RESUMO

A taxonomic study was carried out on strain yzlin-01T, isolated from Dongshan Island seawater. The bacterium was Gram-stain-negative, catalase-positive, oxidase-negative, rod-shaped, and motile by polar flagella. Growth was observed at temperatures of 10-40 °C, at salinities of 0.5-18 %, and at pH of 6-10. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain yzlin-01T belonged to the genus Halomonas, with the highest sequence similarity to Halomonas malpeensis YU-PRIM-29T (96.7 %), followed by Halomonas johnsoniae T68687T (96.4 %) and Halomonas gomseomensis M12T (96.4 %), and other species of the genus Halomonas (93.4-96.3 %). The ANI and digital DNA-DNA hybridization estimate values between strain yzlin-01T and the closest type strain Halomonas malpeensis YU-PRIM-29T were 77.44 and 21.6 %, respectively. The principal fatty acids were summed feature 8 (consisting of C18 : 1 ω7c and/or C18 : 1 ω6c; 55.7 %), C16 : 0 (20.6 %), C12 : 0 3-OH (6.8 %), summed feature 3 (consisting of C16 : 1 ω7c and/or C16 : 1 ω6c; 5.1 %). The G+C content of the chromosomal DNA was 60.0 mol %. The respiratory quinone was identified as Q-9 (100 %). Phosphatidylethanolamine, phosphatidylglycerol, diphosphatidylglycerol, aminophospholipid, and three unidentified phospholipids were present. Combined genotypic and phenotypic data suggest that strain yzlin-01T represents a novel species within the genus Halomonas, for which the name Halomonas dongshanensis sp. nov. is proposed, with the type strain yzlin-01T (=GDMCC 1.3202T=KCTC 92467T).

OBJECTIVE: Treatment with TNF-α inhibitors improve psoriasis with minimize/minor neutrophils infiltration and CXCL-1/8 expression in psoriatic lesions. However, the fine mechanism of TNF-α initiating psoriatic inflammation by tuning keratinocytes is unclear. Our previous research identified the deficiency of intracellular galectin-3 was sufficient to promote psoriasis inflammation characterized by neutrophil accumulation. This study aims to investigate whether TNF-α participated in psoriasis development through dysregulating galectin-3 expression. METHODS: mRNA levels were assessed through quantitative real-time PCR. Flow cytometry was used to detect cell cycle/apoptosis. Western blot was used to evaluate the activation of the NF-κB signaling pathway. HE staining and immunochemistry were used to detect epidermal thickness and MPO expression, respectively. Specific small interfering RNA (siRNA) was used to knock down hsa-miR-27a-3p while plasmids transfection was used to overexpress galectin-3. Further, the multiMiR R package was utilized to predict microRNA-target interaction. RESULTS AND DISCUSSION: We found that TNF-α stimulation altered cell proliferation and differentiation and promoted the production of psoriasis-related inflammatory mediators along with the inhibition of galectin-3 expression in keratinocytes. Supplement of galectin-3 could counteract the rise of CXCL-1/8 but not the other phenotypes of keratinocytes induced by TNF-α. Mechanistically, inhibition of the NF-κB signaling pathway could counteract the decrease of galectin-3 and the increase of hsa-miR-27a-3p expression whereas silence of hsa-miR-27a-3p could counteract the decrease of galectin-3 expression induced by TNF-α treatment in keratinocytes. Intradermal injection of murine anti-CXCL-2 antibody greatly alleviated imiquimod-induced psoriasis-like dermatitis. CONCLUSION: TNF-α initiates psoriatic inflammation by increasing CXCL-1/8 in keratinocytes mediated by the axis of NF-κB-hsa-miR-27a-3p-galectin-3 pathway.

