Blood feeding and digestion are vital physiological activities essential for the survival and reproduction of ticks. Chemical acaricides viz., ivermectin, amitraz and fipronil, are known to act on the central nervous system, resulting in the mortality of ticks. The present study is focused on the effect of these acaricides on the midgut and gut enzymes of Rhipicephalus microplus. The ultra-thin sections of midgut of ivermectin-treated ticks showed irregular basal membrane and ruptured digestive vesicles. Amitraz treatment resulted in a notable decrease in digestive cells with pleats in the basal membrane, while fipronil-exposed ticks exhibited reduced digestive cells, loss of cellular integrity, and disintegration of the basal membrane and muscle layer. The gut tissue homogenate of ivermectin and fipronil treated ticks showed a significant reduction of cathepsin D level, 76.54 ± 3.20 µg/mL and 92.67 ± 3.72 µg/mL, respectively, as compared to the control group (150.0 ± 3.80 µg/mL). The leucine aminopeptidase level (4.27 ± 0.08 units/mL) was significantly decreased in the ivermectin treated ticks compared to other treatment groups. The acid phosphatase activity (29.16 ± 0.67 µmole/min/L) was reduced in the ivermectin treated group whereas, increased activity was observed in the fipronil and amitraz treated groups. All the treatment groups revealed increased alkaline phosphatase levels (17.47-26.72 µmole/min/L). The present finding suggests that in addition to the established mechanism of action of the tested acaricides on the nervous system, the alterations in the cellular profile of digestive cells and enzymes possibly affect the blood digestion process and thus the synthesis of vital proteins which are essential for vitellogenesis, and egg production in ticks.

Colorado potato beetle (Leptinotarsa decemlineata, Say) is the main pest of Solanaceae and its survival is mainly dependent on the carbohydrate digestion. Characterizing the gut enzymes may help us with finding effective inhibitors for plant protection. Activity measurements revealed that gut extracts contain α- and ß-glucosidase in addition to α-amylase. For larvae, amylase activity was detected only in gut saturated with nutrients. Leptinotarsa decemlineata α-amylase (LDAmy) had optimum pH of 6.0 and was active under 30-40°C temperature measured on a selective α-amylase substrate, 2-chloro-4-nitrophenyl-4-O-α-D-galactopyranosyl-maltoside. HPLC analysis demonstrated dimer, trimer, and tetramer reducing end amylolytic products from 2-chloro-4-nitrophenyl-maltoheptaoside substrate in similar ratio than that of during porcine pancreatic α-amylase (PPA) catalyzed hydrolysis. The 4,6-O-benzylidene-modified substrate (BzG7PNP) is very stable toward hydrolysis by exo-glycosidases, therefore is very useful to monitor the digestion catalyzed by α-amylases exclusively. Similarly to PPA active site, three glycon and two aglycon binding sites are suggested for LDAmy based on the pattern of early hydrolysis products of BzG7PNP. The observed similarity between LDAmy and PPA raises the possibility of using known inhibitors of mammalian α-amylases to protect the potato plant from attack of Colorado potato beetle.

An extracellular chitinase was identified and purified (CS1 and CS2) from Bacillus subtilis. The 16S rRNA sequencing was submitted in GenBank (accession numbers KC336487 and KC412256). The purified crude enzymes were identified through matrix-assisted laser desorption/ionization time-of-flight/time-of-flight mass spectrometry (MALDI-TOF MS) analysis. The peptide sequences were matched with chitinase sequences. The peak m/z with 1297. 592 and 3094.570 mascot search resulted sequence was blasted with NCBI protein sequences and confirmed that it is a chitinase enzyme. The effects of chitinase on gut enzymes lactate dehydrogenase, acid phosphatase, alkaline phosphatase and adenosine triphosphatase of the tobacco cutworm Spodoptera litura larvae were investigated. At all concentrations tested, chitinase decreased the activities of these gut enzymes relative to the control. When chitinase treated leaves were fed to larvae in bioassays, gut tissue and gut enzymes were affected. The histological study clearly shows the chitinase treated larval gut, peritrophic membrane and epithelial cells were affected significantly. Chitinase isolated from B. subtilis has effectively reduced the gut enzyme activity and growth of S. litura. The chitin based bioformulation may serve as an effective biocide against the polyphagous pest like S. litura.

