During the study of ectoparasites (Platyhelminthes) of fish in the floodplain of the Upper Paraná River - Brazil, a new species of Urocleidoides, present in the gill filaments of Hemiodus orthonops (Hemiodontidae), is described using morphological description and molecular data from the mitochondrial region of cytochrome c oxidase, subunit 1 (COI) and the partial region of the 28S rDNA gene. Urocleidoides luquei n. sp. differs from all its congeners by the presence of a filament that joins the accessory piece to the base of the male copulatory organ and resembles U. paradoxus and U. surianoae. Phylogenetic analyses using molecular data revealed that U. luquei n. sp. forms a paraphyletic group concerning the other Urocleidoides species. In this way, as well as contributing to the description of a new species, we seek to encourage and contribute to the increase in research using integrative taxonomy, thus making it possible to elucidate some unresolved questions about the genus Urocleidoides.

Based on an integrative approach, this study describes a new species of Urocleidoides infesting Schizodon nasutus in the Paranapanema River basin, Brazil. The new species can be distinguished from its congeners by specific morphological features, including the shape of the male copulatory organ and accessory piece, the ventral bar shape, and the shape and size of the hooks. Molecular analyses, particularly of the 28S rDNA gene, suggest a close relationship between the new species and Urocleidoides paradoxus. The phylogenetic and taxonomic arrangement of Urocleidoides is discussed, as the analyses of the 28S rDNA and COI mtDNA resolved the genus as non-monophyletic, with Diaphorocleidus, Rhinoxenus, and Cacatuocotyle nested within it. Additional morphological and molecular data of other congeneric species are required to investigate the phylogenetic position and classification of Urocleidoides. This study underscores the significance of using integrative approaches in understanding host-parasite associations and phylogenetic relationships, contributing to the description of the freshwater fish parasite biodiversity in South America, particularly in the Paranapanema river basin.

Protein kinases are enzymes involved in essential biological processes such as signal transduction, transcription, metabolism, and the cell cycle. Human kinases are targets for several drugs approved by the US Food and Drug Administration. Therefore, the identification and classification of kinases in other organisms, including pathogenic parasites, is an interesting subject of study. Monogeneans are platyhelminths, mainly ectoparasites, capable of causing health problems in farmed fish. Although some genomes and transcriptomes are available for monogenean species, their full repertoire of kinases is unknown. The aim of this study was to identify and classify the putative kinases in the transcriptomes of two monogeneans, Rhabdosynochus viridisi and Scutogyrus longicornis, and then to predict potential monogenean drug targets (MDTs) and selective inhibitor drugs using computational approaches. Monogenean kinases having orthologs in the lethal phenotype of C. elegans but not in fish or humans were considered MDTs. A total of 160 and 193 kinases were identified in R. viridisi and S. longicornis, respectively. Of these, 22 kinases, belonging mainly to the major groups CAMK, AGC, and TK, were classified as MDTs, five of which were evaluated further. Molecular docking analysis indicated that dihydroergotamine, ergotamine, and lomitapide have the highest affinity for the kinases BRSK and MEKK1. These well-known drugs could be evaluated in future studies for potential repurposing as anti-monogenean agents. The present study contributes valuable data for the development of new antiparasitic candidates for finfish aquaculture.

In this study, a new dactylogyrid species is described from the gills of two ariid fish species of the Brazilian coast, Genidens barbus and Genidens genidens, by combining morphological characters with partial 18S rDNA sequences. The new species can be distinguished from its congeners by the following morphological characteristics: a male copulatory organ (MCO), a coiled tubular shaft of around one counterclockwise ring, base with a sclerotized cap; MCO with tapered distal region; a T-shaped accessory piece; a vagina, a cup-shaped vaginal vestibule, slightly sclerotized; and a pharynx comprising a muscular, glandular bulb. Supplementary taxonomic data for Chauhanellus velum from Sciades couma (type-host) are also presented and new 18S rDNA sequences of Chauhanellus spp. from other ariid fish from South America are provided. Phylogenetic analyses based on partial 18S rDNA gene sequences placed Chauhanellus riograndinensis n. sp. and Chauhanellus velum as two early divergent lineages within Chauhanellus from South America. Finally, a way to test the monophyly of Chauhanellus and Hamatopeduncularia is also discussed, which may be useful for future studies.