Functional and structural adaptation of common carotid artery could be one of the important causes of postflight orthostatic intolerance after microgravity exposure, the mechanisms of which remain unclear. Recent evidence indicates that long-term spaceflight increases carotid artery stiffness, which might present a high risk to astronaut health and postflight working ability. Studies have suggested that vascular calcification is a common pathological change in cardiovascular diseases that is mainly manifested as an increase in vascular stiffness. Therefore, this study investigated whether simulated microgravity induces calcification of common carotid artery and to elucidate the underlying mechanisms. Four-week-old hindlimb-unweighted (HU) rats were used to simulate the deconditioning effects of microgravity on cardiovascular system. We found that simulated microgravity induced vascular smooth muscle cell (VSMC) osteogenic differentiation and medial calcification, increased receptor activator of nuclear factor κB (NF-κB) ligand (RANKL) and RANK expression, and enhanced NF-κB activation in rat common carotid artery. In vitro activation of the RANK pathway with exogenous RANKL, a RANK ligand, increased RANK and osteoprotegerin (OPG) expression in HU rats. Moreover, the expression of osteogenic markers and activation of NF-κB in HU rats were further enhanced by exogenous RANKL but suppressed by the RANK inhibitor osteoprotegerin fusion protein (OPG-Fc). These results indicated that the OPG/RANKL/RANK system modulates VSMC osteogenic differentiation and medial calcification of common carotid artery in simulated microgravity rats by regulating the NF-kB pathway.

BACKGROUND As we know, chemotherapy resistance is a critical factor leading to recurrence and metastasis of nonsmall-cell lung cancer (NSCLC). To clarify the key target and potential mechanism of resistance to gemcitabine (GEM) in NSCLC, we selected Gene Expression Omnibus Data Set and statistically analyzed a parent cell group and a GEM-resistant cell group. Results showed that the expression of troponin C1, slow skeletal and cardiac type (TNNC1) in GEM-resistant cells was higher than in parent cells, which implies that TNNC1 was associated with GEM resistance in lung cancer cells. MATERIAL AND METHODS TNNC1 expression level was detected by reverse transcription-quantitative polymerase chain reaction or western blot in GEM-resistant patient serum and cell lines. It could reduce or increase autophagy response and GEM resistance accordingly by inhibition of the short interfering ribonucleic acid or by forced overexpression of TNNC1 viruses in A549 cell line and GEM-resistant cell line (A549/GemR) respectively. Blocking autophagy with 3-methyladenine increased the sensitivity of chemotherapy confirmed by flow cytometry and microtubule-associated protein 1A/1B - light chain 3 punctate assay. What's more, in a loss-of-function model, silencing of forkhead box 03 (FOXO3) in A549/GemR cells could rescue the autophagy weakened by TNNC1. RESULTS TNNC1 promoted GEM chemoresistance of NSCLC by activating cytoprotective autophagy, regulated negatively by FOXO3. This research may provide a completely new strategy for NSCLC treatment. CONCLUSIONS Targeting the TNNC1/FOXO3 signaling pathway in NSCLC may be a novel strategy to combat GEM resistance.

Rényi entropy as a generalization of the Shannon entropy allows for different averaging of probabilities of a control parameter α. This paper gives a new perspective of the Kalman filter from the Rényi entropy. Firstly, the Rényi entropy is employed to measure the uncertainty of the multivariate Gaussian probability density function. Then, we calculate the temporal derivative of the Rényi entropy of the Kalman filter's mean square error matrix, which will be minimized to obtain the Kalman filter's gain. Moreover, the continuous Kalman filter approaches a steady state when the temporal derivative of the Rényi entropy is equal to zero, which means that the Rényi entropy will keep stable. As the temporal derivative of the Rényi entropy is independent of parameter α and is the same as the temporal derivative of the Shannon entropy, the result is the same as for Shannon entropy. Finally, an example of an experiment of falling body tracking by radar using an unscented Kalman filter (UKF) in noisy conditions and a loosely coupled navigation experiment are performed to demonstrate the effectiveness of the conclusion.

