RESUMO

Acetamiprid (ACDP) is a widely used neonicotinoid insecticide that is popular for its efficacy in controlling fleas in domestic settings and for pets. Our study aims to offer a comprehensive examination of the toxicological impacts of ACDP and the prophylactic effects of cinnamon nanoemulsions (CMNEs) on the pathological, immunohistochemical, and hematological analyses induced by taking ACDP twice a week for 28 days. Forty healthy rats were divided into four groups (n = 10) at random; the first group served as control rats; the second received CMNEs (2 mg/Kg body weight); the third group received acetamiprid (ACDP group; 21.7 mg/Kg body weight), and the fourth group was given both ACDP and CMNEs by oral gavage. Following the study period, tissue and blood samples were extracted and prepared for analysis. According to a GC-MS analysis, CMNEs had several bioactive ingredients that protected the liver from oxidative stress by upregulating antioxidant and anti-inflammatory agents. Our findings demonstrated that whereas ACDP treatment considerably boosted white blood cells (WBCs) and lymphocytes, it significantly lowered body weight gain (BWG), red blood cells (RBCs), hemoglobin (Hb), hematocrit (HCT), and platelets (PLT). ACDP notably reduced antioxidant enzyme activities: superoxide dismutase (SOD), glutathione peroxidase (GPx), and catalase (CAT) and elevated hydrogen peroxide and malondialdehyde levels compared with other groups. ACDP remarkably raised alanine aminotransferase (ALT), aspartate amino transaminase (AST), and alkaline phosphatase (ALP) levels.Moreover, the histopathological and immunohistochemistry assays discovered a severe toxic effect on the liver and kidney following ACDP delivery. Furthermore, cyclooxygenase 2 (COX-2) + immunoexpression was enhanced after treatment with CMNEs. All of the parameters above were returned to nearly normal levels by the coadministration of CMNEs. The molecular docking of cinnamaldehyde with COX-2 also confirmed the protective potential of CMNEs against ACDP toxicity. Our findings highlighted that the coadministration of CMNEs along with ACDP diminished its toxicity by cutting down oxidative stress and enhancing antioxidant capacity, demonstrating the effectiveness of CMNEs in lessening ACDP toxicity.

Salinity stress is a type of abiotic stress which negatively affects the signaling pathways and cellular compartments of plants. Melatonin (MT) has been found to be a bioactive compound that can mitigate these adverse effects, which makes it necessary to understand the function of MT and its role in salt stress. During this study, plants were treated exogenously with 100 µM of MT for 7 days and subjected to 200 mM of salt stress, and samples were collected after 1 and 7 days for different indicators and transcriptome analysis. The results showed that salt reduced chlorophyll contents and damaged the chloroplast structure, which was confirmed by the downregulation of key genes involved in the photosynthesis pathway after transcriptome analysis and qRT-PCR confirmation. Meanwhile, MT increased the chlorophyll contents, reduced the electrolyte leakage, and protected the chloroplast structure during salt stress by upregulating several photosynthesis pathway genes. MT also decreased the H2O2 level and increased the ascorbic acid contents and APX activity by upregulating genes involved in the ascorbic acid pathway during salt stress, as confirmed by the transcriptome and qRT-PCR analyses. Transcriptome profiling also showed that 321 and 441 DEGs were expressed after 1 and 7 days of treatment, respectively. The KEGG enrichment analysis showed that 76 DEGs were involved in the photosynthesis pathway, while 35 DEGs were involved in the ascorbic acid metabolism pathway, respectively. These results suggest that the exogenous application of MT in plants provides important insight into understanding MT-induced stress-responsive mechanisms and protecting Brassica campestris against salt stress by regulating the photosynthesis and ascorbic acid pathway genes.