Cosmetocleithrum Kritsky, Thatcher & Boeger, 1986 (Dactylogyridae) represents one of the most species-rich groups (22 species currently recognized as valid) of all dactylogyrid parasites infecting Neotropical catfishes. Species of Cosmetocleithrum exhibit a remarkable affinity towards catfishes of the Doradidae and the Auchenipteridae. However, phylogenetic relationships between members of this genus have not been yet analysed. This study analysed newly obtained partial sequences of the 28S ribosomal RNA gene of seven species of Cosmetocleithrum, including its type species C. gussevi Kritsky, Thatcher & Boeger, 1986, along with several other dactylogyrids infecting siluriform, gymnotiform, perciform and characiform fishes. Cosmetocleithrum appeared as an evolutionary recent group, composed of two well-defined lineages: lineage 1 includes parasites of doradids - namely, C. bulbocirrus, C. confusum, C. parvum and C. bifurcum - whereas lineage 2 is composed of species from doradids - that is, C. rarum, C. gussevi, C. gigas, C. trachydorasi and C. falsunilatum - together with parasites of auchenipterids - namely, C. laciniatum and C. baculum. The search for synapomorphies to characterize taxonomic groups within Cosmetocleithrum appears challenging, since the morphology of their haptoral elements is quite conservative, and that of the copulatory complex is highly variable between species. The results of the present study support the recent synonymization of Paracosmetocleithrum Acosta, Scholz, Blasco-Costa, Alves & Silva, 2018 with Cosmetocleithrum. Whereas the 28S ribosomal DNA data resolved Cosmetocleithrum as monophyletic, the statistical support for the lineage was low, rendering its phylogenetic position between other Neotropical dactylogyrids yet undefined.

Phylogenetic framework for the closely related Ancylodiscoidinae and Ancyrocephalinae subfamilies remains contentious. As this issue was never studied using a large molecular marker, we sequenced the first two Ancylodiscoidinae mitogenomes: Thaparocleidus asoti and Thaparocleidus varicus. Both mitogenomes had two non-coding regions (NCRs) that contained a number of repetitive hairpin-forming elements (RHE). Due to these, the mitogenome of T. asoti (16,074 bp) is the longest among the Monogenea; especially large is its major NCR, with 3500 bp, approximately 1500 bp of which could not be sequenced (thus, the total mitogenome size is ≈ 17,600 bp). Although RHEs have been identified in other monopisthocotyleans, they appear to be independently derived in different taxa. The presence of RHEs may have contributed to the high gene order rearrangement rate observed in the two mitogenomes, including the first report of a transposition of rRNA genes within the Neodermata. Phylogenetic analyses using mitogenomic dataset produced Dactylogyrinae embedded within the Ancyrocephalinae (paraphyly), whereas Ancylodiscoidinae formed a sister-group with them. This was also supported by the gene order analysis. 28S rDNA dataset produced polyphyletic Dactylogyridae and Ancyrocephalinae. The phylogeny of the two subfamilies shall have to be further evaluated with more data.

