RESUMO

Homo sapiens was present in northern Asia by around 40,000 years ago, having replaced archaic populations across Eurasia after episodes of earlier population expansions and interbreeding1-4. Cultural adaptations of the last Neanderthals, the Denisovans and the incoming populations of H. sapiens into Asia remain unknown1,5-7. Here we describe Xiamabei, a well-preserved, approximately 40,000-year-old archaeological site in northern China, which includes the earliest known ochre-processing feature in east Asia, a distinctive miniaturized lithic assemblage with bladelet-like tools bearing traces of hafting, and a bone tool. The cultural assembly of traits at Xiamabei is unique for Eastern Asia and does not correspond with those found at other archaeological site assemblages inhabited by archaic populations or those generally associated with the expansion of H. sapiens, such as the Initial Upper Palaeolithic8-10. The record of northern Asia supports a process of technological innovations and cultural diversification emerging in a period of hominin hybridization and admixture2,3,6,11.

GPR101 is an orphan G protein-coupled receptor actively participating in energy homeostasis. Here we report the cryo-electron microscopy structure of GPR101 constitutively coupled to Gs heterotrimer, which reveals unique features of GPR101, including the interaction of extracellular loop 2 within the 7TM bundle, a hydrophobic chain packing-mediated activation mechanism and the structural basis of disease-related mutants. Importantly, a side pocket is identified in GPR101 that facilitates in silico screening to identify four small-molecule agonists, including AA-14. The structure of AA-14-GPR101-Gs provides direct evidence of the AA-14 binding at the side pocket. Functionally, AA-14 partially restores the functions of GH/IGF-1 axis and exhibits several rejuvenating effects in wild-type mice, which are abrogated in Gpr101-deficient mice. In summary, we provide a structural basis for the constitutive activity of GPR101. The structure-facilitated identification of GPR101 agonists and functional analysis suggest that targeting this orphan receptor has rejuvenating potential.

Despite numerous studies demonstrate that genetics and epigenetics factors play important roles on smoking behavior, our understanding of their functional relevance and coordinated regulation remains largely unknown. Here we present a multiomics study on smoking behavior for Chinese smoker population with the goal of not only identifying smoking-associated functional variants but also deciphering the pathogenesis and mechanism underlying smoking behavior in this under-studied ethnic population. After whole-genome sequencing analysis of 1329 Chinese Han male samples in discovery phase and OpenArray analysis of 3744 samples in replication phase, we discovered that three novel variants located near FOXP1 (rs7635815), and between DGCR6 and PRODH (rs796774020), and in ARVCF (rs148582811) were significantly associated with smoking behavior. Subsequently cis-mQTL and cis-eQTL analysis indicated that these variants correlated significantly with the differential methylation regions (DMRs) or differential expressed genes (DEGs) located in the regions where these variants present. Finally, our in silico multiomics analysis revealed several hub genes, like DRD2, PTPRD, FOXP1, COMT, CTNNAP2, to be synergistic regulated each other in the etiology of smoking.

PURPOSE OF REVIEW: In this review, we summarized published articles on the role of tripartite motif (TRIM) family members in the initiation and development of human malignancies. RECENT FINDINGS: The ubiquitin-proteasome system (UP-S) plays a critical role in cellular activities, and UP-S dysregulation contributes to tumorigenesis. One of the key regulators of the UP-S is the tripartite motif TRIM protein family, most of which are active E3 ubiquitin ligases. TRIM proteins are critical for the biological functions of cancer cells, including migration, invasion, metastasis, and therapy resistance. Therefore, it is important to understand how TRIM proteins function at the molecular level in cancer cells. SUMMARY: We provide a comprehensive and up-to-date overview about the role TRIMs play in cancer progression and therapy resistance. We propose TRIM family members as potential new markers and targets to overcome therapy failure.

Ulcerative colitis (UC) is a chronic recurrent inflammatory disease affecting the rectum and colon. Numerous epidemiological studies have identified smoking as a protective factor for UC. Dysbiosis of intestinal microbiota and release of inflammatory factors are well-established characteristics associated with UC. Therefore, we have observed that nicotine exhibits the potential to ameliorate colitis symptoms in UC mice. Additionally, it exerts a regulatory effect on colonic microbiota dysbiosis by promoting the growth of beneficial bacteria while suppressing harmful bacteria. Combined in vivo and in vitro investigations demonstrate that nicotine primarily impedes the assembly of NLRP3, subsequently inhibiting downstream IL-1ß secretion.

