RESUMO

Crohn's disease (CD) is a chronic inflammatory illness of the digestive system with unknown etiology, and its incidence is increasing worldwide. However, there are currently no effective treatments or medications available for individuals with CD. Therefore, novel therapeutic strategies are urgently needed. The bioactive compounds and targets associated with compounds of Qinghua Xiaoyong Formula (QHXYF) were examined using The Traditional Chinese Medicine Systems Pharmacology database, and 5 disease target databases were also used to identify CD-related disease targets. A total of 166 overlapping targets were identified from QHXYF-related and CD-related disease targets and they were found to be enriched in oxidative stress-related pathways and the PI3K/AKT signaling pathway. Molecular docking was then used to predict how the bioactive compounds would bind to the hub targets. It was found that quercetin could be the core bioactive compound and had good binding affinity to the top 5 hub targets. Finally, animal experiments were performed to further validate the findings, and the results revealed that QHXYF or quercetin inhibited 2,4,6-trinitrobenzenesulfonic acid-induced inflammation and oxidative stress processes by inhibiting the PI3K/AKT pathway, thereby improving CD symptoms. These findings suggest that QHXYF and quercetin may be potential novel treatments for CD.

In this study, cotton fabric-reinforced phenolic resin (CPF) composites were modified by adding four two-dimensional fillers: graphitic carbon nitride (g-C3N4), graphite (Gr), molybdenum disulfide (MoS2), and hexagonal boron nitride (h-BN). The tribological properties of these modified materials were investigated under dry friction and water lubrication conditions. The CPF/Gr composite exhibits significantly better tribological performance than the other three filler-modified CPF composites under dry friction, with a 24% reduction in friction coefficient and a 78% reduction in wear rate compared to the unmodified CPF composite. Under water lubrication conditions, all four fillers did not significantly alter the friction coefficient of the CPF composites. However, except for an excessive amount of Gr, the other three fillers can reduce the wear rate. Particularly in the case of 10% MoS2 content, the wear rate decreased by 56%. Scanning electron microscopy (SEM) and X-ray photoelectron spectroscopy (XPS) were employed for the analysis of the morphology and composition of the transfer films. Additionally, molecular dynamics (MD) simulations were conducted to investigate the adsorption effects of CPF/Gr and CPF/MoS2 composites on the counterpart surface under both dry friction and water lubrication conditions. The difference in the adsorption capacity of CPF/Gr and CPF/MoS2 composites on the counterpart, as well as the resulting formation of transfer films, accounts for the variation in tribological behavior between CPF/Gr and CPF/MoS2 composites. By combining the lubrication properties of MoS2 and Gr under dry friction and water lubrication conditions and using them as co-fillers, we can achieve a synergistic lubrication effect.

In this study, microporous calcium silicate was synthesized from a silicon solution of fly ash extracted by soaking in strong alkali as a silicon source. By means of XRD, TEM, FTIR, and thermodynamic calculations, the crystal evolution and growth process of microporous calcium silicate were studied under the synthesis temperature of 295~365 K. The results show that calcium silicate is a single-chain structure of the Si-O tetrahedron: Q1 type Si-O tetrahedron is located at both ends of the chain, and the middle is the [SiO44-] tetrahedron connected by [O2-] coplanar, and Ca2+ is embedded in the interlayer structure of calcium silicate. The formation rate and crystallization degree of calcium silicate hydrate were positively correlated with temperature. When the synthesis temperature was 295 K, its particle size was about 8 µm, and when the synthesis temperature was 330 K, a large number of amorphous microporous calcium silicate with a particle size of about 14 µm will be generated. When the temperature was above 350 K, the average particle size was about 17 µm. The microporous calcium silicate showed obvious crystalline characteristics, which indicate that the crystallization degree and particle size of microporous calcium silicate could be controlled by a reasonable synthesis temperature adjustment.

Calcium silicate slag (CSS) is used as a secondary solid waste produced by aluminum extraction technology from high alumina fly ash, and its resource utilization has always been a key issue to be solved. In this study, CSS was used to replace a portion of fly ash (FA) to prepare a new inorganic binder stabilized material for road base. The unconfined compressive strength (UCS), phase composition, microstructure, durability and performance index of the base of the test section of the CSS pavement base material were studied. The results showed that with the increase in CSS content, the UCS of pavement base materials gradually increased. Under standard curing conditions, the UCS increased 6.90~17.24% after 7 days, and 7.90~28.95% after 28 days. The main reason was that as the hydration time increased from 7 d to 28 d, the hydration products C-A-S-H gel and C-S-H gel increased, the [SiO4] polymerization degree increased, the crystal type changed, and the structure denser, which supported the good development of mechanical strength of CSS pavement base material. In addition, the research has been successfully applied to a pilot test in Hohhot, China. The freeze-thaw resistance, water stability and UCS of the CSS pavement base material were tested to meet the requirements of Chinese road construction standards, indicating that the application of CSS in pavement base is feasible.

INTRODUCTION: A high prevalence of cryptoglandular and Crohn's perianal fistulas has been reported worldwide, and several surgical options are available for the management of anal fistula, with varying clinical efficacy. However, currently, the available evidence for the effectiveness of these surgical approaches are lacking and of concern in terms of the credibility and strength. The purpose of this study is to evaluate the credibility of the published systematic reviews and meta-analyses that assess the efficacy and safety of the surgical options for cryptoglandular and Crohn's perianal fistulas through an umbrella review. METHODS AND ANALYSIS: A systematic search in PubMed, Embase and Cochrane library will be performed from inception to December 2020 without any language restriction. We will include systematic reviews and meta-analyses that investigate the efficacy and safety of surgical approaches in the management of cryptoglandular and Crohn's perianal fistulas. Two reviewers will independently screen search results through reading the titles or abstracts. Relevant information will be extracted from each eligible systematic review or meta-analysis. Based on random effects model summary estimates along with their p values, 95% prediction intervals, between-study heterogeneity, small-study effects and excess significance, we will classify the evidence from convincing (class I) to weak (class IV). Findings will be summarized using quantitative synthesis combined with a narrative approach. Cryptoglandular and Crohn's perianal fistulas will be summarized separately. Two authors will independently perform the literature search, data extraction, and quality assessment of each included systematic review and meta-analysis. Any unresolved conflicts or doubts will be resolved by discussion or by consulting a senior author. The risk of bias of the systematic reviews will be assessed using a 16-item Assessment of Multiple Systematic Reviews 2 (AMSTAR2) checklist. The strength of evidence for the included systematic reviews will be classified as "high", "moderate", "low", or "critically low" quality. ETHICS AND DISSEMINATION: Ethics approval is not required as we will collect data from the published systematic reviews and meta-analyses without using individual patient data. The results of this umbrella review will be published in a peer-reviewed journal and will be presented at an anorectal disease conference. PROSPERO REGISTRATION NUMBER: CRD42020200754.