Ligictaluridus michaelalicea n. sp. (Monogenea: Dactylogyridae, Ancyrocephalinae) is described from the gills of Pylodictis olivaris (Siluriformes: Ictaluridae) from Wisconsin and Iowa portions of the upper Mississippi River. Diagnostic features include a relatively large, strongly curved tubular cirrus with minor terminal flare; an accessory piece with a prominent basal lobe and a simple, thick terminal limb featuring a thin lateral flange and blunt distal tip devoid of recurved hooks. The sinistral vagina, two prostatic reservoirs, and a terminal seminal vesicle of the vas deferens are prominent. The anchors, ventral and dorsal bars, and hooks are similar in overall form to those of other members of the genus. The description includes sequence data for the 18S rRNA gene, which aligned most closely with species of ancyrocephaline monogeneans, with the highest similarity being with Ligictaluridus pricei (Mueller, 1936). Other monogenean species identified from the flathead catfish examined included L. pricei and Ligictaluridus mirabilis (Mueller, 1937). L. michaelalicea n. sp. is the fourth species to be described from P. olivaris within its natural range in central and eastern North America. Implications resulting from taxonomic name changes, including species of Ligictaluridus, and United States Food and Drug Administration treatment regulations are discussed. An updated key to species of the genus Ligictaluridus is presented.

Dawestrema cycloancistrium is the main ectoparasite causing mortality in fingerlings of pirarucu (Arapaima gigas) in Amazonian aquaculture. Very little is known about the D. cycloancistrium reproductive index and appropriate methods of collecting eggs for investigation. This study aimed to determine the oviposition rate of D. cycloancistrium. To achieve this aim, two egg quantification methods were tested: the estimative method (ME) and the total counting method (MT). Compared with the MT, the ME overestimated the number of eggs counted, which were 2943.5 ± 2840.6 and 1041.5 ± 533 eggs, and the oviposition rate, which was 80.1 ± 58.7 and 31.4 ± 16.4 eggs/parasite/day, for ME and MT, respectively. These results show that for studies quantifying D. cycloancistrium eggs, the total eggs in the sample must be counted, as the estimates made using subsamples are not representative. Using the MT, the oviposition rate for D. cycloancistrium was determined to be 31.4 ± 16.4 eggs per adult parasite per day. The present study demonstrates the egg production capacity of a monogenean species parasite of A. gigas, providing basic biological data for D. cycloancistrium.

Dactylogyrid monogeneans of Labeo horie Heckel, 1847 and L. niloticus (Linnaeus, 1758) (Cyprinidae) were surveyed at two sites on the River Nile in Sudan. The present study reports the presence of ten species of Dactylogyrus Diesing, 1850 and three species of Dogielius Bychowsky, 1936 based on a morphometric evaluation of the sclerotised structures. The species found include: Dactylogyrus nathaliae Guégan, Lambert & Euzet, 1988; D. rastellus Guégan, Lambert & Euzet, 1988; D. retroversus Guégan, Lambert & Euzet, 1988; D. senegalensis Paperna, 1969, D. yassensis Musilová, Rehulková & Gelnar, 2009 and five other undescribed species of Dactylogyrus. The genus Dogielius was represented by Dogielius flosculus Guégan, Lambert & Euzet, 1989; the newly identified D. sennarensis n. sp., and one undescribed Dogielius species. While D. sennarensis n. sp. resembles D. intorquens, it differs from this species and other congeners by having a longer ventral bar and anchor points and nosclerotised vagina. In addition, this study redescribes D. flosculus based on the morphology of specimens collected from L. horie. All specimens studied had a single large vagina, in contrast to the original description which reported a vagina composed of two unconnected parts. All dactylogyrid species in this study represent new host and geographical records.

We studied the monogenean communities of 34 species of butterflyfish from the tropical Indo-West Pacific, identifying 13 dactylogyrid species (including two species that are presently undescribed). Monogenean assemblages differed significantly between host species in terms of taxonomic structure, intensity and prevalence. Parasite richness ranged from 0 (Chaetodon lunulatus) to 11 (C. auriga, C. citrinellus and C. lunula). Host specificity varied between the dactylogyrids species, being found on 2-29 of the 34 chaetodontid species examined. Sympatric butterflyfish species were typically parasitized by different combinations of dactylogyrid species, suggesting the existence of complex host-parasite interactions. We identified six clusters of butterflyfish species based on the similarities of their dactylogyrid communities. Dactylogyrid richness and diversity were not related to host size, diet specialization, depth range or phylogeny of butterflyfish species. However, there was a weak positive correlation between monogenean richness and diversity and host geographical range. Most communities of dactylogyrids were dominated by Haliotrema aurigae and H. angelopterum, indicating the importance of the genus Haliotrema in shaping monogenean communities of butterflyfishes. This study casts light on the structure of the monogenean communities of butterflyfishes, suggesting that the diversity and complexity of community structures arises from a combination of host species-specific parameters.

