Free-flowing rivers (FFRs) support diverse, complex and dynamic ecosystems globally, providing important societal and economic services. Infrastructure development threatens the ecosystem processes, biodiversity and services that these rivers support. Here we assess the connectivity status of 12 million kilometres of rivers globally and identify those that remain free-flowing in their entire length. Only 37 per cent of rivers longer than 1,000 kilometres remain free-flowing over their entire length and 23 per cent flow uninterrupted to the ocean. Very long FFRs are largely restricted to remote regions of the Arctic and of the Amazon and Congo basins. In densely populated areas only few very long rivers remain free-flowing, such as the Irrawaddy and Salween. Dams and reservoirs and their up- and downstream propagation of fragmentation and flow regulation are the leading contributors to the loss of river connectivity. By applying a new method to quantify riverine connectivity and map FFRs, we provide a foundation for concerted global and national strategies to maintain or restore them.

Coronavirus disease 2019 (COVID-19), an infectious disease caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has been declared a global pandemic by the World Health Organization, and the situation worsens daily, associated with acute increases in case fatality rates. The main protease (Mpro) enzyme produced by SARS-CoV-2 was recently demonstrated to be responsible for not only viral reproduction but also impeding host immune responses. The element selenium (Se) plays a vital role in immune functions, both directly and indirectly. Thus, we hypothesised that Se-containing heterocyclic compounds might curb the activity of SARS-CoV-2 Mpro. We performed a molecular docking analysis and found that several of the selected selenocompounds showed potential binding affinities for SARS-CoV-2 Mpro, especially ethaselen (49), which exhibited a docking score of -6.7 kcal/mol compared with the -6.5 kcal/mol score for GC376 (positive control). Drug-likeness calculations suggested that these compounds are biologically active and possess the characteristics of ideal drug candidates. Based on the binding affinity and drug-likeness results, we selected the 16 most effective selenocompounds as potential anti-COVID-19 drug candidates. We also validated the structural integrity and stability of the drug candidate through molecular dynamics simulation. Using further in vitro and in vivo experiments, we believe that the targeted compound identified in this study (ethaselen) could pave the way for the development of prospective drugs to combat SARS-CoV-2 infections and trigger specific host immune responses.

Nucleotide Binding Site - Leucine Rich Repeat (NBS-LRR) genes play a significant role in plant defense against biotic stresses and are an integral part of signal transduction pathways. Vat gene has been well reported for their role in resistance to Aphis gossypii and viruses transmitted by them. Despite their importance, Vat like NBS-LRR resistance genes have not yet been identified and studied in cotton species. This study report hundreds of orthologous Vat like NBS-LRR genes from the genomes of 18 cotton species through homology searches and the distribution of those identified genes were tend to be clustered on different chromosome. Especially, in a majority of the cases, Vat like genes were located on chromosome number 13 and they all shared two conserved NBS-LRR domains, one disease resistant domain and several repeats of LRR on the investigated cotton Vat like proteins. Gene ontology study on Vat like NBS-LRR genes revealed the molecular functions viz., ADP and protein binding. Phylogenetic analysis also revealed that Vat like sequences of two diploid species, viz., G. arboreum and G. anomalum, were closely related to the sequences of the tetraploids than all other diploids. The Vat like genes of G. aridum and G. schwendimanii were distantly related among diploids and tetraploids species. Various hormones and defense related cis-acting regulatory elements were identified from the 2 kb upstream sequences of the Vat like genes implying their defensive response towards the biotic stresses. Interestingly, G. arboreum and G. trilobum were found to have more regulatory elements than larger genomes of tetraploid cotton species. Thus, the present study provides the evidence for the evolution of Vat like genes in defense mechanisms against aphids infestation in cotton genomes and allows further characterization of candidate genes for developing aphid and aphid transmitted viruses resistant crops through cotton breeding.

