This study tested the hypothesis that brassinosteroids (BRs) mediate moderate soil-drying (MD) to alleviate spikelet degeneration under high temperature (HT) stress during meiosis of rice (Oryza sativa L.). A rice cultivar was pot-grown and subjected to normal temperature (NT) and HT treatments during meiosis, and two irrigation regimes including well-watered (WW) and MD were imposed to the plants simultaneously. The MD effectively alleviated the spikelet degeneration and yield loss under HT stress mainly via improving root activity and canopy and panicle traits including higher photosynthetic capacity, tricarboxylic acid cycle activity, and antioxidant capacity than WW. These parameters were regulated by BRs levels in plants. The decrease in BRs levels at HT was due mainly to the enhanced BRs decomposition, and the MD could rescue the BRs deficiency at HT via enhancing BRs biosynthesis and impeding decomposition. The connection between BRs and HT was verified by using rice BRs-deficient mutants, transgenic rice lines, and chemical regulators. Similar results were obtained in the open-air field experiment. The results suggest that BRs can mediate the MD to alleviate spikelet degeneration under HT stress during meiosis mainly via enhancing root activity, canopy traits, and young panicle traits of rice.

BACKGROUND: Applying organic fertilizer coupled with chemical fertilizer has been widely adopted to improve crop productivity and quality and develop sustainable agriculture. However, little information is available about the effects of organic fertilizer on the grain quality of rice (Oryza sativa L.), especially nutritional quality and starch quality. In the present study, high yielding 'super' rice cultivars were grown in the field with three cultivation practices, including zero nitrogen application (0N), local high yielding practice with chemical fertilizer (T1) and T1 treatment with additional organic fertilizer (T2). RESULTS: Application of organic fertilizer synergistically improved rice production, nitrogen use efficiency, milling and appearance quality, and nutritional quality, including the contents of glutelin, essential amino acids and microelements, and also increased amylopectin and the ratio of the short chain of amylopectin, leading to a reduction in relative crystallinity, and decreased prolamin content. Application of organic fertilizer also increased the viscosity and breakdown values, whereas it decreased the pasting temperature and gelatinization enthalpy, resulting in better cooking and eating quality. CONCLUSION: Overall, application of organific fertilizer could synergistically improve nitrogen use efficiency and grain quality, including the structure and physicochemical properties of starch, contents of high value protein and amino acids, contents of microelements, and cooking and eating quality. © 2021 Society of Chemical Industry.

BACKGROUND: Brassinosteroids (BRs) are a new group of plant hormones and play important roles in plant growth and development. However, little information is available if BRs could regulate spikelet development in rice (Oryza sativa L.) especially under soil-drying conditions. This study investigated whether and how BRs mediate the effect of soil-drying on spikelet differentiation and degeneration in rice. A rice cultivar was field-grown and exposed to three soil moisture treatments during panicle development, that is, well-watered (WW), moderate soil-drying (MD) and severe soil-drying (SD). RESULTS: Compared with the WW treatment, the MD treatment enhanced BRs biosynthesis in young panicles, increased spikelet differentiation and reduced spikelet degeneration. The SD treatment had the opposite effects. Changes in expression levels of key rice inflorescence development genes (OsAPO2 and OsTAW1), ascorbic acid (AsA) content, and activities of enzymes involved AsA synthesis and recycle, and amount of nonstructural carbohydrates (NSC) in young panicles were consistent with those in BRs levels, whereas hydrogen peroxide (H2O2) content showed opposite trend. Knockdown of the BRs synthesis gene OsD11 or application of a BRs biosynthesis inhibitor to young panicles markedly decreased OsAPO2 and OsTAW1 expression levels, BRs and AsA contents, activities of enzymes involved AsA synthesis and recycle, NSC amount in rice panicles and spikelet differentiation but increased the H2O2 content and spikelet degeneration compared to the control (the wide type or application of water). The opposite effects were observed when exogenous BRs were applied. CONCLUSIONS: The results suggest that BRs mediate the effect of soil-drying on spikelet differentiation and degeneration, and elevated BRs levels in rice panicles promote spikelet development under MD by enhancing inflorescence meristem activity, AsA recycle and NSC partitioning to the growing panicles.