Four new and four previously described species of Annulotrema were collected from the gills of four species (three genera, i.e. Alestes, Hydrocynus and Brycinus) of African tetras from Lake Turkana, Kenya: Annulotrema alestesnursi Paperna, 1973 from Brycinus nurse; Annulotrema ansatum n. sp., Annulotrema besalis Rehulková, Musilová and Gelnar, 2014, Annulotrema bipatens n. sp., Annulotrema cucullatum n. sp., Annulotrema nili Paperna, 1973, and Annulotrema pontile n. sp. from Hydrocynus forskahlii; and Annulotrema elongata Paperna and Thurston, 1969 from Alestes baremoze and Alestes dentex. A. elongata is re-described on the basis of new material from A. baremoze. The sclerotized structures of the haptor and male copulatory organ of A. alestesnursi and A. elongata are illustrated from their type material. H. forskahlii is a new host record for A. besalis. The findings of A. besalis and A. elongata in Kenya represent a new locality records for these helminths. Three Annulotrema spp., namely A. besalis, A. elongata and A. pontile n. sp., share the same type of male copulatory organ, which may indicate a close relationship among these species.

During investigations of gill ectoparasites (Platyhelminthes) parasitising freshwater fish from Central America (Guatemala, Honduras, El Salvador and Panama) and southeastern Mexico (Guerrero, Oaxaca and Chiapas), the following dactylogyrid monogenoidean were found: Urocleidoides simonae sp. n. from Profundulus punctatus (Günther) (type host), Profundulus balsanus Ahl, Profundulus guatemalensis (Günther), Profundulus kreiseri Matamoros, Shaefer, Hernández et Chakrabarty, Profundulus labialis (Günther), Profundulus oaxacae (Meek), Profundulus sp. 1 and Profundulus sp. 2 (all Profundulidae); Urocleidoides vaginoclaustroides sp. n. from Pseudoxiphophorus bimaculata (Heckel) (type host) and Poeciliopsis retropinna (Regan) (both Poeciliidae); and Urocleidoides vaginoclaustrum Jogunoori, Kritsky et Venkatanarasaiah, 2004 from P. labialis, Profundulus portillorum Matamoros et Shaefer and Xiphophorus hellerii Heckel (Poeciliidae). Urocleidoides simonae sp. n. differs from all other congeneric species in having anchors with well-differentiated roots, curved elongate shaft and short point. Urocleidoides vaginoclaustroides sp. n. most closely resembles U. vaginoclaustrum, but differs from this species mainly in the shape of its anchors (i.e. evenly curved shaft and short point vs curved shaft and elongate point extending just past the tip of the superficial anchor root). The complexity of potential hosts for species of Urocleidoides and their effect on its distribution on profundulid and poeciliid fishes are briefly discussed.

Piaractus brachypomus (Pacú) is the main native fish species cultivated in Peru and holds great potential for growth in aquaculture from the Peruvian Amazon. Between October 2021 and January 2022 in two fish producing farms in the Amazon region of San Martín in Peru, P. brachypomus individuals were examined for parasite evaluation. A total of 6366 monogeneans were isolated from the gills of 30 fish, revealing a prevalence of 100%, with an abundance and mean intensity of 212 parasites per fish. Monogeneans were morphologically identified as Mymarothecium viatorum and Anacanthorus penilabiatus. The genetic divergence in the 28S rDNA gene found among A. penilabiatus sequences was 0.1% and among Anacanthorus spp. it ranged from 0.9% to 7.5%. The genetic divergence found among the M. viatorum sequences was 0.3%. These finding represents the first molecular data of M. viatorum and A. penilabiatus in Peru using the 28S rDNA gene of these monogeneans. The new sequences obtained will contribute to future studies on the phylogenetic relationships among dactylogyrids. However, further research with a broader range of host-parasite samples and additional genetic markers is needed to clarify these relationships and provide stronger support for the phylogenetic positions.