Fipronil is a broad-spectrum phenyl pyrazole insecticide that has a high degree of environmental toxicity. Commonly available chilies in the market are treated with fipronil insecticides. Demand for insecticide-free chili has thus been increasing globally. This needs various sustainable and economical methods to remove insecticides from chilies. The present study examined the effectiveness of several cleaning methods to remove pesticide residues in chili fruits. A supervised field trial was conducted in randomized block design at Rajasthan Agricultural Research Institute, Durgapura, Jaipur, India. Chili samples were subjected to seven different household methods. The samples were extracted using the quick, easy, cheap, effective, rugged, and safe (QuEChERS) method. The residues were analyzed using a gas chromatograph-electron capture detector and confirmed by GC-MS. Of the seven methods, the acetic acid treatment removes the maximum residue effect of fipronil and its metabolites (desulfinyl [MB046513]), sulfide (MB045950), and sulfone (MB046136) on chili fruits. By contrast, the tap water treatment was the least effective. The Food Safety and Standards Authority of India (FSSAI) have set the maximum residue limit value of 0.001 mg kg-1 for fipronil on green chili.

BACKGROUND: In this study, the genetic diversity of local mango (Mangifera indica L.) germplasm including 14 genotypes were evaluated by using morphological, biochemical markers and DNA barcoding technique. Morphological characterization is the first step towards utilizing these germplasm in crop improvement studies. The advanced chloroplast based DNA barcode method can be utilized to assess the genetic diversity and phylogenetic structure in such populations. METHODS: The study was carried out during 2018-2019 years to evaluate local mango germplasm including 14 diverse genotypes based on a number of morphological and biochemical traits and chloroplast DNA barcoding as well. The experiment was laid out in one way ANOVA design with fourteen germplasm indicated with indigenous collection number. RESULTS: Among local mango germplasm, IC 589756 was found to be the most promising with respect to high magnitudes of fruit length, fruit width, fruit weight, pulp weight, soluble solid content (SSC)/Acidity ratio, pH and low acidity followed by IC 589746 exhibiting the highest pulp percentage and SSC accompanied with lowest stone weight and stone percent as compared to the other genotypes. Further, the dendrogram and cluster analyses based on sequencing of chloroplast marker i.e., trnH- psbA and trnCD depicted the relationship among mango genotypes and clearly clustered them into two main clusters at a similarity coefficient 0.035 and 0.150, respectively. The first cluster includes only one genotype and cluster-II contains 13 genotypes. CONCLUSIONS: Particularly results revealed that DNA barcoding of local mango germplasm can assist not only in molecular identification but also help in elucidation of their phylogenetic relationship and thus important in maintaining biodiversity inventories.

In this experimental investigation, feasibility and performance of a polymer hybrid bio-nano composite were evaluated to remove malachite green (MG) under controlled environment conditions. The polymer hybrid bio-nanocomposite was characterized using FTIR, SEM and EDS. The influence of operating variables, namely effect of pH (2-11), nanocomposite dosage (20-100 mg), initial MG concentration (10- 200 mg/L), contact time (10-120 min) and temperature (298-318 K) were explored. The maximum removal efficiency (RE) of 99.79% was achieved at neutral pH at the dosage level of 50 mg with the initial MG concentration of 150 mg/L in 40 min. The equilibrium results revealed that the adsorption of MG data fitted to Langmuir isotherm (R2 > 0.970) indicating monolayer adsorption. The maximum adsorption capacity of polymer hybrid nanocomposite was found to be 384.615 mg/g. Kinetic studies were performed using five kinetic models and results showed the pseudo second order model fitted very well with the MG adsorption data (R2 > 0.990). The thermodynamic results confirmed that MG adsorption onto polymer hybrid nanocomposite is feasible and (ΔS ͦ = 0.2893 kJ/mol K), spontaneous (ΔH ͦ = 81.103 kJ/mol K) and exothermic (ΔG ͦ < 0). A mechanism is also proposed for the removal of MG using the polymer nanocomposite and identified that electrostatic attraction and hydrogen bonding as the major mechanism for removal of MG. FTIR results confirmed the presence of carboxyl (-COO) and hydroxyl (-OH) groups which helped in effective binding of cationic dye. The overall results revealed that polymer nanocomposite could be used as a potential adsorbent for removing MG from aqueous solution.

