BACKGROUND: Salmonella, an important foodborne pathogen, was estimated to be responsible for 95.1 million cases and 50,771 deaths worldwide. Sixteen serovars were responsible for approximately 80% of Salmonella infections in humans in China, and infections caused by a few uncommon serovars have been reported in recent years, though not with S. Welikade. This study reports the first clinical case caused by S. Welikade in China and places Chinese S. Welikade isolates in the context of global isolates via genomic analysis. For comparison, S. Welikade isolates were also screened in the Chinese Local Surveillance System for Salmonella (CLSSS). The minimum inhibitory concentrations (MICs) of 28 antimicrobial agents were determined using the broth microdilution method. The isolates were sequenced on an Illumina platform to identify antimicrobial resistance genes, virulence genes, and phylogenetic relationships. RESULTS: The S. Welikade isolate (Sal097) was isolated from a two-year-old boy with acute gastroenteritis in 2021. Along with the other two isolates found in CLSSS, the three Chinese isolates were susceptible to all the examined antimicrobial agents, and their sequence types (STs) were ST5123 (n = 2) and ST3774 (n = 1). Single nucleotide polymorphism (SNP)-based phylogenetic analysis revealed that global S. Welikade strains can be divided into four groups, and these three Chinese isolates were assigned to B (n = 2; Sal097 and XXB1016) and C (n = 1; XXB700). In Group B, the two Chinese ST5123 isolates were closely clustered with three UK ST5123 isolates. In Group C, the Chinese isolate was closely related to the other 12 ST3774 isolates. The number of virulence genes in the S. Welikade isolates ranged from 59 to 152. The galF gene was only present in Group A, the pipB2 gene was only absent from Group A, the avrA gene was only absent from Group B, and the allB, sseK1, sspH2, STM0287, and tlde1 were found only within Group C and D isolates. There were 15 loci unique to the Sal097 isolate. CONCLUSION: This study is the first to characterize and investigate clinical S. Welikade isolates in China. Responsible for a pediatric case of gastroenteritis in 2021, the clinical isolate harbored no antimicrobial resistance and belonged to phylogenetic Group B of global S. Welikade genomes.

BACKGROUND: The diagnosis of myasthenia gravis (MG) in children remains difficult. Circulating small extracellular vesicle (sEV)-derived miRNAs (sEV-miRNAs) have been recognized as biomarkers of various diseases and can be excreted by different cell types. These biomarker candidates also play a vital role in autoimmune diseases via intercellular communication. METHODS: In the present study, we used sEV isolation and purification methods to extract the plasma-derived sEV-miRNAs from children with MG and healthy controls. A small RNA sequencing analysis confirmed the miRNA expression features in plasma-derived sEVs from MG patients. The miRNA expression analysis in vitro was determined using microarray analysis. The enrichment and network analyses of altered sEV-miRNAs were performed using miRNA databases and Database for Annotation, Visualization, and Integrated Discovery website. Quantitative real-time polymerase chain reaction was performed for validation of sEV-miRNA. The diagnostic power of altered sEV-miRNAs was evaluated using receiver operating characteristic curve analyses. RESULTS: Twenty-four sEV-miRNAs with altered expression level were identified between groups by DESeq2 method. The miRNAs were extracted from the sEVs, which were isolated from human primary skeletal muscle cell culture treated with mAb198. The target genes and enriched pathways of sEV-miRNAs partially overlapped between cell supernatant and plasma samples. The significantly downregulated miR-143-3p was validated in quantitative real-time polymerase chain reaction analysis. CONCLUSIONS: For the first time, we report that plasma-derived sEV-miRNAs may act as novel circulating biomarkers and therapeutic targets in pediatric MG.

