BACKGROUND: The sympathoadrenergic system and its major effector PKA (protein kinase A) are activated to maintain cardiac output coping with physiological or pathological stressors. If and how PKA plays a role in physiological cardiac hypertrophy (PhCH) and pathological CH (PaCH) are not clear. METHODS: Transgenic mouse models expressing the PKA inhibition domain (PKAi) of PKA inhibition peptide alpha (PKIalpha)-green fluorescence protein (GFP) fusion protein (PKAi-GFP) in a cardiac-specific and inducible manner (cPKAi) were used to determine the roles of PKA in physiological CH during postnatal growth or induced by swimming, and in PaCH induced by transaortic constriction (TAC) or augmented Ca2+ influx. Kinase profiling was used to determine cPKAi specificity. Echocardiography was used to determine cardiac morphology and function. Western blotting and immunostaining were used to measure protein abundance and phosphorylation. Protein synthesis was assessed by puromycin incorporation and protein degradation by measuring protein ubiquitination and proteasome activity. Neonatal rat cardiomyocytes (NRCMs) infected with AdGFP (GFP adenovirus) or AdPKAi-GFP (PKAi-GFP adenovirus) were used to determine the effects and mechanisms of cPKAi on myocyte hypertrophy. rAAV9.PKAi-GFP was used to treat TAC mice. RESULTS: (1) cPKAi delayed postnatal cardiac growth and blunted exercise-induced PhCH; (2) PKA was activated in hearts after TAC due to activated sympathoadrenergic system, the loss of endogenous PKIα (PKA inhibition peptide α), and the stimulation by noncanonical PKA activators; (3) cPKAi ameliorated PaCH induced by TAC and increased Ca2+ influxes and blunted neonatal rat cardiomyocyte hypertrophy by isoproterenol and phenylephrine; (4) cPKAi prevented TAC-induced protein synthesis by inhibiting mTOR (mammalian target of rapamycin) signaling through reducing Akt (protein kinase B) activity, but enhancing inhibitory GSK-3α (glycogen synthase kinase-3α) and GSK-3ß signals; (5) cPKAi reduced protein degradation by the ubiquitin-proteasome system via decreasing RPN6 phosphorylation; (6) cPKAi increased the expression of antihypertrophic atrial natriuretic peptide (ANP); (7) cPKAi ameliorated established PaCH and improved animal survival. CONCLUSIONS: Cardiomyocyte PKA is a master regulator of PhCH and PaCH through regulating protein synthesis and degradation. cPKAi can be a novel approach to treat PaCH.

Harvesting freshwater from fog is one of the possible solutions to the global water scarcity crisis. Surfaces with both hydrophobic and hydrophilic regions are extensively employed for this purpose. Nevertheless, the longevity of these surfaces is still constrained by their delicate surface structures. The hydrophilic zones may become damaged or contaminated after repeated use, thereby compromising their effectiveness in fog collection. The preparation of generally applicable durable superhydrophobic coatings with self-generated Wenzel sites is reported here for long-term efficient and stable fog collection. The coatings are prepared by depositing the poly(tannic acid) coating as the primer layer on various substrates, self-assembly of trichlorovinylsilane into staggered silicone nanofilaments, and then thiol-ene click reaction with 1H,1H,2H,2H-perfluorodecanethiol. The coatings demonstrate remarkable static superhydrophobicity, robust impalement resistance, and stable self-generated Wenzel sites for water droplets. Therefore, the fog collection rate (FCR) of the coatings reaches 2.13 g cm-2 h-1 during 192 h continuous fog collection, which is triple that of bare substrate and outperforms most previous studies. Moreover, the systematic experiments and models have revealed that the key factors for achieving high FCR on superhydrophobic coatings are forming condensed droplets ≈1 mm in critical radius and a Wenzel site proportion of 0.3-0.4.

