Topography of a place has a significant impact on soil characteristics that ultimately influence soil iodine levels. Lower Himalayan region (LHR) in Pakistan has a wide range of climatic and geological variations. Hence, an investigation was conducted to analyze the iodine concentration and other physicochemical properties of soils in two LHR districts, Haripur and Mansehra. Spatial analysis indicated a decrease in iodine levels in the mountainous regions in comparison to the flat portions of LHR. Soil samples obtained from different locations across Haripur had a stronger affinity for iodine due to variations in solubility and adsorption of iodine to soil clay components, which can be attributed to lower pH, higher organic matter, and a higher cation exchange capacity (CEC). In contrast to the plains of Haripur, elevated locations in the Mansehra district had decreased levels of iodine, along with a higher soil pH and reduced soil organic matter. The soil erosion and depletion of soil micronutrients in the hilly region of Mansehra may be attributed to the unfavorable soil conditions and excessive precipitation. Presence of clay, iron (Fe), and aluminum (Al) in the soil led to a rise in iodine levels. Iodine concentrations exhibited an inverse relationship with soil acidity. Study revealed a direct correlation between soil iodine levels and their cation exchange capacity (CEC) and clay content. This study aims to gather fundamental data for the chosen regions of LHR to address illnesses caused by iodine deficiency.

BACKGROUND: Digital surgery is a new paradigm within the surgical innovation space that is rapidly advancing and encompasses multiple areas. METHODS: This white paper from the SAGES Digital Surgery Working Group outlines the scope of digital surgery, defines key terms, and analyzes the challenges and opportunities surrounding this disruptive technology. RESULTS: In its simplest form, digital surgery inserts a computer interface between surgeon and patient. We divide the digital surgery space into the following elements: advanced visualization, enhanced instrumentation, data capture, data analytics with artificial intelligence/machine learning, connectivity via telepresence, and robotic surgical platforms. We will define each area, describe specific terminology, review current advances as well as discuss limitations and opportunities for future growth. CONCLUSION: Digital Surgery will continue to evolve and has great potential to bring value to all levels of the healthcare system. The surgical community has an essential role in understanding, developing, and guiding this emerging field.

BACKGROUND: Acceptance of COVID-19 vaccination was noticed to be less common among parents of children with autism spectrum disorder (ASD) and other neurodevelopmental disorders. This study aimed to explore the beliefs and willingness of parents of children with neurodevelopmental disorders about COVID-19 vaccine and understand how certain factors influencing the vaccine decision-making process differ between them and other parents' groups. METHODS: A cross-sectional study was conducted between August to November 2021. An Arabic online survey was distributed in August 2021 to collect the study's data. 400 parents from all the major regions in Saudi Arabia participated in and shared their beliefs about the new COVID-19 vaccination for their children. RESULTS: Out of 400 participants, 381 of them were eligible to answer the survey (95.2%). The total number of parents of children with neurodevelopmental disorder was 158 (41.5%), was compared to responses of parents of heathy children 223 (58.5%). 85 (53.8%) of them were ready to vaccinate their children with COVID-19 vaccine. While 36 (22.8%) were hesitant, the rest 37 (23.4%) did not want to vaccinate their children at all. Only a small number 16 (10.1%) have beliefs of vaccines as a cause of their child's neurodevelopmental disorder. A total of 79 out of 131 responses were received from both parents' groups. Fear of long-term side-effects was the most common reason reported by 41 responders out of 64 (64.06%) from parents of healthy children and 38 responders out of 67 (56.71%) from parents of diagnosed children. Another reason reported by parents of younger children in both groups was the child's age. Having a healthcare relative worker was significantly associated with the vaccine decision making (p < .001). CONCLUSION: The acceptance rate of COVID-19 vaccination of parents of children with neurodevelopmental disorders was low compared to the parents of healthy children in Saudi Arabia. Authorities can benefit from this study results to offer more accessible information about the vaccine importance and safety to the targeted population.