Background and Aims: The reliability of end tidal carbon dioxide (ETCO2) as a measure of arterial carbon dioxide (PaCO2) in pediatric laparoscopy is unclear. We evaluated the correlation of arterial to end tidal P(a-ET) CO2 during pediatric laparoscopy at two hours of pneumoperitoneum as the primary objective. We also compared P(a-ET) CO2 and alveolar to arterial oxygen gradient P(A-a) O2 and haemodynamics at fixed time points during surgery. Material and Methods: A cross-sectional study was conducted in 25 children undergoing laparoscopic abdominal surgery. Arterial blood gases were drawn at T0, baseline, T10: ten minutes, T1h: 1 hour, T2h: 2 hours of pnuemoperitoneum and T 10d: 10 mins after deflation. The P(a-ET) CO2, P(A-a) O2, were measured from the blood gas and ETCO2 and FiO2 values on the monitor. The Pearson's correlation coefficient, the Wilcoxon rank test and Chi square test were used for statistical analysis. Results: At T2h moderate correlation of P(a-ET) CO2 (r = 0.605, P = 0.001) with 40% children documenting accurate P(a-ET) CO2, -1 to +1 mm Hg was seen. Moderate correlation was also seen at T0, T10, T 10d but poor correlation at T 1h. The P(A-a) O2 increased progressively with surgery and did not correlate with P(a-ET) CO2. Heart rate was stable, but systolic blood pressures at T 10 and diastolic at T10, T 1h, T 2h were higher than baseline. Conclusion: Moderate correlation was seen between PaCO2 and ETCO2 at 2 h of pnuemoperitoneum and at T0, T 10, and T 10d. P(A-a) O2 increased with surgery but did not correlate with P(a-ET) CO2.

La2NiMnO6-a ferromagnetic (FM) insulator offers tunable charge carriers and spins useful to devise its multiple properties and applications. In this view, we studied a core-shell La2NiMnO6(2-3 nm shell on 65 - 80 nm core) of a Ni2+/Ni3+(d7) to Mn4+/Mn3+(d4) spin-up conversion- revived a new FM phase-2, raising a spin-densityσs = 0.7 s a-1over the Ni2+/Mn4+species (phase-1),σs = 0.5 s a-1, i.e. 2.12µB/f.u. larger spin moment. HRTEM images studied with x-ray diffraction characterizing core-shell structure that plays a crucial role in tuning the high spin FM phase-2 of profound properties. Below 110 K, the dc magnetization and ac magnetic susceptibilityχ(ω,T) reveal a metastable magnetic behavior on an antiferromagnetic canting of a spin-glass nature. The results follow a Vogel-Fulcher type relaxation with a relaxation timeτ0∼ 10-13s, confirming a spin-glass freezing behavior. Uniquely, FM field of phase-1 controls magnetics of phase 2 of a coupled magnet, modulating joint features with small thermal magnetic hysteresis on heating-cooling cycles.

The present work explains the sorption ability of a novel nano-composite, Polypyrrole -iron oxide-seaweed (PPy - Fe3O4 - SW), for Cr(VI) removal. The influence of operating parameters, namely pH, contact time, nanocomposite dosage, initial Chromium concentration and operating temperature, on the hexavalent chromium removal was studied. The novel nano-composite was analyzed using FTIR, SEM and EDS to confirm the sorption of Cr(VI) and to understand the mechanism of sorption. PPy - Fe3O4- SW nano-composite removed 96.36% of Cr(VI) at the optimized conditions of pH = 2, temperature = 30 °C, initial Cr(VI) concentration = 50 mg/L, nanocomposite dosage = 100 mg and contact time = 30min. PPy-Fe3O4-SW nanocomposite has a maximum sorption capacity of 144.93 mg/g. The kinetic studies revealed that the metal adsorption obeys pseudo second order (PSO) model and the sorption was found to be monolayer in nature as confirmed by Langmuir isotherm (R2 > 0.9985). Electrostatic interaction and ion-exchange are identified as the fundamental mechanisms for Cr(VI) sorption on PPy-Fe3O4-SW composite.

