RESUMO

Designing smart textiles for personal thermal management (PTM) is an effective strategy for thermoregulation and energy saving. However, the manufacture of versatile high-performance thermal management textiles for complex real-world environments remains a challenge due to the limitations of functional integration, material properties, and preparation procedures. In this study, an aramid fabric based on in situ anchored copper sulfide nanostructure is developed. The textile with excellent solar and Joule heating properties can effectively keep the body warm even at low energy inputs. Meanwhile, the reduced infrared emissivity of the textile decreases the thermal radiation losses and helps to maintain a constant body temperature. Impressively, the textile integrates superb electromagnetic shielding, near-complete UV protection properties, and ideal resistance to fire and bacteria. This work provides a simple strategy for fabricating multi-functional integrated wearable devices with flexibility and breathability, which is highly promising in versatile PTM applications.

RESUMO

Methods: Participants underwent respiratory muscle training for 24 weeks. The main results were changes in respiratory muscle strength and pulmonary function indices (forced expiratory volume in 1 s (FEV1), forced vital capacity (FVC), FEV1/FVC, peak expiratory flow rate (PEF), forced expiratory flow 25-75% (FEF25-75%), and maximal midexpiratory flow 75/25 (MMEF75/25)) before, 12 weeks after, and 24 weeks after the intervention. The secondary outcomes were changes in the exercise load and work rate, exercise work, Leicester Cough Questionnaire (LCQ) scale, and Fatigue Severity Scale (FSS). Results: Compared with before the intervention, after 24 weeks of respiratory muscle training, the maximal inspiratory pressure (MIP) and maximal expiratory pressure (MEP) were significantly enhanced (P < 0.05), while FVC, FEV1, and PEF were significantly increased (P < 0.01). FEF25-75 and MMEF75/25 values showed significant improvement compared to those before training (P < 0.05). The exercise loading, work, and exercise work rate of expiratory muscle training were significantly improved compared to those before intervention (P < 0.05). The LCQ score increased significantly (P < 0.001), and the FSS score decreased significantly (P < 0.001). Conclusion: Incremental load respiratory muscle training effectively improved children's lung function over the long term, improved the strength of their inspiratory and expiratory muscles, and improved their quality of life.

In this work, a special multilayer structure consisting of polylactic acid (PLA) and a co-continuous PLA/polycaprolactone (PCL)/multiwalled carbon nanotube (MWCNT) (ALM) composite with a double-percolated conductive network was fabricated via layer-assembly coextrusion. It was revealed that PLA domains located at the layer interface could serve as rivets properly linking adjacent layers. Such a nacre-like structure with alternately stacked rigid PLA and flexible ALM increased the fracture strain to 354.4%, nearly quadruple that of the PLA/PCL/MWCNT conventional blending composite with the same composition, while maintaining an excellent strength above 46.0 MPa. In addition, the multilayer composites showed a special frequency-selective electromagnetic interference (EMI) shielding performance, with tunable shielding peak positions controlled by the layer number. Their maximum EMI shielding effectiveness almost contributed by absorption loss could reach 49.8 dB, which originated from two aspects: one was the high electrical conductivity offered by the double-percolated distribution of MWCNTs, and the other was the multiple wave attenuation effect that occurred at the interfaces between PLA and ALM layers and the blend interfaces in ALM layers. This effort paves a new way for developing composites with outstanding mechanical and EMI shielding properties that can be extended to other polymeric composite systems.

Resistance to osimertinib represents a significant challenge for the successful treatment of non-small cell lung cancer (NSCLC) harboring activating mutations in epidermal growth factor receptor (EGFR). N6-methyladenosine (m6A) on mRNAs is critical for various biological processes, yet whether m6A regulates osimertinib resistance of NSCLC remains unknown. In this study, we demonstrated that developing osimertinib-resistant phenotypes depends on m6A reduction resulting from downexpression of m6A methyltransferase METTL14 in EGFR-mutant NSCLCs. Both in vitro and in vivo assay showed that specific knockdown of METTL14 was sufficient to confer osimertinib resistance and elevated expression of METTL14 rescued the efficacy of osimertinib in the resistant NSCLC cells. Mechanistically, METTL14 promoted m6A methylation of pro-apoptotic Bim mRNA and increased Bim mRNA stability and expression, resulting in activating the Bim-dependent pro-apoptotic signaling and thereby promoting osimertinib-induced cell apoptosis. Analysis of clinical samples revealed that decreased expression of METTL14 was observed in osimertinib-resistant NSCLC tissues and significantly associated with a poor prognosis. In conclusion, our study reveals a novel regulatory mechanism by which METTL14-mediated m6A methylation of Bim mRNA inhibited osimertinib resistance of NSCLC cells. It offers more evidences for the involvement of m6A modification in regulation of osimertinib resistance, and provides potential therapeutic targets for novel approaches to overcome the tolerance of osimertinib and other EGFR-TKIs. Implications: This study offers more evidences for the involvement of METTL14-mediated m6A modification in regulation of osimertinib resistance, and provides potential therapeutic targets for novel approaches to overcome the tolerance of osimertinib and other EGFR-TKIs.