A non-linear filtering algorithm based on the alpha-divergence is proposed, which uses the exponential family distribution to approximate the actual state distribution and the alpha-divergence to measure the approximation degree between the two distributions; thus, it provides more choices for similarity measurement by adjusting the value of α during the updating process of the equation of state and the measurement equation in the non-linear dynamic systems. Firstly, an α -mixed probability density function that satisfies the normalization condition is defined, and the properties of the mean and variance are analyzed when the probability density functions p ( x ) and q ( x ) are one-dimensional normal distributions. Secondly, the sufficient condition of the alpha-divergence taking the minimum value is proven, that is when α ≥ 1 , the natural statistical vector's expectations of the exponential family distribution are equal to the natural statistical vector's expectations of the α -mixed probability state density function. Finally, the conclusion is applied to non-linear filtering, and the non-linear filtering algorithm based on alpha-divergence minimization is proposed, providing more non-linear processing strategies for non-linear filtering. Furthermore, the algorithm's validity is verified by the experimental results, and a better filtering effect is achieved for non-linear filtering by adjusting the value of α .

Car ownership in China reached 194 million vehicles at the end of 2016. The traffic congestion index (TCI) exceeds 2.0 during rush hour in some cities. Inefficient processing for minor traffic accidents is considered to be one of the leading causes for road traffic jams. Meanwhile, the process after an accident is quite troublesome. The main reason is that it is almost always impossible to get the complete chain of evidence when the accident happens. Accordingly, a police and insurance joint management system is developed which is based on high precision BeiDou Navigation Satellite System (BDS)/Global Positioning System (GPS) positioning to process traffic accidents. First of all, an intelligent vehicle rearview mirror terminal is developed. The terminal applies a commonly used consumer electronic device with single frequency navigation. Based on the high precision BDS/GPS positioning algorithm, its accuracy can reach sub-meter level in the urban areas. More specifically, a kernel driver is built to realize the high precision positioning algorithm in an Android HAL layer. Thus the third-party application developers can call the general location Application Programming Interface (API) of the original standard Global Navigation Satellite System (GNSS) to get high precision positioning results. Therefore, the terminal can provide lane level positioning service for car users. Next, a remote traffic accident processing platform is built to provide big data analysis and management. According to the big data analysis of information collected by BDS high precision intelligent sense service, vehicle behaviors can be obtained. The platform can also automatically match and screen the data that uploads after an accident to achieve accurate reproduction of the scene. Thus, it helps traffic police and insurance personnel to complete remote responsibility identification and survey for the accident. Thirdly, a rapid processing flow is established in this article to meet the requirements to quickly handle traffic accidents. The traffic police can remotely identify accident responsibility and the insurance personnel can remotely survey an accident. Moreover, the police and insurance joint management system has been carried out in Wuhan, Central China's Hubei Province, and Wuxi, Eastern China's Jiangsu Province. In a word, a system is developed to obtain and analyze multisource data including precise positioning and visual information, and a solution is proposed for efficient processing of traffic accidents.

Gaining knowledge of the intrinsic topology of a complex dynamical network is the precondition to understand its evolutionary mechanisms and to control its dynamical and functional behaviors. In this article, a general framework is developed to recover topologies of complex networks with completely unknown node dynamics based on Taylor expansion and compressive sensing. Numerical simulations illustrate the feasibility and effectiveness of the proposed method. Moreover, this method is found to have good robustness to weak stochastic perturbations. Finally, the impact of two major factors on the topology identification performance is evaluated. This method provides a natural and direct point to reconstruct network topologies from measurable data, which is likely to have potential applicability in a wide range of fields.