Monogeneans are flatworm parasites that infest fish gills primarily but can also infect various other vertebrates, including amphibians, aquatic reptiles, mammals like hippos, and occasionally invertebrates like copepods, isopods, and cephalopods. Despite their remarkable diversity, our knowledge of monogenean parasites in Peru remains significantly limited, resulting in substantial gaps in our comprehension of their taxonomic identities, host associations, and geographic distribution. To address these knowledge deficits, we present an extensively curated checklist of monogeneans associated with aquatic vertebrates in Peru. This comprehensive compilation is derived from meticulous literature surveys, the examination of specimens deposited in both international and national collections, and the inclusion of additional freshly collected specimens. The checklist offers a thorough repository of data encompassing the diversity, host associations, and geographical distribution of these parasites. Taxonomic discrepancies are addressed through a critical review of the existing literature, supplemented by the direct examination of specimens, including type or voucher specimens, deposited within scientific collections. Additionally, we provide data on the DNA sequences of individual taxa. The compiled list comprises records of 358 monogenean species, including 270 valid species and 88 taxa identified at the family or generic level, all reported across 145 host species in Peru. Predominantly, these parasitic species exhibit associations within fish, with 335 infecting teleosts and 20 affecting chondrichthyans. Three monogenean species have been documented as infecting amphibians, namely Mesopolystoma samiriensis, Polistoma sp. and Wetapolystoma almae. Among the monogeneans reported, 141 were found in marine environments and 214 in freshwater environments. The most diverse families were Dactylogyridae and Diplectanidae, comprising 217 and 24 species, respectively. The hosts that harbored the highest number of monogeneans were Pygocentrus nattereri (with 23 species), followed by Stellifer minor (13 spp.) and Triportheus angulatus (11 spp.). We detected many species that do not have any material deposited in a scientific collection due to the loss or deactivation of the collection. These findings represent only a fraction of the potential diversity, considering the wide variety of aquatic vertebrate hosts inhabiting the tropical and subtropical regions of Peru.

Purple-spotted bigeyes Priacanthus tayenus Richardson (Priacanthidae) and bartail flathead Platycephalus indicus (Linnaeus) (Platycephalidae) were collected from the Arabian Gulf and examined for species of Monogenoidea (Polyonchoinea) from February to December 2020. Diplectanum robustitubum Wu & Li, 2003 and an undescribed species of Platycephalotrema Kritsky & Nitta, 2019 were recovered from the gill lamellae of these hosts, respectively. Diplectanum robustitubum from Iraq was redescribed and transferred to Oliveriplectanum Domingues & Boeger, 2008 (Diplectanidae) as Oliveriplectanum robustitubum (Wu & Li, 2003) n. comb. Platycephalotrema parile n. sp. (Dactylogyridae) from Iraq and Kuwait was described and differentiated from the similar species, Haliotrema indicum Tripathi, 1959, Platycephalotrema ogawai Kritsky & Nitta, 2019, and Platycephalotrema platycephali (Yin & Sproston, 1948) Kritsky & Nitta, 2019, based primarily on the comparative morphologies of the vaginal sclerites. Haliotrema indicum was transferred to Platycephalotrema as Platycephalotrema indicum (Tripathi, 1959) n. comb. and Haliotrema swatowense Yao, Wang, Xia, & Chen, 1998 was considered a junior subjective synonym of P. indicum. The finding of O. robustitubum in the Arabian Gulf represents a new geographic record for the species. BACKGROUND: The present paper represents the third installment concerning the monogenoids collected during surveys to explore their diversity on the marine and freshwater fishes of Iraq. Previous installments on the monogenoids emanating from the surveys included the dactylogyrid and gyrodactylid species parasitizing mugilid fishes. PURPOSE: The purpose of this paper is to further document the diversity of monogenoids infecting the fishes of Iraq. METHODS: Marine fishes were necropsied for parasites, and standard procedures for collecting, mounting, drawing, and measuring of monogenoids were employed. RESULTS: Oliveriplectanum robustitubum n. comb. (Diplectanidae) and Platycephalotrema parile n. sp. (Dactylogyridae) were collected. The occurrence of O. robustitubum in the Arabian Gulf represented a new locality record for the species. CONCLUSION: The recorded presence of O. robustitubum and P. parile n. sp. suggests that the diversity of monogenoids in Iraq is under estimated in the literature.