Aeromonas hydrophila, a Gram-negative bacterium, is one of the major pathogens causing bacterial sepsis in aquatic animals due to drug resistance and pathogenicity, which could cause high mortality and serious economic losses to the aquaculture. Sanguisorba officinalis (called DiYu in Chinese, DY) is well known as herbal medicine, which could inhibit the growth of pathogenic bacteria, hemostasis and regulate the immune response. Moreover, the active ingredients in DY could remarkably reduce drug resistance. In this study, we investigated the effects of probiotic fermentation cultures on A. hydrophila through in vitro and in vivo experiments. Three lactic acid bacteria, including Lactobacillus rhamnosus (LGG), Lactobacillus casei (LC) and Lactobacillus plantarum (LP), were selected to ferment the Chinese herbal medicine DY. The assays of antagonism showed that all three fermented cultures could influence the ability of A. hydrophila growth, among which L. rhamnosus fermented DY cultures appeared to be the strongest inhibitory effect. In addition, the biofilm determination revealed that L. rhamnosus fermented DY cultures could significantly inhibit the biofilm formation of A. hydrophila compared to the other groups. Furthermore, protease, lecithinase and urease activities were found in the three fermentation cultures. Three probiotics fermented DY cultures were orally administration with crucian carp to evaluate the growth performance, immunological parameters and pathogen resistance. The results showed that the three fermentation cultures could promote the growth performance of crucian carp, and the immunoglobulins, antioxidant-related enzymes and immune-related genes were significantly enhanced. Besides, the results showed that crucian carp received L. rhamnosus (60.87%), L. casei (56.09%) and L. plantarum (41.46%) fermented DY cultures had higher survival rates compared with the control group after infection with A. hydrophila. Meanwhile, the pathological tissue results revealed that the probiotic fermented cultures could largely improve the tissues damage caused by the pathogenic bacteria. In conclusion, this study proved that the fermentation cultures of three probiotics could effectively inhibit the growth of A. hydrophila, regulate the level of immune response and improve the survival rate against A. hydrophila in crucian carp. The present data suggest that probiotic fermented Sanguisorba officinalis act as a potential gut-targeted therapy regimens to protecting fish from pathogenic bacteria infection.

Scene text detection task aims to precisely localize text in natural environments. At present, the application scenarios of text detection topics have gradually shifted from plain document text to more complex natural scenarios. Objects with similar texture and text morphology in the complex background noise of natural scene images are prone to false recall and difficult to detect multi-scale texts, a multi-directional scene Uyghur text detection model based on fine-grained feature representation and spatial feature fusion is proposed, and feature extraction and feature fusion are improved to enhance the network's ability to represent multi-scale features. In this method, the multiple groups of 3 × 3 convolutional feature groups that are connected like the hierarchical residual to build a residual network for feature extraction, which captures the feature details and increases the receptive field of the network to adapt to multi-scale text and long glued dimensional font detection and suppress false positives of text-like objects. Secondly, an adaptive multi-level feature map fusion strategy is adopted to overcome the inconsistency of information in multi-scale feature map fusion. The proposed model achieves 93.94% and 84.92% F-measure on the self-built Uyghur dataset and the ICDAR2015 dataset, respectively, which improves the accuracy of Uyghur text detection and suppresses false positives.

As a research hotspot in the field of natural language processing (NLP), sentiment analysis can be roughly divided into explicit sentiment analysis and implicit sentiment analysis. However, due to the lack of obvious emotion words in the implicit sentiment analysis task and because the sentiment polarity contained in implicit sentiment words is not easily accurately identified by existing text-processing methods, the implicit sentiment analysis task is one of the most difficult tasks in sentiment analysis. This paper proposes a new preprocessing method for implicit sentiment text classification; this method is named Text To Picture (TTP) in this paper. TTP highlights the sentiment differences between different sentiment polarities in Chinese implicit sentiment text with the help of deep learning by converting original text data into word frequency maps. The differences between sentiment polarities are used as sentiment clues to improve the performance of the Chinese implicit sentiment text classification task. It does this by transforming the original text data into a word frequency map in order to highlight the differences between the sentiment polarities expressed in the implicit sentiment text. We conducted experimental tests on two common datasets (SMP2019, EWECT), and the results show that the accuracy of our method is significantly improved compared with that of the competitor's. On the SMP2019 dataset, the accuracy-improvement range was 4.55-7.06%. On the EWECT dataset, the accuracy was improved by 1.81-3.95%. In conclusion, the new preprocessing method for implicit sentiment text classification proposed in this paper can achieve better classification results.

