RESUMO

Solid Oxide Fuel Cell (SOFC) cathodes operating in ambient atmospheric conditions inevitably encounter CO2 contamination, leading to sustained performance deterioration. In this investigation, we examined the impact of CO2 on three variants of (La,Sr)(Co,Fe)O3-δ cathodes and employed the distribution of relaxation times method to distinguish distinct electrochemical processes based on impedance spectra analysis. We meticulously analyzed and discussed the corrosion resistance of these (La,Sr)(Co,Fe)O3-δ cathodes under high CO2 concentrations, relying on the experimental data. Electrochemical impedance spectroscopy results revealed that La0.6Sr0.4Co0.2Fe0.8O3-δ (LSCF-6428), La0.4Sr0.6Co0.2Fe0.8O3-δ (LSCF-4628), and La0.4Sr0.6Co0.2Fe0.7Nb0.1O3-δ (LSCFN-46271) cathodes exhibited persistent degradation when exposed to CO2 at temperatures of 650 °C or 800 °C during the durability-testing period. An increase in electrode polarization resistance was observed upon CO2 introduction to the electrode, but electrode performance recovered upon returning to a pure air environment. Furthermore, X-ray diffraction and scanning electron microscopy analyses confirmed that CO2 did not cause permanent damage to the (La,Sr)(Co,Fe)O3-δ cathodes. These findings indicate that the (La,Sr)(Co,Fe)O3-δ cathodes exhibit excellent resistance to CO2-induced corrosion.

RESUMO

BACKGROUND: Lung cancer is one of the most common malignant tumors, and asbestos exposure was suggested to contribute to a proportion of lung cancer cases. Previous genome-wide gene-environment interaction analysis reported that rs13383928 was associated with asbestos-related lung cancer. However, the mechanism of this association was still unclear. METHODS: In the present study, we retrieved the genotype data from the 1,000 Genomes Project on single-nucleotide polymorphisms (SNPs) surrounding rs13383928 and analyzed the linkage disequilibrium (LD) pattern of this region. Further functional genomics analyses were performed. RESULTS: The result indicated that no other SNPs were in LD with rs13383928, suggesting that rs13383928 is the causal one. The following dual luciferase assay disclosed that the T allele of rs13383928 presented significantly higher enhancer activity than G in lung cells, thus verifying that this SNP was functional in the lung. Through chromosome conformation capture, the PTH2R (parathyroid hormone 2 receptor)promoter was identified to interact with the segment surrounding rs13383928. By chromatin immunoprecipitation, it was observed that the region spanning rs13383928 could bind transcription factor FOXJ2 (forkhead box J2). CONCLUSION: Our functional genomics evidence supports a link between rs13383928 and asbestos-related lung cancer through regulating PTH2R.

RESUMO

Pu-erh tea belongs to the six tea categories of black tea, according to the processing technology and quality characteristics, is divided into two types of raw tea and ripe tea. Raw tea is made from fresh leaves of tea as raw materials, through the process of greening, kneading, sun drying, steam molding and other processes made of tightly pressed tea. Ripe tea is made from Yunnan large-leafed sun green tea, using a specific process, post-fermentation (rapid post-fermentation or slow post-fermentation) processing of loose tea and tightly pressed tea. TAETEA Prebiotea is Puerh Ripe Tea, TAETEA Prebiotea has the effect of increasing insulin level and improving hyperglycemia in mice, and it also has the effect of regulating blood lipids, which can reduce the level of serum total cholesterol (TC) and triglycerides (TG), increase the level of high-density lipoprotein cholesterol (HDL-C), and improve the metabolism of lipids. Therefore, further experiments were conducted by us, and TAETEA Prebiotea was formulated into a suitable dose for the intervention of non alcoholic fatty liver disease (NAFLD) model rats, and at the end of the experiments, the samples of each group of experiments were analyzed and detected by the method of UHPLC-Q-Exactive LC-MS liquid-mass spectrometry methodology, and the relevant metabolites as well as metabolic pathways were analyzed by the method of Non targeted metabolomics analysis. As a result, 71 differential metabolites could be screened, of which 35 differential metabolites were up-regulated after intervention and 36 differential metabolites were down-regulated after intervention. Based on the KEGG pathway enrichment and Pathway Impact bubble diagram analysis, glycine, serine, threonine metabolism, arginine and proline metabolism, protein digestion and absorption, and central carbon metabolism in cancer may be the main metabolic pathways in which TAETEA Prebiotea exerted preventive effects on NAFLD rats, C00148 (Proline), C00300 (Creatine) and C00719 (Betaine) are the differential metabolites that play important regulatory roles.

RESUMO

Lung cancer is a malignant tumor with high rates of mortality and shows significant hereditary predisposition. Previous genome-wide association studies suggest that rs748404, located at promoter of TGM5 (transglutaminase 5), is associated with lung carcinoma. By analysis of 1000 genomes project data for three representative populations in the world, another five SNPs are identified to be in strong linkage disequilibrium with rs748404, thus suggesting that they may also be associated with lung carcinoma risk. However, it is ambiguous about the actually causal SNP(s) and the mechanism for the association. Dual-luciferase assay indicates that the functional SNPs are not rs748404, rs12911132 or rs35535629 but another three SNPs (rs66651343, rs12909095 and rs17779494) in lung cell. By chromosome conformation capture, it is disclosed that the enhancer encompassing the two SNPs, rs66651343 and rs12909095, can interact with the promoter of CCNDBP1 (cyclin D1 binding protein 1). RNA-seq data analysis indicates that CCNDBP1 expression is dependent on the genotype of these two SNPs. Chromatin immunoprecipitation assay suggests that the fragments spanning rs66651343 and rs12909095 can bind with the transcription factors, cut like homeobox 1 and SRY-box transcription factor 9, respectively. Our results establish the connection between genetic variations at this locus and lung cancer susceptibility.