RESUMO

Five new species of Cosmetocleithrum were described parasitizing the gill filaments of neotropical doradid fishes. Cosmetocleithrum undulatum n. sp., Cosmetocleithrum brachylecis n. sp. and Cosmetocleithrum ludovicense n. sp. are described from Platydoras brachylecis from a market-place of São Luís, State of Maranhão, Brazil. Cosmetocleithrum sacciforme n. sp. and Cosmetocleithrum basicomplexum n. sp. are described from Oxydoras niger from Juruá River, State of Acre, Brazil. Cosmetocleithrum undulatum and Cosmetocleithrum brachylecis resemble Cosmetocleithrum falsunilatum Feronato, Razzolini, Morey & Boeger, 2022 mainly by the unique male copulatory organ (MCO) morphology but differ from these and all congeneric species mainly by the morphology of the MCO, accessory piece and hooks pairs. Cosmetocleithrum ludovicense is closer to Cosmetocleithrum confusus Kritsky, Thatcher & Boeger, 1986 and to Cosmetocleithrum akuanduba Soares, Santos Neto & Domingues, 2018 but differs from those mainly by the morphology of the accessory piece. Cosmetocleithrum sacciforme differs from all congeneric species mainly by the morphology of the accessory piece formed by a single plate of saccular appearance. Cosmetocleithrum basicomplexum also shares morphological characters with Cosmetocleithrum gigas Morey, Cachique & Babilonia, 2019 considering the size of the body and shape of the anchors, but differs mainly in the morphology of the bars and hooks. Besides the new species, new data are presented for Cosmetocleithrum leandroi Soares, Neto & Domingues, 2018, C. akuanduba and C. confusus regarding morphological characteristics and biogeography.

During a research on morphological diversity of gill ectoparasites on native and non-native fishes from tributaries (Palizada, El Recreo and Lacantún rivers) of the Usumacinta River Basin in the states of Campeche, Tabasco, and Chiapas (southern Mexico), the following monogenoids were found: Icelanonchohaptor tropicalis n. sp. on Usumacinta buffalo Ictiobus meridionalis (Günther, 1868) (Catostomidae); Heteropriapulus simplexiodes n. sp. and Heteropriapulus heterotylioides n. sp. on catfishes Pterygoplichthys pardalis (Castelnau, 1855) (Loricariidae) (type host) and Pterygoplichthys disyunctivus (Weber, 1991); Ligictaluridus mirabilis (Mueller 1937; Klassen and Beverley-Burton1985 from the southern blue catfish Ictalurus meridionalis (Günther, 1864) (Ictaluridae); Aristocleidus mexicanus Mendoza-Franco and Vidal-Martínez, 2001 on Eugerres mexicanus (Steindachner, 1863) (Gerreidae) (all monogenoidean species in the Dactylogyridae); and Diplectanocotyla megalopis Rakotofiringa and Oliver1987 (Diplectanidae) on tarpon Megalops atlanticus Valenciennes, 1847 (Megalopidae). The new species of Icelanonchohaptor and Heteropriapulus are herein described for the first time from a native catostomid and non-native Pterygoplichthys spp., respectively. While I. tropicalis n. sp. and L. mirabilis are morphologically comparable with their congeners from the Nearctic (i.e., United States and Canada), all other monogenoids exhibited Neotropical affinities. Present study shown that the gill monogenoids on native and non-native fishes in the Neotropical Mexican transition zone of the Usumacinta River basin are equally represented by species with Nearctic and Neotropical affinities including those adapted to freshwater environment in this area from marine ancestry.

