RESUMO

Heavy metal contamination of soil commonly accompanies problems around gold mine tailings ponds. Fully investigating the distribution characteristics of heavy metals and the survival strategies of dominant plants in contaminated soils is crucial for effective pollution management and remediation. This study aims to investigate the contamination characteristics, sources of heavy metals (As, Cd, Pb, Hg, Cu, Zn, Cr, and Ni) in soils around gold mine tailings ponds areas (JHH and WZ) and to clarify the form distribution of heavy metals (As, Cd, Pb, Hg) in contaminated plots as well as their accumulation and translocation in native dominant plants. The results of the study showed that the concentrations of As, Pb, Cd, Cu, and Zn in soil exceeded the national limits at parts of the sampling sites in both study areas. The Nemerow pollution index showed that both study areas reached extreme high pollution levels. Spatial analysis showed that the main areas of contamination were concentrated around metallurgical plants and tailings ponds, with Cd exhibiting the most extensive area of contamination. In the JHH, As (74%), Cd (66%), Pb (77%), Zn (47%) were mainly from tailings releases, and Cu (52%) and Hg (51%) were mainly from gold ore smelting. In the WZ, As (42%), Cd (41%), Pb (73%), Cu (47%), and Zn (41%) were mainly from tailings releases. As, Cd, Pb, and Hg were mostly present in the residue state, and the proportion of water-soluble, ion-exchangeable, and carbonate-bound forms of Cd (19.93%) was significantly higher than that of other heavy metals. Artemisia L. and Amaranthus L. are the primary dominating plants, which exhibited superior accumulation of Cd compared to As, Pb, and Hg, and Artemisia L. demonstrated a robust translocation capacity for As, Pb, and Hg. Compared to the concentrations of other forms of soil heavy metals, the heavy metal content in Artemisia L correlates significantly better with the total soil heavy metal concentration. These results offer additional systematic data support and a deeper theoretical foundation to bolster pollution-control and ecological remediation efforts in mining areas.

Currently, understanding the structure and function of the microbial community is the key step in artificially constructing microbial communities to control soil heavy metal pollution. Abundant/rare microbial communities play different roles in different levels of concentrations. However, the correlation between heavy metals and rare/abundant subgroups is poorly understood. In this study, we used a metagenomics approach to comprehensively investigate the evolutionary changes in microbial diversity, structure, and function under different heavy metal concentration stress in soils surrounding gold tailings. The results show that the main pollutants were Pb, As, and Zn. Indigenous microorganisms have different responses to heavy metal concentrations. Bacteria are the main components of indigenous microorganisms, mainly including Actinobacteria, Proteobacteria, Chloroflexi, and Acidobacteria. With the increase of heavy metal pollution, the relative abundance of Proteobacteria increased, and that of Actinobacteria decreased. Archaea was significantly inhibited by heavy metal stress and was more sensitive to heavy metal concentration. The response of fungi to heavy metal concentration was not obvious. The results of KEGG pathways showed that carbon fixation was inhibited with increasing heavy metal concentrations, while nitrogen metabolism was in contrast. Abundant subcommunity had a greater correlation mainly with metal resistance mechanisms, and rare subcommunity plays a key role for soil nutrient cycling such as N, S cycling in soils contaminated. Overall, this study provides a comprehensive analysis of the effects of heavy metal stress at different concentrations on microorganisms in farmland around gold tailings and reveals the relationship between heavy metals on KEGG pathways.

A quinoline-degrading strain, C2, which could completely degrade 250 mg/L of quinoline within 24 h, was isolated from coking wastewater. Strain C2 was identified as Ochrobactrum sp. on the basis of 16S rDNA sequence analysis According to 16S rDNA gene sequence analysis, Strain C2 was identified as Ochrobactrum sp. Strain C2 could utilize quinoline as the sole carbon sources and nitrogen sources to grow and degrade quinoline well under acidic conditions. The optimum inoculum concentration, temperature and shaking speed for quinoline degradation were 10%, 30 °C and 150 r/min, respectively. The degradation of quinoline at low concentration by the strain followed the first-order kinetic model. The growth process of strain C2 was more consistent with the Haldane model than the Monod model, and the kinetic parameters were: Vmax = 0.08 h-1, Ks = 131.5 mg/L, Ki = 183.1 mg/L. Compared with suspended strains, strain C2 immobilized by sodium alginate had better degradation efficiency of quinoline and COD. The metabolic pathway of quinoline by Strain C2 was tentatively proposed, quinoline was firstly converted into 2(1H) quinolone, then the benzene ring was opened with the action of catechol 1,2-dioxygenase and subsequently transformed into benzaldehyde, 2-pentanone, hydroxyphenyl propionic acid and others.