RESUMO

Based on the enhanced knowledge on the tumor microenvironment (TME), a more comprehensive treatment landscape for targeting the TME has emerged. This microenvironment provides multiple therapeutic targets due to its diverse characteristics, leading to numerous TME-targeted strategies. With multifaced activities targeting tumors and the TME, vitamin C is renown as a promising candidate for combination therapy. In this review, we present new advances in how vitamin C reshapes the TME in the immune, hypoxic, metabolic, acidic, neurological, mechanical, and microbial dimensions. These findings will open new possibilities for multiple therapeutic avenues in the fight against cancer. We also review the available preclinical and clinical evidence of vitamin C combined with established therapies, highlighting vitamin C as an adjuvant that can be exploited for novel therapeutics. Finally, we discuss unresolved questions and directions that merit further investigation.

RESUMO

Intermittent fasting (IF) is gaining popularity as a therapeutic dietary strategy that regulates metabolism and can alter the development of metabolic disorders. An increasing amount of research has connected ocular diseases to IF and discovered that it has a direct and indirect effect on the eye's physiological structure and pathological alterations. This article summarizes the progress of research on IF in regulating the physiological structures of the ocular vasculature, the anterior segment of the eye, the retina, and the choroid. We explored the therapeutic potential of IF for various common ocular diseases. In the future, a comprehensive study into the fundamental processes of IF will provide a direct and rigorous approach to eye disease prevention and therapy.

RESUMO

The purpose of this study was to ascertain the effect of chondroitin sulphate-modified doxorubicin (Dox) nanoparticles on enhancing the tumour-targeting effect and tumour growth inhibition effect of doxorubicin both in vitro and in vivo. The chondroitin sulphate-doxorubicin conjugate and its poly(lactic-co-glycolic acid) (PLGA) nanoparticles (CS-Dox-PLGA) were successfully synthesised, and then characterized by Fourier-transform infrared spectroscopy (FTIR), proton magnetic resonance (1HNMR), thermogravimetric analysis/differential scanning calorimetry (TGA/DSC), transmission electron microscope (TEM), zeta potential and laser light scattering. Taking advantage of the enhanced permeability and CD44-mediated endocytosis, CS-Dox-PLGA showed excellent capacity for penetrating the peripheral tumour barrier and into the nucleus of tumour cells. The CS-Dox-PLGA cellular uptake was improved and exhibited a significantly higher level of cytotoxicity in U251 cells. After intravenous administration, the CS-Dox-PLGA showed good pharmacokinetic properties and excellent U251-induced tumour inhibition with low cardiac toxicity. Therefore, CS-Dox-PLGA with low cardiac toxicity and good anti-tumour ability might be a better choice for Dox in clinical practice.

RESUMO

BACKGROUND: The aim of the study was to investigate knowledge and use of wild food plants in two mountain valleys separated by Mount Taibai--the highest peak of northern China and one of its biodiversity hotspots, each adjacent to species-rich temperate forest vegetation. METHODS: Seventy two free lists were collected among the inhabitants of two mountain valleys (36 in each). All the studied households are within walking distance of primary forest vegetation, however the valleys differed in access to urban centers: Houzhenzi is very isolated, and the Dali valley has easier access to the cities of central Shaanxi. RESULTS: Altogether, 185 wild food plant species and 17 fungi folk taxa were mentioned. The mean number of freelisted wild foods was very high in Houzhenzi (mean 25) and slightly lower in Dali (mean 18). An average respondent listed many species of wild vegetables, a few wild fruits and very few fungi. Age and male gender had a positive but very low effect on the number of taxa listed.Twelve taxa of wild vegetables (Allium spp., Amaranthus spp., Caryopteris divaricata, Helwingia japonica, Matteucia struthiopteris, Pteridium aquilinum, Toona sinensis, Cardamine macrophylla, Celastrus orbiculatus, Chenopodium album, Pimpinella sp., Staphylea bumalda &S. holocarpa), two species of edible fruits (Akebia trifoliata, Schisandra sphenanthera) and none of the mushrooms were freelisted by at least half of the respondents in one or two of the valleys. CONCLUSION: The high number of wild vegetables listed is due to the high cultural position of this type of food in China compared to other parts of the world, as well as the high biodiversity of the village surroundings. A very high proportion of woodland species (42%, double the number of the ruderal species used) among the listed taxa is contrary to the general stereotype that wild vegetables in Asia are mainly ruderal species. The very low interest in wild mushroom collecting is noteworthy and is difficult to explain. It may arise from the easy access to the cultivated Auricularia and Lentinula mushrooms and very steep terrain, making foraging for fungi difficult.

RESUMO

We purified a novel mannose binding lectin form Musca domestica pupae by affinity chromatography on Con A-Sepharose 4B and DEAE weak anion-exchange chromatography. By SDS-PAGE, MBL-1 yielded a single band with the molecular weight of 24 kDa. It was a glycoprotein detected by periodic acid-schiffs staining reaction, with 97.36% protein and 2.1% oligosaccharide. Meanwhile, the results of beta-elimination reaction, infrared spectroscopy, atomic force microscopy and protein sequencing instrument show that MBL-1 was an ellipsoidal-shaped monomer with 60-100 nm in diameter. N-glycoside bond linked oligosaccharide chain and the N-terminal blocked peptide chain. Further study suggested that MBL-1 promote the proliferation of macrophage in a concentration-dependent manner. The scanning electron microscope analysis shows that MBL-1 promoted the activation of macrophages. These results show that MBL-1 purified from Musca domestica pupae possesses immune regulation effect, serving a reference basis to develop natural immune-modulator.
