PURPOSE: Dietary biomarkers can potentially overcome the limitations of self-reported dietary data. While in ecology and archaeology, stable isotope ratios of carbon and nitrogen are widely used as biomarkers, this is not the case in nutrition research. Since the abundance of the 13C and the 15N isotope differ in food sources from plant and animal origin, stable isotope ratios of carbon and nitrogen (δ13C and δ15N) may differ in human biological material. Here, we investigated the stable isotope ratios of nitrogen and carbon in serum and urine from vegans and omnivores. METHOD: Measurement of δ15N and δ13C in serum and 24 h urine was performed by Elemental Analyzer-Isotope Ratio Mass Spectrometer in the cross-sectional study "Risks and Benefits of a Vegan Diet". The study included 36 vegans and 36 omnivores with a median age of 37.5 years (matched for age and sex), who adhered to their diet for at least 1 year. RESULTS: Both δ15N and δ13C were significantly lower in both the serum and 24 h urine of vegans compared to omnivores. δ15N either in serum or urine had 100% specificity and sensitivity to discriminate between vegans and omnivores. Specificity of δ13C was also > 90%, while sensitivity was 93% in serum and 77% in urine. CONCLUSION: δ15N both in serum and urine was able to accurately identify vegans and thus appears to be a promising marker for dietary habits.

PURPOSE: It has been estimated that most vegans meet the total protein requirements, but whether this is also true for individual essential amino acids (AAs) is unclear. Furthermore, a shift in protein intake is suggested to alter microbiota composition, but this association is unknown in terms of veganism or individual AAs. This cross-sectional study compared vegans and omnivores regarding dietary intake and plasma concentration of AAs. The prevalence of insufficient intake of essential AAs among vegans was determined using estimated average requirements (EAR) of WHO. Moreover, correlations between AAs intake and gut microbiota were investigated. METHODS: Data of 36 vegans and 36 omnivores (30-60 years) were analysed. AA intake, AA plasma concentrations and gut microbiota were ascertained by three-day weighed food protocols, gas/liquid chromatography-tandem mass spectrometry and 16S rRNA sequencing, respectively. RESULTS: At almost the same energy intake, the intake of 9 AAs in vegans was significantly lower than in omnivores, with median differences of - 27.0% to - 51.9%. However, only one female vegan showed total protein and lysine intake below the EAR. Vegans showed lower lysine (- 25.0%), but higher glycine (+ 25.4%) and glutamate (+ 13.1%) plasma concentrations than omnivores. Correlation patterns between AA intake and bacterial microbiota differed between vegans and omnivores. In vegans 19 species and in omnivores 5 species showed correlations with AA intake. CONCLUSION: Vegans consumed apparently sufficient but lower AAs than omnivores. In addition, the different AAs intake seems to influence the microbiota composition. The use of short-term dietary data without considering usual intake limits these findings.

PURPOSE: In several studies, exploratory dietary patterns (DP), derived by principal component analysis, were inversely or positively associated with incident type 2 diabetes (T2D). However, findings remained study-specific, inconsistent and rarely replicated. This study aimed to investigate the associations between DPs and T2D in multiple cohorts across the world. METHODS: This federated meta-analysis of individual participant data was based on 25 prospective cohort studies from 5 continents including a total of 390,664 participants with a follow-up for T2D (3.8-25.0 years). After data harmonization across cohorts we evaluated 15 previously identified T2D-related DPs for association with incident T2D estimating pooled incidence rate ratios (IRR) and confidence intervals (CI) by Piecewise Poisson regression and random-effects meta-analysis. RESULTS: 29,386 participants developed T2D during follow-up. Five DPs, characterized by higher intake of red meat, processed meat, French fries and refined grains, were associated with higher incidence of T2D. The strongest association was observed for a DP comprising these food groups besides others (IRRpooled per 1 SD = 1.104, 95% CI 1.059-1.151). Although heterogeneity was present (I2 = 85%), IRR exceeded 1 in 18 of the 20 meta-analyzed studies. Original DPs associated with lower T2D risk were not confirmed. Instead, a healthy DP (HDP1) was associated with higher T2D risk (IRRpooled per 1 SD = 1.057, 95% CI 1.027-1.088). CONCLUSION: Our findings from various cohorts revealed positive associations for several DPs, characterized by higher intake of red meat, processed meat, French fries and refined grains, adding to the evidence-base that links DPs to higher T2D risk. However, no inverse DP-T2D associations were confirmed.