The toxicity tests of nineteen commonly used pesticides were carried out to compare the sensitivity differences between predatory mite Neoseiulus cucumeris and its prey Tetranychus cinnabarinus by a "leaf spray method" in laboratory microcosms. For two avermectins, emamectin benzoate and abamectin, exhibited high bioactivity against T. cinnabarinusf with LR50 values of 0.04 and 0.05 g a.i./ha, respectively, but these two insecticides showed the opposite toxic effects to N. cucumeris. These two agents showed strong selectivity for the two test species with Selective Toxicity Rate (STR) values of 950 and 620, respectively. However, for five neonicotinoids, the LR50s of dinotefuran, thiamethoxam, imidacloprid, and acetamiprid were all greater than the recommended rates in the field except for clothianidin, and they showed no obvious toxicity difference to the two species with STR values ranging from 0.58 to 2.00. For two organophosphates, malathion is more toxic to N. cucumeris than T. cinnabarinus, however, dimethoate showed a higher toxic effect on T. cinnabarinus. In addition, the toxicity of four pyrethroids, bifenthrin, deltamethrin, cyhalothrin and gamma-cyhalothrin to N. cucumeris was higher than that of T. cinnabarinus, except for alpha-cypermethrin. For five acaricides, spirodiclofen, spirotetramat and pyridaben had no obvious selectivity to the two organisms, while diafenthiuron and chlorfenapyr were found to be highly toxic to T. cinnabarinus than N. cucumeris with STR values of 14.2 and 68.5, respectively. Thus, some pesticides above-mentioned like emamectin benzoate, abamectin, diafenthiuron and chlorfenapyr exhibited potential to be used in the management programs of T. cinnabarinus, especially in organically based production systems where there are fewer chemical control measures available, which need to combine with natural enemies to achieve the best control effect.

The purpose of this study was to evaluate the effects of three toxic ß-glucosides, phlorizin, santonin, and amygdalin, on carbohydrate metabolism in the cotton bollworm, Helicoverpa armigera (Hübner), when diets mixed with ß-glucosides were fed to third-instar larvae. The growth of the larvae was significantly inhibited by exposure to santonin after 96 hr but not obviously affected by phlorizin and amygdalin. The midgut trehalase activities were only 51.7%, 32%, and 42.5% of that of the control after treatment with phlorizin, santonin and amygdalin at 2 mg/ml, respectively. In the hemolymph and fat body, the amount of trehalose decreased in all cases. However, the effects of santonin on the alteration of the glycogen and glucose levels as well as the activities of glycogen phosphorylase, were different than those of the other two ß-glucosides. It appears that the three ß-glucosides have different influences on the carbohydrate metabolism of cotton bollworm.

OBJECTIVE: To study the clinical effects of electrical stimulation therapy on lumbar disc herniation-induced sciatica and its influence on peripheral reactive oxygen species (ROS) level. METHODS: 100 patients with lumbar disc herniation-induced sciatica were selected, and were randomly divided into the control and research group. The control group was treated with traction and other basic therapies, while the research group was treated with electrical stimulation. The pain degrees, peripheral ROS levels and clinical effects prior to treatment and at 4 weeks after treatment were examined. RESULTS: The total cure-remarkable-effectiveness rate of patients in research group was higher than that in control group (p<0.05). Before treatment, the pain rating index (PRI), present pain intensity (PPI) and visual analogue scale (VAS) score had no statistically significant differences between the two groups. After treatment, PRI, PPI and VAS scores in the two groups were lower than those prior to treatment; these indexes in research group were lower than those in control group, and the differences were statistically significant (p<0.05). After treatment, the peripheral ROS levels in the two groups were lower than those before treatment; it was lower in research group than that in control group (p<0.05). CONCLUSION: Electrical stimulation has a significant effect in the treatment of lumbar disc herniation-induced sciatica, which can effectively reduce the pain, alleviate the clinical symptoms and signs of patients, regulate the peripheral ROS level, and prevent the oxidative damage of myocardial tissues.

