6-Formylisoophiopogonone B (7a) and 8-formylophiopogonone B (7b), two natural products isolated from Ophiopogon japonicus, represent a subgroup of rare 6/8-formyl/methyl-homoisoflavonoid skeletons. Herein we report an efficient method for the synthesis of these formyl/methyl-homoisoflavonoids. The synthesized compounds were evaluated for their neuroprotective effects on the MPP+-induced SH-SY5Y cell injury model and showed marked activity. Exploration of the neuroprotective mechanisms of compound 7b led to an increased expression of autophagy marker LC3-II and down-regulation of autophagy substrate p62/SQSTM1. Molecular docking studies showed that 7b may prevent the inhibition of the classic PI3K-AKT-mTOR signaling pathway by interfering with the human HSP90AA1.

Clarifying the spatio-temporal evolution of the ecological environment quality of a watershed and its response to the natural environment and human factors are crucial for policy implementation in the ecological environment of the watershed. Using the Google earth engineï¼GEEï¼ to establish a remote sensing ecological index ï¼RSEIï¼, the spatio-temporal changes in the ecological environment quality of the Huaihe River Basin from 2002 to 2022 were evaluated combined with trend analysis, variation coefficient, and Hurst index. The main driving factors of spatial differentiation of RSEI were explored using the geographic detector. The results showed thatï¼ â In the past 21 years, RSEI of the Huaihe River Basin had generally improved, but it showed a gradual upward-downward trend. Overall, the area of poor and less poor grades decreased, the area of medium grades increased, and the area of good and excellent grades increased. The improved area accounted for 55.93%, and the degraded area accounted for 22.01%. â¡ In terms of spatial distribution, RSEI gradually deteriorated from east to west ï¼except in the northwest and southwest marginal mountainous areasï¼. The stability was better in the east and worse in the western and central areas. In the future, the ecological quality change in the basin was prone to be anti-sustainable and mainly improved. ⢠Factor detection results showed that the spatial differentiation of RSEI in the basin was mainly driven by vegetation factors, followed by altitude. The interaction between two factors enhanced the driving force for RSEI spatial differentiation, in which the interaction between vegetation factor and elevation had the strongest driving force for RSEI spatial differentiation, reaching 86.3%.

Winter wheat is one of the main crops in Anhui Province, to analyze the spatial-temporal variations of climate-induced reduction risk is of great significance to ensure regional food security. Based on the data of winter wheat yield from 1973 to 2014 in 74 districts and counties, nine risk assessment indicators were extracted from annual series of climate-induced reduction rate, and the spatial-temporal variations of climate-induced reduction risk for winter wheat in Anhui was analyzed by using principal component analysis. The results showed that north Anhui, especially the region along Huaihe River, was the higher-risk area for climate-induced yield loss. Results of the barycenter migration model showed that the high value regions of climate-induced reduction rate of winter wheat in Anhui Province had a significant tendency of moving from north to south. In the whole province, climate-induced reduction risk of winter wheat presented a significant interdecadal variation. Furthermore, the number of districts and counties with moderate or more reduction intensity was significantly reducing since the 2000s. Results from S-mode principal component analysis and climate-induced reduction rate series showed that the climate-induced reduction risk for winter wheat in north Anhui had a downward trend and that in southern Anhui had an upward trend. Climate-induced reduction risk of winter wheat in Anhui had notable temporal and spatial variations, the interdecadal fluctuation and north-south regional difference of which should draw our attention.

BACKGROUND AND OBJECTIVES: Isoflurane is halogenated volatile ether used for inhalational anesthesia. It is widely used in clinics as an inhalational anesthetic. Neonatal hypoxic ischemia injury ensues in the immature brain that results in delayed cell death via excitotoxicity and oxidative stress. Isoflurane has shown neuroprotective properties that make a beneficial basis of using isoflurane in both cell culture and animal models, including various models of brain injury. We aimed to determine the neuroprotective effect of isoflurane on hypoxic brain injury and elucidated the underlying mechanism. METHODS: A hippocampal slice, in artificial cerebrospinal fluid with glucose and oxygen deprivation, was used as an in vitro model for brain hypoxia. The orthodromic population spike and hypoxic injury potential were recorded in the CA1 and CA3 regions. Amino acid neurotransmitters concentration in perfusion solution of hippocampal slices was measured. RESULTS: Isoflurane treatment caused delayed elimination of population spike and improved the recovery of population spike; decreased frequency of hypoxic injury potential, postponed the onset of hypoxic injury potential and increased the duration of hypoxic injury potential. Isoflurane treatment also decreased the hypoxia-induced release of amino acid neurotransmitters such as aspartate, glutamate and glycine induced by hypoxia, but the levels of γ-aminobutyric acid were elevated. Morphological studies showed that isoflurane treatment attenuated edema of pyramid neurons in the CA1 region. It also reduced apoptosis as evident by lowered expression of caspase-3 and PARP genes. CONCLUSIONS: Isoflurane showed a neuro-protective effect on hippocampal neuron injury induced by hypoxia through suppression of apoptosis.