BACKGROUND: The consumption of legumes is promoted as part of a healthy diet in many countries but associations of total and types of legume consumption with type 2 diabetes (T2D) are not well established. Analyses across diverse populations are lacking despite the availability of unpublished legume consumption data in prospective cohort studies. OBJECTIVE: To examine the prospective associations of total and types of legume intake with the risk of incident T2D. METHODS: Meta-analyses of associations between total legume, pulse, and soy consumption and T2D were conducted using a federated approach without physical data-pooling. Prospective cohorts were included if legume exposure and T2D outcome data were available and the cohort investigators agreed to participate. We estimated incidence rate ratios (IRRs) and CIs of associations using individual participant data including ≤42,473 incident cases among 807,785 adults without diabetes in 27 cohorts across the Americas, Eastern Mediterranean, Europe, and Western Pacific. Random-effects meta-analysis was used to combine effect estimates and estimate heterogeneity. RESULTS: Median total legume intake ranged from 0-140 g/d across cohorts. We observed a weak positive association between total legume consumption and T2D (IRR = 1.02, 95% CI: 1.01 to 1.04) per 20 g/d higher intake, with moderately high heterogeneity (I2 = 74%). Analysis by region showed no evidence of associations in the Americas, Eastern Mediterranean, and Western Pacific. The positive association in Europe (IRR = 1.05, 95% CI: 1.01 to 1.10, I2 = 82%) was mainly driven by studies from Germany, UK, and Sweden. No evidence of associations was observed for the consumption of pulses or soy. CONCLUSIONS: These findings suggest no evidence of an association of legume intakes with T2D in several world regions. The positive association observed in some European studies warrants further investigation relating to overall dietary contexts in which legumes are consumed, including accompanying foods which may be positively associated with T2D.

By combining results of adsorption/desorption measurements on powders and electrical conductivity studies on thick and thin films, the interaction of indium tin oxide with various ambient gas species and carbon monoxide as potential target gas was studied between room temperature and 700 °C. The results show that the indium tin oxide surfaces exhibit a significant coverage of water-related and carbonaceous adsorbates even at temperatures as high as 600 °C. Specifically carbonaceous species, which are also produced under carbon monoxide exposure, show a detrimental effect on oxygen adsorption and may impair the film's sensitivity to a variety of target gases if the material is used in gas sensing applications. Consequently, the operating temperature of an ITO based chemoresistive carbon monoxide sensor should be selected within a range where the decomposition and desorption of these species proceeds rapidly, while the surface oxygen coverage is still high enough to provide ample species for target gas interaction.

A zirconia-based potentiometric solid electrolyte gas sensor with internal solid state reference was used to study the response behavior of platinum cermet and indium tin oxide sensing electrodes. Target gases included both oxygen and carbon monoxide in nitrogen-based sample gas mixtures. It was found that with the indium tin oxide sensing electrode, the low-temperature behavior is mainly a result of incomplete equilibration due to contaminations of the electrode surface. On the other hand, some of these contaminant species have been identified as being pivotal for the higher carbon monoxide sensitivity of the indium tin oxide sensing electrode as compared to platinum cermet electrodes.

Five copper zinc thiolate complexes [(iPr3 PCu)2 (ZnEt2 )(edt)]2 (1-Et), [(iPr3 PCu)2 (Zn(iPr)2 )(edt)]2 (1-iPr), [(iPr3 PCu)4 (edt)2 (ZnMe2 )]2 (2), [(iPr3 PCu)3 (ZnPh2 )(ZnPh)(edt)2 ]2 (3), and [(iPr3 PCu)2 Zn2 (edt)3 ]6 (4) were prepared by the reaction of [(iPr3 PCu)2 (edt)]2 with ZnR2 (R=Me, Et, Ph, iPr) with or without addition of ethanedithiol (edt2- =ethane-1,2-dithiolate). The molecular structures of these complexes were determined by single crystal X-ray diffraction. The ethanedithiolate ligands coordinate in µ3 -η1 :η2 :η1 (2, 4), µ4 -η1 :η1 :η2 :η1 (1-R, 3), and µ5 -η1 :η1 :η2 :η1 :η1 (2) bridging modes, each sulfur atom binds to two or three metal atoms. Evidence for the presence of the weak Zn-S bonds in solution was provided by NMR spectroscopy. Mixtures of 1-Et, 1-iPr, or 3 with Sn(edt)2 were examined by thermogravimetry up to 600 °C, whereupon volatile thermolysis products were identified by mass spectrometry. In all thermolysis experiments, the formation of Cu2 ZnSnS4 as main product, besides small amounts of binary metal sulfides, was confirmed by X-ray powder diffraction (PXRD) and EDX (energy dispersive X-ray spectroscopy) analysis.

