BACKGROUND: As a chronic inflammatory disease, diabetes mellitus (DM) contributes to the development of atherosclerosis (AS). However, how the NLRP3 inflammasome participates in diabetes-related AS remains unclear. Therefore, this study aimed to elucidate the mechanism through which NLRP3 uses high glucose (HG) levels to promote AS. METHODS: Serum and coronary artery tissues were collected from coronary artery disease (CAD) patients with and without DM, respectively. The expression of NLRP3 was detected, and the effects of this inflammasome on diabetes-associated AS were evaluated using streptozotocin (STZ)-induced diabetic apoE-/- mice injected with Adenovirus-mediated NLRP3 interference (Ad-NLRP3i). To elucidate the potential mechanism involved, ox-LDL-irritated human aortic smooth muscle cells were divided into the control, high-glucose, Si-NC, and Si-NLRP3 groups to observe the changes induced by downregulating NLRP3 expression. For up-regulating NLRP3, control and plasmid contained NLRP3 were used. TNF-α, IL-1ß, IL-6, IL-18, phosphorylated and total p38, JNK, p65, and IκBα expression levels were detected following the downregulation or upregulation of NLRP3 expression. RESULTS: Patients with comorbid CAD and DM showed higher serum levels and expression of NLRP3 in the coronary artery than those with only CAD. Moreover, mice in the Ad-NLRP3i group showed markedly smaller and more stable atherosclerotic lesions compared to those in other DM groups. These mice had decreased inflammatory cytokine production and improved glucose tolerance, which demonstrated the substantial effects of NLRP3 in the progression of diabetes-associated AS. Furthermore, using the siRNA or plasmid to downregulate or upregulate NLRP3 expression in vitro altered cytokines and the MAPK/NF-κB pathway. CONCLUSIONS: NLRP3 expression was significantly increased under hyperglycemia. Additionally, it accelerated AS by promoting inflammation via the IL/MAPK/NF-κB pathway.

The role of artificial intelligence (AI) in pathology offers many exciting new possibilities for improving patient care. This study contributes to this development by identifying the viability of the AICyte assistive system for cervical screening, and investigating the utility of the system in assisting with workflow and diagnostic capability. In this study, a novel scanner was developed using a Ruiqian WSI-2400, trademarked AICyte assistive system, to create an AI-generated gallery of the most diagnostically relevant images, objects of interest (OOI), and provide categorical assessment, according to Bethesda category, for cervical ThinPrep Pap slides. For validation purposes, 2 pathologists reviewed OOIs from 32,451 cases of ThinPrep Paps independently, and their interpretations were correlated with the original ThinPrep interpretations (OTPI). The analysis was focused on the comparison of reporting rates, correlation between cytological results and histologic follow-up findings, and the assessment of independent AICyte screening utility. Pathologists using the AICyte system had a mean reading time of 55.14 seconds for the first 3000 cases trending down to 12.90 seconds in the last 6000 cases. Overall average reading time was 22.23 seconds per case compared with a manual reading time approximation of 180 seconds. Usage of AICyte compared with OTPI had similar sensitivity (97.89% vs 97.89%) and a statistically significant increase in specificity (16.19% vs 6.77%) for the detection of cervical intraepithelial neoplsia 2 and above lesions. When AICyte was run alone at a 50% negative cutoff value, it was able to read slides with a sensitivity of 99.30% and a specificity of 9.87%. When AICyte was run independently at this cutoff value, no sole case of high-grade squamous intraepithelial lesions/squamous cell carcinoma squamous lesion was missed. AICyte can provide a potential tool to help pathologists in both diagnostic capability and efficiency, which remained reliable compared with the baseline standard. Also unique for AICyte is the development of a negative cutoff value for which AICyte can categorize cases as "not needed for review" to triage cases and lower pathologist workload. This is the largest case number study that pathologists reviewed OOI with an AI-assistive system. The study demonstrates that AI-assistive system can be broadly applied for cervical cancer screening.