Microplastic (MP) pollution has become a health concern subject in recent years. Althoughann increasing number of studies about the ingestion of microplastics by fish, research on the oxidative stress response to MPs in natural environments is quite limited. In this study, the identification and characterization of MPs in gill (G), muscle tissues (M), and gastrointestinal tract (GI) of turbot (Scophthalmus maximus) were evaluated. Oxidative damage of MPs on the brain (B), liver (L), gill (G), and muscle (M) tissues as well as their effect on superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), paraoxonase (PON), arylesterase (AR) myeloperoxidase (MPO), and malondialdehyde (MDA) biomarkers were evaluated. The potential transmission of MPs from muscle tissues to humans was examined. Results showed that gills contain the highest amounts of MPs, ethylene propylene is the most dominant polymer type, black and blue are the most common MP color, fiber is the most common shape, and 50-200 µm is the most common MP size. Results showed that MPs cause oxidative stress of tissues with inhibiting effect on enzyme activities and promoting impact on lipid peroxidation. The oxidative damage mostly affected the liver (detoxification organ) followed by gill tissue. The intake of MPS in the European Union was estimated by EFSA as 119 items/year, while in Turkey it is 47.88 items/year. This study shows that more research is needed in terms of ecosystem health and food chain safety. The risk assessment of MPs in living organisms and environmental matrices including food safety and human health should be considered a public health issue.

OBJECTIVE: To comprehensively assess recent data on the effects of orthodontic forces on the dental pulp and to critically evaluate, whether any of the changes are permanent. MATERIALS AND METHODS: Articles published between 2/2009 and 2/2022 were searched electronically on the PubMed, EMBASE and SCOPUS databases. The initial search retrieved 780 publications and, applying the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines, 33 relevant articles were identified. Twenty articles fulfilled the requirements for high (n = 1) or moderate (n = 19) methodological quality and were included. All assessments were made independently by three researchers. RESULTS: Orthodontic forces appeared to cause a reduction in pulpal blood flow and a reduction in tooth sensibility, as indicated by increased response thresholds and increased amounts of negative responses to tooth sensibility tests. In addition, there were increases in the expression or activity levels of enzymes and neuropeptides associated with hypoxia and inflammation. Fibrotic tissue formation in the pulp was also reported. CONCLUSIONS: Except for some histological and morphological alterations, the observed pulpal changes were in most cases only temporary, appearing within days of initiating the treatment and usually lasting for weeks. There were no clear signs of permanent damage.

OBJECTIVE: The aim: To assess the structural and metabolic changes in the sensorimotor cortex of the rat brain under conditions of hemorrhagic stroke. PATIENTS AND METHODS: Materials and methods: The experiment was carried out on rats of the control and experimental groups with a model of hemorrhagic stroke. We used histological, electron microscopic, biochemical methods and biological markers. RESULTS: Results: In the sensorimotor cortex of the ipsilateral cerebral hemisphere of rats under conditions of hemorrhagic stroke, cerebral edema and progression of neurodegenerative changes were observed; an increase in the size of mitochondria, which is caused by edema of their matrix; activation of lipid peroxidation processes and a decrease in the activity of enzymes of the antioxidant system, a decrease in the level of apoptosis markers and inhibition of ERK1/2 expression. The study of DNA fragmentation in the cerebral cortex revealed a significant number of manifestations of necrosis and an insignificant number of cells in a state of apoptosis. CONCLUSION: Conclusions: after modelling a hemorrhagic stroke in the right hemisphere of the brain, perivascular and pericellular edema of the energy apparatus, cell death by necrosis and apoptosis, and activation of lipid peroxidation processes were established as well as a decrease in the activity of enzymes of the antioxidant system.