Trolox is a potent antioxidant and a water-soluble analog of vitamin E. It has been used in scientific studies to examine oxidative stress and its impact on biological systems. Trolox has been shown to have a neuroprotective effect against ischemia and IL-1ß-mediated neurodegeneration. In this study, we investigated the potential protective mechanisms of Trolox against a 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced Parkinson's disease mouse model. Western blotting, immunofluorescence staining, and ROS/LPO assays were performed to investigate the role of trolox against neuroinflammation, the oxidative stress mediated by MPTP in the Parkinson's disease (PD) mouse model (wild-type mice (C57BL/6N), eight weeks old, average body weight 25-30 g). Our study showed that MPTP increased the expression of α-synuclein, decreased tyrosine hydroxylase (TH) and dopamine transporter (DAT) levels in the striatum and substantia nigra pars compacta (SNpc), and impaired motor function. However, Trolox treatment significantly reversed these PD-like pathologies. Furthermore, Trolox treatment reduced oxidative stress by increasing the expression of nuclear factor erythroid-2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1). Lastly, Trolox treatment inhibited the activated astrocytes (GFAP) and microglia (Iba-1), also reducing phosphorylated nuclear factor-κB, (p-NF-κB) and tumor necrosis factor-alpha (TNF-α) in the PD mouse brain. Overall, our study demonstrated that Trolox may exert neuroprotection on dopaminergic neurons against MPTP-induced oxidative stress, neuroinflammation, motor dysfunction, and neurodegeneration.

The Coronavirus (COVID-19) has become a critical and extreme epidemic because of its international dissemination. COVID-19 is the world's most serious health, economic, and survival danger. This disease affects not only a single country but the entire planet due to this infectious disease. Illnesses of Covid-19 spread at a much faster rate than usual influenza cases. Because of its high transmissibility and early diagnosis, it isn't easy to manage COVID-19. The popularly used RT-PCR method for COVID-19 disease diagnosis may provide false negatives. COVID-19 can be detected non-invasively using medical imaging procedures such as chest CT and chest x-ray. Deep learning is the most effective machine learning approach for examining a considerable quantity of chest computed tomography (CT) pictures that can significantly affect Covid-19 screening. Convolutional neural network (CNN) is one of the most popular deep learning techniques right now, and its gaining traction due to its potential to transform several spheres of human life. This research aims to develop conceptual transfer learning enhanced CNN framework models for detecting COVID-19 with CT scan images. Though with minimal datasets, these techniques were demonstrated to be effective in detecting the presence of COVID-19. This proposed research looks into several deep transfer learning-based CNN approaches for detecting the presence of COVID-19 in chest CT images.VGG16, VGG19, Densenet121, InceptionV3, Xception, and Resnet50 are the foundation models used in this work. Each model's performance was evaluated using a confusion matrix and various performance measures such as accuracy, recall, precision, f1-score, loss, and ROC. The VGG16 model performed much better than the other models in this study (98.00 % accuracy). Promising outcomes from experiments have revealed the merits of the proposed model for detecting and monitoring COVID-19 patients. This could help practitioners and academics create a tool to help minimal health professionals decide on the best course of therapy.

Unreliable networks often use excess bandwidth for data integration in smart cities. For this purpose, Messaging Queuing Telemetry Transport (MQTT) with a certain quality of service (QoS) is employed. Data integrity and data security are frequently compromised for reducing bandwidth usage while designing integrated applications. Thus, for a reliable and secure integrated Internet of Everything (IoE) service, a range of network parameters are conditioned to achieve the required quality of a deliverable service. In this work, a QoS-0-based MQTT is developed in such a manner that the transparent MQTT protocol uses Transmission Control Protocol (TCP)-based connectivity with various rules for the retransmission of contents if the requests are not entertained for a fixed duration. The work explores the ways to improve the overall content delivery probability. The parameters are examined over a transparent gateway-based TCP network after developing a mathematical model for the proposed retransmission-based mutant QoS-0. The probability model is then verified by an actual physical network where the repeated content delivery is explored at VM-based MQTT, local network-based broker and a remote server. The results show that the repeated transmission of contents from the sender improves the content delivery probability over the unreliable MQTT-based Internet of Things (IoT) for developing smart cities' applications.

In the last few decades, environmental contaminants (ECs) have been introduced into the environment at an alarming rate. There is a risk to human health and aquatic ecosystems from trace levels of emerging contaminants, including hospital wastewater (HPWW), cosmetics, personal care products, endocrine system disruptors, and their transformation products. Despite the fact that these pollutants have been introduced or detected relatively recently, information about their characteristics, actions, and impacts is limited, as are the technologies to eliminate them efficiently. A wastewater recycling system is capable of providing irrigation water for crops and municipal sewage treatment, so removing ECs before wastewater reuse is essential. Water treatment processes containing advanced ions of biotic origin and ECs of biotic origin are highly recommended for contaminants. This study introduces the fundamentals of the treatment of tertiary wastewater, including membranes, filtration, UV (ultraviolet) irradiation, ozonation, chlorination, advanced oxidation processes, activated carbon (AC), and algae. Next, a detailed description of recent developments and innovations in each component of the emerging contaminant removal process is provided.

