additionally, the health risks for humans by ingesting crayfish were calculated. BPs were frequently detected in crayfish tissues, indicating the wide occurrence of these chemicals. In comparison to other substitutions, BPA remains the dominant bisphenol analog. Most of the BPs were observed to accumulate in the hepatopancreas compared to the muscle, so consuming the hepatopancreas of crayfish is not recommended. With the exception of BPS, the Estimated Daily Intakes (EDIs) of the remaining BPs exceeded the Tolerable Daily Intake (TDI) specified by the European Food Safety Authority (EFSA) by a factor of 1.75-69.0. The mean hazard index (HI) values exceeded 1 for both hepatopancreas and muscle in all provinces, and the mean HI values for hepatopancreas were significantly higher than those for muscle, indicating potential health risks for local consumers.

Water-in-oil droplets have emerged as promising microreactors for high-throughput biochemical analysis due to their features of reduced sample consumption and automated operation. For a typical screening application, droplets are often trapped for continuous monitoring of the reaction over an extended period, followed by the selective retrieval of targeted droplets based on the after-effect of biochemical reactions. While techniques for droplet trapping are well developed, retrieval of targeted droplets mainly demands complicated device fabrication or sophisticated control. Herein, facile and rapid selective droplet release is achieved by utilizing a new class of photoresponsive fluorosurfactant based on plasmonic nanoparticles. The intense photothermal response provided by this novel photoresponsive fluorosurfactant is capable of vaporizing the fluorocarbon oil at the droplet interface under laser illumination, resulting in a bubble releasing a trapped droplet on demand. A fully automated fluorescence-activated droplet release platform has also been developed to demonstrate its potential for droplet-based large-scale screening applications.

In this study, an innovative, integrated nontarget and suspect screening workflow was developed for identifying organophosphate diesters (di-OPEs) using high-resolution mass spectrometry. The workflow featured the utilization of 0.02% acetic acid as a mobile-phase additive, differentiated screening methods for alkyl and aryl di-OPEs, and a combination of electrospray negative ionization and positive ionization. Using this workflow, 18 di-OPEs were identified in the extracts of 75 food contact plastic (FCP) samples sourced from South China. Among these, six alkyl and one aryl di-OPEs were previously unknown (one unequivocal identification and six probable structures based on diagnostic evidence). (Semi)quantification revealed that bis(2,4-di-tert-butylphenyl) phosphate was the major di-OPE in FCPs, with a median concentration of 1079 ng/g (range: 23.4-158,414 ng/g). The migration efficiencies of di-OPEs from an FCP sample to four kinds of food simulants were between 2.58 and 54.3%. This study offered a useful workflow for the comprehensive profiling of di-OPEs in FCPs.

The ingestion of contaminated tea involves the risk of human exposure to residues of neonicotinoids (NEOs). Nevertheless, there is little empirical research about this topic; to bridge the current knowledge gap, we collected 220 samples of various tea products from four geographical areas in China, including unfermented green tea, semi-fermented white tea and oolong tea, completely fermented black tea, and post-fermented dark tea. A total of six NEOs were detected from the tea leaves and infusions, namely, dinotefuran (DIN), thiamethoxam (THM), clothianidin (CLO), imidacloprid (IMI), acetamiprid (ACE), and thiacloprid (THI). The detection frequencies (DFs) and concentrations of all target NEOs were relatively high across the investigated tea samples, and the DIN, IMI and ACE residues measured in some samples exceeded the maximum residue level (MRL) standards for the European Union. Samples representing the Jiangnan area exhibited greater levels of total target NEOs (∑6NEOs) than samples representing the Jiangbei area (p < 0.001). Moreover, dark tea samples were found to have far higher levels of NEO residues than green (p < 0.001), white (p < 0.05), or oolong (p < 0.001) samples. The health risks associated with exposure to NEO residues via tea were small for both children and adults in terms of acute, chronic, and cumulative dietary exposure risk assessments. The transfer rates (TRs) of NEOs observed in white, black, and dark tea infusions gradually decreased after the third brewing time. As such, it is recommended to only consume tea that has been brewed at least three times. The presented results not only describe the extent of NEO contamination in Chinese tea leaves and infusions, but also provide tea drinking guidelines for consumers.