The long-term exposure effects of nanodiamonds (NDs), spanning an organism's entire lifespan and continuing for subsequent generation, remain understudied. Most research has focused on evaluating their biological impacts on cell lines and selected organisms, typically over short exposure durations lasting hours or days. The study aimed to assess growth, mortality, and digestive functions in wild (H) and long-lived (D) strains of Acheta domesticus (Insecta: Orthoptera) after two-generational exposure to NDs in concentrations of 0.2 or 2 mg kg-1 of food, followed by their elimination in the third generation. NDs induced subtle stimulating effect that depended on the strain and generation. In the first generation, more such responses occurred in the H than in the D strain. In the first generation of H strain insects, contact with NDs increased survival, stimulated the growth of young larvae, and the activity of most digestive enzymes in mature adults. The same doses and exposure time did not cause similar effects in the D strain. In the first generation of D strain insects, survival and growth were unaffected by NDs, whereas, in the second generation, significant stimulation of those parameters was visible. Selection towards longevity appears to support higher resistance of the insects to exposure to additional stressor, at least in the first generation. The cessation of ND exposure in the third generation caused potentially harmful changes, which included, e.g., decreased survival probability in H strain insects, slowed growth of both strains, as well as changes in heterochromatin density and distribution in nuclei of the gut cells in both strains. Such a reaction may suggest the involvement of epigenetic inheritance mechanisms, which may become inadequate after the stress factor is removed.

BACKGROUND: The nefarious hemipteran mustard aphid (Lipaphis erysimi) inflicts colossal yield losses in Brassica crops including Indian mustard (Brassica juncea). Lack of an accessible resistance source has been the primary impediment in breeding varietal resistance against aphids. In recent years, in planta RNAi-mediated resistance has been demonstrated in model plants as a potential tool for protection against insect pests. However, translational application in crop species is imperative for critical assessment of this technology in breeding effective resistance. RESULTS: The essential role of sucrase 1 (SUC1) in mitigating osmotic pressure imposed by sucrose-rich phloem sap inside the insect gut is corroborated by its expression pattern in L. erysimi. Transgenic lines of Indian mustard were developed expressing SUC1 hairpin RNA for its host-mediated delivery into the infesting aphids. The expression of the dsRNA encoding cassette, and generation of siRNA molecules in transgenic B. juncea lines were verified by quantitative reverse transcription (RT)-PCR, stem-loop RT-PCR and Northern hybridization. Rearing of L. erysimi on the transgenic lines resulted in 22-40% reduction in aphid fecundity. The observed retardation in aphid reproduction was coherent with the detection of SUC1-specific siRNA molecules and attenuation of the SUC1 transcript level in L. erysimi fed on the transgenic lines. CONCLUSION: Augmenting varietal resistance can substantially reduce usage of toxic agrochemicals in crop protection. This attempt was the first successful demonstration of host-mediated RNAi of an aphid gene in any Brassica crop. It paves the way for more rigorous attempt of engineering RNAi-based resistance against aphids in Brassica crops. © 2021 Society of Chemical Industry.

Gastrointestinal (GI) cancer is a prevalent global health disease with a massive burden on health care providers. Internal and external factors such as obesity, smoking, diet (red meat), low socioeconomic status and infection with Helicobacter pylori are the critical risk factors of GI cancers. Flavonoids are natural phenolic compounds found abundantly in fruits and vegetables. Upon ingestion, 90% of flavonoids consumed require further enzymatic metabolism by the gut microbiome to enhance their bioavailability and absorption. Several epidemiological studies reported that consumption of flavonoids and their enzymatic conversion by gut microbes is strongly associated with the reduced risk of GI cancer development. This review summarizes the current knowledge on the enzymatic conversion of flavonoids by the human gut microbiome. It also addresses the underlying anti-GI cancer effects on metabolic pathways such as apoptosis and cellular proliferation. Overall, metabolites produced from flavonoid's enzymatic conversion illustrate anti-GI cancer effects, but the mechanisms of action need further clarification.