OBJECTIVE: The aim of the study was to evaluate the state of oxidation processes and morphological changes in the heart of castrated rats during the development of epinephrine heart damage (EHD). PATIENTS AND METHODS: Materials and methods. The study was performed on 120 white male Wistar rats. The animals were divided into four series: 1 - control, 2 - castration. For EHD, rats were injected once intraperitoneally with a 0.18% solution of adrenaline hydrotartrate at the rate of 0.5 mg/kg of weight. Castration was performed under anesthesia. The concentration of diene and triene conjugates (DC, TC), Schiff's bases (SB), TBA-active products (TBA-ap), oxidatively modi"ed proteins (OMP), activity of superoxide dismutase (SOD) and catalase (CAT) were determined in the heart. A morphological study of preparations stained with Azantrichrome was carried out. All studies were performed in control, 1, 3, 7, 14 and 28 days after adrenaline injection. RESULTS: Results: In the I series DC and TC increased after 1 day of EHD, fell to control values after 3 days, and then had wave-like character (highest - after 14 days). SB decreased (minimal after 7 days), TBA-ap increase (maximal after 14 days). OMP370 increased after 1 and 3 days, after 7 days they did not differ from the control, after 14 days they were higher than in control, and after 28 days they decreased to the control values. OMP430 and OMP530were greater than the control indicators in all terms, except the last; the maximum was noted after 14 days. The activity of antioxidant enzymes was lower than the control indicators at all times of the study. Castration caused an increase of lipid peroxidation. After 7 days, DC and TC, were lower and SB - higher, than in the I series. Castration caused a decrease in OMP. In EHD all values of OMP, compared to the castrated control rats, were higher at all studed times Castration leads to increase of SOD, and decrase of CAT. All indicators of SOD and CAT exceeded the indicators of animals of the I series at all times of the study. Biochemical changes are consistent with morphological changes. After injection of epinephrine, severe vascular disorders, adventitia edema, perivasal edema, endothelial cell damage, dilatation of hemicapillaries, full blood vessels, stasis, hemorrhages in the surrounding tissues, and sclerosing of the walls of arteries and venules were observed. Cardiomyocytes were swollen, shortening, necrosis was observed, myocytolysis was noted. Edema of the stroma was noted. In the stroma, around the vessels, located cells of connective tissue elements were observed. Indicate more damage to the myocardium in the process of development of EHD in animals of the I series. CONCLUSION: Conclusions: Castration of rats causes an increase of lipid peroxidation products and CAT activity in the heart, but a decrease in the content of OMP. Adrenaline injection causes activation of lipid peroxidation and an increase in the content of OMP. During the development of EHD, the activity of antioxidants is significantly higher in II group. Biochemical changes are consistent with morphological, and indicate more damage to the myocardium in the development of EHD in animals of the I series.

The Triassic-Jurassic boundary marks the third largest mass extinction event in the Phanerozoic, characterized by a rise in CO2 -concentrations from c. 600 ppm to c. 2100-2400 ppm, coupled with a c. 3.0-4.0°C temperature rise. This is hypothesized to have induced major floral turnover, altering vegetation structure, composition and leaf morphology, which in turn are hypothesized to have driven changes in wildfire. However, the effects of elevated CO2 on fuel properties, such as chemical composition of leaves, are also important in influencing fire behaviour, but yet have not been considered. We test this by selecting three Triassic analogue species grown experimentally in different atmospheric compositions, and analyse variations in leaf chemistry, and leaf level flammability. These data were used to inform a fire behaviour model. We find that all three species tested showed a reduction in their volatile component, leading to lower flammability. Accounting for these variations in a model, our results suggest that leaf intrinsic flammability has a measurable impact on modelled fire behaviour. If scaled up to ecosystem level, periods of elevated CO2 may therefore be capable of inducing both biochemical and morphological changes in fuel properties, and thus may be capable of influencing fire behaviour.

Organophosphorus pesticides like Chlorpyrifos 48%EC were widely used to control agricultural pests. The present study aimed to evaluate the toxic effects of Chlorpyrifos 48%EC on B. alexandrina snails, the intermediate host of Schistosoma mansoni. After exposure of snails to serial concentrations to determine the LC50, thirty snails for each sublethal concentration (LC10 2.1 and LC25 5.6 mg/l) in each group were exposed for 24 h followed by another 24 h for recovery. After recovery random samples were collected from hemolymph and tissue to measure the impacts on Phagocytic index, histological, biochemical, and molecular parameters. The current results showed a toxic effect of Chlorpyrifos 48%EC on adult B. alexandrina snails after 24 h of exposure at LC50 9.6 mg/l. After exposure to the sub-lethal concentrations of this pesticide, it decreased the total number of hemocytes and the percentage of small cells, while increased the percentage of hyalinocytes. The granulocyte percentage was increased after exposure to LC10, while after LC25, it was decreased compared to the control group. Also, the light microscopical examination showed that some granulocytes have plenty of granules, vacuoles and filopodia. Some hyalinocytes were contained shrinked nuclei, incomplete cell division and forming pseudopodia. Besides, the phagocytic index of hemocytes was significantly increased than control in all treated groups. Also, these sub-lethal concentrations increased MDA and SOD activities, while, tissue NO, GST and TAC contents were significantly decreased after exposure. Levels of Testosterone (T) and Estradiol (E) were increased significantly after exposure compared with control group. The present results showed that the concentration of DNA and RNA was highly decreased after exposure to LC10, 25 than the control group. Therefore, B. alexandrina snails could be used as a bio monitor of the chemical pollution. Besides, this pesticide could reduce the transmission of schistosomiasis as it altered the biological system of these snails.