Rapid quantification of soil organic matter (SOM) is a great challenge for the health assessment and fertility management of agricultural soil. Laser-induced breakdown spectroscopy (LIBS) with appropriate modeling algorithms is an alternative tool for this measurement. However, the current calibration strategy limits the prediction performance of the LIBS technique. In this study, 563 soil samples from Hetao Irrigation District in China were collected; the LIBS spectra of the soils were recorded in the wavenumber range of 288-950 nm with a resolution of 0.116 nm; a self-adaptive partial least squares regression model (SAM-PLSR) was employed to explore optimal model parameters for SOM prediction; and calibration parameters including sample selection for the calibration database, sample numbers and sample location sites were optimized. The results showed that the sample capacity around 60-80, rather than all of the samples in the soil library database, was selected for calibration from a spectral similarity re-ordered database regarding unknown samples; the model produced excellent predictions, with R2 = 0.92, RPD = 3.53 and RMSEP = 1.03 g kg-1. Both the soil variances of the target property and the spectra similarity of the soil background were the key factors for the calibration model, and the small sample set led to poor predictions due to the low variances of the target property, while negative effects were observed for the large sample set due to strong interferences from the soil background. Therefore, the specific unknown sample depended strategy, i.e., self-adaptive modelling, could be applied for fast SOM sensing using LIBS for soils in varied scales with improved robustness and accuracy.

Benefitting from the special structure of the leaf cuticle layer, plants have natural hydrophobicity and anti-fouling abilities. Inspired by the leaf surface structure, a biomimetic modification strategy was raised to improve the surface hydrophobicity of polyacrylate coating for controlled release fertilizer. Double-layer (polyacrylate and carnauba wax) coated fertilizer was obtained after biomimetic modification. The quality of controlled release fertilizer modified with the carnauba wax was greatly enhanced, and the coating material was effectively saved. The surface appearance of polyacrylate-coated fertilizer was improved for the surface blemish was repaired by the loaded carnauba wax. The characterizations by Fourier transform infrared spectroscopy indicated that the hydrogen bonds were formed between the water-based polyacrylate membrane and the carnauba wax layers. By optimizing the content of polyacrylate and carnauba wax, the release duration of the fertilizer was effectively prolonged, which was improved from 1 month to more than 2 months after the biomimetic modification. Therefore, biological wax as an environmentally-friendly natural material that has showed a broad potential in the application of coated controlled release fertilizer.

Metal-organic frameworks (MOFs), due to their possessing a porous structure, are potential candidates for solid-state ionic conduction materials. Moreover, uncoordinated carboxylic acid groups (-COOH) of MOFs can be used as postsynthetic modification sites, which are favorable for lithium ion exchange. Herein, we synthesized a unique multiple carboxylic zinc metal-organic framework (Zn-MOF-COOH) containing uncoordinated carboxylic acid groups. Zn-MOF-COOLi was synthesized through deprotonation using LiOH via a straightforward acid-base reaction at room temperature (RT), thereby exhibiting better good electrochemical properties. The lithium ionic conductivity (σ) increased from 1.81 × 10-5 to 1.65 × 10-4 S·cm-1, lithium ion transference number (tLi+) rose from 0.67 to 0.77, and the electrochemical window improved from 2.0-5.5 to 1.5-6.5 V. This work offers a new strategy to improve the σ of MOFs and a new perspective toward manufacturing of high-performance solid-state ionic conduction materials.

Homochiral metal-organic frameworks (HMOFs) have garnered considerable attention due to their extrachiral properties and broad application for chiral recognition. However, assembling a pair of high-quality chiral MOFs for sensing enantiomers precisely is a formidable challenge because of the complicated chiral environment and uncontrollable coordinated conditions. Herein, one pair of homochiral UiO-66 analogues, S-1 (l-AP@UiO-66-(COOH)2) and R-1 (d-AP@UiO-66-(COOH)2), are reported for chiral recognition. They were fabricated via a condensation reaction between the carboxyl groups of UiO-66-(COOH)2 and amino groups of l/d-amino propanol (l/d-AP). These novel fluorescent probes exhibited highly enantioselective fluorescence enhancement towards l/d-phenylalaninol (l/d-PA). For example, when S-1 and R-1 were treated with l-PA or d-PA, they displayed different fluorescence responses: the enantiomeric fluorescence enhancement ratio (ef) was 2.51 and 0.41 for S-1 and R-1, respectively. Hence, a visible difference in fluorescence enhancement for l-PA and d-PA and excellent enantioselective behavior between S-1 and l-PA (or R-1 and d-PA) was displayed. Measurements of fluorescence lifetime, powder X-ray diffraction, molecular-dynamic simulations and Benesi-Hildebrand plots were employed to determine the observed high enantioselectivity for l/d-PA. In brief, we found that two post-modified HMOFs, S-1 and R-1, were outstanding enantioselective sensors for detecting l-PA and d-PA. They had a prominent difference in ef and remarkable enantioselectivity factor α and ΔΔG based on steric hindrance and stereochemical difference.