Efficient separation and purification of hemoglobin from blood and other complicated biological fluids still remains a big challenge. Molecularly imprinted polymers (MIPs) of hemoglobin are potential choices; however, they suffer from severe problems including difficult template removal and low imprinting efficiency like other protein-imprinted polymers. Herein, a novel MIP of bovine hemoglobin (BHb) was designed in which a peptide crosslinker (PC), instead of the commonly used crosslinkers, was used. The PC, a random copolymer of lysine and alanine, adopts an α-helical conformation at pH 10 but transits to a random coil conformation at pH 5. The introduction of alanine residues lowers the pH range at which the PC undergoes helix-coil transition. The imprint cavities in the polymers are shape-memorable due to the reversible and precise helix-coil transition of the peptide segments in the polymers. They can be enlarged by lowering pH from 10 to 5, thus allowing complete removal of the template protein under mild conditions. When the pH is adjusted back to 10, their original size and shape will be recovered. Therefore, the MIP binds the template protein BHb with high affinity. Compared with the MIP crosslinked with the commonly used crosslinker, the imprinting efficiency of the PC-crosslinked MIP is significantly improved. In addition, both the maximum adsorption capacity (641.9 mg/g) and imprinting factor (7.2) are much higher than the BHb MIPs reported previously. The new BHb MIP also exhibits high selectivity toward BHb and good reusability. Thanks to the high adsorption capacity and high selectivity of the MIP, when it was applied to extract BHb from bovine blood, BHb in the blood sample was extracted almost completely, and high purity product was obtained.

Neural progenitor cells (NPCs) are essential for in vitro drug screening and cell-based therapies for brain-related disorders, necessitating well-defined and reproducible culture systems. Current strategies employing protein growth factors pose challenges in terms of both reproducibility and cost. In this study, we developed a novel DNA-based modulator to regulate FGFR signaling in NPCs, thereby facilitating the long-term maintenance of stemness and promoting neurogenesis. This DNA-based FGFR-agonist effectively stimulated FGFR1 phosphorylation and activated the downstream ERK signaling pathway in human embryonic stem cell (HESC)-derived NPCs. We replaced the basic fibroblast growth factor (bFGF) in the culture medium with our DNA-based FGFR-agonist to artificially modulate FGFR signaling in NPCs. Utilizing a combination of cell experiments and bioinformatics analyses, we showed that our FGFR-agonist could enhance NPC proliferation, direct migration, and promote neurosphere formation, thus mimicking the functions of bFGF. Notably, transcriptomic analysis indicated that the FGFR-agonist could specifically influence the transcriptional program associated with stemness while maintaining the neuronal differentiation program, closely resembling the effects of bFGF. Furthermore, our culture conditions allowed for the successful propagation of NPCs through over 50 passages while retaining their ability to efficiently differentiate into neurons. Collectively, our approach offers a highly effective method for expanding NPCs, thereby providing new avenues for disease-in-dish research and drug screening aimed at combating neural degeneration.

BACKGROUND: Studies on the effect of sleep duration on cardiovascular health have contradictory findings. Underlying health issues may have led to inconsistent results and warrant consideration. We aim to assess the relationship of night sleep duration with incident cardiovascular disease (CVD) in a general population, taking into consideration underlying chronic diseases. METHODS: Data from Shanghai Suburban Adult Cohort and Biobank with a median follow-up of 5.1 years was used, including 33,883 adults aged 20-74 years old. Incident CVD cases were reported and recorded by the Center for Disease Prevention and Control in Songjiang, Shanghai. We used Cox proportional hazard regression models and restricted cubic spline (RCS) analysis to explore the relationship between different sleep groups and sleep duration with incident CVD outcomes, through stratification by gender and age, as well as different health conditions, with adjustments for potential confounders. RESULTS: Long sleep duration (> 9 h) compared to > 7 to ≤ 8 h was associated with overall incident CVD in participants aged ≥ 50 years old: HR(95%CI) = 2.07 (1.15, 3.74) for 50-59y and 1.43 (1.04, 1.93) for 60-74y. RCS analysis showed a J-shaped relationship between sleep and CVD risk in those ≥ 50y, which was confirmed only in those with a chronic health condition. Non-linear relationships between sleep and CVD risk factors, such as BMI, blood glucose and glycated haemoglobin, were observed. CONCLUSIONS: Long sleep duration is associated with increased risk of CVD in people ≥ 50y. However, CVD risk factors and underlying health conditions such as hypertension, and diabetes, may play a driving role in the relationship.