The seven-spotted ladybird beetle, Coccinella septempunctata L., as a dominant predator of aphids, has played a crucial role in integrated pest management (IPM) strategies in agricultural ecosystems. To study the risk of insecticides to C. septempunctata, the neonicotinoid clothianidin was selected for evaluation of its influence on C. septempunctata at lethal and sublethal doses. The LR50 (application rate causing 50% mortality) in the exposed larvae decreased from 19.94 to 5.91 g a.i. ha-1, and the daily HQ (hazard quotient) values increased from 3.00 to 10.15, indicating potential intoxication risks. We also determined NOERs (No Observed Effect application Rates) of clothianidin on the total developmental time (10 g a.i. ha-1), survival (2.5 g a.i. ha-1) and pupation (5 g a.i. ha-1). Moreover, clothianidin at a NOER of 2.5 g a.i. ha-1 did not profoundly affect adult emergence, fecundity or egg hatchability. The total effect (E) assessment also showed that clothianidin at 2.5 g a.i. ha-1 was slightly harmful to C. septempunctata. These results suggested that clothianidin would impair C. septempunctata when applied at over 2.5 g a.i. ha-1 in the field. Conservation of this biological control agent in agricultural ecosystems thus requires further measures to decrease the applied dosages of clothianidin.

The parasitoid Trichogramma species are indispensable natural enemies of many lepidopterans and it plays an important role in integrated pest management (IPM) programs throughout the world. Laboratory studies were conducted to compare the susceptibility of three Trichogramma egg parasitoid species to ten common insecticides and three herbicides. The adults of Trichogramma dendrolimi, T. chilonis, and T. ostriniae were exposed to the above-mentioned pesticides by a glass-vial residue method. Among the four neonicotinoids, dinotefuran and thiamethoxam exhibited extremely toxic effects on the Trichogramma dendrolimi and T. chilonis, with Risk Quotient (RQ) values ranging from 1471.2 to 5492.5. However, these two neonicotinoids have a relatively low toxicity to T. ostriniae, with RQ values 433.6 and 915.4, respectively. In addition, Imidacloprid and acetamiprid were slightly to moderately toxic to all the tested parasitic wasps and their RQ values are less than 500. For pyrethroids, all the selected compounds were slightly to moderately toxic to three Trichogramma species except that cyhalothrin was dangerously toxic to T. dendrolimi and T. chilonis, with RQ values 2567.6 and 3950.4. Among the three herbicides tested, pendimethalin, butralin and napropamid were slightly to moderately toxic to egg parasitoids, with all RQ values below 1000. For two avermectins, abamectin were slightly to moderately toxic to all three wasps with RQ values 635.6, 148.3 and 254.2, respectively. However, emamectin benzoate was found to be safe for the parasitoids. Furthermore, T. dendrolimi showed higher sensitivity than T. chilonis and T. ostriniae to the pesticides based on the comparison of LR50 (application rate causing 50% mortality) values. The present results provide informative data for implementing biological and chemical control strategies in integrated pest management.

Amblyseius cucumeris (Oudemans) is a beneficial non-target arthropod (NTA) and a key predator of tetranychid mites in integrated pest management (IPM) programs across China. Evaluating the toxic effects of insecticides on such predatory mites is essential for the success and development of IPM. We tested six insecticides to determine the risk of neonicotinoid insecticide toxicity to predatory mites, using the 'open glass plate method' and adult female A. cucumeris in a "worst case laboratory exposure" scenario. A 48-h toxicity test was performed using the hazard quotient (HQ) approach to evaluate the risk of each insecticide. The LR50 values (application rate that caused 50% mortality) of acetamiprid, thiamethoxam, imidacloprid, and dinotefuran were 76.4, 104.5, 84.9, and 224.6 g active ingredient (a.i.) ha-1, respectively, with in-field HQ values of 0.40, 1.28, 0.49, and 0.82, respectively. The HQ values were lower than the trigger value of 2, and were consistent with off-field values. The risks of the four neonicotinoid insecticides to adult female A. cucumeris were acceptable in two exposure scenarios in field and off field. The 48-h LR50 values for bifenthrin and malathion were 0.008 and 0.062 g. a.i. ha-1, respectively, which were much lower than the recommended field application rates. The HQ values were much higher than the trigger values for both in- and off-field, indicating that the risks of these two insecticides were unacceptable. Bifenthrin and malathion posed an extremely high risk to the test species, and their use should be restricted to reduce risks to the field with augmentative releases of A. cucumeris.