We experimentally elucidate the impact dynamics of ethylene glycol (EG) droplets laden with both hydrophilic and hydrophobic SiO2 nanoparticles (NPs) onto a flat heated surface in non-boiling, boiling, and Leidenfrost regimes. We use seven nanofluid concentrations (Cp), ranging from 0.89 to 64.3 wt %, and control the surface temperature (Ts) between 100 and 400 °C, while the nanofluid droplet's impact velocity is constant at 0.22 ± 0.02 m/s. Phase diagrams of impact outcomes are established to illustrate the effect of the additive nanoparticles on the droplets' impact dynamics, revealing that nanoparticles modify droplet impact behaviors differently in each regime. In the non-boiling regime, the droplet spreading profile remains unaffected by nanoparticles up to Cp < 11.9 wt % before reaching the maximum spreading diameter (ßmax). For nanofluid drops with higher nanofluid concentration, the increasing viscosity with concentration is likely to be the primary factor that affects the droplets' spreading profile in the non-boiling regime Ts ≲ Tsat ≈ 200 °C, as the saturation temperature. In the boiling regime 200 °C < Ts ≲ 350 °C, a small amount of nanoparticle addition (Cp = 0.89 wt %) promotes atomization regardless of nanoparticle wettability. Finally, manifested in a complete rebound due to an intervening vapor layer, the Leidenfrost temperature (TL) of the nanofluid droplets is affected by both nanofluid concentration and nanoparticles' wettability. The nanofluid droplets' TL increases with higher nanofluid concentration; moreover, this Leidenfrost temperature increment is more significant for EG droplets laden with hydrophobic nanoparticles. Our results quantitatively reveal the significant influence of nanoparticle concentrations and wettability on drop spreading, impact outcome, and Leidenfrost temperature on heat surfaces, potentially benefiting applications in coating, spraying, and cooling.

The higher prevalence of metabolic syndrome (MetS) in women after menopause is associated with a decrease in circulating 17ß-oestradiol. To explore novel treatments for MetS in women with oestrogen deficiency, we studied the effect of exogenous butyrate on diet-induced obesity and metabolic dysfunctions using ovariectomized (OVX) mice as a menopause model. Oral administration of sodium butyrate (NaB) reduced the body fat content and blood lipids, increased whole-body energy expenditure, and improved insulin sensitivity. Additionally, NaB induced oestrogen receptor alpha (ERα) expression, activated the phosphorylation of AMPK and PGC1α, and improved mitochondrial aerobic respiration in cultured skeletal muscle cells. In conclusion, oral NaB improves metabolic parameters in OVX mice with diet-induced obesity. Oral supplementation with NaB might provide a novel therapeutic approach to treating MetS in women with menopause. Video Abstract.

BACKGROUND: Norepinephrine is effective in preventing spinal hypotension during cesarean birth; however, an optimal regimen has not been determined. We hypothesized that an initial bolus of norepinephrine improves efficacy of spinal hypotension prophylaxis beyond continuous norepinephrine alone. METHODS: In this double-blind, controlled study, 120 patients scheduled for cesarean birth under spinal anesthesia were randomly allocated to receive a norepinephrine bolus at 0.05 or 0.10 µg/kg, followed by norepinephrine infusion at a rate of 0.05 µg·kg -1 ·min -1 . The primary outcome was the frequency of spinal hypotension during cesarean birth. The doses of the rescue drug (phenylephrine), frequency of nausea or vomiting, duration of hypotension, frequency of intraoperative hypertension, frequency of bradycardia, and fetal outcomes were also compared. RESULTS: One-hundred-fifteen patients were included in the analysis. Compared with the 0.05 µg/kg group, the frequency of spinal hypotension was lower in the 0.10 µg/kg group (20.7% vs 45.6%; odds ratio [OR], 0.31; 95% confidence interval (CI), 0.14-0.71; P = .004). Fewer rescue doses of phenylephrine (0 [0,0] vs 0 [0,80]; 95% CI for the difference, 0 (0-0); P = .006) were required, and the frequency of nausea or vomiting was lower (5.2% vs 17.5%; OR, 0.26; 95% CI, 0.07-0.99; P = .04) in the 0.10 µg/kg group. The duration of hypotension was shorter in the 0.10 µg/kg group than that in the 0.05 µg/kg group (0 [0,0] vs 0 [0,2]; 95% CI for the difference, 0 [0-0]; P = .006). The incidence of intraoperative hypertension, frequency of bradycardia, and fetal outcomes were comparable between the 2 groups. CONCLUSIONS: With a fixed-rate norepinephrine infusion of 0.05 µg·kg -1 ·min -1 , the 0.10 µg/kg initial bolus was more effective in reducing the incidence of spinal hypotension compared with the 0.05 µg/kg initial bolus.