Introduction: Accessory breast cancer (ABC) is an extremely rare condition, particularly the presence of triple-negative ABC with ipsilateral invasive in situ breast cancer. Binary breast tumors are controversial in terms of surgical methods and comprehensive treatment. Case presentation: We share the case of a 64-year-old postmenopausal woman who presented with an underarm mass for 3 months. Ultrasonography and computed tomography suggested possible breast cancer with axillary lymph node metastasis. The patient underwent a left modified radical mastectomy combined with axillary lymph node dissection. The postoperative pathology confirmed a binary tumor, prompting us to initiate comprehensive treatment. Conclusion: We present the treatment approach for a rare case of triple-negative para-breast cancer complicated with carcinoma in situ of the breast, hoping to contribute new therapeutic ideas for the treatment of this disease.

Three new polyoxygenated C(18)-dibenzocyclooctadiene lignans, arisanschinins M and N (1 and 2) and schisphenin A (3), together with eight related metabolites (4-11), were isolated from the fruits of Schisandra arisanensis and Schisandra sphenanthera, respectively. The structures of 1-3 were elucidated on the basis of extensive spectroscopic and 2D NMR (HSQC, HMBC, and NOESY) analyses. The configuration of the biphenyl moiety in the octadiene ring was determined by circular dichroism (CD). Compound 1 possessed an unprecedented 3-(1-hydroxypropan-2-yl)-3-methyl-1,4-dioxo-2-one lactonide ring system attaching at C-6/C-14. Pharmacological studies revealed that compounds 3, 4, 6, 7, and 10 exhibited significant anti-hepatic fibrosis activity, while 9 and 11 showed cytotoxicity against HSC-T6 cells. The biogenetic pathway for compound 1 was also proposed.

Active control of ion transport in nanoscale channels is attracting increasing attention. Recently, experimental and theoretical results have verified that depending on the charged surface of nanopores, the solution inside nanopores can contain either negative or positive ions, which does not happen in macroscale channels. However, the control of the surface chemistry of synthetic nanopores is difficult and the design of nanotubes with novel recognition mechanisms that regulate the ionic selectivity of negative and positive charges remains a challenge. We present here a design for an ion-selective nanopore that is controllable by external charges. Our molecular dynamics simulations show that this remarkable selectivity can be switched from predominantly negative to positive ions and that the magnitude of the ionic flux can be varied by changing the distance of the external charges. The results suggest that the hydration structures around ions play a prominent role in the selectivity process, which is tuned by the external charge. These studies may be useful for developing ways to control the behavior, properties and chemical composition of liquids and provide possible technical applications for nanofluidic field effect transistors.

Small-molecule vascular disrupting agents (VDAs) target the established tumor blood vessels, resulting in rapidly and selectively widespread ischemia and necrosis of central tumor; meanwhile, blood flow in normal tissues is relatively unaffected. Although VDAs therapy is considered an important option for treatment, its use is still limited. The tumor cells at the periphery are less sensitive to vascular shutdown than those at the center, and subsequently avoid a nutrient-deprived environment. This phenomenon is referred to as tumor resistance to VDAs treatment. The viable periphery rim of tumor cells contributes to tumor regeneration, metastasis, and ongoing progression. However, there is no systematic review of the plausible mechanisms of repopulation of the viable tumor cells following VDAs therapy. The purpose of this review is to provide insights into mechanisms of tumor surviving small-molecule VDAs therapy, and the synergetic treatment to the remaining viable tumor cells at the periphery.

The goal of visual navigation is steering an agent to find a given target object with current observation. It is crucial to learn an informative visual representation and robust navigation policy in this task. Aiming to promote these two parts, we propose three complementary techniques, heterogeneous relation graph (HRG), a value regularized navigation policy (VRP), and gradient-based meta learning (ML). HRG integrates object relationships, including object semantic closeness and spatial directions, e.g., a knife is usually co-occurrence with bowl semantically or located at the left of the fork spatially. It improves visual representation learning. Both VRP and gradient-based ML improve robust navigation policy, regulating this process of the agent to escape from the deadlock states such as being stuck or looping. Specifically, gradient-based ML is a type of supervision method used in policy network training, which eliminates the gap between the seen and unseen environment distributions. In this process, VRP maximizes the transformation of the mutual information between visual observation and navigation policy, thus improving more informed navigation decisions. Our framework shows superior performance over the current state-of-the-art (SOTA) in terms of success rate and success weighted by length (SPL). Our HRG outperforms the Visual Genome knowledge graph on cross-scene generalization with ≈ 56% and ≈ 39% improvement on Hits@ 5* (proportion of correct entities ranked in top 5) and MRR * (mean reciprocal rank), respectively. Our code and HRG datasets will be made publicly available in the scientific community.