OBJECTIVE: The aim of the study was to study the effect of low and high doses of lead acetate on biochemical parameters and morphological status of rat ovaries in the experiment. PATIENTS AND METHODS: Materials and methods: The study was performed on 36 nonlinear female rats weighing 180-210 g, aged 4 months, divided into 3 experimental groups: I - control (C), II - rats, which were given 30 days to drink a solution of lead acetate with at the rate of 0,05 mg / kg of animal weight, group III - rats, which were given for 30 days to drink a solution of lead acetate at the rate of 60 mg/kg of animal weight. Biochemical research methods were included determination of diene conjugate concentration in animals' blood, concentration of TBA-active products, study of oxidative modification of proteins in blood plasma, determination of superoxide dismutase and catalase activities. Endogenous intoxication was assessed by the definition of medium-mass molecules, the content was expressed in units of extinction. The material for light microscopy investigation from the ovary was performed according to the generally accepted method. RESULTS: Results: Lead acetate causes activation of peroxidation of lipids and proteins in the body of female rats, which is directly dependent on the dose of lead. In response to the activation of free radical oxidation there are changes in the antioxidant system, which depend on the dose of lead acetate: at a dose of 0.05 mg / kg superoxide dismutase and catalase activity increase, at a dose of 60 mg / kg superoxide dismutase and catalase activity. Small doses of lead do not cause endogenous intoxication. Lead acetate causes the development of endogenous intoxication in animals only in large doses: increases the formation of toxic compounds, cell apoptosis, decreased excretory function of the kidneys, which is associated with multiorgan disorders. As a result of the action of lead acetate, morphological changes of the ovaries were observed, which increased with increasing dose of lead acetate. There was a dose-dependent decrease in massometric parameters, the number of follicles and changes in the thickness of the surface structures of the ovary, which is more pronounced at 60 mg/kg. CONCLUSION: Conclusions: Under the influence of small and large doses of lead acetate on biochemical changes in blood and morphological changes in the ovaries in male rats the oxidative stress is developed. Under the influence of small doses, the changes are adaptive, and under the influence of large doses - damaging.

The impact of gamma radiation on the activation of rice innate immunity to blast disease caused by Magnaporthe oryzae is described. In the present study, fenugreek seed extracts radiated with different doses of gamma rays viz. 5Gy, 10Gy, 15Gy, 20Gy and 25Gy were examined for their presence of biocompounds as well as for its ability to induce plant growth promotion and resistance against rice blast disease. The results of GC-MS analysis detected antimicrobial properties in methanolic extract. Enhanced germination (97%) and vigor (2718) was noticed in seeds pretreated with 20 Gy of gamma radiation in comparison with non-irradiated controls. Under greenhouse conditions, a significant disease protection of 56.7% on 3rd and 4th day after inoculation against rice blast was observed in 15Gy-irradiated rice plants challenge-inoculated with M. oryzae. Further, a significant increase in the hydrogen peroxide, phenol and lignin deposition was noticed in 20Gy-irradiated rice plants. Additionally, rice plants pretreated with 15Gy induced maximum activities of peroxidase (POX) and polyphenol oxidase (PPO) compared to untreated control plants. These findings revealed that rice plants-pretreated with gamma radiation elicit resistance against rice blast disease as well as strengthening the growth parameters by modulating cellular and biochemical defense system.

Alzheimer's disease (AD), a well known aging-induced neurodegenerative disease is related to amyloid proteinopathy. This proteinopathy occurs due to abnormalities in protein folding, structure and thereby its function in cells. The root cause of such kind of proteinopathy and its related neurodegeneration is a disorder in metabolism, rather metabolomics of the major as well as minor nutrients. Metabolomics is the most relevant "omics" platform that offers a great potential for the diagnosis and prognosis of neurodegenerative diseases as an individual's metabolome. In recent years, the research on such kinds of neurodegenerative diseases, especially aging-related disorders is broadened its scope towards metabolic function. Different neurotransmitter metabolisms are also involved with AD and its associated neurodegeneration. The genetic and epigenetic backgrounds are also noteworthy. In this review, the physiological changes of AD in relation to its corresponding biochemical, genetic and epigenetic involvements including its (AD) therapeutic aspects are discussed.