The role of Dectin-2 (gene symbol, Clec4n) in house dust mite (HDM) induced Th2 immune response and the exact mechanism remains controversial. In this study, we illustrated that, Clec4n-/- mice had decreased Th2 immune response following HDM challenge, which may ascribe to dramatically reduced type 2 conventional dendritic cells (cDC2s) in lung of Clec4n-/- mice, as cDC2s from lung of Clec4n-/- mice after challenging had less ability to induce Th2 response with decreased production of IL-4/IL-13. Further in vitro experiments showed the activation of Clec4n-/--BMDCs significantly decreased after HDM stimulation accompanied with decreased activation of Syk-NF-κB and Syk-JNK signal pathway. Importantly, Dectin-2 expression in PBMCs from asthmatic patients was significantly higher than that in healthy controls. Taken together, these results demonstrated that Dectin-2 could promote cDC2s activation in lung, which polarizes Th2 immune response outlining a novel mechanism of asthma development.

AIMS: To detect abnormal eye movements in Parkinson's disease and explore its correlation with clinical characteristics and their value for diagnosis. METHODS: We recruited forty-nine Parkinson's disease patients, including 35 early Parkinson's disease patients (Hoehn-Yahr: 1 to 2 stage) and 14 advanced Parkinson's disease patients (Hoehn-Yahr: 3 to 5 stage) and 23 healthy controls. Clinical manifestations in Parkinson's disease patients were recorded. Oculomotor performances including fixation, gaze, saccade in horizontal and vertical direction, and smooth pursuit in horizontal and vertical direction were measured by video-oculography. RESULTS: We found that five oculomotor parameters, namely square wave jerk frequency, latency of downward saccade, latency of upward saccade, accuracy of upward saccade, and gain of horizontal smooth pursuit were significantly different in Parkinson's disease patients and controls. When combining all these five parameters, we got the diagnostic sensitivity of 78.3% and specificity of 95.2%. More deficits in upward saccade than in other directions were associated with disease duration and progression of Parkinson's disease. CONCLUSION: Our primary study suggests that oculomotor examination might serve as an aid in the clinical assessment of Parkinson's disease patients and differentiating between early Parkinson's disease and normal controls.

Objective: To evaluate the efficacy of subcutaneous etonogestrel implants for adenomyosis.Methods: We conducted a clinical observational study of 20 patients suffering from adenomyosis treated with subcutaneous etonogestrel implants from August 2015 to July 2017 and followed up for 36 months. We evaluated the efficacy of subcutaneous etonogestrel implants primarily based on the following indicators: the pictorial blood loss assessment chart (PBAC) for menstrual blood volume, changes in bleeding patterns, the visual analog scale (VAS) pain score for dysmenorrhea, uterine volume, serum cancer antigen 125 (CA125) levels, hemoglobin levels and side effects.Results: During the 3 years of follow-up, subcutaneous etonogestrel implants were removed from six patients, among whom one was diagnosed with endometrial cancer, four had an increased menstrual blood volume, and one entered menopause. In total, 14 patients were treated with subcutaneous etonogestrel implants for 3 years. Among these patients, the number of patients with heavy menstrual bleeding and high PBAC and VAS scores and serum CA125 levels was significantly decreased after implantation compared with that before implantation. In the eight patients with anemia, hemoglobin levels increased gradually. However, the uterine volumes did not significantly change. Bleeding patterns were changed but were tolerable.Conclusion: Subcutaneous etonogestrel implants represent a new option for the clinical treatment of adenomyosis for patients who refuse surgery.

Current works focus on detecting macromolecule crowding effects on the phase separation of the mixture between semi-flexible polymer and crowders (hydrophilic polymers) in confined space by Monte Carlo simulations. With the increasing addition of crowders into the spherical confined space, the semi-flexible polymer was first compressed into a condensed state from the initial coil state, and then the condensed conformation expanded and deposited on the inner surface of the spherical confined space with an extended state. The phase diagram in the phase space of the volume fraction of crowders and the scaled radius of spherical confined space by crowder diameter, and the direct conformation transition of semi-flexible polymer have validated the phase transition process successfully. In addition, the deposition of extended conformation on the inner surface of the spherical confined space was qualified by the vertex density, its curve shifted along the radial direction with the increasing volume fraction of crowder. During the phase separation process, the critical volume fraction φ∗ relates to the crowder diameter approximately linearly and the relation between the critical volume fraction and the crowder diameter strongly depends on the size of the spherical confined space.