Sustainable valorization of industrial and solid wastes by utilizing them as feedstock to generate H2 via the photocatalytic reforming (PR) process holds great promise. It can also be an effective method to treat solid waste that otherwise would require tedious and expensive processes. This approach has the potential to offer energy solutions and form value-added chemicals. In this direction, developing photocatalysts and tuning their properties play an essential role in advancing the H2 generation efficiency. This Review article explores the application of 2D photocatalysts to generate H2 via PR of industrial waste (H2S) and solid waste, such as plastic and biomass. Despite having favorable optoelectronic properties, 2D photocatalysts are not widely employed for the PR process. The latest progress in employing 2D photocatalysts to realize efficient H2 evolution from biomass, plastic, and industrial waste such as H2S is detailed in this Review. A correlation between the properties of 2D photocatalysts with H2 evolution rate is discussed. We also emphasize understanding the mechanism involved in the PR process and the importance of 2D photocatalysts design. Such rational insight aids in further enhancing the H2 generation efficiency by effectively using solid/industrial waste as a feedstock.

We convened a multidisciplinary Working Party on behalf of the Association of Anaesthetists to update the 2011 guidance on the peri-operative management of people with hip fracture. Importantly, these guidelines describe the core aims and principles of peri-operative management, recommending greater standardisation of anaesthetic practice as a component of multidisciplinary care. Although much of the 2011 guidance remains applicable to contemporary practice, new evidence and consensus inform the additional recommendations made in this document. Specific changes to the 2011 guidance relate to analgesia, medicolegal practice, risk assessment, bone cement implantation syndrome and regional review networks. Areas of controversy remain, and we discuss these in further detail, relating to the mode of anaesthesia, surgical delay, blood management and transfusion thresholds, echocardiography, anticoagulant and antiplatelet management and postoperative discharge destination. Finally, these guidelines provide links to supplemental online material that can be used at readers' institutions, key references and UK national guidance about the peri-operative care of people with hip and periprosthetic fractures during the COVID-19 pandemic.

OBJECTIVES: For population-level screening of malnutrition among adults-especially in developing-country settings-the body mass index (BMI) can be impractical because of logistical requirements for weight and height measurement. We analyzed anthropometric data collected from a large-scale nutritional survey on women of rural Bihar to determine the mid-upper arm circumference (MUAC) cutoffs corresponding to standard BMI cutoffs and the predictive accuracies of the determined cutoffs. STUDY DESIGN: It was a cross-sectional study using multistage cluster sampling. METHODS: The current analysis used anthropometric data from a study on dietary practices of rural women (adolescents, lactating mothers, and women in the interpregnancy period). The MUAC (cm) cutoffs corresponding to four standard BMI (kg/m2) values were determined using receiver operating characteristic (ROC) curve analysis. RESULT: We detected a significant positive correlation between BMI and MUAC (r = 0.81, P < 0.0001). In ROC curve analysis, the MUAC cutoffs corresponding to BMI cutoffs of 18.5, 23, 25, and 30 kg/m2 were estimated to be 23.2, 26.0, 27.3, and 30.5 kg/m2, respectively. The predictive accuracy of the determined cutoffs was good, as indicated by the area under the ROC curve for the four different cutoffs-which ranged between 88% and 97%. Other than the cutoff for 'obese' (BMI, 30 kg/m2), the Kappa coefficients for the rest of the MUAC cutoffs showed 'substantial' agreement (>0.6) with their BMI counterparts. CONCLUSION: The results suggest that the cutoffs based on MUAC-a less resource-intensive measure than BMI-can be used for community-based screening of malnutrition among women of Bihar.