PURPOSE: Diffuse midline glioma (DMG), H3 K27M-mutant is a type of diffuse high-grade glioma that occurs in the brain midline carrying an extremely poor prognosis under the best efforts of surgery, radiation, and other therapies. For better therapy, we explored the efficacy and toxicity of a novel therapy that combines apatinib and temozolomide in DMG. METHODS: A retrospective analysis of 32 patients with DMG who underwent apatinib plus temozolomide treatment was performed. Apatinib was given 500 mg in adults, 250 mg in pediatric patients once daily. Temozolomide was administered at 200 mg/m2/d according to the standard 5/28 days regimen. The main clinical data included basic information of patients, radiological and pathological characteristics of tumors, treatment, adverse reactions, prognosis. RESULTS: The objective response rate was 24.1%, and the disease control rate was 79.3%. The median PFS of all patients was 5.8 months, and median OS was 10.3 months. A total of 236 cycles of treatment were available for safety assessment and the toxicity of the combination therapy was relatively well tolerated. The most common grade 3 toxicities were myelosuppression including leukopenia (5.08%), neutropenia (4.24%), lymphopenia (2.12%), thrombocytopenia (1.69%) and anemia (1.27%). Grade 4 toxicities included neutropenia (2.12%), thrombocytopenia (2.12%) and proteinuria (1.69%). All the adverse events were relieved after symptomatic treatment or dose reduction. CONCLUSIONS: Apatinib plus temozolomide could be an effective regimen with manageable toxicities and favorable efficacy and may outperform temozolomide monotherapy, particularly in newly diagnosed adults with tumors located outside the pons. The novel therapy deserves further investigation in adult DMG patients.

Superhydrophobic coatings hold immense promise for various applications. However, their practical use is currently hindered by issues such as poor stability, high costs, and complex preparation processes. Here, we present the preparation of cost-effective and stable superhydrophobic coatings through fluorination of natural attapulgite (F-ATP) nanorods and subsequent solvent-induced phase separation of a silicone-modified polyester adhesive (SMPA) with the F-ATP nanorods dispersed in it. Phase separation of the F-ATP/SMPA system forms a uniform suspension of microaggregates, which can be easily utilized for preparing superhydrophobic coatings via spray coating. The coatings have a low-surface-energy hierarchical micro/nanostructure due to phase separation of SMPA and adhesion of F-ATP to it. Moreover, the effects of the solvent composition (i.e., phase separation degree of SMPA) and the SMPA/F-ATP mass ratio on the morphology, superhydrophobicity, and stability of the coatings were investigated. After systematic optimization, the coatings exhibit excellent static and dynamic superhydrophobicity as well as high mechanical, chemical, thermal, and UV aging stability. Finally, the coatings were applied to the 5G radome surface and showed good rain attenuation prevention performance. Thus, we are confident that the superhydrophobic coatings have great application potential due to their advantages of outstanding performance, straightforward preparation procedures, cost-effectiveness, etc.

Superamphiphobic coatings have good application prospects in many fields but are limited by their low impalement resistance, weak mechanical stability, and easy adhesion of tiny droplets. Here, impalement-resistant, mechanically stable, and antistatic superamphiphobic coatings were fabricated by spraying a mixture of conductive carbon black (CB), silicone-modified polyester adhesive/fluorinated SiO2 microspheres onto Al alloy. The microspheres were obtained by adhesive phase separation and the binding of fluorinated SiO2 to them. The morphology, superamphiphobicity, impalement resistance, and mechanical stability of the coatings could be regulated by using solvents with different boiling points. As a result, the coatings simultaneously exhibited outstanding mechanical stability, impalement resistance, and superamphiphobicity. The addition of conductive CB endowed the coatings with good antistatic and tiny droplet repellent properties. In addition, the coatings exhibited good anti-icing properties due to the steady air layer at the solid-liquid interface and the very small contact area between them. We suppose that the coatings are very promising for practical application in various fields, including anti-icing, due to their outstanding comprehensive properties and simple preparation process.

Frailty syndrome denotes a decreased capacity of the body to maintain the homeostasis and stress of the internal environment, which simultaneously increases the risk of adverse health outcomes in older adults, including disability, hospitalization, falls, and death. To promote healthy aging, we should find strategies to cope with frailty. However, the pathogenesis of frailty syndrome is not yet clear. Recent studies have shown that the diversity, composition, and metabolites of gut microbiota significantly changed in older adults with frailty. In addition, several frailty symptoms were alleviated by adjusting gut microbiota with prebiotics, probiotics, and symbiosis. Therefore, we attempt to explore the pathogenesis of frailty syndrome in older people from gut microbiota and summarize the existing interventions for frailty syndrome targeting gut microbiota, with the aim of providing timely and necessary interventions and assistance for older adults with frailty.