BACKGROUND: Salmonella Wandsworth is a rare serotype of Salmonella. This study analyzed the genotyping, genome structure, and molecular biological functions of Salmonella Wandsworth based on the results of multilocus sequence typing and next-generation sequencing genome assembly analysis. METHODS: Serological typing was performed using the slide-agglutination method. The micro broth dilution method was used to test antibiotic susceptibility. Multilocus sequence typing (MLST) was used to perform the homology analysis, while the second-generation sequencing genome analysis was used to analyze the whole genome of the bacteria. RESULTS: Salmonella Wandsworth is Group Q Salmonella. The MLST of this strain was ST1498. Salmonella Wandsworth was sensitive to antibiotics, such as ceftriaxone, imipenem, chloramphenicol, and colistin, but was resistant to ampicillin, cefalotin, gentamicin, and ciprofloxacin. The second-generation sequencing results showed that the genome sequence length of the bacteria was 5109457bp. Annotated COG library analysis generated 3,746 corresponding genes. After the comparison with the KEGG library, 1,340 genes, which participate in 19 types of metabolic pathways, were obtained. A total of 249 pathogenic factors and 2 disease islands were predicted. 2 CRISPR sites and 8 Cas sites were predicted. It can be seen from the evolutionary tree that Salmonella Wandsworth MLST1498 and Paratyphi B str.SPB7 are gathered together. We identified one resistance gene, namely, aac(6')-Iaa accounting for aminoglycoside resistance. CONCLUSION: Salmonella Wandsworth isolated in this study is Salmonella group Q. Consequently, it is necessary to strengthen the understanding of clinical infections of Salmonella Wandsworth and carry out continuous monitoring and research.

Salmonella Enteritidis is an important foodborne pathogen with high prevalence of resistance to cephalosporins, imposing a serious threat to public health. Therefore, a total of 162 Salmonella Enteritidis isolates collected from child patients in China from 2007 to 2017 were characterized for their resistance to cephalosporins and investigated the transmission characteristics of cephalosporin resistance gene. We found that 15 (9.26%) isolates were all resistant to cefalotin (minimum inhibitory concentration [MIC] ≥512 µg/mL), ceftazidime (MIC 16-128 µg/mL), ceftriaxone (MIC 64 to ≥512 µg/mL), ceftiofur (MIC 64-256 µg/mL), and cefotaxime (MIC 64 to ≥512 µg/mL) with the possession of cephalosporin resistance genes blaCTX-M-55 (n = 13), blaCTX-M-101 (n = 1), and blaCTX-M-153 (n = 1). Molecular typing further revealed that these 15 isolates belonged to sequence type ST11 and shared close pulsed-field gel electrophoresis patterns, suggesting the possibility of clonal spread in Salmonella Enteritidis interspecies. Furthermore, conjugation experiments were successfully performed in 13 of 15 isolates, and blaCTX-M-55 was present on conjugative plasmids with sizes ranging from 54.7 to 173.4 kb. Compared with recipient Escherichia coli C600, transconjugants conferred elevated MICs for cephalosporins ranging from 2- to 2048-fold. The genetic structure surrounding of blaCTX-M-55 gene in transconjugants were ΔISEcp1-blaCTX-M-55-orf477 (n = 8) and ISEcp1-blaCTX-M-55-orf477 (n = 3), respectively. Taken together, blaCTX-M on the plasmids might contribute to cephalosporin resistance in Salmonella Enteritidis, and conjugative transfer of blaCTX-M-55 might facilitate the spread of cephalosporin resistance in Salmonella Enteritidis. Hence, effective mitigation measurements are needed to reduce the threat caused by cephalosporin-resistant Salmonella Enteritidis to public health.