A large number of polyethylene terephthalate (PET) bottles are discarded daily after usage. Thus, plastic bottle recycling has elicited considerable attention in recent years. In this context, this study aims to quantify the environmental and economic impacts of blanket production from 100% recycled waste plastic bottles in China through a life cycle assessment coupled with life cycle costing method. In addition, the environmental impact of replacing coal with natural gas and solar energy was evaluated. Results show that impact categories of global warming and fossil depletion have significant influence on the overall environment. Carbon dioxide, water, iron, coal and chromium (VI) to water are the main contributors to the overall environmental burden. The internal and external costs are $6433/metric ton and $370/metric ton, respectively. Analysis results indicate that the optimization of organic chemicals, recycled polyester filament and steam production processes can reduce environmental and economic burdens substantially. Energy substitutions with natural gas and the use of solar photovoltaic in steam production and electricity generation are effective measures for decreasing environmental impacts. Finally, suggestions based on research results and the current status of waste plastic bottle recycling in China are proposed.

Development of adventitious roots (ARs) at the base of the shoot is an important adaptation of plants to waterlogging stress; however, its physiological mechanisms remain unclear. Here, we investigated the regulation of AR formation under waterlogged conditions by hormones and reactive oxygen species (ROS) in Cucumis sativus L., an agriculturally and economically important crop in China. We found that ethylene, auxin, and ROS accumulated in the waterlogged cucumber plants. On the other hand, application of the ethylene receptor inhibitor 1-methylcyclopropene (1-MCP), the auxin transport inhibitor 1-naphthylphthalamic acid (NPA), or the NADPH oxidase inhibitor diphenyleneiodonium (DPI) decreased the number of ARs induced by waterlogging. Auxin enhanced the expression of ethylene biosynthesis genes, which led to ethylene entrapment in waterlogged plants. Both ethylene and auxin induced the generation of ROS. Auxin-induced AR formation was inhibited by 1-MCP, although ethylene-induced AR formation was not inhibited by NPA. Both ethylene- and auxin-induced AR formation were counteracted by DPI. These results indicate that auxin-induced AR formation is dependent on ethylene, whereas ethylene-induced AR formation is independent of auxin. They also show that ROS signals mediate both ethylene- and auxin-induced AR formation in cucumber plants.

The rational allocation of resources is crucial to suppress the outbreak of epidemics. Here, we propose an epidemic spreading model in which resources are used simultaneously to prevent and treat disease. Based on the model, we study the impacts of different resource allocation strategies on epidemic spreading. First, we analytically obtain the epidemic threshold of disease using the recurrent dynamical message passing method. Then, we simulate the spreading of epidemics on the Erdos-Rényi (ER) network and the scale-free network and investigate the infection density of disease as a function of the disease infection rate. We find hysteresis loops in the phase transition of the infection density on both types of networks. Intriguingly, when different resource allocation schemes are adopted, the phase transition on the ER network is always a first-order phase transition, while the phase transition on the scale-free network transforms from a hybrid phase transition to a first-order phase transition. Particularly, through extensive numerical simulations, we find that there is an optimal resource allocation scheme, which can best suppress epidemic spreading. In addition, we find that the degree heterogeneity of the network promotes the spreading of disease. Finally, by comparing theoretical and numerical results on a real-world network, we find that our method can accurately predict the spreading of disease on the real-world network.

BACKGROUND: Estimates of survival after curative colorectal cancer (CRC) surgery are the basis of patient care and treatment planning. A nomogram is a useful tool for individualized cancer prognosis. METHODS: A total of 2450 patients with nonmetastatic CRC were included to develop a nomogram. Prognostic factors were identified and integrated by the Cox proportional hazards model. Then, we developed and validated a prognostic nomogram. The performance of this model was assessed by the concordance index (C-index) and a calibration curve. The nomogram was internally validated by bootstrapping and externally validated with a separate database of 299 patients from The Cancer Genome Atlas. RESULTS: Age, T stage, N stage, histological type, tumor location, lymph-vascular invasion, preoperative carcinoembryonic antigen, and sample lymph nodes were integrated into the nomogram. The C-index of the nomogram for predicting overall survival was higher than that of the seventh edition American Joint Committee on Cancer TNM staging system (training data set, 0.76 vs 0.68, respectively; P < 0.001; validation data set, 0.78 vs 0.69, respectively; P = 0.003). CONCLUSION: We developed a prognostic nomogram for patients with nonmetastatic CRC, which could provide a more individualized outcome prognostication than that afforded by the TNM staging system by using common clinicopathologic factors.