Many organisms in nature such as beetles and cacti can survive in arid places by their own surface structures that are still able to collect mist. These surfaces have micro-nano structures that maintain a very low adhesion, allowing them to continuously collect and transport water. Here, we used a light curing three dimensional molding process to create a template for a water harvesting system inspired by the back of a beetle, a hydrogel-like beetle back surface for water transport. By changing the curvature structure of the water evacuation channels and altering the hydrophilic and hydrophobic properties of the surface, the designed large-scale artificial water harvesting study was made possible. The results show that if the surface has a proper curvature structure and hydrophobic density, the water collection on the super-impregnated surface is much higher than that on an ordinary hydrophobic surface. Based on this, a new efficient and environmentally friendly water collection scheme is proposed. The data show that the triangular tip structure imitating beetle-backed hydrogel surface collects the highest amount of water with a water weight of 16 g in 2 h. This study offers interesting prospects for designing a new generation of structural materials with a bionic structure distribution for high-efficiency water harvesting. The results of the study are useful for pushing the improvement of environmental-friendly water collection, transport and separation devices. Abbreviations: The dorsal shape of the beetle's back is critical for water collection. In this work, while redesigning the shape of the back of the beetle, the method of 3D printing the beetle back template was used to prepare the beetle back made of hydrogel, which greatly improved the water collection performance and has certain engineering application prospects.

The detection of biomarkers in blood macrophages is a new non-invasive cancer screening method, but its performance in early stage lung cancer screening remains undetermined. We evaluated the Apo10 and TKTL1 levels in blood macrophages of 156 early-stage lung cancer patients and 153 controls. APT (combination of Apo10 and TKTL1) level was significantly higher in the lung cancer group than that in the control group (P < 0.001). AUROC analysis showed that APT has high diagnostic value in differentiating early-stage lung cancer (AUC = 0.9132) and can be considered a biomarker for screening lung cancer patients from individuals with lung nodules.

Objective: This study aimed to assess the efficacy of Endoscopic Retrograde Cholangiopancreatography (ERCP), common bile duct exploration, and percutaneous transhepatic cholangiography combined with common bile duct exploration for treating choledocholithiasis with acute cholangitis, to guide management strategies. Methods: A retrospective evaluation was conducted on a cohort of 283 inpatients diagnosed with choledocholithiasis and acute cholangitis at the affiliated hospital. Patients were categorized into three groups: Group A (ERCP group), Group B(common bile duct exploration group), and Group C(PTCD combine common bile duct exploration group.) Parameters such as hepatic function recovery, inflammation level control, blood loss, postoperative hospital duration, and postoperative complications were compared. Results: All groups exhibited notable reductions in postoperative biochemical parameters including alanine aminotransferase (ALT), aspartate aminotransferase (AST), gamma-glutamyl transferase (GGT), total bilirubin (TBIL), and WBC (P < 0.05). Group A had the least blood loss(P < 0.05), and shortest hospital stay(P < 0.05), but a higher incidence of pancreatitis(P < 0.05), with a total of 8 cases occurred(7.3%). Group C had a shorter hospital stay compared to Group B(P < 0.05). Conclusion: For patients with fewer and smaller common bile duct stones and milder symptoms, it is recommended to primarily choose endoscopic retrograde cholangiopancreatography (ERCP), endoscopic sphincterotomy (EST), and endoscopic nasobiliary drainage (ENBD), it procedures offer quicker recovery and cause minimal trauma. For patients with numerous, larger common bile duct stones but stable conditions, bile duct exploration is recommended. For those with severe conditions and significant inflammation, PTCD and common bile duct exploration are advised.