Biodistribution of nanodiamonds in mice after intravenous administration, activities of AST and ALT, and the level of bilirubin in the blood plasma were studied in 2.5 h and 10, 35, and 97 days after injection of nanodiamonds. In 2.5 h after intravenous injection, nanodiamonds mainly accumulate in the lungs and liver. Then, redistribution of nanodiamonds from all organs to the liver was observed. Activities of AST and ALT and the level of bilirubin in the blood increased after 2.5 h and then decreased to the initial values.

Postmortem structural and biochemical changes in the muscle tissue (MT) of myocardium from the positions of forensic examinations (FE) of the prescription of death coming (PDC) were not studied systematically, this fact determining the purpose of the present research. AIM: The aim of the research consisted in study of structural and biochemical changes in the tissue of myocardium during the early postmortem period (PMP). MATERIALS AND METHODS: The muscle tissue of myocardium within the early PMP (3-13 hours) after the coming of death was studied on 30 human corpses. Six BCM in myocardium muscle homogenates (MMH) were determined: BCM1 - the content of glycogen, BCM2 - the content of acid phosphatase, BCM3 - the content of lactate, BCM4 - the content of lactate dehydrogenase (LDH), BCM5 - the content of lipofuscin, BCM6 - the content of cholinesterase. MT was taken with use of special instruments, MT homogenates were prepared following the standard technique. Cytological studies of MT preparations of myocardium as well as their photographic recording were made on an Axiostar microscope (Zeiss, FRG). The optic density (OD) of nuclei and cytoplasm of cardiomyocytes (CMC) in conventional units of OD was measured using VideoTest program (Russia). RESULTS: It was found out that changes in MT of myocardium during the early PMP were characterized by the morphological, biochemical and biophysical regularities that we revealed; their most demonstrative features were as follows: - a gradual and constant reduction of the relative OD of CMC nuclei (YM-7) and cytoplasm (YM-8) during 3-13 hours from the moment of death, the rate and stage of these dynamics depending nonlinearly upon PDC; we substantiated and received quantitative regularities (polynomials) for the above biophysical indicators. CONCLUSIONS: A comparative morphological study of the ultrastructure of CMC at the early PMP depending upon PDC was performed; - the early PMP is characterized by proper biochemical changes in MT, the most demonstrative of them are as follows: a reduction in the content of glycogen (YM-1)and a dynamic increase in the content of lipofuscin(YM- 5).For all six BCM, representative absolute and relative values of their content in MMH depending upon PDC were obtained; - paired correlative values between biochemical and biophysical markers of the state of MT of myocardium were examined in their systemic relationships and proper SCC were determined by six time intervals of the early PMP, thereby making it possible to substantiate those of them that were criterially significant for increasing the accuracy of diagnosis of PDC.

OBJECTIVE: The aim: To assess the vitamin D level in blood plasma of patients with chronic heart failure and to identify the effect of its deficiency on the state of peripheral blood erythrocytes during physical exertion. PATIENTS AND METHODS: Materials and methods: A total of 25 patients with CHF grade IIA stages II and III functional class were examined. The control group consisted of 25 relatively healthy people. All patients were offered to complete a 6 minutes walking test. The level of 25 (OH) D total in plasma was determined by enzyme immunoanalysis. Morphological studies of erythrocytes were performed on the light-optical (Leica CME) and electron-microscopic level («JEOL-25M-T220¼). RESULTS: Results: Patients with chronic heart failure experienced 22.9% decrease in their vitamin D level (17.2±0.04 ng/ml) compared to the control group (38.4±0.05 ng/ml). Correlation analysis showed a direct proportional relationship between vitamin D deficiency and the number of erythrocytes of a modified form (r = 0.58; p <0.05) and erythrocytes with low osmotic resistance (r = 0.87; p <0.05). During the timed physical evaluation patients who experienced chronic heart failure accompanied by vitamin D deficiency developed an increase in the number of their reversibly and irreversibly deformed erythrocytes and a decrease in the cellular osmotic tability. CONCLUSION: Conclusions: During physical exertion, patients who experienced chronic heart falure accompanied by with vitamin D deficiency experienced morpho-biochemical changes in their red blood cells. These changes indicated structural disturbances in the membranes of their erythrocytes and could potentially have negative consequences for the somatic health of these patients.