BACKGROUND: Many patients with colorectal cancer are elderly with competing comorbidities. When constructing nomogram for assessing survival, we should consider the influence of competing risk. A competing-risks nomogram was developed to estimate the probability of death due to colorectal cancer for patients after curative surgery. METHODS: A total of 2442 patients with non-metastatic colorectal cancer were included to develop competing-risks nomogram. Competing-risks nomogram were established based on the results of Fine and Gray competing-risks proportional hazards model. To maximize the accuracy of prediction, model selection was not carried out, and non-linear continuous variables were flexibly modeled with restricted cubic splines. The nomogram was internal-validated by bootstrapping, and externally validated with a separate database of 299 patients from The Cancer Genome Atlas. The performance of this model was assessed by concordance index and a calibration curve. RESULTS: There were 332 patients died of colorectal cancer and 46 died of other causes during the follow-up period. Age, T stage, N stage, histological type, tumor location, adjuvant chemotherapy, preoperative carcinoembryonic antigen, lymph vascular invasion, lymph node ratio and sample lymph nodes were integrated into competing-risks nomogram. The competing-risks nomogram for predicting probability of death due to colorectal cancer with a concordance index of 0.768, ameliorating the stratification provided by the seventh edition tumor-node-metastasis staging system of the American Joint Committee on Cancer (AJCC). The concordance index for validation dataset was 0.783. CONCLUSION: We developed and externally validated a competing-risks nomogram for Chinese Han patients with non-metastatic colorectal cancer, which could provide probability of death from colorectal cancer in the presence of competing risks.

China, as the world's largest crude steel producer, is suffering from water scarcity and pollution. However, only a few systematic analyses on the environmental burdens and improvements of China's crude steel production have been conducted. Therefore, it is important for research to be done how China's steel industry can be improved in environment management. To help decision-makers understand this, a life cycle water footprint analysis including gray and blue water was performed based on the methodology prescribed in the ISO 14046 standard. A life cycle assessment was also conducted to improve the environmental performance of the steel industry. Results of these assessments revealed that gray water footprint, which is mainly derived from aquatic eutrophication, carcinogens, and non-carcinogens, is higher than blue water footprint. Optimizing indirect processes, including iron ore mining, magnesium oxide production, transportation, and electricity generation, played dominant roles in the reduction of gray water footprint. Furthermore, COD, Cr (VI), phosphate, BOD5, Hg, As, nitrogen oxides, particulates, and sulfur dioxide were the key substances for environmental improvements. The underestimation of direct water footprint showed the importance and urgency of implementing scientific and adequate monitoring indicators. Meanwhile, the environmental burden can be reduced by adopting a reasonable location of the steel industry on the basis of regional water resources and actual transportation status, improving the efficiency of raw material consumption, and optimizing the power structure.

Utilizing halogens as traceless directing goups represents an attractive strategy for C-H functionalization. A two C-H alkylation system, initiated by the oxidative addition of organohalides to Pd0 , has been developed. The first reaction involves an intermolecular alkylation of palladacycles to form C(sp3 )-C(sp2 ) bonds followed by C(sp2 )-H activation/cyclization to deliver alkylated benzocyclobutenes as the final products. In the second reaction, two C-C bonds are formed by the reaction of palladacycles with CH2 Br2 , and provides a facile and efficient method for the synthesis of indanes. The alkylated benzocyclobutene products can be transformed into tricyclic hyrocarbons, and the indane derivatives are essential structural motifs in bioactive and odorant molecules.