Parabens, triclosan (TCS), and triclocarban (TCC) are antimicrobial additives that are widely used in personal care products (PCPs) and may dysregulate infant gut microbiota and induce a series of chronic diseases. Dietary intake may be an underestimated exposure route of such antimicrobial additives in infants, but relevant data remain scarce. Therefore, this study determined five common preservatives, including methyl- (MeP), ethyl- (EtP), propyl- (PrP), butyl- (BuP), and benzyl-paraben (BeP), and two antimicrobials TCS and TCC, in major infant food sources (breastmilk, milk-based infant formula [MIF], and cereal-based complementary food [CCF]) in southern China. The health risks associated with dietary exposure among infants across different months of age were also evaluated. The results demonstrated a high incidence of MeP, EtP, PrP, and BeP in processed infant food products, while TCS and TCC were mainly detected in maternal breastmilk. Notably, MeP and EtP were found in all of the MIFs tested, while MeP, EtP, and BeP were detected in 85.6 %-100 % of the CCFs. By incorporating the human equivalent dose and an additional 10-fold margin of safety for infants into the health risk assessment, the 95th percentile hazard quotient of PrP via the ingestion of breastmilk among neonates exceeded 1. For the first time, the results showed that exposure to PrP via breastmilk intake may pose a considerable health risk to urban neonates in southern China. The health risks caused by antimicrobial exposure via ingesting MIF and CCF among infants were negligible. Thus, we recommend breastfeeding women reduce their consumption of PCPs and processed food, especially during the first month after delivery.

Parabens and triclosan (TCS) have been extensively applied in personal care products (PCPs) as preservatives and antibacterial agents. However, their potentiality to disrupt the neurological system has induced increasing concern. The elderly population is at a higher risk of neurodegenerative disorder, although research on its association with PCP exposure remains scarce. Here, we measured the urinary levels of four parabens, TCS, and an oxidative stress marker among 540 participants from the Shenzhen aging-related disorder cohort during 2017-2018. The Mini-Mental State Examination (MMSE) was used to assess the cognitive status of participants. Their demographic, dietary, and behavioral factors were collected via questionnaire survey. Among the four paraben analogs, the median concentration of methyl parabens (MeP) was the highest (Low-risk group: 1.21 ng/mL, High-risk group: 1.64 ng/mL). TCS and 8-hydroxy-2'-deoxyguanosine (8-OHdG) were detected in more than 90% of the samples. Weighted quantile sum regression and quantile-based g-computation showed that the combined effect of all analytes was positively associated with the level of 8-OHdG. BtP, EtP and MeP were identified as the major contributors to the joint effect. After stratification by gender, females exhibited more pronounced changes in urinary 8-OHdG level than males. However, the positive correlation between co-exposure to parabens and TCS and cognitive impairment was not significant (p > 0.05) in both models, which warrants investigation with the larger sample size.

Organophosphate esters (OPEs) are a group of emerging contaminants with widespread environmental occurrence, yet research on their occurrence in foodstuffs is limited. We collected 100 foodstuff samples in South China using a market basket method, and analyzed food extracts for the presence of OPEs and organophosphite antioxidants (OPAs) by suspect and nontarget screening through high-resolution mass spectrometry. Our analysis resulted in the identification of 30 OPEs, comprised of 25 OPEs with a confidence level (CL) of 1 (unequivocal identification using standards) and five OPEs with CL = 2b (probable structure based on diagnostic evidence). Interestingly, 11 of these identified OPEs had not been previously reported in food. No OPA was identified. The occurrence of identified OPEs within the food samples was further investigated. The highest median concentration of OPEs in all food samples was reached by tris(2-chloroisopropyl) phosphate (TCPP) (1.55 ng/g ww, range < 0.74-12.0 ng/g wet weight (ww)). Cereals demonstrated the highest median concentration of the cumulative 30 OPEs. Tris(2-chloroethyl) phosphate (TCEP), TCPP, and triethyl phosphate (TEP) predominantly contributed to OPEs contamination in most food categories. Eight OPEs, namely TEP, tris(2-ethylhexyl) phosphate (TEHP), TCEP, triphenyl phosphate (TPhP), 2-ethylhexyl diphenyl phosphate (EHDPP), bis(2-ethylhexyl) phenyl phosphate (BEHPP), resorcinol bis(diphenyl phosphate) (RDP), and methyl diphenyl phosphate (MDPP) exhibited significantly higher concentrations in the processed group as compared to non-processed group, suggesting that food processing may result in contamination of these OPEs. The median sum of estimated dietary intake (ΣEDI) of all OPEs was determined to be 161 ng/kg body weight/day. Cereals (38.5 %) and vegetables (23.5 %) were the predominant food categories contributing to ΣEDI, and TEP (29.0 %), TCEP (20.2 %), and TCPP (18.3 %) were three major OPEs contributing to ΣEDI. This study for the first time offered a comprehensive overview of OPE species and revealed their occurrence in foodstuffs from South China.