BACKGROUND: At present, azithromycin has become an effective treatment for severe diarrhea caused by Enterotoxigenic Escherichia coli (ETEC) infection. However, enterobacteria have begun to develop resistance to azithromycin and have attracted attention in recent years. This study conducted to described the emergence of a high proportion of azithromycin-resistant ETEC serogroup O6 strains in Shanghai and to analyzed the mechanisms of azithromycin resistance. RESULTS: Strains from adult diarrhea patients with ETEC serogroup O6 infections were collected by Shanghai Diarrhea Surveillance Network and the Foodborne Surveillance Network from 2016 to 2018. We tested 30 isolates of ETEC O6 serogroup, 26 of which were resistant to azithromycin. Phylogenetic analysis revealed that these ETEC serogroup O6 strains have formed an independent dominant clone. S1-PFGE and southern blotting revealed the presence of the mphA gene on the 103 kb plasmid. Illumina and Nanopore sequencing and plasmid coverage analysis further confirmed that azithromycin-resistant strains carried a novel IncFII plasmid harboring mphA and blaTEM-1 resistance genes. CONCLUSIONS: This is the first study to report a high proportion of azithromycin resistance in a particular ETEC serogroup due to a specific plasmid carrying mphA. Our findings indicate the rapid spread of azithromycin resistance, highlighting the urgency of stringent surveillance and control measure.

In this study, we explored the prevalence of oxacillin-susceptible mecA-positive Staphylococcus aureus (OS-MRSA) in staphylococcal food poisoning outbreak isolates and foodborne isolates, and then investigated their molecular characteristics, classical staphylococcal enterotoxins (SEs), and drug resistance. Eight (2.9%) of 275 isolates from food poisoning outbreaks and 7 (3.8%) of 184 isolates from retail foods were identified as OS-MRSA isolates. Among the 15 OS-MRSA isolates, the most frequently detected toxin genes were hld (100%), hla (93.3%), pvl (80.0%), and hlb (46.7%) followed by seg and seq (33.3%, each), hlg (26.7%), seb and hlgv (20.0%, each), sec, seh, sel, sep, and tst (13.3%, each), and sei, sem, sen, and seo (6.7%, each). None of isolates carried other tested virulence genes. The most frequently detected classical SEs were SEB and SEC (26.7%, each), followed by SEA and SEE (20.0%, each), and SED (6.7%). Resistance was most frequently observed in ampicillin, penicillin, and cefoxitin (100%, each), followed by trimethoprim/sulfamethoxazole (93.3%), erythromycin (73.3%), amoxicillin/clavulanic acid (46.7%), tetracyclines (26.7%), and ciprofloxacin (6.7%). All isolates were susceptible to other tested antibiotics. A dominant molecular type belonged to ST398-IVa-t034 (26.7%), followed by ST59-IVa-t437 (20.0%), ST88-III-t14340 and ST1-IVa-t114 (13.3%, each), and ST5-II-t002, ST630-t4549, ST5-II, and ST4495-t10738 (6.7%, each). Our findings indicated that OS-MRSA strains had a low prevalence rate among outbreak strains and foodborne strains, which frequently harbored SCCmec IVa, and carried a variety of toxin genes, and also expressed numerous classical SEs. In addition, all OS-MRSA isolates were susceptible to the majority of antibacterial agents except ß-lactam. Our study is the first to report that OS-MRSA isolates are associated with food poisoning outbreaks worldwide.

Magnetic rings are the most widely used magnetic material product in industry. The existing manual defect detection method for magnetic rings has high cost, low efficiency and low precision. To address this issue, a magnetic ring multi-defect stereo detection system based on multi-camera vision technology is developed to complete the automatic inspection of magnetic rings. The system can detect surface defects and measure ring height simultaneously. Two image processing algorithms are proposed, namely, the image edge removal algorithm (IERA) and magnetic ring location algorithm (MRLA), separately. On the basis of these two algorithms, connected domain filtering methods for crack, fiber and large-area defects are established to complete defect inspection. This system achieves a recognition rate of 100% for defects such as crack, adhesion, hanger adhesion and pitting. Furthermore, the recognition rate for fiber and foreign matter defects attains 92.5% and 91.5%, respectively. The detection speed exceeds 120 magnetic rings per minutes, and the precision is within 0.05 mm. Both precision and speed meet the requirements of real-time quality inspection in actual production.