MAIN CONCLUSION: Abscisic acid mediates the effect of post-anthesis soil drying on grain filling through regulating the activities of key enzymes and expressions of genes involved in sucrose-to-starch conversion in rice spikelets. This study investigated if abscisic acid (ABA) would mediate the effect of post-anthesis soil drying on grain filling through regulating the key enzymes in sucrose-to-starch conversion in rice (Oryza sativa L.) spikelets. Two rice cultivars were field-grown. Three treatments, well-watered (WW), moderate soil drying (MD), and severe soil drying (SD), were imposed from 6 days after full heading until maturity. When compared with those under the WW, grain filling rate, grain weight, and sink activity, in terms of the activities and gene expression levels of sucrose synthase, ADP glucose pyrophosphorylase, starch synthase, and starch branching enzyme, in inferior spikelets were substantially increased under the MD, whereas they were markedly decreased in both superior and inferior spikelets under the SD. The two cultivars showed the same tendencies. Both MD and SD increased ABA content and expression levels of its biosynthesis genes in spikelets, with more increase under the SD than the MD. ABA content was significantly correlated with grain filling rate and sink activities under both WW and MD, while the correlations were not significant under the SD. Application of a low concentration ABA to WW plants imitated the results under the MD, and applying with a high concentration ABA showed the effect of the SD. The results suggest that ABA plays a vital role in grain filling through regulating sink activity and functions in a dose-dependent manner. An elevated ABA level under the MD enhances, whereas a too high level of ABA under the SD decreases, sink activity.

In wheat stems, the levels of fructan-dominated water-soluble carbohydrates (WSC) do not always correlate well with grain yield. Field drought experiments were carried out to further explain this lack of correlation. Wheat (Triticum aestivum) varieties, Westonia, Kauz and c. 20 genetically diverse double haploid (DH) lines derived from them were investigated. Substantial genotypic differences in fructan remobilization were found and the 1-FEH w3 gene was shown to be the major contributor in the stem fructan remobilization process based on enzyme activity and gene expression results. A single nucleotide polymorphism (SNP) was detected in an auxin response element in the 1-FEH w3 promoter region, therefore we speculated that the mutated Westonia allele might affect gene expression and enzyme activity levels. A cleaved amplified polymorphic (CAP) marker was generated from the SNP. The harvested results showed that the mutated Westonia 1-FEH w3 allele was associated with a higher thousand grain weight (TGW) under drought conditions in 2011 and 2012. These results indicated that higher gene expression of 1-FEH w3 and 1-FEH w3 mediated enzyme activities that favoured stem WSC remobilization to the grains. The CAP marker residing in the 1-FEH w3 promoter region may facilitate wheat breeding by selecting lines with high stem fructan remobilization capacity under terminal drought.

Polyamines and ethylene are key regulators of the growth and development, quality formation, and stress response of cereal crops such as rice. However, it remains unclear whether the application of these regulators could improve the nutritional quality via increasing amino acids in rice grains. This study examined the role of exogenous polyamines and ethylene in regulating amino acid levels in the milled rice of earlier-flowered superior grain (SG) and later-flowered inferior grain (IG). Two rice varieties were field grown, and either 1 mmol L-1 spermidine (Spd) or 50 µmol L-1 amino-ethoxyvinylglycine (AVG) was applied to panicles at the early grain-filling stage. The control check (CK) was applied with deionized water. The results showed that the Spd or AVG applications significantly increased polyamine (spermine (Spm) and Spd) contents and decreased ethylene levels in both SG and IG and significantly increased amino acid levels in the milled rice of SG and IG relative to the CK. Collectively, the application of Spd or AVG can increase amino acid-based nutritional quality and grain yield via increasing polyamine (Spm and Spd) contents and reducing ethylene levels in both SG and IG of rice.

High temperatures (HT) and drought are two major factors restricting wheat growth in the early growth stages. This study investigated the role of glutathione (GSH) amendment (0.0, 0.5, 1.0, and 2.0 mM) to soil in mitigating the adverse effect of HT (33 °C, with 25 °C as a control), water regimes (60% of field capacity and control), and their combinations. HT decreased the length, project area, surface area, volume, and forks of the root, while drought had the reverse effect. Shoot length, leaf area, leaf relative water content, and shoot and root dry matter were significantly decreased by HT and drought, and their combined impact was more noticeable. GSH significantly promoted the root system, shoot growth, and leaf relative water content. The combined treatment reduced chlorophyll a, chlorophyll b, and total chlorophyll. However, 0.5 mM GSH raised chlorophyll a, chlorophyll b, and total chlorophyll by 28.6%, 41.4%, and 32.5%, respectively, relative to 0.0 mM GSH. At combined treatment, 0.5 mM GSH decreased malondialdehyde (MDA) by 29.5% and increased soluble protein content by 24.1%. GSH meaningfully enhanced the activity of superoxide dismutase, catalase, and ascorbate peroxide in different treatments. This study suggested that GSH could protect wheat seedlings from the adverse effects of HT and/or drought stresses.