This study explores the use of Multi-Objective Genetic Algorithm (MOGA) for thermodynamic characteristics of serrated plate-fin heat exchanger (PFHE) under numerical simulation method. Numerical investigations on the important structural parameters of the serrated fin and the j factor and the f factor of PFHE are conducted, and the experimental correlations about the j factor and the f factor are determined by comparing the simulation results with the experimental data. Meanwhile, based on the principle of minimum entropy generation, the thermodynamic analysis of the heat exchanger is investigated, and the optimization calculation is carried out by MOGA. The comparison results between optimized structure and original show that the j factor increases by 3.7%, the f factor decreases by 7.8%, and the entropy generation number decreases by 31%. From the data point of view, the optimized structure has the most obvious effect on the entropy generation number, which shows that the entropy generation number can be more sensitive to the irreversible changes caused by the structural parameters, and at the same time, the j factor is appropriately increased.

Due to aerodynamic resistance, aerodynamic noise, and other problems, the further development of traditional high-speed electric multiple units (EMUs) on the open line has been seriously restricted, and the construction of a vacuum pipeline high-speed train system has become a new solution. In this paper, the Improved Detached Eddy Simulation (IDDES) is used to analyze the turbulent characteristics of the near wake region of EMU in vacuum pipes, so as to establish the important relationship between the turbulent boundary layer, wake, and aerodynamic drag energy consumption. The results show that there is a strong vortex in the wake near the tail, which is concentrated at the lower end of the nose near the ground and falls off from the tail. In the process of downstream propagation, it shows symmetrical distribution and develops laterally on both sides. The vortex structure far from the tail car is increasing gradually, but the strength of the vortex is decreasing gradually from the speed characterization. This study can provide guidance for the aerodynamic shape optimization design of the rear of the vacuum EMU train in the future and provide certain reference significance for improving the comfort of passengers and saving the energy consumption caused by the speed increase and length of the train.

The concept of ethnic medicine is divided into a broad sense and a narrow sense. The broad concept refers to the traditional medicine of the Chinese nation, and the narrow concept refers to the traditional medicine of Chinese ethnic minorities. The external medicine is one of the main forms of ethnic medicine, and it is also the important content of ethnic medicine for external use, which is widely used in clinical practice. As the theory of ethnic medicine is unique, the application methods have certain characteristics, which are the key technical parts of clinical practice. However, the existing traditional Chinese medicine consensus formulation me-thods cannot meet the needs of the consensus formulation of the external ethnic medicine. Therefore, the methods suitable for expert consensus on external ethnic medicine are required. This article took Expert opinion on clinical application of Baimai Ointment as an exa-mple, and explorde a reasonable, effective, multi-dimensional, and multi-stage method to formulate expert consensus on the external ethnic medicine. In this research, three-dimensional sources of information, including ancient classics, clinical research evidence, and expert application experiences, were systematically and scientifically collected. After organization and analysis, the information was formed into comprehensive evidence. In a formal consensus meeting, part of the recommendations reached consensus. As to the issues that did not reach agreement, in-depth interviews were used to explore the reasons for the differences and resolve the disagreements. Finally, unanimous recommendations were reached. There are common problems during the formulation process of Expert opinion on clinical application of Baimai Ointment. This study is expected to provide references for the formulation of expert consensus on other external ethnic medicine.

Lung adenocarcinoma (ADC) is the predominant histological type of lung cancer, and radiotherapy is one of the current therapeutic strategies for lung cancer treatment. Unfortunately, biological complexity and cancer heterogeneity contribute to radioresistance development. Karyopherin α2 (KPNA2) is a member of the importin α family that mediates the nucleocytoplasmic transport of cargo proteins. KPNA2 overexpression is observed across cancer tissues of diverse origins. However, the role of KPNA2 in lung cancer radioresistance is unclear. Herein, we demonstrated that high expression of KPNA2 is positively correlated with radioresistance and cancer stem cell (CSC) properties in lung ADC cells. Radioresistant cells exhibited nuclear accumulation of KPNA2 and its cargos (OCT4 and c-MYC). Additionally, KPNA2 knockdown regulated CSC-related gene expression in radioresistant cells. Next-generation sequencing and bioinformatic analysis revealed that STAT1 activation and nuclear phospholipid scramblase 1 (PLSCR1) are involved in KPNA2-mediated radioresistance. Endogenous PLSCR1 interacting with KPNA2 and PLSCR1 knockdown suppressed the radioresistance induced by KPNA2 expression. Both STAT1 and PLSCR1 were found to be positively correlated with dysregulated KPNA2 in radioresistant cells and ADC tissues. We further demonstrated a potential positive feedback loop between PLSCR1 and STAT1 in radioresistant cells, and this PLSCR1-STAT1 loop modulates CSC characteristics. In addition, AKT1 knockdown attenuated the nuclear accumulation of KPNA2 in radioresistant lung cancer cells. Our results collectively support a mechanistic understanding of a novel role for KPNA2 in promoting radioresistance in lung ADC cells.