Glioblastoma (GBM) is the most prevalent primary malignancy of the central nervous system with obvious aggressiveness, and is associated with poor clinical outcome. Studies have indicated that calcium ion (Ca2+ ) can positively regulate the initiation of malignancy with regard to GBM by modulating quiescence, proliferation, migration and maintenance. Hippocalcin like-1 protein (HPCAL1) serves as a sensor of Ca2+ . However, the understanding of HPCAL1 activity in GBM is limited. The present study revealed that the gene HPCAL1 was up-regulated by Ca2+ in the tissues and cells of GBM. Ectopic expression of HPCAL1 promoted proliferation of cells. Exhaustion of HPCAL1 inhibited cell growth not only in vivo, but also in vitro. In addition, HPCAL1 enhanced the Wnt pathway by stimulating ß-catenin accumulation and nuclear translocation in GBM cells, while ß-catenin silencing significantly inhibited the proliferation and growth of the GBM cells. Our results showed that Ser9 phosphorylation of GSK3ß was significantly decreased after HPCAL1 knockdown in GBM cells, and knockdown of the gene GSK3ß in GBM cells enhanced cell proliferation and promoted transcription of the genes CCND1 and c-Myc. Furthermore, the phosphorylation of ERK was decreased in the cells with HPCAL1 knockdown, while it was promoted via overexpression of HPCAL1. The suppression or depletion of the gene ERK decreased proliferation triggered by overexpression of HPCAL1 and impaired transcription of the genes c-Myc and CCND1. These studies elucidate the tumour-promoting activity of HPCAL1. They also offer an innovative therapeutic strategy focusing on the HPCAL1-Wnt/ß-catenin axis to regulate proliferation and development of GBM.

BACKGROUND: Shigella spp., facultative anaerobic bacilli of the family Enterobacteriaceae, are one of the most common causes of diarrheal diseases in human worldwide which have become a significant public health burden. So, we aimed to analyze the antimicrobial phenotypes and to elucidate the molecular mechanisms underlying resistance to cephalosporins and fluoroquinolones in Shigella isolates from patients with diarrhea in Shanxi Province. RESULTS: During 2006-2016, we isolated a total of 474 Shigella strains (including 337 S. flexneri and 137 S. sonnei). The isolates showed high rates of resistance to traditional antimicrobials, and 26, 18.1 and 3.0% of them exhibited resistance to cephalosporins, fluoroquinolones and co-resistance to cephalosporins and fluoroquinolones, respectively. Notably, 91.1% of these isolates, including 22 isolates that showed an ACTSuT profile, exhibited multidrug resistance (MDR). The resistance rates to cephalosporins in S. sonnei isolates were higher than those in S. flexneri. Conversely, the resistance rates to fluoroquinolones were considerably higher in S. flexneri isolates. Among the 123 cephalosporins-resistant isolates, the most common extended-spectrum beta-lactamase gene was blaTEM-1, followed by blaCTX-M, blaOXA-1, and blaSHV-12. Six subtypes of blaCTX-M were identified, blaCTX-M-14 (n = 36) and blaCTX-M-55 (n = 26) were found to be dominant. Of all the 86 isolates with resistance to fluoroquinolones and having at least one mutation (Ser83Leu, His211Tyr, or Asp87Gly) in the the quinolone resistance-determining regions of gyrA, 79 also had mutation of parC (Ser80Ile), whereas 7 contained plasmid-mediated quinolone resistance genes including qnrA, qnrB, qnrS, and aac(60)-Ib-cr. Furthermore, pulsed-field gel electrophoresis analysis (PFGE) showed a considerable genetic diversity in S. flexneri isolates. However, the S. sonnei isolates had a high genetic similarity. CONCLUSIONS: Coexistence of diverse resistance genes causing the emergence and transmission of MDR might render the treatment of shigellosis difficult. Therefore, continuous surveillance might be needed to understand the actual disease burden and provide guidance for shigellosis.

OBJECTIVES: This study aimed to trace the transmission source of Salmonella enterica serovar Typhimurium and Salmonella enterica serovar Enteritidis strains associated with enteric infections in Shanghainese children, and understand the molecular mechanism of resistance to third-generation cephalosporins and ciprofloxacin. MATERIALS AND METHODS: The profiles of pulsed-field gel electrophoresis (PFGE) were compared among the isolates from children, animal, and environment. Antimicrobial susceptibility was determined using the minimal inhibitory concentrations and Kirby-Bauer disk diffusion method. Extended-spectrum ß-lactamase (ESBL) producing isolates mediated by resistance genes were identified using polymerase chain reaction and sequencing. RESULTS: Based on PFGE patterns, 49 (33.1%) of 148 human Salmonella Typhimurium isolates located in the dominant PFGE clusters were genetically related to the isolates from poultry source, environment water, aquatic products, and reptiles, whereas 97 (97.0%) of 100 human Salmonella Enteritidis isolates were genetically related to isolates from poultry and water. The rates of resistance to ceftriaxone among clinical Salmonella Typhimurium and Salmonella Enteritidis isolates were 42.0% and 14.2%, respectively. Besides, 35.1% of Salmonella Typhimurium isolates displayed resistance to ciprofloxacin; 64.9% of Salmonella Typhimurium isolates and 97.0% of Salmonella Enteritidis isolates displayed reduced susceptibility to ciprofloxacin. Of 64 ESBL/AmpC-producing strains, CTX-M, TEM, DHA, and CMY were detected at frequencies of 86.0%, 62.5%, 7.8%, 3.1%, and 3.1%, respectively. CONCLUSIONS: The transmission sources of Salmonella Typhimurium and Salmonella Enteritidis infections in Shanghainese children were diverse. The high prevalence of resistance to third-generation cephalosporins and ciprofloxacin mediated by multiple molecular mechanisms needs continuous monitoring and intervention.