A series of five heteronuclear zinc tin chalcogenide complexes with the general formula [(tmeda)Zn(SnR2)2E3] (1-R, E = S; R = Me, Ph, tBu; 2-R, E = Se; R = Ph, tBu) have been synthesized and characterized by X-ray crystal structure analysis. In all cases, the six-membered ZnSn2E3 rings exhibit twist boat conformation. The presence of the molecular structures in solution is confirmed by 119Sn and 77Se NMR spectroscopy. Cothermolysis experiments using a mixture of complexes 1-R or 2-R and [(iPr3PCu)2(EC2H4E)]2 as a copper source were monitored by thermogravimetry and temperature dependent X-ray powder diffraction to examine the thermolysis reaction. According to Rietveld refinement, the solid residue consists of Cu2ZnSnS4 (up to 78 wt %) or Cu2ZnSnSe4 (up to 43 wt %) as the main product, respectively.

BACKGROUND: Application of metabolite profiling could expand the etiological knowledge of type 2 diabetes mellitus (T2D). However, few prospective studies apply broad untargeted metabolite profiling to reveal the comprehensive metabolic alterations preceding the onset of T2D. METHODS: We applied untargeted metabolite profiling in serum samples obtained from the European Prospective Investigation into Cancer and Nutrition (EPIC)-Potsdam cohort comprising 300 individuals who developed T2D after a median follow-up time of 6 years and 300 matched controls. For that purpose, we used ultraperformance LC-MS with a protocol specifically designed for large-scale metabolomics studies with regard to robustness and repeatability. After multivariate classification to select metabolites with the strongest contribution to disease classification, we applied multivariable-adjusted conditional logistic regression to assess the association of these metabolites with T2D. RESULTS: Among several alterations in lipid metabolism, there was an inverse association with T2D for metabolites chemically annotated as lysophosphatidylcholine(dm16:0) and phosphatidylcholine(O-20:0/O-20:0). Hexose sugars were positively associated with T2D, whereas higher concentrations of a sugar alcohol and a deoxyhexose sugar reduced the odds of diabetes by approximately 60% and 70%, respectively. Furthermore, there was suggestive evidence for a positive association of the circulating purine nucleotide isopentenyladenosine-5'-monophosphate with incident T2D. CONCLUSIONS: This study constitutes one of the largest metabolite profiling approaches of T2D biomarkers in a prospective study population. The findings might help generate new hypotheses about diabetes etiology and develop further targeted studies of a smaller number of potentially important metabolites.

Due to their amorphous structure, metallic glasses exhibit remarkable properties such as high strength, hardness, and elastic strain limit. Conversely, they also exhibit high susceptibility to brittle fracture, making them less qualified for the use as monolithic structural components. Therefore, they may be preferably used as the reinforcing phase in hybrid materials combined with ductile matrix materials. Especially metal matrix composites with interpenetrating structures are suitable. This requires an open-porous structure of the metallic glass. In the study at hand, an open-porous lattice structure was manufactured from metallic glass powder (Ni60Nb20Ta20) by laser powder bed fusion. A parameter study was carried out with various scanning strategies to manufacture a mechanically stable lattice structure while maintaining the amorphous structure of the metallic glass. Thus, X-ray diffraction measurements were conducted to validate the parameter study. A stable lattice structure with a largely amorphous structure was successfully achieved with a scanning strategy of single scanned lines and a rotation of 90° for each layer. However, nanocrystallization of 7% occurred in the heat-affected zones formed between the individual printed layers during reheating. Conducting compression tests, a compressive modulus of 18 GPa and a maximum strength of 90 MPa in 0°-direction were achieved. In 90°-direction, no compressive modulus could be determined but compressive strength resulted in 15 MPa. Performing nanoindentation with a Young's modulus of 195.1 GPa and Vickers hardness of HVIT = 956.1 was achieved for the printed bulk metallic glass alloy. The resulting lattice structure was further characterized by differential scanning calorimetry for thermal behavior.