BACKGROUND: Unresolved taxonomic classification and paraphyly pervade the flatworm class Monogenea: the class itself may be paraphyletic and split into Polyopisthocotylea and Monopisthocotylea; there are some indications that the monopisthocotylean order Dactylogyridea may also be paraphyletic; single-gene markers and some morphological traits indicate that the family Ancyrocephalidae is paraphyletic and intertwined with the family Dactylogyridae. METHODS: To attempt to study the relationships of Ancyrocephalidae and Monopisthocotylea using a phylogenetic marker with high resolution, we sequenced mitochondrial genomes of two fish ectoparasites from the family Dactylogyridae: Dactylogyrus simplex and Dactylogyrus tuba. We conducted phylogenetic analyses using three datasets and three methods. Datasets were ITS1 (nuclear) and nucleotide and amino acid sequences of almost complete mitogenomes of almost all available Monopisthocotylea mitogenomes. Methods were maximum likelihood (IQ-TREE), Bayesian inference (MrBayes) and CAT-GTR (PhyloBayes). RESULTS: Both mitogenomes exhibited the ancestral gene order for Neodermata, and both were compact, with few and small intergenic regions and many and large overlaps. Gene sequences were remarkably divergent for nominally congeneric species, with only trnI exhibiting an identity value > 80%. Both mitogenomes had exceptionally low A + T base content and AT skews. We found evidence of pervasive compositional heterogeneity in the dataset and indications that base composition biases cause phylogenetic artefacts. All six mitogenomic analyses produced unique topologies, but all nine analyses produced topologies that rendered Ancyrocephalidae deeply paraphyletic. Mitogenomic data consistently resolved the order Capsalidea as nested within the Dactylogyridea. CONCLUSIONS: The analyses indicate that taxonomic revisions are needed for multiple Polyopisthocotylea lineages, from genera to orders. In combination with previous findings, these results offer conclusive evidence that Ancyrocephalidae is a paraphyletic taxon. The most parsimonious solution to resolve this is to create a catch-all Dactylogyridae sensu lato clade comprising the current Ancyrocephalidae, Ancylodiscoididae, Pseudodactylogyridae and Dactylogyridae families, but the revision needs to be confirmed by another marker with a sufficient resolution.

Genomic resources for Platyhelminthes of the class Monogenea are scarce, despite the diversity of these parasites, some species of which are highly pathogenic to their fish hosts. This work aimed to generate de novo-assembled transcriptomes of two monogenean species, Scutogyrus longicornis (Dactylogyridae) and Rhabdosynochus viridisi (Diplectanidae), providing a protocol for cDNA library preparation with low input samples used in single cell transcriptomics. This allowed us to work with sub-microgram amounts of total RNA with success. These transcriptomes consist of 25,696 and 47,187 putative proteins, respectively, which were further annotated according to the Swiss-Prot, Pfam, GO, KEGG, and COG databases. The completeness values of these transcriptomes evaluated with BUSCO against Metazoa databases were 54.1% and 73%, respectively, which is in the range of other monogenean species. Among the annotations, a large number of terms related to G-protein-coupled receptors (GPCRs) were found. We identified 109 GPCR-like sequences in R. viridisi, and 102 in S. longicornis, including family members specific for Platyhelminthes. Rhodopsin was the largest family according to GRAFS classification. Two putative melatonin receptors found in S. longicornis represent the first record of this group of proteins in parasitic Platyhelminthes. Forty GPCRs of R. viridisi and 32 of S. longicornis that were absent in Vertebrata might be potential drug targets. The present study provides the first publicly available transcriptomes for monogeneans of the subclass Monopisthocotylea, which can serve as useful genomic datasets for functional genomic research of this important group of parasites.