This study tested the hypothesis that the interaction between polyamines and ethylene may mediate the effects of soil drying on grain filling of rice (Oryza sativa L.). Two rice cultivars were pot grown. Three treatments, well-watered, moderate soil drying (MD), and severe soil drying (SD), were imposed from 8 d post-anthesis until maturity. The endosperm cell division rate, grain-filling rate, and grain weight of earlier flowering superior spikelets showed no significant differences among the three treatments. However, those of the later flowering inferior spikelets were significantly increased under MD and significantly reduced under SD when compared with those which were well watered. The two cultivars showed the same tendencies. MD increased the contents of free spermidine (Spd) and free spermine (Spm), the activities of S-adenosyl-L-methionine decarboxylase and Spd synthase, and expression levels of polyamine synthesis genes, and decreased the ethylene evolution rate, the contents of 1-aminocylopropane-1-carboxylic acid (ACC) and hydrogen peroxide, the activities of ACC synthase, ACC oxidase, and polyamine oxidase, and the expression levels of ethylene synthesis genes in inferior spikelets. SD exhibited the opposite effects. Application of Spd, Spm, or an inhibitor of ethylene synthesis to rice panicles significantly reduced ethylene and ACC levels, but significantly increased Spd and Spm contents, grain-filling rate, and grain weight of inferior spikelets. The results were reversed when ACC or an inhibitor of Spd and Spm synthesis was applied. The results suggest that a potential metabolic interaction between polyamines and ethylene biosynthesis responds to soil drying and mediates the grain filling of inferior spikelets in rice.

Optimizing the N application amount and topdressing ratio can increase crop yield and decrease N loss, but its internal mechanisms have not been well studied, especially from the aspects of population dynamics and structure, ear fruiting traits. Here, field experiments, with three N rates 120 (N1), 180 (N2), and 240 (N3) kg N ha-1 and three N topdressing ratios T1 (7:3), T2 (6:4), and T3 (5:5) were conducted. At the same N level, results showed that the N accumulation amounts in the leaf, grain, and plant in T2 were higher than in T3 and T1, and increasing 60 kg N ha-1 (N3 compared to N2, N2 compared to N1) significantly enhanced N accumulation amounts. The effect of the N topdressing ratio on partial factor productivity of applied N was consistently T2 > T3 > T1, but T1 was more conducive to improving N utilization efficiency for grain and biomass production. After the jointing stage, compared to T1 and T3, T2 was more conducive to increasing the population growth rate of plant height, leaf area index, leaf area growth rate, dry matter weight, dry matter accumulation rate, light interception rate, and spikelets of population, and the above-mentioned indexes of population could be significantly enhanced by increasing 120 kg N ha-1. T2 increased the fruiting spikelets per ear, grains per ear, grain weight per ear, fruiting rate per ear, grain filling rate per ear, and yield but decreased the sterile spikelets at the top and bottom and imperfect grains per ear. Increasing N from 120 kg ha-1 to 180 kg ha-1 or from 180 kg ha-1 to 240 kg ha-1 significantly enhanced yield. The N accumulation amount in the grain, leaf, plant, leaf area growth rate, dry matter accumulation rate, light interception rate, population spikelets, fruiting spikelets per ear, grain filling rate, and yield were significantly positively correlated with each other. This study demonstrates a suitable N application rate with a N topdressing ratio 6:4 would more effectively improve N efficiency, population dynamics, structure, ear fruiting traits, and yield, but the effect of the N topdressing ratio is not as significant as that of increasing 60 kg N ha-1.