ABSTRACT: Regulated cell death is a controlled form of cell death that protects cells by adaptive responses in pathophysiological states. Ferroptosis has been identified as a novel method of controlling cell death in recent years. Several cardiovascular diseases (CVDs) are shown to be profoundly influenced by ferroptosis, and ferroptosis is directly linked to the majority of cardiovascular pathological alterations. Despite this, it is still unclear how ferroptosis affects the pathogenic alterations that take place in CVDs. Based on a review of the mechanisms that regulate ferroptosis, this review explores the most recent research on the role of ferroptosis in the major pathological changes associated with CVDs, to provide new perspectives and strategies for cardiovascular research and clinical treatment.

This study examined the potential correlation between the immoderate intake of sugar-sweetened beverages (SSBs) and the subsequent rate of diabetes remission (DR). 206 individuals who met the eligibility criteria between January 2019 and June 2022 were recruited. Inquiries were conducted to gather information on the participants' beverage consumption before the onset. Subsequently, the participants were separated into the diabetes remission group (DR group) and nondiabetes remission group (NDR group) depending on whether they met the diagnostic criteria for diabetes remission. Baseline clinical elements within the two groups were juxtaposed, and factors influencing diabetes remission were identified through logistic regression analyses. The cutoff values of each critical factor were determined based on the receiver operating characteristic curve. One hundred and nine patients reported a history of SSB consumption, while the remaining 58 reported no such history. After 1 year, 40 patients achieved remission from diabetes. Compared with the NDR group, a higher SSBs ratio, body mass index (BMI), and blood creatinine (BCr) was observed in the DR group after adjusting for confounders, SSBs (odds ratio [OR] = 3.503; 95% confidence interval [CI] = 1.334-9.202; p = 0.011) and BCr (OR = 1.038; 95% CI = 1.003-1.079; p = 0.042) emerged as independent predictors of DR. The composite index of SSBs and BCr efficaciously predicted DR (area under the ROC curve [AUC] = 0.810, p < 0.001). SSBs and BCr were independent risk factors for DR. The amalgamation of these markers could more accurately predict DR.

Aiming to address the issues of parameter complexity and high computational load in existing fault detection algorithms for transmission lines, which hinder their deployment on devices like drones, this study proposes a novel lightweight model called Leaner YOLOv7-Tiny. The primary goal is to swiftly and accurately detect typical faults in transmission lines from aerial images. This algorithm inherits the ELAN structure from YOLOv7-Tiny network and replaces its backbone with depthwise separable convolutions to reduce model parameters. By integrating the SP attention mechanism, it fuses multi-scale information, capturing features across various scales to enhance small target recognition. Finally, an improved FCIoU Loss function is introduced to balance the contribution of high-quality and low-quality samples to the loss function, expediting model convergence and boosting detection accuracy. Experimental results demonstrate a 20% reduction in model size compared to the original YOLOv7-Tiny algorithm. Detection accuracy for small targets surpasses that of current mainstream lightweight object detection algorithms. This approach holds practical significance for transmission line fault detection.

In the face of complex scenarios, the information insufficiency of classification tasks dominated by a single modality has led to a bottleneck in classification performance. The joint application of multimodal remote sensing data for surface observation tasks has garnered widespread attention. However, issues such as sample differences between modalities and the lack of correlation in physical features have limited the performance of classification tasks. Establishing effective interaction between multimodal data has become another significant challenge. To fully integrate heterogeneous information from multiple modalities and enhance classification performance, this paper proposes a dual-branch cross-Transformer feature fusion network aimed at joint land cover classification of hyperspectral imagery (HSI) and Light Detection and Ranging (LiDAR) data. The core idea is to leverage the potential of convolutional operators to represent spatial features, combined with the advantages of the Transformer architecture in learning remote dependencies. The framework employs an improved self-attention mechanism to aggregate features within each modality, highlighting the spectral information of HSI and the spatial (elevation) information of LiDAR. The feature fusion module based on cross-attention integrates deep features from two modalities, achieving complementary information through cross-modal attention. The classification task is performed using jointly obtained spectral and spatial features. Experiments were conducted on three multi-source remote sensing classification datasets, demonstrating the effectiveness of the proposed model compared to existing methods.