BACKGROUND: Time of diagnosis (TOD) of benign esophageal perforation is regarded as an important risk factor for clinical outcome, although convincing evidence is lacking. The aim of this study is to assess whether time between onset of perforation and diagnosis is associated with clinical outcome in patients with iatrogenic esophageal perforation (IEP) and Boerhaave's syndrome (BS). METHODS: We searched MEDLINE, Embase and Cochrane library through June 2018 to identify studies. Authors were invited to share individual patient data and a meta-analysis was performed (PROSPERO: CRD42018093473). Patients were subdivided in early (≤ 24 h) and late (> 24 h) TOD and compared with mixed effects multivariable analysis while adjusting age, gender, location of perforation, initial treatment and center. Primary outcome was overall mortality. Secondary outcomes were length of hospital stay, re-interventions and ICU admission. RESULTS: Our meta-analysis included IPD of 25 studies including 576 patients with IEP and 384 with BS. In IEP, early TOD was not associated with overall mortality (8% vs. 13%, OR 2.1, 95% CI 0.8-5.1), but was associated with a 23% decrease in ICU admissions (46% vs. 69%, OR 3.0, 95% CI 1.2-7.2), a 22% decrease in re-interventions (23% vs. 45%, OR 2.8, 95% CI 1.2-6.7) and a 36% decrease in length of hospital stay (14 vs. 22 days, p < 0.001), compared with late TOD. In BS, no associations between TOD and outcomes were found. When combining IEP and BS, early TOD was associated with a 6% decrease in overall mortality (10% vs. 16%, OR 2.1, 95% CI 1.1-3.9), a 19% decrease in re-interventions (26% vs. 45%, OR 1.9, 95% CI 1.1-3.2) and a 35% decrease in mean length of hospital stay (16 vs. 22 days, p = 0.001), compared with late TOD. CONCLUSIONS: This individual patient data meta-analysis confirms the general opinion that an early (≤ 24 h) compared to a late diagnosis (> 24 h) in benign esophageal perforations, particularly in IEP, is associated with improved clinical outcome.

OBJECTIVES: 5-Fluorouracil (5-FU), a chemotherapeutic drug, has severe deteriorating effects on the intestine, leading to mucositis. Glycyrrhizic acid is a compound derived from a common herbal plant Glycyrrhiza glabra, with mucoprotective, antioxidant and anti-inflammatory actions, however, associated with poor pharmacokinetics. Owing to the remarkable therapeutic action of glycyrrhizic acid-loaded polymeric nanocarriers in inflammatory bowel disease, we explored their activity against 5-FU-induced intestinal mucositis in mice. Polymeric nanocarriers have proven to be efficient drug delivery vehicles for the long-term treatment of inflammatory diseases, but have not yet been explored for 5-FU-induced mucositis. Therefore, this study aimed to produce glycyrrhizic acid-loaded polylactic-co-glycolic acid (GA-PLGA) nanoparticles to evaluate their protective and therapeutic effects in a 5-FU-induced mucositis model. METHODS: GA-PLGA nanoparticles were prepared using a modified double emulsion method, physicochemically characterized, and tested for in vitro drug release. Thereafter, mucositis was induced by 5-FU (50 mg/kg; IP) administration to the mice for the first 3 days (day 0, 1, 2), and mice were treated orally with GA-PLGA nanoparticles for 7 days (day 0-6). RESULTS: GA-PLGA nanoparticles significantly reduced mucositis severity measured by body weight, diarrhea score, distress, and anorexia. Further, 5-FU induced intestinal histopathological damage, altered villi-crypt length, reduced goblet cell count, elevated pro-inflammatory mediators, and suppressed antioxidant enzymes, all of which were reversed by GA-PLGA nanoparticles. CONCLUSION: Morphological, behavioral, histological, and biochemical results suggested that GA-PLGA nanoparticles were efficient, biocompatible, targeted, and sustained release drug delivery nano-vehicle for enhanced mucoprotective, anti-inflammatory, and antioxidant effects in 5-FU-induced intestinal mucositis.