Trehalase is an indispensable component of insect hemolymph that plays important role in energy metabolism and stress resistance. In this study, we cloned and expressed the gene encoding soluble trehalase (HaTreh-1) of Helicoverpa armigera (cotton bollworm) and characterized the enzyme. HaTreh-1 had a full-length open reading frame encoding a protein of 571 amino acids. Sequence comparison indicated that HaTreh-1 was similar to some known insect trehalases. Two essential active sites (D321 and E519) and three essential residues (R168, R221, and R286) were conserved in HaTreh-1. The recombinant trehalase was expressed in Escherichia coli and purified by nickel exchange chromatography. Molecular weight of the recombinant protein was about 71 kDa, and the optimum HaTreh-1 enzyme activity is at 55°C with pH 6.0. Enzymatic assays showed a Km value of 72.8 mmol/liter and a Vmax value of 0.608 mmol/(liter·min). Inhibition assays in vitro indicated that castanospermine, a polyhydroxylated alkaloid, was an effective competitive inhibitor of trehalase with a Ki value of 6.7 µmol/liter. The inhibitor action of castanospermine was linked to its modification effect on trehalase structure. The circular dichroism spectrum showed that the percentage of α-helix increased under the presence of castanospermine. Results of our study will aid in developing effective trehalase inhibitors for controlling H. armigera in the future.

BACKGROUND The aim of this study was to investigate the effects of sleep duration and bedtime on sperm health, and the possible mechanism involved. MATERIAL AND METHODS We randomly divided 981 healthy Chinese men into groups according to research-set bedtimes (A=8-10 PM, B=after 10 PM, and C=after midnight) and sleep durations: group 1=<6.0 h (short), group 2=7.0-8.0 h (average), and group 3=>9.0 h (long). Sperm morphology, count, survival, and motility were examined according to sleep patterns. Antisperm antibody (ASA) production in semen was determined. RESULTS Sperm counts and their survival rates were lower in the short sleepers as compared to others within each group (all P<0.01). The lower counts and survival rates were observed in different bedtimes, with significant differences found between measurements of C1 vs. A1 and C2 vs. A2 or B2 (all P<0.05 or 0.01). Semen motility was lower in the short sleepers as compared to the average and long sleepers (all P<0.01). There were differences in the bedtime-related results between measurements of C1 vs. A1 or B1 (P<0.05 or 0.01). Additionally, the population proportion for the ASA-positive participates and incidence of the ASA-expressed population obviously increased in the short sleepers as compared to others within each group (all P<0.05). CONCLUSIONS Short and long sleep durations and late bedtime were associated with impaired sperm health in the study cohort, partly through increasing ASA production in the semen.

Plants have a chemically heterogeneous lipophilic layer, the cuticle, which protects them from biotic and abiotic stresses. The mechanisms that regulate cuticle development are poorly understood. We identified a rice (Oryza sativa) dominant curly leaf mutant, curly flag leaf1 (cfl1), and cloned CFL1, which encodes a WW domain protein. We overexpressed both rice and Arabidopsis CFL1 in Arabidopsis thaliana; these transgenic plants showed severely impaired cuticle development, similar to that in cfl1 rice. Reduced expression of At CFL1 resulted in reinforcement of cuticle structure. At CFL1 was predominantly expressed in specialized epidermal cells and in regions where dehiscence and abscission occur. Biochemical evidence showed that At CFL1 interacts with HDG1, a class IV homeodomain-leucine zipper transcription factor. Suppression of HDG1 function resulted in similar defective cuticle phenotypes in wild-type Arabidopsis but much alleviated phenotypes in At cfl1-1 mutants. The expression of two cuticle development-associated genes, BDG and FDH, was downregulated in At CFL1 overexpressor and HDG1 suppression plants. HDG1 binds to the cis-element L1 box, which exists in the regulatory regions of BDG and FDH. Our results suggest that rice and Arabidopsis CFL1 negatively regulate cuticle development by affecting the function of HDG1, which regulates the downstream genes BDG and FDH.