BACKGROUND: Hepatocellular carcinoma (HCC) has very low overall survival. According to global cancer statistics, approximately 905677 new cases were reported in 2020, with at least 830180 of them being fatal. Cluster of differentiation 147 (CD147) is a novel, transmembrane glycoprotein that is expressed in a wide variety of tumor cells and plays an important role in various stages of tumor development. Based on the reports described previously, we theorize that CD147 may be used as a novel biological indicator to predict the prognosis of HCC. To study this possibility, expression profiles of CD147 and corresponding clinical data from The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) databases were analyzed, and a hazard ratio (HR) was established. AIM: To explore the pattern of CD147 expression and its applicability in the prognosis of HCC. To establish HRs and probability points for predicting the prognosis of HCC by correlating CD147 expression with clinical characteristics. To determine if CD147 can be a reliable biomarker in HCC prognosis. METHODS: The CD147 expression profile in HCC and corresponding clinical data were obtained from TCGA database. The expression patterns of CD147 were then validated by analyzing data from the GEO database. In addition, CD147 immunohistochemistry in HCC was obtained from the Human Protein Atlas. CD147 expression patterns and clinical characteristics in the prognosis of HCC were analyzed by accessing the UALCAN web resource. Accuracy, sensitivity, and specificity of the CD147 expression profile in predictive prognosis were determined by the time-dependent receiver operating characteristic (ROC) curves. Kaplan-Meier curves were plotted to estimate the HR of survival in HCC. Univariate and multivariate Cox regression proportional hazards analyses of CD147 expression levels and clinical characteristics as prognostic factors of HCC were performed. Nomograms were used to establish probability points and predict prognosis. RESULTS: Data from TCGA and GEO databases revealed that CD147 was significantly overexpressed in HCC (P = 1.624 × 10-12 and P = 1.2 × 10-5, respectively). The expression of CD147 and prognosis of HCC were significantly correlated with the clinical characteristics of HCC as per the data from the UALCAN web resource (P < 0.05). Kaplan-Meier analysis of CD147 expression in HCC revealed that the high expression groups showed poor prognosis and an HR of survival > 1 [log-rank test, P = 0.000542, HR (in high expression group): 1.856, 95% confidence interval (CI): 1.308 to 2.636]. ROC curves were plotted to analyze the 1-year, 3-year, and 5-year survival rates. The area under the ROC curve values were 0.675 (95%CI: 0.611 to 0.740), 0.623 (95%CI: 0.555 to 0.692), and 0.664 (95%CI: 0.582 to 9.745), respectively. Univariate Cox analysis of CD147 expression and clinical characteristics of HCC and multivariate Cox analysis of CD147 patterns and pathological tumor-node-metastasis stage showed significant differences (univariate Cox, P = 0.00013, HR: 1.424, 95%CI: 1.884 to 1.707 and P = 0.00066, HR: 1.376, 95%CI: 1.145 to 1.654, respectively; multivariate Cox, P = 0.00578, HR: 1.507, 95%CI: 1.126 to 2.018 and P = 0.00336, HR: 1.443, 95%CI: 1.129 to 1.844, respectively). Nomograms were plotted to establish the probability points and predict prognosis. The total points ranged from 0 to 180, and the C-index value was 0.673 (95%CI: 0.600 to 1.000, P < 0.01). CONCLUSION: Overexpression of CD147 was correlated with poor prognosis in HCC. The CD147 expression profile combined with clinical characteristics can reliably predict the prognosis of HCC. CD147 can serve as a biomarker to predict the prognosis of HCC.

With the continuous improvement of living standards, the preferences of consumers are shifting to rice varieties with high eating and cooking quality (ECQ). Milled rice is mainly composed of starch, protein, and oil, which constitute the physicochemical basis of rice taste quality. This review summarizes the relationship between rice ECQ and its intrinsic ingredients, and also briefly introduces the effects of nitrogen fertilizer management on rice ECQ. Rice varieties with higher AC usually have more long branches of amylopectin, which leach less when cooking, leading to higher hardness, lower stickinesss, and less panelist preference. High PC impedes starch pasting, and it may be hard for heat and moisture to enter the rice interior, ultimately resulting in worse rice eating quality. Rice with higher lipid content had a brighter luster and better eating quality, and starch lipids in rice have a greater impact on rice eating quality than non-starch lipids. The application of nitrogen fertilizer can enhance rice yield, but it also decreases the ECQ of rice. CRNF has been widely used in cereal crops such as maize, wheat, and rice as a novel, environmentally friendly, and effective fertilizer, and could increase rice quality to a certain extent compared with conventional urea. This review shows a benefit to finding more reasonable nitrogen fertilizer management that can be used to regulate the physical and chemical indicators of rice grains in production and to improve the taste quality of rice without affecting yield.