Title: Assemblage de novo du transcriptome et identification des récepteurs couplés aux protéines G (RCPG) chez deux espèces de Monogènes parasites de poissons. Abstract: Les ressources génomiques pour les Plathelminthes de la classe Monogenea sont rares, malgré la diversité de ces parasites dont certaines espèces sont hautement pathogènes pour leurs hôtes poissons. Ce travail visait à générer des transcriptomes assemblés de novo pour deux espèces de monogènes, Scutogyrus longicornis (Dactylogyridae) et Rhabdosynochus viridisi (Diplectanidae), fournissant un protocole pour la préparation de la bibliothèque d'ADNc avec des échantillons à faible apport utilisés en transcriptomique unicellulaire, ce qui a permis de travailler avec des quantités inférieures au microgramme d'ARN total avec succès. Seven new species of Dactylogyrus Diesing, 1850 (Dactylogyridae) are described from the gills of seven endemic species of cyprinoids (Cyprinidae, Leuciscidae) inhabiting the Balkan Peninsula: Dactylogyrus romuli n. sp. from Luciobarbus albanicus (Greece), Dactylogyrus remi n. sp. from Luciobarbus graecus (Greece), Dactylogyrus recisus n. sp. from Pachychilon macedonicum (Greece), Dactylogyrus octopus n. sp. from Tropidophoxinellus spartiaticus (Greece), Dactylogyrus vukicae n. sp. from Delminichthys adspersus (Bosnia and Herzegovina), Dactylogyrus leptus n. sp. from Chondrostoma knerii (Bosnia and Herzegovina), and Dactylogyrus sandai n. sp. from Telestes karsticus (Croatia). To delineate species boundaries, we used an integrative taxonomic approach combining morphological and genetic data. With these tools, we illustrate that some species of monogeneans considered as cryptic might be designated as pseudocryptic (morphologically similar, not easily differentiated) after a posteriori detailed morphological examination, as happened with D. romuli n. sp. and D. remi n. sp. Thus, for accurate species characterization, it is particularly important to acquire both morphological and molecular data from the same individual specimens, ideally along with illustrations of taxonomically important structures directly taken from hologenophores. Using phylogenetic reconstruction, we investigated the phylogenetic position of newly described Dactylogyrus species within Dactylogyrus species from Balkan cyprinoids with regard to morphological characteristics, host range, and geographical distribution.


TITLE: Découverte de la diversité des Monogènes (Plathelminthes) des poissons cypriniformes endémiques de la péninsule balkanique : nouvelles espèces de Dactylogyrus et commentaires sur leur phylogénie et les associations hôte-parasite dans un contexte biogéographique. Avec ces outils, nous illustrons que certaines espèces de Monogènes considérées comme cryptiques pourraient être désignées comme pseudocryptiques (morphologiquement similaires, pas facilement différenciées) après un examen morphologique détaillé a posteriori, comme cela s'est produit avec D. romuli n. sp. et D. remi n. sp. Ainsi, pour une caractérisation précise des espèces, il est particulièrement important d'acquérir des données morphologiques et moléculaires à partir des mêmes spécimens individuels, idéalement avec des illustrations de structures taxonomiquement importantes directement prises à partir d'hologénophores. En utilisant la reconstruction phylogénétique, nous avons étudié la position phylogénétique des espèces de Dactylogyrus nouvellement décrites au sein des espèces de Dactylogyrus des cyprinoïdés des Balkans en ce qui concerne les caractéristiques morphologiques, la gamme d'hôtes et la distribution géographique.