Based on the bi-linearly interpolated meteorological reanalysis data from National Centers for Environmental Prediction, USA and by using the leaf area index data derived from the GIMMS NDVI to run the process-based Boreal Ecosystems Productivity Simulator (BEPS) model, this paper simulated and analyzed the spatiotemporal characteristics of the terrestrial ecosystem gross primary productivity (GPP) and net primary productivity (NPP) in East Asia in 2000-2005. Before regional simulating and calculating, the observation GPP data of different terrestrial ecosystem in 15 experimental stations of AsiaFlux network and the inventory measurements of NPP at 1300 sampling sites were applied to validate the BEPS GPP and NPP. The results showed that BEPS could well simulate the changes in GPP and NPP of different terrestrial ecosystems, with the R2 ranging from 0.86 to 0.99 and the root mean square error (RMSE) from 0.2 to 1.2 g C x m(-2) x d(-1). The simulated values by BEPS could explain 78% of the changes in annual NPP, and the RMSE was 118 g C x m(-2) x a(-1). In 2000-2005, the averaged total GPP and total NPP of the terrestrial ecosystems in East Asia were 21.7 and 10.5 Pg C x a(-1), respectively, and the GPP and NPP exhibited similar spatial and temporal variation patterns. During the six years, the total NPP of the terrestrial ecosystems varied from 10.2 to 10.7 Pg C x a(-1), with a coefficient of variation being 2. 2%. High NPP (above 1000 g C x m(-2) x a(-1)) occurred in the southeast island countries, while low NPP (below 30 g C x m(-2) x a(-1)) occurred in the desert area of Northwest China. The spatial patterns of NPP were mainly attributed to the differences in the climatic variables across East Asia. The NPP per capita also varied greatly among different countries, which was the highest (70217 kg C x a(-1)) in Mongolia, far higher than that (1921 kg C x a(-1)) in China, and the lowest (757 kg C x a(-1)) in India.

Resumo Justificativa e objetivos: Isoflurano é um éter volátil halogenado usado para anestesia por via inalatória. É amplamente usado na clínica como um anestésico para inalação. A lesão hipóxico-isquêmica neonatal ocorre no cérebro imaturo e resulta em morte celular tardia via excitotoxicidade e estresse oxidativo. Isoflurano mostrou ter propriedades neuroprotetoras que formam uma base benéfica para o seu uso tanto em cultura de células quanto em modelos animais, incluindo vários modelos de lesão cerebral. Nosso objetivo foi determinar o efeito neuroprotetor de isoflurano em hipóxia cerebral e elucidar o mecanismo subjacente. Métodos: Fatias de hipocampo, em fluido cerebrospinal artificial (CSFA) com glicose e privação de oxigênio, foram usadas como um modelo in vitro de hipóxia cerebral. O pico de população ortodrômica (PPO) e o potencial de lesão hipóxica (PLH) foram registrados nas regiões CA1 e CA3. A concentração de neurotransmissores de aminoácidos na solução de perfusão das fatias de hipocampo foi medida. Resultados: O tratamento com isoflurano retardou a eliminação do PPO e melhorou a recuperação do PPO; diminuiu a frequência do PLH, retardou o início do PLH e aumentou a duração do PLH. O tratamento com isoflurano também diminuiu a liberação de neurotransmissores de aminoácidos induzida pela hipóxia, como aspartato, glutamato e glicina, mas os níveis de ácido γ-aminobutírico (GABA) estavam elevados. Estudos morfológicos mostram que o tratamento de edema com isoflurano atenuou o edema de neurônios piramidais na região CA1. Também reduziu a apoptose, como mostrado pela expressão reduzida da caspase-3 e genes PARP. Conclusões: Isoflurano mostrou um efeito neuroprotetor na lesão neuronal no hipocampo induzida por hipóxia através da supressão de apoptose.