Neutrophil extracellular traps(NETs) are fibrous networks formed by neutrophils after a procedure called NETosis, with the function of capturing and killing pathogens. NETs are widely involved in the pathological processes of major diseases such as immune system diseases, respiratory diseases, metabolic diseases, cancers, and reperfusion injury. Therefore, regulating NETs has become one of the important ways to prevent and treat the above diseases. As an excellent traditional culture in China, traditional Chinese medicine has made outstanding contributions to the treatment of diseases. In recent years, studies have discovered that a variety of active components in traditional Chinese medicines, Chinese medicine compound prescriptions, and single traditional Chinese medicines can alleviate the symptoms by regulating NETs in the pathological process of major diseases. This article reviews the research progress in the regulation of NETs by the active components of traditional Chinese medicines, Chinese medicine compound prescriptions, and single traditional Chinese medicines in the last five years, aiming to serve as a reference for related research.

Poly(ethylene oxide) (PEO)-based composite solid electrolytes (CSEs) are promising to accelerate commercialization of solid-state lithium metal batteries (SSLMBs). Nonetheless, this is hindered by the CSEs' limited ion conductivity at room temperature. Here, we propose design, synthesis, and application of the bioinspired neuron-like nanofillers for PEO-based CSEs. The neuron-like superhydrophobic nanofillers are synthesized by controllably grafting silicone nanofilaments onto montmorillonite nanosheets. Compared to various reported fillers, the nanofillers can greatly improve ionic conductivity (4.9×10-4 S cm-1, 30 °C), Li+ transference number (0.63), oxidation stability (5.3 V) and mechanical properties of the PEO-based CSEs because of the following facts. The distinctive neuron-like structure and the resulting synaptic-like connections establish numerous long-distance continuous channels over various directions in the PEO-based CSEs for fast and uniform Li+ transport. Consequently, the assembled SSLMBs with the CSEs and LiFePO4 or NCM811 cathodes display superior cycling stability over a wide temperature range of 50 °C to 0 °C. Surprisingly, the pouch batteries with the large-scale prepared CSEs kept working after being repeatedly bent, folded, cut or even punched in air. We believe that design of neuron-like nanofillers is a viable approach to produce CSEs with high room temperature ionic conductivity for SSLMBs.

Recombinant adeno-associated virus (rAAV) is a leading gene therapy vector. However, neutralizing antibodies reduce its efficacy. Traditional methods used to investigate antibody binding provide limited information. Here, charge detection mass spectrometry (CD-MS) was used to investigate the binding of monoclonal antibody ADK8 to AAV serotype 8 (AAV8). CD-MS provides a label-free approach to antibody binding. Individual binding events can be monitored as each event is indicated by a shift of the antibody-antigen complex to a higher mass. Unlike other methods, the CD-MS approach reveals the distribution of antibodies bound on capsids, allowing AAV8 subpopulations with different affinities to be identified. The charge state generated by the electrospray of large ions is normally correlated with the structure, and the charge is expected to increase when an antibody binds to the capsid exterior. Surprisingly, binding of the first ADK8 to AAV8 causes a substantial decrease in the charge, suggesting that the first antibody binding event causes a significant structural change. The charge increases for subsequent binding events. Finally, high ADK8 concentrations cause agglutination, where ADK8 links AAV capsids to form dimers and higher order multimers.