This work presents a comprehensive investigation into the optimization of critical process parameters associated with metal fused filament fabrication (Metal-FFF) for the production of copper-based components. The study focused on three different commercial and one self-manufactured filament, each with unique chemical compositions. These filaments were systematically optimized and the density was characterized for all processing steps, as well as the electrical conductivity on the specimen scale. Remarkably, two of the studied filaments exhibited exceptional properties after sintering with forming gas (up to 94% density and 55.75 MS/m electrical conductivity), approaching the properties measured for established manufacturing methods like metal injection molding. Finally, the research was extended to component-scale applications, demonstrating the successful fabrication of inductors with integrated cooling channels. These components exhibited water tightness and were used in induction hardening experiments, validating the practical utility of the optimized Metal-FFF process. In summary, the results show great promise in advancing the utilization of Metal-FFF in industrial contexts, particularly in the production of high-performance copper components.

Besides the optimisation of process parameters such as laser power or scan speed, the choice of the scan path represents a possibility to optimise the laser powder bed fusion process even further. The usual hatching strategy creates a homogeneous microstructure but makes it necessary to switch the laser off and on after each scan vector, which can slow down the fabrication. Moreover, the end of each scan vector is a location susceptible to the creation of keyhole pores. In this work, these disadvantages were meant to be avoided by using scan strategies that consist of longer paths and thus less end of track points. To this end, an open-source tool to tailor the LPBF G-code to geometric part features and advanced path configurations was developed and embedded into a co-visualization platform. With this tool, specimens built with four different types of paths were fabricated and the effect of these alternative scan strategies on pore distributions and path neighbourhood was investigated using micro-computed tomography. In the examined example geometry, a spiral scan pattern reduced the distance the laser had to jump between scanning by 78%. However, with the alternative path patterns, the defect architecture was strongly dependant on the part geometry and increased the overall porosity to 0.42%. Respective alleviation approaches are therefore necessary and are discussed in the remainder of this work.

The exposure of operators, workers, residents and bystanders to pesticides is of high potential concern. Yet, reports on pesticide residues in the environment and near treated fields often spark debates if such findings might indicate a health risk. Although the underlying models are considered conservative, there are only limited field data on systemic exposure available. As a first step to improve the situation, we conducted a scoping review of state-of-the-art pesticide exposure biomonitoring studies in operators, workers, residents or bystanders. In contrast to existing reviews, we focused on target cultures of potential high pesticide exposure such as tree-grown produce, vine or hops. The search was conducted in Web of Science, Scopus and PubMed. Out of 17 eligible articles, a total of 11 studies met our search criteria, and 6 of them quantified the systemic exposure of humans. The analysis revealed that exposure was mainly driven by application of pesticides and reentry work, resulting in a higher exposure of operators and workers than of residents and bystanders. In nearly all cases, the systemic exposure was below the relevant toxicological reference values. The studies were subsequently analyzed to identify key criteria for a reliable design of a biomonitoring study on pesticide exposure.

SCOPE: Vegans might have a higher exposure to mycotoxins due to their heightened consumption of typical mycotoxin containing food sources. Yet, data on internal exposure among vegans in comparison to omnivores are currently lacking. METHODS AND RESULTS: This cross-sectional study includes 36 vegans and 36 omnivores (50% females, 30-60 years). A set of 28 and 27 mycotoxins is analyzed in 24-h urine and serum samples, respectively, by validated multi-mycotoxin methods (HPLC-MS/MS). Ochratoxin A (OTA), 2'R-OTA, and enniatin B in serum as well as deoxynivalenol-glucuronide in 24-h urine are quantified in 57-100% of the samples. Serum OTA levels are twofold higher in vegans than in omnivores (median 0.24 ng mL-1 vs 0.12 ng mL-1 ; p < 0.0001). No further significant differences were observed. Serum OTA levels are associated with intake of "vegan products" (r = 0.50, p < 0.0001) and "pasta & rice" (r = 0.33, p = 0.006). Sensitivity analyses advise cautious interpretation. Furthermore, serum levels of 2'R-OTA are related to coffee consumption (r = 0.64, p < 0.0001). CONCLUSION: The results indicate a higher exposure of vegans to OTA, but not to other mycotoxins. However, larger studies with repeated measurements are required to better evaluate the exposure to mycotoxins from plant-based diets.