BACKGROUND/AIMS: Diabetes is well-known to influence endothelial function. Endothelial function and blood flow regulation might be different in diabetic and non-diabetic pregnancy. However, the impact of umbilical blood flow regulation in gestational diabetes on fetal development is unknown so far. METHODS: In a prospective birth cohort study, we analyzed the association of the umbilical artery Doppler indices (pulsatility index, resistance index and systolic/diastolic ratio) and fetal size measures (biparietal diameter, head circumference, abdominal circumference, femur length and birth weight) in 519 non-gestational diabetes mellitus pregnancies (controls) and 226 gestational diabetes mellitus pregnancies in middle (day 160.32 ±16.29 of gestation) and late (day 268.12 ±13.04 of gestation) pregnancy. RESULTS: Multiple regression analysis considering confounding factors (gestational day of ultrasound examination, offspring sex, maternal body mess index before pregnancy, maternal age at delivery, maternal body weight at delivery and maternal hypertension) showed that umbilical artery Doppler indices (pulsatility index, resistance index and systolic/diastolic ratio) were associated with fetal head circumference and femur length in middle gestational diabetes mellitus pregnancy but not in non-gestational diabetes mellitus pregnancy. Head circumference, biparietal diameter, abdominal circumference and femur length in mid gestation were smaller in fetus of gestational diabetes mellitus pregnancy versus non-gestational diabetes mellitus pregnancy. In contrast to non-gestational diabetes mellitus pregnancy in late gestation, umbilical artery Doppler indices in gestational diabetes mellitus pregnancy were not associated with ultrasound measures of fetal growth. Birth weight was slightly increased in gestational diabetes mellitus pregnancy as compared to non-gestational diabetes mellitus pregnancy. CONCLUSIONS: The impact of umbilical blood flow on fetal growth is time dependent in human gestational diabetes mellitus and non-gestational diabetes mellitus pregnancy. In gestational diabetes mellitus pregnancy umbilical blood flow is critical for organ development in much earlier stages of pregnancy as compared to non-gestational diabetes mellitus pregnancy. The physiological and molecular pathways why there is a catch up growth in later times of gestational diabetes mellitus pregnancy resulting in larger gestational diabetes mellitus babies at birth needs to be addressed in further studies.

Long-term effects of a single application of imidacloprid on ladybird beetle, Coccinella septempunctata L., were studied in indoor laboratory microcosms, starting with the 2nd instar larvae of C. septempunctata but covering the full life cycle. The microcosms comprised enclosures containing a pot with soil planted with broad bean plants and black bean aphid, Aphis craccivora Koch, as food. Exposure doses (0.85-13.66g a.i. ha(-1)) in the long-term microcosm experiment were based on a preliminary short-term (72h) toxicity test with 2nd instar larvae. The measurement endpoints used to calculate NOERs (No Observed Effect application Rates) included development time, hatching, pupation, adult emergence, survival and number of eggs produced. Furthermore, for these endpoints ER50 (application rate causing 50 percent effect) and LR50 (application rate causing 50 percent mortality) values were calculated when possible. The single imidacloprid application affected survival (lowest LR50 4.07g a.i. ha(-1); NOER 3.42g a.i. ha(-1)), egg production (ER50 26.63g a.i. ha(-1)) and egg hatching (NOER 6.83g a.i. ha(-1)). Statistically significant treatment-related effects on the whole development duration, pupation and adult emergence could not be demonstrated (NOER≥13.66g a.i. ha(-1)). The lowest L(E)R50 values and NOERs derived from the laboratory microcosm test with C. septempunctata are lower than the reported field application rates of imidacloprid (15-60g a.i. ha(-1)) in cotton cultivation in China, suggesting potential risks to beneficial arthropods.