This study was performed to determine quality changes of battered and breaded pre-fried snack product developed from large sized (5.5-10.5 kg) Catla (Catla catla) fish stored at chilled temperature (- 2 to - 4 °C). Quality changes were assessed based on biochemical, microbiological and sensory quality evaluation at 2 days interval for a period of 18 days. Results of biochemical analysis showed increase in total volatile base-nitrogen (TVB-N) values from initial value of 4.15-13.74 mg/100 g, free fatty acid (FFA) increased from 0.95 to 2.98% in terms of oleic acid, peroxide value (PV) increased from 2.6 to 3.9 meq of O2/kg of fat and pH increased from 6.5 to 6.79 during 18 days chilled storage. Linearity (r2) of TVB-N, FFA, PV and pH values during storage were found to be 0.98, 0.99, 0.97 and 0.99 respectively. In microbiological quality evaluation total plate count (Log. Values) increased from 2.17 to 4.65, while Escherichia coli, Staphylococcus aureus, Salmonella and spore count were not detected during storage period. Results of sensory analysis reported a negative correlation with storage period concluding the shelf life of battered and breaded fish cutlet from Catla fish to be 12 days at chilled storage conditions.

BACKGROUND: Biochemical and metabolic changes in stored RBC may influence the clinical outcome. We aimed to study the temporal changes in the biochemical parameters and the effect of buffy-coat reduction on RBC storage lesions. MATERIALS AND METHODS: A prospective observational study was conducted on fifteen RBC units five each of buffy coat reduced CPD/SAGM (quadruple bags), non-buffycoat reduced CPD/SAGM (triple bags) and non-buffycoat reduced CPDA (double bags). Biochemical parameters such as K+, LDH, pH plasma hemoglobin and percentage hemolysis were measured sequentially on day 7,14, 21, 28, 35 and 42. The data was analyzed using SPSS version 20. RESULTS: Extracellular K+ and LDH increased rapidly starting from the first week of storage. And the all the parameters including percentage hemolysis were significantly higher in RBC stored in CPDA (double bags) compared to that stored in SAGM (triple and quadruple). The difference observed in buffy-coat reduced units in comparison to the non-leukocyte reduced units were statistically not significant. CONCLUSION: The quality of red cells stored in SAGM was superior to that suspended in CPDA measured in terms of percent hemolysis, plasma hemoglobin, potassium and LDH. There was no effect of buffy-coat leukocyte reduction on the red cell storage lesion.

Attenuated total reflection-Fourier transform infrared (ATR-FTIR) spectroscopy is a label-free, non-destructive analytical technique for biochemical analysis of macromolecular components within tissue samples. Cadmium (Cd) and mono-(2-ethylhexyl) phthalate (MEHP), a primary metabolite of di-(2-ethylhexyl) phthalate, are present ubiquitously in the environment and in organisms, and have adverse impacts on ecosystems and human health. Herein we employed ATR-FTIR analysis to identify biomolecular changes in rat liver, spleen, lung and kidney after prepubertal exposure to Cd and MEHP. Our results showed clear segregations between the 3 mg/kg Cd-, 10 mg/kg, 50 mg/kg, 250 mg/kg MEHP- and binary mixture-treated groups vs. the solvent control group. Following principal components analysis coupled with linear discriminant analysis, biochemical alterations associated with different doses of Cd and MEHP were attributed mainly to lipids, proteins, phosphates and carbohydrates. In addition, the ratios of lipid/protein, C=O stretching/CH2 methylene (lipid oxidation level), amide I/amide II, α-helix/ß-sheet and CH3 methyl/CH2 methylene (acetylation level) in target organs were affected by these toxicants. There seems to be no dose-response effect of Cd and MEHP on target organs. We observed hardly any joint toxic action of these toxicants. This is the first study showing the application of ATR-FTIR spectroscopy to the assessment of toxicity of Cd and MEHP. Possibly, destruction of cell membrane structure and integrity could be the common mechanism of Cd and MEHP toxicity in liver, spleen, lung and kidney.