Rises in trace element contents in rice and wheat flour, which are staple foods for almost all the Chinese population, associated with rapid economic development have raised major concerns. This study aimed to assess trace element concentrations in these foods nationwide in China and associated human exposure risks. For these purposes, nine trace elements were measured in 260 rice samples and 181 wheat flour samples with 17 and 12 widely scattered geographical origins in China, respectively. Mean concentrations (mg kg-1) of the trace elements declined in the following orders: Zn > Cu > Ni > Pb > As > Cr > Cd > Se > Co in rice, and Zn > Cu > Ni > Se > Pb > Cr > Cd > As > Co in wheat flour. Significant regional differences in levels of trace elements in both rice and wheat flour were detected (p < 0.05), which may be related to local economic indicators. The hazard index (HI) of trace elements in rice samples from all origins exceeded 1, mainly due to the contribution of As, suggesting a potential non-carcinogenic risk. The total carcinogenic risk (TCR) for rice and wheat flour of all origins exceeded the safe level.

BACKGROUND: There is great interest to engineer in vitro models that allow the study of complex biological processes of the microvasculature with high spatiotemporal resolution. Microfluidic systems are currently used to engineer microvasculature in vitro, which consists of perfusable microvascular networks (MVNs). These are formed through spontaneous vasculogenesis and exhibit the closest resemblance to physiological microvasculature. Unfortunately, under standard culture conditions and in the absence of co-culture with auxiliary cells as well as protease inhibitors, pure MVNs suffer from a short-lived stability. METHODS: Herein, we introduce a strategy for stabilization of MVNs through macromolecular crowding (MMC) based on a previously established mixture of Ficoll macromolecules. The biophysical principle of MMC is based on macromolecules occupying space, thus increasing the effective concentration of other components and thereby accelerating various biological processes, such as extracellular matrix deposition. We thus hypothesized that MMC will promote the accumulation of vascular ECM (basement membrane) components and lead to a stabilization of MVN with improved functionality. RESULTS: MMC promoted the enrichment of cellular junctions and basement membrane components, while reducing cellular contractility. The resulting advantageous balance of adhesive forces over cellular tension resulted in a significant stabilization of MVNs over time, as well as improved vascular barrier function, closely resembling that of in vivo microvasculature. CONCLUSION: Application of MMC to MVNs in microfluidic devices provides a reliable, flexible and versatile approach to stabilize engineered microvessels under simulated physiological conditions.

Neonicotinoid insecticides can selectively interact with the unique nicotinic acetylcholine receptor subtypes in insects and are considered to be low toxic to mammals. However, there is still insufficient knowledge on human exposure to neonicotinoid insecticides, especially for children. This study aimed to investigate urinary concentrations and profiles of neonicotinoid insecticides in South China children and to analyze potential influencing factors. Six neonicotinoid insecticides, including imidacloprid (IMI), thiamethoxam (THM), acetamiprid (ACE), clothianidin (CLO), thiacloprid (THD) and dinotefuran (DIN), exhibited high detection frequencies (>90%) in urine samples collected from 305 children, suggesting broad exposure in South China children. The median concentrations were determined to be 0.13, 0.21, 0.01, 0.19, 0.002 and 1.64 µg/L, respectively. Among the target neonicotinoids, urinary concentrations of CLO and THM exhibited a significant and positive correlation between each other (p < 0.05), suggesting similar sources of these two chemicals.