OBJECTIVE: To investigate the changes and clinical significance of lymphocyte subsets in infants with bronchitis, bronchopneumonia, and bronchiolitis. METHODS: A total of 111 children with bronchitis, 418 children with bronchopneumonia, and 83 children with bronchiolitis were enrolled as disease groups, and 235 healthy children were enrolled as control group. Flow cytometry was applied to measure lymphocyte subsets. RESULTS: The bronchitis group had significantly lower numbers of T cells and CD3+CD8+ T cells than the control group (P<0.05). The bronchopneumonia group had significantly lower numbers of T cells and CD3+CD8+ T cells, a significantly higher number of T helper (Th) cells, and a significantly higher CD4/CD8 ratio than the control group, as well as a significantly higher number of Th cells than the bronchitis group. Compared with the children with mild bronchopneumonia, those with severe bronchopneumonia showed a reduction in T cells and an increase in B cells (P<0.05). The bronchiolitis group had a significantly higher number of Th cells, a significantly higher CD4/CD8 ratio, and a significantly lower number of CD3+CD8+ T cells than the control group (P<0.01). The disease groups showed a significantly higher number of B cells and a significantly lower number of natural killer cells than the control group (P<0.05). CONCLUSIONS: A low, disturbed cellular immune function and a high humoral immune function are involved in the development and progression of lower respiratory tract infectious diseases. The changes in immune function are related to the type and severity of diseases.

Volatile organic compounds (VOCs) are considered as important precursors of ozone in the air, while the contribution of VOCs from pesticide application (PVOCs) to ozone production is unknown. Utilizing data from the Ministry of Agriculture and Rural Affairs of the People's Republic of China and ChinaCropPhen1km, this paper developed PVOC emission inventories with a resolution of 1 km for the main crops (rice, maize, and wheat) from 2012 to 2019 in China. The results revealed that pesticide application is an important VOC emission source in China. Specially, the PVOC emissions from the major grain-producing regions in June accounted for approximately 30% of the annual total PVOC emissions in the local regions. The simulation with the Weather Research and Forecasting Community Multiscale Air Quality model (WRF-CMAQ) indicated that the PVOC emissions increased the mean maximum daily 8-hour average (MDA8) ozone concentration across China by 2.5 ppb in June 2019. During the same period, PVOCs in the parts of North China Plain contributed 10% of the ozone formation. Under the comprehensive emission reduction scenario, it is anticipated that by 2025, the joint implementation of measures including reducing pesticide application, improving pesticide utilization efficiency and promoting solvent substitution will decrease PVOC emissions by 60% compared with 2019, thereby mitigating ozone pollution.

To obtain high-quality joints of Al/steel dissimilar materials, a new extrinsic-riveting friction stir lap welding (ERFSLW) method was proposed combining the synthesis advantages of mechanical riveting and metallurgical bonding. SiC-reinforced Al matrix composite bars were placed in the prefabricated holes in Al sheets and steel sheets, arranged in a zigzag array. The bars were stirred and mixed with Al sheets under severe plastic deformation (SPD), forming composite rivets to strengthen the mechanical joining. SiC particles were uniformly dispersed in the lower part of the welding nugget zone (WNZ). The smooth transition between the SiC mixed zone and extrinsic-riveting zone (ERZ) ensured the metallurgical bonding. The maximum tensile shear load of the joints reached 7.8 kN and the maximum load of the weld per unit length was 497 N/mm. The fracture occurred at the interface between the rivets and steel sheets rather than the conventional Al/steel joining interface. Moreover, ERFSLW can prolong the service life of joints due to three fracture stages. This method can be further extended to the welding of other dissimilar materials that conform to the model of "soft/hard".

China as a major coal-consuming economy faces the challenge of balancing economic development and carbon neutrality goal. This paper incorporates both efficiency-based and equity-based carbon neutrality policies into a numerical model to quantitatively assess how coal reduction under various carbon-neutral policies affects energy mix, economic growth, and industrial structures by 2060. Results show the nationwide coal intensity will ultimately plunge by over 95% from 2017 to 2060, mainly attributed to the coal-phasing-out in most industries. National Gross Domestic Product losses reaches 4,951 billion USD in efficiency-based scenarios by 2060, and the economic losses are even more severe in less developed provinces, especially provinces in Northern China. Although the equity-based policy can reduce the economic burden for the Northern China, the equity-based policy is accompanied by a significant regional shift in coal across the country: eastern coal-intense industries will be relocated northward, leading to increases in embodied coal consumption.