The present study was conducted to investigate the concentrations of potential toxic metals (PTMs) in agricultural soil (n = 25) and their bioaccumulation in wheat crop (n = 25) collected from alongside the Kurram River, Pakistan. The highest concentrations of Cd, Cr, Cu, Fe, Mn, Ni, Pb, and Zn in soil samples were 0.16, 19.5, 14.7, 46.5, 13.5, 14.5, 14.0, and 19.7 mg kg-1, respectively. In the edible tissues of cultivated wheat crop, the highest concentrations of Cd, Cr, Cu, Fe, Mn, Ni, Pb, and Zn were 0.15, 10.00, 8.75, 22.25, 11.00, 11.25, 10.50, and 7.50 mg kg-1, respectively. The selected PTM concentrations in soil samples were observed within their respective permissible limits set by the Food and Agriculture Organization (FAO) and State Environmental Protection Administration (SEPA) China, while in wheat crops, the Cr and Pb concentrations were above the permissible limits of both FAO and SEPA China. The results showed that the orders of PTMs were Fe > Zn > Cr > Cu > Ni > Pb > Mn in soil and Fe > Ni > Mn > Pb > Cr > Cu > Zn in wheat. The highest PTM concentrations were reported in the sample collected near dumping sites. The results of different soil pollution indices including geo-accumulation index (Igeo), contamination factor (CF), and enrichment factor (EF) indicated that the soil of the study area was moderately to severely contaminated. The ADI values of wheat crops were less than 1, while the HQ varied among different PTMs with the highest value of 2.118 for Pb, and the lowest for Zn (0.007). The results indicated that anthropogenic intervention has made a substantial contribution to soil contamination with PTMs and subsequent uptake by wheat, which may exert potential human health risk.

Biological assays including; phytotoxicity, brine shrimp lethality, antileishmanial and insecticidal activities were carried out on crude methanolic extracts of stems and leaves of R. nervosus and their fraction namely; n-Hexane, CHCl3, EtOAc, and MeOH fractions. The highest significant phytotoxicty activity showed by chloroform, n-hexane fractions and crude extract of leaves, the growth regulation were (95%, 90% and 90%) respectively against Lemna minor, while ethyl acetate fraction and n-hexane fractions of stems displayed significant phytotoxicty (100% and 90%) respectively against Lemna minor at high dose (1000µg/ml). The results obtained from cytotoxicity assays revealed that none of the fractions are cytotoxic. The chloroform fraction of stems was showed good antileishmanial activity against L. major with LC50±S.D: 70.3±1.2 at µg/ml. The crude methanolic extracts of leaves, chloroform fraction and ethyl acetate fraction exhibited low mortality against tested insect Rhyzopertha dominica while, the rest of extracts were found almost inactive against insects species.

PURPOSE: Thromboelastography (TEG) has been recommended to characterize post-traumatic coagulopathy, yet no study has evaluated the impact of pre-injury anticoagulation (AC) on TEG variables. We hypothesized patients on pre-injury AC have a greater incidence of coagulopathy on TEG compared to those without AC. METHODS: This retrospective chart review evaluated all trauma patients admitted to an urban, level one trauma center from February 2011 to September 2014 who received a TEG within the first 24h. Patients were classified as receiving pre-injury AC or no AC if their documented medications prior to admission included warfarin, dabigatran, or anti-Xa (aXa) inhibitors (apixaban or rivaroxaban). The presence of coagulopathy on TEG or conventional assays was defined by exceeding local laboratory reference standards. RESULTS: A total of 54 patients were included (AC, n=27 [warfarin n=13, dabigatran n=6, aXa inhibitor n=8] vs. no AC, n=27). Baseline characteristics were similar between groups, including age (72±13years vs. 72±15; p=0.85), male gender (70% vs. 74%; p=0.76) and blunt mechanism of injury (100% vs. 100%; p=1). There was no difference in the number of patients determined to have coagulopathy on TEG (no AC 11% vs. AC 15%; p=0.99). Conventional tests, including the international normalized ratio (INR) and activated partial thromboplastin time (aPTT), identified coagulopathy in a high proportion of anti-coagulated patients (no AC 22% vs. AC 85%; p<0.01). CONCLUSION: TEG has limited clinical utility to evaluate the presence of pre-injury AC. Traditional markers of drug induced coagulopathy should guide reversal decisions.