BACKGROUND AND AIM: Computational complexities encountered in craniospinal irradiation (CSI) have been widely investigated with different planning strategies. However, localization of the entire craniospinal axis (CSA) and evaluation of adaptive treatment plans have traditionally been ignored in CSI treatment. In this study, a new strategy for CSI with comprehensive CSA localization and adaptive plan evaluation has been demonstrated using cone beam CT with extended longitudinal field-of-view (CBCTeLFOV). MATERIALS AND METHODS: Multi-scan CBCT images were acquired with fixed longitudinal table translations (with 1 cm cone-beam overlap) and then fused into a single DICOM-set using the custom software coded in MatLab™. A novel approach for validation of CBCTeLFOV was demonstrated by combined geometry of Catphan-504 and Catphan-604 phantoms. To simulate actual treatment scenarios, at first, the end-to-end workflow of CSI with VMAT was investigated using an anthropomorphic phantom and then applied for two patients (based on random selection). RESULTS: The fused CBCTeLFOV images were in excellent agreement with planning CT (pCT). The custom developed software effectively manages spatial misalignments arising out of the uncertainties in treatment/setup geometry. Although the structures mapped from pCT to CBCTeLFOV showed minimal variations, a maximum spatial displacement of up to 1.2 cm (and the mean of 0.8 ± 0.3 cm) was recorded in phantom study. Adaptive plan evaluation of patient paradigms showed the likelihood of under-dosing the craniospinal target. CONCLUSION: Our protocol serves as a guide for precise localization of entire CSA and to ensure adequate dose to the large and complex targets. It can also be adapted for other complex treatment techniques such as total-marrow-irradiation and total-lymphoid-irradiation.

Aggregation of protein into bundles is responsible for many neurodegenerative diseases. In this work, we show how two-patch colloidal particles self-assemble into chains and a sudden transition to bundles takes place by tuning the patch size and solvent condition. We study the kinetics of formation of chains, bundles, and networklike structures using patchy Brownian cluster dynamics. We also analyze the ways to inhibit and accelerate the formation of these bundles. We show that in the presence of inert immobile obstacles, the kinetics of formation of bundles slows down. However, in the presence of mobile aggregating particles, which exhibit interspecies hard sphere repulsion and intraspecies attraction, the kinetics of bundle formation accelerates slightly. We also show that if we introduce mobile obstacles, which exhibit interspecies attraction and intraspecies hard sphere repulsion, the kinetics of formation of bundles is inhibited. This is similar to the inhibitory effect of peptide P4 on the formation of insulin fibers. We are providing a model of mobile obstacles undergoing directional interactions to inhibit the formation of bundles.

The present study examines the spatial, seasonal and inter annual variation of biomass burning and its impact on regional aerosol optical properties over Northern India using multi-satellite aerosol observations: Active fire points, AOD (550 nm) and AE (550-860 nm) from MODIS retrievals during January 2003-December 2017 and AAOD (388 nm), SSA (388 nm) and AI from OMI UV retrievals during January 2005-December 2017. Results from MODIS active fire count statistics indicate an increase in the number of fire occurrences (average 1477 fires per year) over India in a period of 15 years (2003-2017). The dominant fire seasons are (i) Pre-monsoon (March to May) accounting to more than 45% and (ii) Post-Monsoon having 24% of total annual fires counts. However, the crop residue burning hotspot region located in Punjab and Haryana, constitutes 26% of the total fires in India. At an average, 15456 (77.08%) fire counts were reported during the paddy season, whereas 3296 (16.44%) fire counts during wheat season respectively. The crop residue burning over the northwest IGP (Punjab) significantly affect the aerosol optical properties locally as well in the downwind regions during post-monsoon season i.e., crop residue fires increased by 4% (170 fires per year) with corresponding AOD, AAOD & AI increased by 8%, 9% & 11% respectively. The satellite observation shows large gradient of aerosol parameters from north-west to south-east along the Himalayan foot-hills which indicates the regional transport of smoke aerosols over the region. This is also supported by ground based AOD observations at four locations (Patiala, Delhi, Dehradun and Kanpur) and Black Carbon measurements at two locations (Patiala and Dehradun). The climatological averaged values of ground based AOD550 for Patiala, Delhi, Dehradun and Kanpur are 0.52 ±â€¯0.26, 0.75 ±â€¯0.40, 0.45 ±â€¯0.24 and 0.57 ±â€¯0.29 respectively whereas BC concentrations are 8.43 ±â€¯3.14 µg m-3 & 3.36 ±â€¯1.26 µg m-3 for Patiala & Dehradun respectively. Comparison of MODIS derived AOD agrees well with ground based AODs (overall R = 0.86 and RMSE = 0.14). In addition, CALIPSO shows the maximum amount of biomass burning smoke aerosols present within the atmospheric boundary layer and some cases it extending up to 2-3 km altitudes. The smoke aerosol transport pathways originated from crop residue burning were analyzed using Hysplit forward trajectories. The results reveal that majority of smoke aerosols are transported to eastern IGP, central India and adjacent oceanic regions during post-monsoon season.