Soil heavy metal contamination is an essential challenge in ecological and environmental management, especially for acidic soils. Microbially induced carbonate precipitation (MICP) is an effective and environmentally friendly remediation technology for heavy metal contaminated sites, and one of the key factors for its realization lies in the microorganisms. In this study, Lysinibacillus capsici TSBLM was isolated from heavy metal contaminated soil around a gold mine, and inferred to be a novel ureolytic bacteria after phylogenomic inference and genome characterization. The urease of L. capsici TSBLM was analyzed by genetic analysis and molecular docking, and further applied this bacteria to the remediation of Cu and Pb in solution and acidic soils to investigate its biomineralization mechanism and practical application. The results revealed L. capsici TSBLM possessed a comprehensive urease gene cluster ureABCEFGD, and the encoded urease docked with urea at the lowest binding energy site (ΔG = -3.43 kcal/mol) connected to three amino acids threonine, aspartic, and alanine. The urease of L. capsici TSBLM is synthesized intracellularly but mainly functions extracellularly. L. capsici TSBLM removes Cu/Pb from the solution by generating heavy metal carbonates or co-precipitating with CaCO3 vaterite. For acidic heavy metal-contaminated soil, the carbonate-bound states of Cu and Pb increased significantly from 7 % to 16 % and from 23 % to 35 % after 30 days by L. capsici TSBLM. Soil pH improved additionally. L. capsici TSBLM maintained the dominant status in the remediated soil after 30 days, demonstrating good environmental adaptability and curing persistence. The results provided new strain resources and practical application references for the remediation of acidic heavy metal contaminated soil based on MICP.

In this study, we used soils with different pollution and nutrient levels (non-polluted S1, highly polluted low-nutrient S2, and highly polluted high nutrient S3) around the gold mine tailing ponds, and combined with metabolic limitation modeling and macro-genomics approaches, aiming to investigate the relationship between soil microbial composition and soil eco-chemometrics characteristics under heavy metal stress. The results showed that heavy pollution resulted in reduced SOC, TN, microbial biomass, and with C- and P- acquisition (BG, CBH, ALP) as well as nitrogen limitation of soil microbial metabolism in soils (S2, S3). Further analysis by macrogenomics showed that heavy metal contamination led to an increase in α-microbial diversity and altered the composition of microbial communities in the soil. The cycling of C, N, and P nutrients was altered by affecting the relative abundance of Anaeromyxobacter, Steroidobacter, Bradyrhizobium, Acidobacterium, Limnochorda (predominantly in the Ascomycetes and Acidobacteria phyla), with the most pronounced effect on the composition of microorganisms synthesizing C-acquiring enzymes, and heavy metals and pH were the main influences on ecological stoichiometry. The results of this study are useful for understanding the sustainability of ecological remediation in heavy metal contaminated areas and for developing ecological restoration strategies.

A newly isolated ureolytic bacteria, Brucella intermedia TSBOI, exhibited microbially induced calcite precipitation (MICP) which is a promising technique for the remediation of heavy metals in polluted environments. Brucella intermedia TSBOI achieved 90-100% removal of 1 mmol/L Cu2+/Pb2+/Zn2+ within 72 h. A distinctive feature lies in B. intermedia TSBOI's capacity for the transport and hydrolysis of urea, considered to be critical for its strong urease activity. This study explored the mechanisms of this capacity at the genetic, molecular and protein levels through complete genome sequencing, molecular docking and enzymatic reaction kinetics. The results revealed that, for urea hydrolysis, B. intermedia TSBOI exhibited a comprehensive urease gene cluster, with the key gene ureC demonstrating an absolute expression level approximating to 4 × 104 copies/RNA ng under optimal conditions. Results also confirmed the strong spontaneous, energy-independent binding ability of it's urease to urea, with the lowest Gibbs free energy binding site linking to the three amino acids, alanine, asparagine and serine. The urea transport gene yut presented and expressed, with the absolute expression enhanced in response to increasing urea concentrations. The significant positive correlation between ureC/yut expression levels and urease activity provided a theoretical basis for B. intermedia TSBOI's heavy metal bioremediation potential. ENVIRONMENTAL IMPLICATION: Heavy metals (Cu, Pb and Zn) were studied in this study. Heavy metals are hazardous due to their toxicity, persistence, and ability to bioaccumulate in living organisms. They can cause severe health issues, harm ecosystems, and contaminate air, water, and soil. A novel ureolytic bacteria, Brucella intermedia TSBOI, exhibited microbially induced carbonate precipitation capability was isolated which removed 90-100% of 1 mmol/L Cu2+/Pb2+/Zn2+ within 72 h. Its advantages in urea hydrolysis and transport facilitate the remediation of actual heavy metal contaminated environments.