Feasible regeneration protocol for economically important plant Eruca sativa was established and 1, 1-diphenyl-2-picrylhydrazyl scavenging activity of regenerated tissues was evaluated and compared with plant material collected from the wild. Leaf portions inoculated onto Murashige and Skoog (MS) medium responded to all plant growth regulators exploited. Optimum callus production was achieved on a combination of 2.0 mg l(-1) 6-benzyladenine (BA) + 1.0 mg l(-1) α-naphthalene acetic acid (NAA) and the lowest response was recorded for 0.5 mg l(-1) gibberellic acid (GA3) + 1.0 mg l(-1) NAA. The callus was subcultured on similar composition/concentrations of plant growth regulators after 4 weeks of culture time. A 5.0 mg l(-1) 6-BA + 1.0 mg l(-1) NAA produced optimum percentage shoot organogenesis after 4 weeks of subculturing. However, optimum number of shoots per explant was recorded for moderate concentrations (1.0 and 2.0 mg l(-1)) of kinetin. Incorporation of NAA into MS medium-containing GA3 also produced a feasible number of shoots/explant. Similar mean shoot length was recorded for 2.0 mg l(-1) kinetin + 1.0 mg l(-1) NAA and optimum concentrations (2.0, 5.0, and 10.0 mg l(-1)) of GA3 + 1.0 mg l(-1) NAA. In vitro generated shoots were shifted to MS medium augmented with indole acetic acid (IAA) for rooting after 4 weeks of subculturing. Moderate concentrations (5.0 mg l(-1)) of IAA produced feasible rooting. Investigation of radical scavenging activity showed that callus possesses higher levels of radical scavengers than other plant tissues tested. Phenolics and glucosides are reported to be active components of Eruca sativa phytochemistry.

Bloodstream infections (BSIs) and sepsis are major health problems, annually claiming millions of lives. Traditional blood culture techniques, employed to identify sepsis-causing pathogens and assess antibiotic susceptibility, usually take 2-4 days. Early and accurate antibiotic prescription is vital in sepsis to mitigate mortality and antibiotic resistance. This study aimed to reduce the wait time for sepsis diagnosis by employing shorter blood culture incubation times for BD BACTEC™ bottles using standard laboratory incubators, followed by real-time nanopore sequencing and data analysis. The method was tested on nine blood samples spiked with clinical isolates from the six most prevalent sepsis-causing pathogens. The results showed that pathogen identification was possible at as low as 102-104 CFU/mL, achieved after just 2 h of incubation and within 40 min of nanopore sequencing. Moreover, all the antimicrobial resistance genes were identified at 103-107 CFU/mL, achieved after incubation for 5 h and only 10 min to 3 h of sequencing. Therefore, the total turnaround time from sample collection to the information required for an informed decision on the right antibiotic treatment was between 7 and 9 h. These results hold significant promise for better clinical management of sepsis compared with current culture-based methods.

r, s, t-spherical fuzzy (r, s, t-SPF) sets provide a robust framework for managing uncertainties in decision-making, surpassing other fuzzy sets in their ability to accommodate diverse uncertainties through the incorporation of flexible parameters r, s, and t. Considering these characteristics, this article explores sine trigonometric laws to enhance the applicability and theoretical foundation for r, s, t-SPF setting. Following these laws, several aggregation operators (AOs) are designed for aggregation of the r, s, t-SPF data. Meanwhile, the desired characteristics and relationships of these operators are studied under sine trigonometric functions. Furthermore, we build a group decision-making algorithm for addressing multiple attribute group decision-making (MAGDM) problems using the developed AOs. To exemplify the applicability of the proposed algorithm, we address a practical example regarding laptop selection. Finally, parameter analysis and a comprehensive comparison with existing operators are conducted to uncover the superiority and validity of the presented AOs.

Due to the fuzziness of the medical field, q-rung orthopair fuzzy 2-tuple linguistic (q-RF2L) set is the privileged way to aid medical professionals in conveying their assessments in the patient prioritization problem. The theme of the present study is to put forward a novel approach centered around the merging of prioritized averaging (PA) and the Maclaurin symmetric mean (MSM) operator within q-RF2L context. According to the prioritization of the professionals and the correlation among the defined criteria, we apply both PA and MSM to assess priority degrees and relationships, respectively. Keeping the pluses of the PA and MSM operators in mind, we introduce two aggregation operators (AOs), namely q-RF2L prioritized Maclaurin symmetric mean and q-RF2L prioritized dual Maclaurin symmetric mean operators. Meanwhile, some essential features and remarks of the proposed AOs are discussed at length. Based on the formulated AOs, we extend the weighted aggregated sum product assessment methodology to cope with q-RF2L decision-making problems. Ultimately, to illustrate the practicality and effectiveness of the stated methodology, a real-world example of patients' prioritization problem is addressed, and an in-depth analysis with prevailing methods is performed.