The potential hazards of polycyclic aromatic hydrocarbons (PAHs) on fetus development have raised widespread concerns in recent years, although current epidemiological findings are conflicting. The aim of this case-control study was to investigate the relationship between PAH concentrations in maternal serum and low birth weight (LBW). A questionnaire survey was conducted among 144 (72 pairs) neonates with normal or LBW and their mothers at four different hospitals in Shenzhen, China. Peripheral blood from primiparas and umbilical cord blood from neonates were collected. Concentrations of PAHs were determined by gas chromatography-mass spectrometry. The detection frequencies of PAHs were relatively higher in the peripheral blood samples, while levels of low molecular weight PAHs were greater than high molecular weight PAHs in both peripheral and umbilical cord blood samples. Phenanthrene was frequently detected in blood samples from the case and control groups. Logistic regression analysis showed that acenaphthene in peripheral blood was positively associated with LBW (p < 0.05). This study found a relationship between exposure to certain PAHs and LBW, although future studies are needed to confirm these results.

The purposes of this study were to measure the concentrations of arsenic speciation in shellfish from South China Sea and evaluate the health risk by local residents through shellfish consumption. The median concentrations (in wet weight) of arsenic speciation in shellfish samples were in the following order: AsB (16.0 mg·kg-1) > DMA (1.30 mg·kg-1) > AsV (0.23 mg·kg-1) > AsC (0.08 mg·kg-1) > AsIII (0.05 mg·kg-1) > MMA (0.01 mg·kg-1). Among shellfish species, Mactra mera and Babylonia areolata were found to accumulate iAs and AsB, respectively. The target hazard quotient values of iAs (THQiAs) in all shellfish samples were lower than 1. However, the carcinogenic risk values of iAs (CRiAs) in the Mactra mera, Mytilus galloprovincialis and Pinctada margaritifera were beyond the acceptable range, implying that continuous exposure to iAs pollution via the consumption of these shellfish would pose a potential cancer risk to local consumers.

Waste incineration is one of the most common emission sources of polycyclic aromatic hydrocarbons (PAHs), causing potential occupational exposure in waste incineration workers. However, relative investigations among waste incineration plant workers are still very limited, particularly in China. Therefore, we collected urine specimens from 77 workers in a waste incineration plant as the exposed group, and 101 residents as the control group in Shenzhen, China. Nine mono-hydroxylated PAH metabolites (OH-PAHs) and 8-hydroxy-2'-deoxyguanosine (8-OHdG) were measured, and their internal relationships were explored. The urinary levels of most OH-PAHs and 8-OHdG in the exposed group exhibited high levels versus another group (p < 0.05). We found negative associations between OH-PAHs and 8-OHdG in the control group (p < 0.05), while most of OH-PAHs were not associated with 8-OHdG in the exposed group, which indicated that the exposure to waste incineration could enlarge the level of individual oxidative stress damage. Nevertheless, PAHs were less likely to trigger obvious health risks in exposed workers through estimation of human intake and exposure risks. This study provides a reference for occupational PAH exposure and strengthen the need of health monitoring among incineration workers.

Perchlorate and chlorate are ubiquitous pollutants in various types of foodstuffs, drinking water and environmental compartments. They have raised great concerns due to potential adverse effects on human thyroid functions. Dietary intake is considered as the predominant pathway for human exposure to perchlorate and chlorate. Nevertheless, data on human exposure to the chemicals above remain limited, particularly for the most vulnerable populations such as infants. In the present study, 62 breast milks, 53 infant formulas, 88 baby supplementary food and 50 tap water samples were collected in South China and the levels of perchlorate and chlorate were measured in these samples. Perchlorate and chlorate were frequently detected in more than 90% of measured samples. In these different types of samples, the median concentrations of perchlorate were 0.65 µg/L, 0.61 µg/kg, 0.56 µg/kg and 1.18 µg/L, respectively, while the median concentrations of chlorate were 1.73 µg/L, 2.48 µg/kg, 2.67 µg/kg and non-detected, respectively. Health risk assessment using hazard quotient suggested that perchlorate and chlorate exposure in the sampled baby food are not expected to increase the risk of an adverse health effect. To our knowledge, this is the first study simultaneously investigating perchlorate and chlorate exposure in Chinese infants via food intake.