The growing demand for wood has resulted in large volumes of wood wastes that are daily released to the soil from the activities of sawmills in South-Western Nigeria. In an attempt to setup a bioremediation model for sawdust, this study therefore aimed at evaluating microbial diversity, and the level of digestive enzymes in the gut of earthworms (Eudrilus eugeniae, Libyodrilus violaceous and Hyperiodrilus africanus) of sawmill origin. Four major sawmills located in Abeokuta (7o9’12” N - 3o19’35” E), namely Lafenwa, Sapon, Isale-Ake and Kotopo sawmills were used for this study. The arboretum of the Federal University of Agriculture, Abeokuta was used as control. Gut microbial analysis was carried out using the pour-plate method while digestive enzyme activities in the earthworm guts were done by the spectrophotometric method. Higher microbial counts (28.5±0.1x10³-97.0±0.1x10³cfu for bacteria and 7.0±0.1x10³-96.0±0.1x10³cfu for fungi) and microbial diversity were recorded in the gut of earthworms of the sawmill locations than those of the control site (17.5±0.1x10³cfu for bacteria and 4.5±0.1x10³cfu for fungi). Streptococcus mutans and Proteus spp.were common in the gut of E. eugeniae, and L. violaceous from the study sawmills, while Streptococcus mutans were also identified in H. africanus, but absent in the gut of E. eugeniae from the control site. Cellulase (48.67±0.02mg/g) and lipase (1.81±0.01mg/g) activities were significantly higher (p<0.05) in the gut of earthworms from the control site than those of the study sawmills. Furthermore, amylase (α and β) activity was highest in the gut of earthworms from the sawmills. Variations observed in the gut microbial and digestive enzyme activities of earthworms from the study sawmills as compared to the control site suggests that earthworms, especially E. eugeniae, could be a better organism for use as bioremediator of wood wastes. La creciente demanda de madera ha resultado en grandes volúmenes de residuos de madera que se lanzan diariamente a la tierra, procedentes de las actividades de los aserraderos en el suroeste de Nigeria. Se realizó este estudio en un intento de configurar un modelo de biorremediación de aserrín, con el objetivo de evaluar la diversidad microbiana y el nivel de las enzimas digestivas en el intestino de las lombrices de tierra (Eudrilus eugeniae, Libyodrilus violáceo y Hyperiodrilus africanus) de origen aserradero. Para este estudio se utilizaron cuatro grandes aserraderos ubicados en Abeokuta (7°9’12 ‘’ N - 3°19’35 ‘’ E), a saber: Lafenwa, Sapon, Isale-Ake y Aserraderos Kotopo. El arboreto de la Universidad Federal de Agricultura, Abeokuta se utilizó como control. El análisis microbiano se llevó a cabo utilizando el método de vertido de placa, mientras que las actividades de enzimas digestivas en los intestinos de las lombrices se realizaron por el método espectrofotométrico. Los recuentos microbianos más altos fueron 28.5±97.0±0.1x10³-0.1x10³cfu para las bacterias y 7.0±0.1x10³-96.0±0.1x10³cfu para los hongos y la diversidad microbiana registrada en el intestino de las lombrices de tierra de los lugares de aserraderos y las del sitio de control fueron: 17.5±0.1x10³cfu para bacterias y 4.5±0.1x10³cfu para hongos. Streptococcus mutans y Proteus spp. eran comunes en el intestino de E. eugeniae, y L. violaceous de los aserraderos de estudio, mientras que también se identificaron Streptococcus mutans en H. africanus, que estuvieron ausentes en el intestino de E. eugeniae en el sitio de control. Actividades de celulasa (48.67±0.02mg/g) y lipasa (1.81±0.01mg/g) fueron significativamente mayores (p<0.05) en el intestino de las lombrices de tierra en el sitio de control que las de los aserraderos de estudio. Por otra parte, la actividad de la amilasa (α y β) fue mayor en el intestino de las lombrices de tierra de los aserraderos. Las variaciones observadas en la microbiota intestinal y la actividad de las enzimas digestivas de las lombrices de tierra de los aserraderos de estudio, en comparación con el sitio de control, sugieren que las lombrices de tierra, especialmente E. eugeniae, podrían ser un mejor organismo para su uso como biorremediador de residuos de madera.