When the amount of reactive oxygen species produced by human metabolism cannot be balanced by antioxidants, this phenomenon is commonly referred to as oxidative stress. It is hypothesised that diets with high amounts of plant food products may have a beneficial impact on oxidative stress status. However, few studies have examined whether a vegan diet is associated with lower oxidative stress compared to an omnivorous diet. The present cross-sectional study aimed to compare the levels of five oxidative stress biomarkers in vegans and omnivores. Data of 36 vegans and 36 omnivores from Germany and of 21 vegans and 18 omnivores from Finland were analysed. HPLC coupled with mass spectrometry or fluorescence detection and ELISA methods were used to measure the oxidative stress biomarkers malondialdehyde (MDA), protein carbonyls and 3-nitrotyrosine in plasma and 8-hydroxy-2'-deoxyguanosine (8-OHdG) and 8-iso-prostaglandin F2α (8-iso-PGF2α) in 24 h urine. Analyses of variance and covariance, considering potential confounders, were used. Vegans and omnivores showed no differences in MDA and protein carbonyl concentrations. In Finnish but not in German vegans, the concentrations of 3-nitrotyrosine were lower compared to those in omnivores (p = 0.047). In Germany, vegans showed lower excretion levels of 8-iso-PGF2α than omnivores (p = 0.002) and with a trend also of 8-OHdG (p = 0.05). The sensitivity analysis suggests lower 8-iso-PGF2α excretion levels in women compared to men, independently of the dietary group. The present study contributes to expanding our knowledge of the relationship between diet and oxidative stress and showed that 3-nitrotyrosine, 8-OHdG and 8-iso-PGF2α tended to be lower in vegans. Furthermore, studies are recommended to validate the present findings.

A vegan diet could impact microbiota composition and bacterial metabolites like short-chain (SCFA) and branched-chain fatty acids (BCFA). The aim of this study was to compare the concentrations of SCFA, BCFA, ammonia, and fecal pH between vegans and omnivores. In this cross-sectional study (vegans n = 36; omnivores n = 36), microbiota composition, fecal SCFA, BCFA, and ammonia concentrations and pH were analyzed in complete stool samples. A random forest regression (RFR) was used to identify bacteria predicting SCFA/BCFA concentrations in vegans and omnivores. No significant differences in SCFA and BCFA concentrations were observed between vegans and omnivores. Fecal pH (p = 0.005) and ammonia concentration (p = 0.01) were significantly lower in vegans than in omnivores, while fiber intake was higher (p < 0.0001). Shannon diversity was higher in omnivores compared to vegans on species level (p = 0.04) only. In vegans, a cluster of Faecalibacterium prausnitzii, Prevotella copri, Dialister spp., and Eubacterium spp. was predictive for SCFA and BCFA concentrations. In omnivores, Bacteroides spp., Clostridium spp., Ruminococcus spp., and Prevotella copri were predictive. Though SCFA and BCFA did not differ between vegans and omnivores, the results of the RFR suggest that bacterial functionality may be adapted to varying nutrient availability in these diets.

As part of an international research project (HiPTSLAM), the development and holistic processing of high-performance tool steels for AM is a promising topic regarding the acceptance of the laser powder bed fusion (PBF-LB) technology for functionally optimized die, forming and cutting tools. In a previous work, the newly developed maraging tool steel FeNiCoMoVTiAl was qualified to be processed by laser powder bed fusion (PBF-LB) with a material density of more than 99.9% using a suitable parameter set. To exploit further optimization potential, the influence of dual-laser processing strategies on the material structure and the resulting mechanical properties was investigated. After an initial calibration procedure, the build data were modified so that both lasers could be aligned to the same scanning track with a defined offset. A variation of the laser-based post-heating parameters enabled specific in-situ modifications of the thermal gradients compared to standard single-laser scanning strategies, leading to corresponding property changes in the produced material structure. An increase in microhardness of up to 15% was thus obtained from 411 HV up to 471 HV. The results of the investigation can be used to derive cross-material optimization potential to produce functionally graded high-performance components on PBF-LB systems with synchronized multi-laser technology.