Compared with the colloidal crystals (CCs) of hard spheres, large-scale, high-quality CCs of soft microgel spheres are easier to be assembled because they are more tolerant to defects. However, to assemble microgel CCs, a microgel dispersion should first be concentrated and then allowed to crystallize, which is tedious and time-consuming. Herein, we demonstrated that a magnetic poly(N-isopropylacrylamide) (PNIPAM) microgel with an Fe3O4 core and a PNIPAM shell can be assembled into CCs quickly by simply applying an external magnetic field to the diluted microgel dispersions. The resulting CCs are highly ordered as revealed by their iridescent color, laser diffraction pattern, and confocal characterization. They display a sharp Bragg peak on their reflection spectra, which shifts to lower wavelength when heated because of the thermosensitivity of the PNIPAM shell. The magnetic assembly is not only simple and fast but also allows control of the CC structure in both horizontal and vertical directions. Using spatially varying magnetic fields, patterned microgel CCs were facilely assembled. More importantly, magnetic microgel spheres with different sizes can be assembled in a layer-by-layer manner by adding them sequentially, and the thickness of each layer can be simply controlled by the amount of spheres added.

Spontaneous oxidative polymerization of dopamine (DA) is widely exploited as a facile and versatile method for surface modification. However, the reaction is very slow and only occurs in alkaline solutions, which severely limit its applications. Herein it is reported that the reaction can be dramatically accelerated by using Fe2+ as catalyst. While it takes hours and days using conventional method, the Fe2+ -catalyzed reaction finishes almost immediately at pH 7.0. In addition, under the catalysis of Fe2+ , the reaction can occur at a pH down to 4.0. The fast Fe2+ -catalyzed polymerization of DA leads to fast deposition of polydopamine (PDA) coating, thus allowing fast surface modification and textile dyeing. The Fe2+ -catalyzed reaction also allows spatial control over the PDA deposition. The fast, simple, and mild surface modification method developed here will find applications in numerous fields.

A new species of Dalyelliidae, Gieysztoria pellucida Wang and You, is described based on material collected in southern China through an integrative approach combining morphological, histological, and molecular (18S and 28S rDNA) data. Gieysztoria pellucida sp. nov. is morphologically characterized by a fan-shaped (about 270° when pressed) stylet, consisting of 13 similar distal spines and a broad girdle without fenestrae region. This stylet is distinct from that of any other similar species in the Aequales group to which this species belongs. In addition, specimens identifiable as Gieysztoria garudae Van Steenkiste, Van Mulken, and Artois, 2012 were discovered from the same location as G. pellucida sp. nov. Gieysztoria garudae has previously been known only from India; the present study thus represents the first record of the species from China.

Chemical investigation of the Hainan soft coral Sinularia hirta resulted in the isolation and identification of a library of sixteen structurally diverse terpenoids, including a dinorditerpenoid with an uncommon 17,19-dinorxeniaphyllane skeleton, namely sinuhirtone A (7), six new xeniaphyllane-type diterpenoids (1-6), one new norxeniaphyllanoid (8), two new norcaryophyllene-type sesquiterpenoids (9 and 10), together with six known related compounds (11-16). Compounds 1-3 are three new furanone-containing xeniaphyllane-type diterpenoids. The structures of the new compounds, including their absolute configurations, were determined by extensive spectroscopic analysis and a series of quantum chemical calculations, including quantum mechanical-nuclear magnetic resonance (QM-NMR), time-dependent density functional theory-electronic circular dichroism (TDDFT-ECD), and optical rotatory dispersion (ORD) methods. A plausible biosynthetic connection between new compounds 1-9 was also proposed. New compounds 2-4, 7, and 8 were evaluated for in vitro cytotoxicity against four cancer cell lines.