The high value-added medical applications surely represent the leading edge of the shape-memory materials (SMPs) field. Herein, the biomedical SMPs were easily prepared via incorporating silk fibroin (SF) into poly(propylene carbonate) (PPC) through directly melt blending. Based on the intrinsic glass transition of PPC at ∼37 °C, the blends showed a body temperature responsiveness without a complex procedure for adjusting the switching temperature. By varying the SF content, the blend exhibited tunable shape-memory effects (SME), with a first enhancing but then worsening shape recoverability and a stable and excellent shape fixity. And the blend with 3 wt% SF achieved the best SME, enabling an efficient shape reconfiguration under a 37 °C water bath. It was revealed that SF acted as physical cross-links to connect the PPC chains forming a shape-memory network, thus can well retard irreversible the chain slipping of PPC, leading to the improvement of recoverability. Moreover, the results obtained from cell compatibility testing showed the huge application potential of this material in the biomedical field. This work proposed a facile preparation strategy for developing biocompatible body heat actuated shape-memory materials.

In this work, inspired by the hierarchical architecture of nacre, we have fabricated poly(propylene carbonate) (PPC)/thermoplastic polyurethane (TPU) alternating multilayer films via layer-multiplying coextrusion. Based on the glass transition at around 37 °C of PPC, the multilayer films exhibited an outstanding body heat-responsive shape-memory effect (SME) with high shape fixation and recovery ratios (96.1 and 93.6%), much better than the conventional cocontinuous blend with the same compositions. It was revealed that the high phase continuity and abundantly two-dimensional interfaces both capable of promoting stress transferring and load distribution maximally contributed to the SME. Furthermore, the multilayer films showed a superior recovery stress storage capacity and the force generated by shape recovery allowed automatic expansion of the spiral in 37 °C water and efficient lifting of a load 880 times its weight. Different from the opacity of the blend, a high optical transparence was observed in the multilayers because of the parallel assembly of transparent PPC and TPU enabling light to directly pass through the films. Besides, the nacre-like films had layer debonding and layer stepwise breaking during stretching, resulting in a 90% increase in tensile strength, a 70% increase in elongation at break, and onefold improvement in yield stress, compared with those of the blend. Our approach paves a new way for developing bioinspired structural materials with excellent optical, mechanical, and shape-memory properties, which can be extended to different amorphous polymers and elastomers. Also, the materials presented herein have great potential in applications of biomedical devices and soft robotics.

Staphylococcus aureus is one of the representative foodborne pathogens which forms biofilm. Antibiotics are widely applied in livestock husbandry to maintain animal health and productivity, thus contribute to the dissemination of antimicrobial resistant livestock and human pathogens, and pose a significant public health threat. Effect of antibiotic pressure on S. aureus biofilm formation, as well as the mechanism, remains unclear. In this study, the regulatory mechanism of low concentration of ampicillin on S. aureus biofilm formation was elucidated. The viability and biomass of biofilm with and without 1/4 MIC ampicillin treatment for 8 h were determined by XTT and crystal violet straining assays, respectively. Transcriptomics analysis on ampicillin-induced and non-ampicillin-induced biofilms were performed by RNA-sequencing, differentially expressed genes identification and annotation, GO functional and KEGG pathway enrichment. The viability and biomass of ampicillin-induced biofilm showed dramatical increase compared to the non-ampicillin-induced biofilm. A total of 530 differentially expressed genes (DEGs) with 167 and 363 genes showing up- and down-regulation, respectively, were obtained. Upon GO functional enrichment, 183, 252, and 21 specific GO terms in biological process, molecular function and cellular component were identified, respectively. Eight KEGG pathways including "Microbial metabolism in diverse environments", "S. aureus infection", and "Monobactam biosynthesis" were significantly enriched. In addition, "beta-lactam resistance" pathway was also highly enriched. In ampicillin-induced biofilm, the significant up-regulation of genes encoding multidrug resistance efflux pump AbcA, penicillin binding proteins PBP1, PBP1a/2, and PBP3, and antimicrobial resistance proteins VraF, VraG, Dlt, and Aur indicated the positive response of S. aureus to ampicillin. The up-regulation of genes encoding surface proteins ClfB, IsdA, and SasG and genes (cap5B and cap5C) which promote the adhesion of S. aureus in ampicillin induced biofilm might explain the enhanced biofilm viability and biomass.