The aim of this study was to identify biochemical changes in sciatic nerve (SN) after crush injury and low-level laser therapy (LLLT) with 660 nm and 808 nm by Raman spectroscopy (RS) analysis. A number of 32 Wistar rats were used, divided into four groups (control 1, control 2, LASER 660 nm, and LASER 808 nm). All animals underwent surgical procedure of the SN and groups control 2, LASER 660 nm, and LASER 808 nm were submitted to SN crush damage (axonotmesis). The LLLT in the groups LASER 660 nm and LASER 808 nm was applied daily for 21 consecutive days (100 mW, 30 s, 133 J/cm2 fluence). The hind paw was removed and the SN was dissected and positioned on an aluminum support to collect dispersive Raman spectra (830 nm excitation, 30 s accumulation). To estimate the biochemical changes in the SN associated with LLLT, the principal component analysis (PCA) was applied. The Raman spectra of the sciatic nerve fragments showed peaks of the major biochemical components of the nerve, especially sphingolipids, phospholipids, glycoproteins, and collagen. The spectral features identified in some of the principal component loading vectors are referred to the biochemical elements present on the SN and were increased in the groups treated with LLLT, mainly lipids (sphingo and phospholipids) and proteins (collagen)-constituents of the myelin sheath. The RS was effective in identifying the biochemical differences in the SN after the crush injury, and LASER 660 nm was more efficient than the LASER 808 nm in cell proliferation and repair of the injured SN.

The distribution of boron (B) in leaves is far from uniform, and tolerance to B toxicity should be varied in different portions of an entire leaf. Here, according to the order and degree of leaf chlorosis, a whole leaf blade of trifoliate orange [Poncirus trifoliata (L.) Raf.] rootstock was divided into two segments-leaf tip and leaf center, and transmission electron microscope (TEM) and fourier transform infrared spectroscopy (FTIR) were used to obtain more detailed information on the cell ultrastructure and component architecture of the two leaf segments under B toxicity. Results revealed that B toxicity led to alterations in pectin network crosslinking structure of leaf tip and destruction of cell wall integrity. Moreover, B toxicity altered protein structure and decreased protein content, while increased carbohydrate content in the two leaf segments, especially in leaf tip. Excess B supply reduced the cellulose content in leaf tip but increased in leaf center. TEM micrographs exhibited chloroplast disintegration and plastoglobulus accumulation in cells of two different leaf sections of B-toxicity plants, with less pronounced changes in leaf center. Furthermore, B toxicity only induced accumulation of starch grains in cells of leaf center. Overall results indicated that the B-toxic-induced biochemical changes of the cell ultrastructure and component architecture greatly differed in leaf tip and center. This study facilitates a better understanding of structural changes in different leaf portions of P. trifoliata under B toxicity stress and provides new ideas for further research on other elements in different plant leaf portions.

OBJECTIVE: Little is known about the natural changes in parathyroid function after successful parathyroid surgery for primary hyperparathyroidism. The association of intact parathyroid hormone (iPTH) and calcium (Ca) with "temporary hypoparathyroidism" and "hungry bone syndrome" (HBS) was evaluated. DESIGN: Potential risk factors for temporary hypoparathyroidism and HBS were evaluated by taking blood samples before surgery, intra-operatively, at postoperative day (POD) 1, at POD 5 to 7, in postoperative week (POW) 8 and in postoperative month (POM) 6. PATIENTS: Of 425 patients, 43 (10.1%) had temporary hypoparathyroidism and 36 (8.5%) had HBS. MEASUREMENTS: The discriminative ability of iPTH and Ca on POD 1 for temporary hypoparathyroidism and HBS. RESULTS: Intact parathyroid hormone (IPTH) on POD 1 showed the highest discriminative ability for temporary hypoparathyroidism (C-index = 0.952), but not for HBS. IPTH was helpful in diagnosing HBS between POD 5 and 7 (C-index = 0.708). Extending the model by including Ca resulted in little improvement of the discriminative ability for temporary hypoparathyroidism (C-index = 0.964) and a decreased discriminative ability for HBS (C-index = 0.705). Normal parathyroid metabolism was documented in 139 (32.7%) patients on POD 1 and in 423 (99.5%) 6 months postoperatively, while 2 (0.5%) patients had persistent hyperparathyroidism, one diagnosed between POD 5 and 7 and another at POW 8. No patients suffered from permanent hypoparathyroidism. CONCLUSIONS: The necessity for Ca and vitamin D3 substitution cannot be predicted with certainty before POD 5 to 7 without serial laboratory measurements. Based on the results, a routine 8-week course of Ca and vitamin D3 treatment seems reasonable and its necessity should be evaluated in a follow-up study.