The COVID-19 pandemic has resulted in the implementation of strict mitigation measures that have impacted the transmission dynamics of human respiratory syncytial virus (HRSV). The measures also have the potential to influence the evolutionary patterns of the virus. In this study, we conducted a comprehensive analysis comparing genomic variations and evolving characteristics of its neutralizing antigens, specifically F and G proteins, before and during the COVID-19 pandemic. Our findings showed that both HRSV A and B exhibited an overall chronological evolutionary pattern. For the sequences obtained during the pandemic period (2019-2022), we observed that the HRSV A distributed in A23 genotype, but formed into three subclusters; whereas the HRSV B sequences were relatively concentrated within genotype B6. Additionally, multiple positively selected sites were detected on F and G proteins but none were located at neutralizing antigenic sites of the F protein. Notably, amino acids within antigenic site III, IV, and V of F protein remained strictly conserved, while some substitutions occurred over time on antigenic site Ø, I, II and VIII; substitution S389P on antigenic site I of HRSV B occurred during the pandemic period with nearly 50% frequency. However, further analysis revealed no substitutions have altered the structural conformations of the antigenic sites, the vial antigenicity has not been changed. We inferred that the intensive public health interventions during the COVID-19 pandemic did not affect the evolutionary mode of HRSV.

Nervous necrosis virus (NNV) infection during larvae and juvenile stage in grouper (Epinephelus coioides) has caused severe economic losses in the aquaculture industry in Asia. The aims of this study were to evaluate the influence of recombinant Reishi protein, rLZ-8, on the innate immune responses and the viral resisting ability in fish. Groupers were fed with rLZ-8 supplemented diet (1.25-37.5 mg (rLZ-8)/kg(diet)), and the cytokine gene expression, innate immune responses, and survival rate after NNV challenge were examined. The fish fed with rLZ-8 diet showed 6- to 11-fold upregulated TNF-α and IL-1ß gene expression, along with significant increased respiratory burst and phagocytic activity. Moreover, feeding the fish with 37.5 mg/kg rLZ-8 diet elicited significant improvement in post viral challenge survival rate (85.7%). These discoveries indicated that rLZ-8 could be utilized as an ant-pathogen immunostimulant, and provided a new candidate to fight against NNV infection in fish.

Fog and moisture in nature are important freshwater resources, and the collection of these fog water is of great significance to arid regions. Inspired by the unique geometric structure of the spindle knot on spider silk, artificial fibers with periodic structures have been fabricated for water collection, which can effectively alleviate the problem of water shortage in arid areas. Traditional manufacturing methods are difficult to replicate the true shape of the spindle knot, and related research has encountered a bottleneck in improving water collection efficiency. 3D printing technology, which is different from traditional subtractive manufacturing, can directly replicate spider silk with periodic knots, making it possible to study water collection by artificial spider webs of various designs. Here, 3D printing technology is used to fabricate artificial spider webs with different geometric structures for efficient transportation and collection of water. In addition, the artificial spider web is treated with hydrophilic surfaces. In the humid environment for 2 h, the spider web with convex-concave multi-size spindle knots and multi-curvature connections has a maximum water collection capacity of 6.2g, and the mass of water collection is 35% higher than the existing best water collection artificial fibers. This work provides a sustainable and environmentally friendly route for the effective collection of humid air, and has certain reference value for the development of environmentally friendly water collection equipment.

The oxidation of a simple p-methoxyphenol derivative by HClO induces an intramolecular charge transfer from the end phenyl units to the middle benzoquinone, which leads to colorimetric and fluorescent changes. This detection can be run in aqueous solution with high selectivity over other reactive oxygen species.