In this study, a sulfate-reducing bacterium, Clostridium thiosulfatireducens (CT) was reported and the performance and removal mechanism of Cr(VI) and Cd(II) removal were investigated. It is noteworthy that the dsrAB gene is absent in this strain, but the strain is capable of producing sulfide. The conversion rate of Cr(VI) by CT was 84.24 % at a concentration of 25 mg/L, and the conversion rate of Cd(II) was 94.19 % at a concentration of 28 mg/L. The complete genome is 6,106,624 bp and the genome consisted of a single chromosome. The GC content of the chromosomes was 29.65 %. The mechanism of heavy metal removal by CT bacteria mainly includes biosorption, electron transfer and redox, with reduction combined with S2- precipitation as the main pathway. The product characterization results showed that the formation of mainly ionic crystals and precipitates (CdS, Cd(OH)2, Cr(OH)3, Cr2O3) after adsorption. Genome-wide techniques have shown that the clearance of Cr(VI) and Cd(II) by CT is largely dependent on sulfate transport, sulfur metabolism, and energy metabolism to some extent. In addition, genes related to ATP binding, electron carrier activity, transporter protein genes, and DNA repair are also important factors to improve the heavy metal resistance and transformation ability of CT strains. Both the Fe-S cycle and the ROS-resistant system can enhance the electron transfer activity and thus slow down the damage of heavy metals to microorganisms. This study fills the gap in the understanding of the basic properties and heavy metal transformation mechanism of CT.

To achieve efficient resource utilization of fly ash and multi-source organic waste, a composting experiment was carried out to investigate the effects of fly ash on co-aerobic composting using kitchens, chicken manure, and sawdust (15:5:2). The effects of different application doses (5 % and 10 %, calculated in total wet weight of organic solid waste) of fly ash on physical and chemical properties, nutrient elements, and bacterial community structure during co-composting were evaluated. The results showed that the addition dose of 5 % and 10 % fly ash significantly increased the highest temperature (56.6 ℃ and 56.9 ℃) and extended the thermophilic period to nine days. Compared with that in the control, the total nutrient content of compost products in the treatments of 5 % FA and 10 % FA was increased by 4.09 % and 13.55 %, respectively. The bacterial community structure changed greatly throughout the composting, and the bacterial diversity of all treatments increased obviously. In the initial stage of composting, Proteobacteria was the dominant phylum of bacteria, with a relative abundance ranging from 35.26 % to 39.40 %. In the thermophilic period, Firmicutes dominated; its relative abundance peaked at 52.46 % in the 5 % FA treatment and 67.72 % in the 10 % FA treatment. Bacillus and Thermobifida were the predominant groups in the thermophilic period of composting. The relative abundance of Bacillus and Thermobifida in the 5 % FA and 10 % FA treatments were 33.41 % and 62.89 %(Bacillus) and 33.06 % and 12.23 %(Thermobifida), respectively. The results of the redundancy analysis (RDA) revealed that different physicochemical indicators had varying degrees of influence on bacteria, with organic matter, pH, available phosphorus, and available potassium being the main environmental factors influencing bacterial community structure. In summary, the addition of fly ash promoted the harmlessness and maturation of co- aerobic composting of urban multi-source organic waste, while optimizing microbial community structure and improving the quality and efficiency of composting.

This study reports the simultaneous oxidation of As(III) and reduction of the Cr(VI) strain Ensifer adhaerens M8 screened from soils around abandoned gold tailings contaminated with highly complex metals (loids). Physiological, biochemical, and genomic techniques were used to explore the mechanism. The strain M8 could simultaneously oxidize 1 mM As(III) and reduce 45.3 % 0.1 mM Cr(VI) in 16 h, and the Cr(VI) reduction rate was increased by 5.8 % compared with the addition of Cr(VI) alone. Cellular debris was the main site of M8 arsenic oxidation. Chromium reduction was dominated by the reduction of extracellular hexavalent chromium (23.80-35.67 %). The genome of M8 included one chromosome and four plasmids, and a comparison of the genomes showed that M8 had two more plasmids than strains of the same genus, which may be related to strong environmental adaptations. M8 had 10 heavy metal resistance genes (HMRs), and plasmid D had a complete cluster of arsenic resistance-oxidation-transport genes (arsOHBCCR-aioSR-aioBA-cytCmoeA-phoBBU-PstBACS-phnCDEE). The genes involved in Cr(VI) detoxification include DNA repair (RecG, ruvABC, and UvrD), Cr(VI) transport (chrA, TonB, and CysAPTW) and Cr(VI) reduction. In summary, this study provides a molecular basis for As (III) and Cr (VI) remediation.