To fully explore the effects of N on enhancing photosynthesis, grain quality, and yield of wheat (Ningmai 13), experiments with four nitrogen levels 0 (N0), 120 (N1), 180 (N2), and 240 (N3) kg N ha−1 and four ratios of basal to topdressing R0 (0:0), R1 (7:3), R2 (6:4), and R3 (5:5) were conducted. The basal N was applied to soil before sowing and the topdressing N was applied at jointing stage. The effect of N topdressing ratio on improving leaf area of photosynthetic efficiency was insignificant under the same N level. The effect of N fertilization level on increasing chlorophyll content was more significant than that of N topdressing ratio. Within the same N level, the canopy photosynthetically active radiation in R2 was higher than that in R1 and R3, and increasing N by 60 kg ha−1 significantly enhanced canopy photosynthetically active radiation. The effect of N topdressing ratio on photosynthetic rate, stomatal conductance, and transpiration rate were consistently R2 > R3 > R1; compared to N1, N3 could significantly increase photosynthetic rate. Increasing 120 kg N ha−1 significantly enhanced grain protein content, wet gluten, and sedimentation value, while the effect of N topdressing ratio was insignificant. Increasing N dose from 120 kg ha−1 to 180 kg ha−1 significantly enhanced yield, and the yields and harvest indexes in 2019, 2020, and 2021 were consistently R2 > R3 > R1. The findings suggested that the effect of increasing N dose (60 kg ha−1) was more considerable than that of N topdressing ratio, N3R2 (within the range of N application in this experiment) was more conducive to improving canopy photosynthesis, yield, and harvest index, and R3 was more conducive to increasing grain protein content, wet gluten, and sedimentation value.

Ongoing increases in atmospheric carbon dioxide (CO2) are expected to stimulate biomass and yield of plants possessing the C3 photosynthetic pathway; however, the extent of stimulation is likely to vary both intra- and inter-species specifically. Meta-analytic approaches can be applied to decrease variation and uncertainty by delineating and characterizing variation, allowing results to be used in modeling plant responses to elevated [CO2]. However, the use of meta-analysis in this effort could be limited by missing measures of variance, including standard deviations (SDs) of the compiled dataset. Here, we examined whether there were differences in effect sizes of elevated [CO2] on plant growth using various weighting and imputation approaches. Our results showed that the efficacy of different weighting functions and data interpolation methods on meta-analysis outcomes depended on the SDs provided by the studies. Comparing different methodologies for [CO2] fumigation as a case study, if the ratio of missing SD was low, the overall trend of effect values and 95% confidence interval (CI) were not changed. For datasets of greenhouse and growth chamber [CO2] methodologies, which had a high ratio of missing SDs, effect sizes and 95% confidence intervals using different weighing and imputation methods were influenced relative to that of the raw dataset, with reduced effect sizes and broader CI. Overall these results suggest that application of meta-analysis to discern general biological responses could be influenced by the number of missing SDs. As such, efforts should be made to check the proportion of missing SDs of the compiled dataset and if necessary, to apply various weighting functions and imputation methods to fully discern meta-analysis implications. Our findings could improve the assessment of methodological choices for future [CO2] experimentation and discerning long-term trends for agricultural productivity and food security.

Evaluating the impact of climate change factors, especially temperature and carbon dioxide (CO2), on rice yield is essential to ensure future food security. Because of the wide biogeographical distribution of rice, such evaluations are conducted exclusively through modeling efforts. However, geographical forecasts could, potentially, be improved by the inclusion of field-based data on projected increases in temperature and CO2 concentration from a given rice-growing region. In this study, the latest version of the ORYZA (v3) crop model was evaluated with additional yield data obtained from a temperature-controlled free-air CO2 enrichment system (T-FACE) in Southeastern China. ORYZA (v3) results were then evaluated in the context of phase five of the Coupled Model Intercomparison Project (CMIP5) for representative concentration pathways (RCP) 4.5 and RCP 8.5 using five global change models (GCMs). Our findings indicate that climate change, i.e., inclusion of CO2 and temperature effects, decreased mean rice yield by 3.5%, and 9.4% for RCP 4.5; and by 10.5 and 47.9% for RCP 8.5 for the scenarios in the 2050s and 2080s, respectively. The CO2 fertilizer effect partially compensated but did not offset the negative impacts of rising temperature on rice yields. Warmer temperatures were the primary factor that influenced yield by adversely affecting the spikelet fertility factor and spikelet number. Overall, climate change would have positive effects on rice yields until the middle-century in Southeastern China but negative effects were noted by the end of the century. These results may be of interest for informing policy makers and developing appropriate strategies to improve future rice productivity for this region of China.