For the past decades, recognition technologies of multispectral palmprint have attracted more and more attention due to their abundant spatial and spectral characteristics compared with the single spectral case. Enlightened by this, an innovative robust L2 sparse representation with tensor-based extreme learning machine (RL2SR-TELM) algorithm is put forward by using an adaptive image level fusion strategy to accomplish the multispectral palmprint recognition. Firstly, we construct a robust L2 sparse representation (RL2SR) optimization model to calculate the linear representation coefficients. To suppress the affection caused by noise contamination, we introduce a logistic function into RL2SR model to evaluate the representation residual. Secondly, we propose a novel weighted sparse and collaborative concentration index (WSCCI) to calculate the fusion weight adaptively. Finally, we put forward a TELM approach to carry out the classification task. It can deal with the high dimension data directly and reserve the image spatial information well. Extensive experiments are implemented on the benchmark multispectral palmprint database provided by PolyU. The experiment results validate that our RL2SR-TELM algorithm overmatches a number of state-of-the-art multispectral palmprint recognition algorithms both when the images are noise-free and contaminated by different noises.

Since the initial discovery of mcr-1 in an Escherichia coli isolate from China, the gene has also been detected in Klebsiella pneumoniae and Salmonella enterica but is rarely reported in other Enterobacteriaceae Here, we report the isolation and identification of a Shigella flexneri strain harboring mcr-1 from stool samples in a pig farm in China from 2009. The MIC of colistin for the isolate is 4 µg/ml. Conjugation assays showed that the donor S. flexneri strain has functional and transferable colistin resistance. Sequencing revealed that mcr-1 was present on a putative composite transposon flanked by inverted repeats of ISApl1IMPORTANCE There are four species of Shigella, and Shigella flexneri is the most frequently isolated species in low- and middle-income countries (LMICs). In this study, we report a functional, transferable, plasmid-mediated mcr-1 gene in S. flexneri We have shown that mcr-1 is located on a novel composite transposon which is flanked by inverted repeats of ISApl1 The host strain is multidrug resistant, and this multidrug resistance is also transferable. The finding of a functional mcr-1 gene in S. flexneri, a human-associated Enterobacteriaceae family member, is a cause for concern as infections due to S. flexneri are the main Shigella infections in most low- and middle-income countries.

BACKGROUND: Among 2179 Salmonella isolates obtained during national surveillance for salmonellosis in China from 2005 to 2013, we identified 46 non-H2S-producing strains originating from different sources. METHODS: The isolates were characterized in terms of antibiotic resistance and genetic variability by pulsed-field gel electrophoresis and multilocus sequence typing. Mutation in the phs operon, which may account for the non-H2S-producing phenotype of the isolated Salmonella strains, was performed in this study. RESULTS: Among isolated non-H2S-producing Salmonella strains, more than 50% were recovered from diarrhea patients, of which H2S-negative S. Gallinarum, S. Typhimurium, S. Choleraesuis and S. Paratyphi A isolates constituted 76%. H2S-negative isolates exhibited a high rate of resistance to ticarcillin, ampicillin, and tetracycline, and eight of them had the multidrug resistance phenotype. Most H2S-negative Salmonella isolates had similar pulsed-field gel electrophoresis profiles and the same sequence type as H2S-positive strains, indicating a close origin, but carried mutations in the phsA gene, which may account for the non-H2S-producing phenotype. CONCLUSIONS: Our data indicate that multiple H2S-negative strains have emerged and persist in China, emphasizing the necessity to implement efficient surveillance measures for controlling dissemination of these atypical Salmonella strains.