Lithium (Li) metal batteries are among the most promising devices for high energy storage applications but suffer from severe and irregular Li dendrite growth. Here, it is demonstrated that the issue can be well tackled by precisely designing the leaf-like membrane with hierarchical structure and exquisite fluidic channels. As a proof of concept, plant leaf-inspired membrane (PLIM) separators are prepared using natural attapulgite nanorods. The PLIM separators feature super-electrolyte-philicity, high thermal stability and high ion-selectivity. Thus, the separators can guide uniform and directed Li growth on the Li anode. The Li//PLIM//Li cell with limited Li anode shows high Coulombic efficiency and cycling stability over 1500 h with small overpotential and interface impedance. The Li//PLIM//S battery exhibits high initial capacity (1352 mAh g-1 ), cycling stability (0.019% capacity decay per cycle at 1 C over 500 cycles), rate performance (673 mAh g-1 at 4 C), and high operating temperature (65 °C). The separators can also effectively improve reversibility and cycling stability of the Li/Li cell and Li//LFP battery with carbonate-based electrolyte. As such, this work provides fresh insights into the design of bioinspired separators for dendrite-free metal batteries.

Recombinant adeno-associated virus (rAAV) vectors carry a cassette of interest retaining only the inverted terminal repeats (ITRs) from the wild-type virus. Conventional rAAV production primarily uses a vector plasmid as well as helper genes essential for AAV replication and packaging. Nevertheless, plasmid backbone related contaminants have been a major source of vector heterogeneity. The mechanism driving the contamination phenomenon has yet to be elucidated. Here we identified cryptic resolution sites in the plasmid backbone as a key source for producing snapback genomes, which leads to the increase of vector genome heterogeneity in encapsidated virions. By using a single ITR plasmid as a model molecule and mapping subgenomic particles, we found that there exist a few typical DNA break hotspots in the vector DNA plasmid backbone, for example, on the ampicillin DNA element, called aberrant rescue sites. DNA around these specific breakage sites may assume some typical secondary structures. Similar to normal AAV vectors, plasmid DNA with a single ITR was able to rescue and replicate efficiently. These subgenomic DNA species significantly compete for trans factors required for rAAV rescue, replication, and packaging. The replication of single ITR contaminants during AAV production is independent of size. Packaging of these species is greatly affected by its size. A single ITR and a cryptic resolution site in the plasmid work synergistically, likely causing a source of plasmid backbone contamination.

The sensors with a wide gas pressure detection range are urgently demanded in many industrial applications. Here, we propose a gas pressure sensor based on an all-solid open Fabry-Pérot interferometer, which is prepared by using optical contact bonding to ensure high structural strength and high-quality factor of 8.8 × 105. The applied pressure induces a change in the refractive index of the air, leading to the shift of the resonant spectrum. The pressure is detected by calibrating this shift. The sensor exhibits a pressure sensitivity of 4.20 ± 0.01 nm/MPa in a pressure range of 0 to 10 MPa and has a minimum pressure resolution of 0.005 MPa. Additionally, it shows a lower temperature cross-sensitivity of -0.25 kPa/°C. These findings affirm that the sensor achieves high-sensitivity pressure sensing across a wide detection range. Moreover, owing to its exceptional mechanical strength, it holds great promise for applications in harsh environments, such as high temperature and high pressure.

Superhydrophobic fabrics have great application potential in many fields including wearable electronic devices, sports textiles, and human health monitoring, but good water impalement resistance and stretching stability are the prerequisites. Here, we report the design of waterborne superhydrophobic fabrics with high impalement resistance and stretching stability by constructing elastic reconfigurable micro-/micro-/nanostructures. Following theoretical analysis, two approaches were proposed and employed: (i) regulating distance between the microfibers of polyester fabrics to decrease the solid-liquid contact area, and (ii) forming reconfigurable two-tier hierarchical micro-/nanostructures on the microfibers by stretching during dipping to further decrease the solid-liquid contact area. The effects of microfiber distance and micro-/nanostructures on microfibers on superhydrophobicity and impalement resistance were studied. The superhydrophobic fabrics show excellent impalement resistance as verified by high-speed water impact, water jetting, and rainfall, etc. The fabrics also show excellent stretching stability, as 100% stretching and 1000 cycles of cyclic 100% stretching-releasing have no obvious influence on superhydrophobicity. Additionally, the fabrics show good antifouling property, self-cleaning performance, as well as high abrasion and washing stability. The experimental results agree with the theoretical simulation very well. We anticipate that this study will boost the development of impalement-resistant and stretching-stable superhydrophobic surfaces.