The abnormal functionality of mitochondria has been linked to many life-threatening diseases such as cancers, failure of cardiovascular functions, and neurodegenerative disorders. Therefore, in vitro analysis of mitochondria has garnered great interest for understanding the mechanism of mitochondrial dysfunction-related disease development and therapeutics. However, due to the intrinsic heterogeneity of cell membrane stiffness, it remains challenging to standardize the protocols for the extraction of mitochondria and adequate disruption of the cellular membrane while retaining the functionality of mitochondria. We have previously developed a microfluidics-based cell shredder capable of serving the purpose. In this protocol, we describe the step-by-step procedures to empirically identify the threshold shear stress using this microfluidics-based cell shredder for mitochondrial extraction. The optimal shear stress to disrupt human embryonic kidney cell (HEK 293) and mice muscle cell (C2C12) has been characterized at around 16.4 Pa, whereas cell lines with stiffer membrane stiffness, for example, neuroblastoma cells (SH-SY5Y), require 27.4 Pa to effectively lyse the cells. This protocol also provides detailed procedures to determine the quality of extracted mitochondria based on the membrane potential and the integrity of extracted mitochondria. A comparison with the widely employed Dounce homogenizer has shown that the proposed microscale cell shredder can yield at least 40% more functional mitochondria and retain higher integrity regarding extracted mitochondria than the counterparts extracted from Dounce homogenizer, especially for low cell concentrations (5-20 × 104 cells/mL) and small sample volume (<200 µL).

Shellfish can absorb and accumulate contaminants. The consumption of shellfish could expose humans to pollutants and increase related health risk. Perchlorate (ClO4-) is a ubiquitous pollutant and could affect thyroid functions, especially for children and pregnant women. However, knowledge on the contamination of perchlorate in aquatic food such as shellfish remains limited. This study aimed to investigate the abundances of perchlorate in shellfish from South China Sea and to assess human exposure risks. A total of 178 shellfish samples from eight species were collected from offshore aquaculture waters in South China Sea. Perchlorate was detected in 99.4% of them, suggesting widespread pollution in coastal waters. Concentrations of perchlorate ranged from not detected (N.D.) to 71.5 µg kg-1, with a median value of 4.33 µg kg-1. Estimated daily intake (EDI) and hazard quotient (HQ) were used to assess human exposure dose and health risks, respectively. The HQ values were determined to be less than 1, indicating no significant health risks to local residents via shellfish consumption. To our knowledge, this is the first study to investigate perchlorate contamination in South China shellfish and assess potential human risks.

Multicellular spheroids have served as a promising preclinical model for drug efficacy testing and disease modeling. Many microfluidic technologies, including those based on water-oil-water double emulsions, have been introduced for the production of spheroids. However, sustained culture and the in situ characterization of the generated spheroids are currently unavailable for the double emulsion-based spheroid model. This study presents a streamlined workflow, termed the double emulsion-pretreated microwell culture (DEPMiC), incorporating the features of (1) effective initiation of uniform-sized multicellular spheroids by the pretreatment of double emulsions produced by microfluidics without the requirement of biomaterial scaffolds; (2) sustained maintenance and culture of the produced spheroids with facile removal of the oil confinement; and (3) in situ characterization of individual spheroids localized in microwells by a built-in analytical station. Characterized by microscopic observations and Raman spectroscopy, the DEPMiC cultivated spheroids accumulated elevated lipid ordering on the apical membrane, similar to that observed in their Matrigel counterparts. Made possible by the proposed technological advancement, this study subsequently examined the drug responses of these in vitro-generated multicellular spheroids. The developed DEPMiC platform is expected to generate health benefits in personalized cancer treatment by offering a pre-animal tool to dissect heterogeneity from individual tumor spheroids.