Considering the advantages of q-rung orthopair fuzzy 2-tuple linguistic set (q-RFLS), which includes both linguistic and numeric data to describe evaluations, this article aims to design a new decision-making methodology by integrating Vlsekriterijumska Optimizacija I Kompromisno Resenje (VIKOR) and qualitative flexible (QUALIFLEX) methods based on the revised aggregation operators to solve multiple criteria group decision making (MCGDM). To accomplish this, we first revise the extant operational laws of q-RFLSs to make up for their shortcomings. Based on novel operational laws, we develop q-rung orthopair fuzzy 2-tuple linguistic (q-RFL) weighted averaging and geometric operators and provide the corresponding results. Next, we develop a maximization deviation model to determine the criterion weights in the decision-making procedure, which accounts for partial weight unknown information. Then, the VIKOR and QUALIFLEX methodologies are combined, which can assess the concordance index of each ranking combination using group utility and individual maximum regret value of alternative and acquire the ranking result based on each permutation's general concordance index values. Consequently, a case study is conducted to select the best bike-sharing recycling supplier utilizing the suggested VIKOR-QUALIFLEX MCGDM method, demonstrating the method's applicability and availability. Finally, through sensitivity and comparative analysis, the validity and superiority of the proposed method are demonstrated.

The idea of probabilistic q-rung orthopair linguistic neutrosophic (P-QROLN) is one of the very few reliable tools in computational intelligence. This paper explores a significant breakthrough in nanotechnology, highlighting the introduction of nanoparticles with unique properties and applications that have transformed various industries. However, the complex nature of nanomaterials makes it challenging to select the most suitable nanoparticles for specific industrial needs. In this context, this research facilitate the evaluation of different nanoparticles in industrial applications. The proposed framework harnesses the power of neutrosophic logic to handle uncertainties and imprecise information inherent in nanoparticle selection. By integrating P-QROLN with AO, a comprehensive and flexible methodology is developed for assessing and ranking nanoparticles according to their suitability for specific industrial purposes. This research contributes to the advancement of nanoparticle selection techniques, offering industries a valuable tool for enhancing their product development processes and optimizing performance while minimizing risks. The effectiveness of the proposed framework are demonstrated through a real-world case study, highlighting its potential to revolutionize nanoparticle selection in HVAC (Heating, Ventilation, and Air Conditioning) industry. Finally, this study is crucial to enhance nanoparticle selection in industries, offering a sophisticated framework probabilistic q-rung orthopair linguistic neutrosophic quantification with an aggregation operator to meet the increasing demand for precise and informed decision-making.

Methotrexate (MTX)-induced intestinal mucositis (IM) is a common side effect in cancer treatment that impairs the immune system and gut microbes, resulting in loss of mucosal integrity and gut barrier dysfunction. The quality of life and outcomes of treatment are compromised by IM. The present study was designed to investigate the mucoprotective potential of the benzimidazole derivative N-{4-[2-(4-methoxyphenyl)-1H-benzimidazole-1-sulfonyl] phenyl} acetamide (B8) on MTX-induced IM in mice. IM was induced by a single dose of MTX in mice and assessed by physical manifestations as well as biochemical, oxidative, histological, and inflammatory parameters. B8 (1, 3, 9 mg/kg) significantly reduced diarrhea score, mitigated weight loss, increased feed intake and, survival rate in a dose-dependent manner. Notably, B8 exhibited a mucoprotective effect evident through the mitigation of villus atrophy, crypt hypoplasia, diminished crypt mitotic figures, mucin depletion, and oxidative stress markers (GSH, SOD, MDA, and catalase concentration). Gene expression analysis revealed that B8 downregulated the mRNA expression of tumor necrosis factor-α (TNF-α), cyclooxygenase-2 (COX-2), interleukin-6 (IL-6), IL-1ß, and nuclear factor-κB (NF-κB) and concurrently upregulated IL-10 expression in contrast to the MTX group. Further, B8 significantly improved the luminal microflora profile by augmenting the growth of Lactobacillus spp. and reducing the number of pathogenic bacteria (E. coli). Additionally, the enzyme-linked immunoassay showed that B8 decreased the levels of pro-inflammatory cytokines. Our findings suggest that B8 had mucoprotective effects against MTX-induced IM and could be used as an adjunct in chemotherapy to deter this side effect.