In national defense, aluminum nanoparticles (Al NPs) have better combustion performance than Al microparticles but are easily oxidized during processing, especially in oxidative liquids. Although some protective coatings have been reported, it is still challenging to obtain Al NPs stable in oxidative liquids (e.g., hot liquids) without scarifying combustion performance. Here, we report ultrastable Al NPs with enhanced combustion performance enabled by the crosslinked polydopamine/polyethyleneimine (PDA/PEI) nanocoating merely ∼15 nm in thickness and ∼0.24 wt % in mass. The Al@PDA/PEI NPs are fabricated by one-step rapid graft copolymerization of dopamine and PEI on Al NPs at room temperature. The formation mechanism of the nanocoating is discussed including reactions between dopamine and PEI and interactions of the nanocoating with Al NPs. The Al@PDA/PEI NPs show excellent stability in hot water, and the mechanism is interpreted by molecular dynamics simulation. The PDA/PEI nanocoating can also enhance the combustion heat and burning rate of Al NPs.

Solar-driven atmospheric water harvesting technology has the advantage of not being limited by geography and has great potential in solving the freshwater crisis. Here, we first propose a purely natural and degradable superhydrophilic composite macroporous hygroscopic material by applying guar gum (GG) to atmospheric water harvesting. The material consists of GG-cellulose nanofibers (CNFs) as a porous substrate material, limiting the hygroscopic factor lithium chloride (LiCl) in its three-dimensional (3D) network structure, and carbon nanotubes (CNTs) play a photothermal conversion role. The composite material has a high light absorption rate of more than 95%, and the macroporous structure (20-60 µm) allows for rapid adsorption/desorption kinetics. At 35 °C and 90% relative humidity (RH), the moisture absorption capacity is as high as 1.94 g/g. Under 100 mW/cm2 irradiation, the absorbed water is almost completely desorbed within 3 h, and the water harvesting performance is stable in 10 cycles. Moreover, liquid water was successfully collected in an actual outdoor experiment. This work demonstrates the great potential of biomass materials in the field of atmospheric water collection and provides more opportunities for various energy and sustainable applications in the future.

Clinical and hematological parameters can predict immune-related adverse events (irAEs) caused by immune checkpoint inhibitors (ICIs). However, the exact correlation between these parameters and irAEs is unclear. This study aimed to establish a prediction model for irAEs in patients with non-small cell lung cancer (NSCLC) treated with ICIs. This retrospective study included patients with NSCLC treated with a minimum of one dose of ICIs at the Tianjin Medical University Cancer Hospital and Shanxi Bethune Hospital from 2016 to 2020. Baseline characteristics, treatment details, and adverse events were evaluated. The Student's t-test, Chi-square test, and logistic regression were used to identify risk factors for irAEs to establish a prediction model. A total of 667 patients were included; the median age was 62.47 (range, 27-85) years. Most patients were men (74.5%) with stage IV cancer (93.1%). The incidence of any grade and grade 3 or higher irAEs was 21.74% (145/667) and 5.25% (35/667), respectively. A total of 145 patients experienced 220 irAEs; the incidence of endocrinopathies (35.91%, 79/220) was highest in all grade irAEs, while that of pneumonitis (7.73%, 17/220) was the highest in grade 3 or higher irAEs. A prediction model based on treatment lines, aspartate aminotransferase (AST), lactate dehydrogenase (LDH), absolute lymphocyte count (ALC), and systemic immune inflammation index was established. The area under the receiver operator characteristic curve was 0.722 (95% confidence interval: 0.650-0.793), with a cut-off value of 0.247 and a sensitivity and specificity of 62.9% and 74.6%, respectively. The multivariate logistic regression analysis showed that the risk of irAEs was higher in patients undergoing second-line therapy than in those undergoing treatment with adjuvant therapy (odds ratio [OR] = 8.239, p = 0.011). AST (OR = 1.053, p = 0.007) and ALC (OR = 2.556, p = 0.001) showed a positive correlation with the risk of irAEs, while LDH showed a negative correlation with irAEs (OR = 0.994, p = 0.007). The model showed good prediction efficiency, whereas the treatment lines, AST, ALC, and LDH were independent risk factors for the onset of irAEs.