Efficacious therapeutic strategies against lymphatic filariasis are always sought after. However, natural products are a promising resource for developing effective antifilarial agents. Azadirachtin, a significant tetranortriterpenoid phytocompound found in Azadirachta indica, was evaluated in vitro for antifilarial potential against the filarial parasite Setaria cervi. Dye exclusion and MTT assay confirmed the antifilarial potential of azadirachtin against S. cervi with a median lethal dose (LC50) of 6.28 µg/ml for microfilariae (mf), and 9.55 µg/ml for adult parasites. Morphological aberrations were prominent in the histological sections of the azadirachtin-exposed parasites. Moreover, alterations in the reactive oxygen species (ROS) parameters in treated parasites were evident. Induction of apoptosis in treated parasites was confirmed by DNA laddering, acridine orange (AO)/ethidium bromide (EtBr) double staining and in situ DNA fragmentation. The downregulation of anti-apoptotic CED-9 and upregulation of proapoptotic EGL-1, CED-4 and CED-3 at both the transcription and translation levels confirmed apoptosis execution at the molecular level. Changes in the gene expressions of nuc-1, cps-6 and crn-1 further clarified the molecular cause of DNA degradation. Furthermore, azadirachtin was found to be non-toxic in both in vitro and in vivo toxicity analyses. Therefore, the experimental evidence detailed the pharmacological effectiveness of azadirachtin as a possible therapeutic agent against filariasis.

Drug induced adverse drug reactions is more common in long term therapy particularly in immune compromised patients. Most of the drugs causing dermatological reactions like skin rashes, pruritus, steven johnson syndrome and exfoliative dermatitis. Nevirapine is the first line drug for human immunodeficiency virus (HIV) infection from the category of non-nucleoside reverse transcriptase enzyme inhibitor (NNRTI). In the present cases, we have reported the nevirapine induced severe exfoliative dermatitis. A patient admitted in the hospital with the symptoms of scaling on the skin and he received Nevirapine as a drug for his diagnosis of HIV for past three years. After clear examination exfoliative dermatitis was conformed and suspected with the cause due to Nevirapine. The drug was stopped and the patient was treated with drugs for symptomatic cure. The patient was recovered from his condition and other antiretroviral drugs were recommended for his treatment.

SUMOylation is a reversible post translational modification of proteins that regulates protein stabilization, nucleocytoplasmic transport, and protein-protein interactions. Several viruses and bacteria modulate host SUMOylation machinery for efficient infection. Plasmodium sporozoites are infective forms of malaria parasite that invade mammalian hepatocytes and transforms into exoerythrocytic forms (EEFs). Here, we show that during EEF development, the distribution of SUMOylated proteins in host cell nuclei was significantly reduced and expression of the SUMOylation enzymes was downregulated. Plasmodium EEFs destabilized the host cytoplasmic protein SMAD4 by inhibiting its SUMOylation. SUMO1 overexpression was detrimental to EEF growth, and insufficiency of the only conjugating enzyme Ubc9/E2 promoted EEF growth. The expression of genes involved in suppression of host cell defense pathways during infection was reversed during SUMO1 overexpression, as revealed by transcriptomic analysis. The inhibition of host cell SUMOylation was also observed during Toxoplasma infection. We provide a hitherto unknown mechanism of regulating host gene expression by Apicomplexan parasites through altering host SUMOylation.