A novel piezoelectric fiber sensor based on polyvinylidene fluoride piezoelectric (PVDF) doped with graphene is presented. The near-field electrospinning technology was used for developing the sensor. The uniform experimental design method was introduced to determine the ranges of experimental parameters, including the applied voltage, the drum speed range, the graphene doping ratios from 0% to 11 wt% in PVDF solution, and the electrode gap. By experimental results, the conductivities of PVDF solutions with different doping ratios of graphene increased from 19.6 µS/cm to 115.8 µS/cm. Tapping tests were performed to measure the voltages and currents produced by the piezoelectric fibers. The maximum output voltage was 4.56 V at 5 wt% graphene doping ratio in PVDF fibers, which was 11.54 times that of the pure PVDF sensors. Moreover, mechanical properties of the proposed sensor were measured. Motion intention and swallowing test, such as saliva-swallowing and eating, were carried out. When the subject spoke normally, the output voltage of the sensor was between 0.2 and 0.4 V, approximately. Furthermore, when the subject drank water and ate food, the output voltage of the sensor was between 0.5 and 1 V, approximately. The proposed sensor could be used to detect signals of the human body and serve as a wearable device, allowing for more diagnosis and medical treatment.

The focus of this work was on developing a green, low-cost, and efficient biosorbent based on the biological structure and properties of MT and applying it to the remediation of cationic dyes in dye wastewater. The adsorption performance and mechanism of MT on methylene blue (MB) and crystal violet (CV) were investigated by batch adsorption experiments. The results demonstrated that the highest adsorption values of MT for MB (411 mg/g) and CV (553 mg/g) were greatly higher than the reported values of other biosorbents. In addition, the adsorption behaviors of methylene blue (MB) and crystal violet (CV) by MT were spontaneous exothermic reactions and closely followed the pseudo-second-order (PSO) kinetics and Langmuir isotherm. Further, the depleted MT was regenerated using pyrolysis mode to convert depleted MT into MT-biochar (MBC). The maximum adsorption of Cu2+ and Pb2+ by MBC was up to 320 mg/g and 840 mg/g, respectively. In conclusion, this work presented a new option for the adsorption of cationic dyes in wastewater and a new perspective for the treatment of depleted biosorbents.

Beclin1 is a key regulator of a family of autophagy-related proteins. The aim of our study was to elucidate the clinicopathological and prognostic significance of Beclin1 expression which is a positive regulator of autophagy in cervical cancer. The results showed that a total of 2682 patients were enrolled in 21 case-control studies. The results showed that, as for Beclin1 expression, significant differences were found in cervical cancer vs. normal cervical tissues (p<.00001) and cancer tissues with vs. no lymph node metastasis (p<.00001); tumour diameter no less than vs. less than 4 cm (p=.001), myometrial invasion depth no less than vs. less than 1/2 and FIGO I vs. II (p=.02); relationship between Beclin1 expression and prognosis of cervical cancer (p=.03). Kaplan-Meier's plotter showed that Beclin1 expression was negative. It was associated with overall, post-progressive and distant metastatic survival. According to the Oncomine database, Beclin1 mRNA expression in cervical cancer tissues was higher than that in normal tissues. Cox multivariate showed that lymph node metastasis and TNM stage were important factors affecting the survival time of patients. Beclin1 expression can be used as an indicator of prognosis in patients, and provide methods and ideas for prevention and treatment.

This study aimed to evaluate the characteristics of Calu-3 cells as a model to examine the toxicological responses of inhalable substances. Calu-3 cells were grown to the confluence at an air-liquid interface (ALI) using a Transwell® permeable support system. The ALI resulted in biomimetic native bronchial epithelium displaying pseudostratified columnar epithelium with more microvilli and secretory vesicles. We further characterized and optimized the Calu-3 cell line model using ALI culturing conditions, immunolabeling of protein expression, ultrastructural analysis using scanning electron microscopy (SEM), and transepithelial electrical resistance (TEER) measurements, and then screened for the cytotoxicity of tobacco flavoring extracts. Calu-3 cells displayed dose-dependent responses when treated with the flavoring extract. Within 8-10 days, cell monolayers developed TEER ≥1000 Ω·cm2. During this time, Calu-3 cells exposed to flavoring extracts X01 and X06 exhibited a loss of cellular integrity and decreased ZO-1 and E-cadherin protein expression. In conclusion, we investigated the Calu-3 cell line culture conditions, culture time, and barrier integrity and tested the effect of six new synthetic tobacco flavoring extracts. Our data demonstrate that the Calu-3 human bronchial epithelial cell monolayer system is a potential in vitro model to assess the inhalation toxicity of inhalable substances.