Perfluoroalkyl substances (PFAS) are a complex group of anthropogenic compounds with exceptional properties. Due to their high persistence and mobility, they have caused ubiquitous environmental contamination and in part accumulate in the food chain. In the general population, diet is the main source of PFAS exposure, with the important sources fish and meat. As a vegan diet implies the complete exclusion of any animal products, it might be expected that vegans have lower blood levels of PFAS compared to omnivores. Furthermore, lower levels of cholesterol is one of the well-documented nutritional effects in vegans, but cholesterol levels were also found to be associated with higher PFAS levels in epidemiological studies. To examine the relations of internal PFAS levels and the levels of cholesterol in vegans and omnivores, the cross-sectional "Risks and Benefits of a Vegan Diet" (RBVD) study was used involving 36 vegans and 36 omnivores from Berlin/Germany. Nine perfluoroalkyl substances were quantified in plasma using a triple-stage quadrupole mass spectrometer. Lower median plasma concentrations were found in vegans compared to omnivores for perfluorooctane sulfonic acid (PFOS) (2.31 vs. 3.57 ng/ml, respectively; p = 0.02) and for perfluorononanoic acid (PFNA) (<0.25 vs. 0.41 ng/ml, respectively; p < 0.0001). No significant differences of the median concentrations were observed for perfluorooctanoic acid (PFOA) (1.69 vs. 1.44 ng/ml, respectively, p = 0.26) and perfluorohexane sulfonic acid (PFHxS) (1.96 vs. 1.79 ng/ml, respectively; p = 0.70). The strongest correlations with food groups, derived from a food frequency questionnaire, were observed between levels of PFOA and water consumption (in case of the total study population, n = 72), and between levels of PFOS as well as PFNA and the consumption of 'meat and meat products' (in case of the omnivores, n = 36). Levels of Low Density Lipoprotein (LDL) cholesterol were confirmed to be considerably lower in vegans compared to omnivores (86.5 vs. 115.5 mg/dl, respectively; p = 0.001), but no associations between the four main PFAS and LDL cholesterol were observed (all p > 0.05) at the low exposure level of this study. According to the results of our study, a vegan diet may be related to lower PFAS levels in plasma. We highlight the importance of the adjustment of dietary factors like a vegan diet in case of epidemiological studies dealing with the impact of PFAS on the levels of blood lipids.

Martensitic steels are tempered to increase the toughness of the metastable martensite, which is brittle in the as-quenched state, and to achieve a more stable microstructure. During the tempering of steels, several particular overlapping effects can arise. Classical dilatometric investigations can only detect effects by monitoring the integral length change of the sample. Additional in-situ diffractometry allowed a differentiation of the individual effects such as transformation of retained austenite and formation of cementite during tempering. Additionally, the lattice parameters of martensite and therefrom the tetragonality was analyzed. Two low-alloy steels with carbon contents of 0.4 and 1.0 wt.% and a high-alloy 5Cr-1Mo-steel with 0.4 wt.% carbon were investigated by dilatometry and in-situ diffractometry. In this paper, microstructural effects during tempering of the investigated steels are discussed by a comparative study of dilatometric and diffractometric experiments. The influence of the chemical composition on the tempering behavior is illustrated by comparing the determined effects of the three steels. The kinetics of tempering is similar for the low-alloy steels and shifted to much higher temperatures for the high-alloy steel. During tempering, the tetragonality of martensite in the steel with 1.0 wt% carbon shifts towards a low carbon behavior, as in the steels with 0.4 wt.% carbon.

OBJECTIVE: Plasma protein N-glycan profiling integrates information on enzymatic protein glycosylation, which is a highly controlled ubiquitous posttranslational modification. Here we investigate the ability of the plasma N-glycome to predict incidence of type 2 diabetes and cardiovascular diseases (CVDs; i.e., myocardial infarction and stroke). RESEARCH DESIGN AND METHODS: Based on the prospective European Prospective Investigation of Cancer (EPIC)-Potsdam cohort (n = 27,548), we constructed case-cohorts including a random subsample of 2,500 participants and all physician-verified incident cases of type 2 diabetes (n = 820; median follow-up time 6.5 years) and CVD (n = 508; median follow-up time 8.2 years). Information on the relative abundance of 39 N-glycan groups in baseline plasma samples was generated by chromatographic profiling. We selected predictive N-glycans for type 2 diabetes and CVD separately, based on cross-validated machine learning, nonlinear model building, and construction of weighted prediction scores. This workflow for CVD was applied separately in men and women. RESULTS: The N-glycan-based type 2 diabetes score was strongly predictive for diabetes risk in an internal validation cohort (weighted C-index 0.83, 95% CI 0.78-0.88), and this finding was externally validated in the Finland Cardiovascular Risk Study (FINRISK) cohort. N-glycans were moderately predictive for CVD incidence (weighted C-indices 0.66, 95% CI 0.60-0.72, for men; 0.64, 95% CI 0.55-0.73, for women). Information on the selected N-glycans improved the accuracy of established and clinically applied risk prediction scores for type 2 diabetes and CVD. CONCLUSIONS: Selected N-glycans improve type 2 diabetes and CVD prediction beyond established risk markers. Plasma protein N-glycan profiling may thus be useful for risk stratification in the context of precisely targeted primary prevention of cardiometabolic diseases.
